La tomate, fruit largement consommé frais mais aussi sous forme transformée, est reconnue pour ses qualités nutritionnelles. Riche en microconstituants, tels les caroténoïdes (le lycopène en particulier), les composés phénoliques et la vitamine C, sa consommation régulière permettrait de réduire les risques de maladies cardiovasculaires et de certains cancers. Cependant, les traitements thermiques appliqués lors de la préparation industrielle de produits à base de tomate peuvent être à l'origine de réactions chimiques entraînant la dégradation de ces antioxydants. D'autre part, l'ajout d'huile végétale, pour la préparation de la sauce tomate notamment, met en jeu des réactions d'oxydation des lipides pouvant contribuer à l'instabilité de ces microconstituants. L’objectif principal de cette thèse est la détermination des cinétiques réactionnelles impliquées dans l’évolution des microconstituants de la tomate lors des procédés industriels de transformation.Dans un premier temps, une étude qualitative et quantitative sur plusieurs produits industriels (concentrés, pulpe, sauces) à base de tomate a été menée afin de déterminer les microconstituants marqueurs de la qualité nutritionnelle : (E)-lycopène et (E)-β-carotène pour les caroténoïdes, rutine, 7-O-glucosyl-naringénine et acide chlorogénique pour les composés phénoliques ainsi que les vitamines C et E ont été retenus. Cette première étude a permis de déterminer l’impact de procédés industriels sur les teneurs en microconstituants pour des produits collectés en entrée et en sortie de process pour deux usines de la région PACA.Par la suite, la stabilité des 7 marqueurs sélectionnés a été étudiée dans trois systèmes modèles modélisant la sauce tomate à des températures simulant des conditions de cold et hot breaks. Ces systèmes modèles comprennent un milieu aqueux acide (pH 3,8) contenant de l’amidon, un milieu lipidique constitué d'huile et de phospholipides végétaux et une émulsion H/E (huile dans eau) intégrant ces divers constituants. L’oxydation des lipides et son éventuelle protection par les microconstituants ont été évaluées par détermination de l'évolution des teneurs en diènes conjugués et trilinoléine. Parmi tous les antioxydants, la vitamine C s'est avérée être le moins stable en émulsion comme en milieu aqueux; sa vitesse d'oxydation est influencée par la teneur en oxygène dans le milieu. Les teneurs en composés phénoliques sont peu affectées, même à haute température (95 °C) tandis que les caroténoïdes s'isomérisent rapidement quelles que soient les conditions appliquées. Dans l'émulsion modèle, l'oxydation lipidique est nettement accélérée quand la température s'élève et tous les marqueurs se dégradent plus rapidement qu'en milieu aqueux ou lipidique.La détermination des constantes de vitesses a été réalisée, soit individuellement pour chaque cinétique, soit par desapproches globales (modèle oxydation lipidique et modèle stoechio-cinétique). Ce dernier modèle a pour but de proposer des conditions limitant l’oxydation lipidique et la dégradation des microconstituants de la tomate au cours des procédés / Tomato is a fruit widely consumed either fresh or processed possessing recognized nutritional qualities. Rich in microconstituents like carotenoids (such as lycopene), phenolic compounds and vitamin C, its regular consumption could reduce risks of cardio-vascular diseases and cancers. However, thermal treatments applied during industrial preparation of tomato products may involve various chemical reactions leading to the degradation of these antioxidants. Besides, addition of vegetable oil for the preparation of tomato sauce may lead to lipid oxidation contributing to the microconstituent instability.The main objective of this thesis is the determination of reaction kinetics for tomato microconstituents during industrial processing.First, a qualitative and quantitative study on several industrial tomato products (pastes, pulp, sauces) was carried out in order to select markers of the nutritional quality: (E)-lycopene and (E)-β-carotene for carotenoids, rutin, naringenin 7-O-glucoside and chlorogenic acid for phenolics as well as vitamins C and E. Additionally, an impact of processing was determined for industrial products collected before and after processing in two different plants in the PACA Region. Then, the stability of the selected markers was studied in model systems mimicking tomato sauce at hot and cold break temperatures. For this purpose, three model systems were developed: an aqueous model containing starch, a lipidic model consisting of vegetable oil and phospholipids and an O/W (oil-in-water) emulsion integrating all these constituents. Lipid oxidation and its possible protection by antioxidants were evaluated by the follow-up of conjugated dienes and trilinolein. Among all the antioxidants, vitamin C proved to be the least stable in both the emulsion and aqueous models. Its oxidation rate was found to be oxygen dependent. Phenolic compounds were little affected, even at high temperature (95 °C) whereas carotenoids rapidly isomerised whatever the conditions applied. In the emulsion model, lipid oxidation is clearly accelerated when the temperature increases and all markers are more rapidly degraded than in the aqueous or lipidic model. Rate constants were determined, either individually for each kinetics, either using global approaches (lipid oxidationmodel and stoichio-kinetic model). Homocystéinémie, apports en vitamines B et facteurs de risque cardiométabolique au Bénin, AfriqueEl Mabchour, Asma 01 1900 (has links)
L'hyperhomoscystéinémie (HHcy) est considérée comme un facteur indépendant de risque cardio-métabolique. Notre travail avait pour objectifs : 1) de déterminer la prévalence de l’HHcy au Bénin; 2) d’étudier sa relation avec les apports de vitamines B12, B9, B6 et B2, la consommation d’alcool, l’âge, le sexe et le niveau socioéconomique (NSE); 3) de vérifier son association avec les facteurs classiques de risque cardio-métabolique.
