Neste trabalho apresentamos uma demonstração detalhada para um conhecido teorema de I. J. Schoenberg que caracteriza certas funções positivas definidas em esferas. Analisamos ainda um critério para a obtenção de positividade definida de uma função a partir de condições de suavidade e convexidade dela, em uma tentativa de ratificar alguns resultados da literatura conhecidos como critérios de Pólya. / In this work we present a proof for a famous theorem of Schoenberg on positive definite functions on spheres. We analyze some results that deduce positive definiteness from diferentiability and convexity assumption on the function, an attempt to ratify some Pólya type conjectures found in the literature.

Identificação molecular e caracterização de cepas isoladas do resíduo de bauxita / Molecular identification and characterization of bacterial strains isolated from bauxite residueNOGUEIRA, Elis Watanabe 24 July 2015 (has links)
O presente trabalho visou identificar e caracterizar três cepas, BRA1, BRA3 e BRA5, isoladas do resíduo de bauxita da região Sul de Minas Gerais. Foi realizado o seqüenciamento do gene 16S RNA ribossomal utilizando dois pares de primers em que foi possível seqüenciar 1300pb de cada cepa isolada. Testes bioquímicos e metabólicos complementares foram realizados a fim de caracterizar os isolados. Os resultados indicaram que a cepa BRA1 foi identificada com 99,7% de similaridade com Bacillus cohnii e caracterizada como: Gram-positiva; redutora de nitrato, catalase, oxidase e amilase positiva; faixa de crescimento entre pH 7 e 10; tolerante às concentrações de 0 a 10% de NaCl, e; faixa de temperatura de 20 a 40ºC. A cepa BRA3 foi identificada com 99,7% de similaridade com Bacillus pseudofirmus e caracterizada como: Gram-positiva; catalase e amilase positiva; oxidase negativa e não redutora de nitrato; faixa de crescimento entre pH 7 e 10; tolerante às concentrações de 5 a 10% de NaCl, dependente do íon Na+ para crescimento, e; faixa de temperatura de 10 a 40ºC. Com os resultados da técnica de seqüenciamento, a cepa BRA5 foi identificada com 99,7% de similaridade com Bacilus clarkii e Bacillus polygoni. Após testes bioquímicos e fisiológicos, a cepa BRA5 ficou mais próxima às características bioquímicas do Bacillus clarkii, caracterizada como: Gram-positiva; catalase e oxidase positiva, redutora de nitrato e amilase negativa; faixa de crescimento entre pH 8 e 10; tolerante às concentrações de 5 a 10% de NaCl, dependente do íon Na+ para crescimento, e; faixa de temperatura de 20 a 40ºC. A utilização dos açúcares: glicose, frutose, sacarose e manitol pelas cepas foi positiva e os ácidos mais produzidos a partir das fontes de carbono foram os ácidos acético, isobutírico, isovalérico, entre outros. As cepas BRA1, BRA3 e BRA5 utilizando glicose como fonte de carbono foram capazes de diminuir o pH do meio de 10,5 para aproximadamente 9,3, 9,5 e 9, respectivamente. A identificação e caracterização dos isolados foram bem sucedidas e os resultados apresentados estão de acordo com os encontrados na literatura. / This study aimed to identify and characterize three strains BRA1, BRA3 and BRA5 isolated from bauxite residue in the Southern region of Minas Gerais. After biochemical and physiological tests, the strain BRA5 was closest to the biochemical characteristics of Bacillus clarkii, characterized as Gram-positive; positive for catalase and oxidase, non-nitrate reduction, and amylase; growth range between pH 8 and 10; tolerant to concentrations of 5 to 10% NaCl, Na + ion dependent for growth, and; growth is observed between 20 and 40 °C. The use of sugars: glucose, fructose, sucrose and mannitol was positive and the higher amount of acids produced from the carbon sources were acetic, isobutyric, isovaleric, among others. Strains BRA1, BRA3 and BRA5 using glucose as a carbon source were able to decrease the medium pH 10.5 to approximately 9.3, 9.5 and 9, respectively. Inicialmente, estudamos alguns resultados clássicos da teoria dos núcleos positivos definidos e alguns resultados pertinentes. Estudamos em seguida, o Teorema de Mercer e algumas de suas generalizações e conseqüências, incluindo a caracterização da transformada de Fourier de um núcleo positivo definido com domínio Rm£Rm, m ¸ 1. O trabalho traz um enfoque especial nos