Characterization of Several Small Biologically Relevant Molecules by Infrared Multiple Photon Dissociation Spectroscopy and Electronic Structure Calculations

Infrared multiple photon dissociation (IRMPD) spectroscopy has been coupled with electronic structure calculations in order to elucidate the structures of several small biological molecules including: uracil, 5-fluorouracil, 5-fluorocytosine, ferulic acid, and a number of their related analogs. IRMPD is a powerful technique, that when combined with electronic structure calculations can provide convincing evidence for the structural characterization of ions in the gas phase. Isomers of uracil and 5-fluorouracil (5-FU) have been characterized by calculations performed at the MP2(full)/aug-cc-pVTZ level of theory; however, infrared multiple photon dissociation spectroscopy experiments proved to be unsuccessful for these species. Geometry optimization and frequency calculations have isolated the dominant isomer(s) for neutral and deprotonated uracil and 5-fluorouracil, along with several cluster interactions involving water, methanol, ammonia, and methylamine. For both uracil and 5-FU, a single relevant neutral isomer was determined, with each isomer existing in the diketo, as opposed to the enol form. Following the deprotonation of this neutral isomer, both uracil and 5-FU were permitted to form anionic cluster ions with water, methanol, ammonia, or methylamine, and based on the relative Gibbs free energies (298 K) of the calculated isomers, relevant cluster interactions were determined. For each cluster, several sites of intramolecular interaction were found to exist; however, interaction at the site of deprotonation was the most favourable in every instance. Ionic hydrogen bond interactions have been found in several clusters formed by 5-fluorocytosine (5-FC). The chloride and trimethylammonium cluster ions, in addition to the cationic and anionic dimers have been characterized by infrared multiple photon dissociation (IRMPD) spectroscopy and electronic structure calculations performed at the B2PLYP/aug-cc-pVTZ//B3LYP/6-311+G(d,p) level of theory. IRMPD spectra in combination with calculated spectra and relative energetics have indicated, quite conclusively, that a single isomer for each 5-FC cluster that is likely being observed experimentally except in the case of the anionic dimer, in which a combination of isomers is probable. For the 5-FC-trimethylammonium cluster specifically, the calculated spectrum of the lowest energy isomer matches the experimental spectrum remarkably well. Interestingly, the cationic dimer of 5-FC was found to have a single energetically relevant isomer (Cationic-IV) in which a unique tridentate ionic hydrogen bond interaction is formed. The three sites of intramolecular ionic hydrogen bonds in this isomer interact very efficiently, leading to a significantly large calculated enthalpy of binding of 180 kJ/mol. The magnitude of the calculated binding energy for this species, in combination with the strong correlation between the simulated and IRMPD spectra, indicates that the tridentate-bound dimer is observed predominantly in experiment. Comparison of the calculated relative Gibbs free energies (298 K) for this species with several of the other isomers considered also supports the likelihood of the dominant protonated dimer existing as Cationic-IV. Protonated ferulic acid has been characterized using infrared multiple photon dissociation spectroscopy and electronic structure calculations at the B3LYP/6-311+G(d,p) level of theory. Neutral ferulic acid has been determined to undergo protonation on the carbonyl oxygen of the acid group, forming an ion of m/z 195. Due to its extensively conjugated structure, protonated ferulic acid (m/z 195) is observed to yield three stable fragment ions in IRMPD experiments. It is proposed that two parallel fragmentation pathways of protonated ferulic acid are being observed. First, proton transfer occurs from the carbonyl oxygen to the hydroxyl oxygen within the acid group, resulting in the loss of water and subsequently carbon monoxide, forming ions of m/z 177 and 149, respectively. The second proposed fragmentation pathway undergoes proton transfer from the phenolic group to the methoxy group resulting in loss of methanol and rearrangement to a five-membered ring of m/z 163. IRMPD spectra have been obtained for the ions m/z 195 and m/z 177, and anharmonic calculations have been performed on these species at the B3LYP/6-311+G(d,p) level of theory. The calculated anharmonic spectra for these ions match the experimental spectrum exceptionally well and strongly support the proposed fragmentation mechanisms.

