For all animals the cold represents a dreadful danger. In the event of severe heat loss, animals
fall into a chill coma. If this state persists, it is inevitably followed by death. In poikilotherms
(e.g. insects), the optimal temperature range is narrow compared to homeotherms
(e.g. mammals), resulting in a critical core temperature being reached more quickly. As a
consequence, poikilotherms either had to develop survival strategies, migrate or die. Unlike
the majority of insects, the Western honeybee (Apis mellifera) is able to organize itself into
a superorganism. In this process, worker bees warm and cool the colony by coordinated
use of their flight muscles. This enables precise control of the core temperature in the hive,
analogous to the core body temperature in homeothermic animals. However, to survive the
harsh temperatures in the northern hemisphere, the thermogenic mechanism of honeybees
must be in constant readiness. This mechanism is called shivering thermogenesis, in which
honeybees generate heat using their flight muscles.
My thesis presents the molecular and neurochemical background underlying shivering thermogenesis
in worker honeybees. In this context, I investigated biogenic amine signaling.
I found that the depletion of vesicular monoamines impairs thermogenesis, resulting in
a decrease in thoracic temperature. Subsequent investigations involving various biogenic
amines showed that octopamine can reverse this effect. This clearly indicates the involvement
of the octopaminergic system. Proceeding from these results, the next step was to elucidate
the honeybee thoracic octopaminergic system. This required a multidisciplinary approach to
ultimately provide profound insights into the function and action of octopamine at the flight
muscles. This led to the identification of octopaminergic flight muscle controlling neurons,
which presumably transport octopamine to the flight muscle release sites. These neurons
most likely innervate octopamine β receptors and their activation may stimulate intracellular
glycolytic pathways, which ensure sufficient energy supply to the muscles.
Next, I examined the response of the thoracic octopaminergic system to cold stress conditions.
I found that the thoracic octopaminergic system tends towards an equilibrium,
even though the initial stress response leads to fluctuations of octopamine signaling. My
results indicate the importance of the neuro-muscular octopaminergic system and thus the need for its robustness. Moreover, cold sensitivity was observed for the expression of one
transcript of the octopamine receptor gene AmOARβ2. Furthermore, I found that honeybees
without colony context show a physiological disruption within the octopaminergic system.
This disruption has profound effects on the honeybees protection against the cold.
I could show how important the neuro-muscular octopaminergic system is for thermogenesis
in honeybees. In this context, the previously unknown neurochemical modulation of the
honeybee thorax has now been revealed. I also provide a broad basis to conduct further
experiments regarding honeybee thermogenesis and muscle physiology. / Kälte stellt für alle Tiere eine lebensbedrohliche Situation dar. Erleiden sie einen schwerwiegenden
Wärmeverlust, stellt sich der Zustand eines Kältekomas ein. Hält dieser Zustand
über einen längeren Zeitraum an, folgt unweigerlich der Tod. Poikilotherme (z.B. Insekten)
weisen ein schmaleres optimales Temperaturfenster als Homoiotherme (z.B. Säugetiere) auf,
wodurch sie ihre kritische Körpertemperatur schneller erreichen. Dadurch waren Poikilotherme
gezwungen entweder Überlebenstrategien zu entwickeln, abzuwandern oder zu
sterben. Im Gegensatz zu den meisten anderen Insektenarten, ist die Westliche Honigbiene
Apis mellifera in der Lage einen Superorganismus zu bilden, in dem Arbeiterbienen durch
den koordinierten Einsatz ihrer Flugmuskeln für Erwärmung oder Abkühlung sorgen. In
Analogie zur Körpertemperatur von Homoiothermen, ermöglicht dies die exakte Kontrolle
der Kerntemperatur des Bienenstocks. Um unter den rauen Bedingungen in der nördlichen
Hemisphäre bestehen zu können, muss eine ununterbrochene Einsatzbereitschaft des thermogenen
Mechanismus der Honigbiene garantiert werden. Dabei ist die Honigbiene in der
Lage durch Zittern der Flugmuskulatur Wärme zu erzeugen.
In dieser Dissertation stelle ich die molekularen und neurochemischen Grundlagen des thermogenen
Muskelzitterns bei Honigbienenarbeiterinnen vor. In diesem Zusammenhang habe
ich die Signalwege von verschiedenen biogenen Aminen untersucht und konnte demonstrieren,
dass eine Erschöpfung vesikulärer Monoamine den Prozess der Thermogenese beeinflusst und
zu einem Absinken der Thoraxtemperatur führt. Unter Einbeziehung veschiedener biogener
Amine, konnten Folgeuntersuchungen zeigen, dass dieser Effekt durch Octopamin rückgängig
gemacht werden kann. Dies weist eindeutig auf eine Beteiligung des octopaminergen Systems
hin. Auf Basis dieser Erkenntnisse folgte die Erforschung des thorakalen octopaminergen
Systems der Honigbiene. Dabei erforderte es einen multidisziplinären Ansatz, um weitere
Einblicke in die Funktion und Wirkung von Octopamin in der Flugmuskulatur zu gewinnen.
Im Zuge dessen, konnten flugmuskelinnervierende octopaminerge Neuronen identifiziert werden,
die mutmaßlich die Flugmuskeln mit Octopamin versorgen. Es sind höchstwahrscheinlich
diese Neuronen, die für eine Stimulation von Octopamin-β-Rezeptoren verantwortlich
sind und wordurch intrazelluläre glykolytische Prozesse eine ausreichende Muskelversorgung gewährleisten. In den darauffolgenden Experimenten habe ich das Ansprechen des thorakalen
octopaminergen Systems auf Kältestress untersucht und konnte zeigen, dass dieses System
nach einem Gleichgewichtszustand strebt. Dies trifft selbst nach einer starken initialen
Stressantwort zu. Meine Ergebnisse verdeutlichen die Bedeutsamkeit des neuromuskulären
octopaminergen Systems und zeigen seine erforderliche Resilienz gegenüber exogenen Faktoren.
