Allergy is one of the most common diseases. Identification of early prognostic markers pointing to an increased risk of allergy development is therefore of increasing importance. Cord blood represents an easily attainable clinical material for searching for prognostic markers signalizing future allergy development. Proportions of Th1 cytokines, Th2 cytokines and regulatory cytokines were tested in cord blood of children of allergic mothers (children in relatively high risk of allergy development) in comparison with cord blood of children of healthy mothers (low risk children). Also the activities of lymphocytes, dendritic cells (DC) and regulatory cells (Tregs) were compared in children of healthy and allergic mothers. The generally increased activity of both in vitro stimulated and non-stimulated mononuclear cord blood leukocytes was proved in children of allergic mothers in comparison with low risk children. The increased activity of DC of high risk children was detectable only after polyclonal stimulation. Significantly less pronounced functional properties of cord blood Tregs were found in children of allergic mothers when compared with children of healthy mothers. The fate of neonate calves. A discussion of the bovine infant health implications of dairying in antiquity, using archaeozoological studies of six Orcadian contexts.

A methodology for ageing foetal and neonatal cattle is developed, involving radiographic examination of infant mandibles for early developmental stages in molariform teeth; tooth-wear methodologies are imprecise at this stage before wear commences. Known-age modern bovine foetal and neonate material are collected as a control assemblage for method development (n=73); six Neolithic to Norse era assemblages from Orkney are examined using the modified technique together with standard tooth-wear analysis and other methodologies. Foetal and died-at-birth material is diagnosed at most sites using the new technique, together with a range of other peri-natal age-groups. Ageing at this early stage is highly relevant in the diagnosis of milking as a palaeoeconomy: the accepted view is that unwanted (male) calves were slaughtered to maximise milk for human consumption, hence a surfeit of neonate calf remains, as at the study sites. The diagnosis of foetal and died-at-birth material challenges this view, suggesting that attritional causes may have contributed to deaths at this stage. Although milking was probably carried out at most of the study sites, this may have been combined with slaughter of cattle for meat in a pragmatic exploitation strategy. Literary research shows possible attritional causes of abortion and early death in calves, in particular dietary insufficiency in pregnant cows, microbial infections, and also inadequate colostrum uptake. Additionally, research is used to consider the challenges to health that early milking might have posed, to the calf as mentioned, but also to the cow, where three main health issues are highlighted: infertility, mastitis and lameness. / The attached files include the Landscape pages and appendices V and VI. Déterminants nutritionnels précoces du neurodéveloppement des enfants de l'étude EDEN : rôle des acides gras polyinsaturés / Early nutritional determinants of neurodevelopment among children from the Eden study : role of polyunsaturated fatty acids

Contexte : De la conception à l’âge adulte, de nombreux déterminants environnementaux influencent le neurodéveloppement. Des études montrent que durant la fin de la grossesse et les premiers mois de vie, une grande quantité d’acides gras polyinsaturés (AGPI), notamment à longue chaîne (AGPI-LC), se fixe dans le tissu cérébral. Un apport optimal durant ces périodes critiques pourrait soutenir précocement le bon développement du cerveau et de ses fonctions.Objectifs : Étudier les relations entre les expositions pré- et postnatales précoces aux AGPI, et le neurodéveloppement d’enfants de 2 et 3 ans.Population : Les données utilisées étaient celles de l’étude EDEN, une cohorte mère enfant ayant recruté 2002 femmes enceintes dans les maternités de Nancy et Poitiers entre 2003 et 2006. L’alimentation maternelle pendant la grossesse a été évaluée par un questionnaire de fréquence alimentaire et une table de composition nutritionnelle. Quatre questionnaires postnatals ont permis d’évaluer la durée d’allaitement maternel, et la composition lipidique du colostrum des mères allaitantes a été analysée par chromatographie en phase gazeuse. Des questionnaires parentaux (2 et 3 ans) et un examen neuropsychologique (3 ans) ont permis d’évaluer