Humoral and cell-mediated immunity in vitamin A-deficient lambs

Antigen-specific and polyclonal serum immunoglobulin G (IgG) concentrations were measured in control (Con), vitamin A-deficient (A-def), and vitamin A-repleted (A-rep) lambs. In Trial, I ewe lambs were injected with primary and secondary antigenic challenges of ovalbumin (1mg) and lysozyme (.1mg). The A-def lambs were then repleted with vitamin A and all lambs were injected with primary and secondary antigenic challenges of human gamma globulin (HGG) (.1mg). In Trial II Con and A-def wether lambs were given primary and secondary antigenic challenges of ovalbumin (20μg). Half of the A-def lambs were then repleted with vitamin A. All lambs were subsequently given a primary and secondary challenge of HGG (20 μg). Spleen wt were similar for all treatments in Trial I while A-def V lambs in Trial II had greater spleen wt (P<.01) than Con or A-rep lambs. Polyclonal serum IgG concentrations were unaffected by treatment in Trial I while in Trial II concentrations were greater (P<.05) in the A-def lambs during the HGG challenge period. Antigen-specific IgG concentrations in both trials tended to be greater in the Con lambs towards the end of both the ovalbumin (Trial I and II) and lysozyme (Trial I) challenge periods. Control and A-rep lambs in Trial I responded similarly to the HGG challenges. In Trial II both the A-def and A-rep lambs had lower (P<.10) HGG specific serum IgG concentrations on the last 3 wk of the HGG challenge period as compared to A-def lambs. Humoral immune function appears to be impaired in A-def lambs and a 2-wk repletion period was not sufficient in this study to restore humoral immune function to normal levels. / Ph. D.

Evaluating the efficacy of exogenous composite microbial enzymes in maize-soybean based broiler chicken feeds.

This research reported here was carried out to examine alternatives to antibiotic growth promoters as a result of their being banned in the animal feed industry. Four experiments were conducted to evaluate the efficacy of non-medicated feed additives as replacements for antibiotic growth promoters in broiler feeds. The additives used were enzymes (a new thermo-tolerant powder enzyme called TXAP, phytase, lipase and a new phytase enzyme derived from E. coli called phyzyme XP), organic acid (Acid Pak), prebiotic (Bio-Mos®) and probiotic (All-Lac XCL). Mashed maize-soya based feeds were used in all the experiments, which were conducted in litter-floor pens. The first experiment was a dose-response trial. Broilers in eight replicate pens of 50 males and 50 females were fed unsupplemented feeds and five additional feeds containing increasing levels of TXAP, from 0.5 to 2.5 g/kg to 42 d. The second experiment used enzyme TXAP with two different enzymes (phytase and lipase), individually or in combination. Six replicate pens of 50 males and 50 females were fed either unsupplemented feeds or one of six additional feeds treated with TXAP, lipase, phytase , a combination of TXAP and lipase, a combination of TXAP and phytase or a combination of all the three enzymes . This trial continued for 42 d. In the third experiment three types of TXAP (Lot 1, 2 and 3) were used, with fixed levels of xylanase and amylase but varying levels of protease activities (4000, 2000 and 1000 U/kg for Lot 1, 2 and 3, respectively) in combination with phyzyme XP for 35 d. The fourth experiment used mannan-oligosaccharide (Bio-Mos®), organic acid (Acid pak 2x), probiotic (All Lac XCL 5x), individually or in combination and an antibiotic growth promoter (Zinc bacitracin) for 42 d. The chickens in this experiment were challenged with Clostridium perfringens (CP) at 21, 22 and 23 d to determine the efficacy of these additives for replacing antibiotics in hindering the effects of CP on the villus surface area. The dose-response trial did not show any significant improvement in broiler performance with any level of inclusion of enzyme TXAP. The results from this study showed some beneficial effects with the use of enzyme TXAP when fed alone and at a young age. Its use when combined with other enzymes and at later stages of growth needs further investigation. Feed additives in experiment 4 prevented the negative effects of CP as the treated chickens did not have lesions on their villus surfaces. The conditions under which these trials were conducted appeared to be such that little benefit was derived from the use of any of the feed additives used. Curvas de crescimento de frangos caipiras submetidos a diferentes níveis de lisina digestível

