/ Abstract : The Interactive School Development Plan of school is a public politics constituted by intersectoral and educational action and programs. It is presented as a support tool to school management elaborated by Ministry of Education (MEC) in partnership with the education secretaries. This paper focuses on the school management in three Basic Education schools in Florianópolis Municipal Education System which received direct transfer of financial resources from the National Funds of Education Development, via Interactive School Development Plan, from of 2012 to 2013. Specific objectives are to explain the Interactive School Development Plan proposals and the MEC?s intention related to this plan and to study on how schools have been managing this educational program concerning its proposed actions and goals. We adopted systematic procedures of data collection by interviewing school participants and Municipal Education secretaries involved in the Plan. The Documental analysis comprehended reports and manuals from the federal government which express this policy, as well as books and reports from the World Bank and UNESCO that approach the theme, and action plans of each participant school. To develop this qualitative research, we are grounded on dialectic and historical materialism, building the analysis from the categories of totality, contradiction and mediation. The Interactive School Development Plan monitors the actions selected by the management team on the strategic planning, modifying educational funding and management. This implicates that all information and data collected, via strategic planning, can be used to phrase new public politics aiming the optimization of public expenses on education. At the same time, Interactive School Development Plan is an accountability educational politics, which means it holds docents liable to schools results. Interactive School Development Plan is the expression of school based management which has in the efficacy and rationalization principles the essential elements of the school management proposed by the World Bank. This model consists in the strategy of decentralize the decision making to the school local levels, involving parents, teachers, students and community participation. We understand, thus, that this educational politics points to a decentralization of educational management to the school local level, while it simultaneously recentralize information, in central organ level, MEC. Although its little impact on educational indicators, this centralization-decentralization movement provokes substantial changes in schools organization and functioning, revealing the management not the education quality as its main goal.

/ Abstract : Document classification provides a way to organize information, providing a better way to understand available data. The classification task is characterized by the association of class labels to documents, aiming to create semantic clusters. The exponential increase in the number of documents and digital data demands for more precise, comprehensive and efficient ways to search and organize information. In this context, the improvement of document classification techniques using semantic information is considered essential. Thus, this paper proposes a knowledge-based approach for the classification of documents. The technique uses terms extracted from documents in association with concepts of an open domain knowledge base. Then, the concepts are generalized to a higher level of abstraction. Finally a disparity value between generalized concepts and the document is calculated, and the best ranked concept is then considered as a class label applicable to the document. The application of the proposed technique offers advantages over conventional methods including no need for training, the choice to assign one or multiple classes to a document and the capacity to classify over different subjects without the need to change the classifier.

Colorectal cancer (CRC) remains a major cause of health burden in Europe and in the western world. It is a disease of lifestyle with the western diet and a sedentary lifestyle widely implicated. This implicates CRC to be associated with other metabolic disorders like obesity and diabetes. The general consensus is that it is a disease that is hugely avoidable and treatable if detected early however reliable detection tools are still lacking. Glycosylation biomarkers have been proposed as potentially a new frontier against this disease. The work covered in this thesis pertains to the influence of O-GlcNAcylation (the transfer of a single N-acetyl glucosamine monosaccharide on serine/ threonine residues), on complex glycosylations in colon cells (HT29, HCT116 and CCD841CoN). O-GlcNAcylation is a nutrient sensor which conveys the metabolic state and indeed is aberrantly regulated in metabolic disease. CRC cells and tissues express higher levels of O-GlcNAcylation and an accompanying phenotype of high proliferation and aggressive tumours. There are numerous studies showing that silencing O-linked N-acetylglucosamine transferase (OGT), the enzyme that catalyses O-GlcNAcylation, decreases the aggressiveness or cancer-like properties in several cancer models. There is little evidence starting to accumulate that shows that in these siOGT cells or tissue there is also a change in other complex glycosylations (N-, O- or glycosphingolipids glycans). O-GlcNAcylation is known to participate in key cell processes like transcription and translation. In this preliminary work, we look at the transcriptomic expression of eighty-six glycosyl transferases and glycosidases gene transcripts when OGT is silenced. Silencing OGT resulted in several of these genes being upregulated or downregulated but only 10 genes were significantly downregulated. The translational influence of OGT on several glycosyl transferases was also explored. We used several techniques namely; fluorescent lectin microscopy, lectin aided FACS analysis and several mass spectrometry techniques (glycomics), to study the complex glycan structures and expression in siOGT colon cells. Silencing O-GlcNAcylation has little effect on the global N- and O-glycans as elucidated by glycomic profiling. However, cell surface expression of several glyco-antigens was altered in siOGT. E-cadherin, a major influencer of the epithelial-to-mesenchymal transition, is upregulated in siOGT transfected HT29 cells. Furthermore, the complex glycans it carries are altered in the process. This could in part explain some of the siOGT phenotypes noted in other studies. Using MALDI mass spectrometry analysis, in HCT116 cells we found that glycosphinogolipid (GSL) expression is altered significantly in siOGT HCT116 cells. There is a reversal of some known cancerous GSL patterns; Globo series, Gb3 and Gb4 GSLs were upregulated, while ganglioside GSLs GM1a, GM2 and GD1a are downregulated. This is the first time an association has been made between O-GlcNAcylation and GSLs. As GSLs play an important part in cell recognition, signal transduction and other cell surface properties this may be one of the key links with the siOGT phenotype. All together there is a decrease in several α2,3-sialylated glycans when OGT is silenced. For the first time in scientific literature we show the global glycan effect of silencing OGT on the expression of complex glycans particularly at the cell surface, adding several new findings.