núcleos cujo domínio é um subconjunto não-compacto de Rm £ Rm, uma vez que os demais casos são considerados de maneira extensiva na literatura. Aplicamos esses estudos na análise do decaimento dos autovalores de operadores integrais gerados por núcleos positivos definidos / Firstly, we study some classical results from the theory of positive definite kernels along with some related results. Secondly, we focus on generalizations of Mercer\'s theorem and some of their implications. Special attention is given to the cases where the domain of the kernel is not compact, once the other cases are considered consistently in the literature. We include a characterization for the Fourier transform of a positive definite kernel on Rm£Rm, m ¸ 1. Finally, we apply the previous study in the analysis of decay rates for eigenvalues of integral operators generated by positive definite kernels

Em uma segunda estratégia, empregou-se como alvo a subunidade B da toxina termolábil (LTB) de E. coli enterotoxicogênica (ETEC) co-expressa ou fusionada à proteína GroEL2 de Mycobacterium bovis. As quantidades de E7 e gDE7 expressas pelas linhagens recombinantes de B.subtilis ficaram abaixo do limite de detecção e não foram suficientes para ativação de células T CD8+ específicas. Por outro lado, linhagens de B. subtilis capazes de expressar LTB e GroEL2 promoveram a ativação de linfócitos T CD8+ específicos. Recently, genetically modified B.subtilis strains were successfully employed as vaccine vehicles but there is no information concerning the activation of antigen specific cytotoxic immune responses (T CD8+ lymphocytes) in mice vaccinated with either vegetative cells or spores. In the present report, we evaluated the activation of CD8+ T cell responses in mice immunized with B. subtilis vaccine strains genetically modified in order to express the human papillomavirus type 16 (HPV-16) E7 protein as a target antigen, isolated or genetically fused to the type I herpes simplex vírus (HSV-1) gD protein. In a second approach, we have also tested the B subunit heat labile toxin, produced by enterotoxigenic E. coli (ETEC) strains, coexpressed or genetically fused to the Mycobacterium bovis GroEL2 protein. E7 and gDE7 expression by B.subtilis vaccine strains were bellow the detection limits and did not allow the activation of antigen-specific CD8+ T cell responses in vaccinated mice. Rio de Janeiro, RJ, Brasil / Células CD8+ participam do controle da infecção pelo HIV-1 através de atividades citolítica e supressora da replicação viral (não-citolítica). Contudo, durante a progressão da infeção, são observadas alterações em sua diferenciação e função. Várias razões para tal são postuladas, incluindo a suscetibilidade do próprio linfócito T CD8+ à infecção pelo HIV-1. Neste estudo, analisamos os efeitos da infecção in vitro pelo HIV-1 sobre o fenótipo de células CD8+. Comparado às culturas não-infectadas, observamos aumento de células CD8+ CD57+ e CD45RO+, bem como daquelas de estágio intermediário, enquanto a expressão de CD28 e o fenótipo naive diminuem, sem que haja aquisição do fenótipo efetor, padrão este mais evidente em culturas infectadas por vírus X4 que R5, e semelhante ao que é encontrado in vivo. A função da população expandida CD8+CD57+ foi avaliada e parece inibir a proliferação celular, e desse modo, a replicação viral. Nosso terceiro estudo objetivou analisar se a infecção produtiva de células CD8+ pelo HIV-1 afetaria resultados de ensaios de supressão viral in vitro. Nestes experimentos, utilizamos citocinas gc que apresentam-se alteradas na infecção pelo HIV-1, como IL-2, IL-7 e IL-15, e paradoxicalmente, IL-2 e IL-15 modulam ambos os papéis exercidos por células CD8+ na infecção: alvos e supressoras de replicação viral. Em parte, isto se explica pela cinética mais lenta do vítus nestas células, tornando-as reservatórios me potencial. Ainda, como células dendríticas (DC) produzem IL-15, avaliamos se esta citocina, endógena ou exógena, afetaria a susceptibilidade e atividade antiviral de células CD8+ frente ao HIV-1 em co-cultura com DC autólogas. De fato, IL-15 parece ser um importante estímulo para a atividade supressora de células CD8+ mesmo neste modelo mais fisiológico de replicação viral, embora também promova a infecção das mesmas células CD8+.