The goal of the present work was to analyze the Gompertz functions in order to describe the growth of hillbilly broilers Color Plume (CPK) subjected to different levels of digestible lysine. On the experiment were used 432 chickens from one day old, was divided into three stages: initial, growth and finale respectively corresponding to the ages from 1 to 21, from 22 to 77 days of age and from 78 to 84 days of age. During the initial phase, the birds were reared on shavings beds with access to water, food and a heating source until after 21 days of age, the birds had access to paddocks. After 21 days of age, the birds which were in terms of the medium weight were distributed randomly in four treatments of 27 birds, with four replicates each one and 10 birds per replication (5 males and 5 females) were chipped with microchip brand Animall TAG and dimensions 12x2 mm. Treatments were represented by different lysine levels (0.850, 0.970, 1.090; 1.210%) on the experimental diet. The diets used were based on corn and bran soybean, and were formulated to meet the nutritional requirements at each stage of creation, as suggested by Rostagno et al. (2011), except lysine. Diets and birds were weighed weekly to determine the performance (weight gain, feed intake and feed conversion). Gompertz curves adjusted showed good correlation coefficients above 90%, and good values of Durbin Watson test accurately, estimating the average weight gain, and the consumption ration of mixed batch of free range broiler chickens. After the tests comparison between Gompertz parameters of the model for each level of lysine was determined that: the only parameter that varied was A, since B = 0.0329 and C = 45.819 parameters presented not significant differences. At the level of digestible lysine 0.850%, A = 4870.3, 0.970% A = 4894.9, 4784.3 A = 1.09%, 1.21% A = 4740. Among them, the parameter A in 0.970% level, differed from the others, ie, showing a higher body weight at maturity. Based the performance results, it is recommended for free range broiler CPK chickens of both sexes, to use a level of digestible lysine up to 0.970% on the period 22-77 days of age. After comparison tests between the Gompertz parameters of the model for each lysine level was determined for female that: the only parameter that varied was A, since B = 0.0356 and C = 45.54 parameters presented not significant differences. In males it has been concluded that only a single equation may represent the growth in four levels of lysine. The lysine levels did not influence (P> 0.05) carcass variables, except the yield of abdominal fat, than for females improved quadratically up to the level of 1.064%, for males there was a quadratic effect. / O presente trabalho objetivou-se utilizar o modelo não linear de Gompertz para descrever o crescimento de frangos caipira da linhagem Colorcpk Plumé (CPK), de ambos os sexos e entre os sexos, submetidos a diferentes níveis de lisina digestível. Foram utilizados 432 frangos com um dia de idade, o experimento foi dividido em três fases: inicial, crescimento e final correspondendo respectivamente às idades de 1 a 21, 22 a 77 e 78 a 84 dias de idade. Na fase inicial as aves foram criadas sobre cama de maravalha com acesso a água, ração e fonte de aquecimento e uma única dieta. Aos 21 dias de idade, as aves em função do peso médio foram distribuídas num delineamento inteiramente casualizado, em quatro tratamentos com quatro repetições de 27 aves por unidade experimental, e dentre elas, 10 aves de cada repetição (5 machos e 5 fêmeas) foram chipadas com microchips da marca Animall TAG e dimensões de 12x2 mm. Os tratamentos foram representados pelos diferentes níveis de lisina digestível (0,850; 0,970; 1,090; 1,210%) na dieta experimental na fase de crescimento. As dietas foram à base de milho e farelo de soja e formuladas para atender as exigências nutricionais em cada fase de criação, conforme sugerido de Rostagno et al. (2011), exceto lisina digestível. As rações e as aves foram pesadas semanalmente para determinação do desempenho (ganho de peso, consumo de ração e conversão alimentar), as aves chipadas também eram pesadas individualmente a cada 7 dias. Aos 84 dias de idade as aves chipadas foram abatidas, totalizando 160 aves. As características de carcaça avaliadas foram calculadas em relação ao peso corporal após jejum. As curvas de Gompertz ajustadas apresentaram bons coeficientes de determinação, acima de 90%, e bons valores do teste de Durbin Watson, estimando com boa acurácia o peso médio, ganho de peso e consumo de ração dos frangos caipira. Após os testes de comparação entre os parâmetros do modelo de Gompertz para cada nível de lisina para o lote misto, determinou-se que somente o parâmetro A deveria variar, pois os parâmetros B=0,0329 e C=45,819 não apresentaram diferenças significativas. No nível de lisina digestível 0,850% A=4870,3; 0,970% A=4894,9; 1,09% A=4784,3; 1,21% A=4740; dentre eles o parâmetro A no nível 0,970% diferiu dos demais, ou seja, maior peso corporal a maturidade. Com base nos resultados de desempenho, recomenda-se, para aves caipiras da linhagem CPK de ambos os sexos o nível de lisina digestível na ração de 0,970% no período de 22 a 77 dias de idade. Após os testes de comparação entre os parâmetros do modelo de Gompertz para cada nível de lisina, determinou-se para fêmeas que somente o parâmetro A deveria variar, pois os parâmetros B=0,0356 e C=42,54 não apresentaram diferenças significativas. Para os machos concluiu-se que apenas uma única equação pode representar o crescimento nos quatro níveis de lisina digestível. Os níveis de lisina digestível não influenciaram (P>0,05) as variáveis de carcaça, exceto o rendimento de gordura abdominal, que para as fêmeas melhorou de forma quadrática até o nível estimado de 1,064%.

Improving Pig Performance and Efficiency by Attenuating Transport and Immune Stress Responses through L-Tryptophan Supplementation

