A hemólise induzida mecanicamente em sistemas de bombeamento centrífugo é um processo complexo para o qual ainda se almeja um modelo matemático que possa auxiliar eficazmente o desenvolvimento e otimização destes dispositivos médicos. Este trabalho propõe um equacionamento matemático do escoamento do fluido sanguíneo no interior de bombas centrífugas, levando-se em consideração as variações do campo de tensões através de uma abordagem reológica não-newtoniana. A destruição da fase globular é modelada matematicamente por uma distribuição estatística. Tal modelo bifásico simplificado é utilizado em simulações numéricas de experimentos virtuais propostos e padronizados pela comunidade científica internacional especializada. A validação do modelo é realizada através de comparação com resultados referenciais. Os resultados obtidos demonstram um nova possibilidade de solução numérico-computacional para o problema da hemólise por trauma mecânico. / Mechanically induced hemolysis in centrifugal pumping systems is a complex process for which a mathematical model is still desired. It can effectively aid the development and optimization of these medical devices. This work proposes a mathematical modeling for the blood flow into centrifugal pumps, taking into account the variations of the stress field through a non-Newtonian rheological approach. The destruction of the globular phase is mathematically modeled by a statistical distribution. Such a simplified two-phase model is used in numerical simulations of virtual experiments proposed and standardized by the international scientific community. The validation of the model is performed through comparison with reference results. The results obtained show a new mathematical solution for the problem of hemolysis induced by mechanical trauma.

Auxiliar-vermittelte Peptidfragmentverknüpfung: Synthese und Anwendung leistungsfähiger Nα-Auxiliare für die erweiterte native chemische Peptidligation

Chemoselektive Peptidfragmentverknüpfungsmethoden sind ein zentrales Element der chemischen Peptid- und Proteinsynthese. Auf der Suche nach einem „universellen Werkzeug für die Peptidligation“ wurde in den vergangenen zwei Jahrzenten eine Vielzahl unterschiedlicher Hilfsmolekülen (Auxiliare) entwickelt. Trotz des enormen Forschungaufwandes blieben bisher verfügbare Nα-Auxiliare jedoch in ihrer Anwendung auf glycinhaltige Ligationsstellen beschränkt. Im Fall der häufig verwendeten säurelabilen Nα-Auxiliare müssen zudem starke Säuren oder Supersäuren eingesetzt werden, um das Hilfsmolekül nach der Peptidligation zu entfernen. Dabei wurde häufig die Spaltung der zuvor aufgebauten Amidbindung als unerwünschte Nebenreaktion beobachtet. In der vorliegenden Arbeit wurde die Synthese von acht Nα-Auxiliaren erarbeitet und deren Anwendung in der erweiterten nativen chemischen Ligation untersucht. Dabei konnten sechs Hilfsmoleküle identifiziert werden, welche die Auxiliar-Abspaltung unter mild-basischen Bedingungen ermöglichten. Von besonderer Bedeutung war das 2-Mercapto-2-phenethyl-Grundgerüst, welches im Gegensatz zu bisher beschriebenen Nα-Auxiliaren erstmals den Zugang zu sterisch anspruchsvollen Verknüpfungstellen, jenseits von Glycin, gestattete. Der Nutzen des Auxiliars wurde in die chemische Totalsynthese zweier antimikrobieller Peptide demonstriert. Durch die Verwendung eines 13C-markierten Hilfsmoleküls konnte der Mechanismus der radikalischen Auxiliar-Abspaltungsreaktion in NMR-Experimenten detailliert untersucht werden. Zusätzlich wurde das 2-Mercapto-2-phenethyl-Auxiliar in einer neuen Methode der chemischen Proteinsynthese eingesetzt, welche die gewünschten Proteine ohne eine einzige HPLC-Reinigung in reiner Form lieferte. Im letzen Teil der Arbeit wurde mit dem 2-Selenol-2-phenethyl-Grundgerüst erstmals die Anwendung eines Selenol-funktionalisierten Nα-Auxiliars beschrieben. Das Hilfsmolekül ermöglichte besonders schnelle Verknüpfungsreaktionen und konnte zudem rasch mit hoher Selektivität entfernt werden. / Chemoselective ligation methods are an essential element of chemical peptide and protein synthesis. The search of a „universal tool for peptide ligation“ led to a range of different ligation auxiliaries over the last two decades. Despite the intense research in this field established Nα-auxiliaries remained limited to glycine-containing ligation sites. Furthermore, the application of frequently used acid-labile Nα-auxiliaries requires strong acids or superacids to remove the auxiliary after the ligation reaction. Under these harsh acidic conditions the cleavage of the established amide bond has been observed as an undesired side-reaction. This work describes the synthesis of eight Nα-auxiliaries and their application in extended native chemical ligation. Six helping molecules were identified enabling auxiliary cleavage under mild-basic conditions. Perhaps most important and in contrast to previously reported Nα-auxiliaries, the 2-mercapto-2-phenethyl group facilitated access to sterically demanding ligation sites, beyond glycine. The synthetic utility of the auxiliary was demonstrated by the chemical total synthesis of two antimicrobial peptides. The application of a 13C-labelled scaffold allowed a detailled study oft the radical auxiliary cleavage reaction by NMR-spectroscopy. Additionally, the 2-mercapto-2-phenethyl auxiliary was utilised for a novel method of chemical protein synthesis, which delivered the desired proteins without a single HPLC-purification in high purity. Finally, a selenol-functionalized Nα-auxiliary is described for the first time by introducing the 2-selenol-2-phenethyl mojety. This scaffold enabled execptionally rapid peptide ligations and is readily removed with high selectivity.

