Rôle de la protéine ScFRK1 dans le développement du sac embryonnaire et son impact sur le guidage des tubes polliniques

Le gène Solanum chacoense Fertilization-Related Kinase 1 (ScFRK1) code pour une protéine de la famille des MAPKK kinases exprimée spécifiquement dans les ovules. Son transcrit s’accumule principalement dans la zone micropylaire du sac embryonnaire à l’anthèse et diminue rapidement après pollinisation. Ces résultats suggèrent un rôle possible avant ou pendant la fécondation. Bien qu'aucune expression ne soit détectée dans le pollen à maturité, la protéine est cependant présente dans les cellules mères de microspores. Des plantes transgéniques sous-exprimant ScFRK1 ne montrent aucun phénotype au niveau des tissus végétatifs, mais présentent de petits fruits dépourvus de graines. L’étude microscopique du gamétophyte femelle révèle que son développement ne progresse pas au-delà du stade de la mégaspore fonctionnelle et une grande proportion de sacs embryonnaires anormaux est corrélée avec une faible expression de ScFRK1. De plus, la production de pollen viable diminue en fonction de la baisse des niveaux d’expression du gène, ce qui pourrait s’expliquer par un problème au cours de la mitose I. Puisque l’intégrité du sac embryonnaire est essentielle au guidage des tubes polliniques, nous avons conçu un système de guidage semi-in vivo permettant d’évaluer la capacité des ovules du mutant ScFRK1 à les attirer. L’attraction est sévèrement affectée dans de telles conditions, ce qui confirme l'implication des cellules de la zone micropylaire comme source attractive. Notre système nous a également permis de démontrer que le guidage est très spécifique à l’espèce et que cette attraction constitue un mécanisme important favorisant la spéciation et la maintenance des barrières interspécifiques dans la reproduction sexuée des végétaux. / The Solanum chacoense Fertilization-Related Kinase 1 (ScFRK1) is a member of plant MAPKKK that is specifically expressed in ovules. ScFRK1 mRNA levels accumulate predominantly in the egg apparatus cells of the embryo sac at mature stage and decrease rapidly following pollination. These results suggest both pre- and post-fertilization roles in ovule development. Although no expression could be detected in mature pollen, FRK1 mRNAs could be detected in pollen mother cells. Transgenic plants expressing sense or antisense ScFRK1 showed no abnormal phenotype in vegetative tissues but produced seedless fruits upon pollination. A microscope-based examination of developing female gametophytes revealed that its formation did not progress further than the functional megaspore stage in affected transgenic plants and, as the levels of ScFRK1 mRNA decreased, the percentage of normal embryo sacs declined. Surprisingly, even in severely affected plants producing no or very few embryo sacs, pollination led to the production of parthenocarpic fruits. Similarly, viable pollen production declined with decreasing levels of ScFRK1 and this could be linked to affected mitosis I. Since embryo sac integrity is a prerequisite for pollen tube guidance, we devised a semi-in vivo pollen tube growth system to assess the ability of the ScFRK1 mutant ovules to attract pollen tubes. As expected, guidance was severely affected, confirming the involvement of the egg apparatus cells as the source of attracting molecules. Attraction was also determined to be highly species-specific and developmentally-regulated with the acquisition of attraction competence on anthesis day.

