Dextranas e seus derivados fosforilados: obten??o e avalia??o do potencial antioxidante, imunomodulat?rio e anticoagulante

Contudo, h? pouco relatos sobre as atividades das dextranas com figura??o do tipo ? (alfa). Nesse contexto, dextranas (?-D-glucanas) extra?das da bact?ria Leuconostoc mesenteroides, com massas moleculares de 10 (D10), 40 (D40) e 147 (D147) kDa e seus derivados fosforilados P10, P40 e P147 foram avaliados quanto ao seu potencial antioxidante, anticoagulante e imunomodulador pela primeira vez, com o intuito de elucidar compostos com atividades potentes e pouco t?xicas. An?lises de espectroscopia de infravermelho, composi??o monossacar?dica e dosagens qu?micas comprovaram que estas dextranas s?o o mesmo polissacar?deo, mas com massas moleculares diferentes, al?m de confirmar o sucesso da fosforila??o. Nenhuma das dextranas tem atividade anticoagulante. No teste do poder redutor verificou-se que D147 e P147 foram duas vezes mais potentes que as outras dextranas. Por outro lado, as seis amostras tiveram atividade semelhante (50%) em sequestrar o radical OH. Frente ao sequestro do ?on super?xido, apenas D10 teve uma atividade pronunciada (50%). D40 foi a ?nica dextrana nativa com a??o imunomodulat?ria, pois estimulou em dobro a prolifera??o de macr?fagos mur?nicos (RAW 264.7) e dobrou a libera??o de NO por essas c?lulas, tanto na aus?ncia como na presen?a de LPS. Al?m disso D40 teve maior atividade sequestradora (50%) de per?xido de hidrog?nio, o que fez com que ela tamb?m fosse a dextrana mais potente em inibir peroxida??o lip?dica (70%). J? P147 apresentou maior atividade quelante de ?ons ferro e cobre (~85%), al?m de aumentar o triplo a libera??o de NO na aus?ncia e na presen?a de LPS. P10 provou ser o composto mais efetivo sobre a prolifera??o de macr?fagos. Os dados indicam que dextranas com massa aproximada de 40 kDa s?o as ideais para serem utilizadas como antioxidantes e imunomoduladores, podendo ter suas atividades potencializadas com o processo de fosforila??o. Contudo, estudos futuros com D40 e outras dextranas de massa semelhante confirmar?o esta hip?tese. / Glucans are polysaccharides with different pharmacological and biological activities described. However, there are some reports about the activities of the glucan type ? (alpha). In this context, a group of ?-D-glucans called dextrans extracted from Leuconostoc mesenteroides bacteria, with molecular weights of 10 (D10), 40 (D40) and 147 (D147) kDa and their phosphorylated derivatives P10, P40 and P147, were evaluated as for their antioxidant, anticoagulant and immunomodulatory potential for the first time, in order to elucidate compounds with potent activities and low toxicity. Infrared spectroscopy analysis, monosaccharide composition and chemical dosages showed that these dextrans are the same polysaccharide, but with different molecular weights, besides confirming the success of phosphorylation. None presented with anticoagulant features. The reducing power test showed that D147 was twice as potent as other dextrans. On the other hand, all six samples showed similar activity (50%) when it came to scavenging the OH radical. To the superoxide ion scavenging, only D10 had a pronounced activity (50%). D40 was the single native dextran that presented with immunomodulatory features since it double stimulated the proliferation of murine macrophages (RAW 264.7) and double the release of nitric oxide by the cells, both in the absence and presence of lipopolysaccharides (LPS). In addition, D40 showed a greater scavenging activity (50%) for the hydrogen peroxide, which caused it to also be the more potent dextran when it came to inhibiting lipid peroxidation (70%). On other hand, P147 showed the highest iron and copper ion chelation activity (~85%). P10 proved be the most effective compound to macrophage proliferation. The results point toward dextrans with a 40 kDa weight as being ideal for antioxidant and immunomodulatory use, could be supplemented with phosphorylated derivatives. However, future studies with the D40 and other similarly dextrans are to confirm this hypothesis.

Animais infectados com a cepa Y que tiveram a inibi??o simult?nea de ROS e NO apresentaram mortalidade inicial precoce (in?cio da mortalidade no 6? D.A.I), foi observado 100% de mortalidade em todos os grupos de animais. Camundongos que tiveram a inibi??o de ROS e NO, apresentaram maior produ??o de IFN-? e TNF-? no soro e tamb?m maior express?o de RNAm destas citocinas no tecido card?aco, quando comparado ao grupo controle. O excesso na produ??o de IFN-? e TNF-? pode conduzir a efeitos delet?rios ao hospedeiro. Quanto ? produ??o de IL-17 e IgG total, n?o houve diferen?a entre os grupos de animais que tiveram ROS ou NO bloqueado. A express?o da iNOS foi menor no grupo de animais tratados com os inibidores de ROS e NO, quando comparado ao controle n?o tratado. Os resultados indicam que as esp?cies reativas de oxig?nio participam da regula??o imunol?gica durante a infec??o experimental pelo T. cruzi, evitando mortalidade precoce ao hospedeiro.

