Emprego de reator com membrana na obtenção de frutose e ácido glicônico a partir da sacarose / Use of membrane reactor to obtain fructose and gluconic acid from sucrose

Frutose e Ácido Glicônico são produtos importados empregados em diferentes setores nas áreas química, farmacêutica e alimentícia, representando um mercado de dois milhões de dólares (US$ 2,0 milhões) por ano. Por sua vez, a sacarose pode ser empregada como matéria-prima para a obtenção destes produtos através de conversão enzimátiva empregando invertase e glicose-oxidase. O uso de biorreatores com membrana (MBR) mostra-se interessante em processos enzimáticos, pois, ao serem empregados em processos contínuos permitem, simultaneamente, produção e separação dos produtos, reduzindo a formação de subprodutos e, eventual, inibição da enzima por excesso de substrato ou produtos. A sacarose é convertida em xarope de açúcar invertido (solução equimolar de frutose e glicose) pela invertase (Bioinvert®, enzima comercial), seguido pela oxidação da glicose em ácido glicônico pela ação da glicose oxidase (GO). O processo de conversão multi-enzimático da sacarose foi obtido através da alimentação de sacarose (50 mM) em reator com membrana (MBR) contendo invertase (24 U/mL), glicose-oxidase (0,5 U/mL) e catalase (470 U/mL) e operando com vazão específica de 6,0 h-1, 35ºC e pH 5,5. As condições operacionais otimizadas possibilitaram a conversão completa da sacarose (X = 100 %) e da glicose resultante (Y = 100%) com velocidades específicas de reação de 4,2 mmol/U.h, 0,60 mmol/U.h e 0,00062 mmol/U.h, respectivamente, para a invertase, glicose oxidase e catalase. A respeito da oxidação da glicose, a adição de catalase no meio reacional se fez necessária para minimizar os efeitos inibitórios sobre a GO através do peróxido de hidrogênio formado. / The fructose and gluconic acid are products of great application in chemical, pharmaceutical and food industry. The actual Brazilian market for these compounds is about US$ 2 millions, here as the sucrose, the raw-material used for their production, represents about 2.4% of the Brazil\'s GNP. This conversion increases the value added to the sugarcane, usually marketed as a commodity, because the fructose and gluconic acid are more valuable products than sucrose. The use of membrane bioreactor (MBR), which operates under mild conditions regarding internal pressure, temperature and pH, has been growing along the years for enzyme catalyzed processes. Moreover, in the MBR the reaction and separation of the products occur simultaneously, avoiding the formation of by-products and the eventual inhibition of the enzyme caused by excess of substrate or products. The sucrose is converted to the inverted syrup (an equimolar solution of fructose and glucose) by invertase (in this work was employed Bioinvert®, a commercial invertase) followed by the oxidation of glucose in gluconic acid by the glucose oxidase (GO). The multi-enzymatic conversion of sucrose was attained when carried out under initial substrate of 50mM and invertase, glucose oxidase and catalase concentrations, respectively, of 24.0 U/mL, 0.5 U/mL and 470 U/mL in a membrane reactor utilizing a dilution rate of 6.0 h-1, 35ºC and pH 5.5. The optimized operational conditions led to a conversion yield of 100% for sucrose hydrolysis and glucose oxidation steps resulting in enzyme productivity of 4.2 mmol/U.h, 0.60 mmol/U.h and 0.00062 mmol/U.h, respectively, to invertase, glucose oxidase and catalase. In regard to the glucose oxidation, the addition of catalase in the reaction medium was necessary, in order to minimize the inhibition of the GO by the hydrogen peroxide formed.

