Caractérisation du métabolisme cérébral chez le rat : étude par spectroscopie RMN du carbone 13 / Characterization of cerebral metabolism in rats : a 13C-NMR study

Cette étude a été réalisée dans le but d’obtenir une meilleure connaissance du métabolisme cérébral qui se trouve perturbé dans de nombreuses maladies neurologiques ou bien par des médicaments présentant des effets toxiques au niveau du système nerveux central. Pour cela, nous avons utilisé notre approche de métabolomique cellulaire qui combine les techniques de dosages enzymatiques et de spectroscopie RMN du carbone 13 avec des modèles mathématiques des voies métaboliques. Dans la première partie de ce travail, nous avons caractérisé le métabolisme des tranches de cerveau de rat en utilisant les principaux substrats du cerveau. Nos tranches utilisent ces différents substrats et conservent une haute capacité d’oxydation pendant la durée d’incubation. La deuxième partie présente une validation de notre modèle d’étude en testant l’effet du valproate, un médicament antiépileptique très utilisé. Nous montrons, non seulement une augmentation de l’accumulation du GABA et une diminution de celle de l’aspartate dues au valproate, mais aussi une perturbation des flux à travers différentes enzymes du métabolisme du cerveau. Dans la troisième partie, nous avons recherché les effets des composés guanidiniques sur le métabolisme cérébral ; ces composés augmentent l’utilisation de glucose et l’accumulation de lactate par les tranches de cerveau de rat. Nos tranches représentent donc un bon modèle pour les études métaboliques in vitro et notre approche permet d’obtenir une vision d'ensemble du devenir métabolique d'un substrat donné et d'identifier les voies métaboliques impliquées en mesurant les flux enzymatiques en absence et en présence d'un agent pharmacologique ou toxicologique / This study was conducted to obtain a better knowledge of brain metabolism which is altered in many neurological diseases or by drugs with toxic effects on the central nervous system. For this purpose, we used our cellular metabolomic approach that combines enzymatic and carbon 13 NMR measurements with mathematical modeling of metabolic pathways. In the first part of this work, we characterized the metabolism of rat brain slices by using the main substrates of the brain. Our slices metabolized and oxidized these substrates at high rates during the incubation period. The second part presents a validation of our model of study by testing the effect of valproate, a widely used antiepileptic drug. We showed that valproate not only increases the accumulation of GABA and decreases that of aspartate, but also alters fluxes through several enzymes involved in brain metabolism. In the third part, we investigated the effects of guanidino compounds on cerebral metabolism; these compounds increase glucose consumption and lactate accumulation by rat brain slices. Thus, our slices are a good model for metabolic studies in vitro and our approach provides an overview of the metabolic fate of a given substrate and allows to identify the metabolic pathways involved by measuring enzymatic fluxes in the absence and the presence of a pharmacological or toxicological agents

Matéria orgânica e a sorção de herbicidas em solos sob plantio direto nas regiões tropical e subtropical do Brasil / Organic matter and herbicides sorption in soils under no-tillage in tropical and subtropical regions of Brazil

O sistema de plantio direto (SPD) é extensivamente utilizado no Brasil por tratar-se de prática conservacionista, capaz de melhorar a qualidade do solo e diminuir os custos da produção em relação ao plantio convencional. Neste sistema as condições edafoclimáticas distintas e a adoção de diferentes culturas na entressafra sugerem diferenças na distribuição e na qualidade da matéria orgânica de solos (MOS), devido ao tempo de resiliência da palha em cobertura. A interação entre a MOS e os pesticidas já foi bastante estudada, mas apenas parcialmente esclarecida uma vez que a maioria dos estudos considera a MOS como um compartimento homogêneo, ignorando o papel de seus diferentes componentes moleculares. Assim sendo, o objetivo principal deste trabalho é avaliar o efeito da composição, distribuição e interação com a fase mineral da MOS na sorção de herbicidas aplicados às culturas de soja e milho, sob sistema plantio direto em diferentes condições edafoclimáticas: tropical (Região Centro-Oeste) e subtropical (Região Sul). Os herbicidas alaclor, bentazon e imazetapir foram selecionados devido às suas importâncias junto a essas culturas e suas propriedades físico-químicas distintas. Para tal, soram amostrados 12 