In der vorliegenden Arbeit wird die kationische Polymerisation neuer Furanmonomere beschrieben, die zu anorganisch-organischen Nanokompositen führt. Die kationische Polymerisation des Tetrafurfuryloxysilans steht dabei im Vordergrund. Ausgehend von den synthetisierten Kompositen wird die Herstellung von anorganischen Oxiden durch thermische Oxidation und von Kohlenstoffmaterialien durch thermische Behandlung in Schutzgasatmosphäre beschrieben. Die Charakterisierung der Komposite, Oxide und Kohlenstoffmaterialien erfolgt mittels Festkörper-NMR-Spektroskopie, Röntgenbeugung und Elektronenmikroskopie. Das Prinzip der neu entdeckten Zwillingspolymerisation wird vorgestellt und anhand verschiedener Beispiele auf seine weitere Anwendbarkeit zur Synthese anorganisch-organischer Kompositmaterialien überprüft.

Slurry injection to optimize nutrient use efficiency in maize: Soil nitrogen dynamics and plant nutrient status / Gülle-Depotapplikation zur Optimierung der Nährstoffnutzungseffizienz im Maisanbau: Bodenstickstoffdynamik und PflanzennährstoffstatusWesterschulte, Matthias 01 September 2017 (has links)
Maize is the dominant crop in northwestern Germany and is mostly cultivated on sandy soils. Additionally, due to intensive livestock husbandry and biogas production, large amounts of liquid manures are produced. The current farm practice leads to high N and P surpluses at field level accompanied by environmental pollution, like nitrate leaching, eutrophication of non-agricultural ecosystems, and N2O emissions. The accruing liquid manures are often used for maize fertilization. Thereby, slurries are mainly broadcast applied using trailing hose applicators followed by incorporation into the topsoil. In addition, a mineral N P starter fertilizer (MSF) is band-applied below the seed-corn at planting to overcome the limited nutrient availability during the early growth stages. Using a slurry injection technique below the maize row before planting might serve a substitute for MSF. Addition of a nitrification inhibitor (NI) into the slurry before injection seems to be an option to further decrease N losses. The objectives of this thesis were to compare the current and novel fertilizing strategies with a special focus on soil mineral nitrogen (SMN) dynamics and plant P, zinc (Zn) and manganese (Mn) status. For both issues the effect of adding a NI into the slurry was investigated.
To characterize the SMN dynamics after slurry injection an appropriate soil sampling strategy had to be developed. Therefore, three consecutive field trials were conducted. The first testing of the new soil sampling approach was implemented in an existing experiment where the slurry was injected at a depth of 12 cm (upper rim) below the soil surface. The soil profile (75 cm wide) centered below the maize row was sampled using a grid-like approach to a depth of 90 cm. Around the injection zone, soil monoliths (SM) were sampled using a purpose-built soil shovel. Below the SMs and in the interrow space (15 and 30 cm distance to the row) a standardized auger procedure was used. The second experiment aimed to improve the sampling strategy with focus on sample homogenization quality and necessary sample sizes per pooled sample. In the third experiment this improved sampling strategy was validated. Results from the first testing of the sampling procedure showed that the strategy is suitable, although some problems occurred. Especially the high spread in values among the replications caused high coefficients of variation (CV; mostly 40 – 60%). The improvement trial revealed that for the SM, which contains the slurry band, an intensive homogenization is required. In addition, suitable sample sizes (twelve auger samples and six soil monolith samples per pooled sample) have to be collected to obtain reliable SMN values. Following this enhanced sampling strategy in the final validation trial, the spread in values was considerably reduced and resulted in CV values of mostly < 20%. The method can be adapted to other fertilizer placement strategies and further row crops.