Esta infeção após contato com DC autólogas HIV-1-pulsadas pode ser explicada pela co-expressão de CD4 e CXCR4, provavelmente facilitada pela interação CD28/B7, já que células CD8+CD28+ apresentam maiores níveis de expressão destes receptores, bem como de produtividade do HIV-1. O desenvolvimento de uma vacina é colocado como a melhor estratégia para o controle da pandemia. No entanto, entre os candidatos vacinais que evoluíram para testes clínicos de eficácia, apenas um mostrou resultados positivos, mas com uma modesta eficácia (31,2%). Além da busca por uma vacina eficiente contra a infecção pelo HIV-1, o desenvolvimento de estratégias para se atingir a cura também é essencial. Considerando esses dois objetivos principais, diversos fatores são colocados como obstáculos, sendo a enorme diversidade genética do HIV-1 e a falta de correlatos de proteção contra a doença ou a infecção, os pontos centrais e nos quais focamos nossa atenção. No primeiro trabalho, foi avaliada a resposta de células T frente a peptídeos derivados de sequências consensos e sequências de isolados virais mais próximos ao consenso (CVI) dos três subtipos prevalentes no Brasil, B, C e F1 e do grupo M. O estudo incluiu 32 indivíduos infectados com o HIV-1, subtipo B (n = 13), subtipo C (n = 11) e subtipo F1 (n = 8). As respostas específicas frente a Gag e Nef foram avalidas usando o ensaio de ELISpot IFN-\03B3. Peptídeos baseados nas sequências CVI apresentaram capacidade de redução da distância genética e de detecção da resposta de células T similar ao observado para as sequências consenso
Em geral, o nível de reatividade cruzada entre os subtipos B e F1 foi maior do que entre os subtipos B e C ou entre os subtipos C e F1. Esses resultados sugerem que o obstáculo imposto pela diversidade genética no desenho de vacinas é dependente dos subtipos envolvidos. No segundo trabalho foi avaliada a resposta de células T de indivíduos controladores da replicação viral frente aos peptídeos consenso, derivados do primeiro estudo. Além disso, foi feita uma avaliação de fatores genéticos e imunológicos nesse grupo de controladores. O estudo incluiu três grupos de controladores: 1) Controladores de Elite (EC, n=7) com carga viral (CV) <80 cópias/mL em todos os pontos; 2) Controladores de Elite com Blips (EEC, n=5) com CV>80 cópias/mL em apenas 30% dos pontos e 3) Controladores Virêmicos (VC, n=7) com CV <5.000 cópias/mL e >80 cópias/mL em mais de 30% dos pontos. Indivíduos infectados pelo HIV-1 não tratados (NC) ou em tratamento antirretroviral (HAART) e indivíduos não infectados pelo HIV-1 foram analisados como controles. A coorte de controladores foi enriquecida em alelos protetores HLA-B, B*27 e B*57, mas não foi observada diferenças significativas entre os três grupos de controladores. Uma frequência maior de células T CD8+ ativadas foi observada em EEC e VC quando comparada a indivíduos não infectados. Os EC demonstraram uma resposta de células T de menor magnitude frente a Gag/Nef e uma proporção de anticorpos IgG anti-HIV significativamente menor (P < 0,05) que os grupos de EEC, VC e NC. Esses resultados demonstram que EEC e VC são capazes de montar uma resposta imune específica mais forte frente a estímulos do HIV-1, mas paralelamente, apresentam maiores níveis de ativação imune, translocação microbiana e maior reservatório viral, quando comparados aos EC. trategies to find a cure is essential though has been hindered by several obstacles
such as t
he enormous genetic
diversity of HIV

1 and lack of correlates of protection against
disease or infection
. These two obstacles were investigated in the present stu
dy by
evaluating the T cell response against peptides based on consensus and closest viral isolate
sequences (CVI) from the three prevalent subtypes in Brazil, B, C and F1
and
from group M
consensus. The study included 32 subjects infected with HIV
-
1 subty
pe B (n = 13), C (n =
11), and F1 (n = 8).
