It is defined as a pruritic allergic skin disease, with genetic predisposition and clinical features, being related to the response to environmental allergens. Interleukin 31 (IL) 31 is a cytokine that participates in inflammatory processes and it is associated with pruritic diseases, especially those involving chronic inflammation such as allergic dermatitis. Produced by mononuclear cells, IL-31 is described to play an important role in the pathogenesis of atopic dermatitis. Two studies were performed in order to correlate clinical features, histopathological changes and the presence of IL-31 in the skin of dogs with atopic dermatitis. 34 dogs were selected from clinical routine, which 31 animals were diagnosed with CAD and three were healthy. The animals were evaluated for pruritus level by the owners' report and by clinical examination according to the CAD extent and severity index (CADESI-4). The dogs were grouped in discreetly, moderately and markedly compromised by CAD following the sum of scores’ values assigned in the clinical examination. Cutaneous samples from the axillary and interdigital regions of each dog were collected and submitted to histopathological (HE and toluidine blue) and immunohistochemical analyzes (IL-31). There was a correlation between the clinical score and the microscopic changes. As well, there was correlation among all the microscopic changes, but not between the degree of pruritus and the clinical score of CAD. Also, the inflammatory infiltrate was more intense in the axillary region in relation to interdigital skin. An increased numbers of cells immunostained for IL-31 was observed in dogs severely compromised by CAD. There was a correlation between the clinical score and the amount of interdigital mast cells, with an amount of cells immunostained for IL-31 in the axilla. It was also verified correlation between the amount of mast cells and cells immunostained for IL-31 in the axilla, as well as between acanthosis and all other histopathological alterations of the skin in the axillary region. We concluded that there is a correlation between severity of dermathological lesions, IL-31 immunostaining, mast cell count and histopathological changes in dogs with atopic dermatitis, but there is no correlation between the degree of pruritus reported by owners and severity of cutaneous lesions in animals with CAD. Moreover, the intensity of the inflammatory process may vary depending on the anatomical site of the lesion. / A dermatite atópica canina (DAC) é a segunda doença mais incidente na rotina de atendimento clínico dermatológico de cães. É definida como doença cutânea alérgica pruriginosa, de predisposição genética e características clínicas definidas, estando relacionada a resposta a alérgenos ambientais. A interleucina 31 (IL) 31 é uma citocina que participa de processos inflamatórios e está associada a doenças pruriginosas, principalmente as que envolvem inflamação crônica como as dermatites alérgicas. Produzida por células mononucleares, a IL-31 é descrita por desempenhar papel importante na patogênese da dermatite atópica.Com o objetivo de correlacionar as alterações clínicas, histopatológicas e a presença da IL-31 na pele de cães com dermatite atópica foram realizados dois estudos. Para isso, foram selecionados 34 cães da rotina de atendimento clínico, sendo 31 com diagnóstico de DAC e três hígidos. Os animais foram avaliados quanto ao nível de prurido a partir do relato de seus proprietários e quanto ao índice de extensão e severidade da DAC (CADESI-4) a partir do exame clínico. Os cães foram agrupados em discretamente, moderadamente e acentuadamente comprometidos pela DAC após somatório dos valores de escores atribuídos no exame clínico. Amostras cutâneas da região axilar e interdigital de cada cão foram colhidas e submetidas às análises histopatológica (HE e azul de toluidina) e imunoistoquímica (IL-31). Houve correlação entre o escore clínico e as alterações microscópicas, assim como entre as alterações microscópicas, mas não entre o grau de prurido e o escore clínico da DAC. Também o infiltrado inflamatório foi mais intenso na região axilar em relação a interdigital. Maior número de células imunomarcadas para IL-31 foi observado nos cães acentuadamente comprometidos pela DAC. Houve correlação entre o escore clínico e a quantidade de mastócitos no interdígito, do mesmo modo com a quantidade de células imunomarcadas para IL31 na axila. Também foi verificada correlação entre a quantidade de mastócitos e células imunomarcadas para IL-31 na axila, assim como entre acantose e todas as demais alterações histopatológicas da pele na região axilar. Conclui-se que há correlação entre a gravidade das lesões, a imunomarcação de IL-31, a contagem de mastócitos e as alterações histopatológicas em cães com dermatite atópica, mas não há correlação entre o grau de prurido relatado pelos proprietários e a gravidade das lesões cutâneas em animais com DAC. Ainda, a intensidade do processo inflamatório pode variar em função do sítio anatômico da lesão.

