THIOL-NORBORNENE HYDROGELS WITH TUNABLE MECHANICAL PROPERTIES FOR ENGINEERED EXTRACELLULAR MATRICES

The extracellular matrix (ECM) governs many cellular processes through biochemical and mechanical cues. Particularly, the effect ECM mechanical properties on cells fate has been well established over the years. Many hydrogel systems have been used to mimic the dynamic stiffening processes occurring in ECM. However, changes in ECM stiffness does not fully recapitulate the mechanics of native ECM, as viscoelasticity is also a major factor contributing to ECM dynamic property. This thesis describes the design and characterization of an enzyme-crosslinked hydrogel system that is not only capable of being stiffened on demand, but also can be tuned to obtain viscoelasticity. The first objective of this thesis was to utilize horseradish peroxidase (HRP) to crosslink thiol-norbornene hydrogel and use mushroom tyrosinase (MT) to create secondary DOPA-dimer crosslinks that stiffened the hydrogel. The cytocompatibility of HRP-mediated thiol-norbornene gelation and the effect of stiffening on cell fate was evaluated. The second objective of this thesis represented the first step towards developing a hydrogel system whose viscoelasticity could be dynamically tuned. Thiol-norbornene hydrogel was designed to yield dynamically adaptable boronic ester bonds via partial enzymatic reaction. Thiol-norborne hydrogel was made to contain hydroxyl phenol as well as boronic acid residues within its network. MT, in this case was used to oxidize the hydroxy phenol moieties into DOPA, which then complexed with boronic acid, created dynamic bonds, introducing viscoelasticity to an initial elastic hydrogel.

Estudo da adição aldólica do éster terc-butílico da n-(difenilmetileno)glicina a alguns aldeídos aromáticos / Study aldol addition of the tert-butyl ester of n- (diphenylmethylene)glycine to some aromatic aldehydes

As reações de adição aldólica entre a cetimina 1 e aldeídos aromáticos foram inicialmente efetuadas à temperatura ambiente, em sistema bifásico constituído por uma fase aquosa básica (KOH 10% ou NaOH 5% m/v) e por uma fase orgânica (aldeído), na ausência de solventes e de catalisadores, observando-se baixa conversão em produto. Porém, quando se utilizou o catalisador aliquat®-336, foi possível reduzir a concentração da base (NaOH 1%), com conversão total da imina em produto que, na maioria dos casos, era uma mistura de duas oxazolidinas isoméricas de estereoquímica cis e trans. Esses compostos puderam ser isolados e purificados por recristalização de etanol ou metanol. Em todas as reações efetuadas com benzaldeído, m-clorobenzaldeído e p-nitrobenzaldeído, não se observou excesso diastereomérico significativo. No entanto, as reações com p-clorobenzaldeído mostraram-se diastereosseletivas, conduzindo, à temperatura ambiente, quase que exclusivamente à oxazolidina de estereoquímica cis. A comparação entre o resultado de reações efetuadas a curto e longo tempo de reação, ou em diferentes temperaturas, permitiu concluir que o aldol de estereoquímica anti é o produto cinético, o qual se transforma lentamente na oxazolidina cis. O produto termodinâmico (aldol syn) cicliza rapidamente, não sendo observado nos espectros de RMN de H dos produtos brutos de reação, mas sim seu produto ciclizado, a oxazolidina trans. Tentativas de obter os produtos de reação com excesso enantiomérico, pelo emprego de catalisadores de transferência de fase assimétricos, não foram bem sucedidas. / The aldol addition reactions of ketimine 1 with four aromatic aldehydes could be performed at room temperature, in the presence of an alkaline aqueous solution (KOH 10% or NaOH 5% m/v), and in the absence of organic solvents or catalysts. Under this conditions, conversion to product was often incomplete. However, when the reaction was performed in the presence of aliquat®-336, the base concentration could be reduced (1% m/v), and the ketimine was completely converted into a mixture of diastereomeric oxazolidines, that could be isolated and purified by crystallization from ethanol or methanol. Although no significative diastereomeric excess was observed for the reactions of 1 with benzaldehyde, m-chlorobenzaldehyde or p-nitrobenzaldehyde, for reactions with p-chlorobenzaldehyde the cis oxazolidine was almost exclusively formed. The comparison of diastereomeric ratio at shorter or longer reaction times seems to indicate that the aldol anti adduct is the kinetic product and that cyclization to the corresponding cis oxazolidine is slower than the retro-aldol reaction. On the other hand, in lieu of the thermodynamic product (aldol syn), only the corresponding cyclized product (oxazolidine trans) could be visualized in the H NMR spectra of the crude product, as a result of a fast cyclization step. Attempts to produce enantiomeric enriched oxazolidines by performing the aldol reactions in the presence of chiral catalysts were unsuccessful.

