Optimizing Conditions of Coagulation for Removal of Natural Organic Matter (NOM) : Comparison of Removal Efficiency of NOM When Using Bench-Scale Inline Coagulation Over Ultrafiltration and Classical Jar Tests / Optimering av koaguleringsförhållanden för avskiljning av naturligt organiskt material (NOM) : Jämförelse av avskiljningseffektivitet för NOM vid användning av inline-koagulering tillsammans med ultrafiltrering i bänkskala samt klassisk testning

Abstract The removal of Natural Organic Matter (NOM) from water supplies is crucial for the provision of clean, safe drinking water. Lab-scale experiments have been extensively utilized in water treatment facilities to optimize this process. The most used lab-scale method is known as jar test. However, with emerging membrane filtration, lab-scale inline coagulation has been recently utilized to mimic the inline coagulation over membrane filtration in water treatment plants (WTPs).     This study aims to compare the jar test with inline coagulation micro pilot methods from different aspects, including the NOM removal, time, and cost needed for each process. Three case studies were conducted using different water sources in Sweden: Katrineholm, Mälaren, and Mjörn.     In the experiments conducted to find the optimal NOM removal condition, the inline coagulation micro pilot method outperforms the jar test in terms of removal efficiency. In the Katrineholm case, the micro pilot method with a coagulant dosage of 4.5mg/l Al at pH 6.7 achieved removal efficiencies of 63% for fluorescent dissolved organic matter (fDOM), whereas the jar test showed 60% for fDOM but with a significantly higher coagulant dosage (7mg/l) . In the Mälaren case, the micro pilot method using a dosage of 2.5mg/l Al at pH 6.4 yielded removal efficiencies of 62% for UV254, absorbing organic matter and the jar test showed almost the same removal (64%) for UV254 but with a significantly higher coagulant dosage. Finally, in the Mjörn case, the micro pilot method with a coagulant dosage of 3.5mg/l Al at a pH of 6.6 resulted in removal efficiencies of 76% for UV254, the jar test also resulted in 76% for UV254 at the same pH level.     While the micro pilot method showed better removal efficiencies, it consumed more chemicals compared to the jar test. The micro pilot experiments required higher volumes of coagulant and additional chemical backwashing after each set of experiments. In contrast, the jar test method used lesser quantities of coagulant and chemicals due to the smaller scale of the experiments.     The findings of this study highlight the superiority of the inline coagulation micro pilot method over the jar test for optimizing NOM removal in water treatment processes. Despite the jar test being quicker and more cost-effective, it often required higher coagulant dosages to achieve comparable results. This was mainly because jar test demands heavy and large enough flocs to be removed from the water. Conversely, the micro pilot method, although more time-consuming and costly, provided more precise coagulant dosage control, resulted in higher removal efficiencies, and offered a more comprehensive understanding of the coagulation process. This is achieved by using a sensor called EXO sensor, which allows for immediate monitoring of the treatment results.

Assessment, Optimization, And Enhancement Of Ultrafiltration (uf) Membrane Processes In Potable Water Treatment

This dissertation reports on research related to ultrafiltration (UF) membranes in drinking water applications. A pilot-scale investigation identified seasonal surface water quality impacts on UF performance and resulted in the development of a dynamic chemically enhanced backwash protocol for fouling management. Subsequent analysis of UF process data revealed limitations with the use of specific flux, transmembrane pressure (TMP), and other normalization techniques for assessing UF process fouling. A new TMP balance approach is presented that identifies the pressure contribution of membrane fouling and structural changes, enables direct process performance comparisons at different operating fluxes, and distinguishes between physically and chemically unresolved fouling. In addition to the TMP balance, a five component optimization approach is presented for the systematic improvement of UF processes on the basis of TMP variations. Terms are defined for assessing process event performance, a new process utilization term is presented to benchmark UF productivity, and new measures for evaluating maintenance procedures are discussed. Using these tools, a correlation between process utilization and operating pressures was established and a sustainable process utilization of 93.5% was achieved. UF process capabilities may be further enhanced by pre-coating media onto the membrane surface. Silicon dioxide (SiO2) and powdered activated carbon (PAC) are evaluated as precoating materials, and the applicability of the TMP balance for assessing pre-coated membrane performance is demonstrated. The first use of SiO2 as a support layer for PAC in a membrane pre-coating application is presented at the laboratory-scale. SiO2-PAC pre-coatings successfully reduced physically unresolved fouling and enhanced UF membrane organics removal capabilities.