Un total de 541 sujets apparemment en santé et vivant dans trois zones du Bénin ont été étudiés. L’Hcy sérique a été analysée par ELISA. Des rappels de 24h ont servi à évaluer les apports nutritifs. L’obésité et l’hypertension ont été définies selon l’OMS, la dyslipidémie et la dysglycémie selon la NCEP-ATPIII. Les autres données ont été récoltées par questionnaire.
La prévalence de l’HHcy était élevée : 52,2% chez les hommes et de 24,7% chez les femmes. Dans les modèles multivariés, l’Hcy était positivement associée à la consommation de bière locale chez les hommes; chez les femmes, elle était associée négativement à l'apport de vitamine B12. L’Hcy était positivement associée à la tension artérielle, au taux de LDL-cholestérol et au cholestérol total chez les hommes, mais seulement avec le rapport cholestérol total/HDL-cholestérol (CT/HDL-c) chez les femmes. Les femmes présentant une HHcy étaient au moins deux fois plus susceptibles de présenter une hypertension ou un rapport CT/HDL-c élevé que celles dont l’Hcy était normale.
Un apport suffisant en B12 ainsi qu’une consommation prudente de boissons alcoolisées pourraient réduire l'HHcy et donc contribuer à réduire le risque cardio-métabolique de cette population du sud du Bénin. / Hyperhomocysteinemia (HHcy) appears to be an independent risk factor for cardiovascular disease. This study aims to determine the prevalence of HHcy in Benin, to explore its relationship with intakes of folate, B12, B6 and B2, with alcohol consumption and with socioeconomic status (SES) and to verify whether it is associated with classical risk factors of cardiovascular diseases.
541 apparently healthy subjects were randomly selected in tree areas of Benin. Hcy was measured in serum using ELISA commercial kits. Nutrient intakes were assessed on the basis of three non-consecutive 24-hour recalls. Alcohol consumption, socio-demographics and SES were documented in personal interviews. Obesity, hypertension, dyslipidemia and hyperglycaemia were defined according to WHO and NCEP-ATPIII.
HHcy (> 12 μmol/L) was detected in 52.2% of men and 24.7% of women. In multivariate models, Hcy in men was positively associated with alcohol intake, but only alcohol in beer. In women, Hcy was negatively related to vitamin B12 intake. HHcy was associated in women with more than twice the odds of hypertension and with the CT/HDL-c ratio. In men, Hcy was positively and independently associated with diastolic blood pressure and with LDL-cholesterol and total cholesterol.
In this Beninese population, the prevalence of HHcy is particularly high among men, and it appears to be related to alcohol consumption. Inadequate intake of vitamin B12 may be a risk factor for HHcy which could be related to some of cardiovascular factors.

Determinants of Prenatal Care and Supplement Use: The Case of HondurasHenze, Catherine E 01 January 2004 (has links)
Context: Literature suggests that prenatal care and prenatal supplement use improves pregnancy outcomes. However, we do not know the factors associated with prenatal care and supplement use in Honduras.Objective: To identify characteristics of Honduran women who are the least and most likely to use prenatal care and supplements.Methods: Data from a 2001 Honduras cross-sectional survey of women was used to assess their use of prenatal care and supplements. All data was weighted, resulting in a sample size of n = 5647 women who had a live birth since January 1996. Bivariate and multivariate analyses were used to examine factors associated with prenatal care and supplement use.Results: Current education level was highly positively related to prenatal care and supplement use. Women who were 35 years or older at the time of their most recent birth, currently unmarried, of non-Catholic religious affiliation, and of low SES were significantly less likely to have used prenatal care and supplements. Women who reported the intentionality of their most recent birth as unwanted also were significantly less likely to have used prenatal care and supplements. Prenatal care was the most significant determinant of prenatal supplement use.Conclusion: There are significant differences between Honduran women who use prenatal care and supplements and women who do not. Efforts to increase prenatal health services among underserved women, especially women who are older, unmarried, with no formal education, of low SES, of a non-Catholic religious affiliation, and at risk for an unwanted pregnancy, may significantly improve pregnancy outcomes in Honduras.