Estudo de propriedades microbiológicas e toxicológicas do xilitol visando a sua aplicação no controle da dermatite atópica / Study of microbiological and toxicological properties of xylitol and its application on atopic dermatitis control

O crescente aumento da resistência microbiana aos antibióticos disponíveis impulsiona a busca por novas substâncias, com características superiores às correntemente usadas. Neste sentido, este trabalho propôs investigar as propriedades terapêuticas do xilitol visando a sua aplicação no controle da dermatite atópica, patologia que acomete a pele e que é agravada pela presença da bactéria Staphylococcus aureus. No presente estudo foram executados ensaios in vitro de atividade antimicrobiana do xilitol e verificado se este composto atua na aderência bacteriana, sobre a bactéria Staphylococcus aureus ATCC 25923. Foi avaliada a ação do xilitol produzido tanto pela via química quanto pela biotecnológica, sendo este último obtido a partir de fermentação do hidrolisado hemicelulósico da palha de trigo, nas concentrações de 1, 5 e 10 % (p/ v). A seguir, foram executados testes de toxicidade dérmica aguda com doses repetidas com xilitol nas concentrações de 5 e 10 % (p/ p), nas formas farmacêuticas de creme e de gel, em coelhos albinos da raça Nova Zelândia e testes de fototoxicidade, na concentração de 10 % (p/ p), nas formas farmacêuticas de creme e de gel, utilizando cobaias albinas da raça Durkin-Hartley. Em relação aos ensaios in vitro, observou-se que o xilitol, nas concentrações testadas, não impediu o crescimento bacteriano, mas inibiu a aderência da bactéria a uma superfície de prova, evidenciando ser este o provável mecanismo de ação desta substância sobre as bactérias. Em testes toxicológicos realizados, todas as formulações contendo xilitol foram classificadas como não irritantes quando foram avaliadas quanto à toxicidade dérmica aguda com doses repetidas. Entretanto, nos testes da fototoxicidade, as formulações testadas apresentaram certa fototoxicidade, sugerindo ser a formulação a base de creme mais fototóxica do que aquela a base de gel. Estes resultados evidenciam a aplicabilidade do xilitol no controle da dermatite atópica, como princípio ativo de formulações seguras, observando que a aplicação deve ser realizada com o uso de protetor solar. Este estudo buscou contribuir de maneira expressiva na elucidação do mecanismo de ação do xilitol e verificar os cuidados que devem ser considerados quando realizada sua aplicação pela via dérmica. / Since the microorganisms\' resistance to antibiotics increases, it is imperative to look for different substances that can combat these pathogens growth, with greater advantages. The propose of this work was to study therapeutic properties of xylitol in order to control atopic dermatitis, a dermal disease characterized by the presence of Staphylococcus aureus bacterium. Xylitol is a substance that can be obtained by chemical and biotechnological means, being the latter a relevant alternative that produces high-value compounds obtained by fermentation of lignocelullosic hydrolysates. In the present study in vitro assays were performed in order to check if wxylitol (obtained by chemical and biotechnological means) has antimicrobial activity at 1, 5 and 10 % (w/ v) concentrations. Other assays were also performed to verify if xylitol properties, at the same concentrations, hinder the adherence of Staphylococcus aureus ATCC 25923 bacterium. Xylitol was produced by biotechnological means using wheat straw hemicellulosic hydrolysate. Afterwards, in vivo assays were performed to investigate if xylitol is safe for skin application. Acute dermal toxicity tests with repeated doses were done with New Zealand rabbits using concentrations of 5 and 10 % (w/ w) xylitol, in either cream or gel. Phototoxicity assays were also performed with Durkin-Hartley guinea pigs, using only 10 % (w/ w) xylitol, in cream and gel forms. It was observed that xylitol does not have antimicrobial properties on S. aureus at all tested concentrations, but this compound has the capability of inhibiting this bacterium adherence on a surface, at all tested concentrations. In relation to toxicity assays, formulations contaning xylitol are nonirritative. However, xylitol has phototoxicity properties, mainly when cream is the base. Xylitol is an adequate alternative to be applied for atopic dermatitis control, and its application on the skin should be done with sunscreen. This study aimed to clarify xylitol action mechanism and to check the cares that should be taken when xylitol is applied on skin.

