[pt] O presente trabalho desenvolve um novo modelo para um problema logístico de remanejamento de placas entre pilhas em um Pátio de Placas de uma Laminação, típico da indústria siderúrgica. Esta abordagem inclui uma nova característica que indica o destino das placas remanejadas. Uma nova metodologia de decisão altera sensivelmente o escopo do problema e traz um novo conceito: o balanço entre a escolha ótima das placas e o possível destino das placas remanejadas, ou seja, deve-se pesar a escolha da placa a ser laminada juntamente com a possibilidade de um bom remanejamento das placas que estão acima da
mesma. Esse modelo não linear é linearizado por técnicas de linearização e resolvido através de programação linear inteira. Com isso, o estudo apresenta uma redução no tempo médio de manuseio de placas (TMM) de 5 minutos por placa laminada encontrado na literatura existente, para menos de 4 minutos. Finalmente o modelo proposto incentiva a construção de uma ferramenta de gestão que possa
resolver o problema SSS em situações práticas do dia-a-dia através de redução de custo de operação como forma de viabilizar o investimento nesta ferramenta. / [en] This work develops a new model for a typical logistic problem in the steel industry known as slab stack shuffling problem inside a Slab Yard of a Rolling Mill. This new approach includes a new feature that indicates the destination stacks of shuffled slabs. A new decision methodology changes significantly thescope of the problem and introduces a new concept: the balance between optimal choice of the rolled slabs and possible destinations for shuffled slabs which means it must weigh the choice of the slab to be rolled engaged with a low cost shuffle operation for the slabs that are above the chosen slab. This nonlinear model is linearized through specific modeling techniques and solved by integer linear programming. Thus, this work shows a reduction in average handling time (TMM) from 5 minutes per rolled slab found in the literature, to less than 4 minutes. Finally, the proposed model encourages the construction of a management tool that can solve the SSS problem in practical daily situations through operating costs reduction as a payback of investment in this tool.

Développement de capteurs à pixels CMOS pour un détecteur de vertex adapté au collisionneur ILC / Development of CMOS pixel sensors for a vertex detector suited to the ILCFu, Yunan 09 May 2012 (has links)
Le travail de thèse a consisté, en priorité, à s’approprier les technologies d’intégration verticale en usage dans l’industrie pour réaliser des mémoires à plusieurs étages, et à en évaluer l’apport pour les capteurs à pixel CMOS (CPS). Cette approche s’appuie sur la capacité de l’industrie à interconnecter des puces amincies empilées les unes sur les autres. Elle ouvre la perspective d’associer plusieurs microcircuits superposés à un même pixel, en dépits de sa taille réduite. L’interconnexion est donc réalisée au niveau du pixel. Ce saut technologique permet de lever la majorité des obstacles à l’obtention de performances optimales des CPS. On peut en particulier combiner des puces réalisées dans des technologies CMOS très différentes, chacune optimale pour une fonctionnalité précise. La collection des charges du signal peut ainsi être réalisée dans une couche dédiée, les microcircuits de conditionnement analogique des signaux peuvent être concentrés dans une autre couche, une troisième couche pouvant héberger les parties numériques assurant la compression puis la transmission des signaux, etc. Ce progrès se traduit notamment par la possibilité de combiner haute résolution spatiale et lecture rapide, avec une amélioration probable de la tolérance aux rayonnements intenses.On s’affranchit de cette manière des limitations provenant des paramètres de fabrication des fondeurs, qui ne permettent pas à l’heure actuelle, de pleinement exploiter le potentiel des CPS à l’aide d’une technologie CMOS unique. / The thesis has been a priority as taking ownership of vertical integration technologies used in the industry to realize a multistage development, and to evaluate the contributions on CMOS pixel sensors (CPS). 3D integration technologies (3DIT) provide a way to mitigate this hampering correlation between speed and resolution, since they allow to staple layers of readout circuitry on top of the sensing layer, which results in a drastic increase of the functionalities located in (the shadow of) each pixel. A multi-layer structure allows for a higher spatial resolution because more and more transistors may be integrated vertically in a relatively small pixel. Moreover, bringing the components of the sensor closer to each other translates in a faster readout, owing to the reduction in the average length of the inner connecting wires. Vertical integration also opens up the possibility of combining different technologies best suited to each of the sensor main functionalities (signal sensing, analog and digital signal processing and transmission). It overcomes the limitations in this way from the foundry manufacturing parameters, which do not allow to fully exploit the potential ofCPS with a single CMOS technology. 3D-CPS are thus expected to overcome most of the limitations of standard 2DCPS, and are therefore suspected to over new perspectives for the innermost layer of the ILC vertex detector.