Infections in critically ill patients are a problem for the healthcare system and at any one time, 70 % of all intensive care unit (ICU) patients are treated with antibiotics. Antibiotics bind toproteins in the blood, but only unbound drug can diffuse over capillary membranes and bindto the targeted receptor. Standard protein binding percentages for antibiotics have been developed from studies on healthy volunteers and dosing regimens for patients are adapted accordingly. The determination of the total concentration of antibiotics in patients’ bloodsamples is, based on the standard percentages, ordinarily representative for the pharmacological effect of the antibiotic. However, certain conditions that are common incritically ill patients can alter protein binding percentages, resulting in a larger or smaller unbound fraction. This in turn can result in toxicity or therapeutic failure. The aim of this project was to develop an analytical method for the determination of the unbound concentration of the Beta-Lactam antibiotics cefotaxime, flucloxacillin, cloxacillin and piperacillin in plasma. A method was successfully developed using ultrafiltration for the extraction of unbound analytes and ultra high performance liquid chromatography tandem mass spectrometry, UHPLC-MS/MS, for their quantification. The method was partly validated according to the European Medicines Agency’s guidelines on bioanalytical method validation. / Kritiskt sjuka patienter med infektioner är en börda för sjukvården och 70 % av alla patienter på intensivvårdsavdelningar är ordinerade antibiotika. Antibiotika binder till proteiner i blodet, men enbart den icke-proteinbundna (fria) fraktionen kan diffundera över kapillära membran och binda till receptorer. Standardproteinbindningsgrad för olika antibiotika har utvecklats från studier på friska frivilliga och doseringen av läkemedlen är anpassade därefter. Den totala koncentrationen av antibiotika i patienters blod är vanligen representativ för den farmakologiska effekten. En metod utvecklades med ultrafiltrering för extraktion av den fria fraktionen och högupplösande vätskekromatografi och tandem masspektrometri, UHPLCMS/MS, för kvantifiering av analyterna. Metoden validerades delvis enligt den Europeiska Läkemedelsmyndighetens riktlinjer för bioanalytisk metodvalidering.

Le son et la paille de blé, riches en hémicelluloses, sont deux co-produits de la récolte de blé, abondants et renouvelables, mais qui restent à mieux valoriser. Dans le but d'extraire ces hémicelluloses, possédant plusieurs propriétés intéressantes (filmogènes, épaississantes, émulsifiantes, stabilisantes…), un procédé de fractionnement a été élaboré. Ce procédé consiste, après extraction alcaline, en la purification des extraits hémicellulosiques obtenus. Deux techniques ont été appliquées : l'ultrafiltration et la chromatographie sur résine échangeuse d'anions. L'ultrafiltration a permis de concentrer les extraits d'hémicelluloses et de les purifier en éliminant dans le perméat la plupart des molécules contaminantes, essentiellement les sucres libres et les sels minéraux. Les principales limitations de cette étape étaient la viscosité des extraits et les phénomènes de polarisation de concentration et de colmatage des membranes. La percolation sur résine, quant à elle, a permis de décolorer les extraits et de produire des poudres d'hémicelluloses plus claires en fixant certaines molécules colorées telles que les molécules possédant des groupements phénoliques. La combinaison des deux techniques a permis de produire des hémicelluloses pures à 60%. L'extrapolation du procédé à l'échelle pilote par l'utilisation d'un extrudeur bi-vis a permis d'obtenir des poudres d'hémicelluloses pures à 40%. / Wheat bran and wheat straw are two by-products abundant and renewable, rich in hemicelluloses, but still not well valued. In order to extract these hemicelluloses characterized by their several interesting properties (film-forming, thickening, emulsifying, stabilizing…), a fractionation process was developed. This process consisted, after alkaline extraction, to purify the hemicellulosic extracts obtained. Two techniques were used: ultrafiltration and chromatography on anion exchange resin. The ultrafiltration allowed to concentrate the extracts and to purify them by removing in the permeate, the contaminating molecules, mostly free sugars and minerals. The main limiting factors of this stage were the extracts viscosity and the fouling of the membranes. The percolation on the resin discoloured the extracts and allowed to produce hemicellulosic powders with lighter colours and that by capturing the coloured compounds such as molecules with phenolic groups. The combination of the two techniques allowed the production of purified hemicelluloses; the purity was about 60%. The extrapolation of the process at a pilot scale using a twin-screw extruder allowed to obtain hemicelluloses with a purity of 40%.

