Transforming Growth Factor Beta 3-Loaded Decellularized Equine Tendon Matrix for Orthopedic Tissue Engineering

Transforming growth factor beta 3 (TGF3) promotes tenogenic differentiation and may enhance tendon regeneration in vivo. This study aimed to apply TGF3 absorbed in decellularized equine superficial digital flexor tendon scaffolds, and to investigate the bioactivity of scaffold-associated TGF3 in an in vitro model. TGF3 could effectively be loaded onto tendon scaffolds so that at least 88% of the applied TGF3 were not detected in the rinsing fluid of the TGF3-loaded scaffolds. Equine adipose tissue-derived multipotent mesenchymal stromal cells (MSC) were then seeded on scaffolds loaded with 300 ng TGF3 to assess its bioactivity. Both scaffold-associated TGF3 and TGF3 dissolved in the cell culture medium, the latter serving as control group, promoted elongation of cell shapes and scaffold contraction (p < 0.05). Furthermore, scaffold-associated and dissolved TGF3 affected MSC musculoskeletal gene expression in a similar manner, with an upregulation of tenascin c and downregulation of other matrix molecules, most markedly decorin (p < 0.05). These results demonstrate that the bioactivity of scaold-associated TGF3 is preserved, thus TGF3 application via absorption in decellularized tendon scaffolds is a feasible approach.

Effects on immune cell viability, morphology and proliferation in a sub-microliter cell sampler system

Today,   most traditional method used in the research of immune cells, such as flow   cytometry and microscopy, are based on average values of cell responses.   However, immune cells are heterogeneous and respond differently to a given   stimuli. There is also a risk that important, but rare, behaviors of   individual cells are missed when a larger population of immune cells is   analyzed. Also, flow cytometry and microscopy do not allow long-term survival   of cells; these methods lack the ability to do dynamic long-term analysis of   motile immune cells, i.e. studies of cell-cell interactions, morphology and proliferation.   In a   patient who is affected by cancer, the cell heterogeneity contributes to the   ability to battle various types of cancer or virus infections. In an   outbreak, immune cells recognize and kill tumor cells. However, the number of   specific immune cells is sometimes too few to kill all the tumor cells in a   successful way. One way to help these patients is to isolate, select out and   cultivate the active immune cells with capacity to kill tumor cells.   The   Cell Physic Laboratory (a part of the department of Applied Physics) at the   Royal Institute of Technology (KTH) has developed a method for single-cell   analysis where the immune cells are trapped in microwells in a silicon chip.   The immune cells are then studied by using fluorescence microscopy in an   inverted setup. The method enables high-throughput experiments due to the   parallelization. Furthermore, since the immune cells survive long periods in   the chip, the cells can be analyzed over several days up to weeks. The   research group has also developed a semi-automatic ‘cell-picker’. The   cell-picker will be used in combination with the developed method for   single-cell analysis, which enables picking of cells of interest. In this report, experiments for the characterization and evaluation of the biocompatibility of two generations of the cell-picker will be presented. The experiments include development of a protocol for the cell-picking process, studies of the survival time of transferred cells for both generation of the cell-picker and studies of surface coating in the chip in order to increase the biocompatibility. The preliminary results indicate that the cell-picker has potential to be used as a selection tool for immune cells of interest.

