The Effects of Glyphosate-based Herbicides on the Development of Wood Frogs, Lithobates sylvaticus

Amphibians develop in aquatic environments where they are very susceptible to the effects of pesticides and other environmental contaminants. Glyphosate-based herbicides are widely used and have been shown to affect survival and development of tadpoles under laboratory conditions. The goal my thesis is to determine if agriculturally relevant exposure to Roundup WeatherMax®, a herbicide formulation containing the potassium salt of glyphosate and an undisclosed surfactant, influences the survival and development of wood frogs tadpoles (Lithobates sylvaticus) under both laboratory and field conditions. In the field, experimental wetlands were divided in half using an impermeable curtain so that each wetland contained a treatment and control side. Tadpoles were exposed to two pulses of this herbicide at environmentally realistic concentration (ERC, 0.21 mg acid equivalent (a.e.)/L) and predicted environmental concentrations (PEC, 2.89 mg a.e./L), after which survival, growth, development, and expression of genes involved in metamorphosis were measured. Results indicate that exposure to the PEC is extremely toxic to tadpoles under laboratory conditions but not under field conditions. Results from both experimental conditions show sublethal effects on growth and development, and demonstrate that ERC of glyphosate-based herbicides have the potential to alter hormonal responses during metamorphosis. My secondary objectives were to compare the effects of Roundup WeatherMax® to the well-studied Vision® formulation (containing the isopropylamine (IPA) salt of glyphosate and POEA), and to determine which ingredient(s) are responsible for the sublethal effects on development. Survival, growth and gene expression results indicate that Roundup WeatherMax® has greater toxicity than Vision® formulation. Contrary to my prediction, results suggest that, under realistic exposure scenarios, POEA is not the sole ingredient responsible for the observed developmental effects. However, my results demonstrate that chronic exposure to the POEA surfactant at the PEC (1.43 mg/L) is extremely toxic to wood frog tadpoles in laboratory. As part of the Long-term Experimental Wetlands Area (LEWA) project, this research contributes to overall knowledge of the impacts of glyphosate-based herbicides on aquatic communities.

Value-laden risk assessment and biotechnology regulation in Canada

<p>Canadas regulatory system is science-based and relies on risk assessment to inform decisions about which products of biotechnology (and other technologies) are safe enough for commercial application. Since regulation involves the loss of certain liberties, it is imperative that any regulatory regime be as objective as possible. Scientific risk assessment seems to be a good way to produce the information, which guides policy makers since it involves quantitative analysis and the production of seemingly objective data.</p><p>The view adopted by regulators and in current risk assessment practices is that objective means value-free. Therefore, because risk assessment data is scientific it is thought to be value-free but this is not the case. Risk assessment necessarily involves value assumptions. Assumptions must be made at all stages of the production of risk data. This does not mean, however, that risk assessment is hopelessly subjective. The notion of value-free objectivity can be replaced with the view that genuine objectivity arises through peer review and social discourse. Regulators can adopt this understanding of objectivity to acknowledge the value-ladenness of risk assessment data.</p><p>At present, the value assumptions made by industry, government and private scientists during risk assessment go largely unnoticed yet have an effect on the outcome of regulatory decisions. Such assumptions must be recognized in order to ensure that the decisions made about the risks society face are not biased. This is particularly true in the case of biotechnology regulation. The development of the science of biotechnology has occurred concurrently with the development of the biotech industry creating the opportunity for industry-biased risk assessments.</p><p>It is possible to make changes to the existing regulatory regime in Canada in order to avoid some of the major problems associated with unrecognized value assumptions in risk assessment. A complete restructuring of the regime is unnecessary, however. Maintaining the current regulatory structure with some minor changes could address these problems. These changes include: creating an independent review board, making explicit that value assumptions are part of risk assessment in government advisory reports, and enhancing the role of regulators. Canadas regulatory system can better address the risks associated with biotechnology if it acknowledges that risk assessment is value-laden.</p>

