Pesquisa de Salmonella spp. em lotes de galinhas de postura comercial vacinadas e não vacinadas contra Salmonella Enteritidis /Galdino, Vânia Maria Cristina Alves. January 2010 (has links)
Orientador: Ângelo Berchieri Jnior / Banca: Luiz Augusto do Amaral / Banca: Nilce Maria Soares / Resumo: A presença de Salmonella spp. foi analisada em lotes galinhas para postura comercial de ovos de mesa por meio da pesquisa bacteriológica em amostras de mecônio colhidas das caixas de transporte de aves para reposição, de um dia de vida, e em fezes frescas cecais e ovos de oito lotes de aves poedeiras comerciais adultas, em período de produção, quatro vacinados e quatro não vacinados com bacterina oleosa contra SE. A colheita das amostras e a rotina bacteriológica seguiram as recomendações de ZANCAN et al., (2000) e de WRAY & DAVIES (1994). Obteve-se o isolamento de Salmonella spp. em 50% dos lotes de aves para reposição avaliados e nas fezes de 25% dos oito lotes de aves adultas. Não houve isolamento do agente nas amostras de ovos examinados. Foram isolados os sorovares: Salmonella enterica subsp. enterica 4,12:r:-; Salmonella Mbandaka; Salmonella enterica subsp. enterica 6,7:z10:-; Salmonella Enteritidis e Salmonella Havana, que foram associados a surtos de enfermidades transmitidas por alimentos para seres humanos, principalmente pelo consumo de produtos de origem avícola como carne de aves, ovos e seus derivados. Os resultados obtidos neste estudo sugerem que há falhas nos programas de monitoria e biossegurança das granjas matrizes e incubatórios; indica que as galinhas estão infectadas, que há presença do patógeno no ambiente de criação de galinhas para postura comercial de ovos e, sugere que medidas de higiene e biossegurança associadas à vacinação contra SE, pode ter contribuído para a ausência do sorovar em lotes vacinados. / Abstract: The presence of Salmonella spp. was analyzed in flocks of laying hens by means of bacteriological samples taken from meconium in transport box of newly hatched chicks, at the moment they were received in the farms, in feces samples and eggs of eight flocks of adult laying hens, in the production period, that four flocks were vaccinated with oil-emulsion bacterin against SE and four unvaccinated. The sampling and routine bacteriological analysis followed the recommendations of ZANCAN et al., (2000) and WRAY & DAVIES (1994). Salmonella spp. was isolated from 50% of the replacement poultry flocks evaluated, and 25% of the feces samples from eight flocks of adult laying hens. There was no isolation of the agent in samples of eggs assessed. Isolated serovars were: Salmonella enterica subsp. enterica 4,12:r:-; Salmonella Mbandaka; Salmonella enterica subsp. enterica 6,7:z10:-; Salmonella Enteritidis and Salmonella Havana whom have been associated with outbreaks of food-borne diseases to humans, especially by the consumption of products of poultry and poultry meat, eggs and egg products. The result of this study shows that there are flaws in monitoring and biossecurity programs in breeder farms and hatcheries. It demonstrated that there is infection and presence of the pathogen into the environment of commercial laying hens, and suggests that measures of hygiene and biossecurity associated with vaccination against SE, may have contributed to the absence of Salmonella Enteritidis in vaccinated flocks. / Mestre

The object of the thesis is to experimentally find the most suitable method of artificial spawning of pikeprech, using hormonal stimulation of ovulation in females hormonal medication containing HCG. After scraping of the broodstock and after artificial fertilization of eggs obtained experimentally test new methods of artificial stickiness removing of eggs and then evaluate the success of artificial incubation of stripped, fertilized and unstickeness eggs of pikeprech. Experiments that are part of the thesis have been divided into several sub-experiments. In the first experiment was observed effect of different doses of hormonal Chorulon, containing the active substance HCG, the basic indicators of reproductive females. Was evaluated as spawning synchronization, spawning success, the number of spontaneous spawnings, fertility of females and hatching rate of eggs. The used HCG doses were 250, 500, 700 and 1000 IU per kilogram of body weight. All eggs were deprived of stickiness using a mixture of talc and milk. In the second experiment was carried out comparing different types of removes stickiness solutions. Here was evaluated especially time and labor intensity and the influence of various solutions on fertilization and hatching rate of eggs. In this trial was all pikeperch females injected the same amount of HCG hormone 500 IU per kilogram of body weight. Tested solutions was a mixture of talc and milk, just eggs wash water and the solution of various concentrations alkalase. The amount alkalase was 0,5; 1; 1,5; 2 and 5 ml, which are mixed with distilled water, so that the total volume of the solution was 1000 ml. The third experiment was supposed to simulate the pilot plant mass propagation of pikeperch females. All females received the same amount of hormone 500 IU of HCG per kilogram of body weight. All eggs were deprived of stickiness using a mixture of talc and milk. In the last experiment was one of the methods tested, which could limit the number of spontaneous spawning during the artificial spawning of pikeperch. To prevent spontaneous spawning were selected females (all were injected with 500 IU of HCG per kilogram of body weight) sewing genitourinary papilla. The incubation of eggs collected during all the experiments were used Zugské bottle.