However, current processes for obtaining it involve high cost, high energy demand and use of toxic and pollutant solvents. This problem occurs due to the formation of an azeotropic mixture of ethanol + water, which does not allow the complete separation by conventional methods such as simple distillation. As an alternative to currently used processes, this study proposes the use of ionic liquids as solvents in extractive distillation. These are organic salts which are liquids at low temperatures (under 373,15 K). They exhibit characteristics such as low volatility (almost zero/ low vapor ), thermal stability and low corrosiveness, which make them interesting for applications such as catalysts and as entrainers. In this work, experimental data for the vapor pressure of pure ethanol and water in the pressure range of 20 to 101 kPa were obtained as well as for vapor-liquid equilibrium (VLE) of the system ethanol + water at atmospheric pressure; and equilibrium data of ethanol + water + 2-HDEAA (2- hydroxydiethanolamine acetate) at strategic points in the diagram. The device used for these experiments was the Fischer ebulliometer, together with density measurements to determine phase compositions. The experimental data were consistent with literature data and presented thermodynamic consistency, thus the methodology was properly validated. The results were favorable, with the increase of ethanol concentration in the vapor phase, but the increase was not shown to be pronounced. The predictive model COSMO-SAC (COnductor-like Screening MOdels Segment Activity Coefficient) proposed by Lin & Sandler (2002) was studied for calculations to predict vapor-liquid equilibrium of systems ethanol + water + ionic liquids at atmospheric pressure. This is an alternative for predicting phase equilibrium, especially for substances of recent interest, such as ionic liquids. This is so because no experimental data nor any parameters of functional groups (as in the UNIFAC method) are needed / O etanol anidro tem ampla aplica??o em ind?strias qu?mica, farmac?utica e de combust?veis. Fiabilité des assemblages sans plomb en environnement sévère

Le mémoire porte sur l’étude de la fiabilité des assemblages utilisant des alliages de brasure sans-plomb en environnement sévère pour des applications électroniques. Une méthode de préparation métallographique fiable et reproductible en vue de l’analyse microstructurale est présentée. L'effet du vieillissement thermique statique sur l’évolution microstructurale de billes de brasure en SAC, la tenue à la fatigue mécanique des assemblages brasés et l'étude de l'endommagement thermomécanique sont développés. Le seul effet notable constaté après vieillissement thermique est l'augmentation des épaisseurs des intermétalliques de contact. A la suite de sollicitations mécaniques, les ruptures sont plus souvent observées dans les pistes que dans les brasures, et ne permettent pas d'incriminer des éléments de la microstructure brasée comme facteurs déterminant de propagation des fissures. Les sollicitations thermomécaniques conduisent à une recristallisation, et les fissures se propagent alors le long des joints de grains recristallisés. / The thesis focuses on the study of the reliability of assemblies using Pb-free alloys under' harsh environment for electronic applications. A method for preparing metallographic reliable and reproducible for the microstructural analysis is presented. The effect of thermal aging on the static microstructural evolution of solder balls in SAC, the resistance to mechanical fatigue of solder joints and the study of thermomechanical damage are developed. The only significant effect observed after thermal aging is the increase in thickness of the intermetallic contact. Following mechanical, ruptures are most often seen in the copper tracks of the assembly, and do not blame the solder microstructure elements as determinants of crack propagation. The thermomechanical stresses lead to recrystallization, and then cracks propagate along the grain boundaries of recrystallized.

La pathophysiologie de la maladie de Ménière au niveau du sac endolymphatique : une étude immunohistochimique de l’aquaporine-2, le récepteur de Vasopressine V2R, NKCC2 et TRPV4