Caractérisation du rayonnement acoustique d'un rail à l'aide d'un réseau de microphones / Spatial characterization of the wheel/rail contact noise by a multi-sensors methodFaure, Baldrik 22 September 2011 (has links)
Le secteur des transports ferroviaires en France est marqué par un dynamisme lié notamment à l'essor du réseau à grande vitesse et à la réimplantation du tramway dans de nombreuses agglomérations. Dans ce contexte, la réduction des nuisances sonores apparaît comme un enjeu majeur pour son développement. Afin d'agir efficacement à la source, il est indispensable d'identifier et d'étudier précisément les sources responsables de ces nuisances au passage des véhicules. Parmi les approches possibles, les antennes microphoniques et les traitements associés sont particulièrement adaptés à la caractérisation des sources ponctuelles mobiles, omnidirectionnelles et décorrélées.Pour les vitesses inférieures à 300 km/h, le bruit de roulement constitue la source principale du bruit ferroviaire ; il résulte du rayonnement acoustique des éléments tels que les roues, le rail et les traverses. Le rail, dont la contribution au bruit de roulement est prépondérante aux moyennes fréquences (entre 500 He et 1000 Hz environ), est une source étendue et cohérente pour laquelle les principes classiques de traitement d'antenne ne sont pas adaptés.La méthode de caractérisation proposée dans cette thèse est une méthode inverse d'optimisation paramétrique utilisant les signaux acoustiques issus d'une antenne microphonique. Les paramètres inconnus d'un modèle vibro-acoustique sont estimés par minimisation d'un critère des moindres carrés sur les matrices spectrales mesurée et modélisée au niveau de l'antenne. Dans le modèle vibro-acoustique, le rail est assimilé à un monopôle cylindrique dont la distribution longitudinale d'amplitude est liée à celle des vitesses vibratoires. Pour le calcul de ces vitesses, les différents modèles proposés mettent en évidence des ondes vibratoires se propageant dans le rail de part et d'autre de chaque excitation. Chacune de ces ondes est caractérisée par une amplitude au niveau de l'excitation, un nombre d'onde structural réel et une atténuation. Ces paramètres sont estimés par minimisation du critère, puis utilisés pour reconstruire le champ acoustique.Dans un premier temps, des simulations sont réalisées pour juger des performances de la méthode proposée, dans le cas d'excitations ponctuelles verticales. En particulier, sa robustesse est testée en présence de bruit ou d'incertitudes sur les paramètres supposés connus du modèle. Les effets de l'utilisation de modèles dégradés sont également étudiés. Concernant l'estimation des amplitudes, les résultats ont montré que la méthode est particulièrement robuste et efficace pour les excitations les plus proches de l'antenne. En revanche, pour l'estimation des autres paramètres, les performances sont supérieures pour les positions d'antenne excentrées. De manière générale, le nombre d'onde est correctement estimé sur l'ensemble des fréquences étudiées. Dans les cas à faible atténuation, un traitement classique par formation de voies en ondes planes suffit. En ce qui concerne l'estimation de l'atténuation, la faible sensibilité du critère limite l'efficacité de la méthode proposée.Enfin, certains résultats obtenus à partir des simulations ont été vérifiés lors de mesures in situ. L'excitation d'un rail expérimental par un marteau de chocs a tout d'abord permis de valider le modèle vibratoire pour la flexion verticale. Pour tester la méthode d'optimisation paramétrique, le rail a également été excité verticalement à l'aide d'un pot vibrant. Les principaux résultats des simulations ont été retrouvés, et des comportements particuliers relatifs à la présence de plusieurs ondes dans le rail ont été observés, ouvrant des perspectives de généralisation du modèle vibratoire utilisé. / In France, railway transport has been boosted by the expansion of the high-speed rail service and the resurgent implantation of tram networks in many city centers. In this context, the reduction of noise pollution becomes a crucial issue for its development. In order to directly act on the source area, it is necessary to precisely identify and study the sources responsible for this nuisance at train pass-by. Among all the potential approaches, microphone arrays and related signal processing techniques are particularly adapted to the characterization of omnidirectional and uncorrelated moving point sources. For speeds up to 300 km/h, rolling noise is the main railway noise source. It arises from the acoustic radiation of various elements such as wheels, rail or sleepers. The rail, which mainly contributes to rolling noise at mid-frequencies (from 500 Hz to 1000 Hz approximately), is an extended coherent source for which classical array processing methods are inappropriate. The characterization method proposed in this thesis is an inverse parametric optimization method that uses the acoustical signals measured by a microphone array. The unknown parameters of a vibro-acoustical model are estimated through the minimization of a least square criterion applied to the entries of the measured and modelled spectral matrices. In this vibro-acoustical model, the rail is considered as a cylindrical monopole whose lengthwise amplitude distribution is obtained from the vibratory velocity one. The different models proposed to obtain this velocity highlight the propagation of vibration waves towards both sides of every forcing point. Each wave is characterized by an amplitude at the forcing point, a real structural wavenumber and a decay rate. These parameters are estimated by the minimization of the least square criterion, and are then used in the vibro-acoustical model to rebuild the acoustical field radiated by the rail. First, simulations are performed in order to appraise the performances of the proposed method, in the case of vertical point excitations. In particular, its robustness to additive noise and to uncertainties in the model parameters that are supposed to be known is tested. The effect of using simplified models is also investigated. Results show that the method is efficient and robust for the amplitude estimation of the nearest contacts to the array. On the other hand, the estimation of the other parameters is improved when the array is shifted away from the contact points. The wavenumber is generally well estimated over the entire frequency range, and when the decay rate is low, a single beamforming technique may be sufficient. Concerning the decay rate estimation, the efficiency of the method is limited by the low sensitivity of the criterion. At last, measurements are performed in order to verify some results obtained from the simulations. The vibratory model is first validated for the vertical flexural waves trough the use of an impact hammer. Then, the parametric optimization method is tested by the vertical excitation of the rail with a modal shaker. The main simulation results are found, and some particular behavior due to other waves existing in the rail can be observed, opening the perspective of a generalized method including more complex vibratory modelings.