This thesis focused on the dynamics of the sexual reproduction of microcrustacean Daphnia (Cladocera, Anomopoda). Sex in Daphnia is induced environmentally after rounds of asexual parthenogenesis in favorable conditions. The onset of sexual reproduction and the occurrence of sexual individuals was investigated with respect to their distribution in the water column and clonal structure. The produced sexual eggs were genotyped and their density and distribution in sediment were analyzed. The impact of massive floods was investigated. The role of sexual reproduction in stable habitats and its relevance and importance for Daphnia communities is further discussed.

When hormonally induced artificial spawning of african catfish (Clarias gariepinus), was several female injected intraperitoneally in one dose preparation Ovopel at doses of 1.5 pellet × kg-1. Females were kept separately in the tanks at a temperature of 21.5 °C. All females were spawned at the same time latency 19.2 hours. Eggs from three spawned females were mixed and divided into 6 doses. Each batch was placed into thermoboxes at temperature 5 °C, 10 °C, 15 °C, 20 °C, 25 °C and 30 °C. These eggs were stored in thermoboxes and after times of storage 0.5 h, 1 h, 1.5 h, 2 h, 3 h, 4 h, 6 h, 8 h, 10h, part of the eggs (approximately 50 to 100 pieces) were taken out from each thermoboxes in three replications and was placed into individuals cups and fertilized by adding 5 drops of sperm and 20 ml of water. In these samples were subsequently observed fertilization, hatching rate and survival rate. When watching fertilization was in individual temperature the highest values and also statistically non-significant difference ( = 0.05) achieved: at 5 °C in times of fertilization 0.5 2 hrs. (61.6 +- 5.81 % - 47.7 +- 1.48 %), at 10 °C in times 0.5 - 1.5 hrs. (70 +- 6.7 % - 62.1 +- 8.9 %), at 15 °C in times 0.5 - 3 hrs. (59.6 +- 9.4 % - 59.6 +- 2.9 %), at 20 °C in times 0.5 - 3 hrs. Previous issue date: 2016-03-26 / Conselho Nacional de Desenvolvimento Científico e Tecnológico (CNPq) / Planícies de inundação são áreas úmidas onde podem ser encontradas as lagoas marginais. Devido ao período de seca e a sua baixa profundidade, estas lagoas podem apresentar caráter temporário com ocorrência sazonal. Tal perturbação pode levar algumas populações zooplanctônicas, como as de cladóceros, a entrarem em estado de dormência, produzindo os ovos de resistência que se depositam no sedimento seco formando um banco de ovos. Quando as lagoas voltam a encher, a comunidade de cladóceros rapidamente se reestabelece, provavelmente como resultado da eclosão dos ovos presentes no sedimento. Detectar espécies de cladóceros que produzem os ovos de resistência como estratégia para recolonização, bem como os fatores que influenciam a eclosão dos ovos, pode auxiliar na compreensão do papel do banco de ovos como fonte de biodiversidade zooplanctônica em lagoas marginais. O objetivo neste estudo foi verificar o potencial de cladóceros para recolonização de lagoas marginais temporárias, por meio da eclosão de ovos de resistência em condições experimentais específicas. A comunidade zooplanctônica foi amostrada em quatro lagoas marginais localizadas na planície de inundação do Rio Turvo. As amostragens dos cladóceros planctônicos foram feitas com rede de plâncton de 50 μm de malha, antes e após um período de seca. Além disso, também foram medidos parâmetros físicos e químicos: profundidade, transparência, temperatura, pH, condutividade, OD, clorofila e material em suspensão. Para a realização