Estudo das propriedades do açúcar líquido invertido processado com radiação gama e feixe de elétrons / Study of the properties of inverted liquid sugar processed with gamma radiation and electron beam

O Brasil ocupa posição de destaque no mercado de açúcar a granel e consumo elevado deste ingrediente na forma líquida, em especial pelas indústrias de bebidas e alimentos. O açúcar líquido apresenta vantagens em relação ao açúcar a granel como facilidade do manuseio e dosagem, espaço reduzido para a armazenagem, redução das perdas, custos e mão-de-obra, melhora na sanitização e grande variação possível nas proporções de diferentes misturas de açúcares. O açúcar Uquido invertido é uma solução de sacarose, glicose e frutose em água e recebe esse nome porque durante a sua produção há a inversão do poder óptico de rotação da solução. No processo produtivo do açúcar invertido existem alguns pontos que podem gerar a contaminação por microrganismos, dessa forma torna-se importante o desenvolvimento de técnicas que propiciem a sanitização eficiente desta matéria-prima. Este trabalho estudou duas técnicas alternativas de controle microbiano: a radiação gama com fonte de cobalto-60 e a radiação com feixe de elétrons. O estudo foi conduzido nas doses de 5, 10, 20, 30 e 50 kGy. O teor de sacarose apresentou decréscimo com a radiação indicando que ocorreu a quebra da ligação glicosídica com a formação de glicose e fioitose, gerando o aumento do teor de sólidos solúveis. Ocorreu a formação de compostos ácidos, confirmado pela redução de pH, que foi proporcional ao aumento da dose de radiação. As maiores doses provocaram alterações na cor do xarope pela formação de polímeros com cadeia molecular longa. A viscosidade nas amostras irradiadas com radiação gama aumentou em relação ao controle, indicando a prevalência de polimerização das moléculas, enquanto que na radiação com feixe de elétrons a polimerização foi observada somente na amostra que recebeu a dose de 50 kGy. Na análise sensorial, os julgadores apontaram diferença significativa (5%) no sabor entre o controle e as amostras irradiadas com 5 kGy, nas duas técnicas de processamento. Não foi possível identificar o efeito da radiação gama no controle de bolores e leveduras. / Brazil occupies position of prominence in the sugar market and high consumption of this ingredient in the liquid form, in special for the beverages and food industries. The liquid sugar presents advantages in relation to the bulk granulated system such as easiness of the handling and dosage, reduced space for the storage, reduction of the losses, costs and hand of workmanship, improvement of the sanitizing and great possible variation in the ratios of different mixtures of sugars. The inverted liquid sugar is a solution of sucrose, glucose and fructose in water. This ingredient receives this name because during its production it has an inversion of the optic power of rotation of the solution. In the productive process of the inverted sugar some points occur that can generate contamination by microorganisms, so its become important the development of techniques that propitiate the efficient sanitizing of this raw material. This work aims at to consider two alternative techniques of microbiologic control: gamma radiation with source of cobalto-60 and radiation with electron beam. The study was lead in the doses of 5, 10, 20, 30 and 50 kGy. Concentration of sucrose presented in syrup decreased with the radiation indicating the scission of glicosidic linkage in addition with the glucose and fiaictose formation, generating the increase in solids soluble. The acid compounds formation occurred, confirmed by the reduction of pH, that it was proportional with the increase of the radiation dose. The biggest doses had provoked change of color in the syrup by the long molecular chain polymers formation. Viscosity in the samples processed by gamma radiation increased in relation to the control, indicating the prevalence of polymerization of molecules, whereas in the radiation with electron beam the polymerization was only observed in the sample that received the dose of 50 kGy, In the sensorial analysis, the judges had pointed significant difference (5%) in the flavor among control and the samples radiated with 5 kGy in the two techniques of processing. The alterations provoked for the irradiation had not been enough to provide significant changes in the inverted sugar, even in the highest dose of 50 kGy. It was not possible to identify the effect of gamma radiation in the control of molds and yeasts.

