Synthesis and anticancer activity evaluation of η5-C5(CH3)4R ruthenium complexes bearing chelating diphosphine ligands

Yes / The complexes [RuCp*(PP)Cl] (Cp* = C5Me5; [1], PP = dppm; [4], PP = Xantphos), [RuCp#(PP)Cl] (Cp# = C5Me4(CH2)5OH; [2], PP = dppm; [5], PP = Xantphos) and [RuCp*(dppm)(CH3CN)][SbF6] [3] were synthesized and evaluated in vitro as anticancer agents. Compounds 1–3 gave nanomolar IC50 values against normoxic A2780 and HT-29 cell lines, and were also tested against hypoxic HT-29 cells, maintaining their high activity. Complex 3 yielded an IC50 value of 0.55 ± 0.03 μM under a 0.1% O2 concentration.

Reconnaissance de surfaces de protéines par des foldamères aromatiques / Protein surface recognition by aromatic foldamers

Les interactions protéine-protéine jouent un rôle primordial dans de nombreux processus biologiques. L’importance de ces interactions a suscité le développement de nouvelles approches thérapeutiques qui ciblent ces complexes protéiques. Nous nous proposons d’inhiber ces interactions en élaborant une stratégie de reconnaissance de surfaces de protéines par des molécules synthétiques de taille intermédiaire, les foldamères d’oligoquinoline. Ces composés se replient en des structures hélicoïdales stables dont chaque élément constitutif peut être fonctionnalisé pour permettre des propriétés de reconnaissance de surface de protéine.Afin de valider ce concept, l’interaction entre l’anhydrase carbonique humaine de type II (HCAII) et son inhibiteur N-benzyl-4-sulphamoylbenzamide (SBB) a été sélectionnée comme système modèle. Plusieurs étapes de synthèse ont permis de concevoir de nouveaux foldamères capables de former un complexe avec l’enzyme par l’intermédiaire de l’inhibiteur SBB et d’un espaceur approprié. Chaque complexe protéine-foldamère a été co-cristallisé et l’affinité des interactions a été caractérisée par dichroïsme circulaire induit et par résonance plasmonique de surface. Ce concept a ensuite été appliqué à une interaction protéine-protéine d’intérêt thérapeutique, le complexe IL-4/IL-4R, dans le cadre du programme européenFOLDAPPI (FP7-PEOPLE-IAPP-2008). / Protein-protein interactions play key roles in many biological processes as well as in many diseases. The importance of these interactions has led to the development of new therapeutic approaches that target protein interfaces. We have developed a protein surface recognition strategy to inhibit protein-protein interactions by using intermediate size organicmolecules called oligoquino line foldamers, that result in very stable and well defined helical structures. These helical backbones are used as templates within each building block can be modulated to allow protein surface recognition.In order to validate this concept, the well-characterized interaction between the enzyme human carbonic anhydrase II (HCAII) and its N-benzyl-4-sulphamoylbenzamide (SBB) inhibitor was selected as a model system. Multi-steps synthesis allowed functionalization of new foldamers able to bind to the enzyme through the SBB inhibitor attached by a spacer.Each foldamer–protein complex was cocrystallized and the affinity of the interactions was assayed using both induced circular dichroïsm and surface plasmon resonance. The concept of using a foldamer against protein-protein interaction was then applied to a protein complex of therapeutic interest, IL-4/IL-4R, within the European FOLDAPPI program (FP7-PEOPLEIAPP- 2008).

A Study about the use of Resources to Strengthen the Network Position : A Comparative Case analysis within the Defense industry

This qualitative research has been performed at Saab Dynamics with the purpose to analyze how resources are used in Saab to improve their processes and increase their market share. The theory used is founded in IMP theory with the implication that companies must move their focus from only considering internal resources into using their network. To analyze the different types resources in the network the 4R model has been used and adopted to two different cases at Saab Dynamics. The first case is about Saabs procurement of one of its suppliers, ACAB, and the second case is about a Country Xs procurement of the man-portable air-defense system RBS 70 NG. To gather data about the cases interviews has been performed with employees at Saab as well as other stakeholders. The result from these interviews were then analyzed based on the 4Rs, answering the questions: What Products are involved in the product development?  Which Production Facilities are involved in the product development? Which Business Units are involved in the product development? What Business relationships are involved in the product development? Once the resources were analyzed and compared to separately, the following question was answered: How have resources been combined to expand and strengthen the network? The result from the research showed that Saab has a great focus on its suppliers and how they should structure the network for the best potential end-result. In the first case, the procurement resulted in factors such as shorter lead-times, smoother processes and better quality products. The second case showed that Business Relationship resources such as trust played a great part in the procurement. To grow a stronger Business Relationship, Saab used a supplier in Country X which in turn also ended up being a part of the creation of a new innovative component.  The end discussion is about the benefits that the procurement of ACAB and weather this could be used as a way for Saab to expand their network while also gaining more control over the Supply Chain. This is something that goes against modern-day literature which often argue that the opposite often is to prefer. In the end there is also a short ethical section with the aim to examine why people chose to work in an industry like Saab Dynamics. To gather the data a few interviews were made with the end result that the respondents believe that the industry is necessary for the protection of Sweden and that it “is a human right to feel safe”.

