Side population is a minor subpopulation (SP) of some cell lines, exporting staining dye Hoechst 33342 out of their cytoplasm. It is discussed as a possible source of "cancer stem cells", "tumour initiating cells" or "metastasis initiating cells". However, broad literature suggest, that stemness and other privileged properties of SP are very variable between different cell types, cell lines and stage of disease. Cell lines TRAMP are the only widely available murine models for testing of prostate cancer therapy. We noticed in literature a mention about existence of 1-2% of cells constituting side population, but detailed characteristic have not been described until now. In this diploma thesis, we worked on characterisation of SP of the TRAMP-C2 cell line in comparison to other cells (nonSP). In the first part, we compared stem properties of SP and nonSP. We started with checking the existence of SP by its verapamil sensitivity. Using mRNA analysis, we showed that neither SP nor nonSP have increased c-Kit expression and that there are no differences in Bmi-1 expression. We found that SP is heterogenic mixture of CD24-CD44-, CD24-CD44+ and CD24+CD44+ cells, while nonSP is almost solely CD24-CD44+. As formigas andarilhas têm adquirido uma notoriedade científica graças aos trabalhos realizados desde a década de 70 quando foi constatada a presença de patógenos nas amostras de formigas coletadas de ambientes hospitalares. Os trabalhos elaborados a partir de então relataram a presença dos gêneros Salmonella, Staphylococcus, Klebsiella e Enterobacter nesses ambientes além de outros microrganismos patogênicos de importância. No entanto, pouco conhecimento foi produzido a partir da sua presença em ambientes em que se manipulam e produzem alimentos para consumo humano. As formigas andarilhas têm sido observadas com freqüência em domicílios, áreas de manipulação e fabrico de alimentos asism como em Unidades de Alimentação e se constituem em uma das principais queixas de consumidores. O presente estudo procurou verificar quais os patógenos de importância para a indústria de alimentos encontrados em formigas coletadas em Unidades de Alimentação. As coletas foram feitas em meio de cultura Agar sangue e os isolamentos nos seguintes meios de cultura:Baird Parker para Staphylococcus, Sulfito de Bismuto para Salmonella e Agar MacConkey para enterobactérias. Os resultados demonstraram a presença de S. aureus e de enterobactérias provenientes de amostras de formigas coletadas em Unidades de Alimentação na região da Grande São Paulo, sugerindo que as formigas andarilhas podem ser importantes vetores de microrganismos de relevância e que interfiram na higidez dos alimentos. / Tramp ants have been scientific recognized due to the investigations conducted since 1970 when pathogens were for the first time encountered in ant samples collected from hospital environments. The surveys conducted since then state the presence of important microorganisms such as Salmonella, Staphylococcus, Klebsiella and Enterobacter. However, very little knowledge has been produced in regards to food preparation areas. Tramp ants have been frequently detected in houses, food preparation areas as well as in industrial and commercial restaurants and are considered as one of the most important pests nowadays. The present survey investigated the food industry important pathogens found in tramp species that were collected from industrial restaurants. The samples were obtained with agar blood plaque baits and were later incubated in several culture means: Baird Parker for Staphylococcus, Bismut Sulfit for Salmonella and MacKonkey for enterobacteria. The results show the presence of S. aureus and enterobacteria in the ant samples collected from the industrial restaurant environment in São Paulo, Brazil. These results indicate the tramp ants can be important mechanical vectors of relevant pathogens for the food industry.

L’errance est thématiquement présente dans le théâtre irlandais du début du XXe siècle. Le personnage errant est tout d’abord un moyen pour le dramaturge d’explorer et de dénoncer la perte ressentie par le peuple irlandais. Les vagabonds mis en scène expriment également le questionnement métaphysique de l’homme sur terre. Dans la Seconde moitié du siècle, l’errance infiltre la structure de la pièce, qu’il s’agisse de pièces irlandaises ou anglaises. Elle demeure, à travers le siècle, un thème de prédilection pour explorer les traumatismes, qu’ils soient économiques, politiques ou éthiques. La crise de valeurs du XXe siècle trouve écho sur des scènes théâtrales qui n’ont de cesse de questionner les certitudes, et qui mettent au jour la « déroute » du sens. L’ampleur et l’évolution de l’errance, à la fois thématique et structurelle, à travers des œuvres dramatiques étalées sur tout le XXe siècle, permettent de confirmer l’existence d’une dramaturgie de l’errance. Forte d’un siècle de remise en cause des certitudes, celle-ci se dessine à travers de nombreux exemples tirés des scènes anglaises et irlandaises du XXe siècle. / Thematically speaking, wandering and erring are particularly noticeable in early XXth century Irish drama. Playwrights stage vagrants, tramps, and Tinkers as a means to explore the loss felt by a large part of the Irish population throughout history. These characters also express metaphysical questions which will be echoed in the second half of the century. From being rather important themes, wandering and errancy gradually invade the very structure of the plays, be they Irish or British. Wandering has remained a major way of exploring trauma-related situations, be they economical, political or ethical. As the stage keeps questioning certainties, directions and sense are played with. The thematical and structural roles of wandering onstage highlight the widespread existence of a skeptical dramaturgy debunking trust and conviction.