Previous issue date: 2007-09-20 / People with mental retardation usually present motor development delays. Nevertheless, these delays seem to be more often related to cognitive factors such as attention and comprehension than to motor or mental deficits. Strategies have been developed to teach motor skills to this population. One of the existing strategies is mental training, which is based on imagining the movement to be executed without the presence of the physical movement. The general objective of this doctoral dissertation was to apply a procedure based on physical and mental training to the teaching of rolling forward, a basic skill for artistic gymnastics practitioners, and to verify its effects in mentally retarded students. In experiment 1 there were two subjects (P1 and P2) aged 15 and 20 respectively, enrolled in a school for the mentally retarded. A single-subject experimental design with alternating treatments was applied. The rolling forward skill was divided in four phases. The results showed that P1
learned all the phases of forward rolling, whereas P2 learned partially phases I and II of the
skill taught. We believe that the difficulty found by this participant was due to his motor
difficulties, once he also presented an associated physical handicap. The purpose of
experiment 2 was to apply the mental training procedure and verifying their effects in the
forward rolling learning process in three students aged 12, 12 and 16, respectively, all of them
with mental retardation. The experimental design applied was of the single subject in a multiple baseline design. The participants of this study learned all phases of forward rolling, and even though they presented different performance levels, all of them benefited from the strategy used. We realized that the mental training elicited other behaviors from the participants like: following rules, self-conversation, attention, concentration and relaxation. To understand the aspects that affect the use of mental training and to establish experimental designs to investigate this strategy were some of the contributions of this work. / Pessoas com deficiência mental apresentam atrasos no desenvolvimento motor. No entanto, esses atrasos parecem estar mais relacionados a fatores cognitivos de atenção e compreensão de instruções do que a déficits motores ou mentais. Estratégias têm sido utilizadas com o objetivo de auxiliar o ensino de habilidades motoras para essa população. Uma das estratégias existentes é o treino mental, ou seja, imaginar o movimento a ser executado sem a presença do movimento físico. Partindo da abordagem comportamental,
utilizando diferentes delineamentos experimentais, o objetivo geral deste trabalho foi de aplicar os procedimentos de treino físico e treino mental e verificar seus efeitos na
aprendizagem em alunos com deficiência mental. O experimento 1 teve como objetivo aplicar
os procedimentos de treino físico e mental e verificar os seus efeitos na aprendizagem do
rolamento para frente, elemento da Ginástica Artística, em alunos com deficiência mental.
Participaram desse estudo dois jovens (P1 e P2), com idade de 15 e 20 anos respectivamente,
estudantes de uma escola especializada. O delineamento experimental utilizado foi o de
sujeito único, com tratamentos alternados. Para fins de análise, a habilidade de rolamento para
a frente foi dividida em quatro fases. Os resultados encontrados mostraram que P1 aprendeu todas as fases e P2 aprendeu parcialmente as fases I e II da habilidade ensinada. Acreditamos que a dificuldade encontrada por esse participante foi decorrente de sua dificuldade motora, pois ele possuía também uma deficiência física associada. O experimento 2 teve como objetivo aplicar o procedimento de treino mental e verificar os seus efeitos na aprendizagem do rolamento para a frente, em três alunos, com idades de 12, 12 e 16 anos respectivamente, com deficiência mental. O delineamento experimental utilizado foi o de sujeito único de linha de base múltipla. Os participantes desse estudo aprenderam todas as fases do rolamento para frente e, apesar de apresentarem níveis de desempenho diferentes, os três se beneficiaram da estratégia utilizada. Percebemos que o treino mental evocou também outros comportamentos dos participantes como: seguir regras, autoconversação, atenção, concentração e relaxamento. Desenvolver um programa de ensino de habilidades motoras a jovens com deficiência mental e investigar, através de delineamentos experimentais específicos, o papel do treino mental no
desenvolvimento dessas habilidades foram algumas das contribuições desse trabalho.