Individual and environmental risk factors for hand eczema in hospital workers

Individual and environmental risk factors in hand eczema have been investigated in a prospective cohort study of 2452 newly employed hospital workers with a follow-up time of 20 months. Current hand eczema was analyzed in 142 wet hospital workers from this cohort with respect to the etiologic importance of irritants, allergens and contact urticants. The density of the microflora and the effect on the microflora of topical treatment with a potent corticosteroid were studied in 20 patients with hand eczema. ’Wet’ hospital work was found to increase the odds of developing hand eczema only twice compared to 'dry' office work. Nursing children under four years old and the lack of a dish-washing machine significantly increased the risk of contracting hand eczema. Unfavourable combinations of these domestic factors increased the risk as much as wet work. A history of atopic dermatitis approximately tripled the odds both in wet as well as in dry work. Histories of earlier hand eczema (HHE), metal dermatitis (HMD) and of atopy were analyzed as risk factors for hand eczema in 1857 women in wet work. HHE increased the odds by a factor of 12.9 and created a subdivision of the population into high risk individuals and normal risk individuals. HHE was found in half of the subjects with atopic dermatitis, in one quarter of the subjects with atopic mucosal symptoms and in one fifth of the non-atopics. A HMD increased the odds by a factor of 1.8. This increase was seen as a high risk level in subjects with HHE and as a normal risk level in subjects with no HHE. A history of atopic disease as a complement to information about HHE and HMD increased the odds by another 1.3 times. The predicted probability of developing hand eczema ranged from 91 % in subjects with a combination of HHE, HMD and atopy to 24% in subjects with none of these risk factors. Subjects with AD were found to suffer a more severe form of hand eczema with significantly higher figures for medical consultation, sick- leave, termination due to hand eczema, early debut, permanent symtoms and vesicular lesions. Amongst the patients investigated for current hand eczema high risk individuals were overrepresented. It was claimed in 92.3% of the cases that trivial irritant factors had elicited the current episodes of hand eczema. In 35% of the cases the exposure to the irritant took place largely at home. Although contact sensitivity and contact urticaria were fairly common, they mostly seemed to be of minor importance in the etiology of the current hand eczema. Staphylococcus aureus colonized eczematous lesions of the hands in 18/20 patients. The density exceeded 105 colony forming units/cm2 in 15/20 patients. Only three of these patients showed signs of clinical infection. Comorbidity, distress, coping and social support in asthma and allergy / Komorbiditet, lidande, hantering och socialt stöd i astma och allergi

Asthma and allergies are some of the most common illnesses worldwide that almost everybody will come in contact with. This thesis studied persons with allergic asthma, non-allergic asthma, allergic rhinitis and atopic dermatitis in a population-based sample. At an early stage, these illnesses were regarded as psychosomatic. Over time, as knowledge about asthma/allergy has increased more of a biomedical perspective was taken by the research field. In considering early documentations well as contemporary research, a psychobiosocial perspective was taken in this thesis when conducting the three studies. Thus, as psychological factors may affect the illness and be a result of the illness, it is important to incorporate these factors to better understand asthma and allergy. Study I examined the co- and multimorbidity in asthma/allergy with the environmental intolerances in the form of chemical and building-related intolerance. Study II investigated psychological distress in the four forms of asthma and allergy. Psychological distress was in this study defined as stress, burnout, anxiety, depression and environmental health worries. Study III examined usage of problem and emotion focused coping strategies and perceived social support from the surrounding in high and low asthma/allergy severity. All studies were performed using data from the Västerbotten Environmental Health Study, a questionnaire-based survey with focus on various environmental hypersensitivities and asthma and allergy. The result showed that the co- and multimorbidity with the environmental intolerances in asthma/allergy was larger than what was statistically excepted. Those with allergic asthma and atopic dermatitis experienced more stress, burnout and anxiety than those with non-allergic asthma, allergic rhinitis and non-asthma/allergy. Moreover, the most common way of coping with asthma and allergy was found to be strategies such as avoiding environments that are believed to affect health, and trying to accept the situation, independent of asthma/allergy severity. Finally, in general, those with asthma and allergy reported receiving most support from their partner, other family members and health care, and least support was perceived by those with low asthma/allergy severity.The findings suggest that co- and multimorbidity with environmental intolerances is relatively common in asthma and allergy, and should therefore be included in the clinical anamnesis for this patient group. The elevated level of distress in allergic asthma and atopic dermatitis evokes the question of use of therapies such as mindfulness maybe beneficial in certain afflicted persons. The results on coping and social support provide a foundation for further research regarding informing the asthma/allergy patient and family members about effective coping strategies and the importance of adequate social support. A metaperspective is taken in which interrelations between important variables in the thesis are discussed.