Effekter av en toxisk cocktail av Roundup och livsmedelsklassad polypropylenplast på vattenloppan Daphnia Magna

Gifter och föroreningar förekommer sällan ensamma och studier om olika toxiska substanser genomförs ofta med endast en substans i taget. Det finns studier som har undersökt exempel på olika cocktails av substanser, men ibland är det svårt att identifiera alla källor som påverkar en organism. Vi har undersökt hur en kombination av ogräsmedlet Roundup och livsmedelsklassad polypropylenplast påverkar det sötvattenlevande kräftdjuret Daphnia magna. Vi fann att i högre doser orsakar Roundup 100% dödlighet innan könsmognad, men att det i små doser inte resulterar i lägre fitness än kontrollgruppen, utan till och med ökade den initiala överlevnaden när den tillsattes i plastbrukar. En cocktail av Roundup och plast orsakade en fördubblad utvecklingstid för äggen. Ägg från individer i plastburkar hade också en längre utvecklingstid men dessa individer hade även ett mycket högre antal ägg. Framtida studier bör undersöka avkommans fitness. Tidigt i studien upptäckte vi att det med stor sannolikhet fanns en tredje faktor som påverkade daphnierna negativt, som inte kunde förklaras av Roundup eller plastburkar, men som vi inte kunde identifiera. Därför vill vi också framhäva att experiment lätt kan påverkas av okända faktorer. / Toxins and pollutants rarely come alone and studies about various toxic substances are often performed with only one substance at a time. There are studies that have examined different cocktails of substances, but sometimes it can be difficult to identify all sources that affect an organism. We have investigated how a combination of the herbicide Roundup and Food Safe polypropylene plastic affect the freshwater crustacean Daphnia magna. We found that Roundup causes 100% mortality before reaching fertility in higher doses, but that in small doses does not result in lower fitness compared to the control group, but even increases the initial survival rate when Roundup is added to plastic jars. A cocktail of Roundup and plastic resulted in a doubled development time for the eggs. Eggs from individuals in plastic jars also had a longer development time but there was also a much higher number of eggs from these individuals. Futiure studies ought to investigate the fitness of the offspring. Early in the study we discovered that there was most likely a third factor influencing the daphnia negatively, that could not be explained by plastic jars or Roundup, but one that we could not identify. Therefore, we also wish to urge that experiments can easily be influenced by unknown factors.