FERRAMENTAS DE USINAGEM EM AÇO RÁPIDO AISI M2 OBTIDO POR CONFORMAÇÃO POR SPRAY. / MACHINING TOOLS IN AISI M2 HIGH-SPEED STEEL OBTAINED BY SPRAY FORMING PROCESS.Edilson Rosa Barbosa de Jesus 07 October 2004 (has links)
O objetivo do presente trabalho foi à obtenção de aço rápido tipo AISI M2 pela técnica da conformação por spray e a avaliação deste, quanto ao seu desempenho quando aplicado como ferramenta de usinagem. O material obtido foi laminado a quente com razões de redução de 50 e 72%, resultando em placas a partir das quais foram confeccionadas pastilhas intercambiáveis para realização de testes de usinagem. O desempenho das pastilhas obtidas a partir do material conformado por spray e laminado a quente, foi confrontado com o de pastilhas confeccionadas a partir de material processado convencionalmente (lingotamento convencional), e com material processado pela técnica da metalurgia do pó (MP). As variações do material obtido por conformação por spray, assim como também os demais materiais, foram caracterizados química, física, mecânica e microestruturalmente. Adicionalmente, foram realizados testes de usinagem para avaliação de desempenho do material quando submetido a condições reais de trabalho. Os resultados da caracterização dos materiais evidenciaram o potencial da técnica de conformação por spray, em possibilitar a obtenção de materiais com boas características e propriedades. Para as condições de processamento, conformação mecânica e de tratamentos térmicos aplicados neste trabalho, às análises dos resultados dos testes de usinagem revelaram um comportamento muito próximo entre todos os materiais de ferramenta utilizados. Procedendo a uma análise mais refinada dos resultados dos testes de usinagem contínua, foi verificado um desempenho ligeiramente superior para o material obtido pela técnica da metalurgia do pó (MP), seguido pelo material obtido pela técnica da metalurgia convencional, o qual apresentou uma vantagem mínima em relação ao obtido pela técnica da conformação por spray e laminado com redução de 72%. Por fim, o desempenho inferior ficou por conta do material obtido pela técnica da conformação por spray e laminado com redução de 50%, o qual apresentou os maiores valores de desgaste. / The aim of the present work was the obtention of AISI M2 high-speed steel by spray forming technique and the material evaluation when used as machining tool. The obtained material was hot rolled at 50% and 72% reduction ratios, and from which it was manufactured inserts for machining tests. The performance of inserts made of the spray formed material was compared to inserts obtained from conventional and powder metallurgy (MP) processed materials. The spray formed material was chemical, physical, mechanical and microstructural characterised. For further characterisation, the materials were submitted to machining tests for performance evaluation under real work condition. The results of material characterisation highlight the potential of the spray forming technique, in the obtention of materials with good characteristics and properties. Under the current processing, hot rolling and heat treatments condition, the analysis of the results of the machining tests revealed a very similar behaviour among the tested materials. Proceeding a criterious analysis of the machining results tests, it was verified that the performance presented by the powder metallurgy material (MP) was slight superior, followed by conventional obtained material (MConv), which presented a insignificant advantage over the spray formed and hot rolled (72% reduction ratio) material. The worst result was encountered for the spray forming and hot rolled (50% reduction ratio) material that presented the highest wear values.

Previous issue date: 2009-03-26 / Samonella spp are Gram negatives bactérias belonging to Enterobacteriaceae family. S. enterica comprise about 2.500 sorovars. These sorovars can infect a broad range, including poultry, cattle, swins and humans, and are agent causative of salmonellosis an important public health issue worldwide. Small multicopy plasmids are frequently isolated from Gram negatives and Gram positives bacterias. In Salmonella, low molecular weight plasmids are found on 10% of Salmonella strains and their biological functions are unknown. However, many plasmids in Salmonella control important properties, such as, virulence factors, heavy metals and antibiotics resistance, and utilizations of alternative carbon sources. The pVCM1 plasmid was extracted from one strain of Samonella enteretidis isolated from broilers carcass. The strains were grown in liquid or solid Luria-Bertani broth at 37 °C. The plasmid was purified, separated on 1% agarose electrophoresis and visualized by ethidium bromide staining for analysis. Plasmid was digested with EcoRI enzyme and subcloned in the pUC18 vector.The plasmidal stability was evaluated, inoculating E. coli cells transformated with pVCM1 plasmid (cloned in pUC18) in liquid Luria-bertani broth supplemented with ampicillin. The pVCM1 was stable after 240 generations. The total DNA sequence of plasmid pVCM1 has 1981 pb. Genbank search resulted that pVCM1 showed 99% of identity with pB and 92% with pJ, which were isolated from Salmonella enteretidis. Only one ORF founded in pVCM1 showed significative similarity with others proteins of GenBank. The protein encoded by this ORF showed homology to Rep proteins of plasmids that replicates by rolling-circle mechanism. The pVCM1 posses three impotents elements: rep gene, single strand origin (SSO) and inverted repeat sequences. Such elements are importants for the rolling circle replication, suggesting that pVCM1 use this mechanism. The rep gene was amplified and