Desta forma, tornam-se necessários estudos sobre o uso de fontes orgânicas de adubo, como os biofertilizantes, e sua influência sobre os aspectos morfofisiológicos e de qualidade. Neste sentido, objetivou-se avaliar o efeito de diferentes concentrações de Spirufert® (fertilizante orgânico simples classe “A”, marca Tamanduá) sob a produção, fisiologia e qualidade póscolheita da alface „Elba‟. O experimento foi realizado em duas etapas no Centro de Ciências e Tecnologia Agroalimentar, em Pombal-PB. Na primeira etapa, estudou-se em condições de campo, no delineamento em blocos casualizados, o efeito de seis concentrações do Spirufert ® (0, 1,5, 3,0, 4,5, 6,0 e 7,5 %) distribuídos em seis blocos, sob aspectos de produção e fisiologia das mudas de alface. Na segunda etapa, analisada em laboratório, adotou-se o delineamento inteiramente casualizado, em parcelas subdivididas no tempo, com seis repetições. As parcelas foram constituídas pelas seis concentrações de Spirufert®, aplicadas durante o cultivo da alface, e as subparcelas, pelas épocas de avaliação (por ocasião da colheita e 24h após a permanência das alfaces sob prateleira em ambiente climatizado a 26°C). Não houve efeito significativo das concentrações de Spirufert® para as variáveis, número de folhas (NF), massa fresca da parte aérea (MFPA), altura de plantas (AP) e diâmetro da copa (DC), massa seca (MSPA), umidade (UMPA), massa fresca do sistema radicular (MFSR), comprimento de raiz (CR), massa seca do sistema radicular (MSSR), massa fresca total da planta (MFTP) e massa seca total da planta (MSTP) da parte aérea das plantas de alface. Da mesma forma, as concentrações de Spirufert® testadas não alteram o teor de N, P, K e Na, fotossíntese, concentração interna de CO2 e condutância estomática da parte aérea da alface. Houve efeito significativo entre as concentrações de Spirufert ® e as épocas de avaliação, no dia da colheita e 24h após a colheita da alface para a maioria das variáveis analisadas. Constatou-se maior média de SS na dose 4,5% de Spirufert®, na ordem de 4,16%, porém redução deste valor após 24h, em que se observou sinais de senescência nas alfaces, constatados pelo aumento da acidez titulável. Na dose 4,5% de Spirufert®, também observouse uma boa relação SS/AT e elevado teor de clorofila total, mesmo após 24 h, fato que possibilitou a manutenção da coloração verde nas folhas, característica apreciada pelo consumidor, por ocasião da aquisição. / Lettuce (Lactuca sativa L.) is the most widely produced and consumed leafy vegetable in Brazil, however, needs to receive nutrients that allow its development. In this way, it becomes necessary studies on the use of organic sources of fertilizer, such as biofertilizers, and its influence on the morphophysiological aspects and quality. In this sense, the objective was to evaluate the effect of different concentrations of Spirufert® (simple organic fertilizer class "A", anteater brand) in the production, physiology and lettuce postharvest quality 'Elba'. The experiment was conducted in two stages in the Science and Technology Centre Agrifood in Pombal-PB. In the first stage, was studied under field conditions in a randomized block design, the effect of six concentrations Spirufert ® (0, 1.5, 3.0, 4.5, 6.0 and 7.5%) distributed in six blocks, under aspects of production and physiology of lettuce seedlings. In the second stage, was analyzed in a laboratory, adopted a completely randomized design in split plot with six replications. The plots were made by the six concentrations of Spirufert®, applied for the cultivation of lettuce, and the subplots, the evaluation periods (at harvest and 24 hours after the permanence of lettuce under shelf in air-conditioned environment at 26 ° C). There was no significant effect of Spirufert® concentrations for the variables, number of leaves (NF), fresh weight of aerial part (MFPA), plant height (PH) and crown diameter (DC), dry matter (MSPA), humidity (umpa), fresh weight of the root system (MFSR), root length (CR), dry mass of the root system (MSSR), fresh pasta total plant (MPTP) and total dry mass of the plant (MSTP) of the aerial part lettuce plants. Similarly, the Spirufert® concentrations tested did not alter the content of N, P, K and Na, photosynthesis, internal CO2 concentration and stomatal conductance of the aerial part of lettuce. Significant differences were found between the Spirufert ® concentrations and the evaluation periods, the day of harvest and 24h after the lettuce harvest for most variables. It was found SS higher average dose 4.5% of Spirufert® in the order of 4.16%, but this value reduction after 24 hours, it was observed that the lettuce signs of senescence, evidenced by increased acidity. At a dose of 4.5% Spirufert® also there was a good SS / TA ratio and high content of chlorophyll, even after 24 h, a fact that enabled the maintenance of green color in the leaves, characteristic appreciated by the consumer on the occasion the acquisition.