Toxicological Impact of Agricultural Surfactants on Australian Frogs

Surfactants are one of the more ubiquitous contaminants in aquatic systems. Their importance as toxic components of pesticide formulations has, however, been largely overlooked. Amphibians particularly, as inhabitants of shallow, temporary and often lentic aquatic environments may be at risk from exposure to these chemicals when they enter aquatic systems. This thesis presents data on the toxicity of surfactants to amphibians. Several experimental exposures were conducted with embryo-larval, tadpole and adult developmental stages of the Australian species- Crinia insignifera, Helcioporus eyrei, Limnodynastes dorsalis and Litoria moorei and the exotic species- Bufo marinus and Xenopus laevis. Animals were variously exposed to glyphosate formulations that contain a high proportion of nonionic surfactants, or commercial pesticide wetting agents (alcohol alkoxylate and nonylphenol ethoxylate (NPE) surfactants). Feeding stage tadpoles of C.insignifera, H. eyrei, L. dorsalis and L. moorei were exposed to three commercial glyphosate formulations, glyphosate isopropylamine and glyphosate acid in static-renewal acute toxicity tests. The 48-h LC50 values for Roundup Herbicide (MON 2139) tested against tadpoles of C. insignifera, H. eyrei, L. dorsalis and L. moorei ranged between 8.1 and 32.2 mg/L (2.9 and 11.6 mg/L glyphosate acid equivalent (ae)), while the 48-h LC50 values for Roundup Herbicide tested against adult and newly metamorphosed C. insignifera ranged from 137-144 mg/L (49.4-51.8 mg/L ae). / Touchdown Herbicide (4 LC-E) tested against tadpoles of C. insignifera, H. eyrei, L. dorsalis and L. moorei was slightly less toxic than Roundup with 48-h LC50 values ranging between 27.3 and 48.7 mg/L (9.0 and 16.1 mg/L ae). Roundup Biactive (MON 77920) was practically non-toxic to tadpoles of the same four species producing 48-h LC50 values of 911 mg/L (328 mg/L ae) for L. moorei and >1000 mg/L (>360 mg/L ae) for C. insignifera, H. eyrei and L. dorsalis. Glyphosate isopropylamine was practically non-toxic producing no mortality amongst tadpoles of any of the four species over 48 h, at concentrations between 503 and 684 mg/L (343 and 466 mg/L ae). The toxicity of technical grade glyphosate acid (48-h LC50, 81.2-121 mg/L) is likely to be due to acid intolerance. Feeding stage tadpoles of B. marinus, X laevis, C. insignifera, H.eyrei, L. dorsalis and L. moorei were exposed to NPE and alcohol alkoxylate in static renewal acute toxicity tests. All species exhibited non-specific narcosis following exposure to both these surfactants. The 48-h EC50 values for NPE ranged between 1.1 mg/L (mild narcosis) and 12.1 mg/L (full narcosis). The 48-h EC50 values for alcohol alkoxylate ranged between 5.3 mg/L (mild narcosis) and 25.4 mg/L (full narcosis). Xenopus laevis was the most sensitive species tested. The sensitivity of the other five species was size dependent with larger species displaying greater tolerance. Replicate acute toxicity tests with B. marinus exposed to NPE at 30 degrees celsius over 96 hours indicated that the narcotic effects were not particularly time dependant. / The mean 24, 48, 72 and 96-h EC50 (mild narcosis) were 3.6, 3.7, 3.5 and 3.5 mg/L respectively. The mean 24, 48, 72, and 96-h EC50 (full narcosis) values were 4.0, 4.1, 4.2 and 4.0 respectively. Acute toxicity tests with B. marinus exposed to NPE at 30 degrees celsius under conditions of low dissolved oxygen (0.8-2.3 mg/L) produced a two to threefold increase in toxicity. The 12-h EC50 values ranged from 1.4 to 2.2 mg/L. The embryotoxicity of NPE was determined in X. laevis, L. adelaidensis and C. insignifera using a Frog Embryo Teratogenesis assay-Xenopus (FETAX). The 96-h LC50, EC50 and MCIG (LOEC) values for X. laevis were 3.9 to 5.4 mg/L, 2.8 to 4.6 mg/L and 1.0 to 3.0 mg/L respectively. The 140-h LC50, ECSO and MCIG values for L. adelaidensis were 9.2 mg/L, 8.8 mg/L and 5.1 to 6.0 ing/L respectively. The 134-h LC50, EC50 and MCIG values for C. insignifera were 6.4 mg/L, 4.5 mg/L and 4.0 mg/L respectively. Teratogenicity indices for the three species ranged between 1.0 and 1.6 indicating either no or low teratogenicity. Xenopus laevis was the more sensitive of the three species and the only species that displayed indisputable terata. The acute toxicity data indicated that the amphibian species tested were of similar sensitivity to fish and some invertebrates. Developmental retardation and oestrogenic effects following exposure to nonylphenol ethoxylate were indicated by sublethal toxicity tests. Crinia insignifera embryos were exposed during early embryogenesis to sublethal concentrations of NPE. / Exposure to NPE did not affect either weight nor size (snout-vent length) at metamorphosis. Exposure to 5.0mg/L NPE resulted in a significant delay in the time required to reach metamorphosis. Also, exposure to 3.0 mg/L NPE for the first 6 days of embryonic development or exposure to 5.0 mg/L NPE from day 2 to day 6 resulted in a statistically significant predominance in the female phenotype amongst metamorphosing froglets. Exposure for the first five days to 1.5 ing/L or 3.0 mg/L NPE had no effect on sex ratio. The results indicated that exposure to NPEs has endocrine disruptive effects in this species and that a narrow window of susceptibility exists for the induction of predominantly female phenotype. This study has also followed the degradation of a mixture of NPE oligomers and the concomitant formation of individual oligomers in static die-away tests with and without illumination in freshwater. Over 33 days in darkness there was a progressive and complete loss of long chain oligomers (NPEO(subscript)8-17), transient increases and subsequent loss of short to medium chain oligomers (NPE0(subscript)4-7), and large persistent increases (approximately 1000%) in short chain oligomers (NPE0(subscript)1-3). In the presence of illumination, biodegradation was retarded and heterotrophic bacterial proliferation was inhibited. After 33 days there was complete loss of long chain oligomers (NPE0(subscript)9-17), incomplete loss of medium chain oligomers (NPE0(subscript)6.8) and increases in short chain oligomers (NPE0(subscript)1-5). / This thesis discusses the importance of persistent metabolites of NPE degradation as it pertains to the habitat, developmental time frame and ecology of amphibians. Degradation of NPE is likely to occur over a time frame that is longer than that required for complete embryogenesis and metamorphosis of many species of amphibians, and may easily encompass those critical stages of development during which oestrogenic metabolites can affect development.