cloned in the pGEMT-easy vector, but the heterologous expression of Rep protein wasn t gotten successfully. / Salmonellas ssp são bactérias Gram negativas pertencente a família das enterobácterias. A S. entérica compreende cerca de 2.500 sorovares. Esses sorovares podem infectar vários hospedeiros incluindo aves, bovinos, suínos e humanos, e são os agentes causais das salmoneloses, um importante problema de saúde pública em todo mundo. Múltiplas cópias de pequenos plasmídeos são frequentemente isolados de bactérias Gram negativas e Gram positivas. Nas salmonelas, plasmídeos com baixa massa molecular são encontradas em 10% das linhagens e suas funções biológicas são desconhecidas. Entretanto, muitos plasmídeos de salmonela controlam propriedades importantes, tais como, fatores de virulência, resistência a antibióticos e metais pesados, e utilização de fontes alternativas de carbono. O plasmídeo pVCM1 foi extraído de uma linhagem S. Enteritidis, isoladas de carcaças de frango. Essa linhagem foi crescida em meio Lúria-Bertani líquido e sólido à 37°C. O plasmídeo foi purificado, separado em gel de agarose 1% e visualizado com coloração por brometo de etídio. O plasmídeo foi digerido com EcoRI e subclonado no vetor pUC18. A estabilidade plasmidial foi avaliada inoculando células de E. coli transformadas com plasmídeo pVCM1 (clonado em pUC18) em meio Lúria- bertani líquido suplementado com ampicilina. O pVCM1 foi estável por mais de 240 gerações A sequência nucleotídica total do plasmídeo possui 1981 pb. Pequisa por sequências no GenBank, mostrou que o pVCM1 apresenta similaridade de 99% com o plasmídeo pB e 92% com plasmídeo pJ, os quais foram isolados de S. Enteritidis.Das 11 possíveis ORFS apenas uma única ORF apresentou similaridade significativa com outras proteínas do GenBank. A proteína codificada por essa ORF apresentou homologia com as proteínas Rep de plasmídeos que replicam via círculo rolante. O pVCM1 apresenta três regiões importantes: gene rep, origem de fita simples (SSO) e regiões repetidas invertidas. Tais regiões são importantes para o mecanismo de replicação via círculo rolante, sugerindo que o pVCM1 utilize esse mecanismo. O gene rep foi amplificado e clonado no vetor pGEM T-easy, mas a expressão heteróloga da proteína Rep não foi obtida com sucesso.

Estudo do comportamento do coeficiente de desgaste e dos modos de desgaste abrasivo em ensaios de desgaste micro-abrasivo. / Wear coefficient and wear mode transition study in micro-abrasive wear testing.Ronaldo Câmara Cozza 24 February 2006 (has links)
Esta Dissertação tem por objetivo estudar o comportamento de diferentes materiais sob a ação de desgaste micro-abrasivo. Como parte do trabalho, foi projetada e construída uma máquina de ensaio desgaste por micro-abrasão por esfera rotativa fixa, com configuração mecânica com diferenças em relação às observadas na literatura (Gee et al., 2005). Como corpos-de-prova, foram utilizadas pastilhas intercambiáveis de metal duro (classe P20) e aço ferramenta M2. As esferas foram de aço AISI 1010 cementado e aço AISI 52100 temperado e revenido. Durante os ensaios, foi inserida entre a esfera e o corpo-de-prova uma pasta abrasiva preparada com carbeto de silício preto, com tamanho médio de partícula de 5 µm. Inicialmente, foram realizados ensaios preliminares, com a finalidade de analisar não só o comportamento do equipamento, mas também estudar a transição entre os modos de desgaste que podem ocorrer durante o desgaste micro-abrasivo. A transição entre os modos de desgaste foi estudada em função da carga normal e dos materiais utilizados durante o ensaio. Os resultados obtidos indicaram boa reprodutibilidade do equipamento e coerência com resultados da literatura. Em seguida, em ensaios denominados definitivos, foram pesquisadas as atuações dos modos de desgaste abrasivo e a obtenção do regime permanente de desgaste. Os resultados mostraram que, com a variação da distância de deslizamento, houve alterações nas ocorrências dos modos de desgaste abrasivo. Por outro lado, em alguns ensaios, o coeficiente de desgaste tendeu a permanecer constante, o que caracteriza a obtenção do regime permanente de desgaste. A micro-abrasive wear testing machine with fixed sphere was designed and constructed, presenting a mechanical configuration with differences with respect to those found in the literature (Gee et al., 2005). M2 tool steel and WC-Co P20 were used as testing specimen materials. Ball materials were cemented AISI 1010 steel and quenched and tempered AISI 52100 steel. During the tests, an abrasive slurry, prepared with black silicon carbide (SiC) particles (average particle size of 5 µm), was supplied to the contact between the specimen and the ball. Initially, preliminary tests were conducted to study the wear mode transitions that can occur during the micro-abrasive wear and to analyze the operational conditions of the equipment. The wear mode transitions were evaluated as a function of the applied normal load and of the materials used. The results indicated good reproducibility and qualitative agreement with those found in the litarature. Later, a new set of tests was conducted, which analyzed the evolution of the abrasive wear modes and the achievement of steady state wear as a function of sliding distance. The results indicated a continuous variation in the abrasive wear modes with sliding distance. Additionally, in some tests, the wear coefficient tended to stabilize in constant value, which characterizes the achievement of steady state regime. However, in other tests, the wear coefficient presented a non constant evolution of wear coefficient with the sliding distance, which denotes that the constant regime of wear was not obtained.