A texturização via feixe laser confere maior aderência às superfícies dos dispositivos médicos implantáveis. Este trabalho teve o objetivo de avaliar o comportamento da superfície do aço inoxidável austenítico ABNT NBR ISO 5832-1 submetido à marcação e texturização com feixe laser de fibra óptica, alterando-se a frequência dos pulsos; frente ao seu comportamento biomecânico, por meio de: ensaios de resistência à tração, fadiga e desgaste; verificar a susceptibilidade à corrosão localizada, por ensaios eletroquímicos em solução que simula os fluidos corpóreos; e caracterizar a sua microestrutura. Os tratamentos alteraram a rugosidade e a dureza dos biomateriais em função do aumento da frequência dos pulsos. A microestrutura e composição química das superfícies sofreram mudanças que afetaram diretamente a camada passiva dos aços inoxidáveis. Este efeito foi comprovado com o uso de SVET, XPS e caracterização de propriedades eletrônicas do filme passivo. Os dois tipos de tratamentos implicaram em aumento de susceptibilidade magnética das superfícies. Os parâmetros utilizados para as marcações e texturizações não causaram diminuição na viabilidade celular, de modo que não apresentou citotoxicidade mesmo após incubação prolongada. Este biomaterial mostrou-se adequado perante os ensaios biomecânicos, uma vez que os tratamentos a laser, nas condições utilizadas, não induziram a formação de tensões superficiais de magnitude capaz de levar à fratura por fadiga, indicando vida em fadiga infinita; tampouco se pôde relacionar a região de fratura por tração com as marcações a laser. O volume de desgaste diminuiu em função do aumento da dureza produzido pela elevação da frequência do pulso nas texturizações. Desenvolvimento de um método simplificado para obtenção de adesão à zircônia / Development of a simplified method for obtaining membership of the zirconia

The method was based on deposition of a reactive silica layer on the ceramic surface followed by heat treatment. The study comprised six steps: (i) preparation of solutions using four concentrations of tetraethyl orthosilicate (TEOS) and zirconium tert-butoxide (ZTB) diluted in hexane; (ii) cutting and polishing of the zirconia substrate; (iii) organic silica-based treatment before (infiltration, INF) or after (coating, COA) zirconia sintering; (iv) analysis by scanning electron microscopy and energy dispersive spectroscopy (SEM-EDS); (v) G-Raman confocal spectroscopy analysis; (vi) shear bond strength to zirconia tested after 24-h and failure analysis. An untreated (control) and a commercial reference (Rocatec Plus, 3M ESPE) groups were tested. Quantitative data were analyzed using ANOVA and Tukey s post hoc test (P < 0.05). SEM micrographs showed that zirconia surface was covered by silica nanoparticle clusters. EDS and G-Raman analyses confirmed composition of this layer. The bond strength results showed that most groups that received coating (COA) and infiltration (INF) presented higher bonding potential than the untreated (control) group. Almost all experimental groups were similar to commercial reference. Mixed failures were predominant. In conclusion, the present study introduces a novel, simple, and cost-effective method to provide adhesion to yttria-stabilized zirconia ceramic. Both the method of silica deposition (if before or after zirconia sintering) and concentration of organic silica precursors have a significant impact on the adhesion of methacrylates to the treated zirconia / O objetivo deste estudo foi desenvolver um método simplificado para obtenção de adesão a uma cerâmica de zircônia estabilizada por ítria. O método foi baseado na deposição de uma camada reativa de sílica na superfície cerâmica seguida por tratamento térmico. O estudo compreendeu seis etapas: (i) preparo de 4 soluções contendo tetraetilortosilicato (TEOS) e tert-butóxido de zircônio (ZTB) diluídos em hexano; (ii) corte e polimento do substrato de zircônia; (iii) tratamento com os precursores orgânicos antes (infiltração, INF) ou depois (cobertura, COA) da sinterização da zircônia; (iv) análise por microscopia eletrônica de varredura e espectroscopia de energia dispersiva (MEV-EDS); (v) análise por espectroscopia confocal G-Raman; (vi) avaliação de resistência de união ao cisalhamento 24h após preparo e análise de falha. Grupos sem tratamento (controle) e uma referência comercial (Rocatec Plus, 3M ESPE) foram testados. Dados quantitativos foram analisados usando ANOVA e teste post hoc de Tukey (P < 0,05). Os resultados de MEV mostraram que a superfície da zircônia foi coberta por aglomerados de nanopartículas de sílica, sendo esta composição confirmada por EDS e análise de G-Raman. Os resultados do teste mecânico mostraram que a maioria dos grupos que receberam cobertura (COA) e infiltração (INF) apresentaram maior resistência de união que o grupo sem tratamento (controle). A maioria dos grupos experimentais foram similares à referência comercial. Falhas do tipo mistas foram predominantes. Em conclusão, o presente estudo introduz um método novo, simples e de baixo custo para promover adesão a cerâmicas de zircônia estabilizada por ítria. Tanto o método de deposição de sílica (se antes ou após sinterização da zircônia) e a concentração de precursores orgânicos de sílica têm impacto significativo na adesão de metacrilatos à cerâmica tratada

