Efeitos da inibição crônica das óxido nítrico sintases na mecânica de tecido periférico, no recrutamento eosinofílico e no remodelamento da matriz extracelular induzida por inflamação crônica pulmonar / Effects of chronic nitric oxide inhibition on lung tissue mechanics, eosinophilic and extracellular matrix responses induced by chronic pulmonary inflammation

INTRODUÇÃO: A importância da resposta mecânica do parênquima pulmonar na fisiopatologia da asma tem sido recentemente reconhecida. O óxido nítrico é um mediador que controla o tônus muscular liso das vias aéreas, porém este efeito no parênquima pulmonar periférico ainda não foi previamente investigado. Nossa hipótese é que a inibição crônica das óxido nítrico sintases por meio do tratamento com L-NAME (falso substrato para todas as óxido nítrico sintases) pode modular a mecânica do parênquima pulmonar, o recrutamento eosinofílico e o remodelamento da matriz extracelular em modelo de inflamação alérgica crônica pulmonar em cobaias. MÉTODOS: Os animais foram expostos a sete inalações com soro fisiológico ou com ovoalbumina em doses crescentes (1~5mg/ml - 4 semanas) e tratadas ou não com L-NAME (60 mg/kg/ por dia /por animal) na água de beber. Setenta e duas horas após a sétima inalação os animais foram anestesiados, exsanguinados e a mecânica oscilatória do parênquima pulmonar foi medida na condição pré e após desafio (0.1%). Utilizando a técnica de morfometria foram avaliadas a densidade de eosinófilos, o número de células nNOS e iNOS positivas, a densidade de actina, das fibras colágenas e das fibras elásticas bem como a proporção de volume de 8-iso-PGF2 no septo alveolar. RESULTADOS: Os animais que foram expostos à ovoalbumina apresentaram um aumento da resistência e da elastância tecidual (resposta basal e após desafio antigênico), na densidade de eosinófilos, no número de células nNOS e iNOS positivas, na densidade de fibras colágenas e de fibras elásticas bem como na expressão de 8-isoPGF2 no septo alveolar comparativamente aos grupos controles (p<0,05). O tratamento com L-NAME em animais expostos à ovoalbumina atenuou todas as respostas de mecânica do tecido pulmonar periférico (p<0, 01), reduziu o número de células nNOS e iNOS positivas (p<0.01), o conteúdo de fibras elásticas (p<0,001) e de 8-iso-PGF2 no septo alveolar (p<0,001). No entanto, este tratamento não afetou o número total de eosinófilos e o conteúdo de fibras colágenas. Este trabalho sugere que o óxido nítrico contribui para a constrição do parênquima pulmonar e para a deposição de fibras elásticas neste modelo. Estes efeitos foram associados à ativação de iNOS e nNOS em células do parênquima distal e aumento na via do estresse oxidativo / The importance of lung tissue mechanical responses in asthma pathophysiology has been recently recognized. Although nitric oxide (NO) is a mediator that controls smooth muscle tonus control in the airways, its effects on lung tissue responsiveness has not been previously investigated. We hypothesized that chronic nitric oxide synthase inhibition by L-NAME (false substrate for all nitric oxide synthases) treatment may modulate lung tissue mechanics, eosinophilic recruitment and extracellular matrix remodeling in a model of chronic pulmonary allergic inflammation. Guinea pigs were submitted to seven normal saline or ovalbumin exposures with increasing doses (1~5mg/mL-4weeks) and treated or not with L-NAME in drinking water. Seventy-two hours after the seventh inhalation the animals were anesthetized, exsanguinated, and oscillatory mechanics of lung tissue strips was performed in baseline condition and after ovalbumin challenge (0.1%). Using morphometry, we assessed the density of eosinophils, the number of iNOS and nNOS-positive cells, the density of actin, the collagen and elastic fibers content and the volume proportion of 8-iso-PGF2 in the alveolar septa. Ovalbumin-exposed animals presented an increase in baseline and maximal tissue resistance and elastance responses, eosinophil density, in the number of iNOS and nNOS positive cells, in the amount of collagen and elastic fibers and in the volume proportion of 8-iso-PGF2 in the alveolar septa compared to controls (p<0.05). L-NAME treatment in ovalbumin-exposed animals attenuated all lung tissue mechanical responses (p<0.01), reduced the number of iNOS and nNOS positive cells (p<0.01), elastic fiber content (p<0.001) and 8-isoPGF2 in the alveolar septa (p<0.001). However, this treatment did not affect the total number of eosinophils and collagen deposition. These data suggest that NO contributes to distal lung parenchyma constriction and to elastic fibers deposition in this model. These effects were associated to iNOS and nNOS activation in pulmonary parenchyma and with an increase in oxidative stress pathway activation