plusieurs aspects du neurodéveloppement. Les analyses statistiques ont été réalisées par régressions linéaires multivariées, après ajustement sur de nombreux facteurs de confusion potentiels.Résultats : Les scores de neurodéveloppement à 2 et 3 ans des enfants allaités étaient plus élevés que ceux des enfants non allaités. Chez les enfants allaités, la durée d’allaitement maternel était associée positivement au neurodéveloppement. Le rapport n 6/n 3 dans l’alimentation maternelle en acides gras en fin de grossesse était négativement associé à plusieurs mesures du neurodéveloppement, et cette association était renforcée chez les enfants non allaités. La composition du colostrum en AGPI et en AGPI-LC reflétait amplement les apports nutritionnels en AGPI et en AGPI-LC en fin de grossesse. L’utilisation de matières grasses alimentaires était aussi associée à la composition du colostrum. Les différences de neurodéveloppement des enfants allaités ne semblait pas être expliquées par la composition du colostrum en AGPI ni en AGPI-LC, à l’exception de la teneur en acide linoléique qui était négativement associée à certaines mesures du neurodéveloppement. Dans l’ensemble, les associations avec le neurodéveloppement étaient plus fréquemment retrouvées avec les questionnaires parentaux, mais certaines évaluations par les psychologues venaient parfois conforter ces résultats.Conclusion : Associés à ceux de la littérature, ces résultats soulignent le rôle notable des AGPI durant les périodes pré- et postnatale précoce pour le neurodéveloppement de l’enfant. Le suivi des enfants de l’étude EDEN jusqu’à leur 5 ans permettra d’étudier la persistance de ces résultats. Dans une perspective de santé publique, ces travaux rappellent la nécessité de promouvoir l’allaitement maternel dans la durée et de surveiller l’équilibre des apports nutritionnels en AGPI pendant la grossesse et la période de lactation. / Context: From conception to adulthood, many environmental determinants influence neurodevelopment. Studies showed that in late pregnancy and the first months of life, a large amount of polyunsaturated fatty acids (PUFA), especially long-chain-PUFA (LCPUFA), accretes in the brain. Optimal intake during these critical periods could support the brain development and its cognitive functions.Aims: To investigate the relationships between pre- and early postnatal exposures to PUFA, and neurodevelopment of 2 and 3 years old children.Population: Data were those from the EDEN mother-child cohort study, in which 2002 pregnant women were recruited between 2003 and 2006 in the maternities of Nancy and Poitiers. Maternal food intake during pregnancy was evaluated by food frequency questionnaire combined with a food composition table. Duration of breastfeeding was assessed by 4 postnatal questionnaires. Lipids in colostrum of breastfeeding mothers were analyzed by gas chromatography. Several aspects of the cognitive and motor development were assessed by both parental questionnaires (2 and 3 y) and neuropsychological examination (3 y). Statistical analyzes were performed by multiple linear regressions, after adjusting for many potential confounders.Results: At 2 and 3 years, scores of neurodevelopment were higher among breastfed children than among never breastfed children. Among breastfed children, breastfeeding duration was positively associated with neurodevelopment. Maternal dietary n 6/n 3 ratio during late pregnancy was negatively associated with measures of neurodevelopment, and this association was reinforced among never breastfed children. Colostrum composition in PUFA and LCPUFA greatly reflected maternal intake during pregnancy. Use of oils/fats for cooking and seasoning was associated with colostrum composition. The differences of neurodevelopment of breastfed children did not seem to be explained by PUFA nor LCPUFA in colostrum, except for total linoleic acid that was negatively associated with some measures of neurodevelopment. In general, associations with neurodevelopment were more frequently found with assessments by parental questionnaires, but measures by psychologists sometimes confirmed the results.Conclusion: Combined with the literature, these results underline the importance of PUFA exposures during pre- and postnatal periods for the child neurodevelopment. Follow-up of children up to 5 years will allow to investigate whether those results persist later in childhood. From a public health perspectives, this work reiterates the need to promote breastfeeding duration and to monitor the balance of PUFA intake during pregnancy and lactation periods.