Nestas atividades, os indivíduos precisam coordenar muitos componentes do corpo, mantendo relações entre eles e destes com o ambiente. Em um sistema dinâmico complexo, as relações entre as partes do sistema limitam ou influenciam o comportamento de outras partes, levando à emergência de padrões de coordenação. Crianças com Transtorno do Desenvolvimento da Coordenação (TDC) têm sido caracterizadas por apresentarem dificuldade em coordenar ritmicamente as partes do corpo em padrões específicos de coordenação intermembros. O presente estudo investigou a emergência e a estabilidade de padrões motores de crianças com TDC em uma tarefa rítmica de coordenação intermembros, analisando suas capacidades de sincronizar os deslocamentos dos membros superiores e inferiores em diferentes superfícies de apoio. Doze crianças com Desenvolvimento Típico (DT) e 12 crianças com TDC realizaram a tarefa de saltar e bater palma, durante 15 segundos, em duas superfícies: rígida (chão) e elástica (mini-trampolim). Foi solicitado à criança saltar e bater palma em quatro condições de coordenação: (i) de livre escolha do participante – Livre; (ii) batendo palma toda vez que os pés tocavam a superfície de apoio – Palma em Baixo; (iii) batendo palma toda vez que o corpo alcançava a máxima altura do salto – Palma em Cima; e (iv) batendo palma toda vez que os pés tocavam a superfície de apoio e toda vez que o corpo alcançava a máxima altura do salto – Duas Palmas. Quando a tarefa foi executada na condição Livre, os resultados mostraram que o padrão emergente das crianças com TDC foi diferente e mais variável na superfície mini-trampolim comparado com o das crianças com DT e com o delas mesmas na superfície chão. To perform these activities, people have to coordinate all body segments at the same time, keeping a relationship between segments and with the environment. In a complex dynamic system, the relationships between its parts limit or influence the behavior of other parts, so coordination patterns emerge. Children with Developmental Coordination Disorder (DCD) have been characterized by showing difficulty in rhythmically coordinate body segments, in specific interlimb coordination patterns. The purpose of this study was to investigate the emergency and stability of coordination patterns in children with DCD when they perform a rhythmic interlimb coordination task, analyzing their abilities to synchronize the movements of upper and lower limbs in two different surfaces. Twelve children with Typical Development (TD) and 12 children with DCD performed a jumping and clapping task during 15 seconds in two surfaces: rigid (on the floor) and elastic (on the mini-tramp). Children were required to clap while jumping (i) in their chosen pattern - Free; (ii) when the feet touched on the surface - Clapping-surface; (iii) when the body reached the maximum jumping height - Clapping-jumping; and (iv) when the feet touched the surface and when the body reached the maximum jumping height - Clapping-both. For the Free condition, the results showed that the DCD children emergent pattern was different and more variable on the mini-tramp than on the floor as compared with TD children coordination pattern. Furthermore, children with DCD were more variable in Clapping-surface and Clapping-jumping conditions compared with TD children. Visualisation des Arbres de Noël de Miller par immunoprécipitation de chromatine (ChIP) et mise en évidence d'un mécanisme de surveillance nucléolaire des ARN ribosomiques

Les terminal balls qui constituent des complexes de maturation sont détectées<p>à l’extrémité 5’-terminale des transcrits ribosomiques naissants dans tous les organismes<p>eucaryotes inspectés à ce jour ;générant les images de référence en « arbres de Noël ».<p>La compaction séquentielle des « terminal balls », à présent également dénommées<p>« SSU-processome », reflète les étapes d’assemblage co-transcriptionnel des ribosomes.<p>Au cours de ma thèse, j’ai développé une stratégie expérimentale basée sur<p>l’immunoprécipitation de chromatine (ChIP) qui m’a permis de valider, et ce pour la<p>première fois, in vivo la structure des branches des arbres de Noël (en particulier un<p>rapprochement du « SSU-processome » à l’extrémité 5’- du gène encodant l’ARNr 25S).<p>Notre stratégie nous permet également d’aborder la composition moléculaire des « arbres<p>de Noël ».<p>La biogenèse du ribosome est un processus complexe et dynamique dont la<p>finalité est la synthèse et l’assemblage de 4 molécules d’ARN et de ~80 protéines<p>ribosomiques dans un processus qui requiert l’intervention transitoire et concertée de non<p>moins de 400 facteurs de maturation. D’une telle complexité a récemment émergé le<p>concept de l’existence de modules pré-assemblés autonomes de facteurs de maturation.<p>Dans le cas du « SSU-processome », les trois sous-complexes UTP-A, UTP-B, UTP-C<p>ont d’ores et déjà été décrits. L’existence de tels sous-complexes renforce la notion d’un<p>mécanisme d’assemblage hautement hiérarchisé. En effet, il s’est avéré que l’extrémité<p>5’- du transcrit naissant est initialement liée par le sous-complexe UTP-A dans une étape<p>qui est un pré-requis indispensable au recrutement et à l’assemblage des autres<p>composants du « SSU-processome ». Avec autant d’étapes distinctes dans le processus<p>d’assemblage, la possibilité d’erreur est conséquente, d’où l’importance de l’existence de<p>mécanismes de contrôles de qualité.<p>Toutes les protéines constituant le « SSU-processome » sont requises au clivage<p>des précurseurs d’ARN ribosomique. Préalablement à mon travail, les 7 sous-unités<p>protéiques du complexe UTP-A avaient, en outre, spécifiquement été impliquées dans la<p>synthèse de l’ARN, c’est-à-dire dans la fonction de l’ARN polymérase I. Ceci leur a<p>conféré leur seconde appellation de tUTP, pour transcription UTP, et offert les prémices<p>de l’existence d’une interface physique et fonctionnelle entre les machineries de synthèse<p>et de maturation des ARNr. Au cours de ma thèse, j’ai démontré qu’il n’en est rien. Une<p>inspection minutieuse m’a en effet révélé que les tUTP/UTP-A ne sont nullement<p>requises à la synthèse des ARN ribosomiques mais bien à leur stabilité. Cette observation<p>m’a mené à proposer que la cellule a développé au cours de l’évolution un mécanisme de<p>contrôle de qualité par lequel elle s’assure de l’intégrité des étapes initiales d’assemblage<p>(liaison du complexe UTP-A ). Wanderers of Empire: The Tropical Tramp in Latin America, 1870-1930

