Cerca de metade dos brasileiros adultos apresentam obesidade ou sobrepeso, sendo o excesso de ganho de peso durante a gestação um dos mais importantes fatores de risco para o desenvolvimento da obesidade em mulheres. Além do ganho de peso excessivo durante a gravidez, o diabetes gestacional é outro problema metabólico enfrentado frequentemente por gestantes. No presente trabalho demonstramos que a expressão de SOCS3 em células que expressam o receptor de leptina (LepR) é essencial para causar plenamente a hiperfagia decorrente da gestação e lactação, que consequentemente, reduz o ganho de peso e o acúmulo de gordura durante estes períodos, garantindo menor retenção do peso pós-parto. Finalmente, a deleção do gene SOCS3 em células que expressam o LepR melhorou significantemente a homeostase glicêmica durante a gestação protegendo as fêmeas contra o desenvolvimento do diabetes gestacional. / About half of brazilian adults are obese or overweight, and excess weight gain during pregnancy among the most important risk factors for the development of obesity in women. Besides the excessive weight gain during pregnancy, gestational diabetes is another metabolic problem often faced by pregnant women. The present work we demonstrated that the expression of SOCS3 in LepR expressing cells has proved essential to fully cause hyperphagia due to pregnancy and lactation that, as a consequence, reduces weight gain and fat accumulation during these periods, ensuring a lower weight retention postpartum. Finally, the conditional deletion of SOCS3 significantly improves glucose homeostasis during pregnant females protecting against the development of gestational diabetes.

Yes / The realisation that unregulated activation of the Janus kinase–signal transducer and activator of transcription (JAK–STAT) pathway is a key driver of a wide range of diseases has identified its components as targets for therapeutic intervention by small molecule inhibitors and biologicals. In this review, we discuss JAK-STAT signalling pathway inhibition by the inducible inhibitor “suppressor of cytokine signaling 3 (SOCS3), its role in diseases such as myeloproliferative disorders, and its function as part of a multi-subunit E3 ubiquitin ligase complex. Influência da sensibilidade à leptina na maturação sexual de camundongos. / Leptin sensibility influences on mouse sexual maturation.

Como a incidência de obesidade em crianças têm aumentado e a leptina é um fator permissivo para o início da puberdade, os níveis circulantes de leptina elevados se tornaram um fator comum que pode causar um adiantamento da puberdade. Entretanto, o aumento no acúmulo de gordura nos organismos está associado à resistência ao hormônio leptina. Atualmente existem poucas informações sobre como a sensibilidade a esse hormônio afeta o desenvolvimento sexual. Portanto, este projeto teve como objetivo investigar como diferentes modelos de sensibilidade ou resistência à leptina agem na puberdade de camundongos fêmeas. Técnicas como acompanhamento in vivo das diferentes fases da maturação sexual e da composição corporal, testes de sensibilidade à leptina, testes de tolerância à glucose e à insulina, e PCR em tempo real foram utilizadas. Animais com maior peso corporal desde a infância apresentaram adiantamento da maturação sexual, maior acúmulo de gordura corporal e alterações na expressão de genes metabólicos. Já animais que sofreram inativação do gene Socs3 no sistema nervoso central ou em células que expressam o receptor de leptina, apresentaram um atraso na maturação sexual e alterações na expressão de genes relacionados ao consumo alimentar, porém nos casos de inativação em células Kiss1, os animais não apresentaram nenhum tipo de alteração. Dessa forma, concluímos que o peso corporal é mais importante para o desencadear da maturação sexual do que a sensibilidade do organismo ao hormônio leptina, sugerindo que a leptina atua na maturação sexual de forma indireta sobre o eixo hipotálamo hipófise gonadal. Além disso, na análise da expressão de genes ao longo do desenvolvimento traçamos um perfil de genes associados à reprodução e metabolismo, que servirão de base para comparação para outros estudos. / As the incidence of obesity in children has increased and leptin is a permissive factor for the onset of puberty, elevated circulating leptin levels have become a common factor that may cause puberty to progress. However, the increase in fat accumulation in organisms is associated with resistance to the hormone leptin. There is currently little information on how sensitivity to this hormone affects sexual development. Therefore, this project aimed to investigate how different models of leptin sensitivity or resistance act in the puberty of female mice. Techniques such as in vivo monitoring of the different stages of sexual maturation and body composition, leptin sensitivity tests, glucose and insulin tolerance tests, and real-time PCR were used. Animals with higher body weight since childhood presented an advance of sexual maturation, greater accumulation of body fat and alterations in the expression of metabolic genes. On the other hand, animals that underwent inactivation of the Socs3 gene in the central nervous system or cells expressing the leptin receptor showed a delay in sexual maturation and alterations in the expression of genes related to food consumption, but in cases of inactivation in Kiss1 cells, animals did not present any type of change. Thus, we conclude that body weight is more important for the onset of sexual maturation than the body\'s sensitivity to the hormone leptin, suggesting that leptin acts indirectly on sexual maturation on the hypothalamus - pituitary - gonadal axis. In addition, in the analysis of gene expression along the development we have traced a profile of genes associated with reproduction and metabolism, which will serve as a basis for comparison to other studies.