Comparação entre a biodisponibilidade do β-caroteno sintético e de fonte natural (couve-manteiga): papel de fibra alimentar em animais de laboratório / Comparison between the bioavailability of synthetic and natural source β-carotene (kale-butter): dietary fiber paper in laboratory animalsZanuto, Marcia Elena 24 November 2000 (has links)
Este trabalho, buscou comparar a biodisponibilidade do β-caroteno sintético e de fonte natural (couve-manteiga), e verificar os efeitos da fibra alimentar (pectina cítrica) sobre a biodisponibilidade do β-caroteno, dentro de 2 experimentos diferentes (ratos e coelhos). No Experimento I (ratos), um grupo de animais recebeu ração com β-caroteno e isenta de pectina cítrica (GC) e outro recebeu ração com β-caroteno e adição de pectina cítrica (GE). No Experimento II (coelhos), um grupo de animais recebeu ração com β-caroteno sintético (GCP), outro recebeu β-caroteno de fonte natural (couve-manteiga) (GV) e ainda outro não recebeu de β-caroteno (GCN); todos os grupos de coelhos receberam pectina cítrica. Após 30 dias de experimento, os animais foram sacrificados para determinações plasmáticas e hepáticas de vitamina A e β-caroteno, por HPLC. Dos resultados obtidos no estudo em ratos pode-se verificar que o grupo de animais que recebeu pectina (GE), apresentou menor (p<0,05) concentração hepática total de vitamina A (µg/peso de fígado) (retinol: GC =47,61 ± 24,24 e GE =23,44 ± 9,68 e palmitato de retinila: GC =935,30 ± 428,19 e GE =282,34 ± 98,86) e β-caroteno (µg/peso de fígado) (GC =9,64 ± 3,07 e GE =1,01 ± 0,66) que o grupo que não recebeu pectina (GC). E dos resultados obtidos no experimento em coelhos, verificou-se que o grupo que recebeu β-caroteno de fonte natural (GV), obteve concentração hepática total de vitamina A (mg/peso de fígado) (retinol: GV = 2,30 ± 0,67, GCP =2,07 ± 0,57 e GCN = 0,11 ± 0,08; palmitato de retinila: GV =4,42 ± 2,17, GCP =2,77 ± 0,73 e GCN =0,04 ± 0,02) e β-caroteno (mg/peso de fígado) (GV =0,04 ± 0,01, GCP =0,03 ± 0,01 e GCN =não detectado), maior (p<0,05) que o grupo que recebeu β-caroteno sintético (GCP). Conclui-se que no Experimento I (ratos), a pectina cítrica provavelmente interferiu na absorção do p-caroteno, e no Experimento II (coelhos), que o β-caroteno de fonte natural (GV), foi melhor absorvido comparando-se com o β-caroteno sintético, na presença de pectina cítrica. / This work, looked for to compare the bioavailability of synthetic p-carotene and of natural source (kale), and to verify the effects of the alimentary fiber (citrus pectin) on the bioavailability of β-carotene, inside of 2 different experiments (rats and rabbits). In the Experiment I (rats), a group of animals received diet with β-carotene and it exempts of citrus pectin (CG) and another received diet with β-carotene and addition of citrus pectin (EG). In the Experiment II (rabbits), a group of animals received diet with synthetic β-carotene (PCG), another received β-carotene of natural source (kale) (VG) and still another didn\'t receive from β-carotene (NCG); all the groups of rabbits received citrus pectin. After 30 days of experiment, the animals were sacrificed for determinations plasmatics and vitamin liverworts A and β-carotene, by HPLC. Of the results obtained in the study in rats it can be verified that the group of animals that received pectin (EG), it presented smaller (p <0,05) concentration hepatic vitamin total A (liver mg/weight) (retinol: CG = 47.61 ± 24.24 and EG = 23.44 ± 9.68 and retinyl palmitate: CG = 935.30 ± 428.19 and EG = 282.34 ± 98.86) and β-carotene (liver mg/weight) (CG = 9.64 ± 3.07 and EG = 1.01 ± 0.66) that the group that didn\'t receive pectin (CG). And of the results obtained in the experiment in rabbits, it was verified that the group that received β-carotene of natural source (VG), obtained concentration hepatic vitamin total A (liver mg/weight) (retinol: VG = 2.30 ± 0.67, PCG = 2.07 ± 0.57 and NCG = 0.11 ± 0.08; retinyl palmitate: VG = 4.42 ± 2.17, PCG = 2.77± 0.73 and NCG = 0.04 ± 0.02) and β-carotene (liver mg/weight) (VG = 0.04 ± 0.01, PCG = 0.03 ± 0.01 and NCG = not detected), larger (p <0,05) that the group that received synthetic β-carotene (PCG). Ended that in the Experiment I (rats), the citrus pectin probably interfered in the absorption of the p-carotene, and in the Experiment II (rabbits), the β-carotene of natural source (VG), it was absorbed being compared with the synthetic β-carotene better, in the presence of citrus pectin.

