La machinerie de biosynthèse de la cellulose : une cible pour améliorer l’utilisation de la biomasse végétale / The cellulose synthase machinery as a target to improve biomass use

La production de biocarburants de deuxième génération basée sur la transformation de la biomasse végétale est une question d’actualité. La biomasse végétale est représentée par les parois des cellules, qui consistent en un réseau de microfibrilles de cellulose et de polysaccharides enchâssés dans de la lignine. Pour exploiter pleinement le potentiel de cette biomasse, il est nécessaire d’apporter des connaissances complémentaires sur les mécanismes de biosynthèse de ces polymères pariétaux. Par exemple, il est important d’améliorer le rendement de saccharification des microfibrilles de cellulose afin de produire de plus grandes quantités de bioéthanol. Nous avons donc combiné des études basées sur le modèle bien connu Arabidopsis et sur Brachypodium distachyon, la nouvelle espèce modèle pour les graminées tempérées et les céréales monocotylédones dédiées à la production de biocarburants. La cellulose est synthétisée par des complexes membranaires de cellulose synthases (CSC) qui contiennent les sous-unités catalytiques de cellulose synthase (CESAs), et cela requiert d’autres partenaires parmi lesquels KOR1, une endo-β-1,4-glucanase. Le trafic intracellulaire des CESAs semble jouer un rôle crucial dans la régulation du niveau de la synthèse de la cellulose. Nous avons étudié en détails le trafic intracellulaire de KOR1 dans des hypocotyles d’Arabidopsis cultivés à l’obscurité. En parallèle, lors d’un crible visuel de la collection de mutants de Brachypodium de l’INRA de Versailles, nous avons sélectionné un mutant nommé spa. Ce mutant partage des caractéristiques avec les mutants brittle culm du riz et de l’orge, comme par exemple des tiges cassantes, un xylème irrégulier, et une importante déficience en cellulose, surtout au niveau des tiges, qui contiennent 50% de la quantité retrouvée dans une plante sauvage. Des dosages de lignine ont montré une augmentation significative chez spa. De façon itnéressante, ce mutant présente un défaut flagrant du port érigé, au contraire des mutants brittle culm qui sont parfaitement érigés. Les défauts mécaniques du mutant spa s’illustrent par un module de Young trois fois inférieur à celui d’une plante sauvage. Des approches complémentaires ont été mise en œuvre afin d’identifier les défauts génétiques responsables de ce phénotype : le séquençage de gènes candidats reliés à la synthèse de la cellulose a été réalisé ainsi qu’une approche de NGS. De plus, dans le cadre du projet Européen RENEWALL et du projet KBBE CellWall, et grâce à l’outil BradiNet (M.Mutvil, KBBE CellWall) permettant d’accéder aux réseaux de co-expression, des stratégies RNAi sont en cours afin d’inactiver certains gènes seléctionnés selon des critères d’expression spécifiques, et selon leur implication potentielle dans la synthèse de la cellulose, spécifiquement chez les monocotylédones. Parmi ces gènes nous nous sommes concentrés sur la famille des MAP65 (Microtubules Associated Proteins), qui pourraient, au vu de la relation étroite entre microtubules et microfibrilles, jouer un rôle dans la déposition de la cellulose. / The production of second-generation biofuels based on the transformation of plant biomass is a pressing issue. Biomass is represented by cell walls of the plant cells consisting of a network of cellulose microfibrils and polysaccharides encrusted by lignin. To enhance the potential of plant biomass, we need to provide insights on the mechanisms of the biosynthesis of cell wall polymers. For example, it is important to improve the saccharification yield of cellulose microfibrils to produce the highest amount of bioethanol. We therefore combine studies on the well-known model plant Arabidopsis and Brachypodium distachyon, the new model species for temperate graminae and monocotyledonous crops dedicated to biofuel production. Cellulose is synthesized by plasma membrane-bound cellulose synthase complexes (CSC) containing cellulose synthase proteins (CESAs) and requires other partners among which the endo-beta1,4 glucanase KOR1. The intracellular trafficking of CESAs seems to be crucial to regulate the cellulose synthesis rate. We investigated in detail the intracellular trafficking of KOR1 in Arabidospis dark-grown hypocotyls.In parallel we selected by visual screening of the Versailles collection of mutagenized Brachypodium distachyon a mutant called spa. This mutant shares characteristics of the brittle culm mutants of rice and barley, such as brittleness, irregular xylem, and a cellulose content deficiency especially in stems, with 50% of the amount found in the wild type. Lignin assays indicate a higher amount of lignin in spa. Interestingly, this mutant is also "floppy" unlike others brittle culm mutants which are fully erected and the mechanical strength defects of spa is illustrated by a Young’s modulus three times lower than that of WT. Complementary approaches were used to identify the SPA gene: sequencing of candidate genes related to cell wall synthesis or co-expressed with secondary cell wall cellulose synthases and a classical mapping strategy combined with NGS methods. Moreover within the framework of the European RENEWALL and KBBE CellWall projects and thanks to the co-expression network tool BradiNet (M. Mutwil, KBBE project), RNAi strategies are in progress to inactivate a few genes selected according to specific expression criteria and potentially involved in cell wall synthesis specifically in monocots. Among these genes we are focusing on the MAP65 family (Microtubules Associated Proteins), which could play a role in cellulose deposition according to the close relationship between microfibrils and microtubules.

