Framework For Cost Modeling A Supply Chain

Researchers are interested in value chain analysis to identify the different opportunities for cost savings. The literature have been narrow in scope and addressed specific problems; however none has addressed the need for a general framework that can be used as a standard template in the supply chain cost management and optimization, though Dekker and Goor (2000) said that the goal was to develop a model that would allow direct comparison of specific activities between firms, such as warehousing activities costs. There was no indication in the literature of a cost model that can identify all costs and cost drivers through the supply chain. Some firms built models to analyze the effect of changes in activities but only with limited activities such as logistics. The purpose of this research is to create a general framework that can express the cost data for the partners of the supply chain in similar terms. The framework will layout the common activities identified within the firm and the relationship of these activities between the partners of the supply chain, and the framework will identify the effect of changes in activities on other partners within the supply chain. Cost information will help in making decisions about pricing, outsourcing, capital expenditures, and operational efficiency. The framework will be able to track cost through the chain, which will improve the flexibility of the supply chain to respond to rapidly changing technology. The framework will help in developing product strategy paradigms that encompass the dynamics of the market, in particular with respect to the technology adoption lifecycle.

Biochemical Studies Of Abce1

The growth and survival of all cells require functional ribosomes that are capable of protein synthesis. The disruption of the steps required for the function of ribosomes represents a potential future target for pharmacological anti-cancer therapy. ABCE1 is an essential Fe-S protein involved in ribosomal function and is vital for protein synthesis and cell survival. Thus, ABCE1 is potentially a great therapeutic target for cancer treatment. Previously, cell biological, genetic, and structural studies uncovered the general importance of ABCE1, although the exact function of the Fe-S clusters was previously unclear, only a simple structural role was suggested. Additionally, due to the essential nature of ABCE1, its function in ribosome biogenesis, ribosome recycling, and the presence of Fe-S within ABCE1, the protein has been hypothesized to be a target for oxidative degradation by ROS and critically impact cellular function. In an effort to better understand the function of ABCE1 and its associated Fe-S cofactors, the goal of this research was to achieve a better biochemical understanding of the Fe-S clusters of ABCE1. The kinetics of the ATPase activity for the Pyrococcus abyssi ABCE1 (PabABCE1) was studied using both apo- (without reconstituted Fe-S clusters) and holo- (with full complement of Fe-S clusters reconstituted post-purification) forms, and is shown to be jointly regulated by the status of Fe-S clusters and Mg2+. Typically, ATPases require Mg2+, as is true for PabABCE1, but Mg2+ also acts as a unusual negative allosteric effector that modulates ATP affinity of PabABCE1. Comparative kinetic analysis of Mg2+ inhibition shows differences in the degree of allosteric regulation between the apo- and holo-PabABCE1 where the apparent Km for ATP of apo- iv PabABCE1 increases >30 fold from ~30 µM to over 1 mM when in the presence of physiologically relevant concentrations of Mg2+. This effect would significantly convert the ATPase activity of PabABCE1 from being independent of cellular energy charge () to being dependent on  with cellular [Mg2+]. The effect of ROS on the Fe-S clusters within ABCE1 from Saccharomyces cerevisiae was studied by in vivo 55Fe labeling. A dose and time dependent depletion of ABCE1 bound 55Fe after exposure to H2O2 was discovered, suggesting the progressive degradation of Fe-S clusters under oxidative stress conditions. Furthermore, our experiments show growth recovery, upon removal of the H2O2, reaching a growth rate close to that of untreated cells after ~8 hrs. Additionally, a corresponding increase (~88% recovery) in the ABCE1 bound 55Fe (Fe-S) was demonstrated. Observations presented in this work demonstrate that the majority of growth inhibition, induced by oxidative stress, can be explained by a comparable decrease in ABCE1 bound 55Fe and likely loss of ABCE1 activity that is necessary for normal ribosomal activity. The regulatory roles of the Fe-S clusters with ABCE1 provide the cell a way to modulate the activity of ABCE1 and effectively regulate translation based on both cellular energy charge and the redox state of the cell. Intricate overlapping effects by both [Mg2+] and the status of Fe-S clusters regulate ABCE1’s ATPase activity and suggest a regulatory mechanism, where under oxidative stress conditions, the translational activity of ABCE1 can be inhibited by oxidative degradation of the Fe-S clusters. These findings uncover the regulatory function of the Fe-S clusters with v ABCE1, providing important clues needed for the development of pharmacological agents toward ABCE1 targeted anti-cancer therapy.

Syftet med studien är att undersöka pedagogers syn på konflikter mellan kollegor i förskolan. Genom studien önskar vi belysa konflikter mellan pedagoger i förskolan samt vilken inverkan konflikter kan ha på pedagogers samspel, trivsel och välmående. Vi fann här ett GAP i den forskning som vi hittat inom området konflikt och konflikthantering.   Vi använder oss av en kvantitativ metod genom enkäter för att samla in empirisk data. Enkäterna besvarades av 171 pedagoger i 28 skånska kommuner. Den insamlade empirin analyseras genom deskriptiv statistik och tolkas utifrån Jordans (2015) begrepp: robust samarbetskultur och självinsikt, konfliktnivåerna: individnivå, relationsnivå och systemnivå samt Galtungs ABC-modell.   Resultatet visar att det i stor utsträckning förekommer konflikter mellan kollegor i förskolan samt att dessa kan komma att inverka på samspel, trivseln och välmåendet om de förblir olösta. Trivsel anses vara högt prioriterat av respondenterna och trots förekomsten av konflikter verkar respondenterna trivas på arbetsplatsen. Utifrån resultatet drar vi slutsatsen att majoriteten av respondenterna uppfattar begreppet konflikt som negativt betingat. En annan slutsats är att det behöver finnas kunskap om konflikter och konflikthantering kollegor emellan för att kunna skapa ett gott sampel samt ökad trivseln och välmåendet hos pedagoger på arbetsplatsen.