MORFOLOGIA COMPARADA DA PITIOSE EM CAVALOS, CÃES E BOVINOS / COMPARATIVE MORPHOLOGY OF PYTHIOSIS IN HORSES, DOGS AND CATTLE

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Twenty-one cases of pythiosis involving horses (10), dogs, (nine) and cattle (2) were investigated. In all of them the etiology was confirmed through immunohistochemistry technique. Data related to clinical course, localization and macroscopic features of the lesions and cases closure were obtained from biopsies and necropsies protocols. For compared histomorphological and histochemical characterization, hematoxilin-eosin, Grocott‟s, Schiff periodic acid, sirius red and Grocott-Sirius red techiques were performed. The aspects analyzed were intralesional hyphal quantity, distribution and integrity, angioinvasive capability, Splendore-Hoeppli (SH) and inflammatory reactions inter-species. The equine macroscopic lesions were characterized by sole focally extensive wounds composed by abundant fibrous tissue surrounding yellowish, dense, firm and irregular branching coral-like masses (kunkers). Histologically, lesions consisted of well-collagenized connective tissue with multiple often coalescing areas of immature granulation tissue encircling coarsely round eosinophil cores (corresponding to kunkers). Dogs presented gastrointestinal and/or cutaneous lesions. The gastrointestinal ones usually consisted of transmural and segmental thickening of the organs walls and mesenteric tissue. Mesenteric lymph nodes were always involved. Cutaneous lesions consisted of ulcers in the lips mucocutaneous junction, masses in the thorax and prepuce and small nodules in chest, thorax and fore limbs. Two main patterns of inflammation were seen in dogs, alone or combined, independently of the tissue: one granulomatous/pyogranulomatous and another necro-eosinophilic. In cattle, cutaneous lesions were multiple and usually affected one or more limbs, ventral neck and sternum and, eventually, tail or muzzle. Microscopically, there were multifocal to coalescing discrete granulomas along with proliferated dermal collagen. The sirus red technique was quite useful to localize eosinophils in tissue and confirm the SH reaction nature, seen in the three species. Histomorphological aspects as quantity, distribution and integrity of intralesional hyphae, angioinvasive potential and the type of inflammatory and SH reactions seem to be implicated in the different kinds of lesions and clinical courses in different animal species suffering from pythiosis. Evidences that pythiosis cure is associated to granulomatous inflammation were observed. / Foram estudados vinte e um casos de pitiose em cavalos (dez), cães (nove) e bovinos (dois). Todos os casos tiveram a etiologia confirmada pela técnica de imuno-histoquímica. As informações relativas aos dados de anamnese, forma clínica, localização e macroscopia das lesões e desfecho dos casos foram obtidas a partir de protocolos de biópsia e necropsia. Para a comparação histomorfológica e histoquímica foram realizadas as técnicas de hematoxilina e eosina, impregnação pela prata de Grocott, ácido periódico de Schiff, sirius red, Grocott-sirius red e azul de toluidina. Foram analisadas a quantidade, a distribuição e a integridade das hifas intralesionais, a capacidade de angioinvasividade, a reação de Splendore-Hoeppli (SH) e o tipo de resposta inflamatória interespécies. Nos cavalos, as lesões macroscópicas caracterizavam-se por feridas únicas e focalmente extensas constituídas por tecido fibroso abundante circundando massas amarelas, compactas, firmes e com ramificações grosseiras lembrando corais (kunkers). Histologicamente, as lesões eram