Webbserver från ASP.NET 4.8 till Blazor Server

This report aims to address the design of a web server with Blazor Server for the company FunRock and their mobile strategy game MMA Manager. The web server is intended to administer settings of the game's different components, where staff can, for example, search for a user or a user's belongings. The report is limited to the design of specific pages from the most frequently used ones on the web server. Furthermore, the report wishes to address usability and accessibility analysis of the previous web server due to the basis for the design of the new Blazor Server application. The purpose of converting to Blazor Server has been to contribute to more flexible hosting, faster development and better performance than their previous one, written in ASP.NET 4.8. Finally, the later part of the report addresses a more subjective analysis of the author around the work performed where reflections on the project's results are treated. / Denna rapport önskar behandla utformningen av en webbserver med Blazor Server åt företaget FunRock och deras mobila strategispel MMA Manager. Webbservern ämnar till att administrera inställningar av spelets beståndsdelar, där personal kan exempelvis söka efter en användare eller en användares tillhörigheter. Rapporten erhåller avgränsning till utformning av specifika sidor från den tidigare webbservern som används mest frekvent. Vidare önskar rapporten behandla användbarhets- och tillgänglighetsanalys av den tidigare webbservern med anledning att agera grund till den nya Blazor Server applikationens design. Syftet med utformningen av den nya webbservern har varit att bidra till mer flexibel hostning, snabbare utveckling och bättre prestanda än deras tidigare som var skriven i ASP.NET 4.8. Slutligen tar rapportens senare del upp en mer subjektiv analys av författaren kring det utförda arbetet där reflektioner över projektets resultat behandlas.

Antibiotic Overuse in the Geriatric Population

The Centers for Medicare and Medicaid are requiring long-term care facilities (LTCFs) to implement antibiotic stewardship programs (ASPs) to alleviate overuse of antibiotics in the nursing home population. Current research shows that the benefits of ASPs include improved patient outcomes, reduced adverse events related to Clostridium difficile (C-diff) infection, improvement in rates of antibiotic susceptibilities, and optimized resource utilization. This project addressed the problem of antibiotic overuse and misuse in the geriatric population and whether the implementation of an ASP reduced the overuse of antibiotics, C-diff infection, and resistance rates in the LTCF. Application of the Johns Hopkins nursing model and Centers for Disease Control framework informed this project. An ASP was implemented by the organization. This project evaluated the program preASP and postASP over a 10-month period. A descriptive analysis was used to compare the number of new antibiotic starts, C-diff cases, and resistant cases before and after ASP implementation. The total number of cases of resistance declined from 12 to 10 cases after the ASP was implemented, which was a 16.67% decline. The number of monthly new antibiotic orders for the time period evaluated declined from 120 to 110 respectively, which was an 8.3% change. There was no change in the number of C-diff infections. The results demonstrated that implementing the ASP led to a decline in antibiotic misuse, overuse, and resistance cases. This project supports social change by expanding the healthcare team's knowledge regarding the project problem and informing future interventions to be implemented to help reduce antibiotic overuse and misuse in the geriatric population.

The diploma thesis describes the analysis, specification, origin and transformation of the current editing system N.E.S.P.I, which is the intellectual property of the company WebRex s.r.o. The main aim is to separate the administration of the web sites from their content, which can be seen on the Internet and by doing so to achieve the unified and complex administration interface for the customers. By bringing the service into work we do not only save time and simplify the work of the programmers, but we are also able to reach the stage when the editing system is not only the intellectual property of the company, but also its physical property, because the customer owns only the web sites, but the possibility to administrative them is offered as further service. The service should also provide the customers with modern access to informational technologies and propose enhanced assistance for their entrepreneurial plans and ideals.

Nutrient Management in On-Site Wastewater Treatment

Groundwater and surface water contamination has been linked in the past to inadequate or failing on-site wastewater treatment and disposal systems. The on-site wastewater systems installed in coastal areas have more potential for inflicting this kind of environmental damage. This work studied the regulatory compliance and environmental protection of the four types of on-site wastewater disposal systems found on the Mississippi Gulf Coast; i.e., vegetative rock filter, subsurface drip irrigation, sand mound, and sprinkler systems, by statistical techniques. Compliance was also evaluated for groundwater samples collected from monitoring wells installed at four corners of a disposal field. This work eventually culminated in formulation of strategy for modifying the aerobic treatment prior to disposal to help reduce nitrogen loading on the discharging environment. Process modeling and simulations were performed to optimize conditions for biological nitrogen reduction in the treatment unit by efficient management of aeration. Two separate proposals were developed, such as either running the aerator unit in a low operating dissolved oxygen concentration or intermittent aeration mode.