Pillared Paddle-Wheel Frameworks als stationäre Phasen für gaschromatographische Trennungen

Metal-organic Frameworks (MOFs) sind eine neue Klasse poröser und kristalliner Feststoffe, die durch ihren modularen Aufbau aus organischen und anorganischen Einheiten einzigartige Eigenschaften unter den porösen Materialien besitzen. Im Mittelpunkt dieser Arbeit steht ihre Anwendung im Bereich der Gaschromatographie, die bislang nur wenig erforscht ist. Dazu werden drei verschiedene MOFs aus der Reihe der „Pillared Paddle-Wheel Frameworks“ (PPFs) in GC Kapillarsäulen abgeschieden und untersucht. Durch systematische Messungen kann gezeigt werden, dass PPFs nicht nur zur Analyse flüchtiger organischer Verbindungen, sondern auch für spezielle Anwendungen wie Größenausschlusschromatographie und Enantiomerentrennungen anwendbar sind. Weiterhin wurden Adsorptionsenthalpien und -entropien sowie Diffusionskonstanten und Massenübergangskoeffizienten für ein breites Analytenspektrum bestimmt.

Analyse des nanoparticules de dioxyde de cérium à l’aide de l’ICP-MS en mode particule unique

Due to their unique properties, engineered nanomaterials are now widely used in numerous commercial products. Cerium oxide (CeO2) nanoparticles (NPs) are among the most commonly used engineered NPs, with applications in surface coatings, catalysis, the manufacturing of semiconductors, biomedicine and agriculture. With the significant increase in the production and use of CeO2 NPs, concern is increasing over their release into the environment and their subsequent fate and toxicity. In order to evaluate their environmental risk, it is necessary to detect, quantify and characterize the NPs in all environmental compartments. Unfortunately, analyses of NPs in natural systems are challenging due to their small sizes, their low concentrations (∼ ng L-1) and the complexity of environmental matrices, which also contain natural colloids. Single particle inductively coupled plasma mass spectrometry (SP-ICP-MS) is a specific and sensitive technique that enables the detection of very low concentrations (∼ ng L-1) of NPs and it can provide information on their number concentrations, sizes, and size distributions. This technique is often limited by high size detection limits (SDL). However, it is especially important to obtain rigorous size, concentration, and fate data for the smallest NPs, since they are expected to have the greatest environmental risk. To that end, the specific objective of this thesis was to develop an improved method for the detection, quantification, and characterization of CeO2 NPs in complex natural waters using SP-ICP-MS. The project was then divided into several objectives: (1) decrease the SDL for CeO2 NPs; (2) optimize the preparation method for natural water samples; (3) apply the preparation and the analysis methods to detect, quantify, and characterize CeO2 NPs in several natural water samples and commercial products, such as a paint and a stain; (4) identify the origin (natural or engineered) of the detected CeO2 NPs; (5) quantify and characterize the release of CeO2 NPs from paint and stain under natural weathering scenarios; and (6) evaluate the effect of different physicochemical conditions (pH, ionic strength, and NOM) on the fate of CeO2 NPs after their release. A high sensitivity sector field ICP-MS (SF-ICP-MS) with microsecond dwell times (50 μs) was used to lower the SDL of CeO2 NPs to below 4.0 nm. While filtration is often used as a preparation method for SP-ICP-MS, its effect on the concentrations and sizes of NPs is unknown. For this purpose, the interactions between six different membrane filters and CeO2 NPs in aqueous samples were examined. The highest recoveries were observed for polypropylene membranes, where 60 % of the pre-filtration NPs were found in a rainwater and 75% were found in a river water. Recoveries could be increased to over 80% by pre-conditioning the filtration membranes with a multi-element solution. Similar recoveries were obtained when samples were centrifuged at low centrifugal forces (≤1000xg). SF-ICP-MS was then used to detect CeO2 NPs in Montreal rainwater, St. Lawrence River water, a paint, and a stain. A significant decrease in the concentrations of CeO2 NPs, initially contained in paint and stain, was measured over time under different conditions, which was attributed to agglomeration and/or dissolution. Finally, when painted and stained panels were placed outside, the released Ce in the precipitation