Gene Regulation with O-GlcNAc Glycosylation by dCas9-OGT/OGA Fusion Proteins:Chou, Yu Shu January 2022 (has links)
Thesis advisor: Jia Niu / O-linked β-N-acetylglucosamine (O-GlcNAc) is a post-translational modification plays important roles in cellular network/diseases such as autophagy, transcriptional activity, protein stability, phosphorylation modification competition, cancers, neurodegenerative disorders, etc. However, there are still lots of unknown roles of it in gene regulation especially while there is existence of more than 1000 transcription factors and many other coactivators/repressors which might have influence in expression level with or without O-GlcNAc modification. In this project, we use CRISPR technology with catalytically inactive Cas9 from Streptococcus pyogenes fused with the only writer and eraser of O-GlcNAc to selectively target to the DNA sequence of interest by guide RNA to see what the result of additional modification or cleavage of O-GlcNAc on proximal proteins in gene regulation is.
A cost and time efficient way for guide RNA construction is developed in the project with one-piece PCR and Gibson assembly for 2 different backbones of guide RNA: 11 guide RNA for each. Other different cloning methods have also been used for the future work. For the future work, second reporter will be introduced to normalize the luciferase signal. In addition, new metabolic O-GlcNAc reporter and Y289L GalT/ UDP-GalNAz could be used to find out proteins which might be modify by dCas9-OGT/OGA with click reaction. Furthermore, split OGA/Turbo ID system could also be used to reduce the background and find out proteins/modification sites which might be important to the gene regulation for DNA-protein or protein-protein interaction. / Thesis (MS) — Boston College, 2022. / Submitted to: Boston College. An Investigation of I/O Strategies for MPI WorkloadsAttari, Sanya 19 January 2011 (has links)
Different techniques could be used for improving application performance in parallel systems. Studies have been shown that I/O communication delay is the main reason for different behavior of I/O intensive applications with specific requirements for performance optimization. So, using common strategies, generally defined and effective for computationally intensive applications may not have the same effect on performance improvement for these applications. Moreover, background system configuration effects on the behavior of the application and its performance. Growing use of parallel multi-core systems is an important factor in increasing performance and speeding up the applications. Since changing multi-core systems hardware is not an efficient method in satisfying different expectations of unique application, it is application developer's responsibility to design flexible and scalable code that is compatible with different environments. On the other hand, predicting application behavior and I/O requirements for I/O intensive applications with irregular communication patterns is a complicated and time-consuming task that pushes the problem to runtime impacts.
Addressing this issue, we provided an overview on different techniques used for solving this problem.
We have studied I/O bound parallel applications that use MPI as the communication method in order to define a general perspective to optimize cost performance ratio. Our designed experiments cover different setups for these applications in order to define various criteria that should be considered in design stage as well as runtime. Moreover, targeting one of the popular I/O intensive applications, we have discussed some possible solutions to speed it up on a multi-core system. / Master of Science