Gag and Nef

specific T cell responses were evaluated by IFN
-
γ
-
ELISpot
assay. Peptides based on CVI sequences were similar to consensus in both reducing genetic
distance and detecting T cell responses
. Overall, the level of cross

subtype response
between su
btypes B and F1 was higher than between subtype C and B or C and F1. Our
results suggest that the barrier of genetic diversity in HIV
-
1 group M for vaccine
design may
be dependent on the subtypes involved. The second study evaluated the T cell response

genetics and immunologic patterns in this cohort of controllers. The study

1 DNA reservoir compared to EC.
These results highlight
the heterogeneity of the HIV controllers and reinforce the needs
for
a precise classification of
patients with different levels of viral suppression.

Rio de Janeiro, RJ, Brasil / Alterações no perfil de glicosilação de proteínas, células e tecidos têm sido utilizadas como marcadores biológicos para processos fisiológicos e patológicos. Glicoconjugados contendo o ácido siálico (Neu5Ac) estão envolvidas em interações celulares e parasito-hospedeiro, tráfego de linfócitos e regulação do sistema imune. O Trypanosoma cruzi, agente etiológico da doença de Chagas (DC), expressa em sua superfície e libera no plasma do hospedeiro, uma enzima restrita ao gênero Trypanosoma, a trans-sialidase (TS). Esta é capaz de transferir Neu5Ac de moléculas doadoras e realocá-las em moléculas na superfície do parasito ou sialilar glicoproteínas de células do hospedeiro. A molécula CD43 é um importante aceptor de Neu5Ac em células T e um provável sítio de ação da TS. Na infecção experimental pelo T. cruzi, as células T CD8+ apresentam, majoritariamente, perfis segregados (i) perforina+ (Pfn+), com ação citotóxica, ou (ii) interferon-gama+ (IFN\03B3+), com perfil inflamatório. Estas células estão diferencialmente compartimentalizadas e atuam de forma antagônica, enquanto as Pfn+ se acumulam na inflamação cardíaca e contribuem para a injúria tecidual, as IFN\03B3+ estão majoritariamente na periferia (sangue e baço) e atuam de forma benéfica. Neste trabalho, propomos que na infecção pelo T. cruzi há alterações de glicofenótipos em proteínas séricas e subpopulações de linfócitos T CD8+ funcionalmente segregadas. Camundongos C57BL/6 foram infectados pela cepa Colombiana do T. cruzi e analisados clinicamente. O glicofenótipo de proteínas séricas foi analisado por espectrometria de massas (MALDI-TOF) e o glicofenótipo de tecido cardíaco e de células do baço foi analisado usando lectinas por histoquímica e citometria de fluxo, respectivamente
Não detectamos modificação no perfil glicosídico em proteínas séricas em animais infectados. Observamos, no tecido cardíaco, reduzida marcação para a lectina Peanut agglutinin (PNA), aumento para Sambucus nigra (SNA) e não alteração para Maackia amurensis (MAL) em células musculares e inflamatórias. Notamos nas fases aguda (42 dpi) e crônica (120 dpi) aumento na frequência de células PNA+ nas células T CD8+CD43+ e CD4+CD43+, redução no percentual de células SNA+ nas células T CD8+ e CD4+ e diminuição na proporção de células MAL+ nas células T CD4+ e CD8+CD4+. Células T CD8+ totais ou CD8+PNA+ apresentaram perfis semelhantes de ativação (CD44+CD62L-), migração (LFA-1+CCR5+ ou LFA-1+CCR1+) e funcionalidade (Pfn+, Pfn+IFN\03B3+ ou IFN\03B3+). Em 120 dpi, detectou-se enriquecimento de células CD8+PNA+IFN\03B3+ no baço. Assim, durante a DC experimental, células T CD8+PNA+ apresentam-se ativadas e com potencial de migrar e exercer atividade inflamatória (IFN\03B3+), contudo estas não foram detectadas em tecido cardíaco. Resta-nos esclarecer se estas células não migram para este tecido, acumulando-se na periferia, ou se ao entrarem