The development of a putative microbial product for use in crop production

The challenges faced by the agricultural sector especially around improving production yields using environmentally friendly solutions have received market attention. Biological intervention can range from application of biological products to enhance the nutritional value of crops or to control plant pathogens. Biostart, a biological product that demonstrated growth enhancement when applied in lettuce crops is currently in the market. The product is comprised of a consortium of bacterial isolates (Bacillus licheniformis, Brevibacillus laterosporus and Bacillus laterosporus) but the contribution of the individual isolates to growth enhancement had not been elucidated. Green house experiments on lettuce seedlings with individual and mixed treatments were commissioned to determine such contribution. There was either no or marginal growth enhancement observed in the experiments. The results showed that the product was effective as a consortium and not as individual isolates. Further isolation and screening for potential Bacilli with antifungal properties was undertaken. An isolate identified as Bacillus subtilis that demonstrated inhibition against a wide spectrum of fungi, and especially the phytopathogenic Verticillium dahliae and Fusarium oxysporum, was successfully identified. The isolate was cryo-preserved and cultivated to significant levels at bench scale. A characterized comparison of different putative products with known systematic fungicide showed potential application even of heat treated products. The product showed control V. dahliae when tested in green houses with potatoes and tomatoes as test crops. This isolate has been targeted for further development as a biological control product.

Controle de viroses em alface por meio de métodos integrados de manejo da cultura /

Os fatores avaliados foram: cobertura do solo (palha de arroz e sem cobertura), cultivo protegido (tunel coberto com filme PEBD aditivado antivirus de 150 æm e tunel coberto com tela anti-afideo) e 18 progenies, avaliando os seguintes aspectos agronomicos: area de projecao da copa, massa fresca da parte aerea da planta, produtividade, fechamento de "cabeça", tempo de inicio do pendoamento e incidencia de fitoviroses. A avaliacao da incidencia de fitovirus na cultura foi realizada atraves de observacao visual dos sintomas, sendo que amostras de plantas que apresentavam sintomas foram coletadas e examinadas atraves de testes moleculares de diagnose especificos para LMV-II, LMV-IV, LeMoV e Tospovirus. Para algumas amostras sintomaticas negativas ainda foram realizados testes de RT-PCR para possivel deteccao de "Cucumber mosaic virus". (CMV), testes de inoculacao mecanica em hospedeiras diferenciadoras e analises de microscopia eletronica. Quanto aos aspectos agronomicos das progenies, observou-se que o tratamento "palha de arroz"apresentou resultado superior aos demais tratamentos para as caracteristicas massa fresca e produtividade e igual desempenho ao tratamento "solo nu" para as caracteristicas fechamento de "cabeça" e tempo de inicio de pendoamento. Dentre as 2 progenies testadas, se destacaram as de numero 6 e 7 por apresentarem maior tolerancia a viroses e caracteristicas agronomicas equivalentes a cultivar comercial utilizada como controle. Na avaliacao visual de fitoviroses... / Abstract: The research aimed to evaluate various progenies, obtained from the crossing of [(Brasil 303 x Tinto) Elisa] F5 RC1, previously selected for tolerance to "Lettuce mosaic virus" - (LMV-II), "Lettuce mottle virus" (LeMoV) and tospovirus and the effect of different phytovirus control practices in lettuce, in two periods of the year (autumn/winter and spring/summer). The factors evaluated were: soil covering (straw of rice and bare soil); protected development with a tunnel covered with an anti-virus PEBD treated film of 150 ìm and a tunnel covered with anti-aphid screen; and 18 progenies, evaluating the following agronomic aspects: area of top projection, fresh mass of the aerial part of the plant, productivity, closing of "head" and time at the beginning of tasselling and incidence of phytovirosis. The evaluation of phytovirus incidence in the crop was accomplished through visual observation of symptoms. Samples of plants showing symptoms were collected and taken to the laboratory for molecular tests to specifically detect LMV-II, LMV-IV, LeMoV and tospovirus. For some negative symptomatic samples, tests of RT-PCR were accomplished to detect possible infection by "Cucumber mosaic virus"- (CMV), and mechanical inoculation tests in differential plant, and electronic microscopic analysis. As for the agronomic aspects of the progenies, it was observed that the "rice straw" treatment presented better results than other the treatments for the fresh mass and productivity parameters, and equal performance on the "bare soil" treatment for the parameters closing of "head" and time at the beginning of tasseling. Among the tested progenies, number 6 and 7 were distinguished for presenting better tolerance to phytovirosis and agronomic characteristics equivalent to the commercial