The objectives of this project were to investigate the critical factors impacting the physicochemical and antibacterial properties of β-chitosan based films derived from jumbo squid (Dosidicus gigas) pens, and to evaluate the feasibility of improving water solubility of β-chitosan through Maillard reaction. The studies examined the effect of molecular weight (1,815 and 366 kDa), acid (formic, acetic, propionic, and lactic acid), and plasticizer (glycerol and sorbitol) on the film properties, as well as reducing sugar (fructose and glucosamine) and heat treatment (high temperature short time (HTST), low temperature long time (LTLT)) on water solubility of chitosan. Results on β-chitosan were compared with α-chitosan in both studies. Tensile strength (TS) and elongation (EL) of β-chitosan films were influenced by molecular weight (Mw), acid and plasticizer types (P < 0.05). High molecular weight (Hw) β-chitosan films had an overall TS of 44 MPa, 53% higher than that of low molecular weight (Lw) β-chitosan films (29 MPa) across all acid types used. The mean TS of β-chitosan acetate and propionate films (43 and 39 MPa) were higher (P < 0.05) than that of β-chitosan formate and lactate films (34 and 29 MPa). Films incorporated with plasticizer (32 MPa) had lower TS than those without plasticizer (48 MPa). Mean EL of Hw β-chitosan films was 10% versus approximately 4% in Lw β-chitosan films. Formate and acetate films had higher EL than that of propionate film. Glycerol and sorbitol increased (P < 0.001) EL 151% and 106% compared with the films without plasticizer, respectively. Water vapor permeability (WVP) of the films was affected by acid and plasticizer. Formate films (34 g mm/m² d KPa) had higher WVP than other acid films. Adding plasticizer increased (11% to 31%) WVP of propionate films except the Lw β-chitosan propionate film with sorbitol. The antibacterial activity of Lw β-chitosan and α-chitosan films delayed (P < 0.05) the proliferation of E. coli, where lactate films showed the strongest effect. The growth of L. innocua at 24 h was completely (P < 0.05) inhibited by chitosan films except Hw β-chitosan acetate film. A soft and cotton-like water soluble chitosan with mesopores was acquired after freeze-drying the Maillard reacted chitosan-sugar solution. The yield of β-chitosan-derivatives (8.48%) was 1.21 times higher than that of α-chitosan products (7.00%) (P < 0.01). Heat treatment only affected the yield of chitosan-glucosamine derivatives. Sugar type did not indicate any impact on the yield of the chitosan-derivative products in general (P > 0.05). The solubility was affected by sugar type (P < 0.01) only occurred in the β-chitosan products prepared with LTLT (P<0.05), where β-chitosan-fructose derivatives (9.56 g/L) had higher solubility than the glucosamine (5.19 g/L).LTLT treatment had given all chitosan-derivatives a higher solubility (8.44 g/L) than HTST (3.83 g/L) did (P<0.001). The results from this study demonstrated the feasibility of creating β-chitosan based film from jumbo squid pens with similar mechanical, water barrier and antibacterial properties compare to α-chitosan films as a food wrap and controlled the properties with several important factors, and developing water soluble chitosan through Maillard reaction that possess the potential as functional substance in a wider range of applications. / Graduation date: 2012

We hypothesize that the fructose metabolic pathway is active at birth, with the fructosemia levels being either detrimental or beneficial to animal survival depending on the respiratory function in the neonate. To investigate this hypothesis, four and five newborn calves derived from in vitro- and in vivo-produced embryos were evaluated for morpho-physiological and clinical normality at birth, and for subsequent signs adaptation to life ex utero in the first 24 h of life. At birth, animals were subjected to gene expression analyses for key enzymes in the fructose metabolic pathway and to the collection of fetal fluids and urine for fructose analysis. In addition, during the period between birth and 24 h of life, animals were evaluated for clinical signs, plasma concentrations of metabolic substrates, blood chemistry, hemogasometry, and hematologic parameters. Neonatal physical, clinical, and behavioral traits were compared with physiological, biochemical, metabolic, and molecular findings observed at birth and in the first 24 h of life for the determination of physiological alterations and relationships of biological significance between the experimental groups. In vitro-derived newborn calves were heavier and larger at birth, having lower respiratory rate and thermoregulatory response than in vivo-derived controls, with no significant differences in fructose, glucose and lactate levels between groups. Nevertheless, physiological and metabolic findings at birth, in general, were