Estudo de aplicação de lipases e transaminases como catalisadores na síntese de compostos organoborônicos quirais / Study of application of lipases and transaminases as catalysts in the synthesis of chiral organoboron compounds

Neste trabalho, foi avaliada a resolução cinética de ésteres carboxílicos β-borilados, catalisada por lipases, através de hidrólise e transesterificação. Foram sintentizados ésteres contendo diferentes cadeias carbônicas. A hidrólise enantiosseletiva catalisada por CAL-B forneceu o correspondente ácido carboxílico β-borilado com excesso enantiomérico moderado (e.e. 62%), enquanto os ésteres β-borilados metílico e etílico foram recuperados com excelente pureza enantiomérica (e.e.>99%). A reação de transesterificação enantiosseletiva entre ésteres carboxílicos β-borilados e álcoois, catalisada por CAL-B, demonstrou uma alta seletividade somente quando etanol e (β-pinacolilboronato)-butanoato de metila foram usados como substratos, levando a formação de (β-pinacolilboronato)-butanoato de etila enantioenriquecido (e.e.>99%). Entretanto, ésteres contendo longas cadeias carbônicas ligadas à carboxila não sofreram as reações desejadas, mesmo em temperaturas mais elevadas. Adicionalmente, foi investigada a reação de aminação redutiva assimétrica, catalisada por ω-transaminases, de cetonas pró-quirais contendo grupos borônicos (ácido, éster ou trifluoroborato de potássio). Após as sínteses de cetonas aromáticas e alifáticas, os melhores resultados para a biotransformação foram obtidos com ω-transaminases (R)-seletivas de Arthrobacter sp. e Aspergillus terreus e (S)-seletiva de Chromobacterium violaceum. Sob as condições otimizadas, foram obtidas altas conversões (94%) com perfeitas enantiosseletividades (e.e.>99%), proporcionando assim uma nova via de acesso a aminas (R) ou (S) contendo boro em condições reacionais suaves. / In this work, the kinetic resolution of β-borylated carboxylic esters via lipase-catalyzed hydrolysis and transesterification reactions was evaluated. Esteres with different carbon chains were synthesized. The enantioselective hydrolysis catalyzed by CAL-B furnished the corresponding β-borylated carboxylic acid with reasonable enantiomeric excess (ee 62%), while both methyl and ethyl β-borylated carboxylic esters were recovered with excellent enantiomeric purity (ee>99%). The enantioselective transesterification reaction of β-borylated carboxylic esters and several alcohols, catalyzed by CAL-B, only indicated a high selectivity when ethanol and methyl-(β-pinacolylboronate)-butanoate were used as substrates, which gave the enantioenriched ethyl-(β-pinacolylboronate)-butanoate (ee >99%). However, esters with longer carbon chains or steric hindrance at the carboxyl group did not undergo the desired reactions, even at high temperatures. Additionally, the asymmetric reductive amination ω-transaminase catalyzed of boron-containing (acid, ester or potassium trifluoroborate) prochiral ketones was investigated. After the syntheses of aromatic and aliphatic ketones, the best results of biotransformation were obtained with (R)-selective ω-transaminases from Arthrobacter sp. and Aspergillus terreus and (S)-selective one from Chromobacterium violaceum. Under the optimized conditions, high conversions (94%) with perfect enantioselectivities (ee >99%) were found, providing a new access to (R) or (S) boron-containing amines under mild reaction conditions.