Introdução: Gestações múltiplas apresentam alta morbidade relacionada a fatores como prematuridade, baixo peso ao nascer e sepse. Em gemelares, a aquisição de imunidade passiva por meio do cordão umbilical e do colostro ainda não é bem conhecida. O objetivo geral do estudo foi descrever a concentração de IgG total e específico anti-Streptococcus B, antilipopolissacarídeos de Klebsiella pneumoniae e antilipopolissacarídeos de Pseudomonas aeruginosa no cordão umbilical de recém-nascidos gemelares e a concentração de IgA secretora total e específica destes anticorpos no colostro. O objetivo específico foi analisar a associação entre infecção neonatal e concentração dos anticorpos no sangue de cordão umbilical e no colostro. Métodos: estudo prospectivo transversal de uma coorte de recémnascidos gemelares internados no Centro Neonatal do Instituto da Criança do Hospital das Clínicas da FMUSP no período de 20 meses. Foram excluídos: recém-nascidos com malformação; mães com infecção ou imunodeficiência. Variáveis analisadas: idade gestacional; peso de nascimento; classificação gestacional; concentrações de IgG total e específicos e IgA total e específicas anti-Streptococcus B, antilipopolissacarídeos de Klebsiella pneumoniae e antilipopolissacarídeos de Pseudomonas aeruginosa no sangue de cordão umbilical e no colostro; nutrição enteral; episódios de infecção; desfecho. As dosagens de IgG total foram realizadas por nefelometria e dos demais anticorpos através de ensaio imunoenzimático. Os testes t-Student pareado ou teste de Wilcoxon pareado, teste de Mann-Whitney e teste de Kruskal-Wallis, foram utilizados, considerando-se significante p< 0,005. Resultados: Foram incluídos 57 pares de gêmeos, compondo a casuística de 114 recém-nascidos. A idade gestacional foi 36±1,65 semanas (média±DP) e o peso de nascimento foi 2.304,8 ± 460g (média±DP). As concentrações de anticorpos encontradas foram (média±DP): IgG total 835,71 ± 190,73 mg/dL, IgG anti-Streptococcus B 295,1 ± 250,66 UA/mL, IgG antilipopolissacarídeos de Pseudomonas aeruginosa 280,04 ± 498,66 UA/mL e IgG antilipopolissacarídeos de Klebsiella pneumoniae 504,75 ± 933,93 UA/mL; IgA total 210,2 ± 285,3 g/L, IgA anti-Streptococcus B 6640 ± 9526,8 UA/mL, IgA antilipopolissacarídeos de Pseudomonas aeruginosa 3231,0 ± 2935,2 UA/mL, IgA antilipopolissacarídeos de Klebsiella pneumoniae 3070,1±2886,6 UA/mL. A concentração de IgG total foi menor nos recém-nascidos prematuros (p < 0,001). Em gemelares com idade gestacional < 34 semanas a concentração de IgG total (p=0,013) e IgG antilipopolissacarídeos de Pseudomonas aeruginosa (p=0,032) foi menor. A concentração de anticorpos IgG antilipopolissacarídeos de Klebsiella pneumoniae foi menor nos recém-nascidos pequenos para a idade gestacional (p=0,013). No colostro, houve detecção de IgA total, IgA anti-Streptococcus B, IgA antilipopolissacarídeos de Klebsiella pneumoniae e antilipopolissacarídeos de Pseudomonas aeruginosa em 98,1% das mães. A concentração de IgA antilipopolissacarídeos de Pseudomonas aeruginosa foi menor no colostro das mães de recém-nascidos prematuros (p=0,013). A concentração de IgA antilipopolissacarídeos de Klebsiella pneumoniae foi menor no colostro das mães com parto antes de 34 semanas (p=0,001). Houve infecção em cinco recém-nascidos, cuja concentração de IgG total foi menor (p<0,05). Os neonatos que receberam colostro apresentaram menor incidência de infecção (p < 0,001). Conclusões: Os anticorpos IgG total e específicos foram detectados no sangue do cordão umbilical em todos os gemelares, comprovando sua passagem transplacentária. Houve diminuição significativa na concentração de anticorpos IgG total nos gemelares prematuros e de IgG total e IgG antilipopolissacarídeos de Pseudomonas aeruginosa nos gemelares com idade gestacional inferior a 34 semanas. No colostro houve detecção de IgA total e específica em 98,1% das mães. A concentração de IgA anti-lipopolissacarídeos de Pseudomonas aeruginosa foi menor no colostro das mães de gemelares prematuros. Nas mães com parto antes de 34 semanas houve diminuição da concentração de IgA antilipopolissacarídeos de Klebsiella pneumoniae, sugerindo que a prematuridade possa ter influenciado a transferência de anticorpos maternos pelo colostro. In twins, acquisition of passive immunity through the umbilical cord and colostrum is not yet known. The overall aim of the