<p>  </p> <p>In commercial settings, piglet weaning and transportation occur concurrently due to the expansive application of multi-site production systems across the United States and the combination of these events can be defined as an early life stressor. Early life stress is known to reduce pig performance, efficiency, and immune resilience contributing to reduced welfare and increased production losses. To combat the deleterious effects of stress on pigs, the essential amino acid tryptophan (Trp) supplemented above current National Research Council (NRC 2012) recommended levels, improves neuroendocrine responses to stress, lowers plasma cortisol and norepinephrine concentrations, and improves hypothalamic-pituitary-adrenal axis recovery time following stress. However, there are discrepancies concerning the Trp requirement for nursery pigs, suggesting the 2012 recommendations for Trp may be inaccurate for optimizing growth performance and health in modern pigs. Three experiments were conducted to determine the effects of supplemented standard ileal digestible Trp above NRC (2012) recommended levels on performance, feed efficiency, immune vigor, and stress tolerance. The objective of experiment 1 was to eliminate or reduce short- and long-term, transport-induced reductions in piglet feed efficiency and growth by supplementing Trp above NRC (2012) recommendations pre-weaning and/or during the nursery phase. An oral gavage of Trp or a control milk carrier was provided to pre-allotted piglets beginning at day 5 of lactation and continuing to weaning. At weaning all pigs were blocked by sex, weaning weight and pre-wean treatment and randomly assigned to transport and post-wean treatments. Pigs were fed in four nursery phases with diets containing 1X or 2X NRC recommended concentrations of SID Trp and a common grower diet fed in 6 phases during the grow-finish period. At market, loin characteristics were measured via ultrasound and carcass data was collected from the packing plant. Pre-weaning Trp supplementation had no effect on pre-weaning growth performance; however, post-wean Trp increased overall body weight and average daily gain in nursery pigs when Trp was supplemented pre-wean. The objective for experiment 2 was to mitigate the adverse effects of early life transport stress on subsequent immune challenges by providing supplemental Trp during the nursery period. At weaning, pigs were transported for 8 hours and assigned to treatments of vaccine-induced immune challenge and dietary treatment. Pigs were fed standard nursery diets, in four phases, over 35 days with pigs receiving 1X or 2X the NRC (2012) recommended Trp concentration. Half the pigs on each dietary treatment were subjected to a 3-wk vaccine challenge consisting of circovirus, mycoplasma, and influenza vaccines administered in wk 2, 3, and 4 post-wean, respectively. At market, loin characteristics were measured via ultrasound and carcass data was collected.  At the culmination of the nursery period, unchallenged pigs supplemented with Trp were heavier compared to control pigs. This resulted from an overall improvement in average daily gain for 2X Trp fed pigs. However, increasing Trp did not affect market weight, loin eye area, or lean percent in market pigs, although 2X Trp increased back fat. The objective of experiment 3 was to determine what ratio of SID tryptophan, relative to lysine, maximizes growth performance and feed efficiency in weaned pigs during the nursery period. Pigs were blocked by sex and weaning weight and randomly allotted to one of five dietary treatments with Trp increasing stepwise by 0.33%-units, respectively. Pigs were fed standard nursery diets, in four phases, over 35 days with pigs receiving 1X, 1.33X, 1.66X, 2X, or 2.33X the NRC (2012) recommended Trp concentration.  Overall, there were no differences in performance or efficiency across dietary treatments during a 35-day nursery period in unstressed healthy pigs.  In conclusion, increasing Trp in swine diets prior to and/or following stress events can mitigate stress-related perturbations in performance and efficiency.      </p>

The effect of extrusion on the degradability parameters of various vegetable protein sources