U.S. public and private imperial interests confronted the problem of labor and labor power in the late nineteenth and early twentieth century as the U.S. empire expanded into Latin America and the Caribbean. The question of how to make an empire work spurred the creation of new labor regimes reliant on black West Indians who traveled to work in the Panama Canal Zone and on United Fruit Company (UFCO) banana plantations. Just as importantly, new labor regimes engendered new categories for troublesome laborers. One of these classifications, “tramp,” surfaced in the United States after the U.S. Civil War as a shorthand for vagrant, vagabond, and hobo. This thesis examines the so-called “tramp crisis” of the late nineteenth century to show how questions of labor invariably shaped problems of empire. As a category, the tramp moved outside of the United States where various U.S. foreign policymakers, writers, and business officials created the idea of the “tropical tramp” in U.S. imperial spaces. This label, tropical tramp, offers scholars a different starting point to analyze larger issues of whiteness, masculinity, sexuality, class, and the U.S. empire. By following discursive formations of the tramp and tropical tramp into Central and South America, this thesis argues that the figure of the tramp represented someone unbefitting the U.S. empire’s desired sociopolitical order.

Kostnadseffektivisering och kvalitetsförbättring gällande hantering av skärvätska

Projektet bestod i att ge underlag för lämpliga och användbara lösningar för effektivisering av Kockums Maskins skärvätskehantering och kostnadsrelaterade faktorer gällande företagets skärvätska. En grundlig undersökning av lämpligt material inom projektets huvudsakliga område gjordes till en början för att därefter resultera i en vidare analys av företagets nuvarande system och hantering av skärvätskor. Utifrån denna analys påbörjades en vidare undersökning om möjliga förslag om förbättring gällande de berörda problemen. Genom direktkontakt med flertalet företag och personer inom hantering/inköp samt befintlig dokumentation gällande skärvätska, erhölls ett flertal lösningar och förbättringsförslag som senare utvärderades och användes vid framtagning av det sökta projektresultatet. Problematiken gällande valet av lösningar har att göra med det faktum att det krävs omfattande kompetens inom varje typ av lösningsgrupp för att uppfylla de krav och förhållanden som finns hos Kockums Maskin. Därmed har avgränsningar gjorts gällande direkta val av lösningar för att istället ge förslag på lämpliga lösningar gällande förbättring av kvalitet, som indirekt bör sänka den årliga kostnaden av skärvätska, utifrån Kockums Maskins egentliga behov. / The project was to provide the basis for appropriate and useful solutions for any eventually streamlining of Kockums Maskin's cutting fluid management. Cost-related factors regarding the company's cutting fluid are also included in this part. A detailed examination of suitable material within the project's main area were initially and subsequently result in a further analysis of the company's existing systems and management, regarding cutting fluids. Based on this analysis a further investigation on possible proposals was started for improvement regarding the issues involved with the solutions and the cutting fluids. Through direct contact with many companies and individuals in management/purchasing and existing documentation regarding cutting fluid, obtained a several solutions and suggestions for improvement regarding the searched area. This result was then later evaluated and used in the development of the project results. The problem regarding the choice of solutions has to do with the fact that it requires extensive expertise in every type of area to meet the requirements and conditions that Kockums Maskin demands. Thus, the boundaries claimed the election of solutions to instead provide suggestions for suitable solutions regarding the improvement of quality, which indirectly should lower the annual cost of cutting fluid, based on Kockums Maskin's actual needs.