was mainly in the dissolved form with no significant release of CeO2 NPs. / En raison de leurs propriétés uniques, les nanomatériaux manufacturés sont maintenant largement utilisés dans de nombreux produits commerciaux. Les nanoparticules (NPs) de dioxyde de cérium (CeO2) sont parmi les NPs manufacturées les plus couramment utilisées, avec des applications dans les revêtements de surface, la catalyse, la fabrication de semi-conducteurs, la biomédecine et l’agriculture. Avec l’augmentation significative de la production et l’utilisation des NPs de CeO2, l’inquiétude grandit quant à leur rejet dans l’environnement et à leur devenir et toxicité subséquente. Afin d’évaluer leur risque environnemental, il est nécessaire de détecter, quantifier et caractériser les NPs dans tous les compartiments environnementaux. Malheureusement, les analyses des NPs dans les systèmes naturels sont difficiles en raison de leurs petites tailles, de leurs faibles concentrations (∼ ng L-1) et de la complexité des matrices environnementales, qui contiennent également des colloïdes naturels. La spectrométrie de masse à plasma à couplage inductif en mode particule unique (SP-ICP-MS) est une technique spécifique et sensible qui permet la détection de très faibles concentrations (∼ ng L-1) de NPs en fournissant des informations sur leurs concentrations en nombre, leurs tailles et leurs distributions de taille. Cette technique est souvent limitée par des limites de détection de taille (SDL) élevées. Cependant, il est particulièrement important d’obtenir des données précises sur la taille, la concentration et le devenir des plus petites NPs, qui présentent un plus grand risque environnemental. À cette fin, l’objectif spécifique de cette thèse était de développer une méthode améliorée pour la détection, la quantification et la caractérisation des NPs de CeO2 dans les eaux naturelles complexes à l’aide de SP-ICP-MS. Le projet a ensuite été divisé en plusieurs parties : (1) diminuer la SDL pour les NPs de CeO2; (2) optimiser la méthode de préparation des échantillons d’eau naturelle; (3) appliquer les méthodes de préparation et d’analyse pour détecter, quantifier et caractériser les NPs de CeO2 dans plusieurs échantillons d’eau naturelle et produits commerciaux, telles qu’une peinture et une teinture; (4) identifier l’origine (naturelle ou manufacturée) des NPs de CeO2 détectées; (5) quantifier et caractériser le relargage des NPs de CeO2 de la peinture et de la teinture sous différents scénarios météorologiques; et (6) Évaluer l’effet des différentes conditions physicochimiques (pH, force ionique, et la présence de la matière organique naturelle) sur le devenir des NPs de CeO2 après le relargage. Un ICP-MS à secteur magnétique (SF-ICP-MS) à haute sensibilité avec des temps d’acquisition (i.e. ‘dwell times’) à l’échelle de microsecondes (50 μs) a été utilisé pour diminuer la SDL des NPs de CeO2 à moins de 4.0 nm. Alors que la filtration est souvent utilisée comme méthode de préparation pour le SP-ICP-MS, son effet sur les concentrations et les tailles des NPs est inconnu. Pour cela, les interactions entre six filtres avec des membranes différentes et les NPs de CeO2 dans des échantillons aqueux ont été examinées. Les recouvrements les plus élevés ont été observés pour la membrane de polypropylène, où 60% des NPs de pré-filtration ont été trouvées dans l’eau de pluie et 75% dans les eaux de rivière. Les recouvrements pourraient être augmentés à plus de 80% en préconditionnant les membranes des filtres avec une solution multi-éléments. Des recouvrements similaires ont été obtenus lorsque les échantillons ont été centrifugés à une faible vitesse de rotation (≤1000xg). Le SF-ICP-MS a ensuite été utilisé pour détecter des NPs de CeO2 dans une eau de pluie de Montréal, une eau du fleuve St. Laurent, une peinture et une teinture. En se basant sur le rapport de cérium/lanthane (Ce/La), les NPs de CeO2 détectées dans la pluie sont majoritairement d’origine naturelle alors que celles dans la peinture et la teinture sont manufacturées. Une diminution significative des concentrations des NPs de CeO2, originairement contenues dans la peinture et la teinture, a été mesurée avec le temps, sous l’effet de différentes conditions, ce qui a été attribué à l’agglomération et/ou la dissolution. Finalement, lorsque des panneaux peints et teints ont été placés à l’extérieur, le Ce relargué était principalement sous la forme dissoute, sans relargage significatif des NPs de CeO2.