Application of HTML/VRML to Manufacturing Systems Engineering

Manufacturing systems are complex entities comprised of people, processes, products, information systems and data, material processing, handling, and storage systems. Because of this complexity, systems must be modeled using a variety of views and modeling formalisms. In order to design and analyze manufacturing systems, the multiple views and models often need to be considered simultaneously. However, no single tool or computing environment currently exists that allows this to be done in an efficient and intelligible manner. New tools such as HTML and VRML present a promising approach for tackling these problems. They make possible environments where the different models can coexist and where mapping/linking between the models can be achieved. This research is concerned with developing a hybrid HTML/VRML environment for manufacturing systems modeling and analysis. Experiment was performed to compare this hybrid-modeling HTML/VRML environment to the traditional database environment in order to answer typical design/analysis questions associated with manufacturing systems, and to establish the potential advantages of this approach. Analyzing results obtained from the experiment indicated that the HTML/VRML approach might result in better understanding of a manufacturing system than the traditional database approach. / Master of Science

Propomos métodos de resolução baseados em uma redução para o problema da satisfazibilidade probabilística (PSAT) e um método de redução de Turing ao ASP. / This dissertation introduces a technique called Probabilistic Answer Set Programming (PASP), that allows modeling complex theories and check its consistence with respect to a set of statistical data. We propose a method of resolution based in the reduction to the probabilistic satisfiability problem (PSAT) and a Turing reduction method to ASP.

Die vorliegende Dissertation präsentiert eine umfassende Analyse und Strukturierung des Marktes für netzbasierte Dienste. Basierend auf einer detaillierten Untersuchung des ASP-Angebotes und der ASP-Nachfrage in Europa werden geeignete Institutionen und Informationssubstitute für die Anpassung (Matching) des ASP-Angebotes und der ASP-Nachfrage identifiziert und getestet. Die Strukturierung der Angebotsseite umfasst neben der Bewertung der Marktlage die Entwicklung der ASP-Wertschöpfungskette, die Einordnung in die Systematik der digitalen Güter, die Beschreibung der ASP-Technologien und -Geschäftsmodelle und den Vergleich des ASP-Geschäftsmodells mit der klassischen Softwarenutzung und dem Konzept der virtuellen Organisationen. Die Ergebnisse der Untersuchungen zeigen vorhandene Defizite in der Informations- und Kommunikationspolitik der ASP-Angebotsseite auf. Für die Strukturierung der Nachfrageseite für netzbasierte Dienste werden die Theorien der Auslagerung (Outsourcing) von Informationstechnologien, die Methoden und Instrumente der Kosten- und Nutzenmessung und die Theorie der Transaktionsphasen der Auslagerung angewendet. Kern der wissenschaftlichen Untersuchung bildet die empirische Untersuchung des ASP-Nachfragemarktes in Deutschland und anderen europäischen Ländern. Die Erkenntnisse der Analyse des ASP-Angebotes und der ASP-Nachfrage werden gegenübergestellt und Methoden zur Abstimmung von ASP-Angebot und Nachfrage mit Hilfe der Transaktionskostentheorie und den Theorien der Informations- und Institutionenökonomie abgeleitet. Barrieren, Intransparenzen und Informationsasymmetrien werden auf beiden Seiten aufgezeigt und darauf aufbauend Informationssubstitute für netzbasierte Dienste identifiziert. Erfahrungs- und Vertrauenseigenschaften werden mit Hilfe von Informationssubstituten in einem Onlinebefragungsexperiment hinsichtlich der Wahrnehmung, Wirkung und möglicher Zahlungsbereitschaften überprüft. Aus den Ergebnissen des Onlinebefragungsexperimentes werden Anforderungen an die Informations- und Kommunikationspolitik und Strategien zur Reduzierung der Informationsasymmetrie für netzbasierte Dienste entwickelt. Das Ergebnis der Arbeit ist die Ableitung relevanter Kriterien für die Darstellung und Bewertung der Leistungsbündel netzbasierter Dienste und die Entwicklung von Strategien zur Reduzierung der Transaktionsunsicherheit durch die Identifizierung und Integration von Informationssubstituten für netzbasierte Dienste. / This dissertation presents a comprehensive analysis and conceptual framework of the web-based Application Service Provider (ASP) market. Based on a detailed analysis of ASP supply and demand in Europe, appropriate institutions and information substitutes for the matching process on the ASP market are identified and evaluated. The conceptual framework for the supply side is developed by evaluating the market situation, designing an ASP value chain, classifying digital goods, describing the ASP technologies and business models. These are then compared with the classic model of software utilization and the concept of virtual organizations. The results indicate present deficits in the supply-side information and communication policy. For ASP demand, theories of outsourcing of information technologies, methods and instruments of cost-and-utility-measuring and the theory of transaction phases of outsourcing are used. The basis for both supply-side and demand-side evaluations is an empirical analysis of the European ASP market. We compare the outcome of the supply and demand analyses and derive methods for their coordination by the transaction cost theory and the theories of information and institution economy. Barriers, lack of transparencies and information asymmetries are identified on both sides and thus, information substitutes for web-based services are developed. Information substitutes for experience and trust characteristics are evaluated with an online-questionnaire completed by key participants based on their perception, action and willingness to pay. From those results, we deduce strategies for reducing the information asymmetry. The result of the work is the deduction of relevant criteria for the presentation and evaluation for the supply of web-based services and the development of strategies for reducing the transaction uncertainty by identifying and integrating information substitutes.