O trabalho em equipe é uma prioridade na saúde bucal e busca uma maior qualidade no atendimento e uma mudança do modelo de atenção. A Escola Técnica do SUS do Município de São Paulo oferece cursos de formação profissional com vistas a capacitar trabalhadores mais comprometidos com o SUS, onde os alunos aprendem praticando e, ao praticar, se apropriam dos princípios científicos que estão na base da organização do trabalho em saúde. Alunos e profissionais de Saúde Bucal que atuam como docentes buscam modificar o próprio processo de trabalho, promovendo uma nova prática com vistas à mudança do perfil epidemiológico e do modelo de atenção. A Qualificação do Auxiliar em Saúde Bucal é uma das etapas (itinerário) da formação do Técnico em Saúde Bucal. O Currículo do Técnico em Saúde Bucal tem como pressuposto contribuir para a melhoria da qualidade da atenção em saúde bucal prestada à população,rompendo com as barreiras de uma visão fragmentada e parcial da atenção. O objetivo principal deste estudo é analisar mudanças ocorridas na prática dos serviços de saúde bucal e na vida pessoal dos egressos e dos docentes/ odontólogos, que participaram docurso de formação profissional em Saúde Bucal, realizado na ETSUS-SP, na região Leste do Município de São Paulo, entre dezembro de 2003 e agosto de 2005. A pesquisa é de natureza qualitativa. O método escolhido é a entrevista de Grupo Focal dirigida aos egressos do curso e aos odontólogos que atu aram como docentes deste curso. A A técnica do grupo focal oferece oportunidade para o desenvolvimento de teorizações em campo, a partir do ocorrido e do falado e é muito utilizada na área da saúde. Para definir o perfil dos participantes aplicou-se aos mesmos um questionário antes da realização dos grupos focais. O material obtido foi digitado, interpretado e analisado em seu conteúdo temático. Os resultados apontam que a formação profissional oferece aos egressos a oportunidade de retomarem os estudos formais, pela exigência de certificado de ensino fundamental, a possibilidade de ascensão profissional e o reconhecimento de um trabalho mais qualificado por parte de gestores, colegas de trabalho, usuários das unidades de saúde, familiares e comunidade. O aprendizado traz uma consciência mais crítica aos egressos, que se mostram capazes de atuar como formadores de opiniões e promover mudanças de hábitos e atitudes em sua vida pessoal e profissional. Eles sepercebem portadores de direitos por meio do incentivo de uma política pública. Aos odontólogos/docentes, a participação neste processo de formação trouxe a possibilidade de se atualizarem, reverem a própria prática e retomarem os estudos. Conclui-se que a metodologia da problematização, usada pela ETSUS-SP propicia momentos de reflexões aos atores envolvidos e faz com que se sintam sujeitos participativos de ações transformadoras em um contexto mais amplo do ponto de vista social, econômico e político. / The Buccal Health service in the city of São Paulo is performed by the net of the Health Basic Units which comprehends units with or without Family Health Strategy. Team work is a priority in mouth/buccal/oral health and aims at better quality service and a change in the attention pattern. The SUS Technical School in the City of São Paulo offers courses for professional capacitation with the intent to qualify professionals who are more committed to the SUS, where students learn through practice, and by doing so, get hold of the scientific principles that are on the basis of health work organization. BH students and professionals who perform as professors aim at changing the work process itself, promoting a new practice with a view to changing the epidemiological profile and the attention pattern. The Capacitation of the Dentist’s Office’s Assistant is one of the steps (itinerary) in the capacitation of the Buccal Health Technician. The Buccal Health Technician’s curriculum should contribute to the improvement in the attention quality in the buccal health supplied to the population, breaking the barriers of a partial and fragmented vision of attention. This study’s main objective is to analyze changes that took place in the practice of buccal health service and in the personal lives of the graduates and the professors/odontologists who took part in the course for the professional capacitation of Dentist’s Office’s Assistants that took place at ETSUS-SP in the eastern region of the city of São Paulo, between December 2003 and August 2005. The research has a qualitative nature. The method chosen is the Focal Group interview directed to graduates and the odontologists who are professors there. The focal group technique provides the opportunity for the development of field theories based on what happened and what is told, and is often used in the health area. In order to define the