Konstrukční návrh dřevoobráběcího stroje pro přípravu přířezů / Construction design of woodworking machinery for preparing sawn timber

This thesis deals primary with design of a woodworking machine focused on woodworking technology by cutting – rip saw. The introduction to the work is get to acquainted with the material to be processed – wood, and also the search of associated machines with the same technology of workpiece processing. Next part is focused on the safety of machining on woodworking machines and the individual safety features that the machines must contain are also analyzed. From this survey, a saw model with all the necessary features is created. The conclusion of the thesis is devoted to the risk analysis of the proposed machinery and their reduction.

Epoxy filling simulation of an RIP bushing

The manufacturing process for the core of an RIP bushing was studied. Simulations of a test model was performed, due to the non-linear nature of the fluid filling process. Varying of governing parameters in the model, that could realistically be varied, was carried out and their affect was observed. The model was then compared to real measurements to view its practical reliability. Proposals to optimize the filling speed of the process in order to avoid the creation of bubbles was discussed and conducting it would be the next step for further research of the subject.

Modélisation morphodynamique des plages sableuses

Le long des littoraux sableux tels que la côte Aquitaine, les vagues et principalement les courants moyens induits par celles-ci jouent un rôle majeur dans l’évolution morphologique des plages. Que ce soit d’un point de vue socio-économique (aménagement des plages, protection de l’environnement) ou sécuritaire (noyade), comprendre la dynamique de tels environnements est devenu un réel enjeu scienti?que ces dernières années. Le développement d’une plateforme de modélisation morphodynamique opérationnelle a été menée au cours de cette thèse. Ce modèle est basé sur le couplage du modèle spectral de houle SWAN (Delft University of Technology) avec le modèle MARS 2DH (Ifremer), initialement conçu pour calculer les courants induits par la marée et les vents. Après avoir ajouté les phénomènes liés à la houle, un modèle sédimentaire a été développé pour estimer les évolutions morphodynamiques. La campagne de mesures Biscarrosse-PréECORS menée en juin 2007 sur la plage de Biscarrosse a permis de valider le modèle pour une large gamme d’état de mer. Le modèle a montré son e?cacité et sa robustesse pour décrire les courants et des circulations induites par les vagues au dessus de fortes hétérogénéités bathymétriques. Nous avons mis en évidence la forte modulation tidale des courants sagittaux qui sont intenses même pour de faibles conditions de vagues. La plateforme de modélisation morphodynamique a permis d’analyser la formation et le développement de morphologies tridimensionnelles rythmiques pour des systèmes simple et double barre. Pour les environnements à double barre comme la côte Aquitaine, l’intense couplage morphologique entre la barre externe et la barre interne a été étudié en réponse aux conditions de forçages. Nous avons mis en évidence l’in?uence de la marée sur la croissance des barres et révélé la corrélation existant entre croissance des barres et dissipation d’énergie des vagues sur les barres. Cette thèse a abouti au développement d’un modèle complet permettant d’appréhender la dynamique des environnements littoraux sableux. / Along many sandy environments as the Aquitanian Coast, waves and wave-induced currents play a key role on the beach morphodynamics. Within a socio-economic (human interventions, natural environment protection) and touristic (human safety) framework, understand the complex dynamics of these systems is a real scienti?c challenge these last decades. The development of an operational morphodynamical modeling platform was performed during this PhD thesis. The modeling strategy consists in coupling the spectral wave model SWAN (Delft Univer- sity of Technology) with the non-linear shallow water model MARS 2DH (Ifremer). The MARS model was initially designed to compute tide- and wind-induced currents. Thus, the wave processes and a sed- imentary unit were implemented inside MARS to compute morphological evolutions of wave-dominated sandy beached. The Biscarrosse-PréECORS ?eld experiment carried out in June 2007 at Biscarrosse Beach was used to validate our modeling platform for a large range of o?shore wave conditions. Results have proved the e?ciency and the robustness of our model to calculate wave-induced currents and circulations over alongshore non-uniformities. The strong tidal modulation of currents and the intense rip currents for low-energy conditions were highlighted. The morphodynamical modeling platform was also used to analyze the generation and the development of rhythmic three-dimensional morphologies for one- and double-barred systems. For double-barred en- vironments, the intense morphological coupling between the inner and outer bar was studied in the response of wave and tide forcing. We have highlighted the in?uence of the tide on the bar growth and showed the correlation between the growth and the wave dissipation over the bar. This thesis has succeeded in the development of a complete model for understanding the dynamics of sandy coastal environments.

