Comparative evaluation of different extenders of bull semen stored under different conditionsRaseona, Andrea Motswetla 16 July 2015 (has links)
MSCAGR / Department of Animal Science / Preservation of semen is an important process to ensure that semen quality is sufficient for use in assisted reproductive technologies. This study evaluated the effectiveness of three different extenders to preserve bull semen stored under different conditions, as an alternative to frozen-thawed semen straws used for artificial insemination. Semen samples were collected from two Nguni bulls using an electro-ejaculator and transported to the laboratory at 37 °C for evaluation. Pooled semen was first aliquoted into three extenders namely Triladyl, Ham’s F10 and M199 at a dilution ratio of 1:4 (semen:extender), and then stored at controlled room temperature 24 °C. Secondly pooled semen was aliquoted into four groups of Ham’s F10 extender and diluted at a ratio of 1:4, then stored at 24 °C, 17 °C, 12 °C and 5 °C respectively. Sperm motility rates were analysed after 0, 24, 48 and 72 hours. Morphology, viability and sperm DNA fragmentation were analysed after 72 hours. The study was replicated four times and data was analysed by ANOVA. Triladyl had higher sperm viability rate and total motility rate for 72 hours (P<0.01). However, Ham’s F10 had higher progressive motility rate as compared to the other extenders. There was no significant difference (P<0.01), in the viability rate between Ham’s F10 and M199. No significant difference was also observed in total sperm abnormalities (absent tails, coiled tails and bent tails), except for reacted acrosomes (P>0.05), between the two Nguni bulls. Lower temperatures than 24 °C influenced sperm motility and viability in Ham’s F10. There was no significant difference in sperm DNA fragmentation rates (P<0.01), between all the four storage temperatures which indicated that temperature did not have an influence on sperm DNA fragmentation. In conclusion, bull semen can be preserved in Triladyl or Ham’s F10 and M199 culture media stored at 24 °C and stay alive for 72 hours. Triladyl proved to be the best suitable extender showing higher sperm viability and total motility rates as compared to Ham’s F10 and M199. Lower temperatures than 24 °C noticeably decreased sperm motility and viability in Ham’s F10 culture medium.

Department of African Studies / PhD (African Studies) / One of the contemporary themes which dominated discourse in the education fraternity is the integration of IKS in the curriculum of South African education system. There is a growing call for the recognition of contribution by locals and indigenous societies in making education relevant to African children. The aim of the study was to explore the viability of integrating indigenous knowledge into Life Orientation (LO) Curriculum in the intermediate phase. Objectives were to explore Indigenous Knowledge that could be integrated into LO Curriculum in the intermediate phase and to generate knowledge exchange platforms where community and other stakeholders could be involved in the discussion about Indigenous Knowledge integration. A qualitative method and an exploratory-interpretive approach were adopted in the study. Data were gathered through focus group discussions. Data analysis and field notes were clustered thematically. The study indicated that, to integrate Indigenous Knowledge into LO effectively, it could require a shift from the western knowledge focused curriculum taking into cognisance the knowledge of the local inhabitants. The study showed that the integration of Indigenous Knowledge in the South African school curriculum in LO will bring new life into the content and the delivery of tuition. Although there were opportunities to integrate indigenous knowledge in the curriculum, educators felt that the curriculum does not create sufficient space, time and training for the integration of indigenous knowledge. Some of the reasons put forward were that there was inadequate content knowledge on Indigenous Knowledge integration, inadequate support and insufficient allocation of resources. Without a deliberate educator capacity development focusing on indigenous knowledge systems, the viability of IKS integration remains questionable. This is exacerbated by the growing discouragement of Indigenous Knowledge amongst the parents who also felt that there is no practical relevance for IKS in the current education trajectory. Vigorous efforts need to be executed to fast track the process of integrating Indigenous Knowledge in teaching LO in the intermediate phase.