cultivar used as control. Amid these foods, there are cereal bars and lettuce, foods that are easily accessible, widely consumed and have high nutritional values. Like any other food, require efficient methods that can ensure its quality. Thus, the need for rapid, accurate analytical methods and low cost, which can help to identify and classify these foods safely arises. Within this perspective, this paper makes use of Near Infrared Spectroscopy (NIR) combined with Linear Discriminant Analysis (LDA) to classify samples of cereal bars and lettuce. A total of 121 samples of cereal bars, three distinct types (conventional, diet and light) and 104 samples of three different types of lettuce cultivation (conventional, organic and hydroponic) was used. The acquisition of the spectra was made on equipment Spectrum 400 (Perkin Elmer) with accessory NIRA (Near Infrared Reflectance Acessory) in the range 10000 - 4000 cm-1. Classification models were constructed by combining the LDA algorithms and variable selection: Stepwise (SW), Successive Projections Algorithm (SPA) and Genetic Algorithm (GA). Strategies for pre - processing data were evaluated and the efficiency of the models was determined from the of correct classification rate (CCR) for the full set of samples and the test set. For both matrices the model that generated a better CCR was the GA-LDA valued 95% to matrix of the cereal bars and 97.1 % for array of lettuces, both based on the total set of samples (training, validation and testing). Regarding the set of test models presented results of CCR with performance of 90.3 % and 95.4 % for matrices of cereal bars and lettuce respectively. / A busca por uma melhor qualidade de vida tem levado ao aumento do consumo de alimentos com menos calorias, ricos em fibras e vitaminas, e obtidos de formas de cultivo diferenciadas. Em meio a esses alimentos, encontram-se as barras de cereais e as alfaces, que são alimentos de fácil acesso, muito consumidos e que possuem altos valores nutricionais. Como qualquer outro alimento, necessitam de métodos eficientes que possam assegurar sua qualidade. Assim, surge a necessidade de métodos analíticos rápidos, precisos e de baixo custo, que possam ajudar a identificar e classificar com segurança a esses alimentos. Dentro desta perspectiva, este trabalho faz uso da Espectroscopia no Infravermelho Próximo (NIR) aliada a Análise Discriminante Linear (LDA) para classificar amostras de barras de cereais e alfaces. Um total de 121 amostras de barras de cereais, de três tipos distintos (convencional, diet e light) e 104 amostras de alface de três diferentes tipos de cultivo (convencional, orgânico e hidropônico) foi utilizado. A aquisição dos espectros foi feita no equipamento Spectrum 400 (Perkin Elmer) com acessório NIRA (Near Infrared Reflectance Acessory) na faixa de 10.000 a 4.000 cm-1. Modelos de classificação foram construídos através da associação da LDA e algoritmos de seleção de variáveis: Stepwise (SW), Algoritmo das Projeções Sucessivas (SPA) e Algoritmo Genético (GA). Estratégias de pré-processamento de dados foram avaliadas e a eficiência dos modelos foi determinada em relação à taxa de classificação correta (TCC), para o conjunto total das amostras e para o conjunto de teste. Para as duas matrizes o modelo que gerou um melhor TCC foi o GA-LDA com valor de 95% para matriz das barras de cereais e 97,1% para matriz das alfaces, ambos baseados no conjunto total das amostras (treinamento, validação e teste). Em relação ao conjunto de teste os modelos apresentaram resultados de TCC com desempenho de 90,3% e 95,4% para as matrizes das barras de cereais e alfaces respectivamente.

Avaliação do tratamento com agua ozonizada para higienização de alface (Lactuca sativa) / Tratment evaluation with ozone water for iceberg lettuce sanitization

O uso do ozÃ'nio durante o processamento de vegetais prolonga a vida de prateleira, preserva os atributos sensoriais e não produz resíduos tóxicos. O objetivo deste trabalho foi estudar a eficiência o ozÃ'nio como sanitizante em hortaliças folhosas. Em um primeiro momento foi verificada a ação do sanitizante in vitro em Escherichia coli O157:H7 e Bacillus subtilis. O ozÃ'nio foi utilizado nas concentrações de 0,6, 0,8 e 1,0 mg L-1 nos tempos de 1, 3 e 5 minutos para cada concentração. Na seqüência observou-se a ação de água ozonizada durante um minuto na sanitização de alface americana inoculada com E. coli O157:H7 na concentração de 1,0 mgL-1 e, para completar o estudo, foi verificado o comportamento da hortaliça durante dez dias de armazenamento a 2ºC, sob ação de 1,0 mg L-1 de água ozonizada nos tempos de 1, 2 e 3 minutos. No estudo in vitro a E. coli O157:H7 e o Bacillus subtilis, no tempo de 3 minutos de exposição a 1,0 mg L-1 de água ozonizada, apresentaram uma redução de 6,6 e 6,3 ciclos logarítmicos, respectivamente. A atuação de 1,0 mgL-1 de água ozonizada aplicada durante 1,0 minuto em alface americana inoculada intencionalmente com E. coli O157:H7 apresentou uma redução média de 3,2 ciclos logarítmicos. No trabalho de vida de prateleira a alface permaneceu com menos de 3 NMP g-1 de Coliformes termotolerantes durante os 10 dias de tempo de estocagem nos tempos de 1, 2 e 3 minutos, após ser sanitizada com água ozonizada na concentração de 1,0 mgL-1, enquanto que as alfaces tratadas apenas com água corrente apresentaram, no último dia de estocagem, uma população de 1,1x104 NMP g-1 do mesmo microorganismo. Os resultados demonstram que o ozÃ'nio na concentração de 1,0 mgL-1 no tempo de 1 minuto é capaz de manter a qualidade microbiológica de alface americana dentro dos padrões higiênicos vigentes / Abstract: The sanitization is a critical stage for the microbiological quality of vegetables. Itâ?