Evaluation of Various Herbicides for Saw Greenbrier [Smilax bona-nox L.] and Southern Dewberry [Rubus trivialis Michx.] Control and Bermudagrass [Cynodon dactylon (L.) Pers.] Tolerance and Sharppod Morningglory [Ipomoea trichocarpa var. trichocarpa Ell.] Control in Roundup Ready Flex® and LibertyLink® Cotton Systems

Field studies were conducted during 2006 and 2007 to evaluate control of saw greenbriar and southern dewberry by various pasture herbicides and to assess forage tolerance of Tifton 85 bermudagrass to these herbicides. Herbicides evaluated in each study included triclopyr, picloram, 2,4-D, fluroxypyr, dicamba, aminopyralid, metsulfuron methyl and various combinations of the above. Visual ratings were taken on each herbicide efficacy experiment. Visual evaluations of phytotoxicity, measurements of dry matter yield, and forage quality were quantified for each of the bermudagrass tolerance trials. Saw greenbriar was best controlled at approximately one year after treatment by triclopyr at 10.9% ae v/v with diesel as the carrier (88-98%), although the lower rate of triclopyr + diesel at 0.87% ae v/v + 5% v/v and triclopyr alone at 0.87% ae v/v provided 49 to 86% control. Triclopyr + fluroxypyr at 0.25% ai v/v + 0.086% ai v/v gave best control of southern dewberry in both years when applied as an individual plant treatment (IPT) six weeks after shredding. In general, shredding 45 days prior to herbicide application gave an advantage to southern dewberry control versus not shredding. In 2006, triclopyr + fluroxypyr (IPT) was the only treatment to decrease Tifton 85 dry matter yield at the first harvest, with no effect observed at the second harvest. In 2007, both broadcast treatments containing triclopyr + fluroxypyr and the IPT treatment of triclopyr decreased dry matter yield at the first harvest, with triclopyr (IPT) being the only treatment to lower dry matter yield at the second harvest. Field studies were also conducted in 2006 and 2007 to assess sharppod morningglory control in Roundup Ready Flex® and LibertyLink® cotton systems. Herbicides evaluated included glyphosate, glufosinate, prometryn, fluometuron, and diuron. Visual ratings of percent weed control and sharppod morningglory plant counts were taken to assess control. Prometryn at 1.8 kg ai ha⁻¹ and fluometuron at 1.8 kg ai ha⁻¹ provided significant preemergence control (33-81%) of seedling sharppod morningglory. All rates of glyphosate (1.06 and 1.54 kg ai ha⁻¹) and glufosinate (0.45 and 0.6 kg ai ha⁻¹) controlled sharppod morningglory from 55 to 100% at both application timings. The addition of diuron at 1.12 kg ai ha⁻¹ to glyphosate and glufosinate at the late season application enhanced sharppod morningglory control by 3 to 16%. Additionally, in both years, no reduction in cotton yield was observed in the morningglory infested treatment when compared to the weed free treatment.