Latossolos, na camada de 0 a 5 cm excluindo a palha de superfície, sendo seis representativos do Estado do Paraná (clima subtropical) e seis representativos do Estado do Mato Grosso do Sul (clima tropical). A determinação dos atributos físico e químicos, formas de ferro e alumínio, estudo da qualidade da MOS por espectroscopia de Ressonância Nuclear Magnética do 13C e fracionamento físico dos solos foram os métodos utilizados. Foi observado maior acúmulo de carbono orgânico (C.O) nos solos subtropicais devido ao maior teor de óxidos de alumínio de baixa cristalinindade, além da maior precipitação. Para os solos de ambas as regiões, a estabilização da MOS ocorre através da formação de complexos nos agregados tamanho argila ( ~ 50% do teor de C.O). Em relação às interações com a fase mineral, observou-se que os óxidos de alumínio estão relacionados à proteção física da MOS enquanto que os óxidos de ferro parecem participar das reações de decomposição e humificação através da formação de complexos com grupos carboxílicos. Para o alaclor, a presença de formas de alumínio de baixa cristalinidade diminuiu a sua sorção a MOS, entretanto a sorção deste herbicida não foi correlacionada a nenhum grupo funcional. Para o bentazon, foi observada correlação positiva entre a sorção e materiais orgânicos mais decompostos e também, com o alumínio complexado a MOS. Para o imazetapir, houve correlações positivas com as formas de ferro e alumínio e o grupo funcional carboxil. Para os três herbicidas a maior sorção foi observada nos agregados de tamanho silte (53 a 2 ?m) e, de maneira geral, a sorção nos solos tropicais foi maior que nos subtropicais. O efeito da exposição de sítios de carga após o fracionamento físico dos solos foi evidente para o imazetapir incrementando sua sorção em comparação a sorção no solo inteiro o que evidencia a maior quantidade de sítios bloqueados nos solos tropicais. / No-tillage System (NT) is widely used in Brazil due to improve soil quality and reduce production costs compared to conventional tillage. Different climatic conditions and the adoption of different cultures in the offseason suggest differences in the distribution and quality of soil organic matter (SOM) due to the resilience time of straw in coverage in NT. Interactions between SOM and pesticides has been widely studied, but only partially elucidated. In most of studies, the MOS as a homogeneous compartment and there is not the knowledge about the role of its various molecular components. Therefore, the aim of this work is to evaluate the effect of the composition, distribution and interaction with the mineral phase of SOM on sorption of herbicides applied to soybean and corn under no-tillage in tropical (Central-west) and subtropical (South Region) soils. Alachlor, bentazon and imazethapyr herbicides were selected because of its large use in these cultures and different physicochemical properties. For this, it was sampled 12 soils at 0-5 cm layer excluding the straw surface, six representing the Paraná State (subtropical) and six representative of Mato Grosso de Sul State (tropical). Physical and chemical attributes, iron and aluminum forms, study of quality of SOM by Nuclear Magnetic Resonance Spectroscopy of 13C and soil physical fractionation methods were used for this purpose. Greater accumulation of organic carbon (OC) was observed in subtropical soils due to higher low crystalline aluminum oxides contents, in addition to lower temperatures and higher moisture in this region. For soils of both regions, the SOM stabilization occurs through the formation of complexes in the clay fraction (~ 50 % of the OC content). Regarding interactions with the mineral phase, it was observed that aluminum oxides are related to physical protection of SOM while iron oxides appear to participate in the decomposition and humification reactions by forming complexes with carboxylic groups. For alachlor, low crystallinity aluminum forms decreased its MOS sorption but sorption of this herbicide was not correlated with any functional group. For bentazon, positive correlation was observed between sorption and more decomposed organic material and with the complexed aluminum in MOS. For imazethapyr, there were positive correlations with iron and aluminum forms and carboxyl functional group. For the three herbicides better sorption was observed in the silt size fractions (53-2 ?m) and in general, sorption in tropical was higher than in subtropical soils. The effect of charge sites exposure after soil physical fractionation was evident for imazethapyr increasing its sorption compared to the whole soil which shows the greatest amount of blocked sites in tropical soils.

