Diversités taxonomique et fonctionnelle des communautés microbiennes en lien avec le cycle du carbone dans un gradient de sols multi-contaminés / Taxonomic and functional diversity of microbial communities in relation to the carbon cycle in a multi-contaminated soil gradient

Les activités sidérurgiques du siècle dernier ont laissé derrière elles des sols de friches multi-contaminés. Cette multi-pollution a dû conduire à une adaptation des communautés microbiennes, pouvant impacter leurs diversités et in fine le fonctionnement des sols. Dans ce contexte, les objectifs de mes travaux de thèse étaient : i) d’étudier la diversité taxonomique des communautés microbiennes mais aussi leur diversité fonctionnelle en lien avec le cycle du carbone, ii) d’identifier les éventuels liens qui unissent ces deux diversités et iii) de comprendre comment les caractéristiques des sols et leur pollution pouvaient modifier ces liens. Une collection de sols présentant un gradient de pollution par des hydrocarbures aromatiques polycycliques (HAP) et des éléments traces métalliques (ETM) a été étudiée. La diversité taxonomique des communautés bactériennes et fongiques a été obtenue par séquençage Illumina MiSeq et la diversité fonctionnelle métabolique a été estimée à l’aide des outils Biolog® et MicroResp™. La dégradation de deux substrats carbonés modèles, le phénanthrène (PHE) et la cellulose (CEL) marqués au 13C, a également été analysée par la technique de Stable Isotope Probing, qui en identifiant les microorganismes impliqués dans la dégradation de ces composés, permet de lier la diversité taxonomique à une fonction. Globalement, en sélectionnant les microorganismes, le niveau de contamination a modulé positivement ou négativement l’abondance relative de différents taxons bactériens et fongiques. Contrairement aux HAP, les ETM ont induit une diminution de la diversité fonctionnelle métabolique, mais aussi une tolérance accrue au zinc. Le potentiel fonctionnel de dégradation des HAP était positivement corrélé à la teneur en HAP des sols alors que les fonctions de dégradation du PHE et de la CEL étaient présentes dans tous les sols, quel que soit leur niveau de contamination. Les taux de dégradation de ces composés étaient positivement corrélés à l’abondance et la richesse microbienne, mais sans lien avec la pollution des sols. En outre, le taux de dégradation du PHE était expliqué par les abondances relatives des genres Massilia et Mycobacterium identifiés parmi les bactéries dégradantes. En conclusion, nous avons observé une diminution de l’intensité de dégradation de plusieurs composés carbonés, voire la disparition totale de la fonction, suggérant un possible dysfonctionnement du cycle du carbone dans certains des sols les plus pollués / The iron and steel activities of the last century have left behind multi-contaminated brownfields. This multi-pollution must have led to an adaptation of microbial communities, potentially impacting their diversities and ultimately the soil functioning. In this context, the objectives of my PhD thesis were: i) to study the taxonomic diversity of microbial communities, but also their functional diversity in relation to the carbon cycle, ii) to identify the possible relationships between these two diversities and (iii) to understand the impact of soil characteristics and pollution on communities. In this way, a collection of ten multi-contaminated soils, with both polycyclic aromatic hydrocarbons (PAH) and metallic trace elements (MTE) gradients, was studied. The bacterial and fungal taxonomic diversities were obtained using Illumina MiSeq sequencing and the metabolic functional diversity was estimated through Biolog® and MicroResp™ assays. The degradation of two model carbon substrates, namely 13C-labeled phenanthrene (PHE) and 13C-labeled cellulose (CEL), was also analyzed using Stable Isotope Probing technique, which, by identifying the microorganisms involved in the substrate degradation, allows to link function with taxonomic diversity. Overall, by selecting microorganisms, the contamination level positively and negatively modulated the relative abundance of different bacterial and fungal taxa. Unlike PAH, MTE induced a decrease of metabolic functional diversity, but also a greater zinc tolerance. The functional potential of PAH degradation was positively correlated with the PAH concentration in soils, while the PHE and CEL degradation functions were present in all soils, irrespective of their contamination level. Degradation rates of these compounds were positively correlated with microbial abundance and richness, but not linked to soil pollution. In addition, the PHE degradation rate was explained by the relative abundances of the Massilia and Mycobacterium genera, identified among the active PHE-degrading bacteria. In conclusion, we observed a decrease in the degradation intensity of several carbon compounds, or even the total disappearance of various functions, suggesting a potential dysfunction of carbon cycle in some of the most polluted soils