面對未來全球人口成長、可耕地減少等現象,生物技術在農業上的應用日益增加。其中,基因改造技術等基因層次相關的平臺應用技術更是提升農作物價值的關鍵—透過跨物種功能應用,大幅提升育種效率。自1996年基改作物商業化種植開始至今,美國都是全球最大種植國家,也是基改作物研發先驅國家。生技農業政策以及和基因改造作物有關法規之制訂使基改作物在美國蓬勃發展,尤其是專利保護對於種子產業之影響尤為深遠。專利權所提供的完善保護使大量資本進入種子產業,投入資源將農業生物技術應用於種子培育上,此舉也促成Monsanto等跨國農業生技公司之興起,主導全球基因改造作物之市場。 Monsanto將研發重心放在基改種子之研發,其在生技種子相關營業比重遠高於同業,同時,其投入特殊性狀之基改種子研發之回收遠高於其他公司之相同營業項目之投資報酬率。Monsanto積極藉由併購取得基因、基因轉殖技術以及種子種源。掌握關鍵基因、基因轉殖技術以及大量且優良種源使含有Monsanto轉殖基因作物在市場上佔有極大的比例。在美國,超過一半以上之主要作物種植面積為基因改造作物,尤其是基因改造大豆,佔大豆總種植面積之比例高達94%。市面上絕大多數基改大豆係Monsanto的抗嘉磷塞(RounupReady,RR1Y)基改大豆種子。藉由智慧財產保護策略,Monsanto並進一步限制RR1Y及其他基改作物之使用方式。 惟RR1Y專利將在2014年到期,農民可在2014年時留種種植基改大豆種子而不用每年購買種子,或購買學名(generic)抗嘉磷塞轉基因大豆種子。在美國所種植之大豆約有四成會外銷,而外銷國家基改作物規範法規是出口與否之關鍵。若未取得歐盟、中國等主要外銷國家基改作物主管機關之批准,呈交包括基因之轉殖植物對環境衝擊之風險評估、包含該基因之轉殖植物所製成食品之安全性評估等基改作物資訊,則抗嘉磷塞基因大豆無法進入該國糧食市場。然而,在目前美國農業生技基改作物相關規範下,學名基改種子廠必須在專利種子專利到期後,才能進行實驗及田間試驗,呈交相關資料以符合基改作物主管機關之要求。透過建立學名基改作物快速獲得核准查驗登記之程序,允許學名廠依賴專利基改作物原廠之實驗及田間試驗資料以建立其學名基改作物之安全性與性狀表現有效性,同時,允許學名基改作物在原基改作物開發廠專利期滿前即可開始進行試驗,可以使學名抗嘉磷塞大豆種子以及其他學名基改種子能在原專利基改作物種子專利到期後順利進入市場,促使基改作物種子價格競爭,並對於專利基改作物研發公司進行適度之補償,以促進產業發展。 / The development of new technologies in plant breeding has led to improvements in the efficiency scientists produce improved plant varieties. Genetic modification is among the developments that support plant breeding. The introduction of genetically modified crops has revolutionized the agriculture industry. With patent protection available on GM traits, varieties and other aspects of seed production, private R&D investments in the seed industry have increased tremendously. Monsanto has been the leading company in investing agricultural biotechnology and has obtained a dominant position in the GM traits found in soybeans, cotton, corn and other commercialized crops. Currently, over 90 percent of soybeans planted in the United States were herbicide-resistant, with Monsanto’s Roundup Ready being the dominant soybeans planted. In 2014, however, the agriculture industry will be facing the expiration of a patent for Roundup Ready soybeans. Despiate the fact that the patent expiration date is approaching, the agricultural biotech industry has no guideline in place to tell its players exactly how to transition seamlessly from patent monopolies to generic competition. The transition problem is espectially significant for soybeans exported to oversea markets as more than 40 percent of the soybeans grown in the U.S. are exported. In order for those soybeans to be accepted in the grain channel, regulatory approvals are required in countries that import soybeans. Obtaining data, which includes scitific data on the trait being developed, for regulatory approvals can take years to complete. If a generic trait is to reach market immediately after the original trait go off patent, the data generation process should begin before patent expiration. A Hatch-Waxman type patent infringement defense for activities necessary to obtain regulatory approvals for biotech traits can ensure that seed companies have sufficient time to obtain registrations for a generic Roundup Ready trait or other generic traits. Current legislation should be modified to adequately oversee the transition to the generic use of genetically modified crops and, in the mean time, ensure the availability of generic modified crops.