Es konnte die Kälteempfindlichkeit eines Transkriptes des Octopaminrezeptorgens
AmOARβ2 nachgewiesen werden. Zusätzlich konnte ich zeigen, dass Honigbienen ohne den
sozialen Kontext der Kolonie eine starke physiologische Störung innerhalb des untersuchten
Systems und damit auch in Bezug auf ihre Kälteresilienz aufweisen.
Meine Dissertation verdeutlicht die enorme Bedeutung des neuromuskulären octopaminergen
Systems im Kontext der Thermogenese im Organismus Honigbiene. In diesem Rahmen
konnte die bisher unerforschte neurochemische Modulation des Honigbienenthorax aufgeklärt
werden. Darüber hinaus bietet meine Arbeit eine Grundlage für künftige Experimente zur
Thermogenese und Muskelphysiologie der Honigbiene.

Engineering principles for synthetic biology : from concept to practiceCarbonell Ballestero, Max, 1988- 13 June 2016 (has links)
Synthetic Biology is a relatively new and multi-faceted interdisciplinary emergent field of research that combines biology with technology in novel and exciting ways. One of its main branches aims to see living systems engineered in a rational and straightforward bottom-up approach, like in other engineering disciplines. The inherent complex nature of living systems turns them into a difficult and challenging substrate where to apply common engineering principles such as standardization, abstraction and modularity. Efforts to overcome these limitations and adapt such principles for working upon living systems have been devoted, though yet with relative success. The aim of this Thesis is to critically explore what is Synthetic Biology and how far it is from a veritable engineering discipline. In this Thesis, we first present a review that thoroughly explores and discusses this scenario. Then, we present two works that shall contribute to this ambitious and hard goal. First, within the context of standardization, we address the need for better genetic parts characterization by providing an example of a biologically grounded framework inspired by classical enzymology theory. Second, and in relation with the principle of
modularity, we provide a theoretical framework, in this case inspired by the Ohm’s law of electric theory, that describes the unintended coupling of the coexisting genetic loads within a given host cell due to sharing a limited common pool of machinery and resources. Together, both works contribute, on one hand, to increase our understanding of the organizing principles of living systems, and on the other hand, to improve how engineering principles are applied to synthetic circuit design. Finally, these works emphasize the need to find better experimentally backed-up theoretical frameworks or models that should allow us to jump from the current time-consuming, trial-error and ad hoc Synthetic Biology to a well-established engineering discipline as fruitful and efficient with the living systems realm as other engineering disciplines are. / La Biologia Sintètica és un camp de recerca emergent relativament nou i multi-facètic que combina la biologia amb la tecnologia de formes innovadores i emocionants. Una de les seves principals branques té com a objectiu aconseguir ingenieritzar els sistemes vius des d’abaix de manera racional i senzilla, tal com passa en altres tipus d’enginyeria. La naturalesa inherentment complexa dels éssers vius els converteix en un substrat difícil sobre el qual aplicar principis d’enginyeria com l’abstracció, l’estandardització i la modularitat. S’han dedicat esforços per superar aquestes limitacions i adaptar aquests principis perquè funcionin sobre sistemes vius, tot i que encara que amb un èxit relatiu. L’objectiu d’aquesta tesi és explorar críticament què és la Biologia Sintètica i quan lluny està de ser una veritable enginyeria. En aquesta tesi, primer presentem un article de revisió que explora i discuteix a fons aquest escenari. Després presentem dos treballs que han de contribuir a aquest ambiciós i difícil objectiu. En primer lloc, en el context de l’estandardització, adrecem la necessitat d’una millor caracterització de les parts genètiques oferint un exemple de marc teòric amb fonaments biològics que esta inspirat en teoria enzimològica clàssica. En segon lloc, i relacionat amb el principi de modularitat, oferim un marc teòric, aquest cop inspirat en la llei de Ohm de la teoria elèctrica, que descriu l’aparellament no intencionat de les carregues genètiques coexistents dins d’una cèl.lula hoste qualssevol degut al fet de compartir un conjunt comú limitat de recursos i maquinària cel•lular. Ambdós treballs contribueixen, per un cantó, a incrementar el nostre coneixement sobre els principis d’organització dels éssers vius, i per l’altre, a millorar com s’apliquen els principis d’enginyeria pel disseny de circuits sintètics. Finalment, aquests treballs emfatitzen la necessitat de trobar millors marcs teòrics o models recolzats experimentalment que haurien de permetre’ns fer un salt des de l’actual Biologia Sintètica ad hoc, farregosa i basada en assaig-error, a un tipus d’enginyeria ben establerta que pugui ser tan profitosa i eficient en el reialme dels éssers vius com ho són les altres enginyeries.