Novel Superacidic Ionic Liquid Catalysts for Arene Functionalization

There is a continuing interest in the subject of arene carbonylation, especially in strong acids and environmentally-benign alternatives are sought to HF/BF3 and to AlCl3 as conversion agents. Ionic liquids offer a powerful solvent for useful conversion agents such as aluminum chloride. The ILs permit AlCl3 to be used at lower HCl partial pressures than with other solvents. The superior reactivity demonstrated by acidic, chloroaluminate ILs is probably due to their enhanced solvation power for HCl and CO. Addition of HCl gas increased reactivity of the system by forming Brnsted acids, and toluene carbonylation is a Brnsted demanding reaction. It was found that reaction is stoichiometric in Al species and only intrinsically acidic ILs are active for toluene carbonylation, therefore it was possible to correlate observed conversion with predicted amounts of Lewis + Brnsted acids. Molecular modeling provided information about the different species present in these ILs and predicted 1H NMR, and 27Al NMR spectrum. Predictions suggested that three types of HCl species are present; and these predictions were confirmed using data of 13C-labeled acetone and its 13C-NMR spectra. These data showed that only one of the three types of HCl in the IL were super acidic. Reactivity towards arene formylation can be tuned by adjusting the ligands R and R in the organic cation and by changing the anion. This reactivity tuning can be exploited in a process where high acidity is required for the conversion of substrate but where separation of product from IL is facilitated by low acidity.

Furthermore, the convergence of syntons main purpose was to study the glycosylation, ie been explored two methods of forming glycosidic linkages, the method of trichloroacetimidate and method of isomerization of the allyl glycoside donor. The preparation of donors and acceptors glycosidic sequences involving protection and deprotection of hydroxyl groups from adenosine and D-glucose, respectively, favoring the formation of armed donors and observation of stereochemistry at anomeric carbon. The and anomers formed in this synthesis sequence were separated, but the only reason anomers were characterization by usual methods of analysis of 1D and 2D NMR spectroscopy, mass and infrared. Thus, we synthesized two glycoside acceptors (17) and (18) and five donor glycoside (9), (11), (11), (13) and (13), all substances are novel. Among the glycosylation assays only the method of tricloroacetimidate gave the desired product (glycosylation between 11 and 17) that is the object of characterizing and whose data has not yet been completed and conclusive.