MOCVD and electrochemical polymeric thin films : elaboration, characterization, properties ans applications / FIlms minces polymères par CVD et électrodeposition : élaboration, caractérisation, propriétes et applications

Ce mémoire traite de deux types de polymères en films minces: le poly (méthacrylate de méthyle) (PMMA) et le polypyrrole (PPy). Ces films minces ont été déposés par voie sèche et par voie humide. La voie sèche consiste à faire croitre les films polymères par un procédé original de dépôt chimique en phase vapeur assisté par photons (Chemical Vapor Deposition, CVD). La croissance implique l'activation UV des espèces monomères dans la phase gazeuse. Les deux polymères PMMA et PPy ont été obtenus pour la première fois par ce procédé de photo-CVD. La caractérisation des propriétés a mis en évidence des applications possibles en microélectronique, micro-optique et les dispositifs générant de la chaleur. La voie humide mise en œuvre pour déposer des films minces de polymères et d’hybrides organiques/inorganiques est une méthode électrochimique. Des films de PPy (organique) et de TiO2 nanostructuré (inorganique) ont été obtenus et caractérisés par différentes techniques électrochimiques. Des aspects supplémentaires de la croissance de PPy ont été mis en évidence par la résonance des plasmons de surface. / The thesis deals with two types of polymeric thin films: poly (methyl methacrylate) (PMMA) and polypyrrole (PPy). The thin films were grown by a dry and a wet route. The dry route involved the growth of the polymeric films by an original process of Chemical Vapor Deposition, namely Photo-CVD. The growth involves the UV activation of the monomer species in the gas phase. Both PMMA and PPy were obtained for the first time by this Photo-CVD. The characterization highlighted properties with possible applications in microelectronics, micro-optics and as heat generating devices. The wet route involved the growth of polymeric and hybrid organic/inorganic thin films by an electrochemical approach. Organic PPy and inorganic TiO2 nanostructures were obtained and characterized by various electrochemical techniques. The growth aspects of PPy were supplementary highlighted by the Surface Plasmon Resonance.

Synthèse et caractérisation d’hydrogels de fibrine et de polyéthylène glycol pour l’ingénierie tissulaire cutanée / Synthesis and characterization of fibrin/polyethylene glycol based for skin tissue engineering

Depuis plus d’une cinquantaine d’années, de formidables avancées ont été initiées dans le domaine de l’ingénierie tissulaire cutanée menant à la reconstruction in vitro de substituts de peau. La plupart sont des substituts dermiques destinés à être utilisés comme aide à la cicatrisation des plaies aigües et chroniques en complément des traitements de greffes conventionnels ainsi que pour l’augmentation des tissus mous. Bien qu’un nombre croissant de patients aient pu bénéficier de ces matrices dermiques, leur application clinique reste encore restreinte, en raison de leur coût élevé mais également à cause de résultats cicatriciels parfois peu satisfaisants. Par conséquent, il reste un défi de taille, celui de développer des substituts dermiques stimulant activement la cicatrisation, présentant un faible coût de production, sans propriétés antigéniques et possédant des propriétés mécaniques adaptées. Dans ce cadre, les hydrogels à base de fibrine constituent des candidats prometteurs, en particulier en raison du rôle central de cette protéine dans la cicatrisation. Le principal inconvénient est qu’à concentration physiologique, ces hydrogels sont faibles mécaniquement, ce qui les rend difficilement manipulables. L’objectif de cette thèse a été la mise au point ainsi que la caractérisation de différents hydrogels destinés à être utilisés comme substituts dermiques. Ces derniers présentent l’avantage d’associer les propriétés biologiques de la fibrine avec les propriétés mécaniques d’un polymère synthétique, le polyéthylène glycol dans une architecture de réseaux interpénétrés de polymères (RIP). Les résultats obtenus ont permis : - de confirmer les propriétés physico-chimiques des RIP développés initialement par nos collaborateurs de l’université de Cergy-Pontoise, - de valider en trois étapes (in vitro, ex vivo puis in vivo) la biocompatibilité de ces nouvelles matrices, destinées à être utilisées comme supports de culture 2D et pour l’augmentation des tissus mous, - d’élaborer et de caractériser des matrices macroporeuses, optimisées pour la culture 3D de fibroblastes de dermes humains. / Over the past five decades, we assisted in extraordinary advances in the field of skin tissue engineering which led to the in vitro reconstruction of a wide range of skin substitutes. Most of them are dermal substitutes: Their clinical application ranges from treating acute and chronic wounds to soft tissue augmentation. Although increasing numbers of patients have been treated with dermal substitutes, their clinical application has been limited by their substantial cost and some poor healing outcomes. Hence, there is still a challenge to produce a dermal substitute which enhance sufficiently wound healing. To this end, the substitute should exhibit suitable properties for enabling the repair process. Other requirements such as excellent biocompatibility, minimal antigenicity, ease to handle and cost-effective production are also essential. In this context, fibrin hydrogels constitute promising candidates for skin tissue engineering since fibrin fibers form a physiological and provisional backbone during wound healing. However, the poor mechanical properties of fibrin-based hydrogels at physiological concentration are an obstacle to their use. In this study, our aim was to design and characterize mechanically reinforced fibrin-based hydrogels by combining the intrinsic properties of a fibrin network with the mechanical features of a polyethylene glycol network using an interpenetrating polymer network (IPN) architecture. They are intended to be used as dermal scaffolds. The results obtained in this thesis: - Confirmed the suitable physico-chemical properties of IPN, first developed by our partner of the University of Cergy-Pontoise. - Validated their biocompatibility using a three-step approach (in vitro, ex vivo and in vivo assays). - Led to the synthesis and characterization of a new type of fibrin-based macroporous matrices, optimized for 3D dermal fibroblast culture.