Annexin IV : a pronephros specific gene with a role in pronephric developmentSeville, Rachel Alice January 2001 (has links)
The Xenopus pronephros is a simple paired organ; each nephron consists of a single large glomus, one set of tubules and a single duct. Previous work done in our laboratory demonstrates that the tubules and duct are specified at stages 12.5 and 14 respectively and the glomus is specified at stage 12.5. The kidney unit functions to control water balance and dispose of wastes. The simple organisation of the pronephros and the amenability of Xenopus laevis embryos to manipulation make the Xenopus pronephros an attractive system in which to study organogenesis. It has been shown that pronephric tubules can be induced to form in presumptive ectodermal tissue by treatment with RA and activin. Members of this laboratory have used this system in a subtractive hybridisation screen that resulted in the cloning of Xenopus laevis annexin IV (Xanx-4). In this thesis it has been demonstrated that Xanx-4 transcripts are specifically located to the developing pronephric tubules, and the protein to the luminal surface of these tubules. Temporal expression shows zygotic transcription is upregulated at the time of pronephric tubule specification. The temporal and spatial expression pattern of Xanx-4 suggests it may have a role in pronephric tubule development. Overexpression of Xanx-4 yields no apparent phenotype. Depletion of Xanx-4 dsRNA was tested and was shown to specifically reduce levels of Xanx-4 protein in oocytes. Xanx-4 siRNA was also tested for efficacy and was shown to cause a reduced pronephric tubule phenotype. However analysis of the effects of dsRNA and siRNA on mRNA levels have proved inconclusive and cast doubt on the usefulness of dsRNAs in Xenopus laevis. Further studies on RNAi in Xenopus will be required before it can be judged a reliable method for interfering with gene expression. Xanx-4 depletion, using morpholinos produces a shortened, enlarged tubule phenotype. The phenotype observed can be rescued by coinjection of Xanx-4 mRNA. Thus, Xanx-4 can be successfully depleted in embryos using morpholino oligos.

Les cèl.lules T reguladores naturals (nTregs) són limfòcits que es generen durant la selecció tímica
i que formen part dels mecanismes que té el sistema immunitari per mantenir la seva homeostasi i
controlar respostes autoreactives que poden conduir a l’autoimmunitat. Les nTregs són emigrants
tímics que expressen de forma estable FOXP3, gràcies a canvis epigenètics que ocorren durant la
seva maduració i que li permeten mantenir les funcions supressores. Les nTregs expressen també
la cadena α del receptor de la IL-2, CD25 i això els hi confereix un fenotip activat. A més de CD25,
les nTregs expressen altres marcadors com són: CTLA-4, GITR, CD127lo, CCR6, entre altres, i
que són molècules que també s’indueixen en cèl.lules T activades durant la resposta. Tot i ser un
marcador exclusiu al timus, l’expressió intracel·lular de FOXP3 el fa incompatible amb el seu ús
per la purificació de les nTregs. Més endavant es va descriure que les cèl.lules T humanes
activades també poden convertir-se en FOXP3+ i adquirir un fenotip supresor, tot i que la seva
expressió és transitòria. Aquest fet, juntament amb l’absència de marcadors exclussius, fa que l’ús
d’aquestes cèl.lules en immunoteràpia representi encara un repte. Per això, a partir de diverses
aproximacions hem estudiat el fenotip de les cèl.lules T reguladores al timus i a la perifèria.
De l´estudi exhaustiu d´una àmplia col.lecció de timus pediàtrics (n=35) d´edats compreses entre
els 2 dies i els 8 anys, hem confirmat que la freqüència de les poblacions tímiques varia amb
l´edat, tal i com s´havia descrit prèviament. A més, i per primera vegada, s´ha observat que la
freqüència de les nTregs disminueix amb l´edat. El fenotip d´aquesta població és estable amb el
temps, a excepció de CD95, membre de la familia del TNF involucrat en apoptosi i activació
cel.lulars. La freqüència de cèl.lules CD95+ dins de la població de nTregs incrementa amb l´edat, i
suggeriria un augment en susceptibilitat a l´apoptosi mediada per CD95 que explicaria la davallada
d´aquesta població amb l´edat.
Un cop les nTregs s´exporten des de timus a la perifèria, es barregen amb cèl.lules T activades i
cèl.lules T de memòria amb les que comparteixen diversos marcadors. En el nostre laboratori vam
abordar la cerca de molècules característiques de nTreg fent servir una aproximació proteòmica
seguida d´una anàlisi d´expressió gènica. De la comparació de cèl.lules nTreg de timus i nTreg de
perifèria vam definir la galectina3 com a marcador d´aquesta població que s´adquireix a perifèria.
En cèl.lules Treg activades, funcionalment supressores, la seva expressió es troba a nivells
similars de dos dels marcadors característics d´aquestes cèl.lules, FOXP3 i CTLA4.
Finalment, es va abordar l´estudi d´aquesta població en pacients amb diabetes tipus 1. Es va fer
un estudi exhaustiu de caracterització fenotípica en mostres de sang de pacients en comparació
amb controls, que va revel.lar una disminució de GITR en les Tregs. Aquest defecte no afectava a
la seva capacitat de supressió, en canvi podria estar associat a una major susceptibilitat a
l´apoptosi. / Natural regulatory T cells (nTreg) are lymphocytes generated during thymic selection and are part
of the mechanisms that the immune system uses to keep homeostasis and control autoreactive
responses that can lead to autoimmunity. nTreg are thymic emigrants that stably express FOXP3
thanks to epigenetical changes that take place during its maturation, which allow them to maintain
their supressor functions. Besides, nTregs express the alpha chain of the IL-2 receptor, CD25,
which confers them an activated phenotype. Other Treg markers are CTLA-4, GITR, CD127low
and CCR6, but they are also induced in activated T cells during the immune response. Despite
being an exclusive marker in the thymus, the intracelular expression of FOXP3 makes it non
compatible with its use for Treg purification. Later it was described that activated human T cells can
also express FOXP3 and even acquire a suppresive phenotype, although transitory. This fact,
together with the absence of exclussive markers, makes difficult the use of these cells in
immunotherapy. Thus, using several approaches, we have studied the phenotype of these cells in
the thymus and in the periphery.
After studing a numerous cohort of paediatric thymus (n=35) from donors aged from 2 days until 8
years old, we confirmed that the frequency of thymic populations changes with age, as previously
described. Also, and for the first time, we observed that nTreg frequency diminishes with age. The
phenotype of this population is stable during time, except for CD95, a member of the TNF family
involved in apoptosis and cellular activation. The frequency of CD95+ cells in nTreg population
increases with age, which suggests a higher susceptibility to CD95-mediated apoptosis, and that
could explain the decrease of nTreg with time.