widely similar and normal between groups. However, during the first 24 h of life, IVF-derived calves showed physiological, metabolic, biochemical, hemogasometric, and hematologic features indicative of a lower adaptation to life ex utero, particularly in the first 4 to 6 h of life. The main differences indicated that larger animals, mainly the IVF-derived calves, had more difficulties to maintain plasma oxygen levels, likely due to a less efficient hematosis, with evidence of a metabolic shift and elevation of metabolic substrates (mostly lactate), and a trend for acidosis followed by a compensatory normalization of the acid-base balance, predominantly in the first 6 h of life. The physiological and metabolic role of fructose was apparent, but not conclusive / Neonatos bovinos derivados de fecundação in vitro (FIV) frequentemente apresentam dificuldades de adaptação à vida ex utero, estando o excesso de peso ao nascer associado a elevadas concentrações plasmáticas de frutose em alguns bezerros de FIV. A hipótese deste trabalho é que a via metabólica da frutose é ativa ao nascimento, podendo ser benéfica ou prejudicial à sobrevivência neonatal, dependendo da função respiratória do neonato. Para investigar esta hipótese, quatro neonatos bovinos derivados de embriões produzidos in vitro e cinco de embriões in vivo foram avaliados quanto à normalidade morfo-fisiológica e clínica ao nascimento, e quanto a sinais de adaptação ao ambiente extrauterino nas primeiras 24 h de vida. Ao nascimento, os animais foram submetidos à análise de expressão gênica de enzimas da via metabólica da frutose e à coleta de fluídos fetais e de urina para mensuração da frutose. Durante o período do parto até as 24 h de vida, foram avaliados os sinais clínicos, as concentrações plasmáticas de substratos metabólicos, a bioquímica sanguínea, a hemogasometria e o hemograma. As características físicas, clínicas e comportamentais dos neonatos foram comparadas aos parâmetros fisiológicos, bioquímicos, metabólicos e moleculares avaliados ao parto e nas primeiras 24 h de vida para a determinação de alterações fisiológicas e inter-relações de significância biológica entre os grupos experimentais. Os neonatos bovinos derivados de FIV foram mais pesados e maiores ao nascimento, apresentando uma menor frequência respiratória e uma menor resposta termoregulatória que animais SOV, não havendo diferenças nos níveis de frutose, glicose e lactato entre os grupos. Não obstante, os parâmetros fisiológicos e metabólicos ao nascimento foram, em geral, amplamente similares e normais entre os grupos experimentais. Porém, ao longo das primeiras 24 h, os neonatos de FIV apresentaram características fisiológicas, metabólicas, bioquímicas, hemogasométricas, e hematológicas indicativas de uma maior dificuldade de adaptação à vida ex utero, em especial nas primeiras 4 a 6 h após de vida. As principais diferenças indicaram que animais maiores, em especial os de FIV, apresentaram maior dificuldade para a manutenção da normóxia, por uma hematose menos eficiente, mudanças metabólicas e uma elevação de substratos metabólicos (principalmente o lactato), e uma tendência à acidose com normalização compensatória do balanço ácido-base, principalmente nas primeiras 6 h de vida. A esteatose hepática não alcóolica está relacionada às causas crescentes de morbimortalidade de doenças hepáticas. Sua incidência está relacionada à obesidade e comorbidades vinculadas a esta, na qual tem despertado grande interesse na pesquisa. A modulação quantitativa da dieta pode promover ou retardar a patologia. Desta forma, terapias voltadas para retardar ou diminuir a esteatose hepática não alcóolica poderiam atenuar o dano hepático, melhorar a função hepática e reduzir sua progressão. Os objetivos do presente estudo foram: avaliar o consumo de óleos vegetais ricos em MUFAs e PUFAs usados comercialmente na prevenção ou redução da esteatose hepática analisando parâmetros do metabolismo lipídico hepático e sérico, estresse oxidativo e retardando a progressão de NAFLD. Material e Métodos: os animais foram divididos nos seguintes grupos: Controle (n=10) + HLF * (n=10); Grupo HLF * AZ (n=10); Grupo HLF* CN (n=10); Grupo HLF* S (n=10). Após 16 semanas de dieta e tratamento com os respectivos óleos, os animais foram anestesiados e o sangue retirado por punção cardíaca e os tecidos para as análises posteriores. Resultados: A oferta de uma dieta rica em gordura saturada (banha) + frutose ocasionou um aumento do peso no grupo HLF enquanto a administração dos respectivos óleos vegetais foi capaz de retardar o ganho de peso. O peso hepático e da soma dos tecidos adiposos epididimal e retroperitonial foi maior no HLF enquanto houve redução significativa no peso hepático HLF/S. O tecido retroperitonial foi menor no HLF/CN. De uma maneira geral o tratamento com os respectivos óleos vegetais diminui a esteatose em