no tocante as a??es do curso.
This research had as an objective to identify how the dialogue occurs between the
undergraduate program in Management Engineering from UFRN and its alumni, for planning
on continuing educational activities. The research, characterized as a case study, of varied
approach, adopted a quadripolar method, in which is recommended for scientific
investigations in the area of Applied Social Sciences. Regarding the literature review, it was
considered concepts as management and measuring of information and many aspects of
continued education, moreover the authors that studied the importance of the relationship with
alumni and the Higher Education Institutions. Aiming to make feasible a proposal for the
development of a communication channel between the managers of the program in
Management Engineering from UFRN and its alumni, for planning on continuing educational
activities, the research used two techniques for collecting information: an online survey for
the graduates, and the interview, for the managers of the Management Engineering. The
online survey obtained 27,78% return, being the data processed by means of descriptive
analysis, with the aid of Microsoft Excel worksheets. For the analysis of the interviews, we
used the method of content analysis. As search results, in general, the potential of building a
canal to mediate the relationship between graduates, considering that most graduates proved
willing to maintain relationship with the course, electing through social networks the most
convenient for the occurrence of this relationship. In addition, a significant part of the
graduates demanded actions of continued education upon completion of the course, pointing
to the area of engineering as the most sought. In the interviews, the managers highlighted the
importance of keeping a relationship with the alumni, providing sources on decisions
regarding actions on the program. Despite this, there are few actions of effective
communication with the alumni, and the existing ones are operated poorly. That being said,
it is understood that this study can contribute so that the institution can develop more effective
ways of contact with its alumni, not only regarding the continued education, but also in other
institutional actions.

Information management (IM) has a prominent role in all types of organizations, including the higher education institutions. This highlighted role is derived from the perception of information as a strategical resource that resulted in significant changes in the informational behavior of the associates and in the structures of the organizations. Other factors contribute to the use of IM in universities, mainly when it is observed the growth and expansion of federal higher education institutions, especially due to the employment of the Plan of Restructuring and Expansion of Federal Universities (REUNI), that resulted in significant impacts on their demands, mostly in the management of the administrative activities and the overload of information. Such demands require that universities use tools that better manage searching, selecting, processing, storing, sharing and using information, highlighting practices of information management with more relevance. The problem?s context identified in the Undergraduate Program?s Office of the Federal University of Rio Grande do Norte (PROGRAD/UFRN) was to optimize the information management in the process of registering the incoming students utilizing the available electronic tool? This study is justified for the importance that PROGRAD/UFRN has in the institution when it comes to the number of users of its services, volume of processes, activities and documents processing through. Thus, the objective is to propose the optimization in the information management of a strategical process of PROGRAD/UFRN. The specific objectives are: a) identify research on models of IM applied to other HEI; b) characterizing the information management of PROGRAD/UFRN, with basis on the information ecological approach; c) mapping an existing strategical process in PROGRAD/UFRN with the purpose to optimize it. To fulfill such objectives a qualitative, descriptive and applied study was developed, having the research-action as a strategy. The study was divided in four phases: diagnosing the problem?s context, planning the study, intervention and evaluation. To acquire the data, it was conducted a literature review, documental analysis, structured surveys applied, semi-structured interviews, focal groups organized and performed the participant observation. The results obtained indicate that PROGRA/UFRN has been employing partially the principals of Information Ecology regarding the external environments, organizational and informational, thus the information management is not integrated yet. To optimize the information management in this institution it is necessary that the people are taken into account when it comes to productivity, the use and sharing of information. The intervention proposal was built in a collaborative, implemented and evaluated manner that subsidized a more consistent and broad proposition for its use. It is understood that the enhancement of the strategical process of registering incoming students in the institution, utilizing the available functionality in the academic system still faces some obstacles. Finally, it is recommended that other studies are continued for the research of tools that make it possible the information management in public organizations, such as UFRN, become a strategical resource to reach its complete mission and objectives.
