Desenvolvimento e validação de métodos bionalíticos para dosagem de antimicrobianos em plasma humano

Para tanto é necessário que métodos de análise em matrizes biológicas com uma maior seletividade e sensibilidade para quantificação de fármacos e/ou metabólitos sejam desenvolvidos e validados para posterior utilização em estudos de farmacocinética comparada (bioequivalência). No Brasil ainda é escasso o número de profissionais com formação acadêmica (Técnico-científica) para a condução de estudos de biodisponibilidade comparada, sobretudo na etapa analítica. Com o intuito de colaborar com a implementação desta política, o presente trabalho tem como objetivo desenvolver e validar métodos de análise de duas classes de antibióticos (Sulfonamidas e Fluoroquinolonas) em plasma humano e posterior aplicação na avaliação biofarmacêutica de diferentes formulações. Durante a formação foi realizado um aumento da complexidade das técnicas utilizadas desde a extração líquido-líquido (ELL) e utilização de cromatografia líquida de alta eficiência com detecção por absorção no ultravioleta (CLAE-UV), extração em fase sólida (EFS) com auxílio de CLAE-UV e EFS seguida de cromatografia líquida de alta eficiência com detecção por espectrometria de massas em tandem (CL-EM/EM). O desenvolvimento e validação de métodos bioanalíticos vai desde a seleção de sistemas cromatográficos e de detecção, técnica de extração e clean-up que gerem cromatogramas de qualidade, avaliação da estabilidade de fármacos na matriz biológica até o cumprimento dos critérios de aceitação das seqüências analíticas contendo amostras dos voluntários. Dentre as fluoroquinolonas o objeto de estudo foi o norfloxacino (NFX) comprimido de 400mg. Para extração e clean-up das amostras em plasma humano utilizou-se o método por ELL seguido de quantificação por CLAE-UV. Em meio às muitas sulfonamidas o objeto de estudo foi composto de duas formulações da associação de sulfametoxazol + trimetoprima (SMZ + TMP). Determinou-se simultaneamente a SMZ + TMP em plasma humano após administração da forma farmacêutica (FF) cápsula (400mg + 80mg) através de método de extração e clean-up em fase sólida off-line com posterior quantificação por CLAE-UV e após a administração da FF suspensão (400mg + 80mg / 10mL) por CL-EM/EM com ionização à pressão atmosférica através da técnica de eletronebulização ( Electrospray, ESI ) em modo positivo. A biodisponibilidade dos fármacos, a parir das formas farmacêuticas, foi avaliada através da análise estatística de parâmetros como Área sob a Curva (ASC), concentração plasmática máxima (Cmáx) segundo as normas (RE 898 de 29 de maio de 2003) da ANVISA. Os métodos analíticos foram desenvolvidos e validados segundo a RE 899 de 29 de maio de 2003 da ANVISA mostrando-se lineares, precisos, exatos e robustos frente ao elevado throughput de amostras. O método EFS-CL-MS/MS mostrou-se mais robusto pela maior limpeza dos interferentes da matriz plasmática e pelo menor tempo de análise (2.5 min) por amostra, diminuindo os resíduos de solventes da fase móvel bem como no procedimento de extração mostrando-se menos tóxico aos analistas e ao meio ambiente. Todos os métodos desenvolvidos se configuraram como ferramentas suficientemente capazes de discriminar a biodisponibilidade comparada dos medicamentos testados