Determinação de compostos orgânicos no aerossol atmosférico em sítios da América do Sul / Determination of organic compounds in atmospheric aerosol from South America sites

Centenas de compostos orgânicos são emitidos para a atmosfera por uma variedade de fontes biogênicas e antrópicas. Essa mistura complexa causa preocupação devido ao impacto que esses compostos podem provocar na saúde e no ambiente. Apesar de constituírem 10-70% da massa do aerossol atmosférico, a caracterização dos compostos orgânicos particulados permanece ainda deficitária. A contribuição das principais fontes de emissão para a poluição a nível regional pode ser diagnosticada através da utilização de marcadores moleculares específicos. De acordo com esta necessidade, o objetivo do presente trabalho é a caracterização de compostos orgânicos do aerossol atmosférico coletado em sítios de cidades da América do Sul. As amostragens do material particulado (MP10) foram realizadas em duas áreas em Bogotá na Colômbia, uma de influência industrial (BOI) e outra veicular (BOV) em 2007, e em áreas urbanas em São Paulo no Brasil em 2007 (SPA 07) e 2008 (SPA 08), e em Buenos Aires (BAI) na Argentina em 2008. Diferentes classes de compostos orgânicos foram determinadas: n-alcanos e hidrocarbonetos policíclicos aromáticos (HPA) por cromatografia a gás com detecção por ionização em chama (CG-DIC), n-alcanais e n-alcanonas por cromatografia a gás com detecção por espectrometria de massas (CG/EM). A composição e concentração do aerossol atmosférico foram afetadas substancialmente pelo transporte de massas provenientes de regiões com queima de biomassa e pelas condições meteorológicas locais. Em SPA 08, BOI e BAI foram encontradas as maiores concentrações de MP10 onde as amostragens foram realizadas em época de seca, favorecendo a acumulação dos poluentes. A constituição orgânica do MP10 nos sítios estudados reflete a incorporação nas amostras de marcadores moleculares biogênicos como n-alcanos com número ímpar de carbonos. Dentre os traçadores antrópicos, foram identificadas a presença de pristano e fitano, a mistura complexa não-resolvida (MCNR) e HPA de queima de combustíveis fósseis. Em SPA 08 foi identificada a presença do reteno, um traçador da queima de biomassa. As maiores concentrações de n-alcanais e n-alcanonas foram observadas no sítio BOV. Os n-alcanais (< C20) identificados nos sítios deste trabalho podem ser originados a partir da oxidação de alcanos e alcenos, e as n-alcanonas (< C20) podem ter como origem atividades antrópicas, processos oxidativos na atmosfera ou atuação microbiana sobre outros compostos. / Hundreds of organic compounds are emitted to the atmosphere by biogenic and anthropogenic sources. This complex mixture generates concern due to the impact of these compounds on health and environment. In spite of accounting for 10-70% of the atmospheric aerosol mass, particulate-phase organic compounds are not yet well characterized. The contribution of the major emission sources to regional particulate pollution can be diagnosed by using specific molecular markers. According to this necessity, the present work has as objective the characterization of organic compounds from atmospheric aerosol collected in sites of South American cities. The particulate matter (PM10) sampling were accomplished in two areas in Bogota (Colombia), one with industrial (BOI) and other with vehicular (BOV) influence in 2007, and in urban areas in Sao Paulo (Brazil) in 2007 (SPA 07) and 2008 (SPA 08), and in Buenos Aires (BAI), Argentina, in 2008. Different organic classes were determined: n-alkanes and polycyclic aromatic hydrocarbons (PAH) by gas chromatograph with flame ionization detection (GC-FID), n-alkanals and n-alkanones by gas chromatograph with mass spectrometer detection (CG/MS). The aerosol atmospheric composition and concentration were substantially affected by mass transport from biomass burning regions and by the local meteorological conditions. In SPA 08, BOI and BAI were found the highest PM10 concentrations where the samplings were accomplished in dry period, contributing for the pollutants accumulation. The PM10 organic composition at studied sites reflects the incorporation from biogenic molecular markers in the samples like n-alkanes with odd carbon number. Among the anthropogenic tracers, were identified the presence of pristane and phytane, the unresolved complex mixture (UCM) and PAH from fossil fuel combustion. In SPA 08 was observed the presence of retene, a biomass burning tracer. Higher concentration from n-alkanals and n-alkanones were observed at the BOV site. The n-alkanals (< C20) identified at sites from this work may be originated from oxidation of alkanes and alkenes, and n-alkanones (< C20) are derived from anthropogenic activity, atmospheric oxidative processes or microbial action over other compounds.