Transplante experimental, subcutâneo e intraperitoneal, de ovário em suínos: estudo histomorfométrico e imunoistoquímico / Experimental transplantation, subcutaneous and intraperitoneal, ovary in pigs: immunohistochemical and histomorphometric study

O transplante autólogo de tecido ovariano constitui alternativa relevante na preservação da fertilidade e da função hormonal ovariana em mulheres sujeitas à falência ovariana prematura e infertilidade, por causas malignas, tratamentos adjuvantes ou cirurgias. É a única opção para crianças, fase pré-puberal e para mulheres que não podem retardar a quimioterapia ou não podem ser submetidas à estimulação do ciclo. O transplante ovariano autólogo pode ser, quanto ao local de reimplantação, ortotópico ou heterotópico e, quanto à conservação, a fresco ou após o período de criopreservação. As várias etapas envolvidas neste transplante são estudadas mundialmente na atualidade, como a retirada e preservação do tecido ovariano, as técnicas de criopreservação, o local apropriado para o reimplante e as possibilidades de redução da perda folicular. A avaliação da apoptose - morte celular programada - é útil na avaliação da rejeição e viabilidade dos enxertos de transplantes estabelecidos na prática clínica, tanto autólogos como heterólogos. Com o intuito de utilizar animais de maior porte, conseguir seguimento de médio prazo e realizar os procedimentos cirúrgicos por via laparoscópica, padrão ouro em humanos, o presente estudo utilizou como modelo experimental fêmeas suínas, em idade reprodutiva, da raça Minipig. Este projeto teve como propósito avaliar a influência da criopreservação e do local de implante na qualidade e na viabilidade do transplante autólogo de ovário, a fresco e após criopreservação, no tecido celular subcutâneo e na região intraperitoneal peri-infundibular. Foram avaliados a quantidade e a densidade folicular dos implantes e os aspectos morfológicos e histomorfométricos, bem como a apoptose, por meio da imunoexpressão de proteínas proapoptóticas- Bax e antiapoptóticas-Bcl-2, além da Caspase 3-clivada, fase final das vias extrínseca e intrínseca dos mecanismos de apoptose.Quarenta animais foram divididos em cinco grupos: Controle com ooforectomia (Grupo I), ooforectomia e transplante a fresco subcutâneo (Grupo II), a ooforectomia e transplante fresco intraperitoneal (Grupo III), ooforectomia e transplante criopreservado subcutâneo (Grupo IV) e ooforectomia e transplante criopreservado intraperitoneal (GrupoV). Os resultados mostraram que independente da técnica empregada, havia folículos em desenvolvimento e corpos lúteos em todos os tecidos ovarianos transplantados; que a contagem de folículos antrais não degenerados foi menor nos grupos após criopreservação em relação ao grupo controle e que a imunoexpressão sugestiva de apoptose ocorreu em todos os grupos transplantados, sendo maior nos transplantes intraperitoneais. Concluiu-se que a técnica utilizada para o transplante de ovário e criopreservação foi viável no modelo suíno, em tecido celular subcutâneo e na região intraperitoneal peri-infundibular. O transplante autólogo heterotópico subcutâneo apresentou melhores taxas de apoptose que o transplante ortotópico. / Autotransplantation of ovarian tissue is an important alternative to preserve fertility and hormonal ovarian function in women undergoing ovarian failure and premature infertilidade, because of cancer or surgery. It is the only option for infants, pre-pubertal patients and for women who can not delay chemotherapy or not may be subjected to stimulation of the cycle. The various steps involved in the transplant are studied worldwide today, as the removal and preservation of ovarian tissue, the techniques of cryopreservation, the appropriate site and mechanisms to reduce follicular loss. Assessment of apoptosis - programmed cell death-is useful in the study of the viability of the grafts and rejection of transplants established in clinical practice, both autologous and heterologous. In order to use larger animals, getting following medium term (over 21 