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / Conselho Nacional de Desenvolvimento CientÃfico e TecnolÃgico / Este trabalho relata o estudo fitoquÃmico das sementes de Aspidosperma pyrifolium (Apocinaceae), descrevendo o isolamento e a determinaÃÃo estrutural de um alcaloide inÃdito com esqueleto plumerano rearranjado, o (-)-(3S,7S,21R)-rel-(3αH)-15(14→3)-abeo-2,16,17,20,6,7-hexahidro-15H,8aH,16a,20a-etano-1H-indolizino[3,1-cd]carbazol, alÃm de onze compostos conhecidos, seis alcaloides plumeranos identificados como aspidospermina, desmetoxiaspidospermina, pirifolina, 15-desmetoxipirifolina, aspidofractinina e N-acetilaspidofractinina; um alcaloide tetra-hidro-β-carbolÃnico, a N-metilacuamidina; dois iridoides glicosilados, Ãcido logÃnico e loganina; um derivado do Ãcido salicÃlico, o Ãcido 2-hidrÃxi-3-O-β-D-glicopiranosilbenzÃico; e um derivado metilado do inositol, o 2-O-metil-L-quiro-inositol. Os compostos foram isolados por tÃcnicas cromatogrÃficas, principalmente a Cromatografia LÃquida de Alta EficiÃncia, e suas estruturas foram determinadas atravÃs de anÃlises por RMN, uni e bidimensionais, IV e EMAR, alÃm da comparaÃÃo com dados da literatura. Apesar de jà descritos na literatura, cinco compostos sÃo relatados pela primeira vez para a espÃcie, e um està sendo relatado pela primeira vez como produto natural, de origem vegetal. A revisÃo dos dados de RMN da literatura de alguns alcaloides isolados, e as correspondentes correlaÃÃes estruturais, juntamente com um levantamento bibliogrÃfico (de 1973 a 2013) dos dados de RMN 13C de alcaloides plumeranos isolados da famÃlia Apocinaceae, tambÃm sÃo descritos neste trabalho. A fraÃÃo aquosa resultante da partiÃÃo lÃquido-lÃquido do extrato etanÃlico das sementes de A. pyrifolium, fonte da maioria dos compostos isolados, apresentou atividades antinociceptiva e anti-inflamatÃria nos testes de formalina, contorÃÃo abdominal induzida pelo Ãcido acÃtico e edema de pata por carragenina. / This work reports the phytochemical analysis from seeds of Aspidosperma pyrifolium (Apocynaceae), describing the isolation and the structural characterization of a unknown alkaloid with a rearranged plumeran skeleton, the (-)-(3S,7S,21R)-rel-(3αH)-15(14→3)-abeo-2,16,17,20,6,7-hexahydro-15H,8aH,16a,20a-ethano-1H-indolizino[3,1-cd]carbazole, in addition to eleven known compounds, six plumeran alkaloids identified as aspidospermine, demethoxyaspidospermine, pirifoline, 15-demethoxypirifoline, aspidofractinine and N-acetylaspidofractinine; a tetrahydro-β-carboline alkaloid, the N-methylakuammidine; the glycosides of two iridoids, loganic acid and loganin; a salycilic acid derivative, 2-hydroxy-3-O--D-glucopyranosylbenzoic acid; and a methyl inositol derivative, the 2-O-methyl-L-chiro-inositol. The compounds were isolated by chromatographic techniques, especially High Performance Liquid Chromatography, and their structures were characterized by 1D and 2D NMR spectroscopy, FT-IR and HRESIMS, and comparison with data from literature. Altough already reported in the literature, five compounds are being reported for the first time for the species, and one is being reported from a natural source. The NMR data, and the correspondent assignments of some alkaloids already reported in the literature, together with a bibliographic survey (from 1973-2013) of the 13C NMR data of plumeran alkaloids isolated from the Apocynaceae family, are presented in this work. The residual aqueous fraction of the liquid-liquid partition of the ethanol extract from seeds of A. pyrifolium, source of most compounds isolated, showed antinociceptive and anti-inflammatory activities in the formalin test, abdominal writhing induced by acetic acid, and paw edema induced by carrageenan.

