EFEITO DO Harpagophytum procumbens SOBRE PARÂMETROS DE ESTRESSE OXIDATIVO E A VIABILIDADE CELULAR IN VITRO / EFFECT OF Harpagophytum procumbens ON OXIDATIVE STRESS PARAMETERS AND CELL VIABILITY IN VITRO

Schaffer, Larissa Finger 04 October 2013 (has links)
Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Harpagophytum procumbens, known as devil's claw, is an African plant with numerous therapeutic effects, among which stands out for its powerful anti-inflammatory action, but the mechanism by which exerts its effect is not well elucidated. It is known that reactive oxygen species and / or nitrogen are generated in acute and chronic inflammatory diseases causing high cytotoxicity that leads to tissue damage. Substances with antioxidant action can contribute to control and/or treat the inflammatory process. Thus, the goal of this study was to evaluate the effect of the infusion, the crude extract and fractions of H. procumbens on lipid peroxidation induced by different pro-oxidant agents (Iron or sodium nitroprusside). Furthermore, the effects of the ethyl acetate fraction was also tested on the antioxidant defenses (catalase activity and thiol protein and non-protein levels) and on cellular damage (cerebral cortex slices) induced by different pro-oxidant agents. All extracts prevented lipid peroxidation induced by Fe2+ and sodium nitroprusside. Ethyl acetate fraction of H. procumbens showed great power to protect against lipid peroxidation, prevention of cellular death, and shows ability to maintain the thiol levels and catalase activity in basal standarts. In conclusion, this study demonstrates that one of the possible mechanisms for therapeutic action presented by H. procumbens may be related to its antioxidant action, being was able to protect against oxidative stress and loss of cell viability. Once the relationship of inflammation process with oxidative stress is already well described in the literature, the antioxidant effect can also be found here related to potent anti-inflammatory effect exerted by H. procumbens. However more studies are needed on the mechanisms by which H. procumbens exerts its therapeutic effects.

The study showed an economic return to the producer of 22% compared to a control area without Precision Agriculture. It was expected an increase in the potassium concentration in the resampling but not found, raising the possibility of doing a maintenance fertilization on crop instead of making a soil amendment.

EFICIÊNCIA DE GRADES AMOSTRAIS E ZONAS DE MANEJO NA ACERTIVIDADE DA CORREÇÃO E FERTILIZAÇÃO EM ÁREA DE AGRICULTURA DE PRECISÃO CONSOLIDADA / EFFICIENCY OF SAMPLE GRIDS AND MANAGEMENT ZONES IN THE ASSERTIVENESS OF CORRECTION AND FERTILIZATION IN CONSOLIDATED PRECISION AGRICULTURE AREA

Pias, Osmar Henrique de Castro 17 February 2016 (has links)
Coordenação de Aperfeiçoamento de Pessoal de Nível Superior / Soil samples from sampling grids is currently the leading precision farming tool used in Brazil. But it still has doubts about the economic feasibility of using grids with high density points. New technologies such as the use of management zones (MZ) to guide soil samples have been studied in order to reduce the number of samples and consequently the sampling costs while maintaining the quality of the information obtained. However, this method lacks scientific information about its efficiency. The aim of this study was to evaluate the technical efficiency (chapter I) mapping chemical attributes of soil and economic viability (chapter II) of the recommendations of fertilizers and from the parameters obtained by conventional sampling, sample grids and MZ delimited by different methodologies. The study was carried out in Boa Vista Missões - RS, on an area of 118,9 ha. The soil sampling was carried out in 2015, using a sample grid of 0.5 ha. From the systematic elimination of sample points simulated the use grids; 1, 2, 3 and 4.5 ha. The MZ were defined based on data altimetry of the area and the yield maps. To Chapter I used the data of P, K and pH which were submitted to descriptive statistical analysis. Data collected in sampling grid was submitted to geostatistical analysis. The accuracy of thematic maps was evaluated from the Pearson correlation and coefficient of relative deviation. For chapter II we used the P, K and SMP index to generate recommendations of P2O5, K2O and lime, respectively. The recommendation of errors were calculated in relation to the reference grid (0.5 ha) in application area (ha) kg of fertilizer and in real (R$), later smpling costs were compared with the total cost of errors recommendation. The increase in the size of the sampling grid resulted in loss of information on the spatial variability of attributes and concenquentemente raised the deviations in both fertilizer and corrective recommendations. For a proper assessment of the spatial variability of soil chemical attributes and an accurate prescription of inputs recommends the use of sampling grid of 0.5 ha.