Análise da associação do consumo de frutas, legumes e verduras e de micronutrientes com marcadores de estado oxidativo, inflamatório e de resistência à insulina em indivíduos de risco cardiometabólico / Analysis of the association of fruits and vegetables and micronutrients intakes with markers of oxidative and inflammatory status and insulin resistance in individuals at cardiometabolic riskFolchetti, Luciana Dias 22 August 2012 (has links)
Introdução: O atual estilo de vida traz consequências negativas no que se refere aos padrões dietéticos, nível de atividade física (AF), uso de tabaco e estresse psicossocial, os quais predispõem ao aumento de doenças crônicas não-transmissíveis. Dieta rica em frutas, legumes e verduras (FLV) pode atenuar os efeitos sobre o risco cardiometabólico, havendo evidências consistentes de benefícios na prevenção da obesidade, dislipidemia e diabetes tipo 2. O consumo de FLV, fontes de vitaminas e minerais essenciais para a homeostase corporal está aquém do desejado. Estes alimentos contribuem para um perfil cardiometabólico favorável, atenuando o estresse oxidativo, inflamação e resistência à insulina. Objetivo: Este estudo analisou a associação entre o consumo FLV e de certos micronutrientes com marcadores do estado oxidativo, inflamatório e de resistência à insulina em indivíduos de risco cardiometabólico. Métodos:Nesta análise transversal foram incluídos 205 participantes (65 por cento mulheres; média de idade de 54,1 anos) do Estudo de Prevenção de Diabetes do CS-Escola da FSP-USP, com pré-diabetes ou de síndrome metabólica sem diabetes. Foram submetidos a questionários e coletas de sangue. Entre as dosagens, a superóxido dismutase (SOD) e a LDL oxidada (LDLox) serviram para indicar o estado anti/pró-oxidativo. O nível de AF foi medido pela versão longa do IPAQ. Três recordatórios alimentares de 24h foram empregados para cálculo da ingestão de micronutrientes e de FLV. Três categorias de consumo de FLV consumidas foram criadas considerando-se a recomendação internacional e a ingestão de micronutrientes estratificada segundo tercis de consumo, analisados por ANOVA. Coeficiente de Pearson e regressão linear múltipla foram também empregados. Resultados: Os participantes consumiram uma média de 1800 kcal/dia e 3,7 porções/1000 kcal de FLV. Ao longo das categorias de ingestão de FLV, os valores médios de circunferência da cintura (p=0,06) e pressão arterial diastólica (p=0,05) tenderam a diminuir e adiponectina (p=0,05) a aumentar. Indivíduos no tercil mais alto de ingestão de zinco apresentaram valores mais baixos de massa gorda e HOMA-IR, enquanto as concentrações de PCR foram marginalmente significantes (p=0,06). O HOMA-IR associou-se inversamente com ingestão de zinco e magnésio em todos os modelos. Associações diretas foram encontradas entre as concentrações de SOD com ingestão de FLV, bem como de magnésio em modelos ajustados. A concentração de LDLox se associou inversamente a ingestões de magnésio, vitaminas C e E em modelos ajustados. Resultados similares foram encontrados entre a concentração de LDLox e FLV, mas perdeu a significância após ajustes. Associação direta entre LDLox e ingestão de selênio manteve-se nos modelos ajustados. Conclusão: Nosso estudo sugere que a análise do consumo de FLV e/ou de certas vitaminas e minerais, ainda que estejam abaixo dos níveis recomendados, pode ser útil para identificar estresse oxidativo e resistência à insulina. / Introduction: Current lifestyle has deleterious consequences in dietary habits, physical activity, smoking and psychosocial stress, which have predisposed to non-communicable chronic diseases. A diet rich in fruits and vegetables can attenuate the effects on cardiometabolic risk, and there is consistent evidence of its benefit on the prevention of obesity, dyslipidemia and type 2 diabetes. Worldwide the consumption of fruits and vegetables, sources of vitamins and minerals, which are essential for body homeostasis, is low. These foods contribute to a favorable cardiometabolic profile, attenuating oxidative stress, inflammation and insulin resistance. Objective: Associations between the consumption of certain micronutrients and fruits and vegetables with markers of oxidative status, inflammation and insulin resistance were examined in individuals at cardiometabolic risk. Methods: This cross-sectional analysis included 205 participants (65 per cent women, mean age 54.1 years) with pre-diabetes or with metabolic syndrome without diabetes of the Diabetes Prevention Study of the FSP-USP Health Care Unit. They were submitted to questionaires blood collection. Superoxide dismutase (SOD) and oxidized LDL (oxLDL) were used to indicate the oxidative status. The physical activity level was measured by the long version of IPAQ. Three 24-hour dietary recalls were used to calculate intake of micronutrients and of fruits and vegetables. Three categories of fruits and vegetables consumption were created taking into consideration international recommendations, and micronutrient intake were grouped according tertiles of consumption, these categories analyzed by ANOVA. Pearsons correlation coefficient and multiple linear regression were used. Results: Participants consumed a mean of 1,800 kcal per day and 3.7/1000 kcal servings of fruit and vegetables. Mean values of waist circumference (p=0.06) and diastolic blood pressure (p=0.05) tended to decrease, and adiponectin (p=0.05) to increase across the categories of fruits and vegetables intake. Individuals in the highest tertile of zinc intake showed lower values of fat mass and HOMA-IR, while CRP concentrations were marginally significant (p=0.06). HOMA-IR was inversely associated with zinc and magnesium intakes in all the models. Direct associations were found between SOD concentrations and fruits and vegetables as well as magnesium intakes in adjusted models, while oxLDL concentration was inversely associated with magnesium, vitamin C and E intakes. Similar results were found between oxLDL concentration and fruits and vegetables intake but significance disappeared after adjustments. A direct association between oxLDL and selenium intake was detected after multiple adjustments. Conclusion: Our study suggest that fruits and vegetables and/or selected vitamins and minerals intakes albeit below recommended levels may be useful to identifying oxidative stress and insulin resistance.