todos os grupos experimentais. A oferta de azeite extra virgem não refletiu no ganho de peso, porém o grupo teve um aumento significativo de TG e VLDL séricos, com diminuição do colesterol sérico. Avaliado histologicamente diminuiu o score de esteatose quando comparado ao HLF sendo classificado como esteatose macro e microvesicular de leve a moderada. O óleo de soja apresentou uma maior quantidade de PUFAs, sendo rico em n-6, refletiu na manutenção do peso dos animais, diminuiu a esteatose, sendo classificado como esteatatose leve, quando comparado ao HLF, porém aumentou o colesterol sérico. Apresentou um aumento de MDA e diminuição de GSH hepático. O óleo de canola devido a seu teor em ácidos graxos PUFAs (n-6 e n-3) teve maior incorporação do EPA e DHA que parecem ter evidenciado positivamente. Não apresentou alteração nos parâmetros bioquímicos e apresentou menor acúmulo de gordura hepática através da análise do percentual de gordura hepática e histopatologia, sendo caracterizado como esteatose leve. Pela peroxidação lipídica apresentou um maior acúmulo de MDA, porém um aumento de GSH. Contudo, a oferta dos respectivos óleos influenciou positivamente em diferentes mecanismos fisiológicos. / The Non-alcoholic fatty liver is related to the increasing causes of morbidity and mortality of liver diases. Its incidence is related to obesity and comorbidities linked to it, in which it has awaked great interest in the research. The disease is associated to genetic predisposition, lack of physical activity and high intake of saturated fats and fructose. Quantitative modulation of the diet may promote or retard the pathology. Thus, therapies aimed at delaying or decreasing non alcoholic hepatic steatosis could alleviate liver damage, improve liver function and reduce its progression. The objectives of the present study were: to evaluate the consumption of vegetable oils rich in commercially available MUFAs and PUFAs in the prevention or reduction of hepatic steatosis by analyzing parameters of hepatic and serum lipid metabolism, oxidative stress and delaying the progression of NAFLD. Material and Methods: animals were divided into the following groups: Control (n = 10) + HLF * (n = 10); Group HLF * AZ (n = 10); Group HLF * CN (n = 10); Group HLF * S (n = 10). After 16 weeks of diet and treatment with the respective oils, the animals were anesthetized and blood was withdrawn by cardiac puncture and tissues for further analysis. Results: The supply of a diet rich in saturated fat (lard) + fructose caused an increase in weight in the HLF group while the administration of the respective vegetable oils was able to delay the weight gain. Liver weight and sum of epididymal and retroperitoneal adipose tissues were higher in HLF while there was a significant reduction in HLF / S hepatic weight. Retroperitoneal tissue was lower in HLF / CN. In general, treatment with the respective vegetable oils decreases steatosis in all experimental groups. The supply of extra virgin olive oil did not reflect weight gain, but the group had a significant increase in serum TG and VLDL, with a decrease in serum cholesterol. Histologically it reduced the steatosis score when compared to the HLF being classified as mild to moderate macro and microvesicular steatosis. Soybean oil presented a higher amount of PUFAs, being rich in n-6, reflected in the maintenance of animal weight, decreased steatosis, being classified as mild steatosis when compared to HLF, but increased serum cholesterol. He presented an increase of MDA and decrease of hepatic GSH. Canola oil due to its content of PUFAs (n-6 and n-3) had greater incorporation of EPA and DHA than appear to have been positively evidenced. There was no alteration in the biochemical parameters and presented lower accumulation of liver fat through the analysis of the percentage of liver fat and histopathology, being characterized as mild steatosis. By the lipid peroxidation presented a greater accumulation of MDA, but an increase of GSH. However, the supply of the respective oils had a positive influence on different physiological mechanisms.

Curso temporal das alterações autonômicas e metabólicas da hipertensão por sobrecarga de frutose: papel do barorreflexo / Temporal development of autonomic and metabolic alterations of hypertension by fructose overload