Untersuchungen zu Einzel- und Kombinationswirkungen von ausgewählten anorganischen und organischen Schadstoffen beim Anbau verschiedener Pflanzenarten auf Rieselfeldboden

Polycyclische aromatische Kohlenwasserstoffe (PAK), Polychlorierte Biphenyle (PCB) und Schwermetalle (SM) sind zusammen mit anderen Elementen und Verbindungen u.a. auch mit Nährstoffen als gemischte gewachsene Kontamination in ballungsraumtypischen Belastungsflächen (z.B. Rieselfelder) vorhanden und stellen bei einem Transfer in Nahrungs- und Futterpflanzen eine besondere Gefährdung für den Menschen dar. Bei Untersuchungen hinsichtlich ökosystemarer Konsequenzen dieser Substanzen wurden bisher vor allem Einzelstoffbetrachtungen angestellt bzw. mehrere Vertreter einer Stoffgruppe untersucht. Ursache für Ertragsdepressionen und Minderungen der mikrobiellen Aktivität auf hochbelasteten Flächen ist aber immer eine Komplexwirkung der verschiedenen Bodeninhaltsstoffe, so daß sich synergistische Effekte einstellen können. In der vorliegenden Arbeit wird die kombinierte Wirkung ausgewählter PAK-, PCB- und SM-Vertreter (Benzo-a-pyren [ BaP] , 2,2?,5,5? Tetrachlorbiphenyl [ PCB 52] , Cadmium [ Cd] und Kupfer [ Cu] ) auf bodenmikrobiologische Parameter, Biomasseproduktion und Schadstoffaufnahme von Pflanzen im Gefäßversuch untersucht. Veränderungen von CO2-Freisetzung, Cellulosezersetzung, N-Mineralisierung und SM-Pflanzenverfügbarkeit in einem schwachbelasteten Rieselfeldboden (RefB) nach dessen Einzelstoff- bzw. Mischanreicherung mit den genannten Schadstoffen in Konzentrationen, wie sie in hochbelasteten Rieselfeldern vorliegen, werden dargestellt. Desweiteren erfolgt eine Auswertung der Daten zu Erträgen und Schadstoffgehalten der auf den Versuchsvarianten geprüften Pflanzen (Roggen, Mais und Kartoffeln). Wechselbeziehungen der Schadstoffe untereinander im Pfad ² Boden-Pflanze² werden diskutiert. Auf allen Cu-Anreicherungsvarianten von RefB reagierten die drei Versuchspflanzen mit Ertragsdepressionen. Als Ursache hierfür und für die Störungen der mikrobiellen Aktivität im Boden konnte Cu als Einzelstoff erkannt werden. Die Ertragsdepressionen gingen mit erhöhten Cu-Gehalten aber auch mit erhöhten Cd-Gehalten der Pflanzen einher. Zugeführtes Cu bewirkte eine Erhöhung der Pflanzenverfügbarkeit von Cd im Boden. PCB 52 bzw. (und) BaP verursachten bei ihrer gleichzeitigen Zufuhr mit Cu dessen Mobilisierung im Boden und erhöhten damit die Cu-Pflanzenaufnahme. Neben PCB 52 dem Boden gegebenes Cd führte zu höheren PCB 52-Gehalten im Pflanzensproß. Die kombinierte Anreicherung von RefB mit Cd, PCB 52 und BaP bewirkte noch höhere PCB 52- und auch höhere BaP-Pflanzengehalte. / Together with polycyclic aromatic hydrocarbons (PAH), polychlorinated biphenyls (PCB) and heavy metals (HM) other elements and compounds as well as nutrients are present in typical contaminated areas like sewage fields. They are found as mixed undisturbed contamination and the transfer of these pollutants into food or forage plants is dangerous for people. So far investigations of ecological effects of these substances are focused mainly on single agents or on some agents of the same chemical group. But in most cases the mixture of different chemical compounds in extremely contaminated fields cause synergistic effects resulting in yield reductions and decreases of microbiological activity. Therefore the aim of the present work is to study combined effects of selected PAH-, PCB- and HM- (benzo-a-pyren [ BaP] , 2,2?,5,5? tetrachlorbiphenyl [ PCB 52] , cadmium [ Cd] und copper [ Cu] ) on parameters of soil biology, biomass production and pollutant uptake by plants in pot experiments. Resulting changes of cellulose decomposition, CO2-release from soil, N-mineralization and availability for plants of heavy metals after separated and combined enrichment of weackly polluted sewage field soil (RefB) with said pollutants up to concentrations of extremely polluted sewage field soils are presented. Data of yields and pollutant contents of plants (rye, maize and potatoes) cultivated on experimental soils are summarised. Interactions of pollutants ² soil-plant² are discussed. Cultivated plants showed yield reductions on all treatments enriched with copper. In this context copper only produced the impairments of microbial activity in the soil and caused its phytotoxicity. The yield reductions were connected with higher copper contents and also higher cadmium contents of plants. Added copper led to increasing availability for plants of cadmium in the soil. PCB 52 or (and) BaP together enriched with copper produced a mobilization of copper in the soil and a higher copper uptake by plants. Added cadmium caused in addition to PCB 52 higher PCB 52 contents of plants. The combined enrichment of RefB with Cd, PCB 52 and BaP led to higher contents of PCB 52 and BaP in plants.