Once nTregs are exported from the thymus to the periphery, they are mixed with activated T cells
and memory T cells with which they share several markers. In our laboratory we searched for
characteristical Treg molecules using a proteomic approach followed by a genetic expression
analysis. From the comparison of thymic nTreg and peripheral Treg we defined galectin 3 as a
marker acquired in periphery by nTregs. In activated Treg, functionally supressive, galectin 3
expression is found at similar levels as those of FOXP3 and CTLA-4, very characterical Treg
markers.
Finally, we studied this population in type 1 diabetes patients. We carried out an extensive
phenotypical study in PBMCs from patients compared to donors that revealed a diminished
proportion of GITR+ cells among nTregs. This defect did not affect to their suppressive ability,
instead could be associated to an increased susceptibility to apoptosis.

Influencia de los lípidos de la dieta en la iniciación del cáncer de mama experimental: Ontogenia de las enzimas de detoxificación de xenobióticos. Metabolism ode carcinógeno y daño en el ADNManzanares Serrano, Miguel Angel 18 December 2014 (has links)
El cáncer de mama es el más frecuente en las mujeres de todo el mundo. Además de los factores genéticos, epigenéticos y hormonales, existen evidencias epidemiológicas y experimentales de que los factores nutricionales y ambientales tienen un papel en la etiología y el desarrollo de esta enfermedad. Los lípidos de la dieta se han relacionado directamente con el cáncer, fundamentalmente, el de mama. El grupo de investigación ha contribuido al mejor conocimiento de los efectos y de los mecanismos de acción de las dietas hiperlipídicas de aceite de maíz, rico en ácidos grasos poliinsaturados, PUFA n-6, y el aceite de oliva virgen extra, rico en ácidos grasos monoinsaturados, MUFA n-9, y diversos compuestos bioactivos, en el cáncer de mama experimental, obteniendo un efecto estimulador y potencialmente protector, respectivamente. Estas dietas actuaron fundamentalmente sobre la etapa de la promoción de la carcinogénesis. El objetivo de este trabajo ha sido investigar si estos lípidos podían intervenir, además, sobre la iniciación mediante la modulación de la metabolización y/o detoxificación de los agentes iniciadores carcinogénicos, tanto a nivel hepático como de la propia glándula mamaria. Los resultados proceden de dos series experimentales diferentes, desarrolladas en el modelo experimental de cáncer de mama inducido en la rata con el hidrocarburo aromático policíclico 7,12-dimetilbenz(a)antraceno (DMBA). En ambas se estudiaron las enzimas metabolizadoras de xenobióticos (XMEs) de la fase I: CYP1A1, CYP1A2 y CYP1B1, y de la fase II: GSTP1 y NQO1. En la primera serie se evaluó el efecto de los lípidos de la dieta sobre la expresión en hígado y glándula mamaria de las XMEs en edades clave del desarrollo (prepuberal, puberal y postpuberal/adulta). En la segunda, se estudiaron específicamente las horas inmediatamente posteriores a la administración del carcinógeno y se analizó el efecto de los lípidos de la dieta sobre el metabolismo hepático, la generación de compuestos reactivos y el daño genómico provocado en la glándula mamaria. Los resultados han mostrado que los lípidos de la dieta son capaces de modular la expresión de las XMEs en distintas etapas del desarrollo postnatal. Además, la dieta rica en PUFA n-6 incrementó la expresión de las enzimas de fase I, en edades previas a la administración del DMBA, y aumentó la actividad de los CYP1s en las horas inmediatamente posteriores (12 y 24 horas post inducción), al mismo tiempo que disminuyó la actividad enzimática de la fase II, principalmente de NQO1. El número de metabolitos reactivos generados en hígado y el daño genómico en la glándula en los animales alimentados con la dieta rica en aceite de maíz fue mayor que en los otros dos grupos. Por otro lado, la dieta rica en aceite de oliva virgen extra y la dieta control normolipídica presentaron una actividad de fase I y II mejor coordinada, presentando una menor producción de metabolitos reactivos y menor daño en el ADN en la glándula mamaria. La concordancia de estos resultados con los parámetros de la carcinogénesis en ambas series demostró el efecto de los lípidos sobre la iniciación de la carcinogénesis mamaria experimental, además de la ya descrita modulación durante las fases de promoción/progresión. Los resultados de este trabajo, junto con otros previos sobre los efectos de los lípidos durante la maduración sexual y la diferenciación de la glándula mamaria, sugieren que estos nutrientes, en función de la cantidad y del tipo, pueden modificar diferencialmente la susceptibilidad o resistencia de la glándula mamaria al desarrollo del cáncer y, por tanto, modular las ventanas de susceptibilidad frente a la exposición a carcinógenos ambientales. / Breast cancer is the most common malignancy among women all over the World. Besides the genetic, epigenetic and hormonal factors, there are epidemiologic and experimental evidences that nutritional and environmental factors have a role in the etiology and the development of this cancer. The dietary lipids are directly related with cancer, mainly breast cancer. The research group has contributed to the best understanding of the effects and the mechanisms of action of the high corn oil, rich in polyunsaturated fatty acid (PUFA) n-6, and the high extra virgin olive oil diets, rich in monounsaturated fatty acids (MUFA) n-9 and several bioactive compounds, in the experimental breast cancer, resulting in a stimulating and a potentially protective effect, respectively. These diets mostly acted on the carcinogenesis promotion stage. The aim of this work has been to research on the intervention of these lipids, furthermore, over the initiation through the modulation of the bioactivation and/or detoxification