SUSTAINABLE LUBRICATION FOR FUTURE TRANSMISSIONS : Micropitting performance of Glycerol-based lubricants

Achieving sustainable lubrication by using environmentally friendly formulated lubricants has became an essential component of the transition process from fossil-powered vehicles to electrified transportation. Mixtures, or aqueous solutions of molecules such as polyhydroxy alcohols, and glycols usually known as green lubricants make it possible to achieve low friction coefficients under different lubrication conditions, which constitutes a potential alternative to improve the tribological performance of moving parts in automotive systems, at the same time that the environmental requirements are satisfied. There is a need to improve the protection against mild-wear and micropitting offered by green-lubricants before consider using them in the transmission gear box of battery electric vehicles BEVs, where their low shear-stress resistance can potentially help to improve the vehicle efficiency. Therefore, this research work aims to gain understanding of the tribological behaviour of rolling-sliding elements under glycerol-based lubrication, with focus on assessing the influence of different glycerol-based lubricant formulations on the wear modes associated to rolling contact fatigue such as micropitting, and macropitting.  Micropitting tests were carried out by using a micropitting rig, with a roller on ring planetary configuration. The evolution of microcracks, and micropitting was studied for several glycerol-based lubricants at different slide-to-roll ratios (SRRs) 5-30%, and different load contact conditions 1.5-2.5 GPa. A comparison against a fully formulated transmission oil has been performed. A relationship between the surface damage morphology and the operating conditions has been established. The lubrication regimes of DLC coated contacts and uncoated contacts in presence of glycerol-based lubricants were investigated through the construction of 3D friction maps, and Stribeck curves in a wide range of rolling speeds, and SRR testing conditions. The capability of two different DLC coating systems to prevent micropitting onset due to rolling contact fatigue in presence of glycerol-based lubricants was studied: Cr/a-WC:H/a-C:H and a-C:Cr.  Under mixed-lubrication regime it was found that a reduction up to 51% of friction can be achieved by using glycerol-based lubricants compared to a commercial transmission oil. The initial low friction coefficients at low contact cycles was attributed to the low pressure-viscosity coefficient of the glycerol-based lubricants. Friction was even further reduced with the increasing contact cycles since glycerol aqueous solutions were found to promote mild-wear, causing a smoothing of the surface asperities and therefore an enhancement of the hydrodynamic effect due a higher lambda ratio (Λ), making possible to trigger superlubricity in the contact. Surface micro-cracking was found to decrease at low SRR values under glycerol-based lubrication contrasted to the contacts lubricated with a fully formulated transmission oil. Once microcracks were nucleated, micropitting progressed faster in the contacts lubricated with glycerol-based lubricants, while micropitting was null or significantly delayed in presence of transmission oil. With the increasing SRR surface microcracks density and micropitting was found to became higher. The reduction of microcracks density with a glycerol-glycol based lubricant was attributed to the adsorption of long glycol molecules on the steel surface that avoid the direct interaction between asperities. The faster micropitting progression was attributed to the presence of dissolved water in the contact. Thin DLC a-C:Cr coatings were shown to significantly mitigate the penetration of surface initiated pits into the steel substrate, which was translated in a lower volume loss. Local micro-delamination was found to be the main reason behind coating failure in presence of glycerol-based lubricants, as a result, pitting took place on the steel exposed areas.