FundaÃÃo Cearense de Apoio ao Desenvolvimento Cientifico e TecnolÃgico / O presente trabalho relata o estudo quÃmico do caule de Cestrum laevigatum Schlecht (Solanaceae), coletado no municÃpio de Pacoti-CE. A partir da investigaÃÃo do extrato etanÃlico utilizando tÃcnicas clÃssicas de cromatografia de adsorÃÃo (gel de sÃlica e sÃlica flash) e exclusÃo (Sephadex LH-20) foi possÃvel o isolamento de esterÃides e um triterpeno. Os esterÃides de esqueleto espirostano foram caracterizados como uma mistura de sapogeninas 25(S)-5-espirostano-2,3-diol e 25(R)-5-espirostano-2,3-diol (gitogenina) (CL-1), a mistura de epÃmeros (25R)-5-espirostano-2,3-diol-3-O--D-galactopiranosÃdeo e (25S)-5-espirostano-2,3-diol-3-O--D-galactopiranosÃdeo (CL-2), e a mistura epimÃrica da saponina de carÃter inÃdito na literatura (25R,S)-5-espirostan-2,3-diol-3-O--D-galactopiranosil(1→4)- -D-galactopiranosil(1→4)--D-glicopiranosil (CL-3). AlÃm destes compostos, foram isolados ainda a mistura dos esterÃides -sitosterol e estigmasterol (CL-5) e seus derivados glicosilados (CL-6), alÃm do triterpeno pentacÃclico de esqueleto do tipo lupano β-lup-20(29)-en-3-ol (lupeol) (CL-4). A caracterizaÃÃo estrutural dos compostos isolados foi possÃvel atravÃs do uso de tÃcnicas espectroscÃpicas, principalmente RMN, incluindo seqÃÃncias de pulso uni e bidimensionais, e comparaÃÃo com dados relatados na literatura. Das saponinas investigadas quanto ao potencial antimicrobiano, apenas CL-3 mostrou-se ativa contra Bacillos subtilis, Pseudomonas aeruginosa e Enterobacter aerogenes. A atividade leishmanicida foi realizada com CL-1, CL-2 e CL-3, porÃm, apenas CL-3 mostrou-se ativa contra Leishmania amazonensis. Em relaÃÃo à atividade antifÃngica, CL-1 mostrou-se ativa apenas contra Microsporum canis. CL-2 apresentou excelentes resultados contra Candida albicans, Trichophyton rubrum e Trichophyton mentagrophytes, e uma atividade relativamente boa contra Microsporum canis. Jà CL-3 aparece revelou uma excelente atividade antifÃngica contra todas as cepas testadas. Os resultados promissores obtidos atà o momento a partir do extrato etanÃlico do caule de C. laevigatum confirmam o grande potencial quÃmico/farmacolÃgico das espÃcies deste gÃnero, e justificam a importÃncia do trabalho realizado / This work reports the chemical study of the stem of Cestrum laevigatum Schlecht (Solanaceae), collected at the Pacoti-CE. The investigation of the ethanol extract using conventional techniques of adsorption chromatography (silica gel and silica flash) and exclusion (Sephadex LH-20) was possible the isolation of steroids and triterpene. The spirostan steroids have been characterized as the mixture at the sapogenins 25(S)-5-espirostan-2,3-diol and 25(R)-5-espirostan-2,3-diol (gitogenin) (CL-1), a mixture of epimers (25R)-5-espirostan-2,3-diol-3-O--Dgalactopyranoside and (25S)-5-espirostan-2,3-diol-3-O--D-galactoyiranoside (CL-2), and the mixture of new epimer (25R,S)-5-espirostan-2,3-diol-3-O--Dgalactopyranosil(1→4)--D-galactopyranosil(1→4)--D-glicopyranosil (CL-3). Besides these compounds, were also isolated the mixture of steroids β-sitosterol and stigmasterol (CL-5) and its glycosides (CL-6), and the pentacyclic triterpene β-lup-20(29)-en-3-ol (lupeol) (CL-4). The structural characterization of the isolated compounds was possible through the use of spectroscopic techniques, mainly NMR, including sequences of uni and bidimensional pulse, and comparison with literature data. CL-3 was shown to be active against Bacille subtilis, Pseudomonas aeruginosa and Enterobacter aerogenes. The leishmanicide activity was performed with CL-1, CL-2 and CL-3, however, only CL-3 was shown to be active against Leishmania amazonensis. For the antifungical activity, CL-1 was shown to be active only against Microsporum canis. CL-2 showed excellent results against Candida albicans, Trichophyton rubrum and Trichophyton mentagrophytes, and a relatively good activity against Microsporum canis. CL-3 appears with excellent activity antifungical against all strains tested. The promising results obtained so far from the ethanol extract of the stem of C. laevigatum confirm the great potential chemical / pharmacological species of this genus, and justify the importance of work