of the carcinogenic initiator agents, both in hepatic level and the mammary gland itself. Results proceed from two different experimental series, developed in the experimental breast cancer model induced in the rat with the polycyclic aromatic hydrocarbon 7,12-dimethylbenz(a)anthracene (DMBA). In both series the xenobiotic metabolizing enzymes (XMEs) were studied, CYP1A1, CYP1A2 and CYP1B1 from the phase I, and GSTP1 and NQO1 from the phase II. The first series analyzed the effect of the dietary lipids on the XMEs expression in the liver and the mammary gland in critical development ages (prepubertal, pubertal and postpubertal/adult). The second series studied specifically the hours immediately after the carcinogen administration and analyzed the effect of the dietary lipids on its hepatic metabolism, the generation of reactive compounds and the genomic damage in the mammary gland. Results showed that dietary lipids may modulate the XMEs expression in different stages of the postnatal development. Additionally, the diet rich in PUFA n-6 increased the expression of phase I enzymes, in ages previous to DMBA administration, and raised CYP1s activity in the hours immediately after induction (12 and 24 hours), while reducing the enzyme activity of phase II, mainly NQO1. The number of reactive metabolites generated in the liver and the genomic damage in the mammary gland of the animals fed the high corn oil diet was higher than in the other groups. On the other hand, the high extra virgin olive oil diet and the normolipidic control diet exhibit a better coordinated phase I and phase II activity, showing a lower production of reactive metabolites and smaller DNA damage in the mammary gland. Concordance between these results and the carcinogenesis parameters in both series showed the effect of lipids on the initiation of the experimental mammary carcinogenesis, besides the already described modulation during promotion/progression stages. Results of this work, along with previous others on the effects of lipids during sexual maturity and mammary gland differentiation suggest that these nutrients, depending on the amount and type, can differently modify the mammary gland susceptibility or resistance to cancer development, and therefore, modulate the windows of susceptibility over the exposure to environmental carcinogens.

Epidemiology and antimicrobial resistance of Salmonella spp. and Campylobacter spp. from wild birds and poultry reared outdoorsAntillés Silva, Noelia 11 April 2014 (has links)
Campylobacter i Salmonella són els bacteris enteropatogens transmesos pels aliments
més importants a nivell mundial. Les infeccions causades per aquests bacteris
representen un greu problema econòmic i de salut pública. Ambdós bacteris tenen la
capacitat d’infectar diferents espècies d’animals domèstics i silvestres. El contacte
proper entre aquests animals i l’home és un factor de risc d’infeccions humanes. Tot i
així, hi ha poques dades sobre la incidència, susceptibilitat antimicrobiana i diversitat
genètica de Campylobacter i Salmonella en aus silvestres i aus de corral criades a l’aire
lliure al sud d’Europa. Per aquest motiu es va realitzar un ampli mostreig amb la
finalitat d’avaluar quina és la contribució de les aus domèstiques (aus de corral criades
a l’aire lliure) i silvestres (ànecs i gavines) en l’epidemiologia i resistència
antimicrobiana de Salmonella spp. i Campylobacter spp. a l’àrea del Mediterrani
occidental i Atlàntic oriental.
En aquests estudis vam trobar que les aus de cria a l’aire lliure i algunes espècies
d’ànecs i gavines (en especial la gavina corsa) són un reservori important de
Campylobacter. L’espècie de Campylobacter aïllada amb més freqüència en aus
domèstiques i gavines va ser C. jejuni, en canvi en els ànecs ho va ser C. coli. D’altra
banda, ni les aus de cria a l’aire lliure ni els ànecs són reservoris importants de
Salmonella spp., mentre que les gavines (gavina corsa i gavià argentat) són reservoris
importants d’aquest patogen. Es van aïllar una gran diversitat de serotips de
Salmonella procedents de gavines, alguns d’ells de gran importància per a la salut
pública. A la majoria de les colònies de gavines estudiades s’hi van aïllar S. Enteriditis i
S. Typhimurium, els dos serotips més importants que causen malalties transmeses per
aliments a l’home. És destacable la diferència entre les diferents espècies d’aus
portadores de Campylobacter o Salmonella. Les espècies amb hàbits copròfags i/o
carronyers van presentar una gran prevalença d’aquests bacteris.
Les aus silvestres no entren en contacte amb antimicrobians de manera natural. No és
sorprenent, doncs, que tots els aïllats de Campylobacter procedents d’ànecs fossin
susceptibles a tots els antimicrobians estudiats. En canvi, les soques de Salmonella i
Campylobacter aïllades de gavines i aus de cria a l’aire lliure van presentar resistència a
diversos antimicrobians. Les resistències principals van ser a fluoro(quinones) i a
tetraciclines, cosa que representa un problema de salut pública important ja que són
els agents més utilitzats per a tractar infeccions entèriques humanes.
Es va avaluar la diversitat genètica de Campylobacter i Salmonella mitjançant ERIC-PCR
i PFGE. La gran diversitat de soques de Campylobacter i Salmonella trobades en aus
silvestres suggereix l’existència de més d’una font d’infecció. Per altra banda, en
diverses ocasions es va detectar la mateixa soca de Salmonella en diferents colònies i
espècies de gavines. Aquest resultat suggereix un origen comú de la infecció o una
dispersió de soques de Salmonella mitjançant moviments migratoris o de dispersió de
les gavines. La detecció del mateix patró PFGE de Salmonella Kottbus tant en aus
domèstiques com en gavines suggereix l’existència d’una circulació d’aquests bacteris
entre aus de granja i aus silvestres. Tot i així, calen més estudis per a confirmar-ho.