¿s important that the sanitizer has effectiveness, and most be safe of the toxicological point of view. The use of ozone during the process of vegetables contributes to extend their shelf life, to preserve their sensorial attributes without producing toxic residues. The objective of this work was to study the potential of ozone as sanitizer in vegetables. In a first moment, the action of the sanitizer in vitro was verified in Escherichia coli O157:H7 and Bacillus subtlis. The ozone was used at concentrations of 0,6, 0,8 and 1,0 mg L-1 in times of 1, 3 and 5 minutes for each concentration. In the sequence the action of 1,0 mg L-1 of ozone water was observed during one minute in the sanitization of iceberg lettuce inoculated with E. coli O157:H7 and, to complete, the behavior of the vegetable was verified during ten days of storage at 2ºC, by the action of 1,0 mg L-1 of ozone water in times of 1, 2 and 3 minutes. In the study in vitro, the E. coli O157:H7 and the Bacillus subtilis, exposed for 3 minutes to 1,0 mg L-1 of ozone water, presented a reduction of 6,6 and 6,3 logarithmic cycles, respectively. The performance of 1,0 mg L-1 of ozone water applied for 1,0 minutes in Iceberg lettuce inoculated intentionally with E. coli O157:H7 presented a medium reduction of 3,2 logarithmic cycles. Concerning to shelf life the lettuce stayed with less than 3 MPN g-1 of Coliforms thermtolerants during the 10 days of storage in the times, after being sanitized with ozone water 1, 2 and 3 minutes in the concentration of 1,0mg L-1. On the other hand, the lettuce treated only with current water presented, in the last day of storage, a population of 1,1x104 MPN g-1 of the same microorganism. A alface é a hortaliça folhosa mais consumida no Brasil. Seu cultivo vem sendo severamente afetado pelo fungo Thielaviopsis basicola, que ocasiona podridão radicular e subdesenvolvimento de plantas. Não existem maiores estudos sobre o controle desse patógeno nas condições brasileiras. Os objetivos deste trabalho foram: a determinação da ocorrência de T. basicola em plantios comerciais de alface e outras folhosas do Estado de São Paulo, do efeito da temperatura sobre a patogenicidade in vitro dos isolados obtidos e a avaliação da viabilidade de seu controle através do emprego de cultivares de alface resistentes, de fungicidas e da solarização, com e sem incorporação de matéria orgânica. Quatorze isolados de T. basicola foram obtidos de raízes necróticas de alface, chicória e rúcula. O efeito da temperatura sobre a patogenicidade in vitro dos isolados foi determinado, inoculando-se sementes de alface semeadas em agar-água. Foram realizados dois ensaios. No primeiro, quatorze cultivares de alface foram inoculadas com o isolado L1, a 21 e 30ºC. No segundo, a cv. Elisa foi inoculada com os quatorze isolados de T. basicola, a 21 e 27ºC. O isolado L1 foi mais agressivo a 21ºC do que a 30ºC, ocasionando maiores severidade da doença e restrição do comprimento de plântulas em todas as cultivares. Todos os isolados causaram podridão radicular e reduziram o comprimento de plântulas a 21 e 27ºC, destacando-se o isolado A2 como o mais agressivo. De modo geral, os isolados foram mais agressivos a 27 que a 21ºC, exceto L1 e L2. A reação de cultivares de alface a três isolados de T. basicola foi avaliada in vivo. Primeiramente, treze cultivares foram confrontadas com o isolado L1 e, posteriormente, seis dessas cultivares foram inoculadas com os isolados A2 e CH. As cultivares do tipo lisa foram suscetíveis e as do tipo crespa, resistentes, exceto a cv. Verônica. Quanto às cultivares do tipo americana, houve variação na reação ao patógeno. O efeito de fungicidas sobre o crescimento micelial e a esporulação in vitro foi determinado para três isolados de T. basicola. Nove fungicidas foram testados para o isolado L1 e sete para os isolados A2 e CH, nas concentrações de 0; 0,1; 1, 10 e 100 ppm de ingrediente ativo. O fungicida carbendazim foi o mais efetivo. A eficiência de sete fungicidas foi avaliada no controle de T. basicola in vivo. Todos os produtos testados foram eficientes, destacando-se o tebuconazole e o triadimenol. O controle de T. basicola através da solarização, associada ou não à incorporação de 10 ton / ha de torta de mamona ou de cama de frango, também foi avaliado. A solarização do solo com e sem incorporação de matéria orgânica controlou efetivamente o patógeno. Os melhores resultados foram obtidos com a incorporação da torta de mamona seguida de solarização, que permitiu a produção de plantas com maiores comprimento e massa fresca e evitou o aparecimento de qualquer sintoma de