La connaissance du comportement sous irradiation γ et en présence d'eau des Résines Echangeuses d'Ions est nécessaire pour prévoir leur impact sur l'environnement pendant la phase d'entreposage et dans un éventuel stockage en profondeur géologique. Les REI étudiées sont la résine MB400 en lit mélangé ainsi que ses composants anionique et cationique « purs ». La stratégie expérimentale suivie a été basée sur l'utilisation d'outils chimiométriques qui ont permis d'étudier l'effet du milieu d'irradiation, du débit de dose, de la dose et de la température de lixiviation. Les produits de radiolyse gazeux et hydrosolubles ont été analysés par Spectrométrie de Masse gaz et Chromatographie Ionique. Les REI génèrent principalement du H2g, du CO2g et des amines dont les quantités dépendent de la nature de la résine et des conditions d'irradiation. L'analyse des résines solides irradiées a été effectuée par spectroscopie Infrarouge à Transformée de Fourrier et par Résonance Magnétique Nucléaire. Ces techniques révèlent des modifications structurales différentes suivant les conditions d'irradiation. Le comportement sous eau des REI a été étudié sur une période de 143 jours en caractérisant la matière organique relarguée après lixiviation post-irradiation. Les études cinétiques montrent qu'au premier contact avec l'eau, toutes les espèces hydrosolubles sont relarguées. La quantité de Carbone Organique Total dépend, selon la nature de la résine, soit de la dose, soit du milieu d'irradiation. Le débit de dose n'a pas d'effet sur la dégradation et la lixiviation de la résine MB400 qui, néanmoins se comporte d'une façon différente de ses composants pris séparément. / The knowledge of the behavior under irradiation and in presence of water of Ion Exchange Resins (IER) is very necessary to predict their impact on the environment during the storage phase and in a possible deep geological disposal. The IER studied are the MB400 mixed bed resin and its « pure » anionic and cationic components. The experimental strategy used in this work was based on the use of chemometric tools permitting to estimate the effect of the irradiation atmosphere, the dose rate, the absorbed dose and the leaching temperature. The gaseous and water-soluble radiolysis products were analyzed by gas Mass Spectrometry (MS) and Ion Chromatography (IC). The IER generated principally H2g, CO2g and amines for which quantities depended of the resin nature and the irradiation conditions. The analysis of solid irradiated resins was investigated by Fourier Transformed Infrared (FTIR) and Nuclear Magnetic Resonance (13C NMR) techniques. The last ones revealed structural modifications of the IER solid matrix in function of the experimental conditions. Their behavior in presence of water was studied during 143 days by characterization of the organic matter released after their post-irradiation leaching. The kinetics showed that all the water-soluble components were releasing at the first contact with water. The Total Organic Carbon (TOC) quantity released depends, according to the resin nature, either on the dose, either on the irradiation atmosphere. The dose rate has no effect on the degradation and the leaching of the MB400 resin, which behaved differently than its pure components.