In vitro kultivace motolice Trichobilharzia regenti / In vitro cultivation of the trematode species Trichobilharzia regenti

Vrbová, Kristýna January 2017 (has links)
The class Trematoda includes many pathogenic representatives. Main subject of this thesis, avian schistosome Trichobilharzia regenti, is a close relative to the important human pathogen Schistosoma mansoni (family Schistosomatidae). In vitro cultivation of trematodes enables closer understanding of their biology and parasite- host interactions; however, no trematode species has been successfully kept in vitro from the egg stage to the adults producing eggs. Many studies are focused on the problematic of S. mansoni cultivation, but data concerning T. regenti cultivation remain scarce. Only the ability of T. regenti cercariae to transform into schistosomula in vitro was documented, with following survival in a culture medium for a few days. Comparison of eight transformation methods was performed with T. regenti cercariae. Based on the number of tailless cercarial bodies obtained, five transformation methods were selected for further evaluation of the early schistosomula characteristics (glycocalyx shedding, penetration glands emptying and survival in vitro). It was observed that the largest quantity of cercarial bodies can be obtained by using a syringe needle or the BeadBeater cell disrupter. The largest quantity of schistosomula meeting the criteria of early schistosomulum was recorded after...

Population genetics and population ecology in management of endangered species

Hens, H. (Hilde) 24 May 2017 (has links)
Abstract Knowledge of the determinants of the viability of populations is essential in order to undertake effective conservation and management of endangered species. In this study, long-term demographic data was combined with genetic data to study the viability of an endangered orchid species, Epipactis atrorubens. The genetic analyses revealed low levels of genetic variation and the presence of population genetic differentiation independent of the spatial scale. Low levels of seed-mediated gene flow, possibly linked to low seedling recruitment, is the likely cause of the low levels of gene flow. Indications of slow post-glacial colonisation rates were found, which together with the low gene flow predict a limited capacity of the species to shift its range to more suitable habitats after environmental change. Low genetic variation as a proxy for low evolutionary potential also suggests that the species has limited capacity to adapt to new environmental conditions. Furthermore, poor seedling recruitment lowers population viability in small populations, as highlighted by the low population growth rates. In addition, we found a strong effect of stochasticity that limits the viability of populations. Both the genetic and demographic analyses indicated low viability of the studied species and that seedling recruitment could be the main determinant for the viability. / Tiivistelmä Luonnonsuojelun perusta on populaatioiden elinkykyyn vaikuttavien tekijöiden tuntemus.

Population biology of the <em>Primula sibirica</em> group species inhabiting frequently disturbed seashore meadows: implications for management

Rautiainen, P. (Pirjo) 29 March 2006 (has links)
Abstract Many plant species inhabiting the seashore meadows of the Bothnian Bay, especially early successional ones, have become threatened. Isostatic land uplift creates virgin land for early successional species to colonise. However, at the same time it gradually elevates the habitat and eventually makes the habitat unsuitable for them. Disturbances of the waterfront may slow down succession and create new empty sites. In order to persist on the shores, pioneer species have to be able to colonise new sites by seeds, vegetative propagules or growth. In this thesis I studied the status of an endangered early successional grass species, A. fulva var. pendulina, at the Liminka Bay. According to a matrix population model based on eight years of observations (1992–1999), the population seemed not to be in immediate danger of extinction. However, simulations based on four-year field observations (2000–2003) indicated that if the current trend continues, the species will decrease considerably in area in the next 30 years. In the field studies no seedlings or viable seeds of A. fulva were found. In spite of this, high genotypic diversity was found in the A. fulva population, suggesting that sexual reproduction has taken place at some time during the history of the population. Analysis of the population structure revealed a low level of genotypic differentiation between subpopulations and significant sub-structuring within subpopulations. The overall pattern of genetic variation suggests that the population has characters of both stepping-stone and metapopulation models. The results of the study on the ability of a seashore plant Potentilla anserina ssp. egedii to change its allocation of resources to sexual and vegetative reproduction according to competitive stress implied that the species can modify the allocation of resources to different life-history traits. For a plant living in disturbance-prone environment, it may be beneficial to be able to rapidly track the competition-free space formed by disturbances by changing its reproductive pattern. Management studies on three endangered seashore plant species showed that deterioration of suitable habitats of A. fulva and Primula nutans var. jokelae could be slowed down by management, and the vegetative and/or sexual reproduction of these species was enhanced. However, in the case of Puccinellia phryganodes, no positive response to management was observed.

