Abordagens múltiplas sobre a modulação fenotípica em pupas de Heliconius erato phyllis (Lepidoptera; Nymphalidae)

A presente tese, através de quatro manuscritos, aborda aspectos variados sobre a modulação fenotípica na cor da pupa em Heliconius erato phyllis. O primeiro manuscrito compara e avalia dois métodos de categorização para a cor da pupa através da estimativa da herdabilidade (h2). O primeiro deles, o método qualitativo, considera quatro escores, variando de 2 até 5, em uma ordem crescente de melanização. O segundo método, quantitativo, utiliza a densidade óptica. Após a captura, as imagens foram analisadas por um software de onde foram obtidos os valores para o grau de melanização de cada exúvia. A partir dos valores obtidos por cada metodologia estimou-se a h2 pelos métodos da regressão linear e da análise da variância, com o n variando entre 7 e 18 proles. As estimativas da h2 resultaram em valores semelhantes para ambas às metodologias de categorização das pupas. Desta forma conclui-se que a metodologia qualitativa, por sua praticidade, torna-se a mais indicada para a tarefa. A partir dos baixos valores de h2, discute-se o por quê destes resultados. A primeira hipótese considera o tamanho amostral e a segunda a prioridade na alocação de recurso. O segundo manuscrito avalia a influência do fotoperíodo na cor da pupa em H. erato phyllis. Três proles tiveram seus ovos distribuídos entre cinco fotoperíodos: 11L:13E, 12L:12E, 14L:10E, 9L:15E e 24L:00E. Os três primeiros representam fotofases para Porto Alegre (RS - Brasil). A última fotofase indicada é utilizada pelo nosso grupo como tratamento padrão e foi considerado um controle. Os resultados encontrados, através da ANOVA bifatorial indicam uma diferença entre as proles (p = 0,069) e não indicam diferença entre os tratamentos. Uma segunda análise foi feita através da ANOVA unifatorial considerando apenas a cor da pupa como dependente dos fotoperíodos. O resultado indicou uma dependência entre os fatores (p = 0,047). Uma terceira análise foi feita a fim de verificar se a diferença observada devia-se aos tratamentos ou fatores genéticos. Para isto comparou-se o tratamento 24L:00E das três proles com um conjunto de 22 proles criadas sob este mesmo fotoperíodo. O resultado aponta para uma diferença genética, concluindo que a cor da pupa em H. erato phyllis não é afetada pelo fotoperíodo. A adaptação de pupas em campo é o tema do terceiro manuscrito. Duas localidades foram utilizadas como área de estudo: Estação de Fitotecnia de Águas Belas (ABE) e Instituto de Pesquisas Hidráulicas (IPH). Cem pupas foram colocadas em cada localidade; 50 próximas ao solo e 50 acima de um metro. Dentro de cada nível de altura foram formados 25 pares, sendo 25 camufladas e 25 não-camufladas. Entre as 50 pupas de cada nível havia 25 pupas claras e 25 pupas escuras. Os resultados encontrados indicam uma relação direta da sobrevivência com o nível de altura em que as pupas foram inseridas. A camuflagem e a cor não se mostraram significantes para a sobrevivência da pupa em campo. Pupas implantadas próximo ao solo têm uma chance entre 8 e 13 vezes maior de serem predadas do que pupas acima de 1,0 metro de altura. Estes resultados foram vinculados ao tipo de predador observado em campo: formigas. Duas espécies e um gênero de formigas foram observados predando pupas. Conclui-se que, para pupas próximas ao solo, a camuflagem conferida pela cor não é adaptativa, visto que os principais predadores não dependem da visão. O último artigo trata da descrição do primeiro caso de metilação em uma borboleta: H. erato phyllis. Estudos prévios mostraram que as estimativas da herdabilidade para a cor da pupa nesta espécie, determinadas pelo método da regressão linear, apresentavam valores muito diferentes conforme fossem considerados machos e suas proles ou fêmeas e suas proles. Uma causa possível para esta discrepância poderia ser devido à ocorrência de metilação do DNA e possível silenciamento dos genes. Através das técnicas MSRE e PCR bissulfito com primers aleatórios investigamos a presença de metilação em H. erato phyllis. A análise, utilizando DNA de diferentes fases do desenvolvimento, mostrou que a técnica de PCR bissulfito foi particularmente eficiente para indicar a presença de metilação no DNA desta espécie. Isto abre a possibilidade de investigações futuras direcionadas para avaliar se este fenômeno tem relação com a determinação dos padrões diferenciais de coloração das pupas em H. erato phyllis. Por fim esta tese apresenta três anexos. O primeiro trata de um processo de seleção para fenótipos extremos para a cor da pupa em H. erato phyllis. Resultados preliminares apontam que o