Revêtement en LiAlO2 sur des particules d’un matériau d’électrode positive LiNi0,6Mn0,2Co0,2O2 pour batterie aux ions lithium

Des progrès dans les batteries aux ions lithium sont en cours de développement afin de répondre, entre autres, à la demande croissante des hautes densités d'énergie et de puissance pour le réseau électrique et en particulier pour l'application dans les véhicules électriques. Ces derniers remplacent écologiquement les véhicules à moteur à combustion interne et leurs succès est principalement dû à leur efficacité énergétique supérieure, à leurs faibles coûts d'exploitation et à leur profil respectueux de l'environnement par rapport aux véhicules à essence. Parmi les différents matériaux de cathode, les composés d'intercalation LiNixMnyCo1-x-yO2 (NMC) sont les meilleurs candidats pour des applications dans les batteries aux ions lithium à hautes performances. Des efforts sont en cours pour mettre en oeuvre des matériaux cathodiques à base de NMC riches en nickel pour répondre aux besoins environnementaux et énergétiques. Aussi séduisants soient-ils, ces matériaux de cathode présentent certains inconvénients liés à une forte réactivité, notamment à l'interface avec l'électrolyte. Pour contourner ces problèmes, des modifications de surface sont étudiées comme des solutions accessibles pour protéger le matériau actif et améliorer ses performances. Bien que diverses chimies et stratégies de revêtement soient publiées dans la littérature, notre approche consistant à combiner la synthèse et la modification de surface du matériau actif en une étape est aussi simple qu'efficace. Le présent manuscrit porte sur l’étude de ce composé. Deux méthodes de revêtement de surface ont été étudiées et leur matériau revêtu résultant a été comparé au matériau non revêtu. Après une caractérisation détaillée de ces matériaux, des études électrochimiques ont été menées afin d’évaluer leurs performances. Enfin, notre NMC622 revêtu de LiAlO2 en une seule étape s'est avéré efficace pour contrer la dégradation de la capacité du NMC et pour améliorer la stabilité structurelle des particules, améliorant ainsi leur cycle de vie. / Advances in lithium-ion batteries are being developed in order to meet, among other things, the increasing demand for high energy and power densities for the electric power grid and especially for application in electric vehicles. The latter are a green replacement for internal combustion engine vehicles, and their success is mostly due to their higher energy efficiency, low operating costs and eco-friendliness compared to gasoline-powered vehicles. Among various cathode materials, LiNixMnyCo1-x-yO2 (NMC) intercalation compounds are the best candidates for applications in high performance lithium-ion batteries. Efforts are underway to implement nickel-rich NMC-based cathode materials to meet environmental and energy needs. As appealing as they are, these cathode materials present certain drawbacks associated with high reactivity, especially at the interface with the electrolyte. To circumvent these issues, surface modifications are investigated as accessible solutions to protect the active material and enhance its performance. Although various coating chemistries and strategies are published in the literature, our approach of combining synthesis and surface modification of the active material in a single pot is as simple as it is efficient. The following manuscript will be covering the study of this material. Two methods of surface coating were studied, and their resulting coated material was compared to the uncoated material. After a detailed characterization of these materials, electrochemical studies were carried out to evaluate their performance. Finally, our resulting one pot LiAlO2- coated NMC622 has shown to be effective in counteracting NMC capacity degradation and improving the structural stability of the particles, thereby improving their cycle- life.