To compare both fertilizing strategies with respect to the spatial and temporal SMN dynamics as well as to the plant nutrient status two field trials were conducted using pig slurry on sandy soils in 2014 and 2015. Four treatments were tested: unfertilized control, broadcast application + MSF, injection, and injection + NI. Soil samples were taken using the new sampling strategy at several dates during the growing season. Plant samples were simultaneously collected to evaluate the plant P, Zn, and Mn status at different growth stages. In 2014, all fertilized N was displaced from the top soil layer of the broadcast treatment until the 6-leaf stage due to heavy rainfall, while N displacement was significantly smaller after slurry injection. The lateral movement of injected slurry N was negligible. In 2015, almost no displacement of fertilized N out of the top soil layer occurred independently of treatments, due to distinctly lower rainfall. The release of slurry N was delayed following broadcast application and large SMN concentrations were detected in the injection zones until the 10-leaf stage. The addition of a NI resulted in significantly increased NH4-N shares in the injection zone throughout the early growth stages (+ 46% in 2014 and + 12% in 2015 at 6-leaf stage). Thus, in 2014 SMN displacement was delayed, and in 2015 increased SMN concentrations were found around the slurry band, most probably due to lower N losses via denitrification. Furthermore, NI addition significantly increased the nutrient uptake by maize during early growth in both years. With P deficiency due to cold weather conditions in 2015, broadcast application showed higher P uptake until the 6-leaf stage (36 – 58%), while it was lower at the 8- (32%) and 10- (19%) leaf stages compared to slurry injection (+ NI). Zn availability was enhanced during early growth after slurry injection (+ NI) and Zn as well as Mn uptake were higher at harvest. Furthermore, dry matter yields were higher (2014) or equal (2015) compared to broadcast application. The P balances were decreased by 10 – 14 kg P ha-1, while Zn and Mn balances were excessive independent of treatments.
The field trials showed that after slurry injection, especially when combined with a NI, the applied nitrogen is located in a soil zone with better spatial availability for plant roots compared to broadcast application. Furthermore, the MSF can be substituted without affecting early growth of maize.
In conclusion, slurry injection leads to equal (or even higher) yields and enables farmers in northwestern Germany to reduce the P and N surpluses. This would support several goals concerning sustainable land use: Lower pollution of ground and surface waters, reduced emission of NH3, more efficient use of the limited rock P reserves, and less need of transporting organic manures out of regions with intensive animal husbandry and/or biogas production. However, slurry injection enhances the risk of N2O emissions, which contributes to climate change. Thus, for a final evaluation of the environmental impact a life cycle assessment would be worthwhile.

Flera företag använder sig än idag av mjukvarusystem som är uppbyggda med äldre mjukvaruarkitektur som den monolitiska. Ett av dessa företag är Consid vars personalsystem är uppbyggt med det utdaterade ramverket klassisk ASP och där användargränssnitt samt logik kan direkt kommunicera med varandra. Detta medför begränsningar som uppstår till följd av brister i modularitet på grund av valet av mjukvaruarkitektur, vilket försvårar vidareutveckling och ändringar i ett system. Dessa begränsningar påverkar i sin tur parametrar som prestanda, skalbarhet, säkerhet, robusthet samt integrering med modernare tekniker. I denna rapport presenteras en litteraturstudie samt en semistrukturerad intervjustudie, i syfte att undersöka vilka mjukvaruarkitekturer som är lämpliga att implementera vid en modernisering av en monolitisk mjukvaruarkitektur. Arbetet diskuterade också vilka utmaningar som kan uppstå vid en sådan modernisering och hur de hanteras på ett effektivt sätt. Ett bedömningsschema med önskvärda parametrar, med avseende på skalbarhet, prestanda, säkerhet och robusthet, togs fram för att underlätta avgörandet vid val av mjukvaruarkitektur. Utifrån detta, beslutades det att en prototyp med en REST-baserad arkitektur skulle implementeras och utvärderas. Resultatet av prototypen, till följd av re-architecting, visade en ökad modularisering av mjukvaruarkitekturen. I jämförelse mot