days) and to perform surgical procedures by laparoscopy (gold standard in humans), this study used an experimental model sows, reproductive age, Minipig race. This project aims to evaluate the influence of cryopreservation and implantation site of the quality and viability of ovarian autografts, fresh and after cryopreservation, at subcutaneous site and at intraperitoneal site. We analyzed the quantity and density of follicular implants and the morphological and histomorphometric as well as apoptosis, by proteins immunoexpression antiapoptotic and proapoptotic. Forty animals were divided into five groups: Control with oophorectomy (Group I), oophorectomy and fresh transplantation to subcutaneous site (Group II), oophorectomy and fresh transplantation to intraperitoneal site (Group III), oophorectomy and transplantation of cryopreserved ovarian tissue to subcutaneous site (Group IV) and oophorectomy and transplantation of cryopreserved ovarian tissue to intraperitoneal site (Group V). We concluded that the autologous ovarian transplantation was feasible in the technical proposals, in subcutaneous and intraperitoneal site in the porcine model; that regardless of the technique, there was developing follicles and corpora lutea in all ovarian tissue transplanted; that antral non-degenerate follicle count was lower in groups after cryopreservation that in the control group and that the immunoexpression of apotposis occurred in all transplanted groups, more evident in intraperitoneal transplants

A biodisponibilidade de ferro de diferentes fontes foi avaliada pelo método de recuperação de hemoglobina em suínos anêmicos. O ensaio teve duração de 13 dias e os grupos foram homogeneizados de acordo com o produto do peso (kg) x hemoglobina (g/dL). As fontes de ferro estudadas foram: ferro microencapsulado com polímero de carboximetilcelulose sódica (NaCMC), ferro microencapsulado com polímero de alginato, ferro quelado com metionina e ferro reduzido eletrolíticamente. Com a finalidade de corrigir respostas devidas à variação na ingestão de ferro foram acrescentados três grupos chamados de grupos-padrão, cuja fonte dietética de ferro foi FeSO4.7H2O. A porcentagem média de absorção de ferro para os grupos experimentais variou de 12,8 (4,3) % (ferro eletrolítico) a 15,1 (3,8) % (ferro alginato), não sendo significativamente diferentes (P>0,05). A porcentagem média de absorção relativa ao FeSO4.7H2O variou de 89,5 (25,9)% (ferro NaCMC) a 105,9 (60,5)% (ferro quelado com metionina), sem diferenças significativas (p>0,05). Também não houve diferença significativa nos índices de conversão alimentar, eficiência alimentar e coeficiente de eficácia proteica (CEP). Não houve diferenças significativas (p>0,05) nas concentrações de ferro hepático entre as diferentes fontes estudadas. Nas condições deste ensaio, houve similaridade entre as fontes de ferro testadas quanto à biodisponibilidade. A microencapsulação proporcionou rneltlOr distribuição do produto na mistura quando comparada ao FeSO4.7H2O. / The iron availability from different sources was evaluated by the hemoglobin regeneration method in anemic pigs. The assay took 13 days and the animais were selected according to the weight (kg) x hemoglobina (g/dL) product. The iron sources studied were: microencapsulated iron with sodium carboximetilcelulose polymer (NaCMC), microencapsulated iron with alginato polymer, metionin-quelated iron and electrolytically reduced iron. To COITect answers due to iron ingestion variation three control groups were added, whose dietary iron source was FeSO4.7H2O. The average percentage of iron absorption for the experimental groups ranged from 12,8 (4,3) % (electrolytic iron) to 15,1 (3,8) % (iron alginato), without significant differences (p>0,05). The average percentage of FeSO4.7H2O relative absorption ranged from 89,5 (25.9)% (NaCMC iron) to 105,9 (60,5)% (metionin quelated-iron), without significant differences (p>O,OS). Also it did not have significant difference in the alimentary conversion and efficiency indexes and proteic effectiveness coefficient (CEP). No significant differences (p>0,05) were found in the hepatic iron concentrations among the different studied sources. Based on this assay\'s conditions, there were similarities among the tested iron sources regarding to their bioavailability. Microencapsulation provided better distribution of the product in the mixture when compared to the FeSO4.7H2O.