Thesis (MScAgric)--University of Stellenbosch, 2004. / ENGLISH ABSTRACT: The objective of this study was to determine the effect of extrusion, as a method of heat treatment, on the dry matter (DM) and crude protein (CP) degradability parameters of various vegetable protein sources commonly used in the Western Cape Province, South Africa. The feedstuffs used were lupins (LUP), full fat soybeans (SB), full fat canola seeds (FCS), soybean meal (SBM), canola meal (CM) and sunflower meal (SFM). In the first trial, the degradability parameters were determined according to an in sacco degradability procedure. Four non-lactating Holstein cows, fitted with rumen cannulae, were used in the trial and all cows received the same basal lactation diet. The samples were incubated in dacron bags and bags were removed at intervals of 0, 2, 4, 8, 16, 24 and 48 hours. Dry matter and CP disappearance values were determined and fitted to a onecompartment model by means of an iterative least-square procedure in order to determine the DM and CP degradability parameters. Results indicated that extrusion significantly lowered the effective degradability of the DM-fraction (20.1% on average) of all the feedstuffs, except LUP, and the effective degradability of CP in all the raw materials (27% on average). The second trial was an in vitro degradability trial that ran parallel with the in sacco degradability trial and was done with the aid of a DaisyII Incubator (ANKOM Technology Corp., Fairport, NY). The same feedstuffs were tested in both trials. A composited sample of rumen liquor from two of the cows used in the in sacco trial was used for in vitro incubation of the samples. The data obtained in this trial were analyzed in a similar way to that of the in sacco trial. Due to a limited amount of residue left after incubation, CP disappearance could not be calculated at each time interval for SB and SBM in the in vitro trial. In this case, actual disappearance values after 8h were used to compare treatments. Extrusion significantly lowered the effective degradability (as determined in vitro) of DM in all the feedstuffs tested (16.8% on average), as well as the effective degradability of CP in LUP, FCS, CM and SFM (21.8% on average). A comparison of the actual disappearance values after 8 hours incubation indicated that extrusion also lowered the rate of CP disappearance for SB and SBM. The values obtained in the in vitro trial and those from the in sacco trial, for the same feedstuffs, were compared. It appeared as if the in vitro determined values were overestimations of the in sacco determined values. A regression analysis showed a high correlation between the actual in vitro CP disappearance values after 8h incubation and in sacco determined effective degradability values. The third part of this study was a set of chemical analysis to determine the effect of extrusion on certain nitrogen fractions of the feedstuffs tested in the above mentioned trials. Solubility in a mineral buffer solution was determined to estimate the potential rumen degradability of the protein. The buffer insoluble nitrogen (BIN) fraction of all the feedstuffs, except FCS, was significantly increased by extrusion. Extrusion lowered the acid detergent insoluble nitrogen (ADIN) content of all feedstuffs, except FCS, which could imply that the temperature reached during extrusion (115°C - 120°C) was not high enough to cause damage to the protein. The neutral detergent insoluble nitrogen (NDIN) fraction of extruded SB, SBM, CM and SFM was significantly higher than that of the raw feedstuffs. Extrusion left the NDIN-fraction of FCS and LUP unaltered. Comparison of the NDIN : ADIN ratio of extruded with that of the raw feedstuffs provided reason to believe that extrusion had a positive effect on all feedstuffs (except FCS). Extrusion appears to be a useful method to decrease rumen degradation of vegetable protein sources, without causing heat damage. Furthermore, this means that protein sources of which the use have been limited due to its high rumen degradable protein (RDP) content, could be included in diets at higher levels following extrusion. The protein sources mentioned are also good sources of energy and the combination of energy and rumen undegradable protein (RUP) in the diet of the high-producing dairy cow could only be beneficial. / AFRIKAANSE OPSOMMING: Die doel van hierdie studie was om die effek van ekstrusie op die droëmateriaal (DM) en ruproteïen (RP)-degradeerbaarheidsparameters van verskeie plantaardige proteïenbronne wat algemeen in die Wes-Kaap (RSA), gebruik word, te bepaal. Ekstrusie is ‘n metode van hitteprosessering wat algemeen gebruik word deur plaaslike en internasionale veevoervervaardigers. Die volgende grondstowwe is geëvalueer: lupiene, volvet sojabone, volvet canolasaad, sojaboon-oliekoekmeel, canola-oliekoekmeel en sonneblom-oliekoekmeel. In die eerste proef is die degradeerbaarheidsparameters met behulp van ‘n in sacco studie bepaal. Vier droë Holstein koeie met rumen kannulas is in die studie gebruik en al vier koeie het dieselfde basale dieet ontvang. Monsters is in dacronsakkies geïnkubeer en die sakkies is uit die rumen verwyder na onderskeidelik 0, 2, 4, 8, 16, 24 en 48 uur intervalle. Die waardes van DM- en RP- verdwyning is bereken en dan met ‘n iteratiewe kleinste kwadraat prosedure op ‘n een-kompartement model gepas om die in sacco DM- en RPdegradeerbaarheidsparameters te bepaal. Die resultate van die studie het getoon dat ekstrusie die effektiewe degradeerbaarheid van die DM-fraksie van al die grondstowwe, behalwe lupiene, betekenisvol verlaag het (met gemiddeld 20.1%), asook die effektiewe degradeerbaarheid van die RP-fraksie van al die grondstowwe (met gemiddeld 27%). Die tweede proef was ‘n in vitro-degradeerbaarheidsstudie wat met behulp van ‘n ANKOM DaisyII Inkubeerder uitgevoer is en wat parallel met die in sacco-studie gedoen is. Die in sacco- en in vitro-bepaalde waardes vir elke grondstof is vergelyk en dit kom voor asof die in vitro-waardes oorskattings van die in sacco-waardes is. ‘n Regressie-analise het verder getoon dat daar ‘n hoë korrelasie was tussen die waargenome in vitro RPverdwyningswaardes na 8 ure inkubasie en die beraamde effektiewe degradeerbaarheid, soos in sacco bepaal. Die derde deel van die studie was ‘n stel chemiese analises wat uitgevoer is om die effek van ekstrusie op sekere stikstof (N)-fraksies van die grondstowwe, wat in bogenoemde proewe gebruik is, te bepaal. Die oplosbaarheid van N in ‘n mineraal-bufferoplossing kan gebruik word as aanduiding van die potensiële rumendegradeerbaarheid van die proteïen. Die bufferonoplosbare N-fraksie van al die grondstowwe (behalwe volvet canolasaad) is betekenisvol verlaag deur ekstrusie. Ekstrusie het ook die suur-onoplosbare N-fraksie (ADIN) van al die grondstowwe (behalwe volvet canolasaad) betekenisvol verlaag. Dit kan moontlik daarop dui dat die temperatuur wat tydens ekstrusie (115°C - 120°C) bereik is, nie hoog genoeg was om die proteïen in die grondstowwe te beskadig nie. Ekstrusie het die N-fraksie wat onoplosbaar was in ‘n neutrale oplossing (NDIN) betekenisvol verhoog in volvet sojabone, sojaboonoliekoekmeel, canola-oliekoekmeel en sonneblom-oliekoekmeel en dit onveranderd gelaat in lupiene en volvet canolasaad). Die verhouding van NDIN : ADIN van die geëkstrueerde grondstowwe is vergelyk met dié van die rou grondstowwe. Dit blyk dat ekstrusie wel ‘n positiewe effek op al die grondstowwe (behalwe volvet canolasaad) gehad het. Dit wil dus voorkom asof ekstrusie wel aangewend kan word om die rumendegradeerbaarheid van plantaardige proteïenbronne te verlaag sonder om die protein te beskadig. Dit kan daartoe lei dat proteïenbronne waarvan die gebruik voorheen beperk was as gevolg van die hoë rumen-degradeerbare proteïen-inhoud daarvan nou wel in rantsoene ingesluit kan word na die ekstrusie daarvan. Die proteïenbronne, soos genoem, is ook redelike bronne van energie en die kombinasie van energie en rumen nie-degradeerbare proteïen in die rantsoen van die hoog-produserende melkkoei kan slegs voordelig wees.