Objectifs: La pathophysiologie de la maladie de Ménière (MM) demeure mal comprise. Nous avons identifié dans la littérature un groupe de protéines exprimées sur le sac endolymphatique (SEL) et impliquées dans la régulation du volume endolymphatique : l’Aquaporine-2 (AQP2), le récepteur V2R de vasopressine (AVP), le Co-transporteur de Sodium Potassium et Chlorure type 2 (NKCC2) et le canal TRP type V4 (TRPV4). Notre objectif est de déterminer si leur expression sur le SEL est altérée dans la MM, pour améliorer notre compréhension de la physiologie de l’hydrops endolymphatique. Méthodes: Recrutement des cas de MM et schwannomes vestibulaires (SV) comme contrôles, le jour de leurs chirurgies respectives. Prélèvement de biopsies de SEL et sang pour AVP. L’immunohistochimie pour AQP2, V2R, NKCC2 et TRPV4 fut effectuée, et les lames scannées pour analyse digitale de densité d’expression par un logiciel spécialisé (VIS par Visiopharm®). Résultats: Total de 27 cas MM et 23 contrôles. Les scores générés par le logiciel représentent la densité d’expression totale et relative des protéines, exclusivement sur l’épithélium du SEL. Les scores d’AQP2 sont élevés de façon significative dans la MM comparée aux contrôles (p = 0.018). Nous ne rapportons aucune variation significative pour AVP, V2R, NKCC2 et TRPV4. Conclusion: Cette étude originale évalue l’expression simultanée de AQP2, V2R, NKCC2 et TRPV4 sur le SEL dans la MM, avec un groupe contrôle (SV). Nos résultats démontrent une augmentation isolée de l’AQP2 dans la MM. Nous proposons une surexpression constitutive de cette dernière, indépendante de son axe de régulation (AVP-V2R). Une mutation somatique au niveau des séquences régulatrices pourrait justifier nos observations. / Objectives: Endolymphatic sac (ELS) pathophysiology in Ménière’s Disease (MD) remains poorly understood. We identified from the literature a group of proteins expressed on the ELS and involved in endolymph volume regulation: Aquaporin-2 (AQP2), vasopressin receptor V2R, Sodium Potassium Chloride Cotransporter type 2 (NKCC2) and TRP channel type V4 (TRPV4). Our objective was to determine whether their ELS expression was altered in MD, to better understand the pathophysiology of endolymphatic hydrops. Methods: Patients with definite MD undergoing endolymphatic duct blockage surgery were recruited, as well as controls undergoing surgery for vestibular schwannomas (VS). ELS biopsies and blood samples for plasma Arginine Vasopressin (AVP) were obtained. Immunohistochemistry for AQP2, V2R, NKCC2 and TRPV4 was performed. Slides were scanned digitally for highly sensitive pixel density analysis by specialized software (VIS by Visiopharm®). Results: 27 definite MD patients and 23 VS controls were included. Global scores generated by the software represent total and relative protein expression density of 3 staining intensity levels, exclusively on ELS epithelium. AQP2 expression density was significantly elevated in MD compared to VS (p = 0.018). There was no significant difference in plasma AVP, V2R, NKCC2 and TRPV4 expression. Conclusion: This original study evaluates simultaneous in-situ expression of AQP2, V2R, NKCC2 and TRPV4 on the human ELS in MD, with a VS control group. Our results show only AQP2 up regulation on the ELS of MD patients. We suggest a constitutively increased expression of AQP2 in MD, independent of its regulatory axis (AVP-V2R). Acquired regulator sequence mutations could support this model.

Exploration du rôle de signalisation des Mitogen-Activated Protein Kinases lors de la fécondation chez les plantes

La reproduction est un événement crucial pour la vie des plantes. Ce processus nécessite la formation du pollen et des ovules. Les cellules germinales vont subir la méiose puis une succession de mitoses, deux pour le pollen et trois pour l’ovule, ce qui va leur permettre d’acquérir leurs structures finales. Une fois formés, ces deux gamètes doivent se rencontrer. Pour cela, le grain de pollen va germer sur le stigmate puis former le tube pollinique. Le tube pollinique en croissance va traverser les différents tissus femelles et ainsi tracter les deux cellules spermatiques jusqu’à l’ovule, permettant la reproduction. Un important réseau de signalisation cellulaire est nécessaire pour permettre ces événements. Les cascades des Mitogen-Activated Protein Kinases (MAPKs) sont l’un des réseaux de signalisation les plus étudiés chez les plantes. Ces kinases sont impliquées dans de très nombreux processus développementaux tels que la formation de l’embryon ou des stomates. Pour autant, leurs rôles restent encore peu caractérisés pendant de la fécondation. Ce projet a pour objectif de mieux comprendre le rôle que jouent les MAPK lors de la formation des gamètes mâles et femelles ainsi que lors de la croissance des tubes polliniques. Plusieurs membres de la superfamille des MAPKs ont été caractérisés pour leurs rôles dans la reproduction sexuée des végétaux. De précédents travaux dans le laboratoire de Daniel P. Matton, ont démontré l’implication de deux MAPK Kinases Kinases (MAP3K), la Solanum chacoense Fertilization-Related Kinase 1 (ScFRK1) et ScFRK2. Ces deux kinases sont nécessaires pour le développement de l’ovule et du pollen chez S. chacoense, une espèce de pomme de terre sauvage diploïde. Dans un premier temps, nous avons étudié la fonction d’une nouvelle ScFRK, la