Quantificação à beira do leito do potencial de recrutamento alveolar através da tomografia de impedância elétrica em modelo experimental síndrome do desconforto respiratório agudo / Bedside quantification of alveolar recruitment potential using electrical impedance tomography in an experimental model of acute respiratory distress syndrome

Introdução: A síndrome do desconforto respiratório agudo (SDRA) tem como parte do seu tratamento uma ventilação mecânica adequada. A manobra de recrutamento alveolar (MRA) faz parte de um grupo de estratégias empregadas nos quadros moderados e graves. A MRA consiste na aplicação de um aumento de pressão transitório e controlado nas vias aéreas no intuito de abrir alvéolos previamente colapsados. A Tomografia de Impedância Elétrica (TIE) é capaz de avaliar o potencial de recrutamento alveolar (PRA) a beira leito. Objetivos:1) Comparar o PRA calculado pela TIE (através do ganho de complacência regional) com a TC. 2) Avaliar o \"deslocamento vertical de volume\" como índice de recrutamento alveolar.3) Estimar precocemente o PRA, através das manobras de recrutamento de rastreio propostas, utilizando a TIE. Método: Avaliamos o PRA em um modelo experimental de SDRA. Utilizamos 15 suínos da raça Landrace. Os animais foram sedados e intubados, em seguida, submetidos ao modelo experimental de SDRA desenvolvido na Faculdade de Medicina da Universidade de São Paulo (LIM-09). Ao término da lesão, um grupo de 7 animais recebeu uma sequência randomizada de manobras de recrutamento de rastreio propostas (Pressões inspiratórias de 30, 35 e 40 cmH2O) seguidas da manobra de recrutamento máxima (Pressão inspiratória de 60 cmH2O). Os animais foram monitorados com TIE e Tomografia computadorizada por raio X (TC) durante todas as manobras de recrutamento. Outro grupo de 8 animais ,submetidos a mesma lesão e com medidas de TC e TIE, foi retirado do banco de dados do LIM-09 e também analisados. Utilizamos o programa IBM® SPSS® Statistics 9.0 e 20.0.Resultados:1) PRA calculado pela TIE atráves do ganho de complacência regional corrigido para hiperdistensão, comparado com TC, apresentou um R2=0,76. 2) PRA calculado pelo ganho de complacência regional corrigido para hiperdistensão combinado com o deslocamento vertical apresentou R2 = 0,91 comparado a TC .3) As manobras de rastreio não conseguiram predizer quantitativamente o PRA mas auxiliariam na correção da hiperdistensão. Conclusões: A TIE é capaz de avaliar o recrutamento alveolar a beira leito. O deslocamento vertical combinado com o ganho de complacência regional corrigida para hiperdistensão representam de forma semelhante a TC o comportamento pulmonar durante uma MRA. A aplicação de uma manobra de recrutamento de rastreio pode ser útil para uma manobra de recrutamento máxima mais segura / Introduction: The acute respiratory distress syndrome (ARDS) treatment demands a proper mechanical ventilation strategy. The alveolar recruitment maneuver (ARM) is an intervention applied in moderate and severe cases of ARDS. ARM is a transitory and controlled increase in mechanical ventilator pressure delivered to the lungs aiming to open previously collapsed alveoli. The electrical impedance tomography (EIT) is a valuable tool at bedside; it is able to monitor and to help during an ARM performance through the estimation of the alveolar recruitment potential (ARP). Objectives: 1) To compare the ARP with the EIT as a regional compliance improvement quantification adjusted for lung hyperdistention with CT. 2) To evaluate the \"volume vertical displacement\" at the same pressure as alveolar recruitment index using EIT and CT. 3) To estimate earlier the ARP using the EIT through a screening recruitment maneuver. Methods: We evaluated the ARP in an experimental model of ARDS. We studied 15 Landrace race pigs. Subjects were sedated, intubated and submitted to the ARDS experimental model developed at Medical investigation laboratory n ° 09, University of São Paulo. In the end of the lung injury, a group of 7 pigs received a randomized sequence of screening recruitment maneuvers (inspiratory pressures of 30, 35 and 40 cmH2O) followed by a maximum recruitment maneuver (inspiratory pressure of 60 cmH2O).EIT and x-ray computed tomography (CT) monitored the steps of each recruitment maneuver. Another group of 8 pigs, submitted to the same lesion and with measures of EIT and CT, were extracted from our data