Adubação e biofortificação: caraterização química e física do arroz (Oryza sativa L.) / Biofortification and fertilization: chemical and physical characterization of rice (Oryza sativa L.)Boliani, Evelise 11 April 2012 (has links)
O arroz (Oryza sativa L.) é um dos cereais mais produzidos e consumidos no mundo, caracterizando como principal alimento para mais da metade da população mundial. Sua importância é destacada principalmente em países em desenvolvimento, tais como o Brasil, desempenhando papel estratégico em níveis econômico e social. Atualmente, é crescente o número de pesquisas que integram desde o melhoramento vegetal e o manejo da adubação até o aproveitamento dos nutrientes pelo organismo humano. O melhoramento de plantas pode ajudar a melhorar a dieta humana, com o desenvolvimento de plantas com maiores teores de vitaminas e micronutrientes. A introdução de produtos agrícolas biofortificados como o arroz, complementará as intervenções em nutrição existentes. Sendo assim, tornase imprescindível conhecer como as condições de cultivo influenciam na qualidade do grão e na importância para obtenção de um alimento com maior valor biológico e nutricional e com menores concentrações de antinutricionais. O objetivo deste trabalho foi analisar as características físico-químicas e nutricionais do arroz (Oryza sativa L.) integral, promovidas por diferentes adubações (experimento I) e aos grãos biofortificados (experimento II). Foram realizadas análises de composição centesimal, composição mineral, diálise in vitro de minerais, vitaminas do complexo B, conteúdo de compostos fenólicos e fitatos em grão, além das análises de parâmetros de qualidade em arroz como conteúdo de amilose aparente, propriedade de pasta e análise instrumental de cor. Os resultados obtidos no estudo assemelham-se aos citados nas literaturas quanto á composição centesimal e mineral, enfatizando a influência da cocção na composição nutricional do grão e em seus compostos como fenólicos e fitatos. Para o dialisado de cálcio e zinco no arroz biofortificado, a variação nos valores foi em média de 1,14% a 3,26% para cálcio e 0,80% a 3,43% para zinco. No experimento II, as perdas de vitaminas para o arroz integral biofortificado após o cozimento foi em torno de 33,4%, 52,17% e 76% para vitamina B1, vitamina B2 e vitamina B6, respectivamente. Por fim, conclui-se que a produção de variedades melhoradas seja através de adubações ou da escolha de melhores cultivares, que apresentam maior teor de minerais e vitaminas, foram eficientes para a melhora da disponibilidade nutricional dos grãos. / Rice (Oryza sativa L.) is one of most cereals produced and consumed in the world, characterized as the primary food for more than half the world population. Its importance is highlighted especially in developing countries such as Brazil, playing a strategic role in economic and social levels. There is increasing the number of studies since that integrate plant breeding and fertilizer management to use the nutrients the human body. Plant breeding can help improve the human diet, with the development of plants with higher levels of vitamins and micronutrients. The introduction of biofortified crops like rice, will complement existing nutrition interventions. Thus, it becomes essential to know as the cultivation conditions influence the quality of grain and the importance to obtain a food with higher nutritional and biological value and with lower concentrations of antinutritional. Threfore, the aim of this study was to analyze the pshycochemical and nutritional properties of rice (Oryza sativa L.) in full, caused by different fertilization (experiment I) and subjected to biofortification (experiment II). Were performed to analyze chemical composition, mineral, dialysis in vitro of minerals, vitamins B, content of phenolic compounds and phytates in grain, in addition to the analysis of quality parameters in rice as apparent amylase content, folder property and instrumental analysis of color. The results obtained in this study are similar to those cited in other literature as to the mineral and chemical composition, emphasizing the influence of cooking on the nutritional composition of grain and as phenolic compounds and phytates. For the dialysate calcium and zinc in rice biofortified crops, the variation in the values averaged 1.14% to 3.26% for calcium and 0.80% to 3.43% for zinc. Losses of vitamins to the rice after cooking was around 33.4%, 52.17% and 76% for vitamin B1, vitamin B2 and vitamin B6, respectively. Finally, we conclude that the production of improved varieties is through fertilization or the best choice of cultivars, that have a higher content of minerals and vitamins, were effective to improve the availability of nutrients of the grains.