Le cadre global des persécutions juives en France, les mécanismes de la Shoah sont largement connus. Sur 330 000 Juifs qui vivaient France en 1940, 80 000 ont été victimes des persécutions d’État et des déportations. En deçà de cette histoire nationale, André Kaspi s’étonne en 1991 que des centres aussi importants que Lyon, Toulouse, Grenoble n’aient pas fait l’objet d’étude attentive et scientifique (Les Juifs pendant l’Occupation, Édition du Seuil, 1991, 150 p.). Les travaux locaux ont comblé ce manque depuis. Mais le déroulement sur le terrain au quotidien, au « ras des individus », mérite encore de faire l’objet de nouvelles investigations, sans perdre de vue la diversité des situations que l’on soit de part et d’autre de la ligne de démarcation. Lyon, en zone libre jusqu’en novembre 1942, n’est pas à considérer comme Paris occupée dès juin 1940. Dans cette étude nous n’avons eu de cesse de nous interroger sur ce qui fait les spécificités de Lyon. Globalement le sort des Juifs dans la capitale des Gaules a été proche de leurs coreligionnaires de la zone sud. Toutefois, écrire l’histoire des Juifs à Lyon de l’avant-guerre à la Libération, revient à s’intéresser à des itinéraires de vie et de survie dans une ville dont certaines caractéristiques lui sont propres. L’histoire des Juifs à Lyon de l’avant-guerre à la Libération, en plus de parler de la Shoah dans la cité rhodanienne, cherche à raconter les ondes de choc d’une Europe en guerre sur les individus pour comprendre ce qui leur arrive. C’est en étant plus attentifs au tissu de la vie quotidienne, dans sa diversité individuelle que nous nous proposons dans cette étude de restituer la dimension humaine d’un monde qui a été au bord du gouffre. / The global framework of the Jew's persecutions in France as well as the mechanisms of the Shoah are widely known. 80 000 Jews out of the 330 000 who were living in France in 1940 have been the victims of state persecutions and deportations. On this side of this national history, Andre Kaspi was surprised in 1991 at seeing that cities as populated as Lyon, Toulouse or Grenoble had not been given an active and scientific consideration (Les Juifs pendant l'Occupation, Édition du seuil, 1991, 150 p.). Local research have since then enabled to address this lack. However, the daily course of operations, as close as possible to each individual, still deserves to be submitted to new investigations, without losing sight of the diversity of situations on both sides of the line of demarcation. The city of Lyon, which was within the unoccupied zone until November 1942, is not to be compared with the city of Paris which had been occupied from June 1940.In this essay, we kept wondering about the causes related to the specificities of the city of Lyon. On the whole, the fate of the Jews in the capital of the Gauls was almost the same as for their co-religionists in the south zone. Nevertheless, writing about the history of the Jews in Lyon from the pre-war years to the Liberation comes down to taking an interest in different journeys though life and survival within a city which has its own features.Besides tackling the Shoah in the Rhone city of Lyon, the history of the Jews in Lyon from the pre-war years to the Liberation, also aims at telling about the shock waves experienced by individuals in a Europe in war and perceiving what was happening to them. By paying more attention to the fabric of daily life seen in its individual diversity, we thereby intend to reconstruct the human dimension of a world which was once on the brink of the abyss.