Resonant Excitation Of Ions In Paul Trap Mass Spectrometer

A Paul trap mass spectrometer has a three-electrode geometry mass analyzer consisting of two identical end cap electrodes and a ring electrode. Traditionally, the two end cap electrodes are electrically grounded and an RF potential is applied to the central ring electrode to generate the "trapping field". Ions of the analyte sample are formed in situ by electron bombardment and mass analysis of the fragment ions is performed by mass selectively destabilizing the ions from the trap. The inhornogeneities present in the trapping field (introduced either by misalignment of the trap geometry or by applying a dipolar auxiliary excitation across the end cap electrodes) give rise to various interesting phenomena including, resonance ejection of the trapped ions This thesis is concerned with taking a look into the experimental aspects associated with resonance ejection of ions caused by the dipolar excitation Additionally, u also reports the work undertaken to develop necessary instrumentation for resonant excitation experiments and my contribution to operational>zc the Paul trap mass spectrometer fabricated in the laboratory. The thesis is divided into 5 chapters. Chapter 1 is an introductory chapter. After discussing the conditions for stability of the trapped ions, it goes on to present a brief survey of a variety of applications in literature, which have used resonant excitation. Towards the end, the motivation of the present effort and the scope of work in the thesis have been spelt out. This includes (a) redesign of the ion detector electronics, (b) design of an auxiliary excitation generator, and (c) studies on resonance ejection. Chapter 2 outlines the design considerations, circuit description and fabrication details for the ion detector electronics. The circuits presented in this chapter include (a) electrometer amplifier and (b) -3 kV DC supply for the electron multiplier detector. The electrometer amplifier amplifies the ion current signal from the electron multiplier detector and it needs to have a high input impedance and a high slew rate. The electron multiplier detector requires -3 kV DC power supply for operation. The -3 kV DC power supply is required to have a regulated output voltage with low ripple in the output. Chapter 3 presents the design considerations, circuit description and fabrication details for the auxiliary excitation generator. The auxiliary excitation generator is a three channel DDS (Direct Digital Synthesis) oscillator with independent control of frequency amplitude, and phase of the output signal. Chapter 3 also discusses the micro controller based control sub-system that allows the user to set above mentioned output parameters. The control sub-system provides a user-friendly keyboard interface and 2-line alphanumeric LCD display per channel. It also provides various bus interfaces (such as I2C and SPI) to interface with DDS oscillator ICs, amplitude control DAC, and LCD displays. The chapter then goes on to describe the implementation details of the software written for the control sub-system. The hardware design is simplified by using a micro controller as heart of the control sub-system and employing the software to handle the complex functions. As an example, the design of the keyboard interface is simplified by directly connecting a matrix keyboard to the input/output port of the micro controller. The software is used to scan the keyboard, detect key press and find out the key pressed. Nonetheless, in order to meet specific performance required for the present work, the software needs to have a sense of time, be portable and scalable. Details of the "layered" architecture adopted by as to meet these specific requirements, the lower level "driver" functions implemented for various interfaces of the control sub-system, and the higher level or the "application" software, are described. The application software uses the driver functions to accomplish various tasks required to be executed by the control sub-system. Finally, the chapter presents the design consideration and fabrication details of the coupling transformer used to couple the output of the auxiliary excitation generator to the Paul trap Chapter 4 describes the resonant excitation experiments performed as part of the present work. First of all the chapter presents the improvement in the performance of the Paul trap mass spectrometer as a result of redesigned ion detector electronics It is seen that the resolution is improved significantly due to the improved response time of the electrometer amplifier. The chapter then describes the effect of the resonant excitation on the ions and also that the frequency of the applied auxiliary excitation should be between 500 kHz to 125 kHz. Next, a number of mass spectra for different frequencies of the applied auxiliary excitation are presented. These mass spectra indicate that the resonant ejection sets in for lower masses even at lower amplitude of the auxiliary excitation where as higher amplitude is required for the resonant ejection of the higher masses. It is seen that the resonant excitation of ions improves resolution of the mass spectrum. Moreover, the auxiliary excitation results in ejection of the ions at lower amplitude of the RF voltage and thus allows extending the mass range of the mass spectrometer. We present the mass spectrum of CCI4 which is not possible to normally record in our instrument. We also present results intended to understand the relation between frequency and amplitude of the auxiliary excitation on the mass spectra of benzene. Finally, results of an interesting experiment are presented which indicates the presence of the non-linear resonance points in the Paul trap. Chapter 5 presents the concluding remarks. Molekulare Analyse des probiotischen Stamms Escherichia coli Nissle 1917