Neste trabalho, é proposto um método de separação e determinação das vitaminas hidrossolúveis utilizando-se eletroforese capilar na modalidade cromatografia eletrocinética micelar (MEKC). MEKC pode ser empregada para separar ambas moléculas carregadas ou neutras, individual ou simultaneamente. O método desenvolvido consistiu-se de 20 mmol L-1 de tetraborato de sódio (TBS), 20 mmol L-1 de dodecilsulfato de sódio (SDS) e metanol 15% (v/v). As soluções foram introduzidas no sistema eletroforético sob pressão de 0,5 psi por 3 s. A separação foi conduzida em um capilar de sílica fundida de 50 cm, sob potencial de 20 kV, à temperatura de 22ºC e monitorada em 200, 254 e 270 nm. O método analítico apresentou-se seletivo com separação em linha de base, preciso com áreas relativas no intervalo de 1,0 a 11%, apresentou baixos limites de detecção (entre 1,6 e 27,7 µmol L-1) e quantificação (entre 5,0 e 92,2 µmol L-1), além de ser simples e com alta frequência analítica, possibilitando separar as nove vitaminas hidrossolúveis em 15,50 min. Ele foi aplicado em amostra de farinha láctea, na qual foi possível determinar 4 vitaminas (B3, B5, B6 e C). Um método de separação para as vitaminas lipossolúveis, utilizando cromatografia a líquido, também foi proposto. Foi possível obter a separação em linha de base de sete vitaminas desta classe. A validação e a aplicação do método ainda devem ser investigadas. / In this paper, we propose a method for separation and determination of water-soluble vitamins using capillary electrophoresis method in micellar electrokinetic chromatography (MEKC). MEKC can be employed to separate both charged end neutral molecules, individually or simultaneously .The method consisted in 20 mmol L-1 sodium tetraborate (TBS), 20 mmol L-1 sodium dodecyl sulfate (SDS) and 15% methanol (v / v). The solutions were introduced into the electrophoretic system under pressure of 0.5 psi for 3 seconds. The separation was performed in a fused silica capillary of 50 cm under potential of 20 kV at a temperature of 22 ° C and monitored at 200, 254 and 270 nm. The analytical method was presented with selective separation line basis with precise relative areas in the range of 1.0 to 11%, showed low detection (1.6 to 27.7 µmol L-1) and quantification (from 5.0 to 92.2 µmol L-1) limits, besides being simple and high sampling rate, allowing to separate nine water-soluble vitamins in 15.50 min. The method was applied to a sample of milky flour, which it was possible to determine 4 vitamins (B3, B5, B6 e C). A method for separating soluble vitamins using liquid chromatography has also been proposed. It was possible to obtain a baseline separation of seven vitamins this class. The validation and application of the method must still be investigated.