Surface coating is widely used in microelectronic industry to produce thin films over surfaces with uneven topography. Such processes are used in fabricating integrated circuits, storage devices, such as magnetic disks, memory devices and optical disks as well as for manufacturing adhesives, magnetic tapes, magazines which can produce thicker films over patterns of similar depth and width at higher speeds. Other applications of film flow over uneven surfaces come from specific designs of surfaces of heat-exchangers and the surfaces of various structured packings used to improve heat and mass transfer operations. The one-dimensional, gravity-driven film-flow of a linear or exponential PTT liquid, flowing either on the outer or on the inner surface of a vertical cylinder or over a planar wall is analyzed. Numerical solution of the governing equations is generally possible. Analytical solutions are derived only for: (i) linear PTT model in cylindrical and planar geometries in the absence of solvent and the affinity parameter set at zero; (ii) linear or exponential PTT model in a planar geometry in the absence of solvent and the affinity parameter the affinity parameter obtains nonzero values; (iii) exponential PTT model in planar geometry in the absence of solvent and the affinity parameter set at zero. Then, the two-dimensional, steady flow of a viscoelastic film over a periodic topography under the action of a body force is studied. It is examined the interplay of elastic, viscous, inertia and capillary forces on the film thickness and planarization efficiency over steep topographical changes of the substrate. The code is validated by verifying that in isolated topographies the periodicity conditions result in fully developed viscoelastic film flow at the inflow/outflow boundaries and that its predictions for Newtonian fluids over 2D topography under creeping flow conditions coincide with those of previous works. Finally, the steady film-flow of a Newtonian fluid has been studied over a trench examining the various types of inclusions that can be formed. It can be distinguished three possible flow configurations when (a) the triple contact points are ‘pinned’ at the lips of the cavity, (b) the triple contact points are at the left side and the bottom of the cavity so that the cavity is not filled with liquid only around its left concave corner and (c) the two triple contact points are at the two sides of the cavity so that its bottom remains empty. / Η μελέτη ρευστών υμένων κατέχει σημαντική θέση στη σύγχρονη επιστήμη και τεχνολογία και συναντάται ευρύτατα σε βιομηχανικές διεργασίες (π.χ. κατασκευή ηλεκτρονικών εξαρτημάτων όπως είναι μικροεπεξεργαστές, ολοκληρωμένα κυκλώματα και συσκευές μνήμης, κατασκευή ψηφιακών αποθηκευτικών μέσων όπως μαγνητικοί δίσκοι, δισκέτες, κασέτες και οπτικοί δίσκοι, διεργασίες φωτολιθογραφικής επικάλυψης και εκτύπωσης όπως επικαλυπτικές ουσίες, μαγνητικές ταινίες και περιοδικά κ.α.). Επιπλέον, οι ρευστοί υμένες χρησιμοποιούνται για την βελτιστοποίηση διεργασιών μεταφοράς θερμότητας και μάζας (εναλλάκτες θερμότητας). Πρώτα αναλύεται η μονοδιάστατη ροή λόγω βαρύτητας ενός υμένα ιξωδοελαστικού ρευστού που ακολουθεί το καταστατικό μοντέλο Phan-Thien and Tanner (PTT) σε γραμμική ή εκθετική μορφή. Το ρευστό ρέει είτε στην εξωτερική είτε στην εσωτερική επιφάνεια ενός κατακόρυφου κυλίνδρου ή σε επίπεδο τοίχωμα. Η αριθμητική επίλυση των διεπουσών εξισώσεων είναι πάντοτε δυνατή. Αναλυτικές εκφράσεις εξάγονται μόνο για (i) γραμμικό μοντέλο PTT σε κυλινδρικές και επίπεδες γεωμετρίες απουσία διαλύτη και η παράμετρος συγγένειας είναι μηδενική, (ii) γραμμικό ή εκθετικό μοντέλο PTT σε επίπεδη γεωμετρία απουσία διαλύτη με μη μηδενική παράμετρο συγγένειας και (iii) εκθετικό μοντέλο PTT σε επίπεδη γεωμετρία απουσία διαλύτη και με μηδενική παράμετρο συγγένειας. Στη συνέχεια μελετάται η δισδιάστατη μόνιμη ροή ενός ιξωδοελαστικού υμένα κατά μήκος περιοδικής τοπογραφίας υπό την επίδραση είτε της βαρύτητας είτε της φυγοκέντρου δύναμης. Εξετάζεται τόσο η επίδραση των ιξωδοελαστικών ιδιοτήτων υπό συνθήκες έρπουσας ροής όσο και των τριχοειδών και αδρανειακών δυνάμεων καθώς και της γεωμετρίας του υποστρώματος στο πάχος του υμένα και στη δυνατότητα εξομάλυνσής του παρά τις απότομες αλλαγές του υποστρώματος. Ο κώδικας ελέγχθηκε μέσω της σύγκρισης των προβλέψεών του με προηγούμενες εργασίες που αφορούν σε Νευτωνικά και σε ιξωδοελαστικά ρευστά. Τέλος, εξετάζεται η δισδιάστατη μόνιμη ροή ενός Νευτωνικού υμένα πάνω από παρόμοια μεταβαλλόμενο υπόστρωμα, ενώ επιτρέπεται να σχηματίζονται εγκλείσματα αέρα μεταξύ του υμένα και του υποστρώματος. Διακρίνονται οι εξής δυνατότητες σχηματισμού εγκλεισμάτων αέρα ανάλογα με τη θέση της κάτω επιφάνειας του υμένα: (α) εμφανίζονται σταθερά σημεία επαφής στις άνω (κυρτές) γωνίες του υποστρώματος, (β) σχηματίζεται έγκλεισμα γύρω από την πρώτη κοίλη γωνία του υποστρώματος και (γ) εμφανίζονται δύο τριπλά σημεία επαφής με τις παράπλευρες επιφάνειες του υποστρώματος.