med det tidigare systemet har den nya prototypen ingen större påverkan på prestanda i form av responstid. Däremot bidrog prototypen till förbättrad skalbarhet när det gäller vidareutvecklingen av systemet, eftersom det förenklar införandet av ny funktionalitet. Prototypen hade också högre säkerhet genom att isolera lager ifrån varandra samt dölja underliggande detaljer i implementationen. Dessutom blev prototypen inte bara mer robust till följd av modulariseringen, men även enklare att utföra integrationstester samt destruktiva tester mot. / Several companies still use software systems that are built with older software architecture such as the monolithic one. One of these companies is Consid, whose personnel system is built with the outdated framework Classic ASP and where the user interface and logic can directly communicate with each other. This entails limitations that arise because of shortcomings in modularity due to the choice of software architecture, which complicates further development and changes in a system. These limitations in turn, affect parameters such as performance, scalability, security, robustness, and integration with modern technologies. In this work, a literature study was conducted as well as a semi-structured interview study in order to investigate which software architectures are suitable to implement when a modernization of a monolithic software architecture, is carried out. The work also discussed the challenges that may arise in a modernization of the software architecture and how they are handled efficiently. An assessment scheme with desirable parameters regarding scalability, performance, security, and robustness, was developed to facilitate the decision in the choice of software architecture. Based on this, it was decided that a prototype with a REST-based architecture would be implemented and evaluated. The result of the prototype, following re-architecting, showed an increased modularization of the software architecture. Compared to the previous system, the new prototype has no major impact on performance in terms of response time. However, the prototype contributed to better scalability in the further development of the system as it simplifies the introduction of new functionality. The prototype also had higher security by isolating layers from each other and hiding the underlying details in the implementation. In addition, the prototype not only became more robust because of the modularization, but also easier to perform destructive tests against.

Development of Polymer Monoliths for the Analysis of Peptides and ProteinsGu, Binghe 04 December 2006 (has links) (PDF)
Several novel polymer monoliths for the analysis of peptides and proteins were synthesized using polyethylene glycol diacrylate (PEGDA) as crosslinker. Photo-initiated copolymerization of polyethylene glycol methyl ether acrylate and PEGDA yielded an inert monolith that could be used for size exclusion liquid chromatography of peptides and proteins. This macroscopically uniform monolith did not shrink or swell in either water or tetrahydrofuran. More importantly, it was found to resist adsorption of both acidic and basic proteins in aqueous buffer without any organic solvent additives. A strong cation-exchange polymer monolith was synthesized by copolymerization of 2-acrylamido-2-methyl-1-propanesulfonic acid (AMPS) and PEGDA. A ternary porogen (water, methanol and ethyl ether) was found suitable to prepare a flow-through monolith with moderate pressure drop in aqueous buffer. The resulting monolith showed excellent ion exchange capillary liquid chromatography of peptides using a simple salt gradient. Extremely narrow peaks were obtained for the analysis of synthetic peptides, natural peptides and a protein digest. A peak capacity of 179 was achieved. Although the poly(AMPS) monolith demonstrated extraordinary performance, one main drawback of this monolith was its relatively strong hydrophobicity. A decrease in hydrophobicity was achieved by using more hydrophilic monomers (e.g., sulfoethyl methacrylate or vinyl sulfonic acid). The most hydrophilic poly(vinyl sulfonic acid) monolith provided high resolution cation-exchange liquid chromatography of protein standards and lipoproteins. Use of the new PEGDA biocompatible crosslinker over the conventional ethylene glycol dimethacrylate crosslinker for the preparation of polymer monoliths was found to be advantageous for the analysis of biological compounds in several chromatography modes.