processo é exitoso, porém lento, visto que a variância para a característica permaneciam relativamente alta quando o processo foi interrompido. O segundo anexo aborda o efeito do vírus de poliedrose nuclear na coloração das pupas desta espécie. A hipótese surgiu após o aparecimento de pupas escuras no processo de seleção para pupas claras quando este fenótipo já não era mais esperado. Assim investigou-se o efeito do vírus sobre a cor da pupa bem como sobre a sobrevivência de lagartas de 4º e 5º instar. Os resultados apontam que o vírus não influência a cor da pupa. Por outro lado, há uma relação direta entre sobrevivência e indivíduos não infectados. O terceiro anexo descreve um fenótipo incomum para H. erato phyllis, onde os adultos não apresentavam pigmentos vermelho e amarelo, deixando o imago com um padrão preto e branco ao invés de preto e vermelho. / This thesis, through four manuscripts, covers various aspects on the phenotypic modulation in the pupal color of the in Heliconius erato phyllis. The first manuscript evaluate by heritability (h2) estimative two methods to measure the degree of melanization of pupal exuviae. The first of them, the qualitative method, considers four scores, ranging from 2 to 5, in an ascending order of melanization. The second method, quantitative, uses the optical density. After the capture, the images were analyzed by software from which were obtained values for the degree of melanization of each exuviae. From the values obtained for each methodology the h2 were estimated by the methods of linear regression and analysis of variance, with n ranging between 7 and 18 broods. Estimates of the h2 resulted in similar values for both methods of pupal categorization. Thus it is concluded that the qualitative methodology, for its usefulness, it is the most suitable for the task. From the low values of h2, we have two hypotheses. Three broods had their eggs distributed between five photoperiods: 11L:13E, 12L:12E, 14L:10E, 9L:15E and 24L:00E. The first three represent photoperiod to Porto Alegre (RS-Brazil). The last indicated photoperiod is used as standard treatment by our group and was considered a control. The results found through the ANOVA twofactors to indicate a difference between the broods (p = 0,069) and do not indicate the difference between treatments. A second analysis was done by ANOVA one-factor considering only the pupal colour as a dependent on photoperiod. The results show a dependency among the factors (p = 0,047). A third analysis was performed to determine if the difference observed was due to the treatments or because of the genetic factors. To do this, a comparison at the treatment 24L:00E between the three broods with a set of 22 broods, reared under the same photoperiod, was performed. The result points to a genetic difference, concluding that the pupal colour of the in H. erato phyllis is not affected by photoperiod. The adaptation of pupas in field is the subject of the third manuscript. Two locations were used as the study area: Estação de Fitotecnia de Águas Belas (ABE) and Instituto de Pesquisas Hidráulicas (IPH). One hundred pupas were placed in each locality; 50 next to the ground and 50 above one meter. Each level of height were made by 25 pairs (25 cryptic and 25 non-cryptic). Among the 50 pupas of each level 25 pupae were light and 25 were dark. The results indicate dependence between survival and the level of when the pupas were placed. The camouflage and the colour not shown to be significant to the survival of the pupa in the field. Pupae placed near the ground have a chance between 8 and 13 times larger to be preyed than pupae over 1.0 meter tall. These results were linked to the type of predator observed in the field: ants. Two species and one genus of ants were observed preying pupae. Thus it is concluded that for pupae close to the ground, the camouflage provided by the colour is not adaptive, as the main predators not depend on the vision. The last article deals with the description of the first case of methylation in a butterfly: H. erato phyllis. Previous studies have shown that the estimates of heritability for the pupal colour in this species, determined by the method of linear regression, had different values when were considered male and their broods and female and their broods. One possible cause for this discrepancy could be due to the occurrence of DNA methylation and the possible silencing of genes. Through technical of MSRE and random primers PCR bisulfite we investigate the presence of methylation in H. erato phyllis. The analysis, using DNA from different stages of development, showed that the technique of PCR bisulfite was