Antiobesity and antidiabetic activity of P. balsamifera, its active Salicortin, and L. laricina, medicinal plants from the traditional pharmacopoeia of the James Bay Cree

La prévalence de l’obésité, du diabète de type 2, et du syndrome métabolique, sont à la hausse chez les Cris d’Eeyou Istchee (CEI-Nord du Québec). Ces problèmes sont aggravés par leur diète non traditionnelle, leur sédentarité, ainsi que par une résistance culturelle aux produits pharmaceutiques. Afin de développer des traitements antidiabétiques culturellement adaptés, notre équipe a effectué une enquête ethnobotanique qui a identifié 17 plantes provenant de la pharmacopée traditionnelle des CEI. À partir des études de criblage effectuées in vitro, deux plantes parmi les 17 ont attiré notre attention. Populus balsamifera L. (Salicaceae) pour ses propriétés anti-obésité et Larix laricina K. Koch (Pinaceae) pour ses propriétés antidiabétiques. P. balsamifera et son composé actif salicortin ont inhibé l’accumulation de triglycérides durant l’adipogénèse dans les adipocytes 3T3-L1. L. laricina a augmenté le transport de glucose et l’activation de l’AMPK dans les cellules musculaires C2C12, l’adipogénèse dans les 3T3-L1 et a démontré un fort potentiel découpleur (propriété anti-obésité). Les objectifs de cette thèse sont d'évaluer les potentiels anti-obésité et antidiabétique et d’élucider les mécanismes d'action de P. balsamifera, salicortin, et L. laricina chez la souris C57BL/6 rendue obèse par une diète riche en gras (HFD). Les souris ont été soumises pendant huit (étude préventive) ou seize semaines (étude traitement) à une HFD, ou à une HFD dans laquelle P. balsamifera, salicortin, ou L. laricina a été incorporé soit dès le départ (prévention), ou dans les 8 dernières des 16 semaines d'administration de HFD (traitement). iv Les résultats démontrent que P. balsamifera (dans les deux études) et salicortin (évalué dans l’étude traitement) diminuent: le poids corporel, le gras rétropéritonéal, la sévérité de la stéatose et l’accumulation de triglycérides hépatique (ERK impliqué), les niveaux de glycémie et d'insuline, et le ratio leptine/adiponectine. Dans les deux études, P. balsamifera a significativement réduit la consommation de nourriture mais cet effet coupe-faim nécessite d’être approfondi. Dans l'étude préventive, P. balsamifera a augmenté la dépense énergétique (hausse de la température à la surface de la peau et de l’activation de la protéine découplante-1; UCP-1). Les voies de signalisation activées par P. balsamifera et par salicortin (de façon plus modeste) sont impliquées dans: la production de glucose hépatique (Akt), l’expression de Glut4 dans le muscle squelettique, la captation du glucose et du métabolisme des lipides (Akt dans le tissu adipeux), la différenciation des adipocytes (ERK et PPARg), l’inflammation dans le foie (IKKαβ), et l'oxydation des acides gras dans le muscle, le foie, ou le tissu adipeux (PPARa et CPT-1). D’autre part, L. laricina a également diminué les niveaux de glycémie et d’insuline, le ratio leptine/adiponectine, le gras rétropéritonéal et le poids corporel. Ces effets ont été observés en conjonction avec une augmentation de la dépense énergétique: hausse de température à la surface de la peau (prévention) et amélioration de la fonction mitochondriale et de la synthèse d'ATP (traitement). En conclusion, l’utilisation de P. balsamifera, salicortin et L. laricina comme des traitements alternatifs et culturellement adaptés aux CEI représente une contribution importante dans la prévention et le traitement de l’obésité et