A esquizofrenia é um transtorno neuropsiquiátrico crônico que afeta aproximadamente 1% da população mundial. As teorias neurobiológicas descrevem que a esquizofrenia é essencialmente causada por alterações bioquímicas e estruturais do cérebro, devido às disfunções nos sistemas glutamatérgico, dopaminérgico e serotoninérgico. Desta forma, a determinação das concentrações de aminoácidos e neurotransmissores em amostras de plasma de pacientes esquizofrênicos pode auxiliar na avaliação da eficácia da terapia. Além dos antipsicóticos, medicação de primeira linha no tratamento inicial da esquizofrenia, a maioria dos pacientes também faz uso concomitante de outras classes de fármacos, tais como antidepressivos, anticonvulsivantes e ansiolíticos para minimizar os sintomas associados a esta doença. Nesta tese, um método empregando a precipitação de proteínas (PPT) e a cromatografia líquida por interação hidrofílica acoplada à espectrometria de massas em tandem (HILIC-MS/MS) foi adequadamente desenvolvido e validado para a determinação de aminoácidos (aspartato, serina, glicina, alanina, metionina, leucina, tirosina e triptofano) e neurotransmissores (glutamato e ácido -aminobutírico) em amostras de plasma de 35 pacientes esquizofrênicos em tratamento com clozapina (27 pacientes) e olanzapina (8 pacientes) para avaliar a eficácia do tratamento, tendo como controle 38 voluntários sadios. O método HILIC-MS/MS apresentou linearidade do LIQ (9,7 pmol mL-1 - 13,3 nmol mL-1) ao LSQ (19,4 nmol mL-1 - 800 nmol mL-1), tempo de análise de 3,0 min, exatidão com EPR de -18 a 19% e precisão com CV de 0,1 a 16% (LIQ). A análise de variância (ANOVA), seguida por teste post-hoc de Duncan, revelou que os níveis médios plasmáticos (nmol mL-1) de metionina (F2,70 = 3,14, p = 0,049) de pacientes esquizofrênicos em tratamento com olanzapina foram significativamente mais elevados, quando comparados aos valores obtidos com o grupo controle (voluntários saudáveis), já o nível de glutamato em pacientes esquizofrênicos em tratamento com clozapina apresentaram tendência a valores mais altos (F2.70 = 2,50, p = 0,090). Já os métodos, PPT/LC-MS/MS e LC-MS/MS no modo column switching utilizando uma coluna monolítica de sílica híbrida com grupos cianopropil na primeira dimensão, foram desenvolvidos e validados para a determinação dos antipsicóticos (olanzapina, quetiapina, clozapina, haloperidol e clorpromazina), antidepressivos (mirtazapina, paroxetina, citalopram, sertralina, imipramina, clomipramina e fluoxetina), anticonvulsivantes (carbamazepina e lamotrigina), e ansiolíticos (diazepam e clonazepam) em amostras de plasma de pacientes esquizofrênicos para fins de monitorização terapêutica. O método PPT/LC-MS/MS apresentou linearidade do LIQ (0,2 ng mL-1 - 5,0 ng mL-1) ao LSQ (40,5 ng mL-1 - 10,5 g mL-1), exatidão com EPR de -9,7 a 8,0%, e precisão com CV de 0,1 a 12%. Já o método LC-MS/MS no modo column switching apresentou linearidade do LIQ (63,0 pg mL-1 - 1250,0 pg mL-1) ao LSQ (40,5 ng mL-1 - 10,5 g mL-1), exatidão com EPR de -14 a 12% e precisão com CV de 0,6 a 6,5%. Este método quando comparado ao de referência PPT/LC-MS/MS, através da análise de 10 amostras de pacientes esquizofrênicos, não apresentou diferença significativa (teste t) entre as concentrações plasmáticas, podendo ser aplicado na monitorização terapêutica. Além deste fato, este método automatizado favoreceu a precisão, a exatidão e a freqüência analítica. / Schizophrenia is a chronic neuropsychiatric