particularly efficient to indicate the presence of DNA methylation in this species. This opens the possibility of future research directed to assess whether this phenomenon has connection with the determination of differential colour patterns of the H. erato phyllis pupae. Finally this thesis has three appendices. The first is a process of selection for extreme phenotypes for the colour of the pupa in H. erato phyllis. Preliminary results indicate that the procedure is successful, however slow, as the variance for the feature remained relatively high when the process was interrupted. The second appendix addresses the effect of nuclear polyhedrosis virus on the pupal colour of this species. The event came after the emergence of dark pupae in the selection process for light pupae, when this phenotype was not more expected. So were investigated the effect of the virus on the pupal colour and on the survival of caterpillars, 4 th and 5 th instars. The results indicate that the virus does not influence the pupal colour. Moreover, there is a direct relationship between survival and individuals not infected. The third appendices describes an unusual phenotype to H. erato phyllis, where the adults had not red and yellow pigments, leaving the imago with a pattern black and white instead of black and red.

O terceiro anexo descreve um fenótipo incomum para H. erato phyllis, onde os adultos não apresentavam pigmentos vermelho e amarelo, deixando o imago com um padrão preto e branco ao invés de preto e vermelho. / This thesis, through four manuscripts, covers various aspects on the phenotypic modulation in the pupal color of the in Heliconius erato phyllis. The first manuscript evaluate by heritability (h2) estimative two methods to measure the degree of melanization of pupal exuviae. The first of them, the qualitative method, considers four scores, ranging from 2 to 5, in an ascending order of melanization. The second method, quantitative, uses the optical density. After the capture, the images were analyzed by software from which were obtained values for the degree of melanization of each exuviae. From the values obtained for each methodology the h2 were estimated by the methods of linear regression and analysis of variance, with n ranging between 7 and 18 broods. Estimates of the h2 resulted in similar values for both methods of pupal categorization. Thus it is concluded that the qualitative methodology, for its usefulness, it is the most suitable for the task. From the low values of h2, we have two hypotheses. The first considers the sample size and the second the allocation priority resource. The second manuscript evaluates the influence of photoperiod on the pupal colour of H. erato Phyllis. Three broods had their eggs distributed between five photoperiods: 11L:13E, 12L:12E, 14L:10E, 9L:15E and 24L:00E. The first three represent photoperiod to Porto Alegre (RS-Brazil). The last indicated photoperiod is used as standard treatment by our group and was considered a control. The results found through the ANOVA twofactors to indicate a difference between the broods (p = 0,069) and do not indicate the difference between treatments. A second analysis was done by ANOVA one-factor considering only the pupal colour as a dependent on photoperiod. The results show a dependency among the factors (p = 0,047). A third analysis was performed to determine if the difference observed was due to the treatments or because of the genetic factors. To do this, a comparison at the treatment 24L:00E between the three broods with a set of 22 broods, reared under the same photoperiod, was performed. The result points to a genetic difference, concluding that the pupal colour of the in H. erato phyllis is not affected by photoperiod. The adaptation of pupas in field is the subject of the third manuscript. Two locations were used as the study area: Estação de Fitotecnia de Águas Belas (ABE) and Instituto de Pesquisas Hidráulicas (IPH). One hundred pupas were placed in each locality; 50 next to the ground and 50 above one meter. Each level of height were made by 25 pairs (25 cryptic and 25 non-cryptic). Among the 50 pupas of each level 25 pupae were light and 25 were dark. The results indicate dependence between survival and the level of when the pupas were placed. The camouflage and the colour not shown to be significant to the survival of the pupa in the field. Pupae placed near the ground have a chance between 8 and 13 times larger to be preyed than pupae over 1.0 meter tall. These results were linked to the type of predator observed in the field: ants. Two species and one genus of ants were observed preying pupae. Thus it is concluded that for pupae