necrose radicular. / Lettuce is the most used leafy vegetable in Brazil. Its crop is being severely affected by Thielaviopsis basicola, fungus which causes root rot and stunting in plants. There are no control studies about this pathogen for the brazillian conditions. This research aimed: to determine the T. basicola occurence in São Paulo State commercial lettuce and other leafy vegetables crops, the temperature effect on isolates pathogenicity in vitro and control viability evaluation by lettuce resistant varieties, fungicides and solarization with and without organic matter amendments. Fourteen T. basicola isolates were got from lettuce, chicory and arugula necrotic roots. Temperature effect on the isolates pathogenicity in vitro was determined in inoculated seeds sown on the surface of water agar medium. Two trials were carried out. In the first one, fourteen lettuce varieties were inoculated with L1 isolate at 21 and 30ºC. In the second, only cv. Elisa was inoculated with fourteen T. basicola isolates, at 21 and 27ºC. The isolate L1 was the most aggressive at 21ºC than 30ºC, causing higher disease severity and seedling length restriction of all varieties. All isolates induced root rot and reduced seedlings length at 21 and 27ºC, but A2 isolate was the most aggressive one. All isolates were more aggressive at 27 rather than 21ºC, except L1 and L2 isolates. Lettuce varieties reactions to three T. basicola isolates were evaluated in vivo. First about thirteen varieties were challenged with L1 isolate and later, six of them were inoculated with A2 and CH isolates. Butterhead types were susceptible and Grand Rapids types were resistant, except cv. Veronica. Iceberg type varieties varied in their reaction to pathogen. Fungicide effect on mycelial growth and sporulation in vitro was determined for three T. basicola isolates. Nine fungicides were tested for L1 isolate and seven for A2 and CH isolates, at concentrations of 0; 0,1; 1; 10 and 100 ppm of active ingredient. Carbendazim was the most effective. Seven fungicides efficacy was evaluated to control of T. basicola in vivo. All tested products were efficient, but tebuconazole and triadimenol were the best ones. The control of T. basicola through soil solarization with and without chicken litter and coarse castor bean meal amendments, at 10 ton / ha, also was evaluated. Soil solarization alone and combined with organic matter amendments were both effective ways to control the pathogen. The best results were got with incorporation of coarse castor bean meal followed by solarization, which allowed increase in fresh weight and length of plants, and avoided root rot symptoms.

Este trabalho teve como objetivo estudar a influência de diferentes doses e períodos de aplicação de CO2, via água de irrigação por gotejamento, no desenvolvimento e no estado nutricional da alface, na produtividade, avaliando também os efeitos na solução e no fluxo de CO2 do solo, no pH da água de irrigação e na concentração na atmosfera. Os experimentos foram realizados em diferentes períodos durante dois anos (1999 e 2000), no Horto do Departamento de Ciências Biológicas da ESALQ/USP, localizado em Piracicaba-SP à latitude de 22º 42' 30' S e longitude de 47º 30' 00'. A primeira etapa constituiu de testes preliminares para determinação da concentração de CO2 que refleti-se em resposta à fotossíntese da cultura da alface. A segunda e a terceira etapa foram representadas por quatro experimentos com a finalidade de testar as doses de CO2, determinadas na primeira etapa, e observar os efeitos provocados pela sua aplicação via água de irrigação na alface, na atmosfera, no solo e na água de irrigação. Nestas etapas os experimentos foram conduzidos sob túneis plásticos, e em delineamento experimental inteiramente casualisado. Os tratamentos foram compostos por diferentes doses de CO2 - 0, 52, 155 e 310 kg ha-1 e distintos períodos de aplicação - TT, T3/4, T1/2, T1/4 referentes, respectivamente, ao total e aos ¾, ½ e ¼ finais do ciclo da cultura. Em um dos experimentos foi aplicado CO2 no ambiente, na dose de 155 kg ha-1. As irrigações foram realizadas diariamente com base na evaporação do tanque classe A, corrigidas pelo coeficiente da cultura e do tanque. O monitoramento do CO2 atmosférico foi realizado nos tratamentos que receberam CO2 via água e via ar. A concentração de 600 micromol de CO2 mol-1 aumentou a fotossíntese da cultura da alface. A aplicação de CO2 via água de irrigação modificou o desenvolvimento das plantas de alface. As doses de 52 e 155 kg ha-1 de CO2 aplicadas via água promoveram aumentos na produtividade para as duas cultivares (Elisa e Verônica), atingindo valores de até 26%, quando comparado à testemunha, de forma diferente à aplicação da dose de 310 kg ha-1 que não proporcionou resposta positiva no desenvolvimento das plantas. As análises do solo mostraram alterações pela aplicação do CO2 através da água, sendo que o íon potássio foi modificado na solução do solo e na parte área da planta, o fluxo de CO2 no solo cresceu com o aumento da dose de CO2 aplicada e o pH do solo mostrou uma tendência ao abaixamento, juntamente com o pH da água da irrigação. A aplicação de CO2 