study was to describe the concentration of total and specific IgG antibodies anti-Streptococcus B, anti-lipopolysaccharide of Klebsiella pneumoniae and anti-lipopolysaccharide of Pseudomonas aeruginosa in the umbilical cord of newborn twins and the concentration of total and specific secretory IgA antibodies in the colostrum. The specific aim was to analyze the association between neonatal infection and antibody concentration in the umbilical cord blood and colostrum. Methods: A prospective cross-sectional study of a cohort of newborn twins admitted to the Neonatal Center of Children\'s Institute, University of São Paulo during the period of 20 months. Patients with malformations and mothers with infection or immunodeficiency were excluded from our analysis. Variables analyzed: gestational age; birth weight; sex; gestational classification; antibody concentrations of total and specific IgG and IgA anti-Streptococcus B, anti-lipopolysaccharide of Klebsiella pneumoniae and antilipopolysaccharide of Pseudomonas aeruginosa in umbilical cord blood and colostrum; enteral nutrition; infection episodes; outcome. Total IgG measurements were performed using nephelometry and the specific antibodies were evaluated by enzyme immunoassay. We used paired Student t test or Wilcoxon test, Mann-Whitney test and Kruskal-Wallis test considering significant p < 0.005. Results: During the study period a total of 57 pairs of twins were included, making the sample of 114 newborns. Gestational age was 36 ± 1.65 weeks (mean±SD) and birth weight was 2304.8 ± 460g (mean±SD). We found the following antibody concentrations (mean±SD): total IgG 835.71 ± 190.73 mg/dL, anti-Streptococcus B IgG 250.66 ± 295.1 AU/mL, anti-lipopolysaccharide of Pseudomonas aeruginosa IgG 280.04 ± 498.66 AU/mL and anti-lipopolysaccharide of Klebsiella pneumoniae IgG 504.75±933.93 AU/mL; total IgA 210.2 ± 285.3 g/L, anti- Streptococcus B IgA 6640±9526.8 AU/mL, anti-lipopolysaccharide of Pseudomonas aeruginosa IgA 3231.0±2935.2 AU/mL, antilipopolysaccharide Klebsiella pneumoniae IgA 3070.1±2886.6 AU/mL. The concentration of total IgG was lower in preterm infants (p < 0.001). In twins with gestational age < 34 weeks both total IgG concentration (p = 0.013) and anti-lipopolysaccharide of Pseudomonas aeruginosa IgG concentration (p = 0.032) were lower. The concentration of anti-lipopolysaccharide of Klebsiella pneumoniae IgG was lower in small for gestational age newborns (p=0,013). There was detection of total IgA and specific antibodies in the colostrum of 98.1% mothers. The concentration of anti-lipopolysaccharide of Pseudomonas aeruginosa IgA was lower in the colostrum of premature\'s mothers (p = 0.013). The concentration of anti-lipopolysaccharide of Klebsiella pneumoniae IgA was lower in the colostrum of mothers with delivery before 34 weeks (p = 0.001). Five newborns were diagnosed with sepsis and in this group the concentration of total IgG was lower (p < 0.05). Neonates receiving colostrum had a lower incidence of infection (p < 0.001). Conclusion: This study showed detection of total and specific IgG antibodies in umbilical cord blood for all twin newborn, proving its transplacental passage. There was a significant decrease in the concentration of total IgG antibodies in premature twins and antilipopolysaccharide of Pseudomonas aeruginosa IgG in twins with gestational age less than 34 weeks. There was detection of total and specific IgA in the colostrum of 98.1% mothers. The concentration of anti-lipopolysaccharide of Pseudomonas aeruginosa IgA was lower in mothers of premature twins. Among the newborns with gestational age less than 34 weeks there was a decrease in the concentration of anti-lipopolysaccharide Klebsiella pneumoniae IgA, suggesting that prematurity may have influenced the transfer of maternal antibodies through colostrum. The highest incidence of infection in the newborn group who did not receive colostrum and in those who had significantly lower total IgG serum antibodies reinforces the importance of anti-infectious protection afforded by passive immunity transferred from the mother