Hiperhomocisteinemia e o risco cardiovascular / Hyperhomocysteinemia and cardiovascular riskVani, Gannabathula Sree 13 May 2002 (has links)
Nível elevado de homocisteína (Hcy) no plasma é considerado fator de risco de doença cardiovascular. Consumo reduzido de vitaminas B6, B12 e ácido fólico tem sido relacionado com hiperhomocisteinemia. O objetivo desse estudo foi verificar o consumo de vitaminas B6, B12 e ácido fólico nas populações urbana e rural, bem como a correlação dos níveis plasmáticos dessas vitaminas com os níveis plasmáticos de Hcy. Também determinamos os níveis séricos de lipídeos e avaliamos o risco cardiovascular das populações frente a hiperlipemia. O consumo de B6 e ácido fólico é maior na população urbana, com p=0,00 e p=0,04 respectivamente, sendo o consumo de B12 maior na população rural, com p=0,47. As correlações são significativamente negativa entre Hcy e as vitaminas B12 e ácido fólico . A população rural apresenta Hcy com valor médio de 16,5±9,2µmol/L, classificada como hiperhomocisteinemia moderada, e a população urbana 12,8±5,5 µmol/L, o qual está dentro da faixa de referência. O valor médio de LDL sérica é maior na população urbana (3,4±0,8mmoI/L) do que na população rural (2,8±0,9mmoI/L), com valor de p=0,00. Como fator de risco cardiovascular, consideramos Hcy plasmática >14µmol/L e LDL sérica >3,38mmol/L. Neste caso, 41,4% da população rural e 7,4% população urbana apresentam Hcy maior que 14µmol/L. O inverso ocorre em relação a LDL, onde 43,2% da população urbana e 11% na população rural apresentam níveis acima de 3,38mmol/L. Concluímos que o risco cardiovascular decorrente de hiperhomocisteinemia é maior na população rural que na urbana e este risco poderia reduzir mediante o consumo de vitaminas. / Elevated levels of plasma homocysteine (Hey) are considered a risk factor for cardiovascular diseases. Low intake of vitamins 86, 812 and folic acid have been related to hyperhomocysteinemia. The purpose of the present study is to determine the consumption of the vitamins B6, B12 and folic acid in two Brazilian urban and rural populations, along with the plasmatic levels of these vitamins and plasmatic homocysteine. In addition, the serum levels of lipids have been determined to evaluate the cardiovascular risk in the two populations regarding their hyperlipidemie comdition. The consumption of B6 and folic acid is higher in the urban population (p=0.00 and p=0.04 respective/y), while the consumption of B12 is not significantly different (p=0.47). There is a negative correlation between B12 and folic acid with Hcy. The rural population shows mean Hcy value of 16.5±9.2µmol/L and is classified as having moderate hyperhomocysteinemia, while for the urban population, the mean value is 12.8±5.5µmol/L and is well within the normal range. The mean value of the serum LDL is higher in the urban population (3.4±0.8mmol/L) compared to the rural population (2.8±0.9mmol/lL) with a significance of p=0.00. Plasma Hcy values >14µmol/L and serum LDL >3.38mmol/L were considered as the risk factors for cardiovascular disease. With in the reference values, 41.4% of the rural population and 7.4% of the urban population showa Hcy as a risk factor. For LDL, the inverse is true, i.e 43.2% of urban and 11% of the rural population are at risk. We conclude that the cardiovascular risk arising from hyperhomocysteinemia is higher in the rural population and that this can be reduced by increased consumption of vitamins.

Introdução: O atual estilo de vida traz consequências negativas no que se refere aos padrões dietéticos, nível de atividade física (AF), uso de tabaco e estresse psicossocial, os quais predispõem ao aumento de doenças crônicas não-transmissíveis. Dieta rica em frutas, legumes e verduras (FLV) pode atenuar os efeitos sobre o risco cardiometabólico, havendo evidências consistentes de benefícios na prevenção da obesidade, dislipidemia e diabetes tipo 2. O consumo de FLV, fontes de vitaminas e minerais essenciais para a homeostase corporal está aquém do desejado. Estes alimentos contribuem para um perfil cardiometabólico favorável, atenuando o estresse oxidativo, inflamação e resistência à insulina. Objetivo: Este estudo analisou a associação entre o consumo FLV e de certos micronutrientes com marcadores do estado oxidativo, inflamatório e de resistência à insulina em indivíduos de risco cardiometabólico. Métodos:Nesta análise transversal foram incluídos 205 participantes (65 por cento mulheres; média de idade de 54,1 anos) do Estudo de Prevenção de Diabetes do CS-Escola da FSP-USP, com pré-diabetes ou de síndrome metabólica sem diabetes. Foram submetidos a questionários e coletas de sangue. Entre as dosagens, a superóxido dismutase (SOD) e a LDL oxidada (LDLox) serviram para indicar o estado anti/pró-oxidativo. O nível de AF foi medido pela versão longa do IPAQ. Três recordatórios alimentares de 24h foram empregados para cálculo da ingestão de micronutrientes e de FLV. Três categorias de consumo de FLV consumidas foram criadas considerando-se a recomendação internacional e a ingestão de micronutrientes estratificada segundo tercis de consumo, analisados por ANOVA. Coeficiente de Pearson e regressão linear múltipla foram também empregados. Resultados: Os participantes consumiram uma média de 1800 kcal/dia e 3,7 porções/1000 kcal de FLV. Ao longo das categorias de ingestão de FLV, os valores médios de circunferência da cintura (p=0,06) e pressão arterial diastólica (p=0,05) tenderam a diminuir e adiponectina (p=0,05) a aumentar. Indivíduos no tercil mais alto de ingestão de zinco apresentaram valores mais baixos de massa gorda e HOMA-IR, enquanto as concentrações de PCR foram marginalmente significantes (p=0,06). O HOMA-IR associou-se inversamente com ingestão de zinco e magnésio em todos os modelos. Associações diretas foram encontradas entre as concentrações