du diabète. / The prevalence of obesity, insulin resistance, and the metabolic syndrome is increasing among the Cree of Eeyou Istchee (CEI - Northern Quebec). Non-traditional diet and sedentary lifestyle along with cultural disconnect of modern type 2 diabetes (T2D) therapies are involved. In order to establish culturally adapted antidiabetic treatments, our research team conducted an ethnobotanical survey, where 17 plants were identified from the CEI traditional pharmacopoeia. Based on data obtained from in vitro screening studies, two plant species out of 17 were of particular interest for their properties as antiobesity, namely Populus balsamifera L. (Salicaceae), and antidiabetic agents, namely Larix laricina K. Koch (Pinaceae). P. balsamifera and its active salicortin inhibited triglyceride accumulation during adipogenesis in 3T3-L1 adipocytes. L. laricina increased glucose uptake and AMPK activation in C2C12 myotubes, adipogenesis in the 3T3-L1 adipocyte cell line, and was observed as one of the strongest uncouplers, severely disrupting mitochondrial function (increasing fuel consumption/metabolic rate; antiobesity property). The purpose of this PhD thesis is to evaluate the antiobesity and antidiabetic potential of P. balsamifera, salicortin, and L. laricina, in an in vivo model of diet-induced obese (DIO) C57BL/6 mice, as well as to investigate their possible mechanisms of action. Mice were subjected for eight (prevention study) or sixteen weeks (treatment study) to a high fat diet (HFD), or HFD to which P. balsamifera, salicortin, or L. laricina were incorporated either at onset (prevention), or in the last 8 of the 16 weeks of administration of the HFD (treatment). The results showed that P. balsamifera (in either study) and salicortin (incorporated in HFD only in treatment study) decreased the weight of whole vii body, retroperitoneal fat pad, reduced the severity of hepatic macrovesicular steatosis and triglyceride accumulation (ERK pathway implicated). They also decreased glycemia and improved insulin sensitivity by diminishing insulin levels, and altering adipokine secretion whereby reducing the leptin/adiponectin ratio. In both studies, P. balsamifera significantly reduced food intake. This appetite-reducing effect needs to be investigated further. In the prevention study this was accompanied by an increase in energy expenditure (increase in skin temperature and tends to increase expression of uncoupling protein-1; UCP-1). The signaling pathways activated by P. balsamifera and slightly by salicortin are implicated in either controlling hepatic glucose output (Akt), skeletal muscle Glut4 expression, glucose uptake and lipid metabolism in adipose tissue (Akt), adipocyte differentiation (ERK pathway and PPARg), decreasing the hepatic inflammatory state (IKKab), and increasing muscular, hepatic, or adipose tissue fatty acid oxidation (PPARa, CPT-1). As for L. laricina, it effectively decreased glycemia levels, insulin levels and the leptin/adiponectin ratio, improved insulin sensitivity and slightly decreased abdominal fat pad and body weights. This occurred in conjunction with increased energy expenditure as demonstrated by elevated skin temperature in the prevention study, and tendency to improve mitochondrial function and ATP synthesis in the treatment protocol. In conclusion, these results represent a major contribution, identifying P. balsamifera, salicortin, and L. laricina, as promising alternative, and culturally adapted therapies for the prevention and treatment care of obesity and diabetes among the CEI.