close to the ground, the camouflage provided by the colour is not adaptive, as the main predators not depend on the vision. The last article deals with the description of the first case of methylation in a butterfly: H. erato phyllis. Previous studies have shown that the estimates of heritability for the pupal colour in this species, determined by the method of linear regression, had different values when were considered male and their broods and female and their broods. One possible cause for this discrepancy could be due to the occurrence of DNA methylation and the possible silencing of genes. Through technical of MSRE and random primers PCR bisulfite we investigate the presence of methylation in H. erato phyllis. The analysis, using DNA from different stages of development, showed that the technique of PCR bisulfite was particularly efficient to indicate the presence of DNA methylation in this species. This opens the possibility of future research directed to assess whether this phenomenon has connection with the determination of differential colour patterns of the H. erato phyllis pupae. Finally this thesis has three appendices. The first is a process of selection for extreme phenotypes for the colour of the pupa in H. erato phyllis. Preliminary results indicate that the procedure is successful, however slow, as the variance for the feature remained relatively high when the process was interrupted. The second appendix addresses the effect of nuclear polyhedrosis virus on the pupal colour of this species. The event came after the emergence of dark pupae in the selection process for light pupae, when this phenotype was not more expected. So were investigated the effect of the virus on the pupal colour and on the survival of caterpillars, 4 th and 5 th instars. The results indicate that the virus does not influence the pupal colour. Moreover, there is a direct relationship between survival and individuals not infected. The third appendices describes an unusual phenotype to H. erato phyllis, where the adults had not red and yellow pigments, leaving the imago with a pattern black and white instead of black and red.

Sistemática e biogeografia de Labrundinia Fittkau, 1962 (Diptera: Chironomidae : Tanypodinae): uma abordagem morfológica e molecular

The basis of this study rested on the examination of the type material of all previously described Labrundinia species, including species deposited in national and foreign museums. Field work were carried out to collect more specimens and to secure associations between larvae, pupae and adults. In this study 14 known species of Labrundinia were redescribed and 25 new species were described from the Neotropical region. Morphological and molecular analyses were conducted to infer phylogenetic relationships among all species of the genus and identification keys were developed for all life stages. Based on the results of the phylogeny and distribution of the species, a biogeographic analysis was performed to provide the most probable biogeographic history for the group. The interpretations of the phylogenetic results indicate Labrundinia as a monophyletic group. The group is shown in all resulting trees. Based on these phylogenetic reconstructions, biogeographical analyses were run and indicated Labrundinia ancestor as having its initial diversification in the Neotropical region. / Esta pesquisa de doutorado enfocou a sistemática e a biogeografia das espécies de Labrundinia (Diptera: Chironomidae: Tanypodinae), sendo que a base deste estudo incidiu no exame do material-tipo de todas as espécies descritas para o gênero, incluindo espécies depositadas em coleções nacionais e estrangeiras. Adicionalmente, trabalho de campo foi realizado com objetivo de coletar novas espécies e garantir associações seguras entre larvas, pupas e adultos. Neste estudo, foram redescritas 14 espécies conhecidas de Labrundinia e 25 espécies novas para a ciência foram descritas para região Neotropical. Dados morfológicos e moleculares foram utilizados para inferir relações filogenéticas entre espécies do gênero e chaves de identificação foram preparadas para os adultos machos, pupas e larvas. Com base nos resultados da filogenia e distribuição das espécies, análises biogeográficas foram conduzidas na tentativa de determinar a mais provável história biogeográfica para o grupo. As interpretações das análises filogenéticas indicam o gênero Labrundinia como um grupo monofilético, com presença em todas as árvores resultantes. Com base nestas reconstruções filogenéticas, análises biogeográficas foram processadas e indicam que o ancestral de Labrundinia teve diversificação inicial na Região Neotropical.