Les dades proporcionades en aquesta tesi confirmen la importància de les aus
domèstiques i silvestres com a portadores i disseminadores de Campylobacter i
Salmonella així com de resistències antimicrobianes a l’ambient, al sud d’Europa. Cal
millorar els esforços de vigilància i desenvolupar estratègies de control adequades per
a reduir l’exposició de l’ésser humà a Campylobacter i Salmonella. / Campylobacter y Salmonella son las bacterias enteropatógenas transmitidas por los
alimentos más importantes a nivel mundial. Las infecciones causadas por dichas
bacterias representan un grave problema económico y de salud pública. Ambas
bacterias tienen la capacidad de infectar distintas especies de animales domésticos y
silvestres. El estrecho contacto entre éstos y el hombre constituye un riesgo de
infecciones humanas. Sin embargo, hay pocos datos sobre la incidencia, susceptibilidad
antimicrobiana y diversidad genética de Campylobacter y Salmonella en aves silvestres
y aves de corral criadas al aire libre en el sur de Europa. Por ello, se realizó un amplio
muestreo con el fin de evaluar cuál es la contribución de las aves domésticas (aves de
corral criadas al aire libre) y silvestres (ánades y gaviotas) en la epidemiología y
resistencia antimicrobiana de Salmonella spp. y Campylobacter spp. en el área del
Mediterráneo occidental y Atlántico oriental.
En estos estudios encontramos que las aves de cría al aire libre y algunas especies de
ánades y gaviotas (en especial las gaviotas de Audouin) son un reservorio importante
de Campylobacter. La especie de Campylobacter aislada con más frecuencia en aves
domésticas y gaviotas fue C. jejuni, mientras que en las ánades fue C. coli. Por el
contrario, ni las aves de cría al aire libre ni las ánades constituyen un reservorio
importante de Salmonella spp, mientras que las gaviotas (gaviota de Audouin y gaviota
patiamarilla) son reservorios importantes de este patógeno. Se aislaron una gran
diversidad de serotipos de Salmonella procedentes de gaviotas, algunos de ellos de
gran importancia para la salud pública. En la mayoría de las colonias de gaviotas
estudiadas se aislaron S. Enteriditis y S. Typhimurium, los dos serotipos más
importantes que causan enfermedades transmitidas por los alimentos en humanos. Es
destable la diferencia entre las distintas especies de aves portadoras de Campylobacter
o Salmonella. Las especies con hábitos coprófagos y/o carroñeros presentaron una alta
prevalencia de estas bacterias.
Las aves silvestres no entran en contacto con antimicrobianos de manera natural. No
es pues sorprendente que todos los aislados de Campylobacter procedentes de ànades
fuesen susceptibles a todos los antimicrobianos estudiados. Sin embargo, las cepas de
Salmonella y Campylobacter aisladas de gaviotas y aves de cría al aire libre
presentaron resistencia a varios antimicrobianos. Las principales resistencias fueron a
fluoro(quinolonas) y a tetraciclinas, lo que representa un problema de salud pública
importante puesto que éstos son los agentes más usados para tratar infecciones
entéricas humanas.
Se evaluó la diversidad genética de Campylobacter y Salmonella mediante ERIC-PCR y
PFGE. La gran diversidad de cepas de Campylobacter y Salmonella encontradas en aves
silvestres sugiere la existencia de más de una fuente de infección. Por otro lado, en
diversas ocasiones se detectó la misma cepa de Salmonella en diferentes colonias y
especies de gaviotas. Este resultado sugiere un origen común de la infección o una
dispersión de cepas de Salmonella mediante movimientos migratorios o de dispersión
de las gaviotas. La detección del mismo patrón PFGE de Salmonella Kottbus tanto en
aves domésticas como en gaviotas sugiere que existe una circulación de estas bacterias
entre aves de granja y aves silvestres. Sin embargo, se necesitan más estudios para
confirmarlo.
Los datos proporcionados en esta tesis confirman la importancia de las aves
domésticas y silvestres como portadoras y diseminadoras de Campylobacter y
Salmonella así como de resistencias antimicrobianas al ambiente, en el sur de Europa.
Es necesario mejorar los esfuerzos de vigilancia y desarrollar estrategias de control
adecuadas para reducir la exposición del ser humano a Campylobacter y Salmonella. / Campylobacter and Salmonella are the most important foodborne enteropathogenic
bacteria worldwide. Infections caused by these bacteria are of significant economic
and public health concern. Both bacteria have the ability to infect a variety of domestic
and wild animal species. The close contact between humans, domestic and wild
animals is an important factor contributing to human infections with these bacteria.
However, limited data exists on the occurrence, antimicrobial susceptibility and
genetic diversity of Campylobacter and Salmonella in wild birds and poultry reared
outdoors in southern Europe. Therefore, a wide sampling was performed in order to
assess the contribution of domestic (poultry reared outdoors) and wild birds
(waterfowl and seagulls) in the epidemiology and antimicrobial resistance of
Salmonella spp. and Campylobacter spp. in the western Mediterranean and eastern
Atlantic Ocean.
In these studies, we found that poultry reared outdoors, as well as certain waterfowl
species and seagulls (particularly Audouin’s gulls) are an important reservoir for
Campylobacter. The most common Campylobacter species isolated from poultry
reared outdoors and seagulls was C. jejuni, while from waterfowl was C. coli. On the
contrary, poultry reared outdoors and waterfowl seems not to be an important
reservoir of Salmonella spp, while seagulls (yellow-legged gulls and Audouin’s gulls) are
important carriers of this pathogen. A great diversity of Salmonella serotypes was
isolated from seagulls, some of them of important public health concern. The two
most important serotypes causing human food-borne disease, S. Enteriditis and S.
Typhimurium, were isolated in most of the studied seagull colonies. It is noteworthy
the differences of Campylobacter or Salmonella carriage among different birds species.
Those with coprophagic and/or scavenging habits showed a high occurrence of these
bacteria.