Desenvolvimento de procedimentos analíticos rápidos para o controle de qualidade de biodiesel / Fast analytical procedures for biodiesel quality control

A presente dissertação apresenta os resultados referentes ao desenvolvimento de procedimentos analíticos para a determinação de índice de iodo em biodiesel e óleo vegetal e do teor de éster em biodiesel e em misturas biodiesel:diesel. O primeiro procedimento para a determinação de índice de iodo foi baseado na descoloração de uma solução de I3- monitorada por espectrofotometria. Para amostras de biodiesel e óleo vegetal, foram obtidas respostas lineares nos intervalos de 10-106 g I2/100 g e 20-144 g I2/100 g, respectivamente. O coeficiente de variação (n = 10) e o limite de detecção (99,7% nível de confiança) foram estimados em 5,0% e 2,5 g I2/100 g para amostras de biodiesel, enquanto para amostras de óleos vegetais os valores foram 3,0% e 7 g I2/100 g, respectivamente. Foram consumidos ca. 1,2 mL de amostra, 0,365 mg de I2 e 40 mg de KI, e gerado ca. 2,2 mL de resíduo por determinação. Efeitos de matriz foram constatados e superados por calibração com compatibilização de matriz. Alternativamente, foi desenvolvimento um spot test utilizando imagens digitais, também baseado no consumo de I2 pelos compostos insaturados presentes na amostra, porém envolvendo a deposição de alíquotas de reagente em papel de filtro e a medida do I2 remanescente, na forma do complexo com amido. As medidas foram realizadas com a câmera de um celular e tratadas como reflectância. Resposta linear foi obtida entre 10-106 g I2/100 g de biodiesel, com coeficiente de variação (n = 10) e limite de detecção (99,7% nível de confiança) estimados em 5,0% e 8 g I2/100 g, respectivamente. Foram consumidos 40 ?L de amostra e 50 ?g de I2 e gerado 65 ?L de resíduo por determinação. A determinação do teor de éster baseou-se na formação do complexo violeta entre Fe(III) e hidroxamato, gerado pela reação entre os ésteres de alquila da amostra e hidroxilamina. Foi utilizado um sistema de análises de fluxo, explorando o processo lab-in-syringe, com etanol como solvente mediador para gerar uma única fase entre a amostra hidrofóbica e os reagentes hidrofílicos. Respostas lineares foram obtidas nos intervalos 4-99% (v/v) e 2-40% (v/v) para biodiesel e mistura biodiesel:diesel, respectivamente. Para as amostras de biodiesel, o coeficiente de variação, o limite de detecção e a frequência de amostragem foram estimados em 0,80% (n = 10), 0,36% e 15 h-1, respectivamente, enquanto para a mistura biodiesel:diesel foram 0,20%, 0,03% (v/v) e 12 h-1, respectivamente. Foram consumidos 40 ?L de amostra de biodiesel e 100 ?L da mistura biodiesel:diesel, 0,860 mg de hidroxilamina, 0,145 mg Fe e gerado ca. 3 mL de resíduo. Os resultados obtidos em todos os procedimentos foram concordantes com os obtidos em procedimentos de referência com 95% de confiança. Alternatively, a spot test using digital images was developed, also based on consumption of iodine by unsaturated compounds in the samples, but relying on deposition of reagent aliquots on a filter paper followed by measurement of remaining iodine, through its complex formed with starch. The measurements were performed with a cell phone camera and treated as reflectance. A linear response was obtained within 10 and 106 g I2/100 g of biodiesel. The coefficient of variation (n = 10) and limit of detection were (99.7% confidence level) estimated at 4.9% and 8 g I2/100 g, respectively. About 40 ?L of sample and 50 ?g of I2 were consumed per determination, with generation of ca. 65 ?L of waste per determination. Ester determination was based on reaction of alkyl esters from biodiesel and hydroxylamine, yielding hydroxamate, which was measured by spectrophotometry as an iron(III) complex. The flow-based procedure exploited the lab-in-syringe approach, by using ethanol as mediator solvent to generate a single phase between the hydrophobic sample and the hydrophilic reagents. Linear responses were obtained from 4 to 99% (v/v) and 2 to 40% (v/v) for biodiesel and biodiesel:diesel blends, respectively. For biodiesel samples the coefficient of variation (n = 10), detection limit (99.7% confidence level) and sampling rate were estimated at 0.80%, 0.36%, and 15 h-1, respectively, whereas for biodiesel:diesel blends, the corresponding values were 0.20%, 0.03% (v/v), and 12 h-1, respectively. About 40 µL of biodiesel or 100 µL biodiesel:diesel blends, 0.860 mg of hydroxylamine, and 0.145 mg Fe were consumed per determination, with generation of ca. 3 mL of waste. The results obtained by all the proposed procedures agreed with the reference ones at the 95% confidence level. The proposed procedures are practical and feasible for on-line or at-line analysis, by consuming only low amounts of organic solvents and reagents, and generating low waste volumes