Der probiotische Stamm E. coli Nissle 1917 ist ein Fäkalisolat, das in der Medizin traditionell zur Behandlung verschiedener gastrointestinaler Erkrankungen eingesetzt wird. Durch erfolgversprechende klinische Studien zur Remissionserhaltung bei Colitis ulcerosa, bei denen EcN als therapeutische Alternative zur Standardmedikation eingesetzt wird, ist das Interesse an den Wirkmechanismen von Probiotika stark gestiegen. EcN gehört derzeit zu den am besten untersuchten Probiotika. Einige Wirkmechanismen konnten dadurch schon aufgeklärt werden. So sind vermutlich Strukturkomponenten und stammspezifische Syntheseleistungen an der Ausprägung des probiotischen Phänotyps von EcN beteiligt. Schlüssige Konzepte, die über Gene, Genprodukte und molekulare Mechanismen den probiotischen Effekt von EcN erklären, fehlen bislang. Im Rahmen dieser Arbeit wird das Genom von EcN analysiert und auf der Basis der Genomsequenz mit anderen E. coli-Stämmen verglichen. Mit Hilfe einer Promotor-Reporter-Fusionsbibliothek (Promotorbank) werden intestinal in vivo regulierte Gene identifiziert und dadurch neue Ansätze zur Untersuchung der probiotischen Eigenschaften von EcN geschaffen. Die Grundlage für die molekulare Analyse von EcN ist die manuelle Nachannotation seines sequenzierten Genoms. Die EcN-Sequenz wird mit 13 weiteren annotierten E. coli-Sequenzen verglichen. Nach dieser Analyse kodiert EcN derzeit 121 stammspezifische Gene. Die Genomstruktur ist mit den enthaltenen genomischen Inseln und Prophagen dem Genom des uropathogenen E. coli CFT073 sehr ähnlich. Mit wenigen Ausnahmen kodiert EcN alle in E. coli CFT073 vorhandenen Virulenz- und Fitnessfaktoren, so dass auf der Nukleotidebene die nahe Verwandschaft dieser beiden Stämme bestätigt werden kann. Zudem kann gezeigt werden, dass EcN in artifiziellen Systemen wie der Zellkultur oder gnotobiotischen Mäusen ein pathogenes Potenzial hat, obgleich die Kolonisierungsfähigkeit pathogener Bakterien durch Inkubation mit EcN herabgesetzt wird. Eine wichtige Rolle bei der Besiedlung des Intestinaltrakts und der Immunstimulation von Darmepithelzellen spielt auch die globale Regulation der Genaktivität bei EcN durch den alternativen Sigma-Faktor RpoS, der im Gegensatz zu rpoS-Deletionsmutanten zu einer gesteigerten mRNA-Expression des Tight-junction Proteins ZO-1 führt. Des Weiteren führte die Untersuchung von EcN-Deletionsmutanten zu der Schlussfolgerung, dass einige genomische Inseln für Eigenschaften, die das probiotische Verhalten erklären können, eine Rolle spielen. Durch den Einsatz einer Promotorbank von EcN in konventionellen und gnotobiotischen Mäusen werden erstmalig Sequenzen von intestinal in vivo aktiven Promotoren identifiziert. Der Aufbau eines Promotor-Reportergen-Assays mit dem Biolumineszenz erzeugenden luxCDABE-Operon ermöglichte die Untersuchung ausgewählter Promotoren in vitro. Mit einem In Vivo Imaging System (IVIS) kann in weiteren Experimenten die Aktivität dieser Promotoren in lebenden Mäusen untersucht werden. Im Rahmen dieser Arbeit wird gezeigt, dass EcN kein vollkommen harmloser probiotischer Stamm ist. Weitere Informationen über EcN sind dehalb wichtig für eine optimierte Anwendung