Molecularly imprinted polymers for detection of volatile organics associated with fuel combustion

Magister Scientiae - MSc (Chemistry) / Pollutants such as polycyclic aromatic hydrocarbons (PAHs) are known for their toxic effects which may lead to the cause of degenerative diseases in both humans and animals. PAHs are widespread in the environment, and may be found in water, food, automotive industry and petrochemical industries to name but a few sources. Literature reports have highlighted industrial workplace exposure to PAHs as a leading cause for development of cancer in workers. Particularly, workers in the petrochemical industry are adversely affected and the incidence of skin and lung cancer in this population group is high. The United States of America in its guidelines developed by environmental protection agency (EPA) has identified 18 PAHs as priority pollutants. Among these are anthracene, benzo[a]pyrene and pyrene which have been selected as the focal point of this study due to their significance in the petrochemical industry. Due to the carcinogenic and mutagenic properties reported in literature for certain PAHs, there have been monitoring procedures taken in most countries around the world. The commonly used analytical methods for the detection of PAHs from industrial samples are high performance liquid chromatography (HPLC) coupled to fluorescence detection, membrane filtration, ozonation and reverse osmosis. Analysis of PAHs from the petrochemical industry is typically performed by HPLC method as well as sono-degredation in the presence of oxygen and hydrogen peroxide.

Synthèse et caractérisation de nouveaux matériaux organophosphorés pour des applications en optoélectronique / Synthesis and characterisation of new organophosphorus materials for optoelectronic applications

Ce manuscrit décrit la synthèse et la caractérisation de nouvelles molécules incluant un cœur organophosphoré, le phosphole. Certaines de ces molécules ont été utilisées pour la fabrication de dispositifs OLEDs ou de cellules photovoltaïques organiques. Le premier chapitre fait un état de l'art de la chimie du phosphole dans le domaine des matériaux organiques entre 2010 et 2013. Le second chapitre décrit la synthèse et l'étude physico-chimique de molécules qui permettent de moduler l'angle de torsion dans les systèmes π conjugués pour faire varier les propriétés optiques et rédox. Une de ces molécules a permis la fabrication d'une diode blanche organique. Le troisième chapitre de ce manuscrit présente une structure tridimensionnelle intéressante, le 1,1-biphosphole. En plus de posséder une structure tridimensionnelle, ces structures présentent un mode de conjugaison original, la conjugaison σ-π, qui permet de réduire l'écart HO-BV de nos systèmes. Une de ces molécules a permis la fabrication de la première cellule photovoltaïque organique avec un dérivé du phosphole inséré dans la couche active. Dans une deuxième partie, ce chapitre traite également de la réactivité originale du 1,1'-biphosphole qui permet de fonctionnaliser l'atome de phosphore par une simple substitution nucléophile, permettant d'insérer une grande variété de substituants pour moduler les propriétés des molécules. Pour finir, ce manuscrit présente un quatrième chapitre qui implique le phosphole comme unité coordinante afin de réaliser des nouveaux complexes qui permettent de réaliser une ortho-métallation par activation C-H. De nouveaux complexes ortho-métallés d'Ir(III) et de Rh(III) ont été synthétisés et caractérisés. / This thesis describes the synthesis and the characterization of new molecules including an organophosphorous unit, the phosphole ring. Some molecules have been used to build devices like organic light emitting diodes or organic photovoltaic cells.The first chapter describes the state of the art of the phosphole chemistry in organic materials between 2010 and 2013. The second chapter describes molecules having a tuneable twist angle allowing a fine control of the properties of the molecules like the HOMO-LUMO gap. One of those molecules has been used to build a white organic light emitting diode. The third chapter of this thesis presents an interesting three dimensional structure, the 1,1'-biphosphole. Beside this three dimensional structure, the molecules possess an original conjugation mode, the σ-π conjugation which allows a decrease of the HOMO-LUMO gap. One molecule from this chapter was used as absorber in organic photovoltaic cell. In the second part of this chapter, the 1,1'-biphosphole structure shows an interesting reactivity toward nucleophilic attack in order to functionalize the phosphorus center. This reactivity has been used to make new molecules and offer the opportunity to attach a wide range of substituents to the phosphorus atom in order to tune the properties of the molecules. The fourth chapter deals with the coordination chemistry of the phosphole in order to realize new ortho-metalated complexes. New Ir(III) and Rh(III) complexes have been synthesized and characterized.