Efeitos da ventilação mecânica e pressão positiva no final da expiração sobre a microcirculação mesentérica em ratos Wistar / Effects of mechanical ventilation and positive end-expiratory pressure on mesenteric microcirculation in Wistar ratsPriscila Aikawa 03 September 2009 (has links)
Ventilação mecânica (MV) com pressão positiva no final da expiração (PEEP) melhora a oxigenação sanguínea e oferta de oxigênio aos tecidos no tratamento da insuficiência respiratória aguda. No entanto, a pressão intratorácica elevada pode alterar o fluxo sanguíneo no mesentério que pode contribuir para complicações gastrointestinais durante a VM. Investigamos os efeitos da PEEP sobre as interações leucócito-endotélio durante a VM em ratos com pulmões normais e sem administração de fluido (Fase I) e os efeitos do volume corrente baixo (LTV) e pentoxifilina (PTX) sobre a microcirculação mesentérica (Fase II). O protocolo e resultados da Fase I são os seguintes: 44 ratos Wistar machos (~240g) foram anestesiados com pentobarbital (I.P., 50mg.kg-1) e com isoflurane inalatório (1.5-2%) após instrumentação, e aleatoriamente divididos em (1) INTACTO (somente anestesia), (2) PEEP0 (PEEP=0 cmH2O), (3) PEEP5 (PEEP=5 cmH2O), e (4) PEEP10 (PEEP=10 cmH2O). Os grupos PEEP foram submetidos à traqueostomia e VM com volume corrente de 10 ml.kg-1, frequência respiratória de 70 rpm e fração inspirada de oxigênio de 1. Após 2-h de VM, realizamos laparotomia mediana e avaliamos as interações leucócito-endotélio por meio de microscopia intravital e inflamação pumonar por meios histológicos. Não observamos alterações significantes na pressão sanguínea arterial média (PAM) entre os grupos ao longo do estudo. A pressão traqueal do grupo PEEP5 foi menor comparada com os grupos PEEP0 e PEEP10 (11, 15, e 16 cmH2O, respectivamente; p<0.05). Após 2-h de VM, não houve diferenças significantes entre os grupos INTACTO, PEEP0 e PEEP5 no número de leucócitos rollers (118±9, 127±14 e 147±26 células/10minutos, respectivamente), aderidos (3±1, 3±1 e 4±2 células/100m de comprimento de vênula, respectivamente), e migrados (2±1, 2±1 e 2±1 células/5,000m2, respectivamente) no mesentério. No entanto, PEEP10 aumentaram (p<0.05) o número de leucócitos rollers (188±15 células/10minutos), aderidos (8±1 células/100m de vênula) e migrados (12±1 células/5,000 m2). Observamos inflamação pulmonar nos grupos PEEP0 e PEEP10. O protocolo e resultados da Fase II são os seguintes: 57 ratos Wistar machos (~253g) foram anestesiados com pentobarbital (I.P., 50 mg.kg-1), submetidos a traqueostomia, anestesia inalatória com isoflurane (1.5-2%), VM com PEEP de 10 cmH2O, fração inspirada de oxigênio de 0,21, e aleatoriamente divididos em (1) LTV (7 ml.kg-1), (2) volume corrente elevado (HTV, 10 ml.kg-1), e (3) PTX (HTV+ PTX, 25 mg.kg-1). Nós registramos a PAM, mecânica respiratória e gases sanguíneos arteriais no basal e após 2-h de VM. Realizamos laparotomia mediana e avaliamos as interações leucócito-endotélio no mesentério, fluxo de artéria mesentérica (FAM), mecânica respiratória e inflamação pulmonar. Não observamos diferenças entre os grupos no basal e após 2-h em PAM (113±15 vs 109± 6 mmHg). Após 2-h de VM, o FAM foi similar em todos os grupos (12.4±2.6 ml.min-1). A pressão traqueal foi menor no grupo LTV (11.2±1.6 cmH2O) comparada com HTV (14.7±1.1 cmH2O) e PTX (14.1±2.4 cmH2O). Em todos os grupos a VM aumentou a elastância pulmonar (~22%, p<0.05) e diminuiu a resistência de vias aéreas (~10%, p<0.05). LTV e PTX apresentaram valores similares de leucócitos aderidos (5±2 e 6±4 células/100m de vênula, respectivamente), e migrados (1±1 e 2±1 células/5,000m2, respectivamente). Contrariamente, HTV aumentou o número de aderidos (14±4 leucócitos/100m de vênula, p<0.05) e migrados (9±3 células/5,000m2, p<0.05) no mesentério. O grupo HTV apresentou infiltrado neutrofílico e edema pulmonar. Em conclusão, nosso estudo mostrou que a pressão intratorácica elevada é prejudicial para a microcirculação mesentérica e pulmões no modelo experimental de ratos com pulmões normais e pressão sanguínea sistêmica estável, LTV previne alterações microcirculatórias e pulmonares, e a administração precoce de PTX atenua as interações leucócito-endotélio no mesentério e inflamação pulmonar durante a VM. Esses achados podem ter relevância na compreensão das complicações induzidas pela VM e prognóstico. / Mechanical ventilation (MV) with positive end expiratory pressure (PEEP) improves blood oxygenation and tissue oxygen delivery during treatment of acute respiratory failure. However, high intrathoracic pressure may alter blood flow at mesentery, which may contribute to gastrointestinal complications during MV. We investigated the effects of PEEP on mesenteric leukocyte-endothelial interactions during MV in rats with normal lungs and without fluid administration (Phase I) and the effects of low-tidal volume (LTV) and pentoxifylline (PTX) on mesenteric microcirculation (Phase II). The protocol and results of Phase I are the following: 44 male Wistar rats (~240g) were anesthetized with pentobarbital (I.P., 50mg.kg-1) and inhaled isoflurane (1.5-2%) after instrumentation, and randomly