The main aim of doctoral thesis is the study on physicochemical properties of humic substances (HS) by modern instrumental techniques. The subject of the study were HS isolated from South Moravian lignite, South Bohemian peat, forest soil Humic Podzol and finally extract from brown sea algae Ascophyllum nodosum. With respect on determination of structure and reactivity of these unique “biocolloids”, standard samples (Leonardite HA, Elliott Soil HS and Pahokee Peat HS) were also studied. These samples were obtained from International Humic Substances Society (IHSS). All mentioned substances were characterized by elemental analysis (EA), molecular absorption spectroscopy in ultraviolet and visible region (UV/Vis), infrared spectroscopy with Fourier transformation (FTIR), nuclear magnetic resonance spectroscopy of carbon isotope 13C (LS 13C NMR), steady-state and time resolved fluorescence spectroscopy. Obtained fluorescence, UV/Vis and 13C NMR spectra were used for calculation of fluorescence and absorption indexes, values of specific absorbance and structural parameters respectively, which were used for fundamental characterization of these “biocolloidal” compounds. Infrared spectroscopy with Fourier transformation was utilized for the identification of functional groups and structural units of HS. Evaluation of infrared spectra is quiet complicated by overlapping of absorption bands especially in fingerprint region. This problem was overcome by Fourier self-deconvolution (FSD). Steady-state fluorescence spectroscopy was used for deeper characterization of HS with respect to origin, structural units, amount of substituents with electron-donor and electron-acceptor effects, content of reactive functional groups, “molecular” heterogeneity, the degree of humification, etc. Parameters of complexation of samples Elliott Soil with heavy metal ions (Cu2+, Pb2+ and Hg2+) were obtained by using modified Stern-Volmer equation. These ions were chosen purposefully, because the interaction of HS with these ions is one of the fundamental criteria for the assessment of the reactivity of HS. Key part of the whole doctoral thesis is time-resolved fluorescence spectroscopy. It is able to determine the origin of emission of HS by method Time-Resolved Area Normalized Emission Spectra (TRANES). The viscosity of micro medium about excited fluorophores of HS was determined by Time-Resolved Emission Spectra (TRES).

Water Resistance of Scots Pine Joints Produced by Linear Friction Welding / Résistance à l'eau du joint de soudure par friction linéaire du Pin Sylvestre

Le soudage du bois est une technique d'assemblage sans adhésif de deux pièces de bois, leur soudure étant produite par friction mécanique sous pression des deux pièces. Ce procédé, applicable à des pièces de bois plates, d'essences identiques ou différentes, se prête à la fabrication de meubles et à la menuiserie. Cependant, le joint obtenu n'est pas de classe "extérieur", ce qui le réserve à un usage "intérieur". En effet, un joint destiné à une utilisation extérieure ou en milieu à humidité variable doit présenter une résistance élevée à l'eau. L'objectif principal de cette thèse est d'étudier la résistance à l'eau du bois soudé. A cet effet, des méthodes d'essais complémentaires et non-destructrices ont été utilisées, comme le scanner ou l'imagerie par résonance magnétique (IRM). L'influence des paramètres de soudage et des propriétés du bois sur, d'une part, la formation et la propagation des fissures dans la ligne de soudure, et sur, d'autre part, la densité et l'absorption d'eau de la soudure a été ainsi étudiée. Les expériences de cette thèse seront menées sur des échantillons de pin (Pinus sylvestris) de dimensions 200 mm x20 mm x 40 mm, coupés dans la direction longitudinale du fil du bois. La Norme Européenne EN 205 a servi de cadre pour déterminer la résistance des échantillons de pin en traction-cisaillement. Les méthodes d'essais (non-destructrices) ont été utilisées selon leur pertinence: le scanner a servi à étudier la formation et la propagation des fissures; l'imagerie par résonance magnétique (IRM) a permis quant à elle de caractériser la pénétration et l'infiltration d'eau dans le bois soudé.Le mécanisme d'adhérence du pin a été étudié grâce à la RMN MAS (spectrométrie à résonance magnétique nucléaire avec polarisation croisée et rotation à l?angle magique) du carbone13 et à la micro-densitométrie par rayons X. Ces différentes méthodes, non destructrices, offrent l'avantage d'une analyse non invasive et l'élimination de facteurs parasites liés à la préparation et à la coupe du bois. Voici en résumé les résultats obtenus les plus marquants: (1) Le scanner et l'imagerie par résonance magnétique (IRM) sont des méthodes de recherche particulièrement polyvalentes et adaptées à l'étude des bois