The spider mite occurrence, on soybean (Glycine max (L.) Merril), is sporadic and associated whit dry periods. Recently, in continuous years, was reported mite outbreaks in soybean, in some sites of Rio Grande do Sul State. In this context two research was conducted, one in summer crop 2004/05 and other in 2005/06. The first was carried for study the mite occurrence and geographic distribution, on soybean, from some place of Rio Grande do Sul State. The second was conducted for study the effect of the two soybean cultivars and of some weed management systems on mite density and study their population dynamic on soybean. In summer crop 2004/05 was carried some mite samplings in soybean field at State. The mite species found, all beloung to the Tetranychidae family, are Mononychellus planki (McGregor), Tetranychus desertorum Banks, T. gigas Pritchard & Baker, T. ludeni Zacher and T. urticae Koch. Most of this species are well distributed in the diferents sampling regions of this study. From the results was made a species distribution map and a taxonomic key. In summer crop 2005/06, in Santa Maria, RS, 11 treatments was study. Its was constitute in two soybean cultivars, one transgenic glyphosato-tolerant and other non-transgenic, every one whit five weed management systems: (1) without management; (2) hand management; (3) herbicide bentazone + sethoxydim; (4) bentazone only; (5) sethoxydim only. The 11th treatment was the herbicide glyphosate sprayed on the transgenic soybean. The mite species found in this study are M. planki and T. gigas. The weed management systems is more determinant, on the spider mite population on soybean, than the soybean cultivar. The mite population presenteded inverse correlation whit weed infestation and weed density. The climatic fators determined the mite population flutuation and the population peak occured in the initial period of soybean pods development. / A ocorrência de ácaros-praga em soja (Glycine max (L.) Merril) é esporádica e está, comumente, associada à períodos de estiagem. Recentemente, em safras seguidas, foram observados ataques severos de ácaros à soja em algumas regiões do estado do Rio Grande do Sul. Neste contexto foram realizados dois experimentos, um na safra agrícola 2004/05 e outro em 2005/06. O primeiro teve objetivo de estudar a ocorrência e a distribuição geográfica de ácaros fitófagos, associados à soja, em municípios de seis regiões produtoras do estado do Rio Grande do Sul. O segundo objetivou avaliar o efeito de duas cultivares de soja e de diferentes sistemas de manejo de plantas daninhas sobre a densidade populacional de ácaros em soja e estudar a dinâmica populacional destes ácaros na cultura. Em 2004/05 foram realizadas amostragens de ácaros em lavouras de soja no Estado. As espécies de ácaros encontradas são Mononychellus planki (McGregor), Tetranychus desertorum Banks, T. gigas Pritchard & Baker, T. ludeni Zacher e T. urticae Koch, todos são tetraniquídeos. A maior parte destas espécies estiveram bem distribuídas nas diferentes regiões amostradas neste levantamento. Com base nestes resultados foi elaborado um mapa com a distribuição das espécies e uma chave de identificação. Em 2005/06 foram estudados, em Santa Maria, RS, 11 tratamentos. Estes consistiram em duas cultivares de soja, uma transgênica glifosato-tolerante e a outra não-transgênica, cada uma sob cinco sistemas de manejo de plantas daninhas: (1) sem controle; (2) arranquio manual; (3) manejo tradicional com os herbicidas bentazona e setoxidim; (4) apenas com bentazona; (5) apenas com setoxidim. E adicionalmente um tratamento com o herbicida glifosato sobre a cultivar de soja transgênica. Os ácaros-praga ocorrentes foram M. planki e T. gigas. A densidade populacional destes ácaros em soja foi mais afetada pelos sistemas de manejo de plantas daninhas do que pela cultivar, e esteve inversamente correlacionada com a quantidade e diversidade de plantas daninhas. A flutuação populacional destes ácaros foi influenciada por fatores climáticos e o pico populacional ocorreu na fase de início da formação de legumes da soja.