Wild birds do not naturally come into contact with antimicrobials. Thus, it was not
surprising the susceptibility to all of the antimicrobials tested of all Campylobacter
isolates from waterfowl. However, Salmonella and Campylobacter strains isolated from
seagulls and poultry reared outdoors showed resistance to several antimicrobial
agents. The main resistances found were to fluoro(quinolones) and tetracycline, which
is of public health concern, since these agents are the ones of choice to treat enteric
infections in humans.
Genetic diversity of Campylobacter and Salmonella was assessed by ERIC-PCR and
PFGE. The high diversity of Campylobacter and Salmonella strains found in wild birds
suggests bird infections by multiple sources. On the other hand, several common
Salmonella strains were detected in different seagull colonies and different seagull
species (yellow-legged gull and Audouin’s gull). This finding suggests a common origin
of infection or the contribution of seagulls to the spread of Salmonella strains by
dispersal or migrating movements. The detection of the same PFGE pattern of
Salmonella Kottbus in poultry reared outdoors and seagulls suggests a circulation of
the bacteria between farm and wild birds. However, more studies are needed in order
to confirm this.
The data provided in this thesis highlights the importance of domestic and wild birds as
carriers and dispersal agents of Campylobacter and Salmonella and antibiotic
resistance traits to the environment, in southern Europe. An improvement of
surveillance efforts and development of appropriate control strategies are needed in
order to reduce Campylobacter and Salmonella exposure to humans.

Uncertainty in stock assessments, spatial distribution and habitat modelling of two small pelagic fich species, sardine (Sardina pilchardus) and anchovy (Engraulis encrasicolus), in the Mediterranean from late autumm Spanish acoustic surveysTugores Ferrà, M. Pilar 25 November 2015 (has links)
Small pelagic fish are species that live in the water column and have little
relationship with the sea bottom. During the day they may form schools with feeding,
defence or energetic efficiency purposes and disperse during the night. The
importance of small pelagic fishes within the marine ecosystem rely in the proportion
of biomass they represent and the clue function they develop as intermediate links in
the energy transfer between lower and upper levels of the trophic chain. Their
populations are particularly sensitive to environmental fluctuations (Cole and
McGlade, 1998; Lloret et al., 2004) and frequently highly exploited by commercial
fisheries. These may occasionally collapse the stocks affecting both the marine
ecosystem and the fisheries they sustain.
In the Mediterranean, almost 50% of the total annual landings are attributable to
small pelagic fishes (Lleonart and Maynou, 2003). In the Western Mediterranean Sea,
sardine (Sardina pilchardus) and anchovy (Engraulis encrasicolus) are the two most
important species in terms of landed biomass and commercial interest (Lleonart and
Maynou, 2003). Despite their importance little is known about the spatial distribution
of these stocks or about the relationship between the spatial distribution and
environmental variables.
Since the ‘90s acoustic surveys are annually performed in the Spanish Mediterranean
continental shelf in late autumn (Abad et al., 1998 a,b) coinciding with anchovy’s
recruitment and the beginning of the spawning season for sardine (Abad and
Giráldez, 1993; Giráldez and Abad, 1995). Although the main goal of these surveys is
to estimate abundance and biomass of sardine and anchovy, data about the whole
pelagic community has also been gathered. Furthermore, in the last decade it was
detected an increasing appearance of other small and medium-sized pelagic species
and the application of a multi-species approach to Mediterranean fisheries
assessment was advised (Lleonart and Maynou, 2003).
The present work is structured in two sections. The first section of the work will
analyse the spatial distribution (1D and 2D) of sardine and anchovy by means of
geostatistical techniques, both transitive and intrinsic methods (Matheron, 1969).
Special attention will be paid to the estimation of the uncertainty associated to
abundance estimations and, concretely, the uncertainty caused by sampling scheme
which is thought to be one of the main contributors to random error (ICES, 1998).
The second section will explore the environmental factors that drive the presence or
absence of anchovy and sardine in late autumn. Satellite environmental data as sea
surface temperature, salinity, chlorophyll-a, photosintethic active radiation, sea
level anomalies or bottom depth will be related to the presence-absence of sardine
and anchovy stocks through Generalised Additive Models (GAM) to try to depict the
relationships that may be found between both (Bellido et al., 2008; Giannoulaki et
al., 2008). The spawning area of sardine will also be studied in order to model the
presence-absence of sardine eggs and try to understand the evolution of their stocks. / Los pequeños peces pelágicos son especies que viven en la columna de agua y que
tienen poca relación con el fondo marino. Durante el día forman bancos de peces con
el objetivo de alimentarse, defenderse de potenciales depredadores o por motivos de
eficiencia energética, dispersándose durante la noche. La importancia de los
pequeños pelágicos en el ecosistema marino radica en la proporción de biomasa que
representan y en la función clave que desempeñan como eslabones intermedios,
transfiriendo energía entre los niveles tróficos inferiores y superiores de la cadena
trófica. Sus poblaciones son particularmente sensibles a las fluctuaciones
ambientales (Cole and McGlade, 1998; Lloret et al., 2004) y frecuentemente se
encuentran en un régimen de explotación elevado por parte de las pesquerías
comerciales. Esto puede comportar de manera ocasional el colapso de sus
poblaciones explotables, afectando al ecosistema marino y a las pesquerías que las
mantinen.
Casi el 50% del total de la biomssa anual desembarcada en los puertos del
Mediterráneo provienen de los pequeños peces pelágicos (Lleonart and Maynou,
2003). En el Mediterráneo Occidental, la sardina (Sardina pilchardus) y el boquerón
(Engraulis encrasicolus) son las dos especies más importantes, tanto en términos de
biomasa como capturada como por su interés comercial (Lleonart and Maynou, 2003).