Magnetic nanoparticles have received extensive attention in the biomedical research, e.g. as prospective contrast agents for T2-weighted magnetic resonance imaging. The ability of a contrast agent to enhance the relaxation rate of 1 H in its vicinity is quantified by relaxivity. The main aim of this thesis is to evaluate the transversal re- laxivity of ε-Fe2−x Alx O3 nanoparticles coated with amorphous silica or citrate - its dependence on external magnetic field, temperature and thickness of silica coating - by means of nuclear magnetic resonance. The aluminium content x = 0.23(1) was determined from XRF, the material was further characterised by XRPD, Möss- bauer spectroscopy, DLS, TEM and magnetic measurements. The size of magnetic cores was ∼ 21 nm, the thickness of silica coating ∼ 6,10,17 and 21 nm. Magne- tization of the ε-Fe2−x Alx O3 nanoparticles increased by ∼ 30 % when compared to ε-Fe2O3. The saturating dependence of relaxivity on external magnetic field and on the linear decrease with increase of thickness of silica coating contravene the theo- retical model of motional averaging regime (MAR); nevertheless, the temperature dependence acquired in 0.47 T and 11.75 T may be explained by MAR. La théorie de la viabilité propose des outils permettant de contrôler un système dynamique afin de le maintenir dans un domaine de contraintes. Le concept central de cette théorie est le noyau de viabilité, qui est l’ensemble des états initiaux à partir desquels il existe au moins une trajectoire contrôlée restant dans le domaine de contraintes. Cependant, le temps et l’espace nécessaires au calcul du noyau de viabilité augmentent exponentiellement avec le nombre de dimensions du problème considéré. C’est la malédiction de la dimension. Elle est d’autant plus présente dans le cas de systèmes incorporant des incertitudes. Dans ce cas-là, le noyau de viabilité devient l’ensemble des états pour lesquels il existe une stratégie de contrôle permettant de rester dans le domaine de contraintes avec au moins une certaine probabilité jusqu’à l’horizon de temps donné. L’objectif de cette thèse est d’étudier et de développer des approches afin de combattre cette malédiction de la dimension. Pour ce faire, nous avons proposé deux axes de recherche : la parallélisation des calculs et l’utilisation de la théorie de la fiabilité. Les résultats sont illustrés par plusieurs applications. Le premier axe explore l’utilisation de calcul parallèle sur carte graphique. La version du programme utilisant la carte graphique est jusqu’à 20 fois plus rapide que la version séquentielle, traitant des problèmes jusqu’en dimension 7. Outre ces gains en temps de calcul, nos travaux montrent que la majeure partie des ressources est utilisée pour le calcul des probabilités de transition du système. Cette observation fait le lien avec le deuxième axe de recherche qui propose un algorithme calculant une approximation de noyaux de viabilité stochastiques utilisant des méthodes fiabilistes calculant les probabilités de transition. L’espace-mémoire requis par cet algorithme est une fonction linéaire du nombre d’états de la grille utilisée, contrairement à l’espace-mémoire requis par l’algorithme de programmation dynamique classique qui dépend quadratiquement du nombre d’états. Ces approches permettent d’envisager l’application de la théorie de la viabilité à des systèmes de plus grande dimension. Ainsi nous l’avons appliquée à un modèle de dynamique du phosphore dans le cadre de la gestion de l’eutrophisation des lacs, préalablement calibré sur les données du lac du Bourget. De plus, les liens entre fiabilité et viabilité sont mis en valeur avec une application du calcul de noyau de viabilité stochastique, autrement appelé noyau de fiabilité, en conception fiable dans le cas d’une poutre corrodée. / Viability theory provides tools to maintain a dynamical system in a constraint domain. The main concept of this theory is the viability kernel, which is the set of initial states from which there is at least one controlled trajectory remaining in the constraint domain. However, the time and space needed to calculate the viability kernel increases exponentially with the number of dimensions of the problem. This issue is known as “the curse of dimensionality”. This curse is even more present when applying the viability theory to uncertain systems. In this case, the viability kernel is the set of states for which there is at least a control strategy to stay in the constraint domain with some probability until the time horizon. The objective of this thesis is to study and develop approaches to beat back the curse of dimensionality. We propose two lines of research: the parallel computing and the use of reliability theory tools. The results are illustrated by several applications. The first line explores the use of parallel computing on graphics card. The version of the program using the graphics card is up to 20 times faster than the sequential version, dealing with problems until dimension 7. In addition to the gains in calculation time, our work shows that the majority of the resources is used to the calculation of transition probabilities. This observation makes the link with the second line of research which proposes an algorithm calculating a stochastic approximation of viability kernels by using reliability methods in order to compute the transition probabilities. The memory space required by this algorithm is a linear function of the number of states of the grid, unlike the memory space required by conventional dynamic programming algorithm which quadratically depends on the number of states. These approaches may enable the use of the viability theory in the case of high-dimension systems. So we applied it to a phosphorus dynamics for the management of Lake Bourget eutrophication, previously calibrated from experimental data. In addition the relationship between reliability and viability is highlighted with an application of stochastic viability kernel computation, otherwise known as reliability kernel, in reliable design in the case of a corroded beam.