Le secteur industriel du traitement de surface (TS), secteur d’excellence en Bourgogne Franche-Comté, est contraint de recourir à de nouvelles méthodes d’épuration des eaux du fait d’une règlementation européenne de plus en plus stricte. En effet, notamment dans le cadre de la Directive Cadre sur l’Eau, les industriels doivent sans cesse améliorer la qualité de leurs rejets et diminuer les impacts générés par ceux-ci. Ce travail de thèse s’inscrit dans cet objectif. Ainsi, trois grands types d’investigations ont été menés : le premier a permis de définir la composition qualitative et quantitative en substances dangereuses (SD) des rejets de TS et d’étudier leur variabilité temporelle ; le second a validé des modifications d’une station physico-chimique de traitement des eaux franc-comtoise pour diminuer le flux de SD ; le dernier a proposé des solutions innovantes par bioadsorption sur des matériaux de cyclodextrine et par procédé d’oxydation avancée (POA) soit par ozone/UV, soit par l’utilisation de catalyseurs Pd-Cu pour atteindre le même objectif.[...] / The industrial sector of surface treatment (ST), sector of excellence in Bourgogne Franche-Comté, is forced to resort to new methods for water treatment due to European regulations increasingly strict. A controllable self-assembly strategy of positively charged polycyclic aromatic hydrocarbons (PCPAH) towards the formation of rectangle sheets and ribbon-like nanostructures has been achieved by choosing divalent anions with different sizes. In contrast, only rod-like nanostructures are obtained from PCPAH with univalent anions. It is revealed that the divalent anions play a key role in guiding the packing of PCPAH, which provides an unprecedented route to fabricate two-dimensional nanostructures.

The aim of this work is detailed characterization of pyrolysis oil by advanced analytical separation methods and mass spectrometry. During the pyrolysis process decomposition of complex organic material take place in the presence of high temperatures and the absence of oxygen. Condensable part of vapours and aerosols forms viscous liquid called pyrolysis oil which is a complex mixture of large amount of compounds. In this work samples from microwave pyrolysis of woodchips and sewage sludge from wastewater treatment plant Brno-Modřice were analysed. Samples were provided by Bionic Laboratories company. Samples were separated by column chromatography on activated silica. For detailed characterization of each fraction gas chromatography with mass spectrometry detection and comprehensive two-dimensional gas chromatography with mass spectrometric detection with time of flight analyser were used. Substances that have been identified can be divided into several groups: alkanes, alkenes, aromatic hydrocarbons, polycyclic aromatic hydrocarbons, alcohols, aldehydes, esters, carboxylic acid and heterocycles.

Cardiotoxic effects of polycyclic aromatic hydrocarbons and abiotic stressors in early life stage estuarine teleosts

<div>Following the 2010 Deepwater Horizon oil spill, extensive research has been conducted on the toxicity of oil and polycyclic aromatic hydrocarbons (PAHs) in the aquatic environment. The location and timing of the Deepwater Horizon surface slick coincided with the spawning seasons of many important pelagic and estuarine fish species. As such, there has been particular emphasis placed on the effects of PAHs on sensitive life history stages in fish, such as the embryonic and larval periods. Additionally, the spill occurred throughout the spring and summer months which, in estuaries, are marked by regular fluctuations in abiotic environmental factors such as dissolved oxygen, salinity, and temperature. Until recently, there has been little work done to elucidate the combined effects that PAHs from oil spills and adverse environmental conditions (hypoxia, increased salinity, and elevated temperatures).</div><div>Work presented in this dissertation uses next generation sequencing technology (RNA Seq) to determine differential gene expression in larval estuarine teleosts following exposure to adverse environmental conditions and PAHs. Downstream canonical pathway and toxicological function analysis were then applied to the identified differentially expressed genes (DEGs) to predict cardiotoxic responses at the organismal level. To verify the predicted responses, a phenotypic anchoring study was conducted and identified a cardiotoxic phenotype (pericardial edema) and reduced cardiac output in embryos exposed to oil. Finally, the mechano-genetic interplay governing the morphological development of the teleost heart was investigated and correlations between developmental gene expression and blood flow forces within the cardiovascular system were identified.</div>

Occupational exposure to combustion by-products and breast cancer risk in postmenopausal women