Chemo-attractants or chemo-stimulants is therefore generally included in feeds for especially slow-feeding species to help reduce water fouling and to promote feed efficiency and growth rate through improved feed intake. Considering this, a study was performed to evaluate the attraction and stimulation potential of selected cereals and free amino acids in diets for Japanese koi carp (Cyprinus carpio). Results are presented on the comparative evaluation of five cereals (maize, sorghum, wheat, rye and triticale), raw and cooked forms of maize and concentrations of betaine and selected free amino acids (alanine, arginine, lysine and methionine), as well as their additive effect.

Lysine, Threonine, Methionine, Isoleucine, Phenylalanine and Leucine were determined as the limiting amino acids for nursing lambs. These amino acids were incorporated into the creep feed treatments CF1 and CF2 representing 157 g CP/kg, 477 g NSC/kg and 179 g CP/kg, 508 g NSC/kg, respectively. A commercial creep feed with no optimisation for EAA is represented as CFC with 139 g CP/kg and 455 g NSC/kg. A negative control (CON) treatment represents lambs receiving no creep feed but with ad libitum access to suckle their dams while feeding on kikuyu pasture. A growth trial was conducted on Merino x Döhne-Merino cross twin lambs averaging a birth weight of 4.42 kg ± 0.11 for 60 days following with a digestibility trial towards the end of the trial. Half of the lamb crop was slaughtered at an average live weight of 23.6 kg ± 0.56. The M. longissimus dorsi was removed on both the left and right half of the carcass between the 2nd - 3rd last thoracic vertebrae and the 4th - 5th lumbar vertebrae. A sample from the rumen wall was taken at the rumino-reticular fold to determine development characteristics. Results indicate that the feed conversion ratio for the CFC lambs were better than CF1 (P = 0.052) but not more than CF2 (P = 0.307). The FCR was 0.88, 1.19 and 1.01 (kilogram feed required to gain 1 kg in bodyweight) for CFC, CF1 and CF2, respectively. Dressing percentage was higher for CF2 than for both CFC (P = 0.012) and CF1 (P = 0.077). Along with BUN data it was concluded that the high CP level of CF2 resulted in the higher fat deposition. The optimised creep feeds had higher nitrogen and energy balances than the commercial CFC, this implicates that the optimised creep feeds were more efficiently utilised. Optimised creep feed treatments had longer papillae than CON (P < 0.0001). Papillae of creep feed 2 was longer than CFC (P = 0.0537). Papillae width decreased as the NSC level increased thus it is surmised that higher NSC levels resulted in longer but thinner papillae. Rumen muscularization was equally developed between all the treatments and was ascribed to the lambs’ access to course roughage. Meat quality in terms of physical and chemical characteristics was found to be in range with that expected for lambs at higher slaughter weights (40 kg). In conclusion, the balancing of the limiting EAA increases the benefits of creep feeding while simultaneously being more efficiently utilised. Intensive sheep production systems may benefit from the feeding of such creep feeds provided it fits economically into their farming system. / AFRIKAANSE OPSOMMING: Die volgende studie is om te bepaal of 'n kruipvoer wat gebalanseerd is vir beperkende essensiële aminosure (EAA) by 'n vasgestelde nie-strukturele koolhidraat (NSK) vlak verbeterde groei sal ontlok teenoor lammers wat grootgemaak word op 'n kommersiële kruipvoer. Die VOV was 0,88 , 1,19 en 1,01 (kilogram voer wat nodig is om 1 kg liggaamsmassa aan te sit) vir CFC, CF1 en CF2 onderskeidelik. Uitslag persentasie vir CF2 was hoër as beide CFC (P = 0,012) en CF1 (P = 0,077). Saam met die bloed, urea en stikstofbalans data is daar tot die gevolgtrekking gekom dat die hoë RP inhoud van CF2 gelei het tot ‘n hoër vetneerlegging. Die geoptimaliseerde kruipvoere het ‘n hoër stikstof- en energiebalans gehad teenoor die kommersiële CFC behandeling wat dus impliseer dat die optimale kruipvoere doeltreffender benut was. Papillae breedte het dunner geraak soos wat die NSK-vlak gestyg het dus word vermoed dat hoër NSK-vlakke langer maar dunner papillae tot gevolg het. Rumen bespiering is ewe ontwikkeld tussen al die behandelings en word toegeskryf aan die lammers se toegang tot growwe ruvoer. Vleis kwaliteit in terme van fisiese- en chemiese eienskappe was inlyn met wat verwag kan word vir lammers by hoër slaggewigte (40 kg). Ten slotte, die balansering van beperkende aminosure verhoog die voordele van kruipvoeding en word terselfdertyd doeltreffender benut. Intensiewe skaap produksie stelsels kan dus voordeel trek uit die voeding van sodanige kruipvoere mits dit ekonomies pas in die boerdery stelsel.