Cette thèse a été consacrée à l'étude en laboratoire plusieurs molécules d'intérêt astrophysique par spectroscopie par transformation de Fourier dans l'infrarouge lointain afin de montrer les possibilités offertes par cette gamme spectrale en matière de spectroscopie vibrationnelle et rotationnelle, notamment grâce à l'utilisation du rayonnement synchrotron. Nous nous sommes intéressés à des molécules relativement lourdes, dérivées du naphtalène, pour lesquelles nous avons pu observer les bandes de vibrations de plus basses fréquences. Nous avons également développé différents dispositifs de décharge permettant d'étudier la spectroscopie rotationnelle de molécules instables : molécules légères à haute température et radicaux de petite taille.

Molecular Interactions in Thin Films of Biopolymers, Colloids and Synthetic Polyelectrolytes

The development of the layer-by-layer (LbL) technique has turned out to be an efficient way to physically modify the surface properties of different materials, for example to improve the adhesive interactions between fibers in paper. The main objective of the work described in this thesis was to obtain fundamental data concerning the adhesive properties of wood biopolymers and LbL films, including the mechanical properties of the thin films, in order to shed light on the molecular mechanisms responsible for the adhesion between these materials. LbLs constructed from poly(allylamine hydrochloride) (PAH)/poly(acrylic acid) (PAA), starch containing LbL films, and LbL films containing nanofibrillated cellulose (NFC) were studied with respect to their adhesive and mechanical properties. The LbL formation was studied using a combination of stagnation point adsorption reflectometry (SPAR) and quartz crystal microbalance with dissipation (QCM-D) and the adhesive properties of the different LbL films were studied in water using atomic force microscopy (AFM) colloidal probe measurements and under ambient conditions using the Johnson-Kendall-Roberts (JKR) approach. Finally the mechanical properties were investigated by mechanical buckling and the recently developed SIEBIMM technique (strain-induced elastic buckling instability for mechanical measurements). From colloidal probe AFM measurements of the wet adhesive properties of surfaces treated with PAH/PAA it was concluded that the development of strong adhesive joints is very dependent on the mobility of the polyelectrolytes and interdiffusion across the interface between the LbL treated surfaces to allow for polymer entanglements. Starch is a renewable, cost-efficient biopolymer that is already widely used in papermaking which makes it an interesting candidate for the formation of LbL films in practical systems. It was shown, using SPAR and QCM-D, that LbL films can be successfully constructed from cationic and anionic starches on silicon dioxide and on polydimethylsiloxane (PDMS) substrates. Colloidal probe AFM measurements showed that starch LbL treatment have potential for increasing the adhesive interaction between solid substrates to levels beyond those that can be reached by a single layer of cationic starch. Furthermore, it was shown by SIEBIMM measurements that the elastic properties of starch-containing LbL films can be tailored using different nanoparticles in combination with starch. LbL films containing cellulose I nanofibrils were constructed using anionic NFC in combination with cationic NFC and poly(ethylene imine) (PEI) respectively. These NFC films were used as cellulose model surfaces and colloidal probe AFM was used to measure the adhesive interactions in water. Furthermore, PDMS caps were successfully coated by LbL films containing NFC which enabled the first known JKR adhesion measurements between cellulose/cellulose, cellulose/lignin and cellulose/glucomannan. The measured adhesion and adhesion hysteresis were similar for all three systems indicating that there are no profound differences in the interaction between different wood biopolymers. Finally, the elastic properties of PEI/NFC LbL films were investigated using SIEBIMM and it was shown that the stiffness of the films was highly dependent on the relative humidity. / <p>QC 20110923</p>

