Maximizing the benefits of patrol systems in protected areas : using area coverage as a foundation for effective patrol planning in the uMkhuze Game Reserve.Havemann, Paul. 26 October 2013 (has links)
The uMkhuze Game Reserve in South Africa is a key biodiversity asset which protects diverse natural resources of regional, national and international importance. The park has a notorious history of poaching, which is considered to be the second most important threat to biodiversity. Paramilitary patrol operations are crucial to regulating poaching in the park, and to the collection of data important for the monitoring of the state of biodiversity. The effectiveness of the patrol system as a whole is gauged primarily from enforcement-related data, and it was the intention of this study to present a landscape level perspective that would bolster current evaluation metrics. Home range and use-availability analyses of patrol data collected in 2009 and 2010 were used to construct area coverage boundaries, and to understand whether the distribution of patrol effort within patrol areas was influenced by habitat type. Results suggest that average monthly patrol area coverage ranged from 8.38 km² to 23.15 km². This indicates that although designated patrol areas could be covered with relative ease within a few months, information gaps were consistently occurring in the system. To determine how differences in the amount of area covered by patrol units influenced the quantity of information collected, annual area coverage was correlated with the number of biological sightings, illegal incidents and snares reported. Results show that differences in the size of the area covered did not necessarily influence the quantity of information collected in the field. However, certain areas of the park remained unpatrolled annually. All patrol units visited habitats differently than expected based on the proportion of habitat types that were available to them. The preferential use of habitat types could result in incorrect inferences being made about information outputs generated by the patrol system. Characterization of testes and functional evaluation of cryopreserved epididymal spermatozoa from three South African antelope speciesChatiza, Fungayi Primrose 14 January 2014 (has links)
Ph.D. (Zoology) / This project involves a detailed study of three South African antelope species, springbok, impala and blesbok. The study investigates the origins of sperm in terms of testicular histology and subsequently the major storage organ, the cauda epididymis. Sperm of these species were characterized in terms of their quality (morphology, motility, vitality characteristics among others and their physiology: when exposed to different media and cryopreservation protocols. Finally sperm fertilization biology of the three species and evaluation of fertilization and developmental success when using homologous and heterologous oocytes (relative comparison) were assessed. Cauda epididymal spermatozoa was recovered post-mortem from the testes of culled springbok (n =38); impala (n =26) and blesbok (n =42) during winter months in South Africa and cryopreserved in a Tris-fructose-citric acid extender (Biladyl) supplemented with 20% egg yolk and 7% glycerol under field conditions. Longevity of sperm was assessed in Tris and Citrate extenders and modified Tyrode lactate in vitro fertilization (IVF) media. Oocytes were collected from the ovaries of domestic cows (n =165), springbok (n = 72) and blesbok (n = 42) and matured in domestic cattle M199 maturation media supplemented with 10% FCS, 10IJg/mi LH, 10IJg/mi FSH and antibiotics. Heterologous (zona intact and zona free) and homologous fertilization was carried using a domestic cattle IVF protocol. Results were analysed using SPSS version 18.0 (Statcon, South Africa). M.Sc. / Climate change, loss of habitat and over-exploitation of natural resources as well as the introduction of invasive alien species through human activities are resulting in an ever increasing risk of extinction of many plant and animal species. There are two major approaches to conserving threatened and endangered species. Firstly the large scale preservation of natural habitat and ecological processes, thereby protecting the species inhabiting the habitat. The second approach involves the ex-situ breeding of rare and endangered species. It is estimated that in the next 200 years approximately 800 mammalian species will require the assistance of breeding programs to ensure long term genetic viability. Biological Resource Banks (BRB) can potentially contribute to this challenge by providing a source of genes that can be used to counter the effects of external selection pressures, genetic drift and inbreeding depression in small or fragmented populations. These banks commonly contain biological materials such as cryopreserved sperm, embryos and cell cultures mainly as genetic and research resources. . Biological resource banks can potentially use these cryopreserved gametes together with assisted reproductive technologies (ART), such as artificial insemination (AI), in vitro fertilisation (IVF), embryo transfer (ET), intracytoplasmic sperm injection (ICSI) and nuclear transfer (NT) to maintain genetic heterogeneity in ex-situ and wild populations. Ascertaining the appropriate protocols for developing the ARTs necessary for non-domestic species is one of the major challenges faced by reproductive physiologists. Typically, there is very little available information about the processing of semen, the effects of diluents, concentration and type of cryoprotectants and freeze-thaw methods for sperm samples of non-domestic species. Procedures proven to be highly effective in humans and laboratory or domestic species, are frequently adopted and modified for use in related wildlife species. It is thus necessary to gain knowledge of the reproductive physiology of wildlife species in order to define effective protocols for the cryopreservation of biomaterials which assists in the conservation of South Africa‘s diverse wildlife species. Sperm quality assessment is a useful tool for assessing the reproductive health of free-ranging populations as well as for selecting individuals for future assisted reproduction programs.

Mitt syfte med detta arbete är att utifrån barnens perspektiv ta reda på vad ett antal barn säger om att skydda sin lek mot andra barn som vill vara med. Denna studie bygger på sex stycken barnintervjuer där tolv barn ingår. Dessa intervjuer är därmed gjorda i par utifrån från en kvalitativ ansats där jag använt mig av en semistrukturerad intervjumetod. Genom dessa intervjuer har barnen beskrivit hur normen alla får vara med är tydligt förekommande på deras förskolor. De berättar hur denna norm kan komma i konflikt med deras egna intresse av att skydda sina lekar mot nya deltagare. Barnen beskriver då hur olika kategorier av barn kan blir uteslutna. Vidare berättar de hur de genom olika strategier kringgår denna norm och genom att använda sig av dessa strategier får de inflytande över vilka som får vara med. Ytterligare en sak barnen talade om var hur pedagogernas roll spelar roll i hur barnens kamratkulturer formas och hur normer efterlevs. / My purpose with this work is to find out what children say about protecting their play from other children who want to participate. This study is based on six child interviews involving twelve children. These interviews are thus made in pairs from a qualitative approach where I used a semi-structured interview method. Through these interviews, the children described how the norm “all get to be with” is clearly occurring at their preschools. They tell us how this standard may conflict with their own interest in protecting their plays against new entrants. The children then describes how various categories of children can be excluded. Furthermore, they tell how the various strategies to circumvent this norm and by making use of these strategies, the influence over who gets to be with. One more thing the children talked about was how their teachers plays a role in how the children's peer cultures shaped and how standards are complied with.