Activating Neuron-Intrinsic Growth Pathways to Promote Spinal Cord Regeneration After Dorsal Root Injury

Primary sensory axons fail to regenerate into the spinal cord following dorsal root injury leading to permanent sensory deficits. Re-entry is prevented at the dorsal root entry zone (DREZ), the CNS-PNS interface. Current approaches for promoting DR regeneration across the DREZ have had some success, but sustained, long-distance regeneration, particularly of large-diameter myelinated axons, still remains a formidable challenge. Our lab has previously shown that induced expression of constitutively active B-RAF (kaBRAF) enhanced the regenerative competence of injured DRG neurons in adult mice. In this study, I investigated whether robust intraspinal regeneration can be achieved by selective expression of kaBRAF alone or in combination with deletion of the myelin-associated inhibitors or neuron-intrinsic growth suppressors (PTEN or SOCS3). To this end, I used LSL-kaBRAF: brn3a-CreERT2 transgenic mice in which kaBRAF can be induced selectively in sensory neurons. I have also bred LSL-kaBRAF: brn3a-CreERT2 mice with triple knock-out mice lacking Nogo, Mag and OMgp or mouse lines carrying floxed alleles of PTEN or SOCS3. Single, double, and triple conditional mice were subjected to cervical DR crush and AAV2-eGFP vectors were used to selectively label regenerating axons of large-diameter neurons. I compared the extent of regeneration at 3 weeks or 2 months after DR injury using conventional anatomical and behavioral analyses. I found that kaBRAF alone promoted axon regeneration across the DREZ but did not produce significant functional recovery by two months. Supplementary deletion of Nogo, MAG, and OMgp did not improve kaBRAF-induced regeneration. Deletion of PTEN or SOCS3 individually or in combination failed to promote axon regeneration across the DREZ. In marked contrast, simultaneous deletion of PTEN, but not SOCS3, dramatically enhanced kaBRAF-mediated regeneration enabling many more axons to penetrate the DREZ and grow deep into the spinal cord. This study shows that dual activation of BRAF-MEK-ERK and PI3K-Akt signaling is an effective strategy to stimulate robust intraspinal DR regeneration and may lead to recovery of sensory function after DR injury. / Biomedical Sciences

Les astrocytes sont des éléments clés de la physiologie cérébrale. Dans les maladies neurodégénératives comme la maladie d’Alzheimer (MA), les astrocytes deviennent réactifs. Cette réactivité astrocytaire (RA) est essentiellement caractérisée par des changements morphologiques. En revanche, les effets de la réactivité sur les fonctions de support des astrocytes sont mal connus. De plus, les cascades de signalisation qui conduisent à la RA restent à déterminer. Les objectifs de ce projet étaient de : 1/ démontrer que la voie JAK2-STAT3 (Janus Kinase 2 - Signal Transducer and Activator of Transcription 3) joue un rôle central dans le contrôle de la RA au cours des maladies neurodégénératives ; 2/ comprendre quelle est l’implication de la RA dans les altérations moléculaires, cellulaires et fonctionnelles observées dans la MA. Nous avons montré que la voie JAK2-STAT3 est une cascade de signalisation centrale dans la RA (Ben Haim et al., 2015). Dans ce projet, nous démontrons en utilisant de nouveaux outils moléculaires basés sur des vecteurs viraux, que cette voie est nécessaire et suffisante à la RA. Nos résultats montrent également que la modulation de la RA dans deux modèles murins de la MA (souris APP/PS1dE9 et 3xTg-AD) influence certains index pathologiques, mais de façon contexte-dépendante. L’ensemble de ce travail a permis de valider de nouveaux outils pour étudier les astrocytes réactifs in situ et souligne l’importance et la complexité de leur fonctions au cours des maladies neurodégénératives. / Astrocytes are emerging as key players in brain physiology. In Alzheimer’s disease (AD), astrocytes become reactive. Astrocyte reactivity (AR) is essentially characterized by morphological changes. But how the normal supportive functions of astrocytes are changed by their reactive state is unclear. Moreover, signaling cascades leading to AR are not yet determined. In this study, we aim to: 1/ demonstrate the JAK2-STAT3 pathway (Janus Kinase 2 - Signal Transducer and Activator of Transcription 3) is responsible for AR in neurodegenerative diseases ; 2/ understand the contribution of reactive astrocytes to molecular, cellular and functional alterations in AD. We already reported that the JAK2- STAT3 pathway is a central cascade for AR (Ben Haim et al., 2015). Here, we demonstrate, with new molecular tools based on viral vectors, that this pathway is necessary and sufficient to AR. Our results also show that the modulation of AR in two AD mouse models (APP/PS1dE9 and 3xTg-AD mice) influence several pathological hallmarks, but in a context-dependent manner. Overall, this work has generated new original tools to study reactive astrocytes in situ and it underlines the importance and complexity of their functions in neurodegenerative diseases.