Variables analyzed: gestational age; birth weight; sex; gestational classification; antibody concentrations of total and specific IgG and IgA anti-Streptococcus B, anti-lipopolysaccharide of Klebsiella pneumoniae and antilipopolysaccharide of Pseudomonas aeruginosa in umbilical cord blood and colostrum; enteral nutrition; infection episodes; outcome. Total IgG measurements were performed using nephelometry and the specific antibodies were evaluated by enzyme immunoassay. We used paired Student t test or Wilcoxon test, Mann-Whitney test and Kruskal-Wallis test considering significant p < 0.005. Results: During the study period a total of 57 pairs of twins were included, making the sample of 114 newborns. Gestational age was 36 ± 1.65 weeks (mean±SD) and birth weight was 2304.8 ± 460g (mean±SD). We found the following antibody concentrations (mean±SD): total IgG 835.71 ± 190.73 mg/dL, anti-Streptococcus B IgG 250.66 ± 295.1 AU/mL, anti-lipopolysaccharide of Pseudomonas aeruginosa IgG 280.04 ± 498.66 AU/mL and anti-lipopolysaccharide of Klebsiella pneumoniae IgG 504.75±933.93 AU/mL; total IgA 210.2 ± 285.3 g/L, anti- Streptococcus B IgA 6640±9526.8 AU/mL, anti-lipopolysaccharide of Pseudomonas aeruginosa IgA 3231.0±2935.2 AU/mL, antilipopolysaccharide Klebsiella pneumoniae IgA 3070.1±2886.6 AU/mL. The concentration of total IgG was lower in preterm infants (p < 0.001). In twins with gestational age < 34 weeks both total IgG concentration (p = 0.013) and anti-lipopolysaccharide of Pseudomonas aeruginosa IgG concentration (p = 0.032) were lower. The concentration of anti-lipopolysaccharide of Klebsiella pneumoniae IgG was lower in small for gestational age newborns (p=0,013). There was detection of total IgA and specific antibodies in the colostrum of 98.1% mothers. The concentration of anti-lipopolysaccharide of Pseudomonas aeruginosa IgA was lower in the colostrum of premature\'s mothers (p = 0.013). The concentration of anti-lipopolysaccharide of Klebsiella pneumoniae IgA was lower in the colostrum of mothers with delivery before 34 weeks (p = 0.001). Five newborns were diagnosed with sepsis and in this group the concentration of total IgG was lower (p < 0.05). Neonates receiving colostrum had a lower incidence of infection (p < 0.001). Conclusion: This study showed detection of total and specific IgG antibodies in umbilical cord blood for all twin newborn, proving its transplacental passage. There was a significant decrease in the concentration of total IgG antibodies in premature twins and antilipopolysaccharide of Pseudomonas aeruginosa IgG in twins with gestational age less than 34 weeks. There was detection of total and specific IgA in the colostrum of 98.1% mothers. The concentration of anti-lipopolysaccharide of Pseudomonas aeruginosa IgA was lower in mothers of premature twins. Among the newborns with gestational age less than 34 weeks there was a decrease in the concentration of anti-lipopolysaccharide Klebsiella pneumoniae IgA, suggesting that prematurity may have influenced the transfer of maternal antibodies through colostrum. The highest incidence of infection in the newborn group who did not receive colostrum and in those who had significantly lower total IgG serum antibodies reinforces the importance of anti-infectious protection afforded by passive immunity transferred from the mother

Silagem de colostro: caracterização do perfil de fermentação anaeróbia e desempenho de bezerros leiteiros / Colostrum silage: dynamics of anaerobic fermentation and dairy calves performance evaluation

O objetivo deste estudo foi caracterizar o perfil de fermentação anaeróbia de colostro e determinar seu valor nutritivo, assim como avaliar o desempenho de bezerros leiteiros alimentados com silagem de colostro como dieta líquida. No primeiro experimento, colostro bovino de segunda e terceira ordenhas foi fermentado em garrafas plásticas tipo PET que foram cheias e ligeiramente pressionadas antes do seu fechamento, criando assim uma condição anaeróbia. As garrafas foram armazenadas em sala escura à temperatura ambiente e cinco garrafas foram abertas 0,1, 2, 3, 4, 5, 6, 7, 14, 21, 28 e 56 dias após, para determinação de pH, acidez titulável, temperatura, ácido lático, nitrogênio total, nitrogênio não-protéico (NNP), caseína, lactose, glicose, gordura e contagem de microorganismos. Os valores de pH, acidez titulável e concentração de ácido lático apresentaram variação durante o armazenamento (P<0,0001). A fração NNP apresentou aumento (P<0,0001), enquanto que os valores de caseína e lactose diminuíram (P<0,0001) e apresentaram baixos valores após 56 dias. O desenvolvimento de bactérias