als Therapeutikum. Die molekulare Analyse ist somit eine unbedingt notwendige Grundlage für weiterführende Untersuchungen der Eigenschaften von EcN, die für seinen probiotischen Charakter verantwortlich sind. / The probiotic E. coli Nissle 1917 is a fecal isolate which is traditionally used for treatment of various gastrointestinal disorders. In clinical trials where EcN was used as therapeutic alternative for remission maintenance of ulcerative colitis compared to standard medication, promising results led to an increased interest in probiotics. Today, EcN is one of the best studied probiotics. Therefore, several mechanisms of action could be enlightened. Structural components and strain-specific products are responsible for its probiotic effects. But conclusive concepts about genes, gene products and molecular mechanisms that really contribute to the probiotic character of EcN have not been offered so far. In order to create new possibilities to elucidate the probiotic traits of EcN the genome is analysed by taking this as a basis for comparison to other E. coli genomes and identification of intestinal in vivo regulated genes using a promoter-trap-library. The sequenced EcN genome is annotated and compared to 13 other so far annotated E. coli genomes. Concerning these analyses EcN encodes 121 strain-specific genes. The genome structure including the genomic islands and prophages is highly homolog to the uropathogenic E. coli CFT073. EcN encodes most of the virulence and fitness factors that are present in E. coli CFT073. Therefore, the close relationship of these two strains is confirmed at nucleotide level. Furthermore, it is shown that in artificial systems like cell culture assays and gnotobiotic mice EcN reveals a pathogenic potential although EcN is able to decrease colonization efficiency of pathogenic bacteria. The alternative sigma factor RpoS that is responsible for global regulation and activity of several genes seems to play an important role during colonization of EcN in the intestine and its immunostimulatory effects on intestinal epithelial cells. Investigation of EcN-deletion mutants lacking genomic islands and prophages lead to the conclusion that some genomic islands may play a role for specific probiotic traits. This is the first time where a promoter-trap-library was used in conventional and gnotobiotic mice for collection of intestinal in vivo active promoters. Constructing and establishing a promoter-reporter gene assay with the bioluminescent luxCDABE operon made the investigation of selected promoters in vitro possible as well as establishing a bioluminescence assay using an In Vivo Imaging System (IVIS) for investigation of promoter activity in living mice. In this research project was shown that EcN is not a completely harmless probiotic. The genome structure and regulatory mechanisms of gene expression are the strain’s molecular traits that lead to probiotic activity and immunostimulatory effects. Therefore, the molecular analyses presented here, together with the complete genome sequence, are a basis for further investigations of mechanisms that are responsible for the probiotic effects of EcN.