Le magnétisme moléculaire est une discipline qui regroupe le magnétisme de molécules relativement simples mais aussi celui de systèmes très complexes : molécules à haut spin, aimants à base moléculaire, molécules-aimants… Le travail présenté ici se place dans ce contexte et abord la synthèse et l’étude détaillée des structures cristallines et des propriétés magnétiques de complexes polymétalliques à base de métaux 3d et 4f. L’une des préoccupations majeure de ce travail concerne la compréhension des relations magnéto-structurales dans ces systèmes. Dans le premier chapitre nous donnons quelques éléments de magnétisme moléculaire et les principaux résultats dans le domaine des matériaux à base moléculaire. Dans le deuxième chapitre de ce travail nous présentons les résultats variés obtenus grâce à l’association d’un ligand simple, la 2-hydrosypyridine, avec des ions de CuII et LnIII. Les synthèses réalisées ont permis d’obtenir deux séries différentes de composés : hexanucléaires de type [Cu4Ln2] et décanucléaires de type [Cu8Ln2]. Dans le troisième chapitre nous avons focalisé notre étude sur la synthèse et caractérisation d’un complexe à valence mixte de manganèse comportant le cation [MnII(MnII3MnIII3)L12]5+, ou L = 2-hydroxybenzimidazole. Dans le dernier chapitre nous présentons les résultats de l’étude théorique et expérimentale des interactions antiferromagnétiques et de l’anisotropie dans un composé dinucléaire de CoII à partir de mesures effectuées sur des monocristaux de grande taille. Cette étude a été complétée par une étude par diffraction de neutrons polarisés selon les trois directions cristallographiques a, b et c d’un monocristal / Molecular magnetism is a field which gathers the magnetism of the simplest molecules but also that of very complex systems : high-spin molecules, molecular-based magnets, single molecule magnets, magnetic supramolecular buildings. Coordination chemistry is one way to obtain compounds with specific magnetic properties : ferromagnetic exchange interactions, uniaxial anisotropy, slow relaxation and quantum tunnelling of magnetization. In this context, the work presented here reports the synthesis and the detailed study of the crystal structures and interest of this study is the understanding of the magneto-structural relationships. In the fist chapter we give some elements of molecular magnetism. In the second chapter we present the results obtained by the association of 2-hydroxypyridine lignad, with cuII et LnIII ions. The syntheses lead to two different series of compounds : [Tu4Ln2] and [Cu8Ln2]. In the third chapiter we foaclized our attention on synthesis and study of new heptanuclear mixed-valence manganese complex with 2-OHBzIm ligand (2-hydroxybenzimidazole). The structure is formed by [Mn7(L]12]5+ core, consisting in a nearly planar Mn7 unit comprising a central Mn held within a Mn6 hexagon. The magnetic properties of this compound show a single molecule magnet behaviour. In the last chapter we report the results of the theoretical and experimental study of the antiferromagnetic exchange interactions ad the anisotropy in a dinuclear CoII complex from measurements made on large size single crystal. Achieving Complex Motion with Fundamental Components for Lamina Emergent Mechanisms

Designing mechanical products in a competitive environment can present unique challenges, and designers constantly search for innovative ways to increase efficiency. One way to save space and reduce cost is to use ortho-planar compliant mechanisms which can be made from sheets of material, or lamina emergent mechanisms (LEMs). This thesis presents principles which can be used for designing LEMs. Pop-up paper mechanisms use topologies similar to LEMs, so it is advantageous to study their kinematics. This thesis outlines the use of planar and spherical kinematics to model commonly used pop-up paper mechanisms. A survey of common joint types is given, as well as an overview of common monolithic and layered mechanisms. In addition, it is shown that more complex mechanisms may be created by combining simple mechanisms in various ways. The principles presented are applied to the creation of new pop-up joints and mechanisms, which also may be used for lamina emergent mechanisms. Models of the paper mechanisms presented in Chapter 2 of the thesis are found in the appendix, and the reader is encouraged to print, cut out and assemble them. One challenge associated with spherical and spatial LEM design is creating joints with the desired motion characteristics, especially where complex spatial mechanism topologies are required. Hence, in addition to a study of paper mechanisms, some important considerations for designing joints for LEMs are presented. A technique commonly used in robotics, using serial chains of revolute and prismatic joints to approximate the motion of complex joints, is presented for use in LEMs. Important considerations such as linkage configuration and mechanism prototyping are also discussed. Another challenge in designing LEMs is creating multi-stable mechanisms with the ability to have coplanar links. A method is presented for offsetting the joint axes of a spatial compliant mechanism to introduce multi-stability. A new bistable spatial compliant linkage that uses that technique is introduced. In the interest of facilitating LEM design, the final chapter of this thesis presents a preliminary design method. While similar to traditional methods, this method includes considerations for translating the mechanism topology into a suitable configuration for use with planar layers of material.