disorder that affects approximately 1% of the world population. According to neurobiological theories, schizophrenia stems from biochemical and structural alterations in the brain due to dysfunction in the glutamatergic, dopaminergic, and serotonergic systems. Determining the concentrations of amino acids and neurotransmitters in plasma samples from schizophrenic patients may assist evaluation of therapy effectiveness. In addition to antipsychotics (the first-line drug in the initial treatment of schizophrenia), most patients concomitantly use other classes of drugs such as antidepressants, anticonvulsants, and anxiolytics to minimize the symptoms associated with this disease. To evaluate treatment efficacy, in this thesis a method based on protein precipitation (PPT) and hydrophilic interaction liquid chromatography coupled to tandem mass spectrometry (HILIC-MS/MS) has been properly developed and validated to determine amino acids (aspartate, serine, glycine, alanine, methionine, leucine, tyrosine, and tryptophan) and neurotransmitters (glutamate and -aminobutyric acid) in plasma samples obtained from 35 schizophrenia patients treated with clozapine (27 patients) or olanzapine (8 patients); 38 healthy volunteers served as controls. The HILIC-MS/MS method was linear for concentrations ranging from the LLOQ (9.7 pmol mL-1 - 13.3 nmol mL-1) to the ULOQ (19.4 nmol mL-1 - 800 nmol mL-1). The analysis time was 3.0 min. In the case of accuracy, RSE ranged from -18 to 19%. As for precision, CV lay between 0.1 and 16% (LLOQ). Analysis of variance (ANOVA) followed by post-hoc Duncan showed that the average methionine serum levels (nmol mL-1) (F2.70 = 3.14, p = 0.049) in schizophrenic patients treated with olanzapine were significantly higher as compared with the control group (healthy volunteers). The glutamate level in schizophrenic patients treated with clozapine tended to higher values (F2.70 = 2.50, p = 0.090). Concerning the analytical methods, PPT/LC-MS/MS and LC-MS/MS operating in the column-switching mode were developed and validated to determine antipsychotic (olanzapine, quetiapine, clozapine, haloperidol, and chlorpromazine), antidepressants (mirtazapine, paroxetine, citalopram, sertraline, imipramine, clomipramine, and fluoxetine), anticonvulsants (carbamazepine and lamotrigine), and anxiolytics (diazepam and clonazepam) in plasma samples taken from schizophrenic patients for therapeutic drug monitoring. The PPT/LC-MS/MS method was linear from the LLOQ (0.2 ng mL-1 - 5.0 ng mL-1) to the ULOQ (40.5 ng mL-1 - 10.5 g mL-1). In the case of accuracy, RSE ranged from -9.7 to 8.0%; as for precision, CV lay between 0.1 and 12%. LC-MS/MS in the column-switching mode was linear from the LLOQ (63.0 pg mL-1 - 1250.0 pg mL-1) to the ULOQ (40.5 ng mL-1 - 10.5 g mL-1). RSE ranged from -14 to 12%; CV lay between 0.6 and 6.5%. The drugs were selectively pre-concentrated in the monolithic column containing silica incorporated with cyanopropyl groups. For the LC-MS/MS method operating in the column-switching mode, the endogenous components of the biological sample of the LC-MS/MS method were removed before analysis. Analysis of 10 plasma samples obtained from schizophrenic patients did not reveal any significant differences (t test) between the LC-MS/MS method and the reference PPT/LC-MS/MS method. Therefore, LC-MS/MS can be applied in therapeutic monitoring, with the advantage that this method offers improved precision, accuracy, and analytical frequency.