Among the many chemicals that have been identified as mutagenic action, include the Androgenic Steroids (AAS), hormones widely used in the search for improved physical performance and increased muscle mass. Objective: In this context, the aim of the development of this study was to evaluate the potential of androgenic anabolic steroid nandrolone decanoate, testosterone propionate and testosterone cypionate to induce chromosome damage, apoptosis and necrosis, using the micronucleus test in exfoliated cells from the oral mucosa of users of AAS with a view to its application as a tool in cancer prevention. Method: The study sample consisted of 55 volunteers, male, divided into two (02) groups, matched for age: 25 subjects (G1) users of nandrolone decanoate, testosterone propionate and testosterone cypionate (alone or simultaneously ) and 30 subjects in the control group (G2). The collection methodology and cytological analysis followed the protocol of Tolbert et al. (1992) and Thomas et al. (2009), which includes, in addition to micronuclei, the computation of degenerative nuclear changes indicative of apoptosis (cariorr?xis, condensed chromatin and pyknosis) and necrosis (karyolysis, cariorr?xis, condensed chromatin and pyknosis). Statistical analysis of the endpoints analyzed (micronucleus, cariorr?xis, condensed chromatin, karyolysis, pyknosis and broken eggs) was performed using the conditional test to compare proportions in situations of rare events. Results: Statistical analysis revealed no significant difference in the occurrence of micronuclei, karyolysis and broken eggs between groups. The occurrence of apoptosis was significantly greater in cells from control individuals. Resultados: A an?lise estat?stica revelou que n?o houve diferen?a significativa na ocorr?ncia de micron?cleo, cari?lise e broken eggs entre os grupos. A ocorr?ncia de apoptose foi, significativamente, maior em c?lulas dos indiv?duos do grupo controle. Conclus?o: Os resultados obtidos mostram inibi??o da apoptose induzida pelo uso de EAA, sugerindo que a associa??o descrita entre uso destas subst?ncias e o processo carcinog?nico possa ser permeada por este mecanismo.