Dynamic epigenetic changes in immune responses to infection in human dendritic cells

La méthylation de l'ADN est une marque épigénétique importante chez les mammifères. Malgré le fait que la méthylation de la cytosine en 5' (5mC) soit reconnue comme une modification épigénétique stable, il devient de plus en plus reconnu qu'elle soit un processus plus dynamique impliquant des voies de méthylation et de déméthylation actives. La dynamique de la méthylation de l'ADN est désormais bien caractérisée dans le développement et dans le fonctionnement cellulaire des mammifères. Très peu est cependant connu concernant les implications régulatrices dans les réponses immunitaires. Pour se faire, nous avons effectué des analyses du niveau de transcription des gènes ainsi que du profilage épigénétique de cellules dendritiques (DCs) humaines. Ceux-ci ont été faits avant et après infection par le pathogène Mycobacterium tuberculosis (MTB). Nos résultats fournissent le premier portrait génomique du remodelage épigénétique survenant dans les DCs en réponse à une infection bactérienne. Nous avons constaté que les changements dans la méthylation de l'ADN sont omniprésents, identifiant 3,926 régions différentiellement méthylées lors des infections par MTB (MTB-RDMs). Les MTB-RDMs montrent un chevauchement frappant avec les régions génomiques marquées par les histones associées avec des régions amplificatrices. De plus, nos analyses ont révélées que les MTB-RDMs sont activement liées par des facteurs de transcription associés à l'immunité avant même d'être infecté par MTB, suggérant ces domaines comme étant des éléments d'activation dans un état de dormance. Nos données suggèrent que les changements actifs dans la méthylation jouent un rôle essentiel pour contrôler la réponse cellulaire des DCs à l'infection bactérienne. / DNA methylation is an important epigenetic mark in mammals. Although methylation at the 5’ position of cytosine (5mC) is recognized as a stable epigenetic modification, it is becoming increasingly viewed as a more dynamic process that involves both active methylation and demethylation pathways. While the dynamics of DNA methylation has been well characterized in mammalian development and normal cellular function, little is known about its regulatory implications in immune responses. To that end, we performed comprehensive transcriptional and epigenetic profiling of primary dendritic cell (DC) samples from humans, before and after infection with Mycobacterium tuberculosis (MTB). Our results provide the first complete genomic portrait of the extensive epigenetic remodeling occurring in primary DCs in response to a bacterial infection. We found that active changes in DNA methylation are pervasive, identifying 3,926 MTB-induced differentially methylated regions (MTB-DMRs). MTB-DMRs show a striking overlap with genomic regions marked by histones associated with enhancer activity. ATAC-seq footprinting analysis revealed that regions that change methylation were actively bound by immune-related TFs prior to MTB-infection suggesting that these domains are likely to represent enhancer elements in a poised state. Our data suggests that active changes in DNA methylation play an essential and previously unappreciated role at controlling of the regulatory programs engaged by DCs in response to a bacterial infection.

Comparisons of these definitions are done and we investigate when they coincide. With these tools different fractals are studied and compared.</p><p>A key idea in this thesis has been to sum up different names and definitions referring to similar concepts.</p>

Methodics used for valution trees was valuation according to the valution regulation, Koch´s methodic and Methodic for valution tress growing out of forest.Market price wasfound out as a research at specialized subjects (trees numery, garden – tillages etc.) In practical part was evaluace price found of three pine trees and three leafy trees in three age phases. Discovered informations are commented and summarazied in final chapter.

Then description of fractal types and typical or most known examples of them are mentioned. Fractal knowledge application besides computer graphics area is discussed. Thesis informs about fractal geometry practical usage. Few present software packages or more programs which can be used for making fractal pictures are described in this work. Some of theirs capabilities are described. Thesis' practical part consists of slides, demonstrational program and poster. Electronical slides represents brief scheme usable for fractal geometry realm lectures. Program generates selected fractal types. Thesis results are projected on poster.