In Vitro Evaluation of Novel N-Acetylalaninate Prodrugs That Selectively Induce Apoptosis in Prostate Cancer CellsMcGoldrick, Christopher A., Jiang, Yu Lin, Brannon, Marianne, Krishnan, Koyamangalath, Stone, William L. 18 September 2014 (has links)
Cancer cell esterases are often overexpressed and can have chiral specificities different from that of the corresponding normal cells and can, therefore, be useful targets for activating chemotherapeutic prodrug esters. Prodrug esters are inactive compounds that can be preferentially activated by esterase enzymes. Moreover, cancer cells often exhibit a high level of intrinsic oxidative stress due to an increased formation of reactive oxygen species (ROS) and a decreased expression of some enzymatic antioxidants. Prodrugs designed to induce additional oxidative stress can selectively induce apoptosis in cancer cells already exhibiting a high level of intrinsic oxidative stress. This study focused on the in vitro evaluation of four novel prodrug esters: the R- and S- chiral esters of 4-[(nitrooxy)methyl]phenyl N-acetylalaninate (R- and S-NPAA) and the R- and S- chiral esters of 4-[(nitrooxy)methyl]naphth-1-yl N-acetylalaninate (R- and S-NQM), which are activated, to varying extents, by oxidized protein hydrolase (OPH, EC 3.4.19.1) yielding a quinone methide (QM) intermediate capable of depleting glutathione (GSH), a key intracellular antioxidant. OPH is a serine esterase/protease that is overexpressed in some human tumors and cancer cell lines.Methods: To evaluate the chiral ester prodrugs, we monitored cellular GSH depletion, cellular protein carbonyl levels (an oxidative stress biomarker) and cell viability in tumorigenic and nontumorigenic prostate cancer cell lines.Results: We found that the prodrugs were activated by OPH and subsequently depleted GSH. The S-chiral ester of NPAA (S-NPAA) was two-fold more effective than the R-chiral ester (R-NPAA) in depleting GSH, increasing oxidative stress, inducing apoptosis, and decreasing cell viability in tumorigenic prostate LNCaP cells but had little effect on non-tumorigenic RWPE-1 cells. In addition, we found that that S-NPAA induced apoptosis and decreased cell viability in tumorigenic DU145 and PC3 prostate cell lines. Similar results were found in a COS-7 model that overexpressed active human OPH (COS-7-OPH).Conclusions: Our results suggest that prostate tumors overexpressing OPH and/or exhibiting a high level of intrinsic oxidative stress may be susceptible to QM generating prodrug esters that are targeted to OPH with little effect on non-tumorigenic prostate cells.