No. of bitstreams: 0 / Os projetos de carrocerias veiculares atuais procuram desenvolver estruturas leves, seja para reduzir o consumo de combustível, no caso dos motores de combustão interna, seja para maior autonomia de bateria, no caso dos veículos elétricos e híbridos. Redução no consumo de combustível significa redução na emissão de poluentes. As estruturas precisam ser leves, mas cada vez mais resistentes e rígidas a fim de proporcionar máximo conforto e segurança aos ocupantes. Estas premissas têm levado ao contínuo desenvolvimento dos materiais. No caso dos aços, um dos processos que tem permitido a melhora significativa das propriedades mecânicas é a estampagem a quente. Nos últimos anos, as peças estampadas a quente têm ocupado lugar de destaque na estrutura das carrocerias veiculares por estarem em sintonia com as demandas mencionadas. Há muitas pesquisas em curso para esta tecnologia, seja nos materiais, nos meios de produção, nos revestimentos e em aplicações. O aço mais utilizado neste processo, 22MnB5, também apresenta o chamado efeito bake hardening; a tensão de escoamento é aumentada após tratamento térmico realizado em temperaturas próximas a 200 °C. Neste trabalho, visando à melhoria nas propriedades mecânicas, amostras foram tratadas termicamente na faixa de temperatura supracitada. Após isso, dados obtidos de ensaios mecânicos foram inseridos em programas de simulação de impacto lateral cujo resultado foi a redução na intrusão na célula de sobrevivência. O efeito bake hardening também propiciou um aumento na absorção da energia de impacto em teste estático feito com barras de proteção lateral.