Contexte : L’exposition professionnelle aux sous-produits de combustion est répandue et peut contribuer à l'étiologie du cancer du sein. Cette étude vise à estimer l’association entre l’exposition professionnelle à certains sous-produits de combustion et le risque postménopausique de cancer du sein. Méthodes : Cette étude cas-témoins populationnelle comprenait des femmes ménopausées âgées de 47-75 ans résidant à Montréal, Québec (2008-2011). Les cas incluaient 695 femmes ayant reçu un diagnostic cancer du sein malin et 608 témoins sélectionnés aléatoirement à partir de la Liste électorale du Québec, appariés aux cas en fréquence (groupes d'âge de 5 ans). L’information sur les facteurs de risque et l'historique professionnel a été recueillie par entrevue. Des hygiénistes industriels ont évalué l'exposition à 293 agents, dont six sous-produits de combustion. Le risque de cancer du sein associé à l'exposition professionnelle à certains sous-produits a été estimé, pour l'ensemble des tumeurs et leurs sous-types moléculaires, par régression logistique inconditionnelle avec rapports de cotes ajustés (RC) et intervalles de confiance à 95 % (IC 95%). Résultats : Des associations positives suggestives ont été trouvées entre l'exposition aux hydrocarbures aromatiques polycycliques et certains sous-types moléculaires de tumeurs : toutes tumeurs, RC=1,18 (IC95%=0,80-1,76), tumeurs luminales A, RC=1,25 (IC95%=0,81-1,93) et tumeurs luminales B, RC=2,09 (IC95%=0,87-4,60). Un risque élevé a été observé avec l'exposition aux fumées de cuisson pour les tumeurs HER2-enrichies (RC=2,63, IC95%=0,98-6,40). Conclusion : L'exposition à certains sous-produits de combustion peut augmenter le risque de certains sous-types moléculaires de cancer du sein. Des études futures explorant cette association sont justifiées. / Background: Postmenopausal breast cancer is the most common cancer among women, yet little is known about its association with occupational exposures. Exposure to combustion by-products is widespread in occupational settings and may contribute to breast cancer etiology. Here, we sought to estimate the association between lifetime occupational exposure to select combustion by-products and the risk of postmenopausal breast cancer. Methods: This population-based case-control study included postmenopausal women residing in Montreal, Quebec (2008-2011). Cases comprised 695 women aged 47-75 years diagnosed with incident malignant breast cancer, and 608 controls randomly selected from the Quebec Electoral List, frequency-matched to cases (5-year age groups). Information on risk factors and employment history was collected by interview. Exposure to 293 agents, including six combustion by-products was assessed by industrial hygienists. Unconditional logistic regression was used to estimate adjusted odds ratios (ORs) and 95% confidence intervals (95% CIs) for breast cancer risk, both overall and by tumor molecular subtypes, to occupational exposure to select combustion by-products. Results: We found suggestive positive associations between exposure to polycyclic aromatic hydrocarbons and certain molecular subtypes of tumors: for all tumors, OR=1.18 (95% CI: 0.80-1.76), Luminal A tumors, OR=1.25 (95% CI: 0.81-1.93) and Luminal B tumors, OR=2.09 (95% CI: 0.87-4.60). Elevated risked were observed for exposure to cooking fumes for HER2-enriched tumors (OR=2.63, 95% CI: 0.98-6.40). Conclusion: Exposure to select combustion by-products may increase the risk of certain hormonal subtypes of breast cancer. Future studies exploring this association are warranted.