Fatores genéticos e ambientais no condicionamento da cor da pupa em Heliconiu erato phylis, Fabricius 1775 (Lepidoptera: Nymphalidae)

Neste trabalho procurou-se analisar a coloração das pupas em Heliconius erato phyllis em relação aos componentes genético e ambiental. Um total de 25 proles foi utilizado neste estudo, sendo 17 a partir de cruzamentos feitos em insetário e 8 provenientes de fêmeas já fecundadas trazidas da natureza. A coloração das pupas seguiu um sistema de escores discretos sendo a cor mais clara categorizada como 2 e a mais escura como 5. Para testar o efeito ambiental (na realidade, apenas um componente ambiental, particularmente a sensibilidade da lagarta à um ambiente claro ou à um ambiente escuro), cada prole foi dividida em três tratamentos: controle, preto, e branco, com as lagartas sendo transferidas para estes tratamentos no início do 5º instar. O tratamento controle consistia na criação das larvas em potes translúcidos, com o fundo coberto por papel de cor bege e à temperatura de 25o C + 1o C com iluminação permanente. Os dois outros tratamentos diferiam do anterior por terem todo o revestimento interno dos potes, ora com papel branco, ora com papel preto; nos demais itens, os tratamentos eram idênticos. As lagartas, individualizadas, eram alimentadas diariamente. As pupas desenvolvidas no tratamento preto obtiveram escore 5 ou 4 e as desenvolvidas no tratamento branco não diferiram estatisticamente daquelas do controle. Este caso pode ser visto com um exemplo de modulação fenotípica na qual o grau de melanização da pupa é influenciado por um ambiente preto. Para obter-se maiores informações em relação ao tempo de sensitividade das lagartas ao ambiente preto, elas foram testadas em diferentes tempos de desenvolvimento. Quando elas foram submetidas ao tratamento preto na fase de pré-pupa, estas não mostraram diferenças em relação ao tratamento controle, mas sim quando colocadas sob este tratamento no instar anterior, indicando que o período crítico na indução da melanização ocorre de forma breve no 5º instar. O componente genético foi estudado através dos cruzamentos onde levouse em conta o grau de parentesco (coeficientes de endocruzamento de irmãos inteiros de 0,0; 0,125; 0,25 e 0,375) e a estimativa da herdabilidade (h2) pelos métodos da regressão e da análise da variância. Quatro proles dos cruzamentos foram endocruzadas e assim descartadas das estimativas de herdabilidade. Todavia, elas mostraram uma variância decrescente nos escores das colorações das pupas quando o coeficiente de endocruzamento aumentava, sugerindo uma base genética para a coloração. As estimativas da herdabilidade situaram-se ao redor de 0,30 na regressão da média dos escores da prole sobre a média dos genitores. A regressão da média da prole sobre o escore dos pais (masculinos) forneceu valores de h2 maiores, variando de 0.40 a 0.73. Esta foi negativa e extremamente baixa quando os escores maternos estavam envolvidos. As estimativas pela análise da variância resultaram em um intervalo de 0.48 a 0.55. Um resultado adicional que reforça a idéia de um componente genético, foi obtido a partir do tratamento preto: mesmo quando larvas de 5º instar foram sujeitas ao ambiente preto, com as pupas resultantes sendo escuras, a análise da variância mostrou diferenças significantes entre as proles, o que sugere uma diferença genética entre estas. Assim, mesmo havendo uma forte influência do ambiente preto na indução de pupas pretas, a resposta das proles mostrou variação que deve ser devida à diferenças genéticas. Os resultados encontrados no presente trabalho indicam que o componente genético na melanização da pupa é compatível com o modelo de controle poligênico. / This work analysed the genetic and the environmental influence on the pupal colouration in the butterfly Heliconius erato phyllis. A total of 25 offspring were used in the study, 17 from crosses made in insectary, and 8 from females brought from nature, already inseminated. Pupal colour followed a system of discrete scores, from 2 for the light pupae to 5 for the darkest ones. To test the effect of environmental alteration (actually only one environmental component, namely the caterpillar sensitivity to a light or to a dark environment), each offspring was subjected to three treatments: control, darkness and whiteness, with caterpillars