A aplicação de uma ou mais camadas de tinta sobre as superfícies metálicas é a maneira mais comum e eficaz de proteger os substratos metálicos contra o fenômeno da corrosão. No entanto, os sistemas de pintura podem vir a falhar precocemente por diferentes motivos, causando um ataque corrosivo inesperado no metal a ser protegido. Por esta razão, o processo de repintura em estruturas metálicas é realizado frequentemente para garantir a integridade da estrutura pintada e aumentar sua vida útil. Como o processo de repintura gera impactos econômicos e ambientais, sistemas de pintura capazes de sofrerem uma reparação sem a necessidade de uma intervenção humana, precisam ser desenvolvidos. O encapsulamento de agentes de reparação, com propriedades de formação de filme, em microcápsulas poliméricas é uma excelente alternativa para que os sistemas de pintura se autorreparem, aumentando os intervalos de repintura. Após o processo de encapsulamento, as microcápsulas contendo o agente de reparação são incorporadas na preparação da tinta, para que o sistema de pintura seja aplicado sobre a estrutura metálica. Este tipo de aditivação confere ao revestimento a propriedade de autorreparação, pois quando o sistema de pintura é danificado as microcápsulas são rompidas e liberam o agente de reparação no local danificado, protegendo novamente o substrato metálico. Neste trabalho foi desenvolvido um sistema autorreparador monocomponente, através do microencapsulamento de uma resina a base de éster de epóxi, pelo método de polimerização in-situ. Também foi desenvolvido um sistema autorreparador bicomponente, através do microencapsulamento de uma resina a base de epóxi, pelo método de emulsão e polimerização in-situ de ureia-formaldeído-melamina e do seu endurecedor a base de poliamida, pelo método de extração de solvente em paredes de poliestireno. Foi realizado um planejamento estatístico para estudar a emulsão precursora das microcápsulas de poli(ureia-formaldeído-melamina) contendo o sistema monocomponente, onde foram estudados: o tipo e a velocidade de agitação, a presença de cloreto de sódio na formulação, o uso de uma sonda ultrassônica após a etapa de dispersão, a concentração de tensoativo na formulação e o tensoativo utilizado. Como variáveis de resposta foram determinadas: a estabilidade visual das emulsões e o diâmetro das gotículas formadas. A melhor condição de emulsificação determinada foi utilizada para a obtenção das microcápsulas de poli(ureia-formaldeídomelamina) contendo a resina éster de epóxi e das microcápsulas de poli(ureiaformaldeído-melamina) contento a resina epóxi. Entre as condições de emulsificação estudadas, apenas a condição utilizando o tensoativo goma arábica possibilitou a obtenção das microcápsulas de poli(ureia-formaldeído-melamina) na faixa de diâmetro desejada. O método escolhido para o encapsulamento do endurecedor possibilitou a obtenção de microcápsulas de poliestireno, porém com uma baixa capacidade de armazenamento. A liberação dos agentes de reparação encapsulados foi observada pela microscopia óptica e comprovada pela técnica de espectroscopia na região do infravermelho (FTIR) e pela técnica de espectroscopia Raman. Os aditivos autorreparadores desenvolvidos (mono e bicomponente) foram adicionados separadamente em uma tinta epóxi, nas proporções mássicas em base seca de 10 e 15 %. O sistema de pintura foi aplicado em um esquema de três camadas e o aditivo de autorreparação foi incorporado na primeira e/ou segunda camada aplicada. O sistema de pintura contendo o aditivo autorreparador monocomponente apresentou um aspecto visual melhor do que o sistema de pintura contendo o aditivo autorreparador bicomponente, porém o sistema bicomponente forneceu melhores propriedades de aderência, de impermeabilidade, anticorrosivas e de autorreparação à tinta aditivada. As medidas com as técnicas eletroquímicas de espectroscopia de impedância eletroquímica (EIE) e de varredura com eletrodo vibratório (SVET) comprovaram que os dois aditivos desenvolvidos proporcionaram o efeito autorreparador aos sistemas de pintura aditivados, quando estes foram danificados mecanicamente com uma microbroca ou com um estilete. Ensaios acelerados de