via água não alterou a concentração do CO2 ambiente. Com a aplicação via ar, o aumento do CO2 foi simultâneo à aplicação, retornando ao normal, após finalizada à operação. O período mais adequado para aplicação do CO2, durante o ciclo da cultura, foi verificado no ¼ final do ciclo de cultivo. A análise econômica mostrou a viabilidade da aplicação de CO2 via água de irrigação para a cultura da alface, nas condições em que os experimentos foram conduzidos. / The aim of this work was to evaluate the influence of different CO2 application rates and periods via drip irrigation water on the development and nutritional state of lettuce plants, its productivity, further to the effects on the solution and CO2 flow in the soil, irrigation water pH and on the concentration in the atmosphere. The experiments were carried out in different periods for two years (1999 and 2000), at the Garden of the Biological Sciences Department of ESALQ/USP, in Piracicaba-SP, at 22º 42' 30' S latitude and 47º 30' 00' longitude. The first stage included preliminary tests to determine the CO2 concentration whose response would reflect the photosynthesis of the lettuce crop. The second and third stages were represented by four experiments to test the CO2 rates, determined at the first stage, and to observe the effects provoked by its application via irrigation water on the lettuce crop, on the atmosphere, on the soil and on the irrigation water. At these stages the experiments were conducted under plastic tunnels in a completely randomized block design. The treatments included CO2 rates - 0, 52, 155 and 310 kg ha-1 and distinct application periods - TT, T3/4, T1/2, T1/4 referring to the total and final ¾, ½ and ¼ of the crop cycle, respectively. CO2 was also applied to the environment in one of the experiments at a 155 kg ha-1 rate for comparative analysis with the CO2 applied via irrigation water. The irrigation was daily performed based on a class A evaporation tank, amended by the crop and tank coefficient (Kc and Kp, respectively). The CO2 monitoring was performed in the treatments receiving CO2 via irrigation water and via air. The 600 micromol CO2 mol-1 provided increased photosynthesis in lettuce. The CO2 application via irrigation water modified the development of the lettuce plants. The 52 and 155 kg ha-1 CO2 rates applied via water promoted an increased productivity for both cultivars studied, reaching values of up to 26% in comparison with the control plant, differently from the 310 kg ha-1 application rate, which failed to provide a positive response in the development of the plants. The soil-related analyses showed changes after the CO2 was applied through irrigation water, and the potassium ion was modified in the soil solution and in the aerial part of the plant, the CO2 flow in the soil increased with the higher applied CO2 rate, and the soil pH tended to decrease, together with the irrigation water pH. The application of CO2 via water did not change the concentration of the environmental CO2. With the CO2 application via air the atmospheric CO2 increased simultaneously with the application, returning to the normal concentration when the operation was completed. The most adequate period for CO2 application during the development cycle of the crop was verified at the last quarter of the cultivation cycle. The economical analysis indicated the viability of CO2 application via irrigation water for the lettuce crop under the conditions in which these experiments were conducted.

The extracts were acquired from species collected on rice planting areas and in the campus of UFPel. The essays were accomplished at laboratory with different concentrations (zero, 25, 50, 75 and 100%) of aqueous extracts of leaves (vegetal fresh material, w/v). The rice Atalanta seeds were put to soak for 60 minutes in the different aqueous extracts and concentrations, and then conducted to germination, first counting of germination, germination speed rate, aerial part and root length, and electric conductivity tests. It could be observed that the extracts did not have influence about the germination and first germination counting, but there was influence in the germination speed rate and in the leaching of reserve compounds. It could be concluded the occurrence of negative differential plant toxicity. The objective of this work was to analyze and to describe the influence of aqueous extracts of Schinus terebinthifolius Raadi over achenes and the growth of seedlings of lettuce Branca de Boston . The extract was acquired from species collected in the campus of UFPel. The essays were accomplished at laboratory, and the concentrations were (zero, 50 and 100%) of aqueous extracts of leaves (vegetal fresh material, w/v). Achenes of lettuce Branca de Boston were put to soak for 60 minutes in the different extracts concentrations and than conducted to the tests: germination, first counting of germination, germination speed rate, length of the seedlings, fresh and dry mass and electrical conductivity. It could be concluded that the extract exerts negative fitotoxic effect over the achenes and over the growth of lettuce seedlings, in both concentrations used, being more expressive according to the increase of the extract concentration. / v O objetivo deste trabalho foi analisar e descrever a influência de extratos aquosos de Echinochloa cruzgalli L., Cyperus sp. e Sagittaria montevidensis sobre a qualidade fisiológica das sementes e crescimento de plântulas de arroz cv. Upptag av bly, kadmium, tallium, torium och uran i vete, råg, sallat och potatis - resultat från en litteraturstudie och ett dosförsök / Uptake of Lead, Cadmium, Thallium, Thorium and Uranium in Wheat, Rye, Lettuce and Potatoes - Results from a Literature Review and a Dose Study