compostas basicamente por tecido conjuntivo fibroso bem colagenizado, com múltiplas áreas, muitas vezes coalescentes, constituídas por tecido de granulação jovem, circundando coleções de eosinófilos densas e grosseiramente circulares (correspondentes aos kunkers). Nos cães foram estudados casos das formas gastrointestinal e/ou cutânea. As lesões do trato gastrointestinal geralmente consistiam de espessamento segmentar transmural da parede das vísceras e da gordura mesentérica adjacente. Em todos os casos havia envolvimento dos linfonodos mesentéricos. As lesões cutâneas se apresentavam como úlceras na junção mucocutânea do lábio, massas no prepúcio e tórax e nódulos pequenos distribuídos pelo peito, tórax e membro anterior. Nos cães, dois padrões principais de inflamação foram vistos, isolados ou combinados, independentemente do tecido analisado: um granulomatoso/piogranulomatoso e um necro-eosinofílico. Nos bovinos, as lesões cutâneas eram múltiplas e aleatórias e geralmente acometiam um ou mais membros, a região ventral do pescoço e esterno e, eventualmente, a cauda ou o chanfro nasal. Microscopicamente havia granulomas multifocais ou coalescentes perfeitamente delimitados em meio ao colágeno dérmico levemente proliferado. A técnica de sirius red foi particularmente útil para localizar os eosinófilos nos tecidos e confirmar a natureza da reação SH vista em todas as espécies. Aspectos histomorfológicos tais como a quantidade, distribuição e integridade das hifas intralesionais, o potencial de angioinvasividade e os tipos de resposta inflamatória e de reação SH parecem estar implicados nos diferentes tipos de lesão e cursos clínicos da pitiose nas diferentes espécies animais acometidas pela doença. Evidências de que a cura da pitiose possa estar associada à inflamação granulomatosa foram observadas.

Avaliação do uso glicerina em dietas para frangos de corte nas fases pré-inicial e final / Evaluation of the use glycerin in diets for broilers in the pre-starter and final phases

Dois experimentos foram conduzidos com o objetivo de avaliar a utilização de dietas com níveis crescentes de glicerina de biodiesel apenas na fase pré-inicial (1 a 7 dias) e apenas na fase final (35 a 42 dias de idade) de frangos de corte, respectivamente. As variáveis analisadas incluíram o desempenho, o rendimento de carcaça e cortes, a retenção de umidade em pintos aos 7 dias, a umidade de cama, a incidência de pododermatite, os níveis plasmáticos de colesterol e triglicerídeos e o custo da ração referente ao ganho de peso. Os tratamentos consistiram de dietas formuladas com 0%, 5% ou 10% de glicerina, sendo isonutritivas e formuladas à base de milho, farelo de soja e óleo de milho. A glicerina usada continha 83,6% de glicerol, 11,2% de água, 1,83% de sódio e 397 mg/kg de metanol, com energia metabolizável estimada de 3258 kcal/kg. No Experimento I, as dietas com glicerina foram fornecidas apenas na fase pré-inicial ou durante todo o ciclo de criação com 1610 aves em um delineamento inteiramente casualizado com 5 tratamentos e 7 repetições. Na primeira semana, as dietas com glicerina promoveram melhora significativa no ganho de peso e na conversão alimentar, sem afetar o consumo de ração e viabilidade. Essa melhora não foi mantida após os 14 dias de idade. Não houve efeito de tratamentos sobre o ganho de peso, consumo de ração e conversão alimentar. Entretanto, houve redução significativa na viabilidade aos 35 dias de idade das aves com 10% de glicerina por todo ciclo, mas não aos 42 dias; ao final do experimento, o índice de eficiência produtiva desse tratamento sofreu redução significativa. A umidade da cama das aves do tratamento com 