corrosão em câmara de névoa salina e ensaios de exposição ao intemperismo natural mostraram que os aditivos desenvolvidos promoveram uma proteção adicional ao aço carbono, quando o sistema de pintura foi danificado mecanicamente. / The application of one or more coating layers on the metallic surfaces is the most common and effective way to protect metallic substrates against corrosion. Nevertheless, the coating layer may fail early for different reasons, leading to an unexpected corrosive attack on the protected metal. For this reason, the coating repair process is performed to ensure the integrity during the service life of the coated metallic structures. Due to the fact that coating repair process generates economic and environmental impacts; there is a great need for the development of systems capable to repair themselves, without human intervention. The encapsulation of repairing agents, with film forming properties, in polymeric microcapsules is an excellent alternative to the coating self-repair, decreasing the coating repair process frequency. After the encapsulation process, the microcapsules containing the repair agent are incorporated into the paint preparation and the coating system could be applied normally to the metallic surface. This kind of additivation confers to the coating the self-healing property, because when the coating system is damaged the microcapsules suffers a rupture and release the repair agent into the damaged site, protecting the metallic substrate from corrosion. In this work, a mono-component self-healing system was developed, through the microencapsulation of an epoxy ester resin, by the in-situ polymerization method. A bi-component self-healing system was also developed, by the microencapsulation of an epoxy resin, through the emulsion and in-situ polymerization method and by the microencapsulation of a polyamide hardener, by the double emulsion and solvent extraction method. A factorial design was developed to study the precursor emulsion of the poly (urea-formaldehyde-melamine) microcapsules containing the monocomponent system, where the studied factors were: the type and speed of the agitation, the presence of sodium chloride in the formulation, the use of an ultrasonic probe after the emulsification, the surfactant type and concentration. The analyzed response variables were: the visual stability of the emulsions and the mean diameter of the formed droplets. The best obtained emulsification conditions were employed to produce the poly(urea-formaldehyde-melamine) microcapsules containing the epoxy ester resin and poly(urea-formaldehyde-melamine) microcapsules containing the epoxy resin. Among the studied emulsification conditions, only using arabic gum surfactant the poly (urea-formaldehyde-melamine) microcapsules were obtained. The selected method for the hardener encapsulation was efficient to obtain polystyrene microcapsules, but with low loading capacity. The release of the encapsulated repair agents was observed by optical microscopy and confirmed by infrared spectroscopy (FTIR) technique and Raman spectroscopy technique. The developed self-healing additives (mono and bicomponent) were added separately in an epoxy commercial coating, using the dry mass ratios 10% and 15 %. The coating system was applied in a three layer coating system and the self-healing additive was incorporated into the first and/or second layer. The coated samples containing the mono-component additive had a better visual appearance than the bi-component additive system; nevertheless the bi-component system provided better adhesion, impermeability, anti-corrosion and self-healing properties to the doped coating. The electrochemical impedance spectroscopy (EIS) and scanning vibrating electrode technique (SVET) measurements proved that the two developed additives provided self-healing properties to the doped coating systems, when they were mechanically damaged with a micro drill or a blade. Accelerated corrosion tests in the salt spray chamber and natural atmospheric corrosion tests showed that the developed additives provided an additional protection to the carbon steel, when the coating system has been mechanically damaged.