soudés. (2) L'utilisation de bois de coeur, une pression de soudage de 1.3 Mpa et un temps de soudage de 1.5 s permettent d'augmenter la résistance à l'eau du pin soudé. (3) Des tests d'optimisation ont montré que la résistance du pin en traction-cisaillement est plus sensible aux variations de temps de soudage qu'au temps de refroidissement et qu'elle peut être optimisée à plus de 9.7 MPa en respectant une pression de 1.3 Mpa, un temps de soudage > 3.5 s et un temps de refroidissement < 60 s. (4) La résistance à l'eau du bois soudé peut être améliorée dans une certaine mesure en faisant varier paramètres de soudage et propriétés des essences, mais dans tous les cas, le recours à un imperméabilisant naturel et écologique reste nécessaire. (5) Le pin soudé possède une résistance à l'eau et en traction-cisaillement inhabituellement élevée, cela pouvant s'expliquer par une teneur en composés extractifs augmentée. (6) Des essais sous IRM ont montré que les causes de rupture du joint varient suivant l'essence: faible résistance à l'eau de la ligne de soudure dans le cas du hêtre soudé, retrait et expansion du bois dans le cas du pin soudé. (7) Les extractifs du pin améliorent nettement la résistance à l'eau du joint soudé, mais à un niveau qui ne lui permet cependant pas la certification "extérieur" sans protection. En revanche, il peut être certifié "semi-extérieur" avec protection. / Wood welding is a mechanical friction process allowing the assembly of timber without any adhesives. The process consists of applying mechanical friction, under pressure, alternately to the two wood surfaces to be welded. This process can be applied to weld two flat pieces of timber, originating from the same or different tree species, and can be used in the manufacture of furniture and wood joinery. The only limitation is that the joint is not exterior-grade, but only suitable for interior joints. Exterior use, or use in an environment with varying humidity demands water resistance of the welded joints. The main objective of this thesis is to study the water resistance of the welded wood. This is complemented with special attention to non-destructive test methods such as X-ray Computed Tomography (CT-) scanning and Magnetic Resolution Imaging (MRI). The influence of welding parameters and wood properties on crack formation and crack propagation in the weldline was investigated. The influence of these parameters on weldline density and water absorption in the weldline were also studied. Investigations in this thesis are based on welded samples of Scots pine (Pinus sylvestris) of the dimensions 200 mm × 20 mm × 40 mm which were cut in the longitudinal direction of the wood grain. The tensile-shear strength of the welded Scots pine samples were determined using European standard EN 205. Different non-destructive methods such as X-ray Computed Tomography (CT-) scanning to study crack formation and propagation, and magnetic Resolution Imaging (MRI) to characterize water penetration and the distribution mechanism in welded wood were used. Solid state CPMAS 13C NMR spectrometry and X-ray microdensitometry investigations were carried out to study the mechanism of adhesion in Scots pine. These various non-destructive methods offer the advantage of non-invasive analysis and the elimination of any artifacts present due to preparation and sectioning. The most important results are summarized as follows: (1) X-ray Computed Tomography (CT-) scanning and Magnetic Resolution Imaging (MRI) are versatile research methods applicable to investigations of welded woods. (2) Water resistance of welded Scots pine can be increased using heartwood, a welding pressure of 1.3 MPa, and a welding time of 1.5 s. (3) Optimization tests showed that the tensile-shear strength of Scots pine was more sensitive to welding time changes than holding time and could be optimized to more than 9.7 MPa using 1.3 MPa welding pressure, > 3.5 s welding time, and < 60 s holding time. (4) Changing welding parameters and wood properties can increase water resistance of welded wood to some extent, but treating the weldline with certain natural and environmentally-friendly water repellents is still necessary. (5) Welded Scots pine shows unusually high water resistance and tensile-shear strength. This may be explained by there being more extractives compounds in Scots pine. (6) MRI experiments showed that the origin of the joint failure in welded beech is poor water resistance of the weldline, while swelling and shrinkage of wood are the main reasons for joint failure of welded Scots pine. (7) Extractives in Scots pine dramatically improve water resistance of the welded joint, but not to a level to classify the joint as an unprotected exterior grade. However, it can qualify as a joint for protected semi-exterior application.