10% de glicerina por todo ciclo de criação foi mais alta que dos demais a partir dos 21 dias, entretanto a incidência de pododermatite foi mais severa tanto para 5% como para 10% de glicerina por todo ciclo. Os níveis e tempos de fornecimento de glicerina não afetaram o rendimento de carcaça e de cortes ou a gordura abdominal. No experimento II, as dietas experimentais foram fornecidas a 819 frangos apenas na semana final de criação em um experimento inteiramente casualizado com 3 tratamentos e 7 repetições. O desempenho e o custo de ração por kg de ganho de peso não foram afetados pelos tratamentos. Nos Experimentos I e II, as rações contendo glicerina durante todo o ciclo ou apenas na última semana não resultaram em alteração nas concentrações plasmáticas de triglicerídeos e colesterol dos frangos. Dietas contendo 5% ou 10% de glicerina na fase pré-inicial ou 5% de glicerina continuamente não afetam a produtividade de frangos de corte, enquanto o nível de 10% durante todo o ciclo foi prejudicial. Esse mesmo nível de glicerina pode ser usado na dieta dos 35 aos 42 dias de idade sem afetar as aves. / Two experiments were conducted to evaluate the use of diets with increasing levels of biodiesel glycerin only in pre-starter (1-7 days) and only in the final phase (35-42 days of age) of broilers, respectively. The variables analyzed included the performance, carcass and parts yield, moisture retention in chicks at 7 days, litter moisture, incidence of foot pad dermatitis, plasma levels of cholesterol and triglycerides and diet cost related to weight gain. Treatments were isonutritive diets based on corn, soybean meal and corn oil containing 0, 5 and 10% glycerin. The glycerin used contained 83.6% glycerol, 11.2% water, 1.83% sodium and 397 mg/kg methanol, with estimated metabolizable energy value of 3,258 kcal/kg. In the first experiment, diets with glycerin were provided only in the pre-starter or for the entire growth period with 1610 birds in a completely randomized design with 5 treatments and 7 replications. In the first week, diets with glycerin resulted in significant improvement in weight gain and feed conversion without affecting feed intake and viability. This improvement was not sustained after 14 days of age. There was no effect of treatments on weight gain, feed intake and feed conversion. However, there was a significant reduction in viability at 35 days of age for the birds with 10% glycerin throughout the cycle, but not at 42 days; at the end of the experiment, the productivity index of this treatment was significantly decreased. Litter moisture for the treatment with 10% glycerin throughout the cycle was higher than the for the others from the 21 days, however the incidence of foot pad dermatitis was more severe for both 5% and 10% glycerin treatments for the entire period. The levels and periods of glycerin feeding did not affect carcass and parts yield and abdominal fat. In experiment II, the experimental diets were provided to 819 chickens only in the final week of growth in a completely randomized design with 3 treatments and 7 replications. The performance and feed cost per kg of weight gain were not affected by treatments. In Experiments I and II, the diets containing glycerin throughout the cycle or just in the last week resulted in no change in plasma concentrations of triglycerides and cholesterol of chickens. Diets containing 5% or 10% glycerin in the pre-starter phase or 5% glycerin continually did not affect the productivity of broilers, while the 10% level throughout the cycle was detrimental. This same level of glycerin can be used in the diet from 35 to 42 days without affecting the birds.