Techniques to assess volume status and haemodynamic stability in patients on haemodialysis

Volume overload is a common feature in patients on haemodialysis (HD). This contributes significantly to the cardiovascular disease burden seen in these patients. Clinical assessments of the volume state are often inaccurate. Techniques such as interdialytic blood pressure, relative blood volume monitoring, bioimpedance are available to improve clinical effectives. However all these techniques exhibit significant shortcomings in their accuracy, reliability and applicability at the bed side. We evaluated the usefulness of a dual compartment monitoring technique using Continuous Segmental Bioimpedance Spectroscopy (CSBIS) and Relative Blood Volume (RBV) as a tool to assess hydration status and determine dry weight. We also sought to evaluate the role of Atrial Natriuretic Peptide (ANP) and B-type Natriuretic Peptide (BNP) as a volume marker in dialysis patients. The Retrospective analysis of a historical cohort (n = 376, 55 Diabetic) showed a significant reduction in post-dialysis weights in the first three months of dialysis (72.5 to 70kg, p<0.027) with a non-significant increase in weight between months 6-12. The use of anti-hypertensive agents reduced insignificantly in the first 3 months, increased marginally between months 3-6 and significantly increased over the subsequent 6 months. The residual urea clearance (KRU) fell and dialysis times increased. The cohort was very different to that dialysing at Tassin and showed a dissociation between weight reduction and BP control. This may relate to occult volume overload. CSBIS-RBV monitoring in 9 patients with pulse ultrafiltration (pulse UF) showed distinct reproducible patterns relating to extra cellular fluid (ECF) and RBV rebound. An empirical Refill Ratio was then used to define the patterns of change and this was related to the state of their hydration. A value closer to unity was consistent with the attainment of best achievable target weight. The refill ratio fell significantly between the first (earlier) and third (last) rebound phase (1.97 ± 0.92 vs 1.32 ± 0.2). CSBIS monitoring was then carried out in 31 subjects, whilst varying dialysate composition, temperature and patient posture to analyse the effects of these changes on the ECF trace and to ascertain whether any of these interventions can trigger a change in the slope of the ECF trace distinct to that caused by UF. Only, isovolemic HD caused a change in both RBV and ECF in some patients that was explained by volume re-distribution due to gravitational shifts, poor vascular reactivity, sodium gradient between plasma and dialysate and the use of vasodilating antihypertensive agents. This has not been described previously. These will need to be explored further. The study did demonstrate a significant lack of comparability of absolute values of RECF between dialysis sessions even in the same patient. This too has not been described previously. This is likely to be due to subtle changes in fluid distribution between compartments. Therefore a relative changes must be studied. This sensitivity to subtle changes may increase the usefulness of the technique for ECF tracking through dialysis. The potential of dual compartment monitoring to track volume changes in real time was further explored in 29 patients of whom 21 achieved weight reductions and were able to be restudied. The Refill Ratio decreased significantly in the 21 patients who had their dry weights reduced by 0.95 ± 1.13 kg (1.41 ± 0.25 vs 1.25 ± 0.31). Blood pressure changes did not reach statistical significance. The technique was then used to examine differences in vascular refill between a 36oC and isothermic dialysis session in 20 stable prevalent patients. Pulse UF was carried out in both these sessions. There were no significant differences in Refill Ratios, energy removed and blood