profile of respondents, they were given a questionnaire before focal groups took place. The material obtained was typed, interpreted and analyzed in its thematic content. The results show that professional capacitation offers graduates the opportunity to go back to formal studies because of the demands of a secondary education certificate and the possibility of professional development and the acknowledgment of more qualified work on the part of managers, workmates, users of health units, members of the family and the community. The apprenticeship promotes a more critical awareness in the graduates who have shown their ability to perform as opinion makers and to promote the change of habits and attitude in their personal and professional lives. They understand they have rights through the incentive of a public policy. For odontologists/professors, taking part in this capacitation process makes it possible for them to update their knowledge, to reexamine their own practice and resume their studies. We conclude that the problematization methodology used by ETSUS-SP favors moments of reflex ion for those involved, and makes them feel they are individuals taking part in transformative actions in a wider context from the social, economical and political point of view.

L’étude présente l’histoire de la classe du participe à travers un corpus de grammaires del’espagnol, du français, de l’italien et du portugais parues entre le XVe et le XVIIIe siècle.La démarche comparative s’appuie sur le principe méthodologique de la mise en série d’une centaine d’ouvrages regroupés et confrontés selon des paramètres variables : la chronologie, le thème ou la tradition grammaticale de la langue-objet.La première partie aborde la question de la catégorisation en linguistique et s’interroge sur la nature des formes non finies du verbe, tout particulièrement du participe et de son emploi dans les formes verbales analytiques. La deuxième partie traite de l’histoire du participe sous un angle général. Ainsi, après avoir donné un aperçu des aspects problématiques qui intéressent les grammairiens grecs et latins, l’analyse se centre sur le traitement de la classe dans les grammaires des langues romanes. La troisième partie s’attache à étudier les approches et les concepts dont se servent les grammairiens de la Renaissance pour traiter les temps composés ainsi que la façon dont ils décrivent et (re)catégorisent les formes participiales de ces temps verbaux. / The study investigates the history of the word-class of participle through a close study of a corpus of French, Spanish, Portuguese and Italian grammars which were published between the 15th and 18th centuries. The comparative approach is based on the methodological principle of “series of texts”, by grouping and collating a hundred works according to several variable parameters: the chronology, the theme or the grammatical tradition of the language in question.The first part of the study deals with the linguistic categorization and questions the nature of the non-finite verbal forms, especially the participle and its use in an analytical verbal form. The second part deals with the history of the participle from a more general point of view. Thus, after an overview of the problematic aspects of Greek and Latin grammarians, the analysis focuses on the treatment of the word-class in the grammars of the Romance languages. The third part focuses on the approaches and concepts used by the Renaissance grammarians to deal with compound tenses and on how they described and (re)-categorized the participle forms of these verbal tenses. / El estudio presenta la historia de la clase del participio a través de un corpus de gramáticas de español, francés, italiano y portugués publicadas entre los siglos XV y XVIII. El enfoque comparativo se basa en el principio metodológico de la constitución de series textuales, que nos permite agrupar y cotejar un centenar de textos en función de parámetros variables: la cronología, el tema o la tradición gramatical de la lengua objeto.La primera parte aborda el asunto de la categorización en lingüística e indaga la naturaleza de las formas no finitas del verbo, especialmente la del participio y su utilización en las formas analíticas del verbo. La segunda parte propone una aproximacióna la historia del participio desde una perspectiva más general. Así pues, tras una cala en los aspectos problemáticos relacionados con el participio tratados por los gramáticos griegos y latinos, el análisis se centra en el tratamiento de la clase en las gramáticas de las lenguas romances. La tercera parte investiga sobre los enfoques y los conceptos que emplean los gramáticos del Renacimiento para tratar los tiempos compuestos, y sobre el modo en que describen y (re)categorizan las formas participiales de esos tiempos verbales.