ácido-láticas (BAL) foi intenso durante o processo fermentativo, enquanto que enterobactérias e leveduras apresentaram declínio. No segundo experimento, colostro bovino de segunda e terceira ordenhas foi fermentado da mesma forma que no primeiro experimento, mas em diferentes condições de temperaturas ambientais. As garrafas foram armazenadas em incubadora BOD com temperatura controlada (32,5±1°C ou 22,5±1°C) ou em sala escura à temperatura ambiente, sendo abertas três garrafas após 0, 1, 7, 14, 21, 28 e 35 dias para avaliação dos mesmos parâmetros avaliados no primeiro estudo. A maioria dos parâmetros apresentou comportamento semelhante ao observado no primeiro experimento. O desenvolvimento de BAL foi intenso, principalmente quando o colostro foi armazenado em temperatura mais elevada. A temperatura também afetou os parâmetros nutricionais (P<0,0001) com acréscimos nos valores de NNP e redução da concentração de caseína e lactose. No terceiro experimento, dezoito bezerros Holandês foram alocados em abrigos individuais até a oitava semana de vida, com livre acesso à água e concentrado inicial, e passaram a receber 4L da dieta líquida, sucedâneo lácteo ou colostro fermentado sob condições anaeróbicas (silagem de colostro), diluído na razão de 1:1. Os animais alimentados com silagem de colostro apresentaram menor consumo de concentrado, ganho de peso diário e peso vivo (P<0,07) durante o período experimental, em comparação aos animais consumindo sucedâneo lácteo. As avaliações quanto à altura na cernelha, perímetro torácico e largura da garupa não apresentaram diferenças (P>0,07). Todos os parâmetros sanguíneos avaliados (glicose, N-uréico, ácidos graxos livres e -hidroxibutirato) foram afetados pelos tratamentos (P<0,07), exceto a concentração plasmática de proteínas totais (P>0,07). O escore fecal foi afetado pelos tratamentos durante a segunda semana de vida (P<0,07), com animais alimentados com silagem de colostro apresentando fezes anormais e muito secas. A dinâmica fermentativa observada mostra que o colostro pode ser conservado através de fermentação anaeróbia de forma eficiente. Entretanto, o fornecimento de silagem de 12 colostro como dieta líquida exclusiva durante o período de aleitamento não resulta em desempenho animal adequado, não sendo uma boa alternativa de substituto de leite. / The objective of this study was to characterize the fermentative dynamic of bovine colostrum, under anaerobic condition, and determine the nutritional value, as well as evaluate dairy calves´ performance receiving colostrum silage as liquid diet. In the first experiment, bovine colostrum from second and third milking was fermented at room temperature in PET plastic bottles that were filled and lightly pressed before its closure, thereby creating an anaerobic condition. The bottles were stored in the dark at room temperature and five bottles were opened on days 0,1, 2, 3, 4, 5, 6, 7, 14, 21, 28 and 56 after for determination of pH, titratable acidity, temperature, lactic acid, total nitrogen, non-protein nitrogen (NPN), casein, lactose, glucose, fat and counts of microorganisms. The pH, titratable acidity and concentration of lactic acid showed large variation during storage (P<0.0001). The NPN fraction increased (P<0.0001), whereas the values of casein and lactose decreased (P<0.0001) and showed very low values after 56d. The development of lactic-acid bacteria (LAB) was intense during the fermentation process, while enterobacteria and yeasts declined. In the second experiment, bovine colostrum from second and third milking was collected, mixed and stored in PET plastic bottles, in the same manner as it was for the first study. However, the bottles were stored in a dark room with temperature controlled by a BOD incubator (32.5±1°C or 22.5±1°C) or at room temperature. Three bottles were opened after 0, 1, 7, 14, 21, 28 and 35 days for evaluation of the same parameters done during the first study. Most parameters presented similar patterns as observed in the first trial. The development of LAB was intense, especially when colostrum was stored at higher temperature. Temperature also influenced the results of nutritional parameters (P<0.0001) with increases in NPN values and reduction of casein and lactose concentration. In the third experiment, eighteen Holstein calves were allocated to individual hutches until the eighth week