In attempt to assuage this problem and trying harnessing the underused waste, some proposals have been emerged, like the use of these residues to produce handcrafts which reduces the waste of wood and adds a great value to a thing that is seen as a trash. Other proposal is the present study, which proposed the phytochemical study of three timber species of Fabaceae family: Dipteryx odorata (cumaru), Swartzia laevicarpa (saboarana) e Andira parviflora (sucupira-vermelha), contributing for the knowledge of the chemical constituents of the wood, and in search for secondary metabolites with biological activity or chemical relevance. The organic crude extracts were fractionated by chromatography column (CC). The fractionation of the dichloromethane extract from residues of S. laevicarpa, resulted in the isolation of 8-hydroxy-3,4,9,10-tetramethoxipterocarpan. The methanolic extract of D. odorata fractionated in CC led the isolation of four isoflavones: 8-O-metilretusin, cladrastrin, 7,3’-dihydroxy-8,4’dimethoxi-isoflavone and the 7,3’-dihydroxy-5,6,4’-trimethoxi-isoflavone, this last one is reported for the first time. The fractionation of the methanolic extract of the alburnum of A. parviflora in CC led the isolation of the isoflavone genistein, and in the methanolic extract of A. parviflora's core were isolated the 7,5’,6’-trihydroxy-4’-methoxi-isoflavan and biochanin A. The resulting mother liquor of biochanin A, was evaluated by HPLC-EFS-NMR, leading to the isolation and identification of substances 3,8-dihydroxy-9-methoxipterocarpan,8-dihydroxy-9-methoxipterocarpan, nissolin, medicarpin, biochanin A and the novel bipterocarpan medicarpin secundiflorol-I (OC 3 → C-3 '). In this work were identified by spectroscopic techniques flavonoids (isoflavone, isoflavan and pterocarpan) typical of the Fabaceae family. / lixo, geralmente descartado em locais inadequados causando grandes problemas ambientais. Na tentativa de amenizar esses problemas e na busca de aproveitamento dos resíduos sólidos têm surgido propostas como a produção de pequenos objetos a qual é uma forma sócio-econômica de agregar valor aos resíduos. Por meio de estudos fitoquímicos dos resíduos, a presente proposta visou proporcionar o conhecimento químico de madeiras das espécies de família Fabaceae Dipteryx odorata (cumaru), Swartzia laevicarpa (saboarana) e Andira parviflora (sucupira-vermelha). Os extratos orgânicos foram fracionados por cromatografia em coluna (CC). O fracionamento do extrato diclorometano dos resíduos de S. laevicarpa resultou no isolamento de 8-hidroxi-3,4,9,10-tetrametoxipterocarpano. O extrato metanólico de D. odorata fracionado em CC forneceu 4 isoflavonas: 8-O-metilretusina, cladastrina, 7,3’-dihidroxi-8,4’dimetoxiisoflavona e a inédita 7,3’-dihidroxi-5,6,4’-trimetoxiisoflavona. Os fracionamentos dos extratos metanólico do alburno de A. parviflora em CC forneceu a isoflavona genisteína e do cerne o isolamento de 7,5’,6’-trihidroxi-4’-metoxiisoflavana e biochanina A, ambas purificadas por recrsitalização. A água-mãe resultante desta última foi avaliada a técnica hifenada CLAE-EFS-RMN, levando ao isolamento e identificação das substâncias 3,8-dihidroxi-9-metoxipterocarpano, 8-dihidroxi-9-metoxipterocarpano, nissolina, medicarpina, biochanina A e o inédito bipterocarpano medicarpina-secundiflorol I (C-3→O-C-3'). Nesse estudo identificou-se por técnicas especteroscópicas flavonóides (isoflavona, isoflavana e pterocarpano) típicos da família Fabaceae.

Pharmacogénétique du DHFR chez les enfants leucémiques

Le dihydrofolate réductase (DHFR) est la principale cible du méthotrexate, un important composant du traitement de la leucémie lymphoblastique aiguë (LLA). Une association des polymorphismes du promoteur de DHFR avec l’issue de la LLA a été mise en évidence au laboratoire. Une survie sans événement (EFS) réduite corrélait avec les allèles A -317 et C -1610, et l’haplotype *1, défini par ces allèles. L’haplotype *1 était aussi associé à une expression élevée du DHFR. Dans cette étude, nous étendons l’analyse à la région régulatrice adjacente, d’environ 400 pb, correspondant au transcrit mineur non-codant du DHFR, qui joue un rôle essentiel dans la régulation de la transcription au niveau du promoteur majeur. Six polymorphismes ont été identifiés, parmi lesquels 5 étaient des SNPs et un polymorphisme de longueur composé d’un nombre variable d’éléments de 9 pb et d’une insertion/délétion de 9 pb. L’analyse d’haplotype, incluant tous les polymorphismes promoteurs, a révélé une diversification de l’haploytpe *1 en 5 sous-types (*1a à *1e). Les variations du promoteur majeur et les sous-types de l’haplotype *1 ont été par la suite analysés pour l’association avec l’issue de LLA. Un EFS réduit corrélait avec l’allèle A du polymorphisme G308A (p=0,02) et avec l’haplotype *1 (p=0,01). Des niveaux élevées d’ARNm étaient trouvés chez les porteurs de l’haplotype *1b (p=0,005) et pas pour les autres sous-types de l’haplotype *1. Alors, la mauvaise issue de LLA associée avec l'haplotype *1 est en effet déterminée par le sous-type *1b. Engelskinlärning i förskolan och grundskolans tidiga år : En kvalitativ studie om lärarnas uppfattningar om tidig engelskinlärning