Ein Video- und Multimediacampus für die sächsische Hochschullandschaft ist im Zeitalter der Digitalisierung unabdingbar. Bei der Auswahl eines neuen Systems für die Bereitstellung von Videos im sächsischen Hochschulraum sollte jedoch neben technischen Betrachtungen unbedingt auch eine vergleichende Untersuchung zu den entstehenden Kosten und möglichen Organisationsmodellen erfolgen, um das System dauerhaft in der sächsischen Bildungslandschaft zu etablieren. Die Erfahrungen und Erkenntnisse aus dem Teilprojekt, welches sich den letztgenannten Aspekten widmete, bilden die Grundlage für die folgenden Abschnitte. Ziel des Teilprojektes Der wirtschaftliche Betrieb eines zentralen Videocampus für die sächsischen Hochschulen im Bildungsportal Sachsen war es – im Sinne eines dauerhaften Betriebes einer solchen zentralen Lösung – frühzeitig relevante Kostenpositionen zu identifizieren und bestmöglich hinsichtlich des zu erwartenden Umfangs zu bewerten. Ergänzend sollte eine Abschätzung möglicher organisatorischer Modelle erfolgen, so dass eine Entscheidungsgrundlage für eine Systemauswahl (in Verbindung mit den Aspekten Anforderungen und Systemleistung) und die Rahmenbedingungen für den späteren Betrieb geschaffen werden können. Der nachfolgende Beitrag zeigt auf, wie die Ermittlung des kostengünstigsten Systems erfolgte und liefert wertvolle Hinweise für andere Hochschulen oder Verbünde, die sich in einer ähnlichen Auswahlsituation befinden.

Muy aparte de ello, poder ir a un supermercado hoy en día en tiempos de pandemia puede significar un contagio, largas colas, perder tiempo valioso para poder compartir en familia. Es por ello, que a partir de este problema nace la idea de EasyMarket, el cual tiene como propósito principal brindar a las soluciones a las necesidades propuestas por la sociedad.
El proyecto EasyMarket consiste en demostrar la viabilidad de nuestra propuesta de negocio, la cual se basa en la creación de un aplicativo móvil llamado EasyMarket, la cual fue creada por la necesidad de muchas personas de poder encontrar precios más baratos de productos en supermercados y que se encuentren cerca a su domicilio. La aplicación puede ser usada entre los usuarios de un rango de edad de 18 a 55 años de Lima metropolitana y servirá para que en un solo lugar puedan comparar precios de los diferentes supermercados, ver qué productos están disponibles, qué ofertas hay, dónde está el supermercado, así como ayudar a identificar los productos idóneos para aquellas personas que sufran de enfermedades como diabetes, sean fitness y deseen consumir diversos productos saludables (libre de gluten, bajas calorías). De esta manera, el cliente podrá conseguir los mejores precios cerca de casa, con el fin de lograr un ahorro en tiempo y dinero. Por medio de esta investigación se indetificó que el principal problema se basa en la dificultad de las personas de no contar con un aplicativo que les permita tener la posibilidad de encontrar precios y ofertas de productos de supermercados al mismo tiempo, de manera fácil y rápida. Para llevar a cabo este proyecto, se realizó un análisis de cada uno de los factores involucrados en el business model canva, tales como segmento de clientes, socios claves, competidores, estructura de costos, entre otros. De la misma manera, se desarrolló el Plan Financiero, que nos permitió identificar si le proyecto era viable o no. Finalmente, pudimos conocer que desde el primer año se obtiene una utilidad neta positiva y a medida que pasa los años, dicho monto se encuentra en aumento, lo cual permitira generar una mayor inversion constantemente. / Being able to shop in a supermarket without knowing the information about specific products today can generate great stress and waste of time on people, as well as money spent too much if there are existing offers on specific days and we do not know them, or perhaps to know in which supermarket the same product is more comfortable. The EasyMarket project consists of demonstrating the viability of our business proposal, which is based on the creation of a mobile application called EasyMarket, which was created by the need of many people to find cheaper prices of products in supermarkets and that are close to their home. The application can be used by users between 18 and 55 years of age in metropolitan Lima and will allow them to compare prices of different supermarkets in one place, see what products are available, what offers are available, where the supermarket is, as well as help identify the ideal products for those who suffer from diseases such as diabetes, are fitness and wish to consume various healthy products (gluten-free, low calories). In this way, the customer will be able to get the best prices close to home, in order to save time and money. Through this research it was determined that