dos experimentos de eclosão dos ovos de resistência em laboratório, amostras do sedimento das lagoas foram obtidas com Corer (4,5 cm de diâmetro), durante a estação seca. Duas metodologias foram testadas para indicar qual a mais eficaz para a eclosão: 1. Tratamento ambiente: frascos com sedimento e água do rio Turvo mantidos em condições ambiente; 2. Tratamento câmara: frascos com sedimento e água do rio Turvo mantidos em condições controladas em câmara de germinação (23ºC e fotoperíodo 12:12). A cada dois dias cada réplica foi analisada quanto aos parâmetros físicos e químicos e para a verificação de cladóceros recém eclodidos. Após duas semanas estes procedimentos foram realizados semanalmente até completar 60 dias. Uma porção do sedimento foi utilizada para a caracterização quanto à granulometria. Antes da seca 23 espécies estavam presentes nas lagoas estudadas, após a seca 16 espécies. Durante o experimento de eclosão o tratamento Câmara obteve 431 eclosões enquanto o tratamento Ambiente obteve 118. Houve a eclosão das espécies Alona glabra, Ephemeroporus hybridus, Ilyocryptus spinifer, Leydigiops ornata, Notoalona sculpta e Oxyurella longicaudis, destas eclodiram 6 e 4 espécies no tratamento Ambiente e Câmara, respectivamente. No tratamento Câmara, após 7 dias, E. hybridus foi a primeira espécie a eclodir, e no tratamento Ambiente após 9 dias eclodiram as espécies I. spinifer e N. sculpta. Com a análise da partição hierárquica verificou-se que no tratamento Câmara a condutividade e o pH apresentaram uma correlação positiva com a eclosão dos ovos de resistência e o oxigênio dissolvido uma correlação negativa. No tratamento Ambiente, os parâmetros condutividade e oxigênio dissolvido apresentaram uma correlação negativa com a eclosão dos ovos. Em conclusão pode-se apontar que as espécies da família Chydoridae e Ilyocryptidae, que são organismos bentônicos ou litorâneos, apresentam um maior potencial para rapidamente eclodir em ambientes lacustres temporários. / Floodplains are wetlands where the oxbow lakes can be found. Due of the dry season and its
shallow depth, these lagoons can present seasonal occurrence. Such disruption can lead some
zooplankton populations, such as cladocerans, to enter in dormancy, producing resting eggs that
are deposited in dry sediment forming a bank of eggs. When the lagoons back to fill the
cladoceran community quickly restores, probably as a result of the hatch of eggs present in the
sediment. To detect species of cladocerans that produce resting eggs as a strategy for
recolonization, as well as the factors influencing the eggs hatch, may help understanding the egg
bank's role as zooplankton biodiversity source in lagoons. Our goal in this study was to
investigate the potential of cladocerans to recolonize of temporary lagoons, through the hatching
of resting eggs in specific experimental conditions. The zooplankton community was sampled in
four lagoons located in the Turvo River floodplain. Sampling of planktonic cladocerans were
taken by a plankton 50 µm mesh net before and after the drying lagoons. Physical and chemical
parameters as depth, transparency, temperature, pH, conductivity, OD, chlorophyll and
suspended material were also measured. For the hatching experiments of resting eggs in
laboratory, sediment from lagoons were sampled using a corer (4.5 cm diameter), during the dry
season. Two methods were tested to indicate the most effective for the hatching: 1. Environment
Treatment: bottles with sediment and water from the Turvo river kept at ambient conditions; 2.