pressure response between the two sessions. The core temperature (CT) of these patients was close to 36oC and administering isothermic HD did not confer any additional benefit. Mean BNP levels in 12 patients during isovolemic HD and HD with UF did not relate to volume changes. ANP concentrations fell during a dialysis session in 11 patients from a mean 249 ± 143 pg/ml (mean ± SD) at the start of dialysis to 77 ± 65 pg/ml at the end of the session (p<0.001). During isolated UF levels did not change but fell in the ensuing sham phase indicating a time lag between volume loss and decreased generation. (136±99 pg/ml to 101±77.2 pg/ml; p<0.02) In a subsequent study ANP concentrations were measured throughout dialysis and in the post-HD period for 2 hours. A rebound in ANP concentration was observed occurring at around 90 min post-HD. The degree of this rebound may reflect the prevailing fluid state and merit further study. We have shown the utility of dual compartment monitoring with CSBIS-RBV technique and its potential in assessing volume changes in real time in haemodialysis patients. We have also shown the potential of ANP as an independent marker of volume status in the same setting. Both these techniques merit further study.

A falência de ultrafiltração (UFF) é uma causa importante de interrupção da diálise peritoneal (DP) enquanto terapia renal substitutiva. Além da inflamação crônica e aguda causadas à membrana peritoneal (MP) pelos produtos de degradação da glicose, produtos avançados da glicosilação, pH ácido das soluções e infecções, -bloqueadores (BB) também foram implicados na gênese da UFF. A vasoconstrição arteriolar esplâncnica é considerada a causa provável da UFF por BB. O nebivolol (NV), um bloqueador 1-adrenérgico altamente seletivo que, diferente de outros BB, possui efeito vasodilatador por aumento de óxido nítrico (NO) por ativar a via L-arginina-NO, foi testado em pacientes idosos com ICC e levou à redução na mortalidade. O objetivo desse estudo é analisar os efeitos do NV sobre a ultrafiltração (UF), MP e características do efluente em um modelo animal de DP, através do estudo de fenômenos envolvidos na degeneração da MP e UFF, como transição epitélio mesenquimal (EMT) e fibrose, além de parâmetros humorais e celulares de inflamação. 21 camundongos C57BL/6 fêmeas, não urêmicos, com 12 a 14 semanas, foram submetidos à colocação de cateter peritoneal. Após uma semana, foram divididos em 3 grupos de 7 animais: grupo controle (observação 30 dias), grupo SDP (2 mL/ dia de solução glicosada de diálise peritoneal a 4,25% através do cateter, por 30 dias) e grupo NV (além da infusão, receberam 8 mg/kg/dia de NV por gavagem, por 30 dias). Após 30 dias, comparou-se espessura submesotelial, volume de UF, velocidades de transporte de pequenos solutos, marcação submesotelial de pan-citoqueratina, para quantificar EMT, contagem de vasos, linfangiogênese diafragmática e concentração de IL-6 e IL-10 no efluente. A espessura da MP foi de 23,14 m no grupo controle, no grupo SDP foi de 102,4 m e no grupo NV, 29,04 m, com p<0,05. O volume de UF foi 1,94mL para o grupo controle, para o grupo SDP, 1,56 mL e, para o grupo NV, 2,05 mL, também com p<0,05. Houve menor EMT, menor angiogênese e tendência a transporte mais lento de solutos no grupo tratado, assim como menor concentração de IL-6 e proporções de populações de linfócitos semelhantes às do grupo controle. Concluímos que a droga impediu o desenvolvimento de UFF, através do bloqueio de fenômenos como EMT, espessamento da MP e neoangiogênese, além de preservar características de imunidade celular e humoral locais, merecendo ser estudada em pacientes submetidos à DP / Ultrafiltration failure (UFF) is a major cause of peritoneal dialysis (PD) discontinuation. Besides peritoneal membrane (PM) acute and chronic inflammation caused by glucose degradation products, advanced glycation end-products, acidic