bank. Analysis was performed at IBM® SPSS® Statistics 20.0. Results: 1) ARP calculated by EIT (regional compliance improvement quantification) reached a R2=0,76 when compared to CT. 2) The combination of regional compliance improvement and volume vertical displacement obtained R2 = 0,91 when compared to CT 3) The screening recruitment maneuvers were not able to predict quantitatively the ARP, but they helped in the lung hyperdistension adjustment. Conclusions: EIT is able to evaluate the ARP at bedside. The combination of regional compliance improvement and volume vertical displacement give information similar to CT about the lung behavior during a ARM. The application of a recruitment screening maneuver might be useful for more safe ARM

Objetivos:1) Comparar o PRA calculado pela TIE (através do ganho de complacência regional) com a TC. 2) Avaliar o \"deslocamento vertical de volume\" como índice de recrutamento alveolar.3) Estimar precocemente o PRA, através das manobras de recrutamento de rastreio propostas, utilizando a TIE. Método: Avaliamos o PRA em um modelo experimental de SDRA. Utilizamos 15 suínos da raça Landrace. Os animais foram sedados e intubados, em seguida, submetidos ao modelo experimental de SDRA desenvolvido na Faculdade de Medicina da Universidade de São Paulo (LIM-09). Ao término da lesão, um grupo de 7 animais recebeu uma sequência randomizada de manobras de recrutamento de rastreio propostas (Pressões inspiratórias de 30, 35 e 40 cmH2O) seguidas da manobra de recrutamento máxima (Pressão inspiratória de 60 cmH2O). Os animais foram monitorados com TIE e Tomografia computadorizada por raio X (TC) durante todas as manobras de recrutamento. Outro grupo de 8 animais ,submetidos a mesma lesão e com medidas de TC e TIE, foi retirado do banco de dados do LIM-09 e também analisados. Utilizamos o programa IBM® SPSS® Statistics 9.0 e 20.0.Resultados:1) PRA calculado pela TIE atráves do ganho de complacência regional corrigido para hiperdistensão, comparado com TC, apresentou um R2=0,76. 2) PRA calculado pelo ganho de complacência regional corrigido para hiperdistensão combinado com o deslocamento vertical apresentou R2 = 0,91 comparado a TC .3) As manobras de rastreio não conseguiram predizer quantitativamente o PRA mas auxiliariam na correção da hiperdistensão. Conclusões: A TIE é capaz de avaliar o recrutamento alveolar a beira leito. O deslocamento vertical combinado com o ganho de complacência regional corrigida para hiperdistensão representam de forma semelhante a TC o comportamento pulmonar durante uma MRA. A aplicação de uma manobra de recrutamento de rastreio pode ser útil para uma manobra de recrutamento máxima mais segura / Introduction: The acute respiratory distress syndrome (ARDS) treatment demands a proper mechanical ventilation strategy. The alveolar recruitment maneuver (ARM) is an intervention applied in moderate and severe cases of ARDS. ARM is a transitory and controlled increase in mechanical ventilator pressure delivered to the lungs aiming to open previously collapsed alveoli. The electrical impedance tomography (EIT) is a valuable tool at bedside; it is able to monitor and to help during an ARM performance through the estimation of the alveolar recruitment potential (ARP). Objectives: 1) To compare the ARP with the EIT as a regional compliance improvement quantification adjusted for lung hyperdistention with CT. 2) To evaluate the \"volume vertical displacement\" at the same pressure as alveolar recruitment index using EIT and CT. 3) To estimate earlier the ARP using the EIT through a screening recruitment maneuver. Methods: We evaluated the ARP in an experimental model of ARDS. We studied 15 Landrace race pigs. Subjects were sedated, intubated and submitted to the ARDS experimental model developed at Medical investigation laboratory n ° 09, University of São Paulo. In the end of the lung injury, a group of 7 pigs received a randomized sequence of screening recruitment maneuvers (inspiratory pressures of 30, 35 and 40 cmH2O) followed by a maximum recruitment maneuver (inspiratory pressure of 60 cmH2O).EIT and x-ray computed tomography (CT) monitored the steps of each recruitment maneuver. Another group of 8 pigs, submitted to the same lesion and with measures of EIT and CT, were extracted from our data bank. Analysis was performed at IBM® SPSS® Statistics 20.0. Results: 1) ARP calculated by EIT (regional compliance improvement quantification) reached a R2=0,76 when compared to CT. 2) The combination of regional compliance improvement and volume vertical displacement obtained R2 = 0,91 when compared to CT 3) The screening recruitment maneuvers were not able to predict quantitatively the ARP, but they helped in the lung hyperdistension adjustment. Conclusions: EIT is able to evaluate the ARP at bedside. The combination of regional compliance improvement and volume vertical displacement give information similar to CT about the lung behavior during a ARM. The application of a recruitment screening maneuver might be useful for more safe ARM