Isolamento e caracterização funcional de uma fosfolipase A2 de Bothrops jararaca: avaliação do potencial antitumoral e inflamatório / Isolation and functional characterization of a phospholipase A2 from Bothrops jararaca snake venom: evaluation of its antitumor and inflammatory potential

As fosfolipases A2 (PLA2s) catalisam a hidrólise de ácidos graxos na posição sn-2 das membranas fosfolipídicas e liberam, como subprodutos, ácidos graxos livres. As PLA2s do grupo IIA são encontradas em peçonhas de serpentes da família Viperidae e desempenham diversas atividades apresentando potencial miotóxico, neurotóxico, hemolítico, edematogênico, citotóxico, hipotensivo, anticoagulante, inibição/ativação da agregação plaquetária, bactericida e pró-inflamatório. Esse trabalho teve como objetivo o isolamento e a caracterização funcional de uma PLA2 isolada da peçonha de Bothrops jararaca. Para a purificação dessa proteína, denominada BJ-PLA2-I, foram necessários três passos cromatográficos consecutivos: cromatografia de exclusão molecular em Sephacryl S-200, cromatografia de troca iônica em Source TM 15Q/50mL e cromatografia de troca iônica em MonoQ TM 5/50 GL. A BJ-PLA2-I apresentou elevado grau de pureza por SDS-PAGE e por cromatografia de fase reversa C18, em HPLC. Apresentou ainda, características ácidas, com pI em torno de 4,4 e teve a sua massa molecular determinada por dois métodos, obtendo-se valores bem próximos de 14,8 kDa (SDS-PAGE) e 14,2 kDa (MALDI-TOF). Esse fato é comum considerando que a espectrometria de massas é um método mais preciso e determina de maneira mais exata a massa molecular. O sequenciamento N-terminal da BJ-PLA2-I resultou em 60 resíduos de aminoácidos. O alinhamento múltiplo com outras fosfolipases A2 de serpentes do mesmo gênero mostrou similaridade entre elas, mostrando identidade de 100% com a BJ-PLA2, fosfolipase A2 Asp-49, também isolada da Bothrops jararaca. Esse dado levanta a hipótese de que a BJ-PLA2-I purificada neste trabalho e a BJ-PLA2 se tratam da mesma proteína, entretanto essa hipótese só poderá ser confirmada quando a sequência completa da BJ-PLA2-I for obtida. Outros dados encontrados neste trabalho reforçam essa hipótese, isso porque, avaliando a atividade fosfolipásica, o efeito sobre as plaquetas e o pI, tanto a BJ-PLA2-I quanto a BJ-PLA2 apresentaram características semelhantes. A BJ-PLA2-I, sendo uma Asp-49, mostrou alta atividade catalítica e efeito inibidor da agregação plaquetária induzida por ADP (20,5 ?g/mL inibiu 50 % da agregação plaquetária). Ela também foi capaz de induzir a migração leucocitária após a administração de diferentes concentrações (5, 10 e 20 ?g/mL) da BJ-PLA2-I. Esse dado também foi encontrado no ensaio em que a concentração de 10 ?g/mL foi fixada e variou-se o tempo de 2, 4 e 24 horas, observando-se principalmente a migração de neutrófilos. Além disso, verificou-se a liberação das citocinas IL-6 e IL-1?, de proteínas totais e de prostaglandina E2 na reação inflamatória induzida pela BJ-PLA2-I. No entanto, não foi observado a produção de TNF-?, IL-10 e leucotrieno B4. A BJ-PLA2-I caracterizou-se como uma PLA2 pró-inflamatória, produzindo inflamação local aguda. A BJ-PLA2-I foi avaliada quanto ao seu potencial antitumoral em três linhagens celulares distintas (PBMC, HL-60 e HepG2). Observou-se que a enzima em questão possui baixo potencial antitumoral para a linhagem HL-60, reduzindo o número de células tumorais em apenas cerca de 20% nas concentrações testadas. Verificou-se pequena alteração na viabilidade celular das células de PMBC, nas maiores concentrações testadas (160 e 80 ?g/mL) e, na linhagem HepG2 não foi encontrada nenhuma alteração. Concluindo, as