The evaluation of larvae of Musca domestica (common house fly) as protein source for broiler production

This was done by investigating the nutritive value of house fly larvae meal together with its total tract digestibility, potential toxicity and carcass characteristics of the broilers supplemented with house fly larvae meal. The proximate analysis of house fly larvae meal show that it contained, on a dry matter basis, a gross energy value of 20.10 MJ/kg, 60.38% crude protein, 14.08% crude fat and 10.68% ash and that the house fly pupae contained a gross energy of 20.42 MJ/kg, 76.23% crude protein, 14.39% crude fat and 7.73% ash. House fly pupae meal had the closest match of amino acid profile when compared with the ideal amino acid profile required by broilers and it has arginine relative to the lysine content closer to the ideal amino acid profile than the house fly larvae meal. The essential fatty acid, linoleic acid, was found at levels of 26.25 and 36.27% of the total fats for the house fly larvae and pupae meal respectively. House fly larvae meal supplementation did not induce gizzard erosion or showed toxicity (regarding the gastro intestinal tract, immune system and organ stress) in broilers. Results revealed that house fly pupae meal had higher total tract digestibilities for most nutrients than of the house fly larvae meal. House fly larvae meal had a crude protein total tract digestibility of 69% and that of pupae meal was 79%. Both larvae and pupae meal had high amino acid total tract digestibilities of all the amino acids analysed. The house fly larvae and pupae meal had an apparent metabolizable energy (AME) value of 14.23MJ/kg and 15.15MJ/kg respectively. The larvae meal total tract crude fat and crude fibre digestibilities were 94% and 62% respectively. The pupae meal total tract crude fat and crude fibre digestibilities were 98% and 58% respectively. House fly larvae meal supplementation in a three phase feeding system significantly increased average broiler live weights at slaughter, total feed intake, cumulative feed intake as well as average daily gain (ADG) when compared to commercial maize: soya oil cake meal diet. In direct comparison of larvae inclusion levels with fishmeal in isonitrogenous and isoenergetic diet, no significant differences were observed between a 10% house fly larvae and a 10% fish meal diets regarding performance characteristic. The 25% house fly larvae meal diet yielded significantly better average broiler live weights at slaughter, total feed intake, cumulative feed intake (from the second week until slaughter) as well as average daily gain when compared to the 25% fish meal diet in the growth phases. Carcass characteristics of the 10% larvae, 10% fishmeal and commercial diets were compared. Chicks that received either the 10% house fly larvae meal or 10% fish meal supplementation produced significantly heavier carcasses and breast muscle portions than the chicks that received the commercial maize: soya oil cake meal. No treatment differences were found regarding breast and thigh muscle colour or pH. This study showed that house fly larvae meal can be regarded as a safe protein source that can be used to replace other protein sources and that has the ability to promote broiler performance without having any detrimental effects on carcass characteristics. / AFRIKAANSE OPSOMMING: Die evaluasie van Musca domestica (gewone huisvlieg) larwe meel as ‘n proteien bron vir braaikuiken produksie Die doel van die studie was om die effek van Musca domestica (gewone huisvlieg) larwe meel, as ‘n protein bron, in braaikuikens te evalueer. Dit was gedoen deur die nutrient waarde van huisvlieg larwe meel saam met die totale spysvertering verteerbaarheid, moontlike toksiesiteit en karkas-eienskappe van braai kuikens te evalueer. Laboratoruim analiese toon dat huisvlieg larwe meel 20.10 MJ/kg bruto energie, 60.38% ru- protein, 14.08% ru- vet en 10.68% as bevat en huisvlieg papie meel 20.42 MJ/kg bruto energie, 76.23% ru- protein, 14.39% ru- vet en 7.73% as bevat. Huisvlieg papie meel stem die meeste ooreen met die idiale amino suur profiel soos wat benodig word deur braaikuikens en dit het ‘n arginien tot lisien verhouding wat die meeste ooreenstem met die idiale amino suur profiel in vergelyking met huis vlieg larwe meel. Die 25% larwe meel dieet het merkwaardig beter gemiddelde braaikuiken lewende gewigte by slag, totale voer iname, sowel as die gemiddelde daaglikse toename getoon wanneer vergelyk word met die 25% vismeel dieet gedurende die verskeie groei fases. Karkas eienskappe van die 10% larwe meel, 10% vismeel en die kommersiele diete was gevergelyk. Kuikens wat 10% larwe meel en 10% vismeel in die diete ontvang het, het swaarder karkasse gelewer met swaarder borsie massas wanneer vergelyk word met die kommersiele mielie- soya olie koek dieet. Geen behandelings verskille was gevind rakende die borsie- en dy spier kleure of pH nie. Die studie toon dat huisvlieg larwe meel as ‘n veillige protein bron kan beskou word, wat gebruik kan word om ander protein bronne te vervang. Huisvlieg larwe meel het ook die vermoë om braaikuiken produksie te verhoog sonder om enige negitiewe effekte rakende die karkas eienskappe te toon nie.