Pathogenesis and clinical significance of AIDA-I-positive <i>E. coli</i> in diarrhea of pigs

AIDA-I adhesin and its gene aidA were first identified and characterized in <i>E. coli</i> isolated from a human case of infantile diarrhea. Recent studies have demonstrated a significant degree of homology between the AIDA-I adhesin isolated from porcine neonatal diarrheagenic <i>E. coli</i> isolates and that from a human <i>E. coli</i> isolate; however, the role of AIDA-I adhesin in the pathogenesis of diarrhea and the clinical significance of the AIDA-I <i>E. coli</i> virotype are unknown in humans or in animals. <p>First, in order to evaluate the role of AIDA-I adhesin, colostrum deprived newborn pigs were infected with: i) a wild strain PD20 (AIDA-I+/STb+) <i>E. coli</i>; ii) a mutant strain PD20M (AIDA-I-/STb+), generated by partial deletion of the aidA gene from the wild strain, iii) a complemented strain PD20C (AIDA-I+/STb+), generated by reintroducing the full length aidA gene into PD20M strain, and iv) a nonpathogenic <i>E. coli</i> strain PD71 used as negative control. Pigs infected with wild type (PD20) and complemented (PD20C) strains developed diarrhea between 15-19 h and 27-31 h after oral inoculation, respectively, in contrast to pigs infected with strains PD20M or PD71 that did not developed diarrhea. Intestinal colonization was evaluated by histology, imunohistochemistry (IHC), transmission electron microscopy (TEM), including immunogold electron microscopy (IGEM), and showed heavy bacterial colonization with biofilm formation in the large intestine with AIDA-I+ strains (PD20 and PD20C), but not in pigs infected with AIDA-I- strains (PD20M and PD71). In vitro assays showed marked diffuse adherence to HeLa cells, enhanced bacterial autoaggregation and significant biofilm formation by AIDA-I+ strains, when compared to AIDA-I- strains.<p>Second, 110 F4 negative <i>E. coli</i> isolates from problematic cases of diarrhea in pigs were subjected to multiplex polymerase chain reaction (M-PCR) for detection of the genes encoding the virulence factors F4, F5, F6, F18, F41, AIDA-I, EAE, STa, STb, LT, EAST1 and Stx2e. In this study, the prevalence of aidA gene among the 110 isolates was 8.2%, and the aidA gene was shown to be associated most commonly with EAST1 and STb genes. The genes for the F4, F5, F6 and F41 fimbriae were absent in all the AIDA-I+ <i>E. coli</i> isolates. <p>The clinical significance of the AIDA-I+ <i>E. coli</i> was studied using clinical data available for 35 of the 110 <i>E. coli</i> isolates, originating from 18 cases of diarrhea. Among these 18 diarrhea cases, 3 cases (5 isolates) were found to have AIDA-I+ <i>E. coli</i> and these were significantly associated with diarrhea cases of post-weaning age group. Enterotoxigenic <i>E. coli</i> strains were isolated from the majority (72.5%) of 18 diarrhea cases and a high proportion (23.1%) of these ETEC cases carried AIDA-I+ <i>E. coli</i>. <p>In conclusion, AIDA-I adhesin appears to be a significant virulence factor for intestinal colonization and induction of biofilm formation. Further, experimental studies and clinical data suggest that the AIDA-I/STb virotype may be important in the pathogenesis of pre-weaning and post-weaning diarrhea in pigs. Desarrollo de métodos rápidos para el análisis de residuos en producción animal