Caracterização e aplicação de filmes finos de acetato butirato carboximetil celulose / Characterization and application of thin film of carboxymehtylcellulose acetate butyrate

Esta tese apresenta o estudo do efeito do solvente acetato de etila e acetona no comportamento em solução dos polímeros acetato butirato celulose (CAB) e acetato butirato carboximetil celulose (CMCAB) e nas características dos seus filmes finos obtidos pela técnica de revestimento rotacional ou por adsorção. As medidas de viscosidade e espalhamento de raio-X a baixo ângulo (SAXS) mostraram que o acetato de etila é um melhor solvente para CAB e CMCAB do que a acetona. A caracterização dos filmes foi feita através de medidas de elipsometria, microscopia de força atômica (AFM), espectrocospia vibracional por geração de soma de freqüências (SFG) e medidas de ângulo de contato. Os filmes de CMCAB obtidos por revestimento rotacional são mais espessos quando preparado em acetona do que em acetato de etila. Imagens de AFM mostraram que os filmes de CMCAB oriundos de soluções em acetato de etila são mais homogêneos e lisos do que aqueles preparados a partir de acetona. Medidas de SFG comprovaram a forte afinidade da acetona com SiO2/Si, mostrando que esse solvente cria uma nova camada para os filmes de CAB e CMCAB. Os valores de energia superficial calculados para CAB e CMCAB foram semelhantes ~ (49,0 ± 0,5) mJ/m², sendo a contribuição da componente dispersiva maior que a da componente polar. A adsorção das proteínas lisozima, albumina do soro bovino (BSA), concanavalina A e jacalina foram mais pronunciadas sobre os filmes de CMCAB do que sobre CAB. Indicando que a presença do grupo carboximetil (CM) contribui significativamente no processo de adsorção das biomoléculas. O efeito da rugosidade dos filmes de CAB e CMCAB sobre o processo de adsorção das proteínas foi estudado. No caso do CMCAB, a adsorção das proteínas foi mais pronunciada sobre o filme rugoso do que sobre o filme mais liso. Entretanto, para os filmes de CAB a rugosidade não teve um efeito significativo na adsorção das proteínas / The effect of ethyl acetate and acetone on the solution behavior of cellulose acetate butyrate (CAB) and carboxymehtylcellulose acetate butyrate (CMCAB) and on the characteristics of films obtained either by spin coating or adsorption was investigated. Viscosity and small angle X-ray scattering (SAXS) measurements showed that ethyl acetate is a better solvent than acetone for CAB e CMCAB. Films were characterized by means of ellipsometry, atomic force microscopy (AFM), sum frequency generation (SFG) and contact angle measurements. Spin-coated films of CMCAB from ethyl acetate solutions were thicker than those deposited from acetone solutions. AFM images revealed that CMCAB spin coated films from ethyl acetate solutions were homogeneous and flat. However, films obtained from solutions in acetone were very rough. SFG spectra showed that acetone binds strongly to SiO2/Si wafers, creating a new surface for CAB and CMCAB films. Surface energy values determined for spin-coated CAB and CMCAB were similar ~ (49,0 ± 0,5) mJ/m² with the dispersive component larger than the polar component. The adsorption of lysozyme, bovine serum albumin (BSA), concanavalin A and jacalin was more pronounced onto CMCAB films than that onto CAB films. Indicating that carboxymethyl group favored the adsorption process. The influence of surface roughness of CAB and CMCAB on protein adsorption has been investigated. In the case of CMCAB, protein adsorption was morepronounced onto rough films than that onto flat films. However, the roughness of CAB films exerted no significant influence on proteins adsorption

Investigations in amine chemistry: Mn-Mediated radical addition approach toward gamma amino esters and synthetic studies of the tubulysins