Characterisation of soluble components and PAH in PM10 atmospheric particulate matter in Brisbane

Fours sets of PM10 samples were collected in three sites in SEQ from December 2002 to August 2004. Three of these sets of samples were collected by QLD EPA as a part of their regular air monitoring program at Woolloongabba, Rocklea and Eagle Farm. Half of the samples were used in this study for the analysis of water-soluble ions, which are Na+, K+, Mg2+, Ca2+, NH4 +, Cl-, NO3 -, SO4 2-, F-, Br-, NO2 -, PO4 -3 and the other half was retained by QLD EPA. The fourth set of samples was collected at Rocklea, specifically for this study. A quarter of the samples obtained from this set of samples were used to analyse water-soluble ions; a quarter of the sample was used to analyse Pb, Cu, Al, Fe, Mn and Zn; and the rests were used to analyse US EPA 16 priority PAHs. The water-soluble ions were extracted ultrasonically with water and the major watersoluble anions as well as NH4 + were analysed using IC. Na+, K+, Mg2+, Ca2+ Pb, Cu, Al, Fe, Mn and Zn were analysed using ICP-AES while PAHs were extracted by acetonitrile and analysed using HPLC. Of the analysed water-soluble ions, Cl-, NO3 -, SO4 2-, Na+, K+, Mg2+ and Ca2+ were high in concentration and determined in all the samples. F-, Br-, NO2 -, PO4 -3 and NH4 + ions were lower in concentration and determined only in some samples. Na+ and Cl- were high in all samples indicating the importance of a marine source. Principal Component Analysis (PCA) was used to examine the temporal variations of the water-soluble ions at the three sites. The results indicated that there was no major difference between the three sites. However, comparing the average concentrations of ions and Cl-/Na+ it was concluded that Woolloongabba had more marine influence than the other sites. Al, Fe and Zn were detected in all samples. Al and Fe were high in all samples indicating the significance of a source of crustal matter. Cu, Mn and Pb were in low concentrations and were determined only in some samples. The lower Pb concentrations observed in the study than in previous studies indicate that the phasing-out of leaded petrol had an appreciable impact on Pb levels in SEQ. This study reports for the first time, simultaneous data on the water-soluble, metal ion and PAH levels of PM10 aerosols in Brisbane, and provides information on the most likely sources of these chemical species. Such information can be used alongside those that already exist to formulate PM10 pollution reduction strategies for SEQ in order to protect the community from the adverse effects of PM pollution.

Caractérisation physico-chimique des particules issues du chauffage domestique au bois / Physicochemical characterization of the particles from domestic wood heating

La problématique des émissions de particules fines, posant des risques sanitaires et environnementaux avérés, a été très médiatisée ces dernières années. La solution mise en place consiste à aider financièrement les particuliers pour renouveler le parc d’appareils. Or, une utilisation non optimisée d’un appareil dégrade de manière significative ses émissions. Afin d’amender entre autres cette réflexion, de nombreuses campagnes expérimentales ont été menées au travers de caractérisations physico-chimiques complètes et innovantes des fumées, en particulier des particules fines : la Microscopie Electronique à Balayage, la microanalyse X et la détermination du type de carbone constitutif (Carbone Elémentaire, Carbone Organique). En premier lieu, la définition des paramètres d’utilisation de l’appareil et leurs niveaux de réponse ont permis de proposer une réduction significative des émissions dont celles de particules fines. Ainsi, les impacts des paramètres comme la nature du combustible, le mode d’allumage et l’injection d’air secondaire ont été quantifiés. Ensuite, les mécanismes de formations des particules ont été définis. Dans un second temps, ce travail de thèse s’est intéressé à l’évolution des caractéristiques des particules entre la chambre de combustion de l’appareil et le champ proche, permettant d’aller au-delà de l’étude normative classique. De nouveaux savoirs sur les particules ont ainsi été mis en évidence. L’ensemble de ces nouvelles connaissances, aidant à mieux connaître les particules produites par la combustion de bois dans des appareils indépendants, permettront dans le futur de créer des systèmes de traitement spécifiques et efficaces / The issue of the emissions of fine particles, known for their health and environmental hazards, has been significantly covered in recent years. The governments made the decision to help mainly financially the private individuals in order to renew the domestic appliances stock. However, a non-optimized use of an energy-efficient appliance can degrade system performances. In order to solve this issue, many experimental campaigns have been achieved through complete and innovative physicochemical characterizations of the smoke, especially of the fine particles : Scanning Electron Microscope, X-ray microanalysis and assessment of the constitutive Carbon (Elemental Carbon, Organic Carbon). First of all, the definition of the use parameters for the appliance and their impacts allowed offering a significant reduction of the fine particles emissions. In this work, the influences of the parameters such as the nature of the fuel, the ignition mode and the air secondary injection were in this way quantified. In order to better understand the origin of these emissions produced regardless of the combustion quality, the mechanisms of formation of all kinds of particles were defined. In a second phase, this work paid attention to the evolution of the characteristics of particles from the combustion room to the near-field. This enabled to go over the classic normative study. New knowledge on particles was brought to light, helping to better know the particles generation from domestic wood appliances. This could allow in the future to create some specific and efficient treatment systems