La utilización de algunas sustancias antimicrobianas y algunos corticosteroides como promotores del crecimiento es una práctica ilegal en la UE. Una nueva aproximación para la detección y el control del suministro de estos compuestos puede ser el análisis del pelo. Esta matriz presenta ciertas ventajas frente a otras matrices de análisis, pero los métodos analíticos así como los principales mecanismos por los cuales estas sustancia se acumulan, no están del todo claros ni bien definidos. En este trabajo se desarrollan protocolos de análisis rápidos y específicos para detectar residuos de sulfametacina (SMZ), enrofloxacino (ENR) y dexametasona (DEX) en pelo, músculo e hígado de animales de producción (vacuno y porcino). Asimismo, se confirma la deposición de estos compuestos en el pelo de animales tratados y se evalúa el pelo como matriz analítica y de control de la administración de estos compuestos en producción animal. / The use of antimicrobial substances and some corticosteroids as growth promoters is an illegal practice in the EU countries. A new approximation to detect and control these practices could be hair analysis. This matrix offers certain advantages if compared to other tissues, but the analytical protocols as well as the mechanisms of drugs incorporation into hair are not well known and defined. The objectives of this work are to develop rapid and specific analytical protocols to detect residues of sulphamethazine (SMZ), enrofloxacin (ENR) and dexamethasone (DEX) in hair, muscle and liver of treated animals (cattle and pigs). The accumulation of these compounds in the hair of treated animals as well as the evaluation of the hair as a tool to control the administration of these compounds is studied and evaluated in this work.

Estudio de otros compuestos relacionados con la presencia de olor sexual no atribuible al escatol y a la 5@-Androst-16-en-3-ona en grasa dorsal de cerdo

En el presente trabajo de tesis se evalúa la posible contribución de otros compuestos al desarrollo del olor sexual mediante la aplicación de técnicas de Head Space dinámico y cromatografía de filtración de gel. El estudio se efectuó en canales de cerdos enteros previamente clasificadas con concentraciónes bajas de escatol (<0,10 µg /g) y androstenona (0,50 µg/ g) , aunque presentaban , según las respuestas de un panel sensorial, olor sexual. La selección de muestras de grasa clasificadas sin olor sexual permitió la comparación estadística de los resultados obtenidos. El análisis de los compuestos volátiles mostró una mayor abundancia de aldehidos (hexanal, heptanal), ácidos grasos de cadena corta (hexanoico, heptanico nonanoico), 1,4-diclorobenceno y estireno en las muestras de grasa con defecto organoléptico, ue puede, debido a los atributos sensoriales de estas sustancias y sus bajos umbrales de detección, favorecer el desarrollo de aromas desagradables asociados al olor sexual por los miembros de un papel sensorial. El análisis de las fracciones obtenidas por filtración de gel mediante cromatografía de gases acoplada a la espectometría de masas permitió la identificación de la 4-fenil-3-buten-2-ona en las muestras clasificadas con olor sexual y concentraciones bajas de escatol y androstenona, que junto a los resultados obtenidos en el estudio sensorial, indican una influencia de este compuesto en el olor sexual. Por una parte la 4-fenil-3-buten-2-ona presentaría una acción sinérgica con la androstenona al potenciar su detección por parte de los panelistas a concentraciones inferiores de 0,01µg/g. Además, el olor a naftalina que presenta la 4-fenil-3-buten-2-ona puede confundir a los miembros del panel debido a que es uno de los descriptores sensoriales que se asocia con la presencia de escatol en las muestras de grasa evaluadas. Así mismo, la baja transferencia a la fase vapor del escatol y, especialmente, de la androstenona determinada mediante técnicas de Head Space estático, indica que la concentración de cada uno de los compuestos en la fase vapor no se corresponde con la concentración real en las muestras de grasa . Según estos resultados, las respuestas del panel test pueden estar influenciadas por la poca volatilidad de los compuestos implicados en el olor sexual y por la presencia en la fase vapor de otros compuestos aromáticos de mayor volatilidad.La validación de métodos de análisis para la determinación de indol-escatol y androstenona-androstenoles en grasa dorsal porcina mediante cromatografía líquida en fase normal y cromatografía de gases acoplada a la espectrometría de masas, respectivamente, fue otro de los objetivos planteados del presente estudio. La simplificación del tratamiento de las muestras de grasa fue uno de los aspectos analíticos que se tuvo en mayor consideración con la finalidad de ofrecer una mayor capacidad de muestras en un intervalo de tiempo corto.