Untersuchung der Erzeugung hochgeladener Ionen in einer Raumtemperatur-Elektronenstrahl-Ionenfalle / Investigation on the production of highly charged ions at a room temperature electron beam ion trap

Hochgeladene Ionen stellen einen extremen Zustand der Materie dar, wie sie vornehmlich in kosmischen Plasmen zu finden ist. Die labormäßige Erzeugung und (spektroskopische) Untersuchung hochgeladener Ionen liefert wichtige Daten und Erkenntnisse für die Astrophysik und Fusionsforschung. Aufgrund ihrer zum Teil exotischen Eigenschaften besitzen hochgeladene Ionen ein großes Potential für eine Vielzahl neuer Anwendungen. Die bisher weltweit einzige Elektronenstrahl-Ionenfalle, die hochgeladene Ionen bis hin zu vollständig ionisierten Ionen unter Raumtemperaturbedingungen erzeugen und bereitstellen kann, die Dresden EBIT, ist Gegenstand der vorgelegten Arbeit. Die Dresden EBIT zeichnet sich neben ihrer Kompaktheit und einer einfachen Bedienung durch ihre Langzeitstabilität aus. Sowohl über Röntgenspektren als auch über die Extraktion der Ionen aus der EBIT konnte für eine Reihe von Elementen der Nachweis der Erzeugung von vollständig ionisierten Ionen bis Z=28 erbracht werden. Für schwere Elemente können Ionenladungszustände bis hin zu neonähnlichen Ionen erzeugt werden. Entscheidenden Einfluss auf den erzielten mittleren Ladungszustand hat die Ioneneinschlusszeit. Die zeitliche Entwicklung der Ladungszustandsverteilung, wie sie im Zusammenspiel der verschiedenen atomaren Prozesse simuliert werden kann, ist sowohl an einer Reihe von röntgenspektroskopischen Messungen als auch anhand von Extraktionsspektren untersucht worden. Neben der Beladung der EBIT mit gasförmigen Elementen ist insbesondere die Beladung mit Metallen, d. h. mit einem möglichst breiten Spektrum an Elementen gefordert. Die Beladung mit leichtflüchtigen metallorganischen Verbindungen, die über das Gaseinlassventil eingebracht werden können, hat sich als erfolgreiche und preiswerte Alternative zu einer MEVVA-Quelle erwiesen. Die Beladung mit Metallionen ist am Beispiel verschiedener Untersuchungen gezeigt. Der monoenergetische Elektronenstrahl gestattet neben der Untersuchung von Anregungs- und Ionisationsprozessen insbesondere die der wichtigen Rekombinationsprozesse des Strahlenden Einfangs und der Dielektronischen Rekombination. Der Einsatz eines Kristalldiffraktionsspektrometers erlaubt trotz einer aufwendigen Kalibrierung und sehr langen Messzeiten die Auflösung einzelner Übergänge in hochgeladenen Ionen. Hauptanwendungsfeld der Dresden EBIT wird der Einsatz als Ionenquelle sein. Aus den Untersuchungen des extrahierten Gesamtionenstroms können die Bedingungen für einen möglichst großen Ionenstrom und einen optimalen Ionenstrahltransport abgeleitet werden. Eine optimale Ausnutzung der Eigenschaften hochgeladener Ionen erfordert die Separation der einzelnen Ladungszustände. Der Nachweis der sehr kleinen Ionenströme erfolgt über die kapazitive Messung in einem Faradaycup. Die Messung der Ladungszustandsverteilung in Abhängigkeit von den Parametern der EBIT gibt zusätzliche Aufschlüsse über die Eigenschaften der Ionenfalle.

Formation of Small Hydrocarbon Ions Under Inter- and Circumstellar Conditions: Experiments in Ion Traps