being transferred to these treatments in the beginnings of their 5th instar. The control treatment consisted in rearing caterpillars in translucent pots, with the bottom covered with beige paper and at a temperature of 25o C + 1o C and with permanent illumination. The two other treatments differed from the previous one by having their pots covered internally with white or with black paper; the remaining procedures being similar. Caterpillars, growing individually, received daily abundant food. Pupae from the dark environment originated pupae that scored 5 or sometimes 4; for those in the white environment, there were no differences with the controls. The case can be viewed as an example of phenotypic modulation, in which the degree of melanization of the pupa is influenced by a dark environment. To get more details on the time the caterpillars were sensible to the darkening of environment, they were tested at different age of development. When they were subjected to the dark treatment at the prepupal stage, the score for the pupae did not differ statistically from those of the control treatment. However, when subjected to the dark environment at the 5th instar, the resulting pupae had scores statistically different from the control treatment. This indicates that the critical period for inducing melanization is shortly around the 5th instar. The genetic component was studied through crosses where the degree of relatedness were taken into account (inbreeding coefficients of the sibship being 0.0 for the unrelated, and 0.125, 0.25, and 0.375). Heritability estimates (h2) were done by the methods of regression and analysis of variance. Four offspring from the crosses were inbred and were discarded in the estimates of heritability. However, they showed a decreasing variance in the scores for pupal colour as the inbreeding coefficient increased, suggesting a genetic background for colour. Estimates of heritability were around 0.30 when the score of the offspring were regressed on the midparental value. When regressed on the male parent, h2 was larger, varying from 0.40 to 0.73 and was generally negative and very low when female parent was the regressor. Estimates by the analysis of variance resulted in the interval 0.48 - 0.55. One additional fact comes from the black treatment. Even when fifth instar larvae were subjected to a dark environment, with the resulting pupae being dark, the analysis of variance showed a significant difference among offspring, which suggests genetic differences among the offspring not totally inhibited by the dark treatment. Our results dealing with the genetic component of pupal melanization are compatible with a polygenic control.

This taxon is commonly cited in ecological studies, always in generic level. In the Neotropical region are known 10 species, however none specifically mentioned for Brazil. The sampling followed the conventional methods of benthonic studies. In laboratory the larvae were kept alive to obtain the three development phases: larva, pupa and adult. The results allowed the identification of five species*, four of which are new species and the first record of Labrundinia tenata Roback (1987) for Brazil. The species are described with their respective diagnoses. Information about species natural history of genus is presented and propels for the identification keys, for the different stages of life, are given. / O objetivo deste trabalho foi estudar espécies de Labrundinia Fittkau, 1962 (Diptera: Chironomidae: Tanypodinae: Pentaneurini) no Estado de São Paulo. Esse táxon é comumente citado em muitos trabalhos de cunho ecológico, sempre em nível genérico. Na região Neotropical são conhecidas 10 espécies, porém nenhuma especificamente mencionada para o Brasil. As coletas em campo seguiram a metodologia convencional para estudos bentônicos. Em laboratório as larvas foram mantidas vivas para a obtenção das três fases de desenvolvimento: larva, pupa e adulto. Os resultados permitiram a identificação de cinco espécies*, das quais quatro são espécies novas para a ciência e o primeiro registro de Labrundinia tenata Roback (1987) para o Brasil. As espécies são descritas com as respectivas diagnoses. Informações sobre a história natural das espécies do gênero são apresentadas e propostas de chaves de identificação, para os diferentes estágios de vida, são dadas.