Chamber Treatment: bottles with sediment and water from the Turvo river kept under controlled
conditions in a growth chamber (23 ° C and photoperiod 12:12). Every two days each replica
was analyzed for physical and chemical parameters and the verification of cladocerans newly
hatched. After two weeks these procedures were performed weekly to complete 60 days. A
portion of the sediment was used for particle size characterization. Before the drought 23 species
were present in the studied lagoons, after drought 16 species. During the hatching experiment
Chamber Treatment had 431 hatching while Environment Treatment had 118. Six species
hatched: Alona glabra, Ephemeroporus hybridus, Ilyocryptus spinifer, Leydigiops ornata and
Notoalona sculpta and Oxyurella longicaudis, being four species in the Chamber Treatment and
six species in the Treatment Environment. In the Chamber Treatment, after 7 days, E. hybridus
was the first species to hatch, and after 9 days Environment Treatment N. sculpta and I. spinifer
hatched. With the analysis of hierarchical partition we found that in the Chamber Treatment
conductivity and pH showed a positive correlation and dissolved oxygen a negative correlation
with the hatching of resting eggs. In the Environment Treatment, conductivity and dissolved
oxygen showed a negative correlation with the eggs hatch. In conclusion we can point that
species of Chydoridae and Ilyocryptidae family, which are benthic or littoral organisms have a
greater potential to quickly hatch into temporary lacustrine environments.

Previous issue date: 2016-03-03 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior (CAPES) / Fundação de Amparo à Pesquisa do Estado de São Paulo (FAPESP) / O objetivo deste trabalho foi avaliar a viabilidade técnica e econômica da utilização de diferentes formas de cálculo e valores de balanço eletrolítico na formulação de rações para poedeiras comerciais no segundo ciclo de produção. Foram utilizadas 216 poedeiras comerciais leves, da linhagem Lohmann, em segundo ciclo de produção, distribuídas em um delineamento inteiramente casualizado em esquema fatorial (2 x 4 + 1) com parcela subdividida no tempo. Duas fórmulas de cálculo do Balanço Eletrolítico (Mongin = [Na+ + K+] – [Cl-] e Ender = [Na+ + K+] - [Cl- + S2-]) e quatro valores de Balanço Eletrolítico (170, 220, 270 e 320 mEq.kg-1) e uma dieta testemunha (sem fórmula ou valor para balanço eletrolítico) perfazendo um total de nove tratamentos com quatro repetições de seis aves cada, dividido em quatro ciclos, sendo que cada ciclo foi composto por 28 dias. Foram analisados parâmetros de desempenho das aves, qualidade dos ovos e qualidade dos ovos na vida de prateleira. As diferenças entre os tratamentos compostos pelas formulas e balanços eletrolíticos e o testemunha foram pouco consistentes sobre o desempenho ou qualidade dos ovo quando comparada a uma ração formulada sem esse conceito, porém a combinação da fórmula de Ender e do balanço eletrolítico de 270 mEq.kg-1 determinou maior retorno econômico. Na fórmula de Mongin o balanço eletrolítico determinou um comportamento quadrático para consumo de ração, massa de ovos, unidade Haugh de ovos frescos e estocados, indicando melhores resultados na faixa de 241,84 mEq.kg-1, porém o BE 170 mEq.kg-1 favoreceu a qualidade de ovos frescos ou armazenados. Para fórmula de Ender, o aumento no balanço eletrolítico determinou diminuição linear no consumo de ração, produção e massa do ovo e aumento linear na unidade Haugh de ovos frescos e armazenados. No balanço eletrolítico de 270 mEq.kg-1 o uso da fórmula de Ender se mostrou eficiente, conferindo melhor conversão alimentar e maior gravidade específica, além de proporcionar um maior retorno econômico. O aumento nos níveis de balanço eletrolítico promoveu maior excreção de água e potássio. / The objective of this study was to evaluate the technical and economic viability of using different forms of calculation and electrolyte balance in feed formulation for laying hens in the second production cycle. They were used 216 light commercial layers, Lohmann, second production cycle, distributed in a completely randomized design in a factorial scheme (2 x 4 + 1) with split-plot in time. Two calculation formulas of Electrolyte Balance (Mongin = [Na+ + K +] - [Cl] and Ender = [Na+ + K+] - [Cl- + S2-]) and four Electrolyte balance values (170, 220, 270 and 320 mEq.kg-1) and a control diet (no formula or value for electrolyte balance) for a total of nine treatments with four replicates of six birds each, divided into four cycles, each cycle consisted of 28 days. Performance parameters were analyzed for hens, egg quality and egg quality in shelf life. The differences between the treatments comprise the formulas and electrolyte balance and the witness were inconsistent about the performance or quality of the egg compared to a formulated diet without this concept, but the combination of Ender's formula and electrolyte balance 270 mEq.kg-1 determined a higher economic return. In Mongin’s formula the electrolyte balance determined a quadratic behavior for feed intake, egg mass, Haugh unit of fresh and stored eggs, indicating better results in the range of 241.84 mEq.kg-1, but electrolyte balance 170 mEq.kg-1 promoteted the quality of fresh or stored eggs. To Ender's formula, the increase in electrolyte balance determined linear decrease in feed intake, production and mass of the egg and linear increase in the Haugh unit of fresh and stored eggs. In the electrolyte balance of 270 mEq.kg-1 the use of Ender's formula proved to be efficient, providing better feed conversion ratio and higher specific gravity, in addition to providing greater economic return. Para isto, 360 poedeiras com 72 semanas de idade foram submetidas a muda forçada pelo método convencional. Após o retorno à produção de ovos (83 semanas de idade), as aves foram selecionadas e alojadas de acordo com um delineamento inteiramente casualizado com seis tratamentos e seis repetições de 10 aves cada uma. O tratamento controle foi constituído de suplementação à dieta de 0,10 % de uma fonte de microminerais inorgânicos, a qual foi substituída proporcionalmente por cinco níveis (110, 100, 90, 80 e 70%) de um complexo de microminerais com 30; 30; 40; 6; 0,61 e 0,3 g/kg do produto de Zn, Fe, Mn, Cu, I e Se, respectivamente. Todas as dietas eram isoprotéicas, isoenergéticas e isoaminoacídicas. Os resultados de desempenho (peso corporal, % de produção de ovos, massa de ovos, consumo de ração e conversão alimentar), incidência de mortalidade e as características ósseas (resistência à quebra, % de cinzas, % de Ca e % de P das tíbias) foram avaliados por ANOVA e teste de Tukey a 5% de significância. Não foi observado nenhum efeito dos tratamentos sobre o desempenho, mortalidade ou características ósseas das poedeiras. Os resultados obtidos sugerem que o nível de 70% de microminerais orgânicos pode substituir 100% de microminerais inorgânicos suplementado na dieta de poedeiras semi-pesadas no segundo ciclo produtivo sem alterar a produtividade e características ósseas. / The study aimed to evaluate post-induced molting performance and bone mineralization of semi-heavy laying hens submitted to diets with different sources and levels of microminerals. 360 layers aged 72-w-old were subjected to conventional induced molting. After returning to production (83-w-old), the layers were selected and allotted in a completely randomized design of six treatments and six replicates of ten birds each one. Control treatment consisted of 0.10% inclusion of inorganic micromineral (IM) premix (100% of IM) to diet (control treatment), which was proportionally replaced with five levels (110, 100, 90 , 80 and 70%) of organic micromineral complex (OM) composed of 30; 30; 40; 6; 0.61; and 0.3 g/kg of Zn, Fe, Mn, Cu, I, and Se, respectively. All diets were isoproteic, isoenergetic and isoaminoacidic. Layer performance (body weight, % production and mass of eggs, feed intake, and feed conversion), % mortality, and bone characteristics (breaking strength, % ash, % Ca, and % P of tibiae) results were tested by ANOVA and Tukey’s test at 5% of significance. No effect of any treatment upon performance, mortality and bone characteristics of the semi-heavy layers were here observed. Results indicate that the lowest level of OM supplementation (70% of IM level) can replace 100% of IM in the diet of semi-heavy laying hens in the second cycle of egg production without any negative effect upon productive and bone characteristics.