divided in (1)NAIVE (only anesthesia), (2) PEEP0 (PEEP=0 cm H2O), (3) PEEP5 (PEEP=5 cmH2O), and (4) PEEP10 (PEEP=10 cmH2O). PEEP groups were submitted to tracheostomy and MV with tidal volume of 10 ml.kg-1, respiratory rate of 70 rpm and inspired oxygen fraction of 1. After 2-hrs of MV, we performed a median laparotomy and evaluated leukocyte-endothelial interactions at the mesentery and lung inflammation by histology. We did not observe significant changes mean arterial blood pressure (MABP) among groups throughout the study. HTV presented lung neutrophil infiltration and edema. In conclusion, our study showed that high intrathoracic pressure is harmful to mesenteric microcirculation and lungs in the experimental model of rats with normal lungs and stable systemic blood pressure, LTV prevents microcirculatory and pulmonary alterations, and early administration of PTX attenuates leukocyte-endothelial interactions at the mesentery and lung inflammation during MV. These findings may have relevance for complications MV-induced and outcome.

Caractérisation et modélisation des propriétés à la fatigue à grand nombre de cycles des aciers cémentés à partir d'essais d'auto-échauffement sous sollicitations cycliques / Characterization and model of high cycle fatigue of carburizing steel with self-heating measurement under cyclic loadGraux, Nicolas 24 November 2017 (has links)
Le dimensionnement en fatigue à grand nombre de cycles d'un contact roulant entre des éléments ayant subi un traitement thermochimique de cémentation s'avère rapidement complexe.D'une part le traitement de cémentation apporte une hétérogénéité de propriété dans les couches supérieures de la pièce qui dépend du protocole utilisé. D'autre part le chargement de contact roulant est un chargement complexe dont le mode de défaillance en fatigue s'initie en sous-couche.Afin de limiter le temps de la caractérisation des champs de propriétés en fatigue, l'utilisation des mesures d'auto-échauffement sous sollicitation cyclique ainsi que leur interprétation par un modèle probabiliste à deux échelles est proposé. Néanmoins de par l'hétérogénéité du matériau et de par la particularité du chargement il peut s'avérer délicat d'appliquer une telle méthode d'évaluation. ll est alors proposé d'explorer ces deux difficultés de manière séparé.Pour prendre en compte l'hétérogénéité matériaux, un protocole d'analyse de courbe d'auto-échauffement basé sur une variante d'un modèle probabiliste à deux échelles et sur les mesures de taux de carbone a été proposé. Les paramètres du modèle ont été identifiés sur une classe d'acier via des mesures d'auto-échauffement réalisées sur des éprouvettes représentatives de l'hétérogénéité du au traitement de cémentation. Enfin le modèle a été validé par comparaison avec des points de fatigue expérimentaux.En ce qui concerne le chargement de contact roulant, les difficultés pour réaliser une mesure d'auto-échauffement ont mené à effectuer une première campagne de mesure sur le cas intermédiaire du contact répété. A l'aide d'un modèle analytique simple, l'évolution du champ de température a pu être reliée à un terme source de chaleur moyen dont le lien avec les mécanismes de fatigue reste à démontrer. Finalement, des prototypes de machine de contact roulant dédiés aux mesures d'auto-échauffement ont été proposés. Les mesures réalisées sur ces dernières et leur interprétation laissent à penser qu'il sera possible d'identifier des propriétés de fatigue à partir de mesure d'auto-échauffement. / The rolling contact fatigue prediction between two carburizing part quickly becomes complex.On one hand, the carburizing treatment give heterogeneous properties in surface layer depending on the treatment protocol. On the other hand, the rolling contact load is a complex load with a fatigue initiation in the sub-layer. To limit the duration of the field fatigue properties characterization, self-heating measurements under cycle load are used and their interpretation by a probabilistic two scales model is proposed. Nevertheless applying this fatigue evaluation method on heterogeneous material and for rolling contact load can be difficult. ln first approach those difficulties are split.To take into account the material heterogeneity, an analysis based on a variation of one probabilistic two scales model and on carbon rate measurement is proposed. Model parameters are identified on one steel class with self-heating measurement made on specimens representative of carburizing material heterogeneity. Finally the model is validated by comparison with experimental fatigue point.Making self-heating measurement for rolling contact load is complex. Consequently a first self-heating measurement campaign is made on the intermediary case of repeated contact. With a simple analytic model, the temperature field evolution can be linked to a mean heat source whose link with fatigue mechanism must be proven. Finally, rolling contact machine prototypes are proposed. Self-heating measurement made on those prototypes and their interpretation suggest that it will be possible to identify fatigue properties with self-heating measurement.