ScFRK3. Ce troisième membre de la classe des FRKs chez S. chacoense, est, elle aussi, impliquée dans le développement des gamétophytes mâles et femelles. Du patron d’expression jusqu’à l’établissement d’une voie de signalisation potentielle, en passant par la caractérisation phénotypique des mutants, plusieurs expériences ont été réalisées dans le but de comprendre le rôle de ScFRK3 lors de la formation des gamètes chez Solanum chacoense. Nous montrons que la ScFRK3 est impliquée dans la formation du pollen ainsi que celui des ovules. Nous avons ensuite poursuivi nos recherches en affinant le phénotypage du mutant de surexpression ScFRK2. En effet, les précédentes études ont permis de montré que la surexpression de ScFRK2 conduit le primordium ovulaire à la formation de structures carpéloïdes. Pour autant, les ensembles des primordius ovulaires ne sont pas devenu des strutures capéloïde. Nous montrons ici que seulement 10 % des ovules dans l'ovaire sont devenu 4 des carpéloïde. Notre étude montre qu’en plus des structures carpéloïdes, un grand nombre d'ovule n'ont pas de sac embryonnaire à l'anthèse ce qui explique le faible nombre de graines par fruit. L’analyse du développement des sacs embryonnaires monte que la surexpression de ScFRK2 entraine l’arrêt au stade mégaspore fonctionnelle. Ce phénotype est similaire à ce qui a pu être observé dans les lignées ARN interférant pour la ScFRK1 et la ScFRK3. De précédentes études faites chez Arabidopsis thaliana semblent montrer que les membres de la superfamille des MAPK ne sont pas essentiels pour la croissance du tube pollinique. Pour comprendre le rôle que jouent les MAPK dans l’élongation du tube pollinique, nous avons utilisé un inhibiteur des MAP Kinase Kinase (MKK), appelé U0126. La présence de cette drogue dans le milieu de croissance des grains de pollen provoque une diminution de la germination et de l’élongation du tube pollinique. L’utilisation de la méthode semi-in vivo montre une perte de la polarisation de la croissance des tubes polliniques causée par l’inhibition des MKK. La présence de l’inhibiteur conduit à la diminution de la quantité de filaments d’actine ainsi qu’à leur désorganisation à l’apex du tube. L’exocytose est aussi affectée par l’inhibition des MKK. Les cascades MAPK sont nécessaires à la croissance polarisée du tube pollinique chez Arabidopsis thaliana. Pour finir, nous avons voulu identifier certains membres de la superfamille des MAPK impliqués dans la croissance du tube pollinique. Nous nous sommes intéressés en premier lieu aux orthologues de la famille ScFRK chez Arabidopsis thaliana. Les AtMAP3K19-20-21 sont les orthologues les plus proches de ScFRK3. Ces AtMAP3K sont exprimées lors du développement des grains de pollen et lors de l’élongation de tube pollinique. L’analyse du pollen des différentes lignées mutantes montre qu’en leur absence, le pollen ne présente aucun problème de développement contrairement à ScFRK3. Par contre, les doubles mutants et le triple mutant pour les AtMAP3K19-20-21 montrent une diminution de la capacité de germination. L’élongation du tube pollinique est affectée lors de la mutation d’au moins une des AtMAP3K. Ces deux études démontrent que les MAPK sont essentielles dans la formation et l’élongation du tube pollinique. / Reproduction is a crucial event for plant life. This process requires the formation of pollen and ovules. The germ cells will undergo meiosis and then a succession of mitosis, two for the pollen and three for the ovule, which will allow them to acquire their final structures. Once formed, these two gametes must meet each other. For this, the pollen grain will germinate on the stigmas to form the pollen tube. The growth of the pollen tube will pass through the different female tissues and thus pull the two sperm cells to the ovum for reproduction. An important cellular signaling network is necessary to allow these events to occur. The Mitogen-Activated Protein Kinase (MAPKs) cascades are one of the most studied signaling networks in plants. These kinases are involved in a wide range of developmental processes such as embryo formation and stomata. However, their roles remain poorly characterized during fertilization. The aim of this project is to better understand the role played by MAPKs during the formation of male and female gametes as well as during the growth of pollen tubes. Several members of the MAPK superfamily have been characterized for their role in the sexual reproduction of plants. Previous work in Daniel P. Matton's laboratory has demonstrated the involvement of two MAPK Kinases (MAP3K), Solanum chacoense Fertilization-Related Kinase 1 (ScFRK1) and ScFRK2. These two kinases are necessary for egg and pollen development in S. chacoense, a diploid wild potato species. In a first step, we studied