Using ion-trapping techniques, selected laboratory experiments on ion-molecule reactions of astrophysical interest have been performed. For the first time a carbon beam source has been integrated into an ion trapping machine for studying collisions between ions and neutral carbon atoms and molecules. Results are presented for the interaction of D3+ ions stored in a ring-electrode trap (RET), with a beam of hot neutral carbon molecules, Cn (n = 1, 2, 3). The measured reaction rate coefficients are up to a factor two smaller than values presently used in astrophysical models. In order to complete our knowledge about the ion chemistry involving three carbon atoms, detailed investigations of reactions of C3+, C3H+ and C3H3+ with H2 and HD have been performed between 15 K and room temperature. These studies have been performed in a second apparatus, a variable-temperature 22-pole trap machine (VT-22PT). Results include reactive collisions, deuteration and radiative association. It is discussed in connection with the increase in lifetime of the C3+ + H2 collision complexes with falling temperature, what could be responsible for producing more C3H+ at 15 K. Tunneling is excluded. In C3+ + HD collisions an isotope effect has been detected, the C3D+ product ions being slightly more abundant than C3H+. Comparison of the reaction of C3H+ primary ions with HD and H2 gas revealed that the deuterated molecules are significantly more reactive. The process of radiative association of C3H+ and for the first time of C3+ with hydrogen molecules has been observed. An analysis of the data shows that radiative association becomes slower, if the neutral reactant is deuterated. Finally, the theoretical prediction from ab initio calculations that C3H3+ does not exchange an H for a D in collisions with HD, has been proven in an ion trap experiment. Careful analysis of all competing processes allows the conclusion that the rate coefficient is smaller than 4x10-16 cm3s-1 at 15 K. / Unter Verwendung von zwei Speicherapparaturen wurden ausgewählte, astrophysikalische wichtige Ionen-Molekülreaktionen untersucht. Durch die Kombination einer Kohlenstoffquelle mit einem Ionenspeicher, in dem so Reaktionen zwischen Ionen und Kohlenstoffmolekülen oder -atomen untersucht werden können, wurde Neuland betreten. Es werden Ergebnisse vorgestellt für die Reaktion von D3+ Ionen, die in einem Ringelektrodenspeicher gefangen sind, mit einem Strahl von heißen Cn (n = 1, 2, 3). Die gemessenen Ratenkoeffizienten sind nur halb so groß wie die Werte, die in astrophysikalischen Modellen verwendet werden. Um die Kenntnis über alle möglichen Reaktionen, bei denen drei C-Atome beteiligt sind, abzurunden, wurden zwischen 15 K und Zimmertemperatur die Reaktionen zwischen C3+, C3H+ und C3H3+ Ionen mit H2 und HD in vielen Details untersucht. Diese Experimente wurden in einer zweiten Apparatur durchgeführt, in der ein temperaturvariabler 22-Polspeicher das zentrale Element ist (VT-22PT). Berichtet werden Ergebnisse zu reaktiven Stößen, zur Deuterierung von Kohlenwasserstoffen und zur Strahlungsassoziation. In der Diskussion bleibt offen, was - in Verbindung mit der von 300 K zu 15 K zunehmenden Lebensdauer - der Grund dafür sein kann, daß die Bildung des exothermen Produkts C3H+ anwächst. Der Tunneleffekt scheidet aus. Bei der Reaktion C3+ + HD wurde ein Isotopeneffekt beobachtet, das C3D+ Produkt wird etwas häufiger gebildet als C3H+. Ein Vergleich der Reaktion zwischen C3H+ Ionen mit HD bzw. H2 zeigt, daß das deuterierte Molekül wesentlich reaktiver ist. Es wurden Ratenkoeffizienten für die Strahlungsassoziation von H2 Molekülen mit C3H+ und erstmals mit C3+ Ionen gemessen. Die Auswertung der Daten zeigt, dass der Prozeß langsamer abläuft, wenn der neutrale Stoßpartner deuteriert ist. Schließlich wurde experimentell die theoretische Vorhersage überprüft, dass C3H3+ keinen H-D Austausch mit HD eingeht. Eine sorgfältige Analyse aller konkurrierenden Prozesse ergab, dass bei 15 K der Raten koeffizient kleiner als 4x10-16 cm3s-1 ist.