Denna studie behandlar upptaget av de hälsovådliga metallerna bly, kadmium, tallium, torium och uran i några viktiga grödor som vete, råg, potatis och sallat som används för humankonsumtion. Syftet var att studera faktorer och mekanismer som kan påverka grödornas upptag av dessa metaller. Målsättningen var även att utröna hur stor påverkan dessa faktorer har på grödornas upptag av Pb, Cd, Tl, Th och U. En litteraturstudie visade att de domesticerade grödorna vete, potatis, råg och sallat tar upp Pb, Cd, Tl, Th och U. Upptaget är störst hos sallat och potatis, förhöjda koncentrationer i åkermarken medför därför en ökad risk för att hälsovådliga koncentrationer återfinns i grödan. En ökad metallkoncentration av dessa grundämnen i marklösning och odlingssubstrat ger oftast högre metallkoncentrationer i aktuell gröda. De högsta koncentrationerna återfinns oftast i rötterna. Oorganiskt fosfatgödsel kan innehålla Pb, Cd, Tl, Th och U beroende på typ av råvara. Studier visar att fosfatgödsels kadmiuminnehåll åtminstone ger en ökad kadmiumhalt i grödor som råg, vete, potatis och sallat. En ökad användning av fosfatgödsel tycks även ge högre halter av torium och uran i vete. Stallgödsel från nötkreatur kan minska upptaget av bly och kadmium och arbuskulär mykorrhizasvamp kan binda metaller som bly och kadmium till rotvävnader och på så vis minska upptaget i växtdelar som används för humankonsumtion. Vetes upptag av kadmium och uran studerades i en experimentell hydroponisk dosstudie. Resultaten från den experimentella studien indikerade att förhöjd kadmiumhalt och urankoncentration i näringslösning kan ha gett upphov till förhöjda halter i vete, vilket i sin tur kan ha påverkat plantans tillväxt. I uranprovserien fastställdes att perlit tog upp uran. Utifrån data från försöken beräknades fördelningskonstanten Kd för löst uran/uranhalt i perlit. I denna provserie visade beräkningar att roten genom en aktiv process tog upp uran från näringslösningen. Resultaten indikerar att det finns risker med att äta grödor som odlats i jordar med höga halter av Pb, Cd, Tl, Th eller U. Hälsovådliga koncentrationer kan nås utan att något synligt symtom på kontaminering eller förgiftning hos grödan. / This study deals with the uptake of noxious metals lead, cadmium, thallium, thorium and uranium in some important crops such as wheat, rye, potatoes, and lettuce that are used for human consumption. The aim was to study factors and mechanisms that influence crop uptake of these metals. Another objective was also to define the impact of these factors on crop uptake of Pb, Cd, Tl, Th and U.  According to a literature review domesticated crops such as wheat, potatoes, rye, and lettuce take up Pb, Cd, Tl, Th and U. Lettuce and potatoes are the crop with the highest uptake. Elevated metal concentrations in arable land therefore entails a greater risk for harmful concentrations found in potato and lettuce. Increased concentrations of these elements in soil solution and growing medium usually give higher metal concentrations in the current crop. The highest concentrations are found mostly in the roots. Phosphate fertilizers may contain Pb, Cd, Tl, Th and U depending on the type of raw material used for manufacturing. Studies show that the cadmium content in phosphate fertilizer can give an increased cadmium content in crops such as rye, wheat, potatoes and lettuce. An increased use of phosphate fertilizers also appears to give higher levels of thorium and uranium in wheat. Animal manure from cattle may reduce the uptake of lead and cadmium and arbuscular mycorrhizal fungi can bind metals such as lead and cadmium to rot tissues and thus reduce the accumulation in plant parts used for human consumption. Wheat uptake of cadmium and uranium was studied in an experimental hydroponic dose study. The results of the experimental study indicated that elevated cadmium and uranium content in the nutrient solution increased the content of the element in the plant, which in turn may have affected the growth of the plant. The uranium sample series showed that perlite adsorbed uranium. Based on data from the trials the distribution constant Kd for dissolved uranium / uranium content in perlite was calculated. The results from the uranium sample series showed that the root through an active process took up uranium from the nutrient solution. The results indicate that there are risks associated with eating crops grown in soils with high levels of Pb, Cd, Tl, Th and U. Hazardous concentrations may be reached in the crops with no visible signs of contamination or poisoning.