the functionality of a new ScFRK, ScFRK3. This third member of the class of FRKs in S. chacoense, is also involved in the development of male and female gametophytes. From the expression pattern to the establishment of a potential signaling pathway, through the phenotypic characterization of mutants, several experiments have been performed in order to understand the role of ScFRK3 in the formation of gametes in S. chacoense. We show that ScFRK3 is involved in the formation of pollen as well as that of the embryonic sac. We then continued our research by refining the phenotyping of the overexpression mutant ScFRK2. Indeed, previous studies have shown that ScFRK2 overexpression leads the ovular primordium to the formation of carpeloid structures. However, the sets of ovular primordia have not become capeloid structures. We show here that only 10% of the eggs in the ovary have become carpeloid. Our study shows that in addition to the carpeloid structures, a large number of ova do not have an embryonic sac in the anthesis, which explains the low number of seeds per fruit. The analysis of the development of the embryonic sacs shows that 7 overexpression of ScFRK2 leads to the cessation of the functional megaspore stage. This phenotype is similar to what has been observed in interfering RNA lines reducing expression of ScFRK1 and ScFRK3. Previous studies in Arabidopsis thaliana suggest that members of the MAPK superfamily are not essential for pollen tube growth. To understand the role that MAPKs play in pollen tube elongation, we used a MAP Kinase Kinase (MKK) inhibitor called U0126. The presence of this drug in the growth medium of pollen grains causes a decrease in germination and elongation of the pollen tube. The use of the semi in vivo method shows a loss of polarization of the pollen tube growth caused by the inhibition of MKK. The presence of the inhibitor leads to a decrease in the number of actin filaments and their disorganization at the apex of the tube. Exocytosis is also affected by MKK inhibition. We show in this chapter that MAPK cascades are necessary for polarized pollen tube growth in Arabidopsis thaliana. Finally, we wanted to identify some members of the MAPK superfamily involved in pollen tube growth. We were first interested in the ScFRK family orthologs in Arabidopsis thaliana. AtMAP3K19-20-21 are the closest orthologs to ScFRK3. These AtMAP3K are expressed during the development of pollen grains and during the elongation of the pollen tube. Pollen analysis of the different mutant lines shows that in their absence the pollen does not present any development problems unlike ScFRK3. On the other hand, double mutants and triple mutant for AtMAP3K19-20-21 show a decrease in germination capacity. Pollen tube elongation is affected when at least one of the AtMAP3Ks is mutated. These two studies demonstrate that MAPKs are essential for the formation and elongation of the pollen tube and that AtMAP3K19-20-21 participate in these biological processes.

[pt] Com recentes avanços em poder computacional, o uso de novos modelos de controle complexos se tornou viável para realizar o controle de quadrotores. Um destes métodos é o aprendizado por reforço profundo (do inglês, Deep Reinforcement Learning, DRL), que pode produzir uma política de controle que atende melhor as não-linearidades presentes no modelo do quadrotor que um método de controle tradicional. Umas das não-linearidades importantes presentes em veículos aéreos transportadores de carga são as propriedades variantes no tempo, como tamanho e massa, causadas pela adição e remoção de carga. A abordagem geral e domínio-agnóstica de um controlador por DRL também o permite lidar com navegação visual, na qual a estimação de dados de posição é incerta. Neste trabalho, aplicamos um algorítmo de Soft Actor- Critic com o objeivo de projetar controladores para um quadrotor a fim de realizar tarefas que reproduzem os desafios citados em um ambiente virtual. Primeiramente, desenvolvemos dois controladores de condução por waypoint: um controlador de baixo nível que atua diretamente em comandos para o motor e um controlador de alto nível que interage em cascata com um controlador de velocidade PID. Os controladores são então avaliados quanto à tarefa proposta de coleta e alijamento de carga, que, dessa forma, introduz uma variável variante no tempo. Os controladores concebidos são capazes de superar o controlador clássico de posição PID com ganhos otimizados no curso proposto, enquanto permanece agnóstico em relação a um conjunto de parâmetros de simulação. Finalmente, aplicamos o mesmo algorítmo de DRL para desenvolver um controlador que se utiliza de dados visuais para completar um curso de corrida em uma simulação. Com este controlador, o quadrotor é capaz de localizar portões