no tecido cardíaco têm seu glicofenótipo alterado ou morrem de modo seletivo. Embora nossos achados não permitam a identificação de biomarcadores de natureza glicosídica de progressão e/ou gravidade da DC, abrem perspectivas para explorar outros modelos experimentais de infecção e para estudos sobre a compartimentalização de células fenotipicamente distintas e funcionalmente relacionadas à proteção contra a injúria cardíaca na infecção pelo T. cruzi / Changes in glycosylation profile of proteins, cells and tissues have been used as biological markers of physiological and pathological processes. Glicoconjugates containing sialic acid (Neu5Ac) are involved in cell and host-parasite interactions, lymphocyte traffic and regulation of the immune system. Trypanosoma cruzi, the causative agent of Chagas disease (CD), expresses on the surface and releases in the host plasma, a restricted enzyme to the genus Trypanosoma, the trans-sialidase (TS). This enzyme transfers Neu5Ac from donor molecules and relocates them on parasite surface molecules or sialylates glycoproteins of the host cells. The CD43 molecule is a major acceptor of Neu5Ac on T cells and a target site for TS. In experimental T. cruzi infection, CD8+ T cells have, mostly, segregated profiles (i) perforin+ (Pfn+) with cytotoxic action, or (ii) interferon-gamma+ (IFN\03B3+), with inflammatory profile. These cells are differentially compartmentalized and play antagonistic roles, while the Pfn+ cells accumulate in cardiac inflammation and contribute to tissue injury, the IFN\03B3+ are, mostly, at the periphery (blood and spleen) and play a beneficial role. In the present study, we propose that in T. cruzi infection there are glycophenotype changes in serum proteins and subpopulations of functionally segregated CD8+ T lymphocytes. C57BL/6 mice were infected with the Colombian strain of T. cruzi and analyzed for clinical changes. The glycophenotype of serum proteins was analyzed by mass spectrometry (MALDI-TOF) and glycophenotypes of the cardiac tissue and splenic cells were analyzed using a lectin-based immunohistochemistry and flow cytometry, respectively
In infected mice, we did not detect changes in glycoside profile of serum proteins. In the cardiac tissue, we observed reduced staining for the lectin Peanut agglutinin (PNA), increased for Sambucus nigra (SNA) and no alterations for Maackia amurensis (MAL) in muscle and inflammatory cells. In the acute (42 dpi) and chronic (120 dpi) phases, we noticed increased frequency of PNA+ cells among CD8+CD43+ and CD4+CD43+ T cells, reduced percentage of SNA+ cells among CD8+ and CD4+ T cells and decreased proportion of MAL+ cells among CD4+ and CD8+CD4+ T cells. Total and PNA+ CD8+ T cells showed similar profiles of activation (CD44+CD62L-), migration (LFA-1+CCR5+ cells or LFA-1+CCR1+) and functionality (Pfn+, Pfn+IFN\03B3+ or IFN\03B3+). At 120 dpi, there is an enrichment in IFN\03B3+ cells among splenic CD8+PNA+. Thus, in experimental CD PNA+ CD8+T cells are activated and potentially able to migrate towards heart tissue and show inflammatory profile (IFN\03B3+); however, PNA+ cells were not detected in this tissue. It remains to be clarified whether these cells do not migrate to this tissue, accumulating in peripheral tissues, or whether they enter the cardiac tissue but have their glycophenotype modified or selectively dye. Although, our finds do not allow identifying glycosidic biomarkers of progression and/or severity of CD, they open new pathways to be explored using other experimental models of infection and studying the compartmentalizationof phenotypically and functionally distinct cells associated with detrimental or protective role in heart injury in T. cruzi infection