the main problem is based on the difficulty of people not having an application that allows them to have the ability to find prices and offers of supermarket products at the same time, easily and quickly. To carry out this project, an analysis of each of the factors involved in the business model canva was performed, such as customer segment, key partners, competitors, cost structure, among others. In the same way, the Financial Plan was developed, which allowed us to identify whether the project was viable or not. Finally, we were able to learn that from the first year a positive net profit is obtained and as the years go by, this amount is increasing, which will allow us to generate a higher investment constantly. / Trabajo de investigación

Le raisonnement de la fertilisation azotée du blé a, depuis 40 ans, été largement orienté par le consensus autour de la méthode du bilan, avec comme principes fondamentaux : une nutrition azotée non limitante tout au long du cycle et l’estimation, de manière indépendante, des différents termes de l’équation du bilan pour caractériser la fourniture du sol et les besoins en azote de la plante. Au regard des enjeux de réduction des pollutions environnementales, de l’évolution des exigences qualitative du marché, ainsi que des difficultés de mise en oeuvre de la méthode, on s’interroge sur l’opportunité de renouveler ce paradigme. Pour proposer une nouvelle méthode qui réponde aux divers enjeux concernant l’azote, qui valorise au mieux les connaissances disponibles et dont la mise en oeuvre est cohérente avec les moyens des acteurs, nous avons mis en oeuvre une démarche de conception innovante, structurée en 3 étapes : un diagnostic des usages des outils actuels, une phase de conception, incluant des ateliers de conception et la mise au point de règle de décision à l’aide d’un modèle, et un test d’usage du prototype conçu.Le diagnostic des usages a mis en évidence plusieurs obstacles à la mise en oeuvre de la méthode du bilan, dont la fixation de l’objectif de rendement, ce qui a orienté la phase de conception vers l’exploration d’un concept de méthode de fertilisation azotée permettant de s’en affranchir. La méthode mise au point est basée sur le suivi régulier de l’état de nutrition azotée de la plante, l’acceptation de carences en azote non préjudiciables et des règles de décision tenant compte des conditions météorologiques au moment de l’apport. Nous montrons que la conception a rendu nécessaire la production de nouvelles connaissances, mais aussi la diversification des ressources et des compétences habituellement mobilisées. Ce travail enrichit les méthodes de conception d’outils d’aide à la décision en montrant comment l’articulation des 3 étapes permet de sortir du paradigme qui domine la fertilisation azotée depuis des décennies et d’élaborer un outil palliant les défauts des outils actuels. / Decisions about nitrogen fertilizer application on wheat have, for the last 40 years, been based on the balance sheet method, with the following underlying principles: non-limiting nitrogen nutrition throughout the crop cycle, and independent estimation of the various terms of the equation, to characterize soil nitrogen supply and plant nitrogen needs. Environmental pollution, changes in the qualitative requirements of the market and difficulties implementing this method have raised questions about whether it might be appropriate to switch to new ways of managing nitrogen fertilizer. We developed a new method meeting the diverse constraints relating to nitrogen use, making the best use of available knowledge and easily applicable by users, through a 3-steps innovative design approach: a diagnosis of the use of current tools, a design phase including design workshops, production of new knowledge, a modelbased prototyping, and the testing of a prototype method.The diagnosis of uses identified several barriers to the implementation of the balance sheet method, including the need to set a target yield. This directed the design phase towards the exploration of new ways of managing nitrogen fertilizer that did not require the fixing of a target yield. The method developed is based on the regular monitoring of plant nitrogen nutrition, the toleration of periods of nitrogen deficiency that are not prejudicial and the use of decision rules taking weather conditions at the time of nitrogen application into account.This design required the generation of new knowledge and a diversification of the resources and skills usually mobilized. This work enriches the methods for designing decision support tools and shows how a combination of 3 steps can be used to develop a tool for managing nitrogen fertilizer applications completely different from the dominant paradigm of the last 40 years, and compensating the defects of current methods.