Desenvolvimento e validação de método para determinação de hidrocarbonetos policíclicos aromáticos em caranguejo-uçá (ucides cordatus)

Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The mangrove plays an important role for the man since prehistoric times because of the abundance of food resources it provides, in addition to being a natural breeding that serves as a shelter for several species of animals, including uçá-crab, which is great importance for the maintenance of this ecosystem. However, this environment is the final destination of many pollutants, including polycyclic aromatic hydrocarbons (PAH). The aim of this work is to develop and validate a method using the extraction technique for matrix solid-phase dispersion (MSPD) and gas chromatography coupled with mass spectrometry (GC/MS) for the determination of priority PAH in hepatopancreas of uçá-crab (Ucides cordatus). The best extraction conditions were obtained using 0.5 g hepatopancreas, 1.5 g florisil as a dispersant and 0.5 g sodium sulfate as the drying agent, along with 0.5 g of C18 as an auxiliary adsorbent, and eluting with 10 mL of acetonitrile. The analytical validation was performed from the parameters: selectivity; linearity, which was estimated from the correlation coefficient, with values greater than 0.990 for all PAH in a concentration range of 5 ng mL-1 to 100 ng mL-1; limits of detection and quantification in the intervals of 0.2 to 3.0 ng g-1 and 0.6 to 9,8 ng g-1 respectively; accuracy and precision, with recovery values between 60-111% and coefficients of variation in the 1-19% range for concentration levels of 10, 25 and 50 ng mL-1; and robustness, which was assessed by external standard, being robust for most of the HPA tested, with the exception of naphthalene. For the application of validated method were collected uçá-crab from mangroves located in the Gameleiro rivers (P1), Vaza-Barris (P2), Poxim (P3) and Sergipe (P4). At point P3 found a concentration of 26.6 ng g -1 of benzo[a]anthracene , which is considered as "probably carcinogenic". Mangrove sediments were also collected the same points and extracted by a method already validated, which is based on ultrasound extraction, and analyzed by GC/MS. The concentration values found for ΣPAH ranged from 12.5 to 813.6 ng g-1 of dry sediment. / O manguezal tem um papel importante para o homem desde a pré-história em razão da abundância de recursos alimentares que fornece, além de constituir um criadouro natural que serve de abrigo para diversas espécies de animais, entre eles o caranguejo-uçá, que é de grande importância para a manutenção desse ecossistema. Contudo, esse ambiente é destino final de muitos contaminantes, entre eles os hidrocarbonetos policíclicos aromáticos (HPA). O objetivo desse trabalho é desenvolver e validar um método, utilizando a técnica de extração por dispersão da matriz em fase sólida (MSPD) e cromatografia gasosa acoplada a espectrometria de massas (GC/MS), para a determinação de HPA prioritários em hepatopâncreas do caranguejo-uçá (Ucides cordatus). As melhores condições de extração foram obtidas utilizando 0,5 g do hepatopâncreas, 1,5 g de florisil como dispersante e 0,5 g de sulfato de sódio como agente secante, juntamente com 0,5 g de C18 como adsorvente auxiliar e eluição com 10 mL de acetonitrila. A validação analítica foi realizada através da obtenção dos parâmetros de: seletividade; linearidade, que foi estimada a partir do coeficiente de correlação, apresentando valores maiores do que 0,990 para todos os HPA, numa faixa de concentração de 5 ng mL-1 a 100 ng mL-1 ; limites de detecção e quantificação, nos intervalos de 0,2 a 3,0 ng g-1 e 0,6 a 9,8 ng g-1 respectivamente; exatidão e precisão, com valores de recuperação variando entre 60-111% e coeficientes de variação na faixa de 1-19%, para níveis de concentração 10, 25 e 50 ng mL-1; e robustez, que foi avaliada por padronização externa, mostrando-se robusto para a maioria dos HPA testados, com exceção do naftaleno. Para a aplicação do método validado foram coletados caranguejos-uçá de mangues localizados nos rios Gameleiro (P1), Vaza-Barris (P2), Poxim (P3) e Sergipe (P4). No ponto P3 foi encontrada uma concentração de 26,6 ng g-1 do benzo[a]antraceno, que é considerado como “provavelmente cancerígeno” ao homem. Os sedimentos de mangue também foram coletados nos mesmos pontos e extraídos por um método já validado, no qual se baseia na extração por ultrassom, e analisados por GC/MS. Os valores de concentração encontrados para o ΣHPA variaram entre 12,5 – 813,6 ng g-1 de sedimento seco.