The effect of different dietary levels of energy and protein on the production and body composition of broiler breeders

Thesis (MScAgric)--University of Stellenbosch, 2000 / ENGLISH ABSTRACT: A flock of 500 Hybro broiler breeders were employed to study the effect of different levels of protein and energy on production and body composition. The daily lysine intake of the birds were 900, 1050, 1200 and 1350 mg respectively, each fed in diets with a daily energy intake of 1800 and 2000 kJ ME to provide a 4 x 2 factorial design. Lysine was used as the reference amino acid in the experiment and all other amino acids were kept in a constant ratio in every experimental diet. The total production was divided into 3 periods of 13 weeks each (week 23 - 35; week 36 - 48 and week 49 - 61) to determine the effect of the treatments over time. Production was evaluated by hen day production; egg weight (g/egg); egg mass (g/day); fertility; hatchability; chicks/hen/week; feed conversion and day old chick weight. Hen day production was significantly (P < 0,05) lower at the high energy intake for period week 49 to 61. Energy and protein levels significantly increased egg weight. Effect of protein was consistent during all three periods of production. Egg mass output had a significant (P < 0,05) response to increasing levels of protein for the total period of production. The birds on the high energy diet produced a significant higher egg mass per hen during the first period of production (week 23 - 35). Hatchability was reduced (P < 0,05) by the higher energy intake for the total period of production and this effect was very significant (P < 0,01) during the final period of production. Similar to hen day production, the higher energy had a significant (P < 0,05) negative effect on the amount of chicks produced. The well-known correlation between egg weight and chick weight was confirmed with the regression equation: Chick weight = 10,5 + 1,22 x Egg weight. Both energy and protein had a significant (P < 0,01) positive effect on chick weight. The feed conversion was lower at increasing levels of amino acid intake (P < 0,01). Higher energy intake significantly (P < 0,05) increased hen weight and protein had a very significant effect during the first period of production (P < 0,01). The isotope dilution technique (tritiated water) was used to estimate the body composition of the breeders. This was done at 5 different periods (week 27, 35, 43, 52 and 61) of the production period to establish changes in requirements over time. Significant correlation were found between carcass moisture and waterspace (R2 = 0,76); fat % and carcass moisture (R2 = 0,78); protein % and waterspace (R2 = 0,35) and fat % and waterspace (R2 = 0,46). The regression equations obtained from these correlation were employed to determine excess energy and lysine consumed at different ages. Energy requirements were calculated according to the effective energy (EE) system and the conventional ME system. According to effective energy the lower energy intake was not sufficient and the maximum requirement was calculated to be 1942 kj EE/day. The metabolizable energy calculations indicated sufficient intakes at every treatment with the maximum requirement 1746 kj ME/day. The lysine requirement was found to be in excess of 1050 mg lysine per day. The recommendation for daily lysine intake is 1200 mg/day and the energy intake of breeders should be 1900 to 2000 kj ME/day from week 23 to 35 and can be reduced to 1800 kj ME/day in the final period of production. / AFRIKAANSE OPSOMMING: Om die effek van verskillende vlakke energie en proteïen op produksie en liggaamsamestelling te ondersoek is 'n trop van 500 Hybro braaikuiken teelouers gebruik. Die daaglikse lisien inname van die henne was onderskeidelik 900, 1050, 1200 en 1350 mg, wat elk by 'n daaglikse energie inname van 1800 en 2000 kJ ME verskaf is, in 'n 4 x 2 faktoriale ontwerp. Alle aminosure is in 'n konstante verhouding met die verwysings aminosuur lisien in die rantsoen ingesluit. Voeromset was laer by stygende vlakke van proteïen inname (P < 0,01). Die hoër energie inname het henmassa ook betekenisvol (P < 0,05) verhoog en proteïen het dieselfde effek slegs in die eerste periode van produksie gehad (P < 0,01). Die liggaamsamestelling van teelhenne is bepaal met behulp van die isotoop verdunnings tegniek (tritium water). Dit is bepaal op 5 verskillende stadiums in die produksie periode (week 27, 35, 43, 52 en 61) om die moontlike verandering in behoeftes oor tyd vas te stel. Betekenisvolle korrelasies is gevind tussen karkasvog en die waterspasie (RZ= 0,76); vet % en karkasvog (R2 = 0,78); proteïen % en waterspasie (R2 = 0,35) en vet % en waterspasie (R2 = 0,46). Die regressie vergelykings van hierdie korrelasies is gebruik om die hoevelheid surplus energie en lisien ingeneem te bepaal. Die" effective energy" sisteem en die konvensionele ME sisteem is gebruik in die bepaling van energie behoeftes. Volgens die " effective energy" was die energie inname nie voldoende by die laer energie innames nie en die maksimum behoefte is bereken as 1942 kJ EE/dag. Die maksimum energie behoefte volgens metaboliseerbare energie is bereken as 1746 kj ME/dag. Die lisien behoefte moet hoër as 1050 mg/dag wees. Die aanbeveling van die daaglikse lisien inname is 1200 mg/dag en die energie inname moet 1900 tot 2000 kj/dag wees tot 35 weke produksie en kan daarna verlaag word na 1800 kJ ME/dag.