pH of the solutions and peritoneal infections, also -blockers (BBs) have been implicated in UFF genesis. Splanchnic arteriolar vasoconstriction has been considered the probable cause of UFF induced by BBs. Nebivolol (NV), a highly selective 1-adrenergic blocker, unlike other BBs, has a vasodilatory effect caused by its ability to increase nitric oxide (NO) through L-arginine-NO pathway activation. NV has been tested in elderly patients with congestive heart failure and led to mortality reduction. The aim of this work is to analyze the effects of NV over ultrafiltration (UF), PM and effluent characteristics in an animal model of PD. For that end, phenomena known to be involved in PM degeneration and UFF, such as epithelial-to-mesenchymal transition (EMT), fibrosis, as well as cellular and humoral parameters of inflammation have been studied. 21 C57BL/ 6 female non uremic mice, ageing 12 to 14 weeks, underwent peritoneal catheter placement. One week later, they were divided into 3 groups of 7 animals: control group (observation for 30 day), PDF group (2 mL/ day of 4.25% dextrose peritoneal dialysis fluid injected through the catheter for 30 days) and NV group (besides the PDF infusion, this group received 8 mg/ kg/ day of NV by gavage, for 30 days). After 30 days, submesotelial thickness, UF volume, small solute transport speed, submesotelial pan-cytokeratin staining (EMT quantification), vessel count, diaphragmatic lymphangiogenesis and IL-6 and IL-10 concentrations in the effluent were compared. PM thickness was 23.14 m in the control group, 102.4 m in the PDF group and 29.04 m in the NV group, p <0.05. UF volume was 1.94 mL in the control group, 1.56 mL in the SDP group, and in the NV group, 2.05 mL, p <0.05. There was less EMT, less angiogenesis and a tendency to a slower solute transport in the treated group. Lower levels of IL-6 and similar lymphocyte populations proportions to the control group were also found. We conclude that the drug can prevent UFF development, through blockade of phenomena such as EMT, PM thickening and neoangiogenesis, while characteristics of local cellular and humoral immunity were preserved. These results warrant a clinical study of the drug in PD patients

Hidrólise contínua de sacarose em um reator enzimático com membrana / Continuous hydrolysis of sucrose on a enzymatic reactor with membrane

Reatores enzimáticos com membrana (REM) combinam muitas das características desejáveis em um bioprocesso, tais como: reator de elevada produtividade, reprodutibilidade no tempo, fácil controle e automação, operação em regime contínuo, eficiente separação de biocatalisador, substratos e produtos, com a viabilidade do uso de enzimas sem necessidade de imobilização. Este estudo faz parte do desenvolvimento e otimização de um bioprocesso que utiliza um reator enzimático com membrana com a enzima invertase, que catalisa a hidrólise da sacarose produzindo uma mistura equimolar de glicose e frutose. Estudou-se a influência de algumas variáveis operacionais e de reação no comportamento do REM. Este reator consiste de um tanque agitado de 50 mL onde células de Saccharomyces cerevisiae, contendo a invertase, ficam retidas por meio de uma membrana (&#216;poro 0,45&#181;m), disposta na parte inferior do sistema. A configuração deste é semelhante a um reator contínuo tipo tanque agitado (CSTR). Inicialmente, avaliou-se a influência das variáveis: concentração de sacarose e concentração de enzima, por meio de experimentos seriais, que possibilitaram fixar estas variáveis em níveis adequados, de 500 mM e 1 mg/mL, respectivamente, para a continuidade dos ensaios por meio de planejamentos experimentais fatoriais. Foi utilizado um planejamento experimental (23 + estrela) e a análise de superfície de resposta para avaliar a influência das variáveis: vazão de alimentação do substrato, temperatura e pH. Para a análise estatística dos