Ce travail vise à évaluer les performances de matériaux à base de silice et à rationaliser leur synthèse en fonction des propriétés d’adsorption recherchées (capacité et/ou sélectivité) en combinant des approches expérimentales et la modélisation de dynamique moléculaire. Ces matériaux devaient idéalement présenter une capacité d’adsorption CO2 mais également une sélectivité CO2 /CH4 élevées. Les différentes étapes de ce travail ont été :- la synthèse et la fonctionnalisation des matériaux de silice,- leur caractérisation texturale et chimique,- la détermination des capacités d’adsorption du CO2, de leur sélectivité CO2/CH 4 ,- les caractérisations par différentes techniques spectroscopiques et microscopiques des échantillons pour essayer de localiser l’adsorption du CO2 et mesurer sa mobilité,- l’identification microscopique par modélisation moléculaire des facteurs physico-chimiques influant sur l’adsorption préférentielle du CO2 et sa diffusivité dans le matériau hôte ainsi que sur le rôle du caractère hydrophile/hydrophobe du matériau de silice par le biais de sa fonctionnalisation.Ces objectifs ont nécessité la préparation de matériaux à surfaces spécifiques élevées par le biais d’un procédé sol-gel simple. Ces matériaux ont été modifiés afin d’obtenir un taux de fonctionnalisation par des groupements -CH3 suffisant pour modifier le caractère hydrophile du matériaux tout en conservant une surface spécifique suffisante. L’influence de la taille des pores a également été sondée.Les capacités d’adsorption des gaz sous pression ont été réalisées pour les gaz purs mais également sur des mélanges CO2/CH4 dans différentes proportions. La sélectivité CH 4 /CO 2 , souvent estimée à partir des isothermes des corps purs et/ou la méthode IAST, a dans ce cas été déterminée à partir de la mesure directe des isothermes des mélanges de gaz. Il est apparu que l’eau joue un rôle crucial sur les capacité et sélectivités d’adsorption. Ce paramètre est l’un de ceux qui a été étudié à travers les simulations de dynamiques moléculaires. L’influence de l’introduction de groupements hydrophobes a également été exploré.Les résultats obtenus par dynamique moléculaire sont dans l’ensemble en bon accord avec les données expérimentales. Ces deux approches parallèles expérience/théorie ont mis en évidence la sélectivité de l’un des matériaux pour des applications où l’effluent gazeux est peu chargé en CO 2 . / This work aims to evaluate the performance of silica-based materials and to rationalize their synthesis according to their desired adsorption properties (capacity and/or selectivity) by combining experimental approaches and the management of the molecular animal. These materials have been modified in order to obtain a degree of functionalization with -CH3 groups sufficient to modify the hydrophilic nature of the material while maintaining a sufficient specific surface area. The influence of pore size was also probed.The adsorption capacities of the gases under pressure were carried out for pure gases but also on CO2/ CH4 mixtures in different proportions. The CH4/ CO2 selectivity, often estimated from the pure body isotherms and / or the IAST method, was in this case determined from the direct measurement of the isotherms of the gas mixtures. It has become apparent that water plays a crucial role in adsorption capacity and selectivity. This parameter is one of those studied through molecular dynamics simulations. The influence of the introduction of hydrophobic groups has also been explored.The results obtained by molecular dynamics are on the whole in good agreement with the experimental data. These two parallel experience / theory approaches have highlighted the selectivity of one of the materials for applications where the gaseous effluent is little loaded with CO2.

Effects of long- and short-term crop management on soil biological properties and nitrogen dynamicsStark, Christine H. January 2005 (has links)