Syftet med undersökningen var att undersöka lärarnas uppfattning om betydelsen av tidig engelskinlärning i förskola och skola. Lärarnas utmaningar med att ta vara på de ungas kontakt med engelska via fritiden och bygga på denna i undervisningen. Vidare lärarnas utmaningar med flerspråkiga elever. Studien utfördes på fem skolor av olika karaktär: En mångkulturell skola. En skola beläget i ett” högstatusområde”, en skandinavisk skola i Spanien, en skola som satsar på projektet ”Engelska spåret” och en förskola där en engelskspråkig pedagog arbetar med barnen tillsammans med svenska pedagoger. Detta urval gjordes för att kunna observera eventuella likheter och skillnader mellan lärarnas uppfattningar. Undersökningen har gjorts genom kvalitativa intervjuer med öppna frågor för datainsamling och resultatet har analyserats med hjälp av frågeställningar. Det framkom olika uppfattningar om betydelsen av en tidig engelskinlärning i skolan. Yngre barns öppna ”språk fönster”, timplanernas utrymme för engelska språket, lärarnas intresse och kunskap för barns språkinlärning samt den mångkulturella skolans inflytande.

The subject of this dissertation is the confessional revivalist organisation Evangeliska Fosterlands-Stiftelsen (EFS – approximately the Swedish Evangelical Mission Society) between 1856 and 1910. EFS was founded in 1856 in a Pietistic tradition, and its aim was to revitalise from within what was seen as a too dormant State church, and also to counteract the influence of the emerging free churches. The study has five main sections. The introductory part consists of the theoretical framework and the historical context. In the second chapter EFS’ aims and expectations are studied on a national level. The third chapter examines the content of published and distributed tracts. The fourth chapter focuses on the activities of the itinerant colporteurs, and the fifth and final part studies the work of a local EFS-congregation. The aim of the dissertation is twofold. The first objective is to describe the transformation EFS underwent during the period studied. This process is described in terms related to Jürgen Habermas’ expression “public sphere”. When EFS was founded, as a board, it can be seen as one of other middle- and upper-class associations, and as such a part of the Swedish public sphere. By 1910 EFS had begun to move away from the Swedish State church and had become more like contemporary popular movements and free churches – it had started to take the shape of an limited alternative sphere, a denomination. The second aim is to use EFS as an example to describe and analyse the changed perception of religion during the second half of the 19th century. This change is described as a process of secularisation. Secularisation is seen here as the process that turned religion into an individual, voluntary and optional act of faith, among other religious and non-religious alternatives, for ordinary people. Of particular interest is the paradoxical relation between revivalism and secularisation. Various revivalist movements emphasised the personal relation to God and the individual right to interpret the word of God. The individual choice for salvation was also stressed within revivalism. These movements also created new alternatives to the all-embracing State church. Thus both the position of the Church, and the universal claims of Christianity in general, were undermined. The transformation EFS underwent is seen as an adaptation to the rise of modern society, which became more pluralistic and hence competitive during the final decades of the 19th century. Modulation of TRPV1 function in sensory neuropathy