The topic of this work is the structural characterization and theoretical modeling of organic single and heterolayers. The growth of sub-monolayers and monolayers (ML) of the two polycyclic aromatic hydrocarbons quaterrylene (QT) and hexa-peri-hexabenzocoronene (HBC) on Ag(111) and Au(111) was investigated. A transition from a disordered, isotropic phase to an ordered phase with increasing coverage was found. The lattice of the ordered phase turned out to be coverage dependent. The intermolecular potential was modeled including Coulomb and van der Waals interaction by a force-field approach. The postulated repulsive character of the potential could be connected to the non-uniform intramolecular charge distribution and to a screening of the van der Waals forces. Furthermore, the influence of the variable lattice constant on the epitaxial growth of HBC was studied. The second part of this work deals with a ML of 3,4,9,10-perylenetetracarboxylic dianhydride (PTCDA) on a ML of HBC. In dependency on the initial lattice constant of HBC, a total of three line-on-line (LOL) and point-on-line coincident phases of PTCDA (with respect to HBC) was found. Following an analysis of the general properties of LOL coincident systems via force-field calculations, a new method to predict the structure of such systems is introduced. / Thema dieser Arbeit ist die strukturelle Charakterisierung von organischen Einfach- und Heterolagen sowie deren theoretische Beschreibung und Modellierung. Es wurden Submonolagen und Monolagen (ML) der polyzyklischen Kohlenwasserstoffe Quaterrylen (QT) und Hexa-peri-hexabenzocoronen (HBC) auf Ag(111) und Au(111) Einkristallen untersucht und ein Übergang von einer ungeordneten, isotropen Phase zu einer geordneten Phase mit steigender Bedeckung beobachtet. Die geordnete Phase wies dabei bedeckungsabhängige Gitterkonstanten auf. Das intermolekulare Potential wurde unter Berücksichtigung von Coulomb und van der Waals Anteilen mittels Kraftfeldmethoden modelliert. Der postulierte repulsive Charakter des Potentials konnte auf die Ladungsverteilung im Molekül und eine Abschwächung des van der Waals Potentials zurückgeführt werden. Weiterhin wurde der Einfluss der variablen HBC Gitterkonstante auf die epitaktische Relation des Gitters zum Metallsubstrat untersucht. Der zweite Teil der Arbeit widmet sich der Untersuchung einer ML 3,4,9,10-Perylenetetracarboxylic dianhydrid (PTCDA) auf einer ML HBC. Dabei wurden, in Abhängigkeit von der HBC Gitterkonstante, insgesamt drei verschiedene Typen von line-on-line bzw. point-on-line Epitaxie nachgewiesen. Im Anschluss an eine Analyse der generellen Eigenschaften solcher epitaktischer Lagen mittels Kraftfeldrechnungen wird eine neue Methode zur Vorhersage der Struktur konkreter Systeme vorgestellt.