La foulée du coureur à pied est souvent assimilée à un ressort surmonté d'un masse. Le coureur prend appui au sol, brièvement, avec la plante du pied, rigidifie son membre inférieur et progresse en rebondissant. Pourtant, plusieurs études soutiennent qu'un appui long après une pose du pied par le talon améliore l'efficacité de la foulée. L'approche Volodalen prétend que les paramètres biomécaniques s'unifient en un système organisé selon deux grands processus d'optimisation : terrien et aérien. Le coureur aérien rebondit comme l'affirme le modèle masse-ressort. Le coureur terrien progresse en minimisant les mouvements donc l’énergie dépensée sur la verticale. Cette thèse confirme l'existence de deux modèles d'efficacité de la foulée.Le coureur terrien minimise le temps de vol et la raideur du membre inférieur et augmente le recrutement des extenseurs de hanche ainsi que l'avancée de son bassin sur l'appui. Le coureur aérien favorise un appui court, un temps de vol allongé, une activation précoce des gastrocnemii et une oscillation verticale du centre masse importante. Les deux coureurs présentent la même économie de course aux vitesses usuelles d'endurance. Selon l’approche Volodalen, ces deux processus d'optimisation sont médiés par le plaisir ressenti qui inciteraitle coureur à s'inscrire dans un environnement donné. Le coureur rebondi préfère courir plus vite. La compréhension de ces relations entre interne et externe pourrait permettre une meilleure individualisation de l'entrainement. Pourtant, les premières observations ne confirment pas l'existence d'une relation simple et directe entre la foulée du coureur à pied et un entrainement privilégié / Human motion is often modelled using a spring-madd model where the body balanced on a compressible spring. The runner bouces from one step to the next whit a mid or forefoot strike pattern and a stiff leg. However, some studies highlight that a long contact time and a rearfoot stike pattern enhance running economy. The Volodalen approach suggests that the biomechanical parameters interact in a holistic system following tow main locomotive optimization processes : terrestrial and aerial. Aerial runners bounce on the forepart of their foot following the spring-mass model, wheras terrestrial runners move forward from heel-to-toe with limites vertical desplacement. Through a series of experiments, this thesis verifies the existence of two biomechanial models to optimize running economy. Terrestrial runners seduce flight time and leg stiffness, uncreasing hip extensors recruitment and horizontal displacement during stance. Aerial runners favour a short contact time, a mid-forefoot strike pattern, a gastrocemius pre-activation, and a high vertical oscillation. Both types of runners exhibit similar running economy levels at sub-maximal running speeds. According to Volodalen, these two optimization process are mediated by runners' perceived pleasure that would guide the runner in preferntial environments. The arial runner prefers to run faster than the terrestrial runner.futher undrstanding the unerlying relation between internal processes and external environments could lead to enhanced invidualization of training. however, results from a trainning intervention indicate no differential benefit in undertaking rebound-based versus pushing-based training based.

Rôle des anomalies de transmission des contraintes dans la pathogénèse des maladies fémoro-acétabulaires / Key role of strains' transmission abnormality as an origin of femoro-acetabular diseases