This thesis examined how and why TRPV1 function is being modulated in sensory neuropathy and explored the potential of its rescue in the urinary bladder of STZ-­‐induced diabetic rats. Diabetes induced a rapid decline in TRPV1 function and changes in neurogenically mediated electrically-­‐evoked responses together with a gradual decline in muscarinic function. Diabetic bladder was also deficient in muscarinic and TRPV1 organ bath temperature-­‐induced changes but not in those affecting spontaneous contractile activity. Exposure to a potential neuropathy causative agent, methylglyoxal was studied and its mechanism of action explored through the use of TRPA1 ligands. Methylglyoxal exposure mimicked some of the effects of diabetes on TRPV1, neurogenic electrically evoked responses and muscarinic function. Methylglyoxal effects were seen to be partly through TRPA1 receptor activation but other as yet undefined pathways were also involved. Use of TRPA1 ligands revealed an unexpected complexity of the interaction of the TRPA1 receptor with TRPV1. Finally the potential of reversing the diminished TRPV1 response was examined through the use of three known sensitising agents, bradykinin, NGF and insulin. Bradykinin was the only agent seen to reverse the TRPV1 diminished response back up to to control equivalent levels and through the use of bradykinin selective ligands, it was seen that the dual activation of BK-­‐1 and BK-­‐2 receptor was necessary to rescue the TRPV1 response. The likely mechanism of action of bradykinin was through prostaglandin production as indomethacin blocked TRPV1 rescue. In the acute stage of diabetes, TRPV1 function is downregulated and may be caused by exposure to a neuropathy-­‐causing metabolite such as methylglyoxal. The TRPV1 function still retains plasticity at this acute stage because function could be enhanced back to control levels by bradykinin receptor activation : a potential for early therapeutic intervention.

För att få ett lokalt signum på uppsatsen kommer den till stor del att handla om en lokal missionär, Olle Hagner från Ovansjö socken, som kom att tillbringa närmare 40 år som missionär i östra Afrika – många av dessa med sin fru som följeslagare.</p><p>Följande frågeställningar har fungerat som utgångspunkt:</p><p>* Varför missionerade EFS i just Eritrea?</p><p>* Vad åstadkom Olle Hagner?</p><p>* Är det möjligt att se en förändring över tid på hur missionärerna såg på den inhemska religionen på plats?</p><p>* Hur gestaltade sig de inter- och intrareligiösa relationerna mellan EFS-missionärer (särskilt Hagner), muslimer, kopter, katoliker och anhängare av den inhemska religionen?</p><p>* Vilka tänkbara orsaker ligger bakom eventuella förändringar i attityder och relationer?</p><p>Kritiken från Hagner och EFS mot andra samfund var bitvis hård, och då framförallt mot den katolska kyrkan. Detta är inte förvånande då den katolska kyrkan gjorde allt för att tränga ut EFS’ missionärer ur Eritrea och lyckades också med detta under en period.</p><p>Olle Hagner anlände till Eritrea första gången 1923 efter att ha lovat att ta vid efter sin broder Johans tidigare missionsgärning i Eritrea som han tvingats avbryta på grund av sjukdom. Hagner kom att arbeta som missionär i Eritrea i närmare 40 år och hans tid som missionär kom att innebära många konflikter, framförallt med den katolska kyrkan och den italienska staten vilka han inte värderade högt. Hagner samarbetade under sin tid i Eritrea med flertalet av andra religiösa samfund. Hagner utövade sin mission i tre olika språkområden i Eritrea. I ett av dom, Kunama, har Hagner utfört översättningsarbeten av bland annat Nya testamentet.</p>

Detta är inte förvånande då den katolska kyrkan gjorde allt för att tränga ut EFS’ missionärer ur Eritrea och lyckades också med detta under en period. Olle Hagner anlände till Eritrea första gången 1923 efter att ha lovat att ta vid efter sin broder Johans tidigare missionsgärning i Eritrea som han tvingats avbryta på grund av sjukdom. Hagner kom att arbeta som missionär i Eritrea i närmare 40 år och hans tid som missionär kom att innebära många konflikter, framförallt med den katolska kyrkan och den italienska staten vilka han inte värderade högt. Hagner samarbetade under sin tid i Eritrea med flertalet av andra religiösa samfund. Hagner utövade sin mission i tre olika språkområden i Eritrea. I ett av dom, Kunama, har Hagner utfört översättningsarbeten av bland annat Nya testamentet.