Ontogeny of the pupal salivary, hypopharyngeal and mandibular glands and the role of apoptosis during metamorphosis in Apis mellifera

abstract: Amongst the most studied of the social insects, the honey bee has a prominent place due to its economic importance and influence on human societies. Honey bee colonies can have over 50,000 individuals, whose activities are coordinated by chemical signals called pheromones. Because these pheromones are secreted from various exocrine glands, the proper development and function of these glands are vital to colony dynamics. In this thesis, I present a study of the developmental ontogeny of the exocrine glands found in the head of the honey bee. In Chapter 2, I elucidate how the larval salivary gland transitions to an adult salivary gland through apoptosis and cell growth, differentiation and migration. I also explain the development of the hypopharyngeal and the mandibular gland using apoptotic markers and cytoskeletal markers like tubulin and actin. A regressão da média da prole sobre o escore dos pais (masculinos) forneceu valores de h2 maiores, variando de 0.40 a 0.73. Esta foi negativa e extremamente baixa quando os escores maternos estavam envolvidos. As estimativas pela análise da variância resultaram em um intervalo de 0.48 a 0.55. Um resultado adicional que reforça a idéia de um componente genético, foi obtido a partir do tratamento preto: mesmo quando larvas de 5º instar foram sujeitas ao ambiente preto, com as pupas resultantes sendo escuras, a análise da variância mostrou diferenças significantes entre as proles, o que sugere uma diferença genética entre estas. Assim, mesmo havendo uma forte influência do ambiente preto na indução de pupas pretas, a resposta das proles mostrou variação que deve ser devida à diferenças genéticas. Os resultados encontrados no presente trabalho indicam que o componente genético na melanização da pupa é compatível com o modelo de controle poligênico. / This work analysed the genetic and the environmental influence on the pupal colouration in the butterfly Heliconius erato phyllis. A total of 25 offspring were used in the study, 17 from crosses made in insectary, and 8 from females brought from nature, already inseminated. Pupal colour followed a system of discrete scores, from 2 for the light pupae to 5 for the darkest ones. To test the effect of environmental alteration (actually only one environmental component, namely the caterpillar sensitivity to a light or to a dark environment), each offspring was subjected to three treatments: control, darkness and whiteness, with caterpillars being transferred to these treatments in the beginnings of their 5th instar. The control treatment consisted in rearing caterpillars in translucent pots, with the bottom covered with beige paper and at a temperature of 25o C + 1o C and with permanent illumination. The two other treatments differed from the previous one by having their pots covered internally with white or with black paper; the remaining procedures being similar. Caterpillars, growing individually, received daily abundant food. Pupae from the dark environment originated pupae that scored 5 or sometimes 4; for those in the white environment, there were no differences with the controls. The case can be viewed as an example of phenotypic modulation, in which the degree of melanization of the pupa is influenced by a dark environment. To get more details on the time the caterpillars were sensible to the darkening of environment, they were tested at different age of development. When they were subjected to the dark treatment at the prepupal stage, the score for the pupae did not differ statistically from those of the control treatment. However, when subjected to the dark environment at the 5th instar, the resulting pupae had scores statistically different from the control treatment. This indicates that the critical period for inducing melanization is shortly around the 5th instar. The genetic component was studied through crosses where the degree of relatedness were taken into account (inbreeding coefficients of the sibship being 0.0 for the unrelated, and 0.125, 0.25, and 0.375). Heritability estimates (h2) were done by the methods of regression and analysis of variance. Four offspring from the crosses were inbred and were discarded in the estimates of heritability. However, they showed a decreasing variance in the scores for pupal colour as the inbreeding coefficient increased, suggesting a genetic background for colour. Estimates of heritability were around 0.30 when the score of the offspring were regressed on the midparental value. When regressed on the male parent, h2 was larger, varying from 0.40 to 0.73 and was generally negative and very low when female parent was the regressor. Estimates by the analysis of variance resulted in the interval 0.48 - 0.55. One additional fact comes from the black treatment. Even when fifth instar larvae were subjected to a dark environment, with the resulting pupae being dark, the analysis of variance showed a significant difference among offspring, which suggests genetic differences among the offspring not totally inhibited by the dark treatment. Our results dealing with the genetic component of pupal melanization are compatible with a polygenic control.