of life, with free access to water and starter feed, and fed 4 liters of liquid diet: milk replacer or colostrum fermented under anaerobic condition (colostrum silage), diluted at a 1:1 ratio. Animals fed colostrum silage had lower intake of starter, average daily gain and body weight (P<0.07) during the experimental period, compared to animals consuming milk replacer. The values of withers height, heart girth and hip width showed no differences (P>0.07). All blood parameters measured (glucose, urea-N, free fatty acids and -hydroxybutyric acid) were affected by treatments (P<0.07), except the total protein plasma concentration (P>0.07). The fecal score was affected by treatments during the second week of life (P<0.07), with animals fed colostrum silage presenting abnormal and very dry feces. The dynamics of fermentation show that colostrum can be efficiently stored under anaerobic conditions. However, feeding colostrum silage as an exclusive liquid diet, during the whole milk-feeding period, results in poor animal performance, being considered as an inadequate alternative as milk replacer.

Déterminants nutritionnels précoces du neurodéveloppement des enfants de l'étude EDEN : rôle des acides gras polyinsaturés

Contexte : De la conception à l'âge adulte, de nombreux déterminants environnementaux influencent le neurodéveloppement. Des études montrent que durant la fin de la grossesse et les premiers mois de vie, une grande quantité d'acides gras polyinsaturés (AGPI), notamment à longue chaîne (AGPI-LC), se fixe dans le tissu cérébral. Un apport optimal durant ces périodes critiques pourrait soutenir précocement le bon développement du cerveau et de ses fonctions.Objectifs : Étudier les relations entre les expositions pré- et postnatales précoces aux AGPI, et le neurodéveloppement d'enfants de 2 et 3 ans.Population : Les données utilisées étaient celles de l'étude EDEN, une cohorte mère enfant ayant recruté 2002 femmes enceintes dans les maternités de Nancy et Poitiers entre 2003 et 2006. L'alimentation maternelle pendant la grossesse a été évaluée par un questionnaire de fréquence alimentaire et une table de composition nutritionnelle. Quatre questionnaires postnatals ont permis d'évaluer la durée d'allaitement maternel, et la composition lipidique du colostrum des mères allaitantes a été analysée par chromatographie en phase gazeuse. Des questionnaires parentaux (2 et 3 ans) et un examen neuropsychologique (3 ans) ont permis d'évaluer plusieurs aspects du neurodéveloppement. Les analyses statistiques ont été réalisées par régressions linéaires multivariées, après ajustement sur de nombreux facteurs de confusion potentiels.Résultats : Les scores de neurodéveloppement à 2 et 3 ans des enfants allaités étaient plus élevés que ceux des enfants non allaités. Chez les enfants allaités, la durée d'allaitement maternel était associée positivement au neurodéveloppement. Le rapport n 6/n 3 dans l'alimentation maternelle en acides gras en fin de grossesse était négativement associé à plusieurs mesures du neurodéveloppement, et cette association était renforcée chez les enfants non allaités. La composition du colostrum en AGPI et en AGPI-LC reflétait amplement les apports nutritionnels en AGPI et en AGPI-LC en fin de grossesse. L'utilisation de matières grasses alimentaires était aussi associée à la composition du colostrum. Les différences de neurodéveloppement des enfants allaités ne semblait pas être expliquées par la composition du colostrum en AGPI ni en AGPI-LC, à l'exception de la teneur en acide linoléique qui était négativement associée à certaines mesures du neurodéveloppement. Dans l'ensemble, les associations avec le neurodéveloppement étaient plus fréquemment retrouvées avec les questionnaires parentaux, mais certaines évaluations par les psychologues venaient parfois conforter ces résultats.Conclusion : Associés à ceux de la littérature, ces résultats soulignent le rôle notable des AGPI durant les périodes pré- et postnatale précoce pour le neurodéveloppement de l'enfant. Le suivi des enfants de l'étude EDEN jusqu'à leur 5 ans permettra d'étudier la persistance de ces résultats. Dans une perspective de santé publique, ces travaux rappellent la nécessité de promouvoir l'allaitement maternel dans la durée et de surveiller l'équilibre des apports nutritionnels en AGPI pendant la grossesse et la période de lactation.

