Estudo epidemiológico e genético dos vírus da imunodeficiência felina identificados no Estado de São Paulo /

Orientador : João Pessoa Araújo Júnior / Resumo: O presente trabalho objetivou determinar a freqüência do vírus da imunodeficiência felina, correlacioná-la a dados epidemiológicos, identificar os diferentes sorotipos das amostras positivas e realizar a análise filogenética das mesmas. Para tanto, foram colhidas 519 amostras de sangue total de gatos domésticos oriundos de diferentes cidades do estado de São Paulo submetidas à técnica de PCR-nested para identificação do vírus. Dessas amostras, 81 (15,6%) apresentaram-se positivas ao teste, das quais 45 (55,5%) pertenciam a gatos com sinais clínicos de alguma enfermidade, sendo 21 fêmeas e 24 machos. Vinte e duas amostras (27,2%) foram provenientes de animais aparentemente normais, com duas fêmeas e 20 machos, e outras 14 (17,3%) a gatos sem histórico clínico definido, com três fêmeas e 11 machos. Foram submetidas ao seqüenciamento genético 50 (61,7%) amostras positivas. Entretanto, em somente 32 destas (64%) realizou-se a inferência filogenética. A análise genética foi baseada em um segmento do gene gag de 459 pb. As relações genealógicas foram realizadas através dos métodos da máxima parcimônia, da evolução mínima e de agrupamento de vizinhos. Os resultados obtidos revelaram uma ampla similaridade entre as três árvores construídas e em todas elas, as 32 amostras estudadas foram agrupadas dentro do mesmo sorotipo, que foi o sorotipo B. / Abstract: The present work aimed to determine the feline immunodeficiency virus frequency, to correlate it to epidemiologic data, to identify different FIV subtypes of positive samples and also to accomplish its phylogenetic analysis. For that, it was collected 519 blood samples of domestic cats from different cities of the São Paulo state, submitting them by PCR-nested for virus identification. Eighty one samples (15.6%) were positive to the test, which 45 (55.5%) of them were from animals with clinical signs of some illness. Those samples, 21 were from female and 24 from male cats. Twenty-two of the positive samples (27.2%) were from healthy cats (2 females and 20 males), and 14 of them (17.3%) from cats without clinical history (3 females and 11 males). Fifty positive samples (61.7%) were submitted to DNA sequencing, but only in 32 of them the phylogenetic inference was done. The genetic analysis was based on a segment of the gag gene with 459pb. The genealogical relationships were established through maximum parcimony, minimum evolution and neighbor-joining methods. The results revealed a wide similarity among the three built trees and, in all of them, the 32 studied samples were allocated inside of the same subtype, that was the subtype B. / Doutor

Estes animais estão expostos constantemente a agentes virais causadores de patologias. Dentre estes, o herpesvírus bovino 1 (BHV1) e 5 (BHV5) apresentam grande importância epidemiológica. O BHV1 causa infecções respiratórias e genitais, enquanto que o BHV5 meningoencefalite. As proteínas de membrana presentes nos herpesvírus os tornam capazes de infectar as células do hospedeiro. Com afinidade a distintas células, desenvolvem sua capacidade de infecção, apresentam mecanismos de sobrevivência à morte celular programada e a resposta imunológica. As interações vírus-hospedeiro são fundamentais no entendimento de aspectos relacionados a transmissão viral, capacidade de infecção e progressão da doença. Para este fim, se faz necessário compreender elementos fundamentais, como a modulação da função mitocondrial e a replicação do BHV, bem como os níveis de apoptose nas células do hospedeiro. De igual importância, é indispensável entender o comportamento de genes apoptóticos que estão relacionados à infecção por BHV5 e seus envolvimentos na viabilidade celular. Estes achados contribuem para o entendimento da imunopatogenia do BHV, auxiliam a evitar a disseminação destes agentes e são de grande valia para o desenvolvimento de vacinas. / Brazil has one of the largest cattle hers worldwide. These animals are constantly exposed to pathological viral agents. Bovine herpesvirus 1 (BHV1) and BHV5 have great epidemiological importance. BHV1 is causative agent of respiratory and genital infections, while BHV5 causes meningoencephalitis. Herpesvirus membrane proteins allow them to infect the host cells. They present affinity to different types of cells, develop mechanisms to survive programmed cell death, and immune response. Virus-host interactions are essential to understand aspects related to viral transmission, capacity of infection and disease progression. To this end, it is necessary to understand the fundamental elements related to the modulation of mitochondrial function and replication of BHV, as well as apoptosis levels in the host cells. It is crucial to understand the behavior of apoptotic genes related to BHV5 infection and cell viability. These findings contribute to understanding the BHV immunopathogenesis, help to prevent the spread of these agents, and are of great value to the development of vaccines.