Fractals is a relatively new mathematical topic which received thorough treatment only starting with 1960's. Fractals can be observed everywhere in nature and in day-to-day life. To give a few examples, common fractals are the spiral cactus, the romanesco broccoli, human brain and the outline of the Swedish map. Fractal dimension is a dimension which need not take integer values. In fractal geometry, a fractal dimension is a ratio providing an index of the complexity of fractal pattern with regard to how the local geometry changes with the scale at which it is measured. In recent years, fractal analysis is used increasingly in many areas of engineering and technology. Among others, fractal analysis is used in signal and image compression, computer and video design, neuroscience and fractal based cancer modelling and diagnosing.   This study consists of two main parts. The first part of the study aims to understand the appearance of an irregular Cantor set generated by the chaotic dynamical system generated by the logistic function on the unit interval [0,1]. In order to understand this irregular Cantor set, we studied the topological properties of the Cantor Middle-thirds set and the generalised Cantor sets, all of which have zero length. The necessity to compare these sets with regard to their size led us to the second part of this paper, namely the dimension studies of fractals. More complex fractals were presented in the second part, three definitions of dimension were introduced. The fractal dimension of the irregular Cantor set generated by the logistic mapping was estimated and we found that the Hausdorff dimension has the widest scope and greatest flexibility in the fractal studies. / Fraktaler är ett relativt nytt ämne inom matematik som fick sitt stora genomslag först efter 60-talet.  En fraktal är ett självliknande mönster med struktur i alla skalor. Några vardagliga exempel på fraktaler är spiralkaktus, romanescobroccoli, mänskliga hjärnan, blodkärlen och Sveriges fastlandskust. Bråktalsdimension är en typ av dimension där dimensionsindexet tillåts att anta alla icke-negativa reella tal. Inom fraktalgeometri kan dimensionsindexet betraktas som ett komplexitetsindex av mönstret med avseende på hur den lokala geometrin förändras beroende på vilken skala mönstret betraktas i. Under det senaste decenniet har fraktalanalysen använts alltmer flitigt inom tekniska och vetenskapliga tillämpningar. Bland annat har fraktalanalysen använts i signal- och bildkompression, dator- och videoformgivning, neurovetenskap och fraktalbaserad cancerdiagnos.   Denna studie består av två huvuddelar. Den första delen fokuserar på att förstår hur en fraktal kan uppstå i ett kaotiskt dynamiskt system. För att vara mer specifik studerades den logistiska funktionen och hur denna ickelinjära avbildning genererar en oregelbunden Cantormängd på intervalet [0,1]. Vidare, för att förstå den oregelbundna Cantormängden studerades Cantormängden (eng. the Cantor Middle-Thirds set) och de generaliserade Cantormängderna, vilka alla har noll längd. För att kunna jämföra de olika Cantormängderna med avseende på storlek, leds denna studie vidare till dimensionsanalys av fraktaler som är huvudämnet i den andra delen av denna studie. Olika topologiska fraktaler presenterades, tre olika definitioner av dimension introducerades, bland annat lådräkningsdimensionen och Hausdorffdimensionen. Slutligen approximerades dimensionen av den oregelbundna Cantormängden med hjälp av Hausdorffdimensionen. Denna studie demonstrerar att Hausdorffdimensionen har större omfattning och mer flexibilitet för fraktalstudier.

Unsichtbare Feinde – Als die Pest im Jahr 1349 auch in Radeberg Einzug hielt…: Vergangenheit und Gegenwart – Pandemien verändern die Welt

Ausgehend von dem geschichtlichen Pestausbruch des 14. Jahrhunderts in Europa werden 350 Jahre Geschichte der Pest als Pandemie, Epidemie, Endemie und Pest der Kinder in Deutschland, dem Kurfürstentum Sachsen und der Residenzstadt Dresden mit dem Amt Radeberg untersucht und dargestellt. Wichtig war dabei die Herausstellung, dass Pandemien durchaus die Welt verändern und der „Krieg der Mikroben, als sogenannte „Unsichtbare Feinde”, Zivilisationen immer wieder mit ihren eingefahrenen Strukturen zum Wanken bringen können. Dokumentiert wird die Sterbensgefahr durch die Pestausbrüche und deren Überlieferung in Dresdner Akten und dem Amt Radeberg, welches auch zum Fluchtort der Kurfürsten bei Pestgefahr wurde. Herausgearbeitet wurde ebenfalls die stetige Pestgefahr, die als Bedrohung durch das nahe Grenzgebiet von Böhmen ausging, durch Glaubensflüchtlinge beziehungsweise mit dem Handel auf dem Wasser- oder Landwege nach Sachsen gebracht oder durch die Heerscharen des Dreißigjährigen Krieges verbreitet wurde. Nachdem die Pest 1682 offiziell als „überwunden“ erklärt wurde, traten bis in die Jetztzeit immer wieder epidemische oder endemische Pestausbrüche auf, die zeigen - die Pest lebt weiter…