Wnt-11 signalling, its role in cardiogenesis and identification of Wnt/β-catenin pathway target genes

Railo, A. (Antti) 30 March 2010 (has links)
Abstract Wnt genes encode secreted signalling molecules that control embryonic development including organogenesis, while dysregulated Wnt signalling is connected to many diseases such as cancer. Specifically, Wnts control a number of cellular processes such as proliferation, adhesion, differentiation and aging. Many Wnt proteins activate the canonical β-catenin signalling pathway that regulates transcription of a still poorly characterized set of target genes. Wnts also transduce their signaling in cells via β-catenin-independent “non-canonical” pathways, which are not well understood. In this study, Wnt-11 signalling mechanisms in a mammalian model cell line and roles of Wnt-11 in heart development were analyzed in detail. In addition the aim was to identify new Wnt target genes by direct chromatin immunoprecipitation and Affymetrix GeneChip assays in the model cells exposed to Wnt-3a. Our studies reveal that Wnt-11 signalling coordinates the activity of key cell signalling pathways, namely the canonical Wnt/β-catenin, the JNK/AP-1, the NF-κB and PI3K/Akt pathways in the CHO cells. Analysis of the Wnt-11-deficient embryos revealed a crucial role in heart organogenesis. Wnt-11 signalling coordinates cell interactions during assembly of the myocardial wall and Wnt-11 localizes the expression of N-cadherin and β-catenin to specific cellular domains in the embryonic ventricular cardiomyocytes. Collectively these studies reveal that the mammalian Wnt-11 behaves as a non-canonical Wnt and that it is a critical factor in the coordination of heart development. Specifically, it controls components of the cell adhesion machinery. Analysis of the Wnt target genes revealed a highly context-dependent profile in the Wnt-regulated genes. Several new putative target genes were discovered. Out of the candidate Wnt target genes, Disabled-2 was identified as a potential new direct target for Wnt signalling.

The Effects of Fire on Spore Viability of Lygodium microphyllum (Old World Climbing Fern)

Sebesta, Nicole 02 July 2015 (has links)
Lygodium microphyllum, native to the Old World tropics, has invaded central and southern Florida, destroying native habitats, reducing biodiversity and altering fire regimes. Prescribed fire, one of several methods used to manage L. microphyllum infestations, reduces fern biomass over large areas, but its effects on spore viability are unknown. To provide tools to evaluate whether fire-dispersed spores are viable, this research determined how heat affects spore viability. Spores were exposed to temperatures of 50°C to 300°C for durations of 5 seconds to 1 hour, then allowed to germinate on agar in petri plates. Percent germination was assayed after two weeks. Temperatures of 50°C had little effect; 300°C killed spores for all durations. Results indicate that while viability of unburnt spores decreases with increasing temperature and duration of heat exposure, spores are killed when exposed to relatively low temperatures compared to those in fires.

Minority Governments in Canada: A Study of Legislative Politics

Gervais, Marc January 2011 (has links)
Despite their prevalence, the study of Canadian minority governments has been the object of few published studies. In particular, the issue of how governments that must rely on the support of one or more opposition parties in Parliament manage to remain in power (viability) and pass their legislative proposals (effectiveness) has not been thoroughly investigated. This study examines the parliamentary dynamics at play in these situations by applying a majority building framework grounded in and supported by three theoretical perspectives, namely the rational choice tradition, new institutionalism, and the role of party politics and party systems, to four minority governments that have occurred in the last 50 years or so: 1- Diefenbaker (1957-1958), 2- Pearson (1963-1965); 3- Clark (1979-1980); and, 4- Harper (2006-2008). The data on the specific circumstances that held during these minority governments has been gathered from archival records, from the recorded debates and votes in the House of Commons, from previous Canadian studies on minority government, from political autobiographies, and from third party accounts of the events at the time. The study finds that majority building is a function of primarily two interrelated variables: 1- bargaining power (interparty dynamics and intra-party cohesion) and 2- agenda control (House business, confidence tests, other institutional features). It also stresses the importance of government concessions as an effective means of achieving desired goals and outcomes. Furthermore, this study highlights the capacity and skill of individual parliamentary actors in the exercise of legislative politics generally and in manipulating institutional and party system levers specifically, as a contributing factor to their government’s duration and legislative output. This study adds to the empirical knowledge of the minority experience in Canada and provides a conceptual framework to better understand legislative politics and its impact on the success of minority governments in Canada and elsewhere.