Therefore, studies on population dynamics under different abiotic conditions, as well as investigations on diperan control are very important. This study was divided into four chapters that approached the subject from different perspectives. The first two chapters are related to post-embryonic development of Chrysomya megacephala and Chrysomya albiceps submitted to different photoperiods (L0:D24, L12:D12, L16:D8, L24:D0). These species responded differently, however both species had a faster development for the different stages and their viabilities were higher in the 24h scotophase ( 24 hours of darkness) when compared to the other photoperiods. There was a trend for deceleration of the development as the photophase increased and it was evident for the larval stage of the both species. However, in C. albiceps was more pronouced for newly- larvae hatched to adult period, presenting a difference of up to four days between the shorter photophase (L0:D24) and the longer photophase (L24:D0). The total period of development of C. megacephala was shorter when the 24hscotophase was compared to the others. Therefore, it was possible to infer that the different photoperiods presented a circadian rhythm of emergence with distinct daily emergence rate, suggesting that this phenomenon was primarily regulated by an endogenous circadian clock, even if they presented modulations by photophases. In the third chapter, the aim was to investigate the biological aspects of Nasonia vitripennis (Pteromalidae) when the female explored hosts buried at different depths ranging from 0.0 to 5.0 cm at 0.5 cm intervals, for two periods of exposition: 24 and 48h. It was observed that two factors, period of exposition and pupation depths, caused differences in the parasitoid progeny and had a relation of dependence. For the group exposed for 48h, the pupae were parasitized only when buried up to 2.0 cm depth, however, for the group exposed for 72h, the parasitic capacity of parasitoids reached pupae buried up to 3.0 cm deep. The group exposed for 48h had a decreasing number of female emerged per pupae as the depth increased. This trend was not observed for the group exposed for 72h. The number of male emerged per pupae did not differ among the depths and periods of expositions. The sex deviation was higher for female for both exposure periods and for all depths. In the fourth chapter was the effect of a cornoside extracte from Parahancornia amapa (Apocynaceae) on post-embryonic development of Chrysomya putoria and progeny of N. vitripennis was investigated. The larval weigh and post-embryonic developmental period of C. putoria were not affected, with exception for the pupal development that was slower with the treatment with 5% cornosideo when compared to the control (distilled water). The newly-hatched larvae to adult period presented a viability of around 60 % for boths treatments. In relation to to the effect of the cornosideo on progeny of the parasitoids the following parameters were analysed: number of parasitoids emerged per pupae, sex ratio and developmental period. None of these were affected in comparison to the control group. The lack of effect of 5% cornoside on calliphorid flies may be a negative aspect when control is being concerned. However the lack of effect on the parasitoid, used as natural controllers of calliphoridas, is a / Califor?deos apresentam import?ncia m?dica-sanit?ria e tamb?m na entomologia forense, pois auxiliam a data??o do intervalo p?s-morte. Em C. megacephala houve diferen?a somente quando o grupo submetido ?s 24h de escotofase foi comparado aos demais fotoper?odos, tendo o desenvolvimento mais acelerado. Foi poss?vel inferir que o ritmo de emerg?ncia teve periodicidade circadiana, embora com taxas di?rias de emerg?ncia distintas nos diferentes fotoper?odos. No terceiro cap?tulo objetivou-se investigar aspectos biol?gicos de Nasonia vitripennis (Pteromalidae) quando expostas aos hospedeiros enterrados em diferentes profundidades, variando de 0,0 a 5,0 cm, a cada 0,5 cm, por dois per?odos, 24h e 48h. Observou-se que a explora??o do pup?rio foi influenciada pelos dois fatores, tempo e profundidade, havendo uma rela??o de depend?ncia entre eles. Em 48h de exposi??o foram observados pup?rios parasitados somente at? a profundidade de 2,0 cm, por?m em 72h a capacidade parasit?ria foi at? 3,0 cm. Houve um decr?scimo do n?mero de f?meas emergidas por pupa com o aumento das profundidades em 48h, no entanto esta tend?ncia n?o foi observada em 72h. O n?mero de machos por pupa n?o diferenciou entre as profundidades e nem entre os dois tempos de exposi??o. No quarto cap?tulo foi investigada a a??o do cornos?deo, a fra??o metan?lica do l?tex de Parahancornia amapa (Apocynaceae), sobre o desenvolvimento de Chrysomya putoria e sobre a prog?nie de N. vitripennis. Foi observado que o peso larval e o tempo de desenvolvimento p?s-embrion?rio de C. putoria n?o foram alterados, com exce??o do per?odo pupal que foi mais lento no tratamento com cornos?deo a 5% quando comparado ao grupo controle (?gua destilada). A viabilidade de neolarva a adulto foi em torno de 60% para ambos os tratamentos. Quanto ? influ?ncia do cornos?deo sobre a prog?nie dos parasit?ides, o n?mero de parasit?ides, raz?o sexual e tempo de desenvolvimento n?o foram alterados quando comparados ao controle. P?de se dizer que o cornos?deo a 5% n?o exerceu efeito sobre os califor?deos, aspecto negativo para o controle, e nem tampouco em seus controladores naturais, aspecto positivo.