Sensory evaluation and quality assessment of an alternative inner coating film in yogurt cartons

The dairy food industry is continuously striving towards products with higher quality and longer shelf-life available to the customer at low prices. Arla Foods in Linköping, Sweden, is currently investigating the possibilities of changing the material in yogurt packaging containers by replacing the currently used carton with a different and cheaper alternative. A successful switch will give the company an economical advantage without affecting the sensory attributes (smell, taste, sight, and consistency), aroma profile or other important trademarks of the yogurt. This study is designed to examine and compare yogurt that has been stored in different packaging cartons, one coated with a single-layered low-density polyethylene (LDPE) and one coated with a currently used multi-layered ethylene-vinyl alcohol (EVOH). The study was based on the analysis and measurement of sensory attributes performed by experts, physical properties in laboratory and chemical composition in GC-FID/MS together with a discriminative test where a group of people would identify any difference between the yogurts. Together, these analyses would provide an explanation about any differences between the packaging materials by connecting physical, chemical and/or sensory characteristics. The collected results would give a better and more comprehensive picture than each analysis would do separately. The results from the study show that there is a difference between yogurts stored in LDPE-based containers and yogurts stored in EVOH-based containers and that the product was chemically affected, mainly by the level of oxygen in contact with the food. The overall assessment is that the largest difference was discovered in the taste.

New Methods to Create Multielectron Bubbles in Liquid Helium

An equilibrium multielectron bubble (MEB) in liquid helium is a fascinating object with a spherical two-dimensional electron gas on its surface. After it was first observed a few decades ago, a plethora of physical properties of MEBs, for example, a tunable surface electron density, have been predicted. In this thesis, we will discuss two new methods to create MEBs in liquid helium. Before the discussion, the way to generate a large number of electrons in a low temperature system will be discussed, including thermionic emission and field emission in helium. In the first new method to make MEBs, we used a dome-shaped cell filled with superfluid helium in which an MEB was created and confined at the dome. The lifetime of the MEB was substantially longer than the previously reported observations of MEBs. In the second method, MEBs were extracted from the vapor sheath around an electrically heated tungsten filament submerged in liquid helium, either by a high electric field (up to 15 kV/cm) or by a sudden increase of a negative pressure in liquid helium. High-speed photography was used to capture the MEB's motion. A method to determine the number of electrons was developed by monitoring the oscillations of the MEBs. Finally, an electromagnetic trap was designed to localize the MEBs created using the second method, which was important for future studies of the properties of MEBs. / Physics

臺南市流浪犬捕捉、絕育、回置政策執行成效影響因素之研究 / Factors Affecting the Implementation of Trap-Neuter-Return Policy for Stray Dogs in Tainan City