informações adquiridas neste trabalho são de suma importância para a melhor compreensão dos mecanismos envolvidos nas atividades biológicas desempenhadas pelas PLA2s. Além disso, a BJ-PLA2-I pode servir como modelo molecular para a formulação de fármacos mais eficazes a serem utilizados no tratamento de várias doenças. / Phospholipases A2 (PLA2s) catalyze the hydrolysis of fatty acids in the sn-2 position of membrane phospholipids, releasing free fatty acids as by-products. PLA2s of group IIA are found in snake venoms of the Viperidae family and perform various activities, including myotoxic, neurotoxic, hemolytic, edematogenic, cytotoxic, hypotensive, anticoagulant, inhibition/activation of platelet aggregation, bactericidal and proinflammatory effects. This work aimed at the isolation and functional characterization of a PLA2 isolated from Bothrops jararaca venom. For the purification of this protein, called BJ-PLA2-I, three consecutive chromatographic steps were used (size exclusion chromatography on Sephacryl S-200, ion exchange chromatography on Source 15Q/50 mL, ion exchange chromatography on MonoQ 5/50 GL). Confirmation of the purity of BJ-PLA2-I was evaluated by SDS-PAGE and reverse phase HPLC using a C18 column. BJ-PLA2-I has acidic characteristics, with pI around 4.4, and its molecular mass was determined by two methods, obtaining values close to 14.8 kDa (SDS-PAGE) and 14.2 kDa (MALDI-TOF). The N-terminal sequencing of BJ-PLA2-I resulted in 60 amino acid residues. Multiple alignment with other phospholipases A2 of snakes of the same genus showed high similarity between them, showing 100% identity with BJ-PLA2, an Asp-49 phospholipase A2 previously isolated from Bothrops jararaca venom. This finding raises the possibility that the PLA2 purified in this work is the same protein previously described (BJ-PLA2), however, this assumption can only be confirmed when the complete sequence of BJ-PLA2-I is obtained. Other data obtained in this study support this hypothesis, considering that the phospholipase activity, the effect on platelets and pI of both BJ-PLA2-I and BJ-PLA2 showed to be similar. BJ-PLA2-I, being an Asp-49 PLA2, showed high catalytic activity and inhibitory effect on the platelet aggregation induced by ADP (20.5 ?g/mL inhibited 50% of the platelet aggregation). It was also able to induce leukocyte migration after the administration of different concentrations (5, 10 and 20 ?g/mL) of BJ-PLA2-I. This fact was also found when the concentration of 10 ?g/mL was fixed and response times were varied (2, 4 and 24 hours), observing especially neutrophil migration. Furthermore, there was a release of IL-6 and IL-1?, total proteins and prostaglandin E2 in the inflammatory reaction induced by BJ-PLA2-I, however, the production of TNF-?, IL-10 and leukotriene B4 was not observed. BJ-PLA2-I was characterized as a proinflammatory PLA2 producing acute local inflammation. BJ-PLA2-I was evaluated for its antitumor potential on three different cell lines (PBMC, HL-60 and HepG2). It was observed that this enzyme showed a low antitumor potential on HL-60 tumor cell line, reducing the number of tumor cells in only about 20% at the concentrations tested. There was little change in cell viability of PBMC cells in the higher concentrations tested (80 and 160 ?g/mL), but no change was found on HepG2 tumor cell line. In conclusion, the information obtained in this work are of utmost importance for better understanding the mechanisms involved in the biological activities induced by PLA2s. Furthermore, BJ-PLA2-I may serve as a molecular model for the formulation of more effective drugs to be used in the treatment of various diseases.