Mn-Mediated radical addition has been developed within the Friestad laboratory as a versatile method toward addition to C=N bonds. N-Acylhydrazones generated by condensation between an aldehyde and an N-acylamine serves as the substrate toward radical addition. A bulky directed group attached with the N-acyl moiety and restricted rotation around N-N bond due to a three point chelation with a Lewis acid differentiates the faces of the C=N bond of the N-acylhydrazones. Radical generation initiated by photolysis of Mn2(CO)10 causing homolysis of C-X bond in alkyl halide serves as the radical donor to the N-acylhydrazones. Radical addition thereafter occurs stereoselectively from the less hindered face of the C=N bond of the N-acylhydrazones. The product N-acylhydrazines can be effectively transformed to α-chiral amines. In this thesis, a new protocol toward generation of non-proteogenic γ-amino esters using Mn-mediated radical addition has been described. Moreover, the utility of the Mn-mediated radical addition has been demonstrated through studies toward synthesis of tubulysin U and V. Chapter 3 describes a new strategy for asymmetric synthesis of γ-amino esters starting from non-amino precursors. The α-substituted γ-amino esters are prevalent in drugs, drug candidates, and in peptidomimetics. As a part of progressing the Mn-mediated radical addition reaction, highly stereoselective reactions were devised for addition to N-acylhydrazonoesters in absence of Lewis acid. Spectroscopic investigations were carried out to decipher the Lewis acid chelation of N-acylhydrazones. Finally, a novel microwave mediated trifluoroacylation of N-acylhydrazinoesters facilitated the cleavage of N-N bond to liberate γ-aminoester. Chapter 4 describes application of Mn-mediated radical addition toward synthesis of tubulysin natural products. Tubulysins are natural products, isolated from myxobacteria, that have exhibited potent cytotoxicity toward cancer cells in the picomolar regime. The Mn-mediated radical addition was used to prepare two chiral amine subunits in highly diastereoselective fashion. The subunits were then assembled after required manipulations into the tetrapeptide structure characteristic of tubulysins. This strategy to synthesize tubulysins is the most stereoselective of all efforts toward the synthesis of this molecule. Synthesis toward tubulysin was achieved in 18 steps as the longest linear sequence with a 31% overall yield to tubulysin V in benzyl protected form. Chapter 5 describes a new strategy toward installation of N-hydroxymethyl unit into a peptide chain. N,O-Acetals are acid-base labile species that is present in some tubulysin natural analogs. This new approach exploits Fleming-Tamao oxidation and hence introduce the hydroxymethyl unit of the N,O-acetal in a masked form. Following peptide construction the masked hydroxy group is released to liberate the N-hydroxymethyl moiety. Acylation of the free hydroxy group furnishes the N,O-acetal moiety in a strategy that is potentially applicable toward synthesis of tubulysin D.

1. Improving the Yield of Biodiesel from Microalgae and Other Lipids. 2. Studies of the Wax Ester Biosynthetic Pathway and Potential Biotechnological Application

The production of biofuels and oleochemicals from renewable sources offers an opportunity to reduce our dependence on fossil fuels. The work contained in this dissertation has focused on developing and improving methods for the production of biodiesel from non-traditional feedstocks and understanding biosynthetic pathways that result in the production of oleochemicals and fuels. Pure vegetable oil can account for 70-80% of the total cost of biodiesel production. Many low-cost oils contain high amounts of free fatty acids, which are unsuitable for base-catalyzed transesterification. Herein an approach is described that efficiently accomplishes the simultaneous esterification and transesterification of both free fatty acids and triglycerides found in low-cost oils. The approach utilizes an acid catalyst and longer-chain alcohols to improve biodiesel yields from oils high in free fatty acids. Microalgae are a promising biodiesel feedstock, due to its high lipid productivity and its ability to be cultivated using resources, land and water, unsuitable for agriculture. As part of this work, reaction conditions were optimized for the direct (or in situ) transesterification of algal biomass to biodiesel. This approach accomplishes the simultaneous extraction and conversion of the total lipids from microalgae and results in increased yields compared to extraction followed by conversion. The use of this process to effectively produce biodiesel from wet algal biomass is also discussed. Wax esters are a class of oleochemicals that can be used for a wide range of applications in diverse industries. The chemical composition of native wax esters from the bacterium Marinobacter aquaeolei was determined. It was found that including small alcohols in the growth medium resulted in the in vivo formation of esters similar to biodiesel. All of the proteins involved in the wax ester biosynthetic pathway are not known. The cloning, purification, and characterization of a putative fatty aldehyde reductase from M. aquaeolei, believed to be involved in the production of wax esters, is reported. Finally, the expression of a ws/dgat (wax ester synthase) gene from M. aquaeolei in the cyanobacterium Synechocystis sp. PCC 6803 is discussed as an approach to producing biodiesel in vivo from sunlight and CO2.