Vitamina E em dietas para reprodutoras de tilápia-do-nilo (Oreochromis niloticus) /Nascimento, Thálita Stefann Ribeiro. January 2010 (has links)
Resumo: A vitamina E funciona como um antioxidante biológico, protegendo os compostos oxidáveis do citoplasma celular, evitando a formação de lipoperóxidos tóxicos. Em fêmeas sua carência determina parada de crescimento do feto, sua retenção e absorção, com morte do embrião em virtude de distúrbios nas produções mesodérmicas que interrompem a circulação vitelina. Este estudo teve como objetivo verificar o efeito da vitamina E sobre as respostas reprodutivas de fêmeas de tilápia-do-nilo (Oreochromis niloticus). Para isso, dietas isoprotéicas (32% PD) e isoenergéticas (3100 kcal kg-1 ED) contendo diferentes níveis de suplementação de vitamina E (200, 300, 400 e 500 mg. Kg-1), foram fornecidas para os grupos de fêmeas (peso médio de 659 ± 121g) por um período de 90 dias. A suplementação de 500 mg influenciou positivamente o volume e o número de ovos produzidos, o índice de desova e a fecundidade relativa e total das fêmeas. Já níveis de suplementação entre 300 e 400 mg afetaram significativamente a taxa de eclosão dos ovos, a sobrevivência e o número de larvas produzidas. A freqüência reprodutiva das fêmeas, taxa de fertilização, bem como o peso e diâmetro dos mesmos não foram influenciados pela suplementação da vitamina nas dietas. O presente estudo mostrou que a vitamina E melhorou o desempenho reprodutivo de fêmeas de tilápia-do-nilo, sendo que o nível de inclusão ideal para essa espécie está entre 300 e 500mg kg-1 da dieta / Abstract: Vitamin E acts as a biological antioxidant, protecting the oxidizable compound the cell cytoplasm, avoiding the formation of toxic lipoperoxide. In females the lack of arrest determines fetal growth, retention and absorption, with embryonic death due to disturbances in the production mesodermal interrupting vitelline circulation. This study aimed to evaluate the effect of vitamin E on reproductive responses of female tilapia (Oreochromis niloticus). To do this, isonitrogenous diets (32% DP) and isocaloric (3100 kcal kg 1-ED) with different levels of vitamin E supplementation (200, 300, 400 and 500 mg. Kg-1), were provided to groups of females (average weight 659 ± 121g) for a period of 90 days. The supplementation of 500 mg has positively influenced the volume and number of eggs produced, the index of spawning and the relative fecundity of females and total. Since levels of supplementation between 300 and 400 mg significantly affected the rate of egg hatching, survival and number of larvae produced. The frequency of reproductive females, fertilization rate, and the weight and diameter of them were not influenced by supplementation of vitamin in the diet. As aves foram alojadas em uma câmara bioclimática com controle automático de temperatura, contendo duas baterias compostas de cinco andares e dez gaiolas, com capacidade para 24 aves cada gaiola, divididas em dois grupos: 240 aves no grupo controle (21°C) e 240 aves no grupo experimental, com temperaturas contínuas de 24, 27, 30, 33 e 36°C em períodos de 14 dias. O período experimental, com duração de 105 dias, foi dividido em cinco ciclos de 21 dias (um ciclo para cada temperatura), sendo 14 dias na temperatura teste e sete dias na temperatura termoneutra. No final de cada ciclo experimental foi avaliado o desempenho e a qualidade dos ovos. Para todos os tratamentos, as rações foram isonutritivas e isocalóricas. Para a avaliação do desempenho e da qualidade dos ovos utilizou-se um delineamento inteiramente casualisado com dois tratamentos (temperatura termoneutra e temperatura teste) e dez repetições de 24 aves por parcela. A elevação da temperatura piora o consumo de ração, reduz o peso dos ovos, a produção e a massa de ovos. Em temperaturas superiores a 27°C houve piora na qualidade interna e externa dos ovos. / In order to evaluate the effect of temperature on performance and egg quality of Japanese quails was conducted an experiment using 480 birds in the post-peak production. The birds were housed in a climate chamber with automatic temperature control, with two batteries composed of five floors and ten cages for up to 24 birds in each cage, divided into two groups: 240 birds in the control group (21oC) and 240 birds in the experimental group, which temperatures of 24, 27, 30, 33 and 36°C. The experiment, lasting 105 days, was divided into five 21-day cycles (one cycle for each temperature), and 14 days at test and seven days a thermoneutral. At the end of each experimental cycle was evaluated performance and egg quality. For all treatments, the diets were isocaloric and isonutritives. For the evaluation of performance and quality of eggs used a completely randomized design with two treatments (temperature and thermoneutral temperature test) and ten replicates of 24 birds per plot. Elevated temperature worsens the feed intake, egg weight, production and egg mass. At temperatures above 27°C worsened the internal and external quality of eggs.