de SOD com ingestão de FLV, bem como de magnésio em modelos ajustados. A concentração de LDLox se associou inversamente a ingestões de magnésio, vitaminas C e E em modelos ajustados. Resultados similares foram encontrados entre a concentração de LDLox e FLV, mas perdeu a significância após ajustes. Associação direta entre LDLox e ingestão de selênio manteve-se nos modelos ajustados. Conclusão: Nosso estudo sugere que a análise do consumo de FLV e/ou de certas vitaminas e minerais, ainda que estejam abaixo dos níveis recomendados, pode ser útil para identificar estresse oxidativo e resistência à insulina. / Introduction: Current lifestyle has deleterious consequences in dietary habits, physical activity, smoking and psychosocial stress, which have predisposed to non-communicable chronic diseases. A diet rich in fruits and vegetables can attenuate the effects on cardiometabolic risk, and there is consistent evidence of its benefit on the prevention of obesity, dyslipidemia and type 2 diabetes. Worldwide the consumption of fruits and vegetables, sources of vitamins and minerals, which are essential for body homeostasis, is low. These foods contribute to a favorable cardiometabolic profile, attenuating oxidative stress, inflammation and insulin resistance. Objective: Associations between the consumption of certain micronutrients and fruits and vegetables with markers of oxidative status, inflammation and insulin resistance were examined in individuals at cardiometabolic risk. Methods: This cross-sectional analysis included 205 participants (65 per cent women, mean age 54.1 years) with pre-diabetes or with metabolic syndrome without diabetes of the Diabetes Prevention Study of the FSP-USP Health Care Unit. They were submitted to questionaires blood collection. Superoxide dismutase (SOD) and oxidized LDL (oxLDL) were used to indicate the oxidative status. The physical activity level was measured by the long version of IPAQ. Three 24-hour dietary recalls were used to calculate intake of micronutrients and of fruits and vegetables. Three categories of fruits and vegetables consumption were created taking into consideration international recommendations, and micronutrient intake were grouped according tertiles of consumption, these categories analyzed by ANOVA. Pearsons correlation coefficient and multiple linear regression were used. Results: Participants consumed a mean of 1,800 kcal per day and 3.7/1000 kcal servings of fruit and vegetables. Mean values of waist circumference (p=0.06) and diastolic blood pressure (p=0.05) tended to decrease, and adiponectin (p=0.05) to increase across the categories of fruits and vegetables intake. Individuals in the highest tertile of zinc intake showed lower values of fat mass and HOMA-IR, while CRP concentrations were marginally significant (p=0.06). HOMA-IR was inversely associated with zinc and magnesium intakes in all the models. Direct associations were found between SOD concentrations and fruits and vegetables as well as magnesium intakes in adjusted models, while oxLDL concentration was inversely associated with magnesium, vitamin C and E intakes. Similar results were found between oxLDL concentration and fruits and vegetables intake but significance disappeared after adjustments. A direct association between oxLDL and selenium intake was detected after multiple adjustments. Conclusion: Our study suggest that fruits and vegetables and/or selected vitamins and minerals intakes albeit below recommended levels may be useful to identifying oxidative stress and insulin resistance.

An Examination of the Population Health Implications of Voluntary Food Fortification and Nutrition-related Marketing Practices in Canada.Sacco, Jocelyn 19 December 2012 (has links)
The 2004 Canadian Community Health Survey (CCHS) revealed many indicators of poor diet quality in Canada which, together with the high rates of obesity and diet-related chronic disease, suggest that shifts in dietary patterns are urgently needed. Given the widespread promotion of foods on the basis of nutrition and health, the aim of this work was to explore the population health implications of voluntary food fortification and nutrition-related marketing in Canada.
Using the CCHS, the potential impact of a proposed discretionary food fortification policy on nutrient inadequacies and excesses was examined, in addition to the relationship between consumption of foods eligible to be fortified under this policy and indicators of dietary quality. To better understand the potential risk associated with liberal fortification practices, the National Health and Nutrition Examination Survey (2007-08) was used to examine potential for risk of excess associated with voluntarily fortified food consumption in the US, where these practices have long been permitted. The results suggest that proposed changes to voluntary fortification may reduce inadequacy and increase excess, and may reinforce poor diet patterns. Excessive nutrient intakes were also found to be associated with consumption of voluntarily fortified foods in the US, particularly among children. Therefore, there appears to be real potential for risk associated with voluntary fortification practices in Canada.
The extent, nature, and population health implications of nutrition marketing in Canada was examined, using a survey of front-of-package nutrition-related marketing on foods within three large grocery stores in Toronto. Nutrition-related marketing was found on 41% of all foods surveyed, and was widely found on highly processed, often fortified foods. References to nutrients of public health concern (e.g. sodium, vitamin D) were infrequently found. Overall, this practice provides limited nutritional guidance.
Current directions in nutrition policy in Canada should be re-evaluated, to ensure that they support healthy diet patterns.