Os combustíveis fósseis representam a principal contribuição, devido à taxa e escala espacial, em que o petróleo tem sido usado como fonte de energia e matéria-prima para a indústria química. O objetivo deste estudo foi avaliar os níveis de concentração e identificar fontes de hidrocarbonetos na Bacia do Alto Iguaçu e, em especial, na área de influência da Refinaria Presidente Getúlio Vargas (REPAR). Além dos fatores de poluição crônica, a área de estudo foi alvo de um derrame acidental de petróleo em julho de 2000. Diversos indicadores em diferentes compartimentos ambientais (água superficial, sedimento, solo e água subterrânea) foram avaliados na fase pós-derrame e no monitoramento ambiental ao longo de várias campanhas por mais de uma década. Os esforços de avaliação foram concentrados nos seguintes indicadores: os nalcanos, os alcanos isoprenoides, os hidrocarbonetos policíclicos aromáticos (HPA), os biomarcadores de petróleo e ainda o total de hidrocarbonetos de petróleo (THP). Os resultados identificaram como principal aporte de matéria orgânica para as águas superficiais dos rios Barigüi e Iguaçu, os fluxos materiais originados em região a montante do acidente, refletindo a contribuição antropogênica crônica da cidade de Curitiba. Em 2007 e 2008, as concentrações de THP e HPA nas águas superficiais e sedimentos dos rios Barigüi e Iguaçu refletem uma expressiva melhoria nas condições desses rios em relação a 2000. Na área interna da refinaria (Ponto Zero), observou-se uma nítida diminuição (atenuação) natural das concentrações de THP no solo em todas as profundidades dos perfis amostrados ao longo do tempo nos Banhados 1 e 4 e, em particular, uma diminuição importante das concentrações entre 2004 e 2007. Para a identificação das fontes de hidrocarbonetos foram utilizadas razões diagnósticas e quimiometria. As razões diagnósticas calculadas a partir de concentrações de HPA sugeriram que, na maioria dos sedimentos dos Rios Barigüi e Iguaçu coletados nas campanhas de 2000 e 2001, a fonte petrogênica é a principal. Somente na estação a montante do acidente no Rio Barigüi, a fonte pirolítica predominou nestas duas campanhas. As razões diagnósticas que apresentaram maior eficiência na identificação de fontes de hidrocarbonetos nos sedimentos dos rios Barigüi e Iguaçu foram: ΣC1-Fenantrenos/Fenantreno; e (ΣHPA parentais de 3-6 anéis)/(Σ5 séries de HPA alquilados). A identificação de fontes através de razões diagnósticas calculadas a partir de áreas e alturas de picos cromatográficos demonstrou sua aplicabilidade verificando a relação entre os compostos encontrados em amostras de solo da área interna da refinaria com a amostra de petróleo derramado no acidente, após quase uma década da ocorrência do vazamento. A identificação de fontes através método de quimiométrico baseado na análise de componentes principais (ACP) de seções pré-processadas e combinadas dos Cromatogramas de Íons Selecionados (CIS) mostrou que as amostras mais contaminadas estão na área interna da refinaria. Essas amostras apresentam um padrão de distribuição petrogênica e diferentes graus de intemperismo. As amostras da área externa à refinaria (Guajuvira, General Lúcio e Balsa Nova) são menos ou não contaminadas e/ou contém uma mistura de contribuições diagenéticas, pirolíticas e petrogênicas onde predominam diferentes proporções. Os locais mais distantes da atividade industrial (Balsa Nova) contem, como esperado, os níveis mais baixos de contaminação por HPA. Os resultados de biomarcadores demonstraram que não há evidências para concluir que as amostras da área externa à refinaria e o óleo Cusiana vazado tenham a mesma origem. Os resultados ao longo dos rios Barigüi e Iguaçu e do Ponto Zero demonstraram que as ações de emergência para a contenção do óleo foram adequadas para os rios, e que a contaminação decorrente do derrame ficou predominantemente contida no Ponto Zero e diminuiu significativamente após uma década. / Hydrocarbons present in the environment consist of complex mixtures of compounds derived from multiple sources. The main contribution lies on fossil fuel inputs due to the rate and spatial scale by which petroleum has been used as an energy source and chemical feedstock. The aim of this study was to assess the concentration levels and identify sources of hydrocarbons in the Upper Iguaçu Watershed and, in particular, in the area of influence of the President Getulio Vargas Refinery (REPAR). In addition to the factors of chronic pollution, the study area was the scenario of an acute accidental oil spill in July 2000. Numerous indicators in different environmental compartments (surface water, sediment, soil and groundwater) were assessed in the post spill phase and during the environmental monitoring programs over the course of several campaigns for more than a decade. Assessment efforts were concentrated on the following indicators: n-alkanes, alkanes isoprenoids, polycyclic aromatic hydrocarbons (PAH), petroleum biomarkers and total petroleum hydrocarbons (TPH). The results identified as the main contribution of organic matter to surface waters of the Barigüi and Iguaçu Rivers the materials flows originated in the region upstream of the accident, reflecting chronic anthropogenic contribution of the city of Curitiba. In 2007 and 2008, the TPH and PAH concentrations in surface waters and sediments of the Barigüi and Iguaçu Rivers revealed a significant improvement in the conditions of these rivers when compared with 2000. Inside the refinery area (Point Zero), it was observed a clear natural decrease (attenuation) of the concentrations of TPH in the soil at all depths sampled over time in Marshes 1 and 4 and, in particular, an important decrease of concentrations between 2004 and 2007. Diagnostic ratios and chemometrics were used to identity hydrocarbon sources. The diagnostic ratios calculated from the concentrations of PAH suggested that, in the majority of sediments from the Barigüi and Iguaçu Rivers collected in 2000 and 2001 campaigns, the main source is petrogenic. Only in the station upstream the accident in the Barigüi River, the pyrolytic source predominated in these two campaigns. The diagnostic ratios that presented higher efficiency in identifying sources of hydrocarbons in sediments of the Barigüi and Iguaçu Rivers were: ΣC1- Phenantrenes/Phenanthrene; and (Σ3-6 rings parental PAH)/(Σ5 alkylated PAH series). The source identification through diagnostic ratios calculated from heights and areas of chromatographic peaks demonstrated its applicability establishing a relationship between the compounds found in soil samples of the internal area of the refinery and the sample of the oil spilled in the accident, after nearly a decade of occurrence of the spillage. The source identification through chemometric method based on principal components analysis (PCA) of preprocessed and combined sections of Selected Ion Chromatograms (SIC) showed that the most contaminated samples are inside the refinery area. These samples present a petrogenic pattern and different weathering degrees. Samples from outside the refinery area (Guajuvira, General Lúcio e Balsa Nova) are either less or not contaminated, and/or contain mixtures of diagenetic, pyrogenic and petrogenic inputs where different proportions predominate. The locations farthest away from industrial activity (Balsa Nova) contain, as expected, the lowest levels of PAH contamination. The biomarkers results do not show any evidences to conclude positive matches between the samples from outside the refinery area and the spilled Cusiana oil. The results along Barigüi and Iguaçu rivers and Point Zero demonstrated that emergency actions to contain the oil were appropriate for the rivers, and that the contamination resulting from the spill was mostly contained in the Point Zero and decreased significantly after one decade