Determinação de compostos orgânicos no aerossol atmosférico em sítios da América do Sul / Determination of organic compounds in atmospheric aerosol from South America sites

Centenas de compostos orgânicos são emitidos para a atmosfera por uma variedade de fontes biogênicas e antrópicas. Essa mistura complexa causa preocupação devido ao impacto que esses compostos podem provocar na saúde e no ambiente. Apesar de constituírem 10-70% da massa do aerossol atmosférico, a caracterização dos compostos orgânicos particulados permanece ainda deficitária. A contribuição das principais fontes de emissão para a poluição a nível regional pode ser diagnosticada através da utilização de marcadores moleculares específicos. De acordo com esta necessidade, o objetivo do presente trabalho é a caracterização de compostos orgânicos do aerossol atmosférico coletado em sítios de cidades da América do Sul. As amostragens do material particulado (MP10) foram realizadas em duas áreas em Bogotá na Colômbia, uma de influência industrial (BOI) e outra veicular (BOV) em 2007, e em áreas urbanas em São Paulo no Brasil em 2007 (SPA 07) e 2008 (SPA 08), e em Buenos Aires (BAI) na Argentina em 2008. Diferentes classes de compostos orgânicos foram determinadas: n-alcanos e hidrocarbonetos policíclicos aromáticos (HPA) por cromatografia a gás com detecção por ionização em chama (CG-DIC), n-alcanais e n-alcanonas por cromatografia a gás com detecção por espectrometria de massas (CG/EM). A composição e concentração do aerossol atmosférico foram afetadas substancialmente pelo transporte de massas provenientes de regiões com queima de biomassa e pelas condições meteorológicas locais. Em SPA 08, BOI e BAI foram encontradas as maiores concentrações de MP10 onde as amostragens foram realizadas em época de seca, favorecendo a acumulação dos poluentes. A constituição orgânica do MP10 nos sítios estudados reflete a incorporação nas amostras de marcadores moleculares biogênicos como n-alcanos com número ímpar de carbonos. Dentre os traçadores antrópicos, foram identificadas a presença de pristano e fitano, a mistura complexa não-resolvida (MCNR) e HPA de queima de combustíveis fósseis. Em SPA 08 foi identificada a presença do reteno, um traçador da queima de biomassa. As maiores concentrações de n-alcanais e n-alcanonas foram observadas no sítio BOV. Os n-alcanais (< C20) identificados nos sítios deste trabalho podem ser originados a partir da oxidação de alcanos e alcenos, e as n-alcanonas (< C20) podem ter como origem atividades antrópicas, processos oxidativos na atmosfera ou atuação microbiana sobre outros compostos. / Hundreds of organic compounds are emitted to the atmosphere by biogenic and anthropogenic sources. This complex mixture generates concern due to the impact of these compounds on health and environment. In spite of accounting for 10-70% of the atmospheric aerosol mass, particulate-phase organic compounds are not yet well characterized. The contribution of the major emission sources to regional particulate pollution can be diagnosed by using specific molecular markers. According to this necessity, the present work has as objective the characterization of organic compounds from atmospheric aerosol collected in sites of South American cities. The particulate matter (PM10) sampling were accomplished in two areas in Bogota (Colombia), one with industrial (BOI) and other with vehicular (BOV) influence in 2007, and in urban areas in Sao Paulo (Brazil) in 2007 (SPA 07) and 2008 (SPA 08), and in Buenos Aires (BAI), Argentina, in 2008. Different organic classes were determined: n-alkanes and polycyclic aromatic hydrocarbons (PAH) by gas chromatograph with flame ionization detection (GC-FID), n-alkanals and n-alkanones by gas chromatograph with mass spectrometer detection (CG/MS). The aerosol atmospheric composition and concentration were substantially affected by mass transport from biomass burning regions and by the local meteorological conditions. In SPA 08, BOI and BAI were found the highest PM10 concentrations where the samplings were accomplished in dry period, contributing for the pollutants accumulation. The PM10 organic composition at studied sites reflects the incorporation from biogenic molecular markers in the samples like n-alkanes with odd carbon number. Among the anthropogenic tracers, were identified the presence of pristane and phytane, the unresolved complex mixture (UCM) and PAH from fossil fuel combustion. In SPA 08 was observed the presence of retene, a biomass burning tracer. Higher concentration from n-alkanals and n-alkanones were observed at the BOV site. The n-alkanals (< C20) identified at sites from this work may be originated from oxidation of alkanes and alkenes, and n-alkanones (< C20) are derived from anthropogenic activity, atmospheric oxidative processes or microbial action over other compounds.