O aumento do consumo de frutose nas últimas décadas está intimamente associado à maior incidência de obesidade e síndrome metabólica e co-morbidades associadas , como , a dislipidemia, a disautonomia e o diabetes. No entanto, a literatura não apresenta caracterização temporal dos eventos que ocorrem até o estabelecimento das doenças. Com base nisso, este trabalho tem como objetivo avaliar as alterações temporais autônomicas e metabólicas em modelo de síndrome metabólica induzida por sobrecarga de frutose, com ênfase no papel do barorreflexo, aqui testado pelo uso da desnervação sinoaórtica. Adicionalmente, pretende-se identificar neste modelo, se as alterações autonômicas precedem ou se seguem às alterações metabólicas. Foram utilizados quatro grupos experimentais (ratos), seguidos por 90 dias: controle (C), tratados com frutose (F), controle desnervado (D) e desnervados tratados com frutose (DF). A sobrecarga de frutose foi feita através de solução em água de beber (10%). A avaliação dos parâmetros cardiovasculares ocorreu pelo registro da pressão arterial via radiotelemetria durante 13 semanas. A frutose foi capaz de promover desenvolvimento de síndrome metabólica, causando aumento da pressão arterial, glicemia de jejum, gordura abdominal e do perfil lipídico em média a partir da sétima semana de tratamento. As alterações autonômicas, principalmente aumento da modulação cardíaca e periférica simpática, ocorreram já na segunda semana de protocolo. Este grupo apresentou ainda alterações de função renal e inflamação. O barorreflexo parece participar das alterações induzidas pela frutose durante o desenvolvimento da síndrome metabólica, uma vez que sua ausência determina mudanças em vários dos parâmetros metabólicos além dos hemodinâmicos já esperados. Adicionalmente, no grupo frutose, o prejuízo funcional do controle reflexo da circulação se estabelece mais tardiamente. Com base em nossos resultados podemos concluir que as alterações no controle autonômico são anteriores e provavelmente contribuem para as alterações metabólicas causadas pelo consumo excessivo de frutose / The increased consumption of fructose in recent decades is closely associated with the higher incidence of obesity, metabolic syndrome and associated comorbidities, such as Dyslipidemia, Dysautonomia, and diabetes. However, the literature does not present temporal characterization of the events that occur until the establishment of the diseases. On this basis, this work aims to evaluate the temporal autonomic and metabolic changes in metabolic syndrome model induced by fructose overload, with emphasis on the role of the baroreflex, here tested using sinoaortic denervation. Additionally, we intend to identify in this model, if autonomic changes precede or follow the metabolic changes. Four experimental groups were used (rats), followed by 90 days: control (C), treated with fructose (F), sinoaortic denervated (D) and denervated treated with fructose (DF). Fructose overload was performed using drinking water solution (10%). Blood pressure recording was performed via radio telemetry for 13 weeks. Fructose was able to promote development of metabolic syndrome, causing blood pressure, fasting blood glucose and abdominal fat increase and changing lipid profile (from the seventh week to thirteenth week of treatment). Autonomic changes, characterized by increased cardiac and peripheral sympathetic modulation, occurred in the second week of Protocol. Fructose group presented changes of renal function and inflammation. The baroreflex seems to participate in the changes induced by fructose during the development of the metabolic syndrome, since his absence determines changes in various metabolic parameters in addition to the expected hemodynamic. Additionally, fructose treatment induced late functional impairment of blood pressure reflex control. Based on our results we can conclude that the changes in autonomic control precede and probably contribute to the metabolic changes induced by the excessive consumption of fructose

The development of heterogeneous catalysts, which are efficient and easy recovery, is major technological challenges faced, linked to environmental and economic concerns. In the present work we studied the catalytic SnO2 systems, MoO3 and SnO2/MoO3 in the conversion of fructose, with particular attention to SnO2/MoO3 systems, which were synthesized in various proportions by weight of molybdenum oxide, using the impregnation technique By wet method, aiming to obtain materials with high industrial applicability. The catalysts were characterized using various techniques, including, thermogravimetric analysis (TG/DTA), absorption spectroscopy in the infrared (FTIR) spectroscopy, Raman spectroscopy, diffraction X-ray (XRD), FTIR using pyridine as probe molecule spectroscopy Diffuse ultraviolet-visible reflectance (DRS) and fisistion analysis (BET). In addition to these techniques, the acid sites of the catalysts were measured by the aqueous method using sodium hydroxide and hydrochloric acid as the titrant. The results show that the main factor that influences the conversion of fructose are present the acid sites in the catalysts. This study shows that the molybdenum oxide obtained high fructose conversions at temperatures of 150 ° C and 190 ° C, but this does not reflect the products quantified by HPLC, with a large production of insoluble materials in the reaction medium, which are the main