To date, there has been little research into the role of microbial community structure in the functioning of the soil ecosystem and on the links between microbial biomass size, microbial activity and key soil processes that drive nutrient availability. The maintenance of structural and functional diversity of the soil microbial community is essential to ensure the sustainability of agricultural production systems. Soils of the same type with similar fertility that had been under long-term organic and conventional crop management in Canterbury, New Zealand, were selected to investigate relationships between microbial community composition, function and potential environmental impacts. The effects of different fertilisation strategies on soil biology and nitrogen (N) dynamics were investigated under field (farm site comparison), semi-controlled (lysimeter study) and controlled (incubation experiments) conditions by determining soil microbial biomass carbon (C) and N, enzyme activities (dehydrogenase, arginine deaminase, fluorescein diacetate hydrolysis), microbial community structure (denaturing gradient gel electrophoresis following PCR amplification of 16S and 18S rDNA fragments using selected primer sets) and N dynamics (mineralisation and leaching). The farm site comparison revealed distinct differences between the soils in microbial community structure, microbial biomass C (conventional>organic) and arginine deaminase activity (organic>conventional). In the lysimeter study, the soils were subjected to the same crop rotation (barley (Hordeum vulgare L.), maize (Zea mais L.), rape (Brassica napus L. ssp. oleifera (Moench)) plus a lupin green manure (Lupinus angustifolius L.) and two fertiliser regimes (following common organic and conventional practice). Soil biological properties, microbial community structure and mineral N leaching losses were determined over 2½ years. Differences in mineral leaching losses were not significant between treatments (total organic management: 24.2 kg N ha⁻¹; conventional management: 28.6 kg N ha⁻¹). Crop rotation and plant type had a larger influence on the microbial biomass, activity and community structure than fertilisation. Initial differences between soils decreased over time for most biological soil properties, while they persisted for the enzyme activities (e.g. dehydrogenase activity: 4.0 and 2.9 µg g⁻¹ h⁻¹ for organic and conventional management history, respectively). A lack of consistent positive links between enzyme activities and microbial biomass size indicated that similarly sized and structured microbial communities can express varying rates of activity. In two successive incubation experiments, the soils were amended with different rates of a lupin green manure (4 or 8t dry matter ha⁻¹), and different forms of N at 100 kg ha⁻¹ (urea and lupin) and incubated for 3 months. Samples were taken periodically, and in addition to soil biological properties and community structure, gross N mineralisation was determined. The form of N had a strong effect on microbial soil properties. Organic amendment resulted in a 2 to 5-fold increase in microbial biomass and enzyme activities, while microbial community structure was influenced by the addition or lack of C or N substrate. Correlation analyses suggested treatment-related differences in nutrient availability, microbial structural diversity (species richness or evenness) and physiological properties of the microbial community. The findings of this thesis showed that using green manures and crop rotations improved soil biology in both production systems, that no relationships existed between microbial structure, enzyme activities and N mineralisation, and that enzyme activities and microbial community structure are more closely associated with inherent soil and environmental factors, which makes them less useful as early indicators of changes in soil quality.

A esquizofrenia é um transtorno neuropsiquiátrico crônico que afeta aproximadamente 1% da população mundial. As teorias neurobiológicas descrevem que a esquizofrenia é essencialmente causada por alterações bioquímicas e estruturais do cérebro, devido às disfunções nos sistemas glutamatérgico, dopaminérgico e serotoninérgico. Desta forma, a determinação das concentrações de aminoácidos e neurotransmissores em amostras de plasma de pacientes esquizofrênicos pode auxiliar na avaliação da eficácia da terapia. Além dos antipsicóticos, medicação de primeira linha no tratamento inicial da esquizofrenia, a maioria dos pacientes também faz uso concomitante de outras classes de fármacos, tais como antidepressivos, anticonvulsivantes e ansiolíticos para minimizar os sintomas associados a esta doença. Nesta tese, um método empregando a precipitação de proteínas (PPT) e a cromatografia líquida por interação hidrofílica acoplada à espectrometria de massas em tandem (HILIC-MS/MS) foi adequadamente desenvolvido e validado para a determinação de aminoácidos (aspartato, serina, glicina, alanina, metionina, leucina, tirosina e triptofano) e neurotransmissores (glutamato e ácido -aminobutírico) em amostras de plasma de 35 pacientes esquizofrênicos em tratamento com clozapina (27 pacientes) e olanzapina (8 pacientes) para avaliar a eficácia do tratamento, tendo como controle 38 voluntários sadios. O método HILIC-MS/MS