Dans le cadre de ce travail de thèse nous nous sommes intéressés à deux pathologies fémoro-acétabulaires dont la pathogénie semble liée aux « conditions biomécaniques » locales. La première partie de cette thèse était consacrée à l’analyse de l’influence de l’anatomie osseuse sur l’apparition de l’ostéonécrose aseptique de la tête fémorale. Nous avons réalisé des analyses analytique de l’anatomie des patients souffrant d’ONA (notre hypothèse était que nous retrouverions un profil anatomique particulier chez les patients souffrant d’ONA par rapport à la population générale.Nos résultats démontrent que les patients souffrant d’ONA présentent fréquemment une anatomie particulière. Notre modèle en éléments finis de la hanche de dix patients souffrant d’ONAi, a permis de valider notre hypothèse puisqu’il existe un recouvrement quasi-parfait, entre la zone de nécrose osseuse et celle supportant les contraintes fémoro-acétabulaires. L’hypothèse selon laquelle une pathogénie mécanique puisse être responsable du développement l’ostéonécrose aseptique de la tête fémorale est vérifiée par nos travaux.Dans la seconde partie de cette thèse nous avons décrit le rôle mécanique du LA par une évaluation multimodale combinant analyse cinématique,de contraintes fémoro-labrales et enfin analyse en éléments finis. Les résultats de ces études ont permis de constater que le LA se déforme lors du mouvement reflétant une transmission de contraintes fémoro-labrales lors du mouvement d’abduction. Ces contraintes ont pu être enregistrées à l’aide de capteurs piezo-resistifs, elles augmentent lors du mouvement d’abduction. Ces éléments ont été confirmés par notre modèle en éléments finis. / As parts of this thesis we focused on two femoro-acetabular diseases whose pathogenesis appears to be related to "biomechanical local conditions". The first part of this thesis was devoted to analyze the influence of bony anatomy on the development of aseptic osteonecrosis of the femoral head. We thus aimed to analyze ONA patients’ anatomy in a matched-controlled fashioned study. Our results demonstrate that patients with ONA often present a particular anatomy. A finite element model of ten iONA hips validated our hypothesis since we found an overlap between bone necrosis area and femoro-acetabular strains bearing area. The assumption that a mechanical pathogenesis may be responsible for ONA development has been confirmed by our results.If the relationship between bony anatomy and bone disease is quite intuitive, acetabular labrum involvement (AL) in the regulation of femoro-acetabular mechanical conditions is unclear. The mechanical role of the AL is widely controversial, though some authors attributed, AL, a key role in hip joint’s mechanics, others advocated its complete resection in case of painful traumatic tears. We tried to demonstrate AL’s mechanical role in a multimodal analysis combining kinematic, strains and finite element analysis. The AL deforms during abduction movement reflecting femoro-labral strains’ transmission. Those strains have been recorded using piezo-resistive sensors, they increase during adduction to abduction movement. These elements were confirmed by our finite element model: labral resection changed the Femoro-acetabular strains to the detriment of cartilage surfaces.

Esta investigação tece relações comparativas entre padrões de crescimento de redes vasculares e argumenta que no conceito de morfogênese, analogamente à biologia, há evoluções e agrupamentos gerativos em sistemas estruturais ramificados. Para a compreensão deste agrupamento gerativo, são utilizados conceitos adjacentes à geometria fractal - estudo de estruturas rugosas, porosas ou fragmentadas, que mantém sua irregularidade em grau similar para todas as escalas. Embora os fractais sejam mais conhecidos como objetos resultantes de algoritmos matemáticos em iterações sucessivas no campo das ciências computacionais, nesta pesquisa as estruturas naturais que se aproximam de modelos de geometria fractal irregular, isto é, classificadas como fractais estatísticos, e que são encontradas em vários sistemas orgânicos e minerais, têm predominância. Pesquisam-se e coletam-se in loco imagens de padrões ramificados para início de produção de trabalhos práticos. Elas são em sua maioria fotografias de seres biológicos naturais com algum grau de ramificação, mas incluem-se também estruturas estatisticamente fractais análogas a sistemas biológicos. Esses estudos de casos de ramificações são então categorizados de acordo com padrões superficiais, colorações, estruturação corporal, funcionalidade no sistema em que atuam e interação com os demais sistemas ou seres vivos presentes num dado ecossistema. A partir de seleção de alguns organismos de acordo com sua estrutura corporal e seus movimentos característicos, são produzidas arte-instalações onde estruturas biomiméticas híbridas são criadas para responder cinética e sensorialmente a estímulos humanos, ambientais e climáticos. / This survey weaves comparative relationships among growth patterns of vascular networks and argues that, in the concept of morphogenesis, in analogy to biology, there are generative clustering and evolutions in branched structural systems. To understand this generative clustering, adjacent concepts to fractal geometry are used - the study of rough, porous or fragmented structures that keep their irregularity to a similar degree at all scales. Although fractals are better known as resulting objects of mathematical algorithms in successive iterations in the field of computer science, in this research the natural structures that approximate to models of irregular fractals, i.e., classified as statistical, and that are found in various minerals and organic systems are predominant. Images of branched patterns are researched and collected in loco for a first production of practical work. They are mostly photographs of natural biological beings with some branching level, but they also include statistical fractal structures analogous to biological systems. These branching study cases are then categorized according to surface patterns, colors, body structure, functionality in the system in which they operate and interaction with other systems or living organisms present in a given ecosystem. From the selection of a few organisms according to their characteristic movements and body structure, installation artworks are experimentally designed where hybrid biomimetic structures are created to sensory and kinetically respond to human, environmental and climate stimuli.