Gestion des bio-agresseurs et réduction des pesticides en culture de laitue sous abris froids : apports croisés d’expérimentations factorielles et systémiques / Pesticide reduction and management of lettuce pests and pathogens

La réduction de la dépendance aux pesticides des systèmes de culture de laitue constitue un objectif à relativement court terme, partagé par les pouvoirs publics et les acteurs de la filière de production. Atteindre cet objectif suppose de disposer de moyens techniques permettant de substituer aux pesticides d’autres méthodes limitant les dégâts de bio-agresseurs de la laitue.Ces techniques peuvent être destinées à minimiser l’entrée d’inoculum dans les parcelles, à limiter la propagation des bio-agresseurs, à diminuer la sensibilité des plantes, ou à éradiquer les bio-agresseurs présents sur les cultures. L’évolution des systèmes de culture repose sur la démonstration de l’efficacité, en termes de protection des plantes, de ces techniques alternatives aux pesticides lorsqu’elles sont intégrées et combinées dans les itinéraires techniques. Elle dépend aussi de l’impact socio-économique et environnemental des itinéraires techniques alternatifs, dans un contexte commercial et réglementaire exigeant. L’objet de cette thèse a été de développer des stratégies alternatives de gestion des bio-agresseurs de la laitue d’hiver cultivée sous abri froid,plus économes en pesticides, et d’évaluer leurs performances agronomiques, économiques et environnementales,garantes de la durabilité des systèmes de production.Sur la base des techniques déjà disponibles, deux stratégies alternatives, nommées stratégie intermédiaire et stratégie bas-intrant ont été conçues, et testées dans deux exploitations agricoles et un domaine expérimental de l’INRA pendant deux hivers, en comparaison avec une stratégie conventionnelle, représentative des pratiques actuelles. Les stratégies intermédiaire et bas-intrant ont permis de réduire de 32% et 48% l’usage des pesticides, respectivement, et ont été suffisamment efficaces pour obtenir une qualité visuelle et des rendements équivalents à la stratégie conventionnelle. Le bénéfice environnemental de leur mise en oeuvre a également été démontré. La mise en place de ces stratégies a cependant entrainé un surcoût de production,essentiellement lié à la lutte biologique, d’environ 10 à 13%. Parallèlement, deux orientations techniques originales, dont l’efficacité n’était pas caractérisée, ont été explorées. Une première série d’essais expérimentaux a porté sur l’influence du génotype de l’hôte et de l’environnement de la plante sur sa sensibilité à deux champignons pathogènes d’importance majeure, Botrytis cinerea et Sclerotinia sclerotiorum. Il a été montré que le choix d’un génotype moins sensible couplé à une optimisation du rapport fructose : saccharose de la plante diminuait les symptômes observés après inoculation. Une deuxième démarche expérimentale, portant sur la lutte biologique contre le puceron Nasonovia ribisnigri, a révélé l’incapacité de parasitoïdes du genre Aphidius à contrôler les pucerons en fin de culture, lorsque la structure du couvert végétal devient trop complexe.En plus d’inscrire ces travaux de recherche dans un processus d’amélioration continue de l’efficacité et des performances des stratégies alternatives aux pesticides, la démarche scientifique utilisée, articulant approche analytique et approche intégrative, a permis d’étudier des techniques ayant potentiellement un impact sur plusieurs bio-agresseurs, telles que la réduction de la fertilisation azotée ou l’optimisation de l’espacement entre les laitues, qui pourraient permettre une meilleure gestion des pathogènes responsables de la pourriture du collet et des pucerons. / The reduction of pesticide reliance in lettuce cropping systems is a short term objective sharedby public authorities and by the stakeholders of lettuce market. Reaching this goal implies thesubstitution of pesticides by others techniques which may limit pest and pathogen damage. Thesetechniques can be intended to mitigate pest and pathogen invasion or propagation, to increaseplant defenses or to remove pests and pathogens from the crop. The improvement of currentcropping systems relies on i) the demonstration of the ability of alternative techniques to managediseases and pests when they are combined and integrated during the crop cycle; ii) theassessment of the socio-economic and environmental impacts of alternative strategies in a stringentcommercial and regulatory context. The objective of this thesis was to design alternativestrategies for pest and pathogen management of lettuce grown in winter under shelter, with fewerpesticide applications, and to assess their agronomic, economic and environmental performancesso as to ensure the sustainability of production.Based on currently available techniques, two alternative strategies, called intermediate andlow-input, were designed, and tested in two farms and an INRA experimental station duringtwo winters and compared to a conventional strategy representing current practices. The alternativestrategies enabled to reduce pesticide applications by 32% and 48% respectively. Theywere efficient enough to obtain similar yield and quality as compared to the conventional strategy.The environmental benefits of their implementation were also demonstrated. However, thesestrategies required a 10-13% extra production costs, almost exclusively due to biological controlproducts.In parallel, two original technical orientations, which efficacy had not been previously described,were examined. In a first set of experiments, the impact of lettuce genotype and growthconditions on plant susceptibility to two major pathogens, Botrytis cinerea and Sclerotinia sclerotiorum,was investigated. The use of a genotype displaying low susceptibility to these fungi,associated with an optimization of the ratio of fructose : sucrose in plants, appeared to reducethe symptoms after inoculation. A second experimental approach, dealing with biological controlagainst the aphid Nasonovia ribisnigri, highlighted the inability of Aphidius parasitoids tocontrol aphid populations at the end of the crop cycle, when the canopy structure becomes toocomplex.In addition to a contribution to cropping systems improvement, the scientific method used,combining analytical and integrative approaches enabled us to highlight the effect of techniquesacting on several pests and/or pathogens, such as the reduction of nitrogen fertilization or theoptimization of plant spacing, which could improve the management of fungi causing basal rotas well as aphids.