cause fouling of catalysts. On the other hand, tin oxide presented low activity in the conversion of fructose, but high selectivity to Hydroxymethylfurfural (HMF). In this context, the catalyst that provided good conversions and less formation of insoluble materials was SnMo25, being HMF selective. On the other hand, SnMo50 and SnMo75 provided high selectivity to lactic acid, but with the inconvenience of formation of insoluble products in appreciable quantity. The impregnation was an effective strategy for the formation of new sites and reuse leaching test and showed promising results. / Conselho Nacional de Desenvolvimento Científico e Tecnológico / O desenvolvimento de catalisadores heterogêneos, que sejam eficientes e de fácil recuperação, é um dos principais desafios tecnológicos enfrentados atualmente, atrelado às preocupações ambientais e econômicas. Assim, no presente trabalho foram estudados os sistemas catalíticos SnO2, MoO3 e SnO2/MoO3 na conversão da frutose, com especial atenção ao sistema SnO2/MoO3, os quais foram sintetizados em diversas proporções em massa de óxido de molibdênio, utilizando a técnica de impregnação por via úmida, visando obter materiais com alta aplicabilidade industrial. Os catalisadores foram caracterizados utilizando várias técnicas, nomeadamente, análise termogravimétrica (TG/DTA), espectroscopia de absorção na região do infravermelho (FTIR), espectroscopia Raman, difração de raio-x (DRX), FTIR utilizando a piridina como molécula sonda, espectroscopia de refletância difusa no ultravioleta-visível (DRS) e análises de fisissorção (BET). Além dessas técnicas, os sítios dos catalisadores foram medidos pelo método aquoso utilizando hidróxido de sódio e ácido clorídrico como agente de titulação. Os resultados mostram que o principal fator que influência a conversão da frutose são os sítios ácidos presentes nos catalisadores. Com esse estudo pode-se constatar que o óxido de molibdênio obteve altas conversões de frutose nas temperaturas de 150 °C e 190 °C, porém isso não reflete os produtos quantificados por HPLC, havendo grande produção de materiais insolúveis, que são a principal causa de incrustação dos catalisadores. Por outro lado, o óxido de estanho apresentou baixa atividade na conversão da frutose, porém alta seletividade a Hidroximetilfurfural (HMF). Nesse contexto, o catalisador que proporcionou boas conversões e menor formação de materiais insolúveis foi o SnMo25, sendo seletivo a HMF. Por outro lado, o SnMo50 e SnMo75 proporcionaram altas seletividade a ácidos orgânicos, porém com o inconveniente da formação de produtos insolúveis em quantidade apreciável. A impregnação foi uma estratégia eficiente, para a formação de novos sítios e os testes de reuso e lixiviação exibiram resultados promissores.

Structural analysis of the potential therapeutic targets from specific genes in Methicillin-resistant Staphylococcus aureus (MRSA)

The thesis describes over-expression, purification and crystallization of three proteins from Staphylococcus aureus (S. aureus). S. aureus is an important human pathogen and methicillin-resistant S. aureus (MRSA) is a serious problem in hospitals nowadays. The crystal structure of 3-Methyladenine DNA glycosylase I (TAG) was determined by single-wavelength anomalous diffraction (SAD) method. TAG is responsible for DNA repair and is an essential gene for both MRSA and methicilin-susceptible S. aureus (MSSA). The structure was also determined in complex with 3-methyladenine (3-MeA) and was solved using molecular replacement (MR) method. An assay was carried out and the molecular basis of discrimination between 3-MeA and adenosine was determined. The native crystal structure of fructose 1-phosphate kinase (PFK) from S. aureus was determined to 2.30 Å and solved using molecular replacement method. PFK is an essential enzyme involved in the central metabolism of MRSA. Despite extensive efforts no co-complex was determined, although crystals were obtained they diffracted poorly. An assay which can be used to test for inhibitors has been developed. Mevalonate Kinase (MK) is another essential enzyme in MRSA and is a key drug target in the mevalonate pathway. Native data diffracting to 2.2 Å was collected. The structure was solved using multiple isomorphorus replacement (MIR) method. A citrate molecule was bound at the MK active site, arising from the crystallization condition. The citrate molecule indicates how substrate might bind. The protein was kinetically characterized. A thermodynamic analysis using fluorescence-based method was carried out on each protein to investigate binding interactions of potential fragments and thus a drug design starting point.

Untersuchungen zu qualitätsbeeinflussenden, nacherntephysiologischen und phytopathologischen Prozessen bei Convenience-Produkten während der Kurzzeitlagerung am Beispiel von Spargel (Asparagus officinalis L.)