O método de polimerização por precipitação foi utilizado para a síntese dos polímeros, devido à simplicidade de preparo, altos rendimentos e obtenção de partículas mais uniformes, devido a não trituração do polímero. O MIP foi sintetizado com ácido metacrílico (MAA) como monômero funcional, trimetacrilato de trimetilolpropano (TRIM) e dimetacrilato de etilenoglicol (EDMA) como agentes de reticulação e o cloridrato de amilorida (AMI) foi escolhido como molécula-molde. Diferentes proporções de MAA, TRIM, EDMA, volume e tipo de solvente foram utilizadas para ajuste das condições ideais de síntese. Os MIP foram avaliados quanto à capacidade de adsorção comparando-se a polímeros sintetizados na ausência da molécula-molde (NIP, polímeros não impressos). O solvente de elevada polaridade empregado na síntese (THF:MeOH:H2O) permitiu o emprego da técnica para moléculas polares como AMI. O controle no volume de solvente permitiu a obtenção de partículas maiores, de modo que a EFS foi realizada em condições usuais, o que confere um potencial para aplicação dessa técnica de polimerização na preparação de adsorventes para EFS. O polímero que apresentou maior capacidade adsortiva no ensaio realizado em tampão citrato-acetato pH 6,5 foi o MIP/NIP 12 (AMI:MAA:TRIM 1:8:10), com uma taxa média de adsorção de 83 e 88% para NIP e MIP, respectivamente. A adsorção foi elevada devido a interação iônica entre MAA e AMI promovida pelo controle de pH, porém foi não específica. O polímero MIP/NIP 12 foi aplicado como adsorvente na EFS, onde a recuperação de AMI foi avaliada nos resíduos de carregamento e eluição com solventes. O carregamento com tampão citrato-acetato pH 6,5 foi o ideal, favorecendo a interação iônica do polímero com o analito. A eluição total de AMI do cartucho somente ocorre após lavagem com o solvente na presença de ácido, que protona os grupos carboxila do polímero, rompendo assim a interação iônica com o analito / Molecularly imprinted polymers (MIPs) were synthesized and applied as adsorbents in solid-phase extraction technique (SPE). The polymers have been synthesized by precipitation polymerization method because of its simplicity, high yields and good control of final size and shape of particles. MIP was synthesized using methacrylic acid (MAA) as functional monomer, trimethylolpropane trimethacrylate (TRIM) and ethyleneglycol dimethacrylate (EDMA) as cross-linker and amiloride hydrochloride (AMI) was chosen as template. Different ratios of MAA, TRIM and EDMA, volume and type of solvent were used to adjust the optimal synthesis conditions. The MIP were tested for adsorption capacity compared to the polymers synthesized in the absence of template molecule (NIP, non-imprinted polymers). The polar solvent mixture used (THF:MeOH:H2O) allowed the synthesis of MIP of polar molecules as AMI. The solvent volume control afforded the larger particles so the SPE was performed in the usual conditions, giving a potential application for this polymerization technique in the preparation of adsorbents for SPE. The polymers with higher adsorption capacity at the test performed in citrateacetate buffer pH 6,5 was MIP/NIP 12 (AMI:MAA:TRIM 1:8:10) with adsorption rate of 83 and 88% for NIP and MIP, respectively. The recognition of MIP was due to ionic interaction between MAA and AMI promoted by pH control, but was not specific. The polymer MIP/NIP 12 was used as a solid-phase extraction sorbent and the recoveries of AMI was evaluated using different loading and elution conditions. The loading with buffer citrate-acetate pH 6,5 was optimal, due to ionic interaction of the polymer with the analyte. Total elution of AMI bound to the polymers only occurs after washing with a acid-containing solvent, because of protonation of the carboxyl groups of the polymer and disrupting the ionic interaction with the analyte