An Experimental Implementation of Action-Based Concurrency

This thesis reports on an implementation of an action-based model for concurrent programming. Concurrency is expressed by allowing objects to have actions with a guard and a body. Each action has its own execution context, and concurrent execution is realized when program execution is happening in more than one context at a time. Two actions of different objects can run concurrently, and they are synchronized whenever a shared object is accessed simultaneously by both actions. The appeal of this model is that it allows a conceptually simple framework for designing and analyzing concurrent programs. To experiment with action-based concurrency, we present a small language, ABC Pascal, which is an experimental attempt as a proof of feasibility of such a model, and also meant to help identify issues for achieving reasonable efficiency in implementation. It extends a subset of Pascal that supports basic sequential programming constructs, and provides action-based concurrency as the action-based model prescribes. This work deals with the specification and implementation of ABC Pascal. The one-pass compiler directly generates assembly code, without devoting efforts to optimization. While the code is not optimized, the results that ABC Pascal has achieved in performance testing are so far comparable to mainstream concurrent programming languages. / Thesis / Master of Science (MSc)

Categorizing Accelerated Bridge Construction Projects for Improving Decision-Making

Accelerated Bridge Construction, also known as ABC, is a methodology that seeks to improve project development of bridges by reducing the overall project schedule and the impact on the traveling public by implementing innovative technologies and strategies in any phase of project development. However, ABC may incur additional direct costs for the project and some risks are associated because of the accelerated constraints implied in this methodology. On the positive side, the opportunity costs and reductions of traffic disruptions costs may overcome the additional costs associated with ABC. Decision-making methodologies for assessment of ABC as an alternative to traditional construction are of great interest for project developers. The topics of research about ABC are diverse but focus mainly on the means and methods, technical aspects, applications, innovations, and decision-making of ABC. Decision-making is of great concern for project developers, especially government organizations, to sustain project goals of serviceability and to validate the additional expenditures in a project. In addition, project developers improve their decisions and project outcomes by reviewing success and failure cases for completed projects in the past. This study seeks to improve the decision-making processes in ABC by finding a more direct correlation of projects to compare by means of a categorization of these ABC projects. Smaller groups in this categorization will help narrow the scope of the characteristics of the projects to consider and to find more relevant lessons learned from the smaller groups of the categorization. To develop the categorization in this study, the data source used is the completed ABC projects database from the Federal Highway Administration (FHWA). The statistical categorization methodology for this study is the Agglomerate Hierarchy Clustering which developed a determined number of cluster based on the closeness among data parameters with "n" number of dimensions of analysis. The number of dimensions for the analysis in this study was established as 13 parameters collected from the database and these were considered critical decision-making parameters and consequential parameters to reflect project decisions and consequences of those decisions. The results of this study rendered 3 categories, and into these categories, 5 sub-categories were distributed according to the same analysis developed. The sub-categories show similarities between the projects according to the parameters established, so the sub-categories help narrow the scope of projects for project developers. As a complement to the categorization, a project matching tool for external projects was also developed to help decision-makers to test their projects according to the analysis in this study and also help developers narrow their review of cases in search for lessons learned. Uses of this study include the prediction of information of parameters according to the variables and ranges in this categorization, and the narrowing of study cases to review. Stakeholders interested can be government organizations seeking to establish the viability of an ABC project, or to improve their project outcomes at any stage of development. Other stakeholders can be designers and contractors that also need to improve their projects at any stage of development. / Master of Science / Accelerated Bridge Construction (ABC) reduces construction time and diminishes traffic disruption by applying strategies and innovations to the way bridges are designed and constructed. However, it implies additional initial costs that need to be justified. Project developers need to evaluate their projects to make decisions to assure the best execution and outcomes. The review of lessons learned from previous bridge projects has been one of the sources that developers have found to find strategies and tips to implement best practices or to avoid mistakes during planning and construction of new bridge projects. This study relies on the lessons learned compiled on the completed ABC project database from the federal transportation authority, the Federal Highway Administration (FHWA), in order to help developers, narrow the scope of projects to be reviewed and find more relevant cases to compare their projects. This scope includes the development of a categorization or a structured hierarchy of completed ABC projects with the FHWA database by applying a statistical analysis and a validation procedure. The results of this study rendered a categorization with 3 categories and 5 subcategories, each with its definition of parameters, which highlight the similarities among projects in the same groups and the differences among different categories. In addition, a framework was developed to match external projects with the categorization in this study to help project developers narrow the projects to review, or even predict missing information according to the parameters in this study. These results may help decision-makers better inform their decisions and support the research in the field of study.