The influence of processing of soyabeans and sunflower seed on their energy and amino acid availability for poultry

The effect of heat-treatment and dehulling of full-fat soya (Glycine max) on AMEn' amino acid availability and broiler performance. Heat treatment and dehulling of full-fat soya were evaluated in terms of their effect on AMEn, apparent amino acid availability and broiler performance. Four products were tested: raw whole soya (RWS), raw dehulled soya (RDS), extruded whole soya (EWS) and extruded dehulled soya (EDS). Trials were conducted to determine AMEn values and apparent amino acid availability of the products. A 42-day broiler trial was conducted to measure the effect of extrusion and dehulling on broiler performance. Test products were added to the diets at levels up to 263.2 g/kg in the starter diet and 260.0 g/kg in the finisher diet. Results indicated that extrusion cooking improved AMEn and apparent amino acid availability. This was reflected in the improved performance of broilers fed extruded soya as compared to raw soya. Increasing amounts of RWS and RDS led to decreases in mass and intake and poor feed conversion ratio (FeR) due to the presence of anti-nutritional factors. Broilers fed EWS were heavier, consumed more feed and had better FeR than those fed RWS or RDS. Dehulling improved AMEn (EWS: 13.75 MJ/kg vs EDS: 15.09 MJ/kg) and rendered a product with slightly higher levels of total amino acids. Broiler performance did not reflect this as there was no difference in mass, intake or FeR between broilers fed increasing levels of EWS or EDS. Lysine and arginine were less available in EDS than EWS indicating the possible over-cooking of EDS. Levels of EWS and EDS of 263.2 g/kg in the starter and 260.0 g/kg in the finisher supported maximum broiler performance. 2. The effect of heat-treatment and dehulling of full-fat sunflower (Helianthus annuus) on AMEm amino acid availability and broiler performance. Heat treatment and dehulling of full-fat sunflower were evaluated in terms of their effect on AMEn, apparent amino acid availability and broiler performance. Four products were tested: raw whole sunflower (RWSF), raw dehulled sunflower (RDSF), extruded whole sunflower (EWSF) and extruded dehulled sunflower (ED SF). Trials were conducted to determine AMEn values and apparent amino acid availability of the products. A 42-day broiler trial was conducted to measure the effect of expansion and dehulling on broiler performance. Test products were added to the diets at levels up to 115.0 glkg in the starter diet and 181.5 glkg in the finisher diet. Dehulling rendered a product with higher crude protein, ether extract, amino acid and AMEn values. AMEn was only slightly improved by expansion. The AMEn values obtained were: RWSF: 16.03 MJ/kg, RDSF: 18.87 MJ/kg, EWSF: 16.22 MJ/kg, EDSF: 19.49 MJ/kg. Dehulling had no effect on apparent amino acid availability. Expansion did not affect apparent amino acid availability of dehulled full-fat sunflower but had a negative influence on apparent amino acid availability of whole sunflower seeds. This highlights the possible dangers of reduced protein quality as a result of overprocessing. No differences were observed in terms of mass, intake and feed conversion ratio of broilers fed any of the products during the 42-day broiler growth trial. All products supported optimum broiler performance at levels up to 115.0 glkg in the starter diet. For the finisher diets, optimum performance was maintained at levels of 181.5 glkg, for RDSF and EDSF, while performance of broilers fed RWSF and EWSF was optimal up to 145.2 glkg. / AFRIKAANSE OPSOMMING: 1. Die invloed van hitte-behandeling en ontdopping van volvetsojabone (Glycine max) op SMEm skynbare aminosuur beskikbaarheid en braaikuikenprestasie. Hitte-behandeling en ontdopping van volvetsojabone is ge-evalueer in terme van hul invloed op stikstof gekorrekteerde skynbare metaboliseerbare energie (SMEn) waardes, skynbare aminosuur beskikbaarheid en braaikuikenprestasie. Vier produkte is getoets: rou heel sojabone (RWS), rou ontdopte sojabone (RDS), geekstrueerde heel sojabone (EWS) en geekstrueerde ontdopte sojabone (EDS). Proewe is uitgevoer om die SMEn waardes en skynbare aminosuur beskikbaarheid van die vier produkte te bepaal. Gedurende 'n braaikuikengroeitoets van 42 dae is die invloed van hittebehandeling (ekstrusie) en ontdopping op braaikuikenprestasie gemeet. Die vier toetsprodukte is ingesluit in die aanvangsdieet teen peile van tot 263.2 g/kg en teen peile van tot 260.0 g/kg in die afrondingsdieet. Resultate het getoon dat ekstrusie die SMEn waardes en skynbare aminosuur beskikbaarheid verbeter het. Braaikuikens wat geekstrueerde soja ontvang het, het beter gepresteer as braaikuikens wat rou soja ontvang het. Verhoogte insluitings van RWS en RDS het gelei tot verlagings in massa toename en voerinname en swak voeromsetverhoudings (VOV), as gevolg van die teenwoordigheid van anti-voedings faktore. Braaikuikens wat EWS ontvang het, was swaarder, het meer ingeneem en het beter vav gehad as die wat RWS of RDS ontvang het. Ontdopping het SMEn verhoog (EWS: 13.75 MJ/kg vs EDS: 15.09 MJ/kg) en ontdopte produkte het klein hoeveelhede meer aminosure bevat. Dit het egter nie in braaikuikenprestasie gewys nie. Skynbare aminosuur beskikbaarheid van ontdopte sonneblomsaad IS me deur hitte-behandeling beinvloed nie, terwyl die van heel sonneblomsaad negatief beinvloed is. Dit beklemtoon die gevare van verswakte proteien kwaliteit as gevolg van oor-prosessering.