Simulation multi-échelles de la propagation des fissures de fatigue dans les rails / Multi-scale fatigue crack propagation simulation in railsTrollé, Benoit 20 March 2014 (has links)
Sous l’effet des sollicitations mécaniques répétées induites par les passages des trains, on observe l’apparition de fissures de fatigue de contact dans les rails. Une fois amorcées, celles-ci peuvent se propager et mener à la rupture du rail. Dans un contexte d’intensification du trafic et d’augmentation globale des vitesses de circulation, il devient stratégique pour SNCF d’optimiser sa politique de maintenance. Afin de définir des pas de surveillance adaptés et une planification optimisée des opérations de maintenance, une meilleure connaissance des mécanismes d’endommagement par fatigue du rail s’avère nécessaire. Tendre vers cette stratégie de maintenance prédictive passe par la mise en place d’outils de simulation numérique adaptés. Dans ce contexte, une chaîne d’outils a été développée : détermination des sollicitations transmises au rail, des champs de contraintes et de déformations résiduelles, localisation des zones critiques vis-à-vis du risque de fissuration. L’étape suivante consiste à estimer le risque lié à la présence de fissures et à étudier leurs propagations. Elle constitue une partie des objectifs de ces travaux de thèse. La résolution du problème tri-dimensionnel d’une structure fissurée, avec contact et frottement entre les lèvres, est effectuée grâce à un modèle tri-dimensionnel éléments finis étendus multi-échelles. Ce modèle fait appel à une formulation mixte stabilisée où chaque champ est écrit à l’aide d’enrichissement. La fissure est représentée grâce à une stratégie implicite-explicite. Le problème est résolu à l’aide du solveur non-linéaire LATIN. Une étude empirico-numérique a permis de proposer des formules a priori assurant à la méthode de résolution un taux de convergence proche de l’optimal. La simulation de la propagation des fissures de fatigue est réalisée à l’aide de critères spécifiques, adaptés à un chargement multi-axial et non-proportionnel, et d’une loi de propagation dédiée en mode mixte. La confrontation des résultats de simulation avec des essais réalisés sur une configuration cylindre-plan a validé la stratégie X-FEM/LATIN à deux échelles. Tous ces développements ont été implémentés dans le code de calcul éléments finis CAST3M. Des contraintes résiduelles réalistes, provenant d’un logiciel externe, ont été introduites. Cette étape a requis la mise en place d’une procédure de transfert des champs entre les deux maillages (celui utilisé pour le calcul des contraintes résiduelles et celui utilisé pour la résolution du problème de mécanique élastique linéaire de la rupture). L’étude de la flexion du rail a révelé l’influence de ce phénomène uniquement lors du passage du chargement sur la fissure.Enfin, une étude numérique a montré la très forte influence de l’orientation du chargement tangentiel, des contraintes résiduelles et de la présence de plusieurs fissures sur la direction et les vitesses de propagation des fissures de fatigue. / To optimize the rail grinding strategy, the prediction of crack growth rates has a vital role. Contact, with friction between the crack faces, notably occurs in rolling contact fatigue (RCF) problems. These time-dependent, multi-axial, non proportional loadings may lead to a crack initiation and propagation, and sometimes to the development of very complex 3D crack network. Numerical simulations of frictional fatigue crack are efficiently performed using the eXtended Finite Element Method (X-FEM). Within this method, the mesh does not need to conform to the crack geometry. Most difficulties associated to complex mesh generation around the crack and the re-meshing steps during the propagation are hence avoided. A 3D two-scale frictional contact fa-tigue crack model developed within the X-FEM framework is presented. It allows the use of a refined discretization of the crack interface independent from the underlying finite element mesh and adapted to the frictional contact crack scale. The model is used here to analyze the crack propagation, rate and direction, under rolling contact fatigue. The wheel-rail contact loading is modeled as a traveling hertzian load. The stress intensity factors are computed at the crack tips during the wheel passage. Criteria for determining crack growth direction under multiaxial non proportional conditions and mixed mode Paris’ law are used. Actual residual stresses are accounted for in the simulation. They are determined thanks to a dedicated model used at SNCF in which the asymptotic mechanical state of the rail is computed when submitted to cyclic loads. A non-uniform elastic-plastic stabilized state is calculated and introduced, by projection of the mechanical fields onto the finite element mesh, in the crack propagation simulation. All this strategy has been implemented in CAST3M and is now used to model 3D frictional crack growth under RCF.