Introduktion: Hyalint brosk består av kondrocyter inbäddade i ett extracellulärt matrix som täcker alla kroppens synoviala leder. I dag är skador på hyalint brosk ett folkhälsoproblem med begränsade behandlingsmöjligheter. Mesenkymala stamceller har i flertalet studier visat sig vara en lovande kandidat för framtida behandling av broskskador, eftersom dessa kan differentiera till broskproducerande kondrocyter. Modern behandling av broskskador innefattar odling av patientens egna kondrocyter eller stamceller för att sedan återföra dem till broskskadan. För att göra detta effektivt behöver optimering av odlingsförhållanden för aktuella celler utföras. Syfte: Att undersöka tillväxt hos kondrocyter samt mesenkymala stromaceller från benmärg i varierande odlingsförhållanden, detta med avseende att utvärdera vilka tillsatser som mest gynnar cellernas viabilitet och proliferation. Material & Metod: Framtagning av kondrocyter och mesenkymala stromaceller från benmärg i denna studie utfördes på överblivet ortopediskt material från en individ. Cellerna kollagenasbehandlades innan de odlades ut i ett standardmedium under tre veckor. Därefter odlades de i närvaro av olika tillsatser under sex dagar. Cellernas viabilitet mättes genom relativ fluorescens med CellTiter-Blue® reagens medan proliferationen mättes genom cellräkning med flödescytometri. Resultat: Det fanns en signifikant skillnad (ANOVA, p<0,05) mellan minst två tillsatser inom grupperna för viabilitetsanalys samt cellräkning för både kondrocyter och benmärgderiverade mesenkymala stromaceller (BMMSC). Kondrocyterna som odlades i närvaro av fetalt bovint serum (FBS) hade högre proliferation och viabilitet än de celler som odlades i frånvaro av FBS. FBS i kombination med insulin-transferrin-selen (ITS) och kondrocytfaktorerna (KF) dexametason, prolin, insulin samt transforming growth factor beta hade störst positiv effekt på proliferationen. Vidare visade Minimum Essential Medium α med tillsats av FBS 10 % ha störst effekt, av den panel som undersöktes, på både viabilitet och proliferation hos BMMSC. Slutsats: FBS i kombination med ITS och KF hade störst stimulerande effekt på proliferation hos kondrocyter. Val av standardmedium hade större betydelse än tillsats av faktorer vid odling av BMMSC in vitro. / Introduction: Articular cartilage consists of chondrocytes and covers all the synovial joints in the body. Articular cartilage injuries are today a public health issue with limited orthopedic treatments. Mesenchymal stem cells have shown to be a promising candidate for future treatment of articular cartilage injuries due to their many abilities. Modern treatment of cartilage damage involves cultivation of the patient´s own chondrocytes and stem cells. To make this treatment as effective as possible the culture conditions for chondrocytes and stem cells must be optimized. Aim: To study cell growth in chondrocytes and mesenchymal stromal cells from bone marrow in varying culture conditions, this regarding to evaluate the most optimal culture conditions for cell viability and proliferation. Method: Cell extraction in this study was performed on leftover orthopedic material. The cells were treated with collagenase and then cultured in a standard medium for three weeks before they were cultured in presence of various factors for six days. Cell viability was measured by relative fluorescence while proliferation was measured by cell counting. Result: There was a statistically significant difference (ANOVA, p<0,05) between at least two additives within the groups for viability analysis and cell count for both chondrocytes and bone marrow-derived mesenchymal stromal cells (BMMSC). The chondrocytes cultured in presence of fetal bovine serum (FBS) had a higher proliferation and viability compared to the cells cultured in the absence of FBS. FBS in combination with insulin-transferrin-selenium (ITS) and the chondrocyte factors (CF) dexamethasone, proline, insulin and transforming growth factor beta had the biggest impact on proliferation. Minimum Essential Medium α with FBS 10% additive showed positive effects on both viability and proliferation of BMMSCs. Conclusion: FBS in combination with ITS and CF had a good effect on the proliferation of chondrocytes. The choice of standard medium seems to have a more important role than growth factor additive when studying BMMSCs expansion in vitro.