Determinação de HPA em ambiente ocupacional de indústria de cimento que co-processa resíduos / Determination of PAH in occupational environment of a cement industry co-processing residues

O co-processamento de resíduos em fornos de cimenteiras é uma alternativa de destinação de resíduos que consiste em substituir combustíveis ou matérias primas usados na fabricação do clínquer por diversos resíduos que apresentem poder calorífico e propriedades similares. Os resíduos são co-processados a temperaturas altas, porém podem ocorrer emissões de diversos poluentes devido a variedade de resíduos que podem ser co-processados. Entre esses poluentes estão os Hidrocarbonetos Policíclicos Aromáticos (HPA), que podem ser emitidos nas chaminés dos fornos das cimenteiras, ou também podem estar presentes no ambiente interno das mesmas. Considerando que os HPA são compostos orgânicos que apresentam potencial carcinogênico e/ou mutagênico, e assim oferecem risco à saúde humana, esse estudo tem o objetivo apresentar uma avaliação preliminar da presença e dos níveis de concentração de HPA em uma indústria de cimento que co-processa resíduos. As análises de HPA no ambiente ocupacional foram realizadas seguindo a metodologia proposta do NIOSH (National Institute for Occupational Safety and Health). Para isso utilizou-se como ferramenta analítica a técnica de cromatografia a líquido de alto desempenho (CLAE) com detecção por fluorescência, acompanhada da otimização e avaliação dos principais parâmetros analíticos do método, possibilitando assim a determinação dos 17 HPA prioritários pela NIOSH com confiabilidade analítica. Os resultados indicaram a presença de vários HPA no ambiente de trabalho estudado, porém numa concentração baixa (faixa de ng.m-3) e dentro da faixa de concentração encontrada em trabalhos em ambientes convencionais. Este estudo foi desenvolvido com a colaboração da FUNDACENTRO/MTE. / The co-processing of residues in furnaces of cement industries is an alternative to dispose residues by substituting conventional combustibles and raw materials used for production of clinker by diverse organic residues of similar properties and calorific power. Co-processing is made at high temperatures, but emission of diverse pollutants may occur as a consequence of the large diversity of residues that are processed. Among these pollutants, Polycyclic Hydrocarbons (PAH) may be either emitted in the chimneys of the furnaces, or be present in the interior ambient of these industries. PAH pose risk to human health because they are organic compounds having potential carcinogenic and mutagenic activities. The present study aims to show the results of a preliminary evaluation of the presence and concentration levels of PAH in a cement industry co-processing residues. PAH analyses in the occupational environment were made according the methodology recommended by NIOSH (National Institute of Occupational Safety and Health). High performance chromatography (HPLC) with fluorescence detection was the analytical technique used for analyses. Optimization and evaluation of the major analytical parameters allowed the determination of the 17 PAH prioritized by NIOSH to be made with analytical confidence. Results showed the presence of several PAH in the studied environment, but at low concentrations (in the range of ng.m-3). These concentrations are of similar magnitude to the ones found in conventional environments. The present study was made in collaboration with FUNDACENTRO/MTE.