Estudo da expressão de filagrina e claudinas 1 e 4 em indivíduos adultos com dermatite atópica / Study of expression of filaggrin and claudin 1 and 4 in adults with atopic dermatitis

Introdução: A dermatite atópica (DA) é uma doença cutânea inflamatória crônica que cursa em surtos. Possui manifestação clínica variável, mas o prurido e a xerose são características frequentes, e pode estar associada a outras manifestações extra-cutâneas de atopia. Pacientes com DA apresentam maior risco de infecções por bactérias e vírus, destacando-se a erupção variceliforme de Kaposi, causada por herpes simples. A DA mostra-se como exemplo de dermatose com comprometimento da barreira cutânea, aliado a disfunção imunológica. São descritas alterações das proteínas da barreira cutânea na DA (filagrina e claudinas ), relacionadas ao maior risco de infecção . Objetivo: Avaliar a expressão de proteínas relacionadas à barreira cutânea como a filagrina, e as claudinas -1 e -4 na pele de pacientes adultos com dermatite atópica, acompanhados no Departamento de Dermatologia da Faculdade de Medicina da Universidade de São Paulo. Métodos: 32 indivíduos com diagnóstico de DA (estabelecido pelos critérios de Hanifin & Rajka) e 23 controles (indivíduos sem DA), maiores de 18 anos, foram submetidos a biópsias cutâneas. Os indivíduos com DA foram biopsiados em dois pontos, tanto na pele lesada, quanto na pele não-lesada. O material obtido foi analisado por imuno-histoquímica, através de marcadores específicos para filagrina, claudina-1 e claudina-4. As lâminas foram digitalizadas pelo Panoramic Scan - 3DHistech - Hungria, e as imagens analisadas pelo software Image Pro Plus 4,5, quanto à intensidade da expressão do marcador. A espessura média da epiderme do local estudado foi também avaliada. O grupo com DA foi também analisado quanto à gravidade da doença (EASI), níveis séricos de IgE e grau de eosinofilia. Resultados: Houve redução da expressão da filagrina na pele de doentes de DA em relação aos controles, tanto na pele com lesão quanto na pele sem lesão. Demonstrou-se correlação inversa na expressão da filagrina, tanto com relação à gravidade da doença quanto à espessura da epiderme. A análise das claudinas -1 e -4 demonstrou redução de ambas na pele dos doentes de DA, mas não houve correlação com a gravidade, espessura da epiderme, níveis de IgE sérica ou eosinofilia. Conclusão: No adulto com dermatite atópica, existe redução da expressão das proteínas relacionadas à barreira cutânea, como a filagrina e as claudinas -1 e -4. A redução da expressão da filagrina relacionou-se inversamente com a gravidade da doença, e com a espessura da epiderme, sugerindo cronicidade das lesões. Its clinical features are variable, but pruritus and xerosis are frequent, and the disease may be associated to extracutaneous atopy. Patients with AD have increased risk for bacterial or viral infection, with emphasis on eczema herpeticum due to herpes simplex. AD is an example of a compromised skin barrier, allied to na imune dysfunction. There are reports on efective proteins of the skin barrier (filaggrin and claudins), related to increased risk for infection. Objectives: To evaluate the expression of proteins related to the skin barrier, such filaggrin and claudins-1 and-4 in the skin of adults with AD, followed at the Department of Dermatology, University of Sao Paulo Medical School. Methods: 32 individuals diagnosed as AD, according to Hanifin & Rajka\'s criteria, and 23 non-atopic controls, above the age of 18, were biopsied. Individuals with AD were biopsied in two different sites (lesional and nonlesional skin). The specimens were analyzed by immunohistochemistry through specific markers for filaggrin, claudins 1 and 4. The slides were scanned utilizing Panoramic Scan - 3DHistech - Hungary, and images analyzed by Image Pro Plus 4,5 for the intensity of each marker. The mean epidermal thickness was also evaluated. AD patients were also analyzed for disease severity (EASI), circulating IgE levels and eosinophilia. Results: In lesional and nonlesional skin of AD patients there was a reduced expression of filaggrin, when compared to nonatopic controls. There was an inverse correlation of filaggrin expression with disease severity and epidermal thickness. In the skin of AD individuals, there was reduced expression of claudins 1-and-4, which did not correlate with disease severity, epidermal thickness or eosinophilia. Conclusion: In adults with AD, there is reduced expression of skin barrier proteins, such as filaggrin, claudins 1 and 4. The reduction of filaggrin expression had an inverse correlation with disease severity and epidermal thickness, suggesting disease chronicity. There was reduction of claudins 1 and 4, with no relation with disease severity, epidermal thickness, circulating IgE levels or eosinophilia