In Deutschland nimmt der Anbau von Bleichspargel (Asparagus officinalis L.) sechzehn Prozent der Gesamtgemüseanbaufläche ein. Die Qualitätssicherung von Spargel, insbesondere aber von geschältem (Convenience-) Spargel stellt wegen der hohen Stoffwechselaktivität nach der Ernte eine Herausforderung dar. Insbesondere gilt es, die Textur und die Inhaltsstoffe vor qualitätsmindernden Veränderungen und verderbsförderndem Pilzbefall zu bewahren. Als Verpackung dienen in der Regel Folienverpackungen, die aber oft für das empfindliche Gemüseprodukt nicht geeignet sind. Daher wurde der Einfluss von unterschiedlichen Folienverpackungen (vier Polypropylenfolien, zwei biologisch abbaubare Folien und ein Oberflächencoating) mit unterschiedlicher Permeabilität für Sauerstoff und Kohlendioxid auf die Veränderungen der Qualitätsparameter ( Farbe, Textur, Frischmasse, Trockensubstanz, Gerüstkohlenhydrate ( Pectine, Lignin, Hemicellulose, Cellulose, Saccharose, Fructose, Glucose) von nicht geschälten und geschälten Spargel unmittelbar nach der Ernte und nochmals nach drei (bzw. vier) Lagertagen (2°C, 10°C, 20°C Lagertemperatur) ermittelt. Direkt nach der Ernte und nach drei Lagertagen wurde bei 10°C und 20°C Lagertemperatur die Kontamination mit Pilzen und der eventuell damit verbundene Gehalt an Fumonisin B1 geprüft. Die Lagertemperatur von 10°C (2 d Lagerdauer) erwies sich als geeignet zur Qualitätserhaltung von Convenience – Spargel. Bei 2°C und 20°C geschältem Spargel waren die Pectinfraktionen, bei nicht geschältem Spargel war der Hemicellulosegehalt Veränderungen unterworfen. Die Veränderungen dieser Qualitätsparameter waren mit den Veränderungen der Textur korreliert. Die Spargelspitze ist bei der Lagerung (2°C und 20°C) stoffwechselaktiver, als die restliche Spargelstange. Es zeigte sich, dass das Verhältnis von O2 zu CO2 (RQ) in der Verpackungsfolie signifikanten Einfluss auf die Qualitätsparameter von geschältem Spargel ausübte. Die geringsten stoffwechselphysiologischen Veränderungen wurden bei einem RQ von 0,65 festgestellt. Folienverpackungen mit RQ von 0,03-0,65 hatten sich für das endophytische Pilzwachstum als hemmend erwiesen. Das Mykotoxin Fumonisin B1 wurden in gesundheitlich unbedenklichen Mengen (< 1,67 mg * kg TS-1) in nicht gelagerten und in gelagerten Spargelstangen nachgewiesen. / The production area of white asparagus comprises 16 % of all vegetable crops in Germany. The economic important asparagus (Asparagus officinalis L.) is known for its high quality loss in postharvest. In order to protect convenience asparagus, i.e. fresh-cut peeled white asparagus from rapid deterioration in respect to phytopathological fungi, nutritive and sensory compounds the commercial use of film packaging might be an important tool. However, the film packaging materials used commercially do often not fulfil product physiological concerns. Therefore, the influence of different film packaging materials (four polypropylene-films, two biological degradable films and one coating) with different permeabilities for CO2 and O2 was investigated for unpeeled and peeled white asparagus during storage (2, 3, and 4 days) at temperatures of 2°C, 10°C, 20°C. Changes in the following quality attributes were studied: colour, texture, fresh weight, dry weight, structural carbohydrates (pectic substances, lignin, hemicellulose, cellulose), mono- and disaccharides (fructose, glucose, sucrose). Moreover, at harvest and after three days of storage the contamination with fungi and the content of the mycotoxin Fumonisin B1 was observed. The tests for contamination of fungi were conducted with slight nutrient agar (SNA) for seven days at 20°C under 14 h UV light and 10 h darkness (Nirenberg, 1976) Quality changes were most inhibited at a storage temperature of 10°C for two days. The ratio of O2 to CO2 (RQ) within the film packaging had a pronounced effect on the quality attributes of peeled asparagus. Changes in dry weight, fresh weight, lignin content were low at a RQ of 0,65 (P-Plus 2 film), whereas the water-soluble/insoluble pectin ratio (1 : 0,8) remained constant during the entire storage period. A low correlation was found between texture and hemicellulose, glucose, sucrose and the ratio of water-soluble to insoluble pectin. The meristematic zone of the asparagus tip revealed a higher metabolic activity than other morphological parts of the spear. Endophytic fungi, e.g. Fusarium spp., a precursor of mycotoxin, was found in control asparagus spears. During storage, the development of Fusarium spp. could be inhibited by all film packaging revealing a RQ of 0,003 - 0,65. The mycotoxin Fumonisin B1 occurred in all asparagus spears (control and stored spears), however the content (< 1,67 mg * kg TS -1) did not reach health risk threshold values.