A avalia??o morfol?gica evidenciou que os caracteres sexuais secund?rios apresentaram diferen?as discern?veis entre os est?gios ontogen?ticos intra e interespecificamente, tanto em machos quanto em f?meas. Al?m disso, o estudo sobre o crescimento do petasma em rela??o ao comprimento do cefalot?rax evidenciou que essas estruturas apresentam rela??es de crescimento distintas entre as esp?cies e entre os grupos ontogen?ticos. Existe tamb?m uma diferen?a bem marcada sobre o tamanho da primeira matura??o entre as duas esp?cies mas n?o entre os grupos de L. vannamei de natureza e de viveiro. A Baccharis trimera (Less.) DC, conhecida popularmente como carqueja, ? uma planta medicinal utilizada na medicina tradicional em v?rias partes do Brasil. Para tal, infus?es, decoc??es e tinturas de suas folhas s?o produzidas e utilizadas para o tratamento da obesidade, diabetes, assim como diur?ticos, agentes digestivos, antiinflamat?rios, dentre outros. Neste trabalho, com intuito de respaldar o potencial medicinal da carqueja, extratos de folhas de Baccharis trimera foram obtidos, caracterizados qu?mica e fitoquimicamente e avaliados com rela??o ? sua atividade antioxidante e antiadipog?nica. Foram obtidos tr?s extratos: aquoso (AE), decoco (AE-D) e metan?lico (ME); a partir destes, foram realizados seis diferentes ensaios antioxidantes in vitro: ensaio do radical super?xido e hidroxila, poder redutor, capacidade antioxidante total e quela??o dos ?ons ferro e cobre. Bem como, foi avaliada sua poss?vel atividade antiadipog?nica com os testes de oil red O, glicerol livre e mensura??o de fatores de transcri??o adipog?nicos C/EBP?, C/EBP? e PPAR?. Na caracteriza??o fitoqu?mica, revelou-se a presen?a de flavonoides (?cido clorog?nico e apigenina) e compostos fen?licos nos extratos AE e AE-D. Quanto a atividade antioxidante, verificou-se uma atividade dose-dependente. Em rela??o ? atividade antiadipog?nica, a Baccharis trimera inibiu significantemente tanto a diferencia??o e ac?mulo de gordura nos adip?citos pelo MDA, quanto a express?o dos fatores de transcri??o C/EBP?, C/EBP? e PPAR?, durante a adipog?nese todas em rela??o dose-dependente. Este foi o primeiro trabalho que demonstrou o potencial efeito de extratos de Baccharis trimera na diferencia??o de adip?citos 3T3-L1 em adip?citos. / Obesity is considered an important public health problem, being a risk factor for several chronic diseases such as cardiovascular diseases, hypertension, diabetes and other. Baccharis trimera (Less.) DC (carqueja) is a medicinal plant used in traditional medicine in various parts of Brazil. Infusions, decoctions and tinctures of its leaves are produced and used for the treatment of obesity and diabetes, and also as diuretics, digestive agents, anti-inflammatory drugs, among others. In this work, in order to support the medicinal potential of carqueja, extracts of Baccharis trimera leaves were obtained, characterized chemically and phytochemically and evaluated in relation to their antioxidant and antiadipogenic activities. Three extracts were obtained: aqueous (AE), decoco (AE-D) and methanolic (ME); From these, six different in vitro antioxidant assays were performed: superoxide and hydroxyl radical assay, reducing power, total antioxidant capacity and chelation of iron and copper ions. As well, its possible antiadipogenic activity was evaluated with oil red O, free glycerol and measurement of adipogenic transcription factors C / EBP?, C / EBP? and PPAR?. Phytochemical characterization revealed the presence of flavonoids (chlorogenic acid and apigenin) and phenolic compounds in extracts AE and AE-D. As for the antioxidant activity, a dose-dependent activity was observed. Regarding antiadipogenic activity, Baccharis trimera significantly inhibited the differentiation and accumulation of fat in adipocytes by MDA in a dose-dependent relationship and significantly reduced the expression of the transcription factors C / EBP?, C / EBP? and PPAR?, during adipogenesis in a relation dose-dependent too. This work suggests that Baccharis trimera has an excellent antioxidant activity, preventing oxidative stress, and may contribute to the decrease of adipogenesis, besides having an excellent antiadipogenic activity. This was the first work that demonstrated the potential effect of Baccharis trimera extracts on the differentiation of 3T3-L1 adipocytes into adipocytes.

Hitherto, 137 species were known for the genus, which is cosmopolitan. The Neotropical region was 68 species, most of them in Brazil. Species of Trichomyia were divided according to the number of palpus segments in two groups without taxonomic value. The subgenera currently known are Apotrichomyia Duckhouse, 1978; Dicrotrichomyia Duckhouse, 1978; Dactylotrichomyia Duchouse, 1978; Gondwanotrichomyia Duckhouse, 1980; Septemtrichomyia Bravo, 1999 and Opisthotrichomyia Bravo, 2001, being the last two exclusively Neotropical. In the first part, thesis is focused on ways and solutions of radiation protection and principles of measuring absorbed dose and intensity of radiation and possibilities of sending measured data through sensor networks. In the second part, the complete schematic of measuring module is solved, critical blocks are tested and design of PCB is done. Detection circuit, blocks for optical and acoustic signaling, power management and control of the entire device are designed. Detection circuit uses a GM Tube. Next, main box of whole device is designed and made by 3D printing technology. The next step is putting hardware into operation and writing test firmware included verification of wireless communication through WiFi.

This thesis describes creation of dynamically evolvable node, which will cooperate with other nodes. Group of these nodes will then create a distributed control system. The MQTT protocol is used for communications purposes between individual nodes. As hardware platform is used ESP32 and ESP8266. Whole operating system is written in MicroPython and supports a live uploading of user applications written in the same language. Later in thesis is decribed creation of monitoring node on Raspberry Pi, which control network. Complete system can be then used to control a intelligent house.

A polycrystalline diamond was tested as a bearing material for a tilting pad thrust bearing to be used in an electric submersible pump, which elevates process fluids from the bottom of well bores. The goal of this study was to compare the PCD to a current best of technology, which is stainless steel with an engineering polymer.This study found that PCD can handle larger loads than current technology but is limited in size due to diamond sintering and manufacturing constraints. The maximum size is Ø75mm.