The impact of exogenous TGFβ1 on male reproductive function.

The TGFβ family of cytokines are potent signalling molecules that regulate tissue development, inflammation and immunity. Previous studies in mice with a null mutation in the Tgfb1 gene (TGFβ1-/- mice) implicate a key role for TGFβ1 in male reproductive function. These mice show profound infertility due to an inability to copulate successfully, associated with reduced testosterone and sperm production. The focus of this project was to 1) further characterize mechanisms underpinning reproductive deficiency in male TGFβ1-/- mice, 2) identify a reliable physiological marker of TGFβ1 availability in vivo, and 3) to determine whether exogenous TGFβ1 administration influences TGFβ1 availability and restores fertility. To investigate the causes of unsuccessful copulation by TGFβ1-/- mice, penis morphometry was examined. Penile organ structure, as assessed by scanning electron microscopy, was comparable between genotypes however a superfluous epidermal covering that impeded penile spine protrusion was evident in TGFβ1-/- mice. The epidermal covering was not due to increased epithelial cell proliferation, as measured by Brdu labelling and immunohistology. Behavioural observations of erectile activity showed that TGFβ1-/- mice achieved spontaneous erections albeit at reduced frequency compared to TGFβ1+/+ mice. The efficacy of exogenous TGFβ1 replacement was evaluated by first identifying measures of in vivo TGFβ1 availability and/or function and selecting an effective route of administration. Serum TGFβ1 and testosterone levels were reliable discriminators of TGFβ1 genotype. Gene expression and phagocytic function of peritoneal macrophages revealed no differences between genotypes. Exogenous sources of TGFβ1 for replacement studies included colostrum, naturally occurring in breast milk and recombinant human latent TGFβ1 (rhLTGFβ1). Colostrum did not increase circulating levels and rhTGFβ1 injection caused only transient elevation of serum levels. Thus mini-osmotic pumps were used to deliver a constant supply of cytokine to TGFβ1-/- mice. The fertility status of TGFβ1-/- mice receiving exogenous TGFβ1 was investigated. Reproductive behaviour in response to normal receptive female mice was assessed twice during treatment, on day 7 and day 14. Blood, liver and reproductive tissues were collected at sacrifice. Circulating TGFβ1 was increased in TGFβ1 treated TGFβ1-/- mice above TGFβ1-/- control levels, although this did not affect circulating testosterone. Erectile activity and sperm production were unchanged. Videotaping behaviour with estrous females revealed that the TGFβ1+/+ mice successfully mounted and intromitted, unlike the TGFβ1-/- controls. The TGFβ1-/- mice receiving exogenous TGFβ1 displayed moderately enhanced mounting and intromission behaviour although this remained less frequent than in the TGFβ1+/+ controls. Ejaculation behaviour was not observed in any TGFβ1-/- mice regardless of TGFβ1 replacement, compared to TGFβ1+/+ controls where >90% mice displayed ejaculated. Modest improvement in the copulation activity of the TGFβ1-/- mice receiving exogenous TGFβ1 suggests that systemic TGFβ1 availability can influence reproductive performance in male TGFβ1-/- mice. However since fertility was not restored, locally produced TGFβ1 in the reproductive tract and/or hypothalamic pituitary axis are also implicated in regulating fertility.