One hundred bovine females, 50 Holstein and 50 Girolando, from two different farms were used for the risk factor study. Experimental units were divided into four groups: healthy Holstein cattle group (GHS) and digital dermatitis Holstein cattle group (GHL), healthy Girolando cattle group (GGS) and digital dermatitis Girolando cattle group (GGL). Ten milliliters of blood were collected from all animals and stored in anticoagulant added tubes. Samples were refrigerated and submitted to hemolytic tests for eight bovine blood systems: A, B, C, F, J, L, S and Z. The frequency of blood factors was determined and compared, in each system and each group. Data were analyzed through non parametric tests: Chi Square and Fisher s exact. The results indicated that the blood factors may contribute to the occurrence of digital dermatitis lesions, but further studies are needed for a more accurate conclusion. Thirty confined, 10 months old calves were selected in order to evaluate the efficacy of 5% barbatimão solution. A biopsy punch was introduced in each interdigital space, up to the dermis, to produce a lesion measuring one centimeter in diameter. The lesions were numbered 120 in total and named experimental units, which were divided into two groups of 60 units each: GB treated with 5% aqueous barbatimão bark extract solution and GC treated with water. Tissue repair was evaluated through macroscopic and histological findings in days three, seven, 14 and 21 of treatment. Macroscopic findings were analyzed descriptively, while histological findings were submitted to Mann Whitney test, with p<0.05. For lesions treated, GB group showed less intense polymorphonuclear cells infiltration on the first 14 days of treatment. A marked collagen deposition was also observed at day 14 while collagen alignment was more expressive on day 21. More specific studies may be necessary in order to determine if blood factors can contribute to the occurrence of digital dermatitis. Barbatiman 5% footbath solution for confined cattle helps qualitatively in modulation of interdigital cutaneous incisional wound repair, but does not avoid the extension of inflammatory phase of healing. / Objetivou-se nesta pesquisa estudar um fator de risco para a dermatite digital bovina e avaliar a eficácia do extrato de barbatimão em solução aquosa a 5% como auxiliar no reparo de feridas cutâneas induzidas no espaço interdigital de bovinos. Para pesquisar o fator de risco utilizaram-se 100 fêmeas bovinas, sendo 50 Holandesas e 50 Girolando, de duas propriedades rurais. Dividiram-se as unidades experimentais em quatro grupos: grupo de animais saudáveis da raça Holandesa (GHS) e grupo com lesões de dermatite digital (GHL); grupo de animais saudáveis da raça Girolando (GGS) e grupo com lesões de dermatite digital (GGL). De todos os animais colheram-se 10 ml de sangue, em tubo a vácuo, contendo anticoagulante. As amostras foram refrigeradas e submetidas a testes hemolíticos, para oito sistemas sanguíneos de bovinos: A, B, C, F, J, L, S e Z. A frequência dos fatores sanguíneos foi determinada e comparada, dentro de cada sistema, em cada grupo de animais. Os dados foram analisados através dos testes não paramétricos de Qui-quadrado e Exato de Fisher. Pelos resultados obtidos concluiu-se que os fatores sanguíneos podem contribuir para a ocorrência de lesões de dermatite digital, contudo são necessários estudos mais específicos para uma conclusão mais precisa. Para avaliar a eficácia da solução de barbatimão a 5%, utilizaram-se 30 animais com dez meses de idade, mantidos em sistema de confinamento. Em cada espaço interdigital dos animais, com auxílio de um punch de um centímetro de diâmetro, produziu-se uma lesão cutânea até a derme, totalizando 120 feridas. As feridas foram denominadas unidades experimentais e distribuídas em dois grupos com 60 unidades cada: grupo GB tratado com solução de extrato de barbatimão a 5% e grupo GC tratado com água. A evolução do processo de reparação tecidual das feridas foi acompanhada por meio de avaliações macroscópicas e histológicas nos dias três, sete, 14 e 21 de tratamento. Os achados macroscópicos foram analisados descritivamente e os histológicos submetidos ao teste de Mann Whitney, com p<0,05. Para as feridas tratadas, observou-se que o grupo GB apresentou menor intensidade de infiltrado de células polimorfonucleares nos primeiros 14 dias de tratamento. Nesse grupo observou-se ainda que a deposição de colágeno mostrou-se mais acentuada aos 14 dias e aos 21 dias as fibras colágenas apresentavam-se mais alinhadas. O uso do extrato de barbatimão a 5% em pedilúvio, em bovinos confinados, auxiliou qualitativamente no processo de modulação da reparação de feridas cutâneas interdigitais incisionais, mas não impediu o prolongamento da fase inflamatória do processo cicatricial.