A pesar de su importancia, la distribución espacial de las poblaciones explotables es
poco conocida así como también lo es la relación de la distribución espacial con los
parámetros ambientales.
Desde principios de los años 90, se llevan a cabo campañas acústica en la plataforma
continental española del Mediterráneo a finales de otoño (Abad et al., 1998 a,b),
coincidiendo con el reclutamiento de la anchoa y la época de puesta de la sardina
(Abad and Giráldez, 1993; Giráldez and Abad, 1995). A pesar de que el principal
objetivo de estas campañas es la estimación de la abundancia y de la biomas de la
sardina y el boquerón, datos de la comunidad pelágica en conjunto también se
recogen periódicamente. Además, a lo largo de la última década se ha detectado una
aparición incremental de otras especies de pequeños y medianos pelágicos y por
tanto, se aconsejó la aplicación de un enfoque multiespecífo en la evaluación de
pesquerías del Mediterráneo (Lleonart and Maynou, 2003).
El presente trabajo se estructura en dos seciones. La primera sección del trabajo
analizará la distribución espacial (1D y 2D) de la sardina y el boquerón por medio de
técnicas geoestadísticas, tanto transitivas como intrínsecas (Matheron, 1969). Se
prestará especial atención a la incertidumbre asociasda a las estimaciones de
abundancia y, concretamente, a la incertidumbre asociada al diseño de muestro que
se considera uno de los factores que contribuyen con mayor intensidad al error
aleatorio (ICES, 1998).
La segunda sección explorará los factores ambientales que condicioinan la presencia
o ausencia de sardina y boquerón a finales de otoño. Datos ambientales obtenidos de
satélite, como la temperatura superficial del mar, la salinidad, clorofila, radiación
fotosintéticamente activa, anomalía del nivel del mar y la profundidad del fondo
marino se relacionarán con la presencia-ausencia de las poblaciones explotables de
sardina y boquerón mediante modelos aditivos generalizados (GAM, del acrónimo
inglés) para determinar las relaciones existentes (Bellido et al., 2008; Giannoulaki et
al., 2008). El hábitat de la puesta de sardina será igualmente estudiada para modelar
la presencia-ausencia de huevos de sardina e intentar entender la evolución de las
especies explotables. / Els petits peixos pelàgics són espècies de peixos que viuen a la columna d’aigua i que
tenen poca relació amb el fons marí. Durant el dia formen bancs de peixos amb
l’objectiu alimentar-se, defensar-se en front potencial depredadors o per eficiència
energètica, dispersant-se durant la nit. La importància dels petits pelàgics a
l’ecosistema marí radica en la proporció de biomassa que representen i en la funció
clau que exerceixen com esglaons intermedis, transferint energia entre les nivells
inferiors i superiors de la cadena tròfica. Les seves poblacions són particularment
sensibles a les fluctuacions ambientals (Cole and McGlade, 1998; Lloret et al., 2004) i
freqüentment es troben baix un règim d’explotació elevat per part de les pesqueres
comercials. Això pot comportar de manera ocasional el col·lapse de les seves
poblacions explotables, afectant a l’ecosistema marí i a les pesqueries que sostenen.
Gairebé el 50% del total de la biomassa anual desembarcada als ports del Mediterrani
prové dels petits peixos pelàgics (Lleonart and Maynou, 2003). Al Mediterrani
Occidental, la sardina (Sardina pilchardus) i l’aladroc (Engraulis encrasicolus) són les
dues espècies més importants, tant en termes de biomassa capturada com pel seu
interès comercial (Lleonart and Maynou, 2003). Malgrat la seva importància, la
distribució espaial de les poblacions explotables és poca coneguda així com també ho
és la relació d’aquesta distribució espaial amb variables ambientals.
D’ençà de principis dels anys 90, es duen a terme campanyes acústiques a la
plataforma continental del Mediterrani espanyol a finals de la tardor (Abad et al.,
1998 a,b), coincidint amb el reclutament de l’aladroc i amb l’època de posta de la
sardina (Abad and Giráldez, 1993; Giráldez and Abad, 1995). Tot i que el principal
objectiu d’aquestes campanyes és l’estimació de l’abundància i de la biomassa de la
sardina i l’aladroc, dades del conjunt de la comunitat pelágica també es recolleixen.
A més a més, al llarg de la darrera dècada s’ha detectat una aparició incremental
d’altres espècies de petits i mitjans pelàgics i per tant, es va aconsellar l’aplicació
d’un enfoc multiespecífic a l’avaluació de les pesqueres del Mediterrani (Lleonart
and Maynou, 2003).
El present treball s’estructura en dues seccions principals. La primera secció del
treball analitzarà la distribució espaial (1D i 2D) de la sardina i l’aladroc per mitjà de
tècniques geoestadístiques, tant transitives com intrínseques (Matheron, 1969). Es
prestarà especial atenció a la incerteza associada a les estimacions d’abundància i,
concretament, a la incertesa originada pel disseny de mostreig que és considerat un
dels factors que contribueixen amb més intensitat a l’error aleatori (ICES, 1998).
La segona secció explorarà els factors ambientals que condicionen la presència o
absència de sardina i aladroc a finals de tardor. Dades ambientals obtingudes de
satèl·lit, com la temperatura superficial de la mar, la salinitat, clorofil·la, radiació
fotosintèticament activa, anomalies del nivell de la mar i la profunditat del fons marí
es relacionaran amb la presència-absència de les poblacions explotables de sardina i
aladroc mitjançant models additius generalitzats (GAM, de l’acrònim anglès) per
determinar les relacions existents (Bellido et al., 2008; Giannoulaki et al., 2008).
L’hàbitat de la posta de la sardina serà igualment estudiada per tal de modelar la
presència-absència d’ous de sardina i intentar entendre l’evolució de les seves
poblacions explotables.