resultados, utilizaram-se como respostas o grau de conversão da sacarose e a produtividade em frutose, determinadas em amostragens feitas durante o tempo de operação do reator, em média de 8 a 9 horas por condição avaliada. Dos resultados obtidos, mostraram-se significativos, com 95% de confiança, os efeitos lineares e quadráticos da vazão e temperatura. As condições ótimas encontradas foram: vazão de alimentação entre 0,4 e 1,0 mL/min e temperatura 51&#176;C. O grau de conversão obtido foi de aproximadamente 95%, com as seguintes condições experimentais: concentração da suspensão de células de 1mg/mL, temperatura de 51&#176;C, pH 5,5; concentrações de sacarose de 500 mM e vazão de alimentação de 1,0 mL/min. Para esta condição obteve-se uma produtividade da ordem de 0,6 mmol frutose/h.mg invertase. Os desvios entre os valores previstos pelo modelo estatístico e pelo modelo experimental foram da ordem de 3%. Em função dos resultados obtidos neste trabalho concluiu-se que esta concepção de reator é eficiente para a bioconversão da sacarose e, portanto, o processo contínuo com reator com membrana é promissor para o desenvolvimento de processos enzimáticos desta natureza. / Enzymatic membrane reactors combine several desirable characteristics in a bioprocess, such as high productivity, reproducibility, easy control and automation, continuous operation, efficient separation of biocatalyst, substrate and products, and the use of enzymes without immobilization. This work is part of the development and optimization of a bioprocess using an enzymatic reactor which utilizes a membrane reactor for the enzymatic hydrolysis of sucrose in a solution of fructose and glucose, containing the enzyme invertase. The influence of some operational and reaction variables on the performance of the membrane reactor was studied. The bioreactor consisted of a 50 mL-stirred tank where intact cells of Saccharomyces cerevisiae, containing invertase in the cell wall, are retained inside the reactor by a microfiltration membrane (&#216;pore 0.45&#181;m). The flat sheet membrane is fixed at the bottom of the device. The reactor configuration is similar to a continuous stirred tank reactor (CSTR). Initially, the influence of sucrose and enzyme concentration was evaluated through a series of experiments in order to determine the suitable levels of these variables. Sucrose was set to 500 mM and enzyme set to 1 mg/mL. This allowed the work to continue by means of experimental factorial designs. It was utilized a 23 full experimental design followed by a 2nd order statistical design and, the surface response methodology in order to evaluate the influence of volume feeding rate of the substrate, temperature and pH on fructose productivity and sucrose conversion. The values were obtained during the reactor operation. The average operation time was trom 8 to 9 hours for all evaluated conditions. From the statistical analysis, it was concluded that the linear and quadratic effects of temperature and flow rate on the results were the most significant with 95% of confidence. The optimum conditions found for this bioprocess were volume feeding rate between 0.4 and 1.0 mL/min and temperature of 51&#176;C. The degree of conversion of sucrose obtained experimentally was 95%, in the following experimental conditions: cell concentration of 1mg/mL, temperature of 51&#176;C, pH 5.5; sucrose concentration of 500 mM and feeding rate of 1.0 mL/min. For this operational condition it was obtained a productivity of about 0.6 mmol fructose/h.mg invertase. The deviation between the predicted values by the statistical model and the experimental data was 3%. Based on the results obtained in this work, it can be concluded that this conception of bioreactor is efficient for the bioconversion of sucrose and, a continuous membrane reactor process is very promising for the development of this kind of enzymatic process.