utilizando uma câmera RGB-D e encontrar uma trajetória que o conduz a atravessar o máximo possível de portões presentes no percurso. / [en] With recent advances in computational power, the use of novel, complex control models has become viable for controlling quadrotors. One such method is Deep Reinforcement Learning (DRL), which can devise a control policy that better addresses non-linearities in the quadrotor model than traditional control methods. An important non-linearity present in payload carrying air vehicles are the inherent time-varying properties, such as size and mass, caused by the addition and removal of cargo. The general, domain-agnostic approach of the DRL controller also allows it to handle visual navigation, in which position estimation data is unreliable. In this work, we employ a Soft Actor-Critic algorithm to design controllers for a quadrotor to carry out tasks reproducing the mentioned challenges in a virtual environment. First, we develop two waypoint guidance controllers: a low-level controller that acts directly on motor commands and a high-level controller that interacts in cascade with a velocity PID controller. The controllers are then evaluated on the proposed payload pickup and drop task, thereby introducing a timevarying variable. The controllers conceived are able to outperform a traditional positional PID controller with optimized gains in the proposed course, while remaining agnostic to a set of simulation parameters. Finally, we employ the same DRL algorithm to develop a controller that can leverage visual data to complete a racing course in simulation. With this controller, the quadrotor is able to localize gates using an RGB-D camera and devise a trajectory that drives it to traverse as many gates in the racing course as possible.

Comparison of drug permeability in rat, pig and human in vitro models / Ruan Joubert

A crucial step in the drug discovery and development process is the assessment of membrane permeability properties of new chemical entities and researchers are constantly searching for cost-effective, high through-put models with as high as possible predictive value. In addition, a thorough understanding of the membrane permeability pathways and metabolism mechanisms are required when evaluating drug disposition and pharmacokinetics. Various in vitro methods/techniques are available to measure the rate of permeation of compounds across epithelial cell membranes to estimate oral drug absorption in humans. The aim of this study is to compare three in vitro models (i.e. excised rat intestinal tissue, excised pig intestinal tissue and Caco-2 human cell cultures) in terms of drug permeability characteristics by means of different techniques including the Ussing type Sweetana-Grass diffusion chamber apparatus, everted sac glass apparatus and the Transwell® plate apparatus. The transport of abacavir sulphate was determined in two directions (i.e. apical-to-basolateral or AP - BL and basolateral-to-apical or BL - AP) across excised rat intestinal tissue, excised pig intestinal tissue and Caco-2 cell monolayers. The test solution was applied to the donor side and samples (200 μl) were drawn from the acceptor side at 20 min intervals for a period of 2 h. The concentration of abacavir in the samples was then measured by means of a validated high performance liquid chromatography (HPLC) method. The transepithelial electrical resistance (TEER) was measured before and after each transport experiment to give an indication of the integrity of the cell membranes. The apparent permeability coefficient (Papp) and efflux ratio (ER) values were calculated and used to compare the different methods and techniques in terms of drug permeation characteristics. All three of the in vitro methods, in all of the techniques employed, showed higher transport of abacavir in the BL - AP direction than in the AP - BL direction. This indicates that all three in vitro methods had intact active efflux transporters over the entire study period. The excised rat intestinal method showed similar drug permeability characteristics in both techniques compared to that of the Caco-2 cell monolayers. In contrast, the excised pig intestinal method only showed similar drug permeability characteristics in the Sweetana-Grass diffusion apparatus when compared to the Caco-2 cell monolayers. This phenomenon can possibly be explained by the relatively large surface area of the pig tissue used in the everted sac technique where the role of physiological and other factors take effect. These factors may include the thickness of the membrane and mucus layer as well as variables such as diet, age, gender and size of the pigs obtained from the abattoir that cannot be controlled. / MSc (Pharmaceutics), North-West University, Potchefstroom Campus, 2015