Simulation of the Filling of Epoxy in Resin Impregnated Paper Bushings : Analysis of Temperature Measurements during Filling of Epoxy and the Generation of New Filling Recipes to Avoid Cavities in the RIP Bushing

The goal of the project was to create a model in COMSOL Multiphysics that can simulate thefilling of resin (epoxy) in Resin Impregnated Paper (RIP) bushings. Using the model new fillingrecipes that avoid problematic double-fronts and thereby avoid the creation of cavities in thebushings should be designed. Another goal was to use Thermocouples and Fiber optics tomeasure the temperature at different positions during the filling to get a better understanding ofthe filling. The first step was to design and determine functions, parameters (such as thepermeability), boundary conditions etc. for the simulation model in COMSOL Multiphysics and tocalibrate the model to align with measurements of fillings previously done. The second part wasto increase the filling time to minimize the maximum difference between filling fronts in thebushings and thereby decrease the risk of problematical double-fronts that create cavities,which decrease the bushing's dielectrical properties. The simulation model aligned somewhatwith the measured data but there is a lot of uncertainty both in the measurements and due toassumptions made in the simulation model. The measurements from Thermocouples and Fiberoptics where almost identical and they indicated that the filling of the bushing is symmetric.

Engineering of the RTB Lectin as a Carrier Platform for Proteins and Antigens

The major obstacle many promising drugs struggle to overcome is the barrier imposed by the outer cell membrane. In addition to technologies such as liposomes and cell-penetrating peptides, more attention is being given to the class of proteins known as lectins to deliver therapeutic and antigenic proteins to the interiors of cells. Lectins bind to but do not modify sugars, and provide an efficient route to endocytosis. The galactose/N-acetyl-galactosamine specific lectin ricin B-chain (RTB) is especially attractive in possibly fulfilling a carrier role due to its well-characterized endocytotic trafficking and its efficacy over a wide range of cell types. By producing RTB recombinantly in plants it is possible to create a fully active, non-toxic carrier that does not rely on the processing of large amounts of toxic material (e.g. castor bean). Payload molecules such as small molecules and proteins can be attached to RTB via chemical conjugation at primary amine groups, without the loss of lectin or uptake activities. The biotin/streptavidin interaction and direct genetic fusion of polypeptides also provide efficient mechanisms for the attachment of payload proteins to RTB. An immunoglobulin domain-based scaffolding mechanism bridges modified RTB and payload proteins when co-expressed in Agrobacterium-infiltrated plant leaves. Carrier and payload proteins expressed in plants and E. coli, respectively, and purified independently are not able to assemble into an efficient carrier/payload arrangement. These findings show that plant cells are able to correctly produce the two components of the carrier/payload system and assemble them into an efficient and flexible capture and carry technology. / Ph. The aim of this work is to explore the joining of inspiration, such as different garments and materials, in relation to commonly used methods in the fashion industry when it comes to joining of different trends and references such as clashing and collaging. The work proposes a new method and framework for join- ing inspiration which generates different results depending on what kind of inspiration that is put in to it. A garment can roughly be broken down to a silhouette and shape, materials and details. The material put in to the method and framework is based on information from trend seminars for SS16, because that is how many of today’s trend-oriented fashion brands get there inspiration. Trendy garment silhouettes are mixed through processes of computational morphing in Adobe Flash by a generation of spin in the mixing process were shape hints are used in a new manner. The new generated silhouettes are further developed and materialized through procedures of interpretation and figuration. Different trendy materials are melded in a direct and concrete way through mixed media techniques such as laminating, fusing and vacuum-techniques. The final steps of the method is a garment shape and material synthesis with starting point in the generated shape with the final material. The projects intention is to let the physical experimentation, interpretation and figuration play a central role in the research process for new types of methods, garments, materials and expressive pos- sibilities.