流浪犬所造成的各種社會問題由來已久,然而過去中央及地方政府大多採取捕捉、撲殺的方式,官方收容所流浪動物所內死亡與安樂死數量皆居高不下,這樣的作法不僅沒有成效,亦造成了生命無辜的消逝。2015年初,立法院三讀《動物保護法》部分條文修正,通過「零安樂死」政策,零安樂死看似美好,但卻衍生出其他問題,包括收容所爆量、收容所照護人力不足、經費拮据、合適安置場所難尋等問題,可見若單採取收容及認養的方式可能會使得流浪動物的處境更加悲慘。許多動保團體倡議應採取捕捉並進行絕育後回置流浪犬(TNR)的做法,在減量與人到之間取得一個平衡,對此,臺南市政府早在2012便領先全國執行流浪犬TNR,因此本研究以臺南市為研究標的,針對臺南市的動物保護專責機關及相關利害關係人為主軸進行個案研究,就流浪犬TNR政策的執行現況做一個深入的剖析與檢視。本研究以臺南市流浪動物管理之背景脈絡與改制過程為基礎,筆者探討了流浪犬TNR政策的執行成效,並以扎根理論為基礎,來審視何種因素影響政策執行成效,最後分析影響政策執行成效之因素,進而提出政策建議;研究方法上主要運用深度訪談法,其中訪談對象包含政府機關、代議士、動物保護團體、學者專家、獸醫師、里長等六大類,共計訪談10位政策利害關係人。 本研究發現流浪犬TNR政策執行的過程中,政策目標有越來越多元的趨勢,TNR的政策目標已不單單僅有減量,隨著零安樂死政策的迫在眉睫,研究者認為,當前最重要的目標便是要避免零安樂死的弊病出現。另外,相關利害關係人對流浪犬TNR政策成效之看法仍有許多差異,整體來說,大台南地區之流浪犬數量已獲得控制,但TNR之具體成效無法單就收容所數據抑或是受訪者的片面之詞來評斷,此亦為本研究之限制;最後,本研究從臺南市政府及臺南市動保團體之觀點來分析影響臺南市流浪犬TNR政策之關鍵因素;在地方政府方面,雖然在賴清德市長的強力推動下,將TNR納入流浪動物管理藍圖中,但臺南市政府認為TNR不是唯一一條路,僅是眾多流浪犬管理政策中的一環,若TNR犬隻與民眾發生爭議,仍會以其他管理手段來安置流浪犬,將民眾權益擺優先;相較於臺南市政府,動保團體則是高度推崇TNR,認為TNR是解決當前流浪犬問題最有效的方法。此外,本研究亦發現諸多因素影響了流浪犬TNR的成效,包括了動保人士與臺南市動保處之間溝統產生問題、動保人士與臺南市動保處之分工不明確、動物管制隊的意願與能力嚴重地影響了該政策的效率、流浪犬TNR政策的配套措施仍不及政策施行的速度、民眾觀念不足、絕育補助經費不足等問題。但眾多問題中,本研究認為當前流浪犬TNR政策之「回置」是最大的問題。 本研究給予下述幾項政策建議: 1.目前臺南市政府所應該做的便是教育與說服標的團體,強而有力的宣導TNR的知識及好處,並且說明零安樂死所帶來的後遺症,能將民眾之疑慮降低;另外藉由媒體宣傳策略也能提升民眾對TNR之認知。 2.臺南市動保處應與在地動保人士配合,將權力下放,補助動保人士對回置的犬隻進行控管、照護,便能大幅降低TNR犬隻造成問題的頻率,進而讓市民接受TNR。 3.針對異地回置之爭議,目前最嚴重的弊病便是動物管制隊未能將回置之詳細情形公開周知,讓TNR的成效大打折扣,對此,動保處應公布回置犬隻之狀況,讓動保團體得以進行監督與後續的照料。 本研究旨在探究上述因素如何影響臺南市流浪犬TNR政策之執行,冀望未來能夠作為臺南市政府、甚至是中央政府的參考依據。

Preemption strategy for traffic signals at intersections near highway-railroad grade crossings

Because the operational characteristics of signalized intersections near highway-railroad grade crossings (IHRGCs) are different from those of signalized intersections located elsewhere in the traffic system, standard operational strategies do not apply. This is because safe operation at IHRGCs takes precedence over all other objectives. Because the prime objective of the current preemption methods is to clear the crossing, secondary objectives such as safe pedestrian crossing time and minimized delay are given less consideration or ignored completely. Consequently, state-of-the-practice strategies may cause serious pedestrian safety and efficiency problems at IHRGCs. Therefore, there is a definite need for research on how to improve traffic signal preemption strategies. An important element of preemption strategy is detection of trains and prediction of arrival times. However, because of the limitations of current detection technologies, estimation algorithms, etc., there is a wide range in these warning times. In this dissertation, a new train-arrival prediction algorithm was developed using detection equipment located farther upstream from the HRGC. The state-of-the-art transition preemption strategy (TPS) was developed to ensure that as preemption is initiated by approaching trains, the signal display does not change in a manner that endangers either pedestrians or drivers. However, because it does not account for the variability of predicted train arrival times, there is still a possibility of failure. Therefore, a new transition preemption algorithm that is specifically designed to improve intersection performance while maintaining or improving the current level of safety is developed. This dissertation developed a preemption strategy (TPS3) that uses better train arrival time estimates to improve the safety and efficiency of IHRGCs. The approach was simulated on a test bed in College Station, Texas, and it was concluded that the new TPS improves the safety and operation of intersections near highway-railroad grade crossings.