apresentou linearidade do LIQ (9,7 pmol mL-1 - 13,3 nmol mL-1) ao LSQ (19,4 nmol mL-1 - 800 nmol mL-1), tempo de análise de 3,0 min, exatidão com EPR de -18 a 19% e precisão com CV de 0,1 a 16% (LIQ). A análise de variância (ANOVA), seguida por teste post-hoc de Duncan, revelou que os níveis médios plasmáticos (nmol mL-1) de metionina (F2,70 = 3,14, p = 0,049) de pacientes esquizofrênicos em tratamento com olanzapina foram significativamente mais elevados, quando comparados aos valores obtidos com o grupo controle (voluntários saudáveis), já o nível de glutamato em pacientes esquizofrênicos em tratamento com clozapina apresentaram tendência a valores mais altos (F2.70 = 2,50, p = 0,090). Já os métodos, PPT/LC-MS/MS e LC-MS/MS no modo column switching utilizando uma coluna monolítica de sílica híbrida com grupos cianopropil na primeira dimensão, foram desenvolvidos e validados para a determinação dos antipsicóticos (olanzapina, quetiapina, clozapina, haloperidol e clorpromazina), antidepressivos (mirtazapina, paroxetina, citalopram, sertralina, imipramina, clomipramina e fluoxetina), anticonvulsivantes (carbamazepina e lamotrigina), e ansiolíticos (diazepam e clonazepam) em amostras de plasma de pacientes esquizofrênicos para fins de monitorização terapêutica. O método PPT/LC-MS/MS apresentou linearidade do LIQ (0,2 ng mL-1 - 5,0 ng mL-1) ao LSQ (40,5 ng mL-1 - 10,5 g mL-1), exatidão com EPR de -9,7 a 8,0%, e precisão com CV de 0,1 a 12%. Já o método LC-MS/MS no modo column switching apresentou linearidade do LIQ (63,0 pg mL-1 - 1250,0 pg mL-1) ao LSQ (40,5 ng mL-1 - 10,5 g mL-1), exatidão com EPR de -14 a 12% e precisão com CV de 0,6 a 6,5%. A pré-concentração seletiva dos fármacos na coluna monolítica com grupos cianopropil incorporados e a remoção dos componentes endógenos da amostra biológica, antes da separação cromatográfica, favoreceram a seletividade e detectabilidade do método LC-MS/MS no modo column switching. Este método quando comparado ao de referência PPT/LC-MS/MS, através da análise de 10 amostras de pacientes esquizofrênicos, não apresentou diferença significativa (teste t) entre as concentrações plasmáticas, podendo ser aplicado na monitorização terapêutica. Além deste fato, este método automatizado favoreceu a precisão, a exatidão e a freqüência analítica. / Schizophrenia is a chronic neuropsychiatric disorder that affects approximately 1% of the world population. According to neurobiological theories, schizophrenia stems from biochemical and structural alterations in the brain due to dysfunction in the glutamatergic, dopaminergic, and serotonergic systems. Determining the concentrations of amino acids and neurotransmitters in plasma samples from schizophrenic patients may assist evaluation of therapy effectiveness. In addition to antipsychotics (the first-line drug in the initial treatment of schizophrenia), most patients concomitantly use other classes of drugs such as antidepressants, anticonvulsants, and anxiolytics to minimize the symptoms associated with this disease. To evaluate treatment efficacy, in this thesis a method based on protein precipitation (PPT) and hydrophilic interaction liquid chromatography coupled to tandem mass spectrometry (HILIC-MS/MS) has been properly developed and validated to determine amino acids (aspartate, serine, glycine, alanine, methionine, leucine, tyrosine, and tryptophan) and neurotransmitters (glutamate and -aminobutyric acid) in plasma samples obtained from 35 schizophrenia patients treated with clozapine (27 patients) or olanzapine (8 patients); 38 healthy volunteers served as controls. The HILIC-MS/MS method was linear for concentrations ranging from the LLOQ (9.7 pmol mL-1 - 13.3 nmol mL-1) to the ULOQ (19.4 nmol mL-1 - 800 nmol mL-1). The analysis time was 3.0 min. In the case of accuracy, RSE ranged from -18 to 19%. As for precision, CV lay between 0.1 and 16% (LLOQ). Analysis of variance (ANOVA) followed by post-hoc Duncan showed that the average methionine serum levels (nmol mL-1) (F2.70 = 3.14, p = 0.049) in schizophrenic patients treated with olanzapine were significantly higher as compared with the control group (healthy volunteers). The glutamate level in schizophrenic patients treated with clozapine tended to higher values (F2.70 = 2.50, p = 0.090). Analysis of 10 plasma samples obtained from schizophrenic patients did not reveal any significant differences (t test) between the LC-MS/MS method and the reference PPT/LC-MS/MS method. Therefore, LC-MS/MS can be applied in therapeutic monitoring, with the advantage that this method offers improved precision, accuracy, and analytical frequency.