Virulência de linhagens de Rhodococcus equi isoladas de linfonodo de suínos e javalis (Sus scrofa) de abatedouros /

Diferentes fatores de virulência são reconhecidos na patogenicidade de Rhodococcus equi. A estrutura da parede celular bacteriana, a viabilidade no interior de fagócitos e na ausência de ferro, a produção de citotoxinas, a resistência aos antimicrobianos convencionais e, recentemente, a presença de proteínas associadas à virulência (Vap) reguladas por plasmídios, são considerados os principais mecanismos de virulência do microrganismo. Diferentes fatores de virulência foram avaliados em 23 (6,1%) linhagens de R. equi isoladas de 378 linfonodos submandibulares e mesentéricos de suínos e javalis (Sus scrofa). Foram realizados exames microbiológicos em 129 linfonodos apresentando lesões (linfadenite) e 129 sem lesões (controle) de suínos, e 60 linfonodos com lesões e 60 sem lesões de javalis. Dentre as 23 linhagens de R. equi, 19 (7,4%) foram isoladas de suínos, das quais 17 obtidas de linfonodos com lesões e duas sem lesões. Das 19 linhagens de suínos, 18 (94,7%) foram obtidos de linfonodos submandibulares e um (5,3%) de mesentérico. As quatro (3.3%) linhagens de R. equi isoladas de javalis foram obtidas exclusivamente de linfonodos com lesões. Destes, três foram obtidos de linfonodos submandibulares e um de mesentérico. Dentre nove antimicrobianos testados, azitromicina (100,0%), gentamicina (100,0%), levofloxacina (100,0%), vancomicina (100,0%), amoxicillina/ácido clavulânico (94,7%), eritromicina (94,7%) e rifampicina (94,7%) foram os fármacos mais efetivos. Baixa ocorrência de resistência aos antimicrobianos nos isolados de suínos foi observada contra os fármacos testados. Different virulence factors are recognized in pathogenicity of the Rhodococcus equi. The structure of bacterial cell wall, the viability inside of phagocytes and in absence of iron, the production of cytotoxins, the resistance to conventional antimicrobials and recently, the presence of proteins associated to virulence (Vap) regulated by plasmids, are considered the most important virulence mechanisms of microorganism. Different virulence factors were evaluated in 23 (6.1%) R. equi strains isolated from 378 submandibular and mesenteric lymph nodes of swine and wild boars (Sus scrofa). Microbiological exams were performed in 129 lymph nodes presenting lesions (lymphadenitis) and 129 without lesions (controls) from swine, and 60 lymph nodes with lesions and 60 without lesions from wild boars. Among 23 R. equi strains, 19 (7.4%) were isolated from swine and, from these, 17 were obtained from lymph nodes with lesions and two without lesions. Avaliação do tratamento experimental de cães naturalmente infectados com vírus da cinomose com ribavirina, prednisona e DMSO através da RT-PCR /

Foram utilizados 60 cães apresentando sinais neurológicos da cinomose com evolução de dez dias. Animais foram internados para tratamento de suporte; avaliados diariamente e submetidos ao hemograma e dosagens bioquímicas. Os grupos 1 e 2 foram tratados com ribavirina associada ao DMSO; os grupos 3 e 4 foram tratados com DMSO e prednisona e os grupos 5 e 6 foram tratados com ribavirina e prednisona, ribavirina, prednisona e DMSO. Os animais foram anestesiados e colhidos líquor, medula óssea e sangue, antes e após o tratamento e realizada a RTPCR das amostras; as negativas foram analisadas pela técnica de hn-PCR. O vírus foi encontrado em 95% das amostras de sangue, 90% de medula óssea e 53,3% de líquor pré-tratamento. O efeito adverso da ribavirina quando associada com a prednisona foi anemia. A prednisona na dose imunossupressora causou aumento da dosagem de proteína e diminuição da celularidade liquórica, leucocitose. Já a dose antinflamatória causou diminuição de proteína no líquor. Baseado nos índices de sobrevida e melhora clínica, o tratamento mais efetivo foi o G2 (80%); seguido do G1, G5 e G3 (70%); o G6 (60%); o G4 com o pior índice (30%). Pós-tratamento, a frequência viral foi 97,7% no sangue, 86,4% na medula óssea e 27,3% no líquor / Abstract: The present study aims at the identification of ribavirin, prednisone and DMSO's treatment effects in dogs with canine distemper, at the identification of the viral presence in the blood, bone marrow and cerebrospinal fluid (CSF) before and after the treatment and also at the identification of side effects of the experimental drugs and its combinations. Sixty dogs presenting canine distemper with neurological signs about ten days evolution were observed. The animals were hospitalized for the support treatment, assessed on daily basis and subjected to blood cells count and biochemical analysis. Groups 1 and 2 were treated with ribavirin and its combination with DMSO; Groups 3 and 4 treated with prednisone and DMSO, Group 5 treated with ribavirin and prednisone, while Group 6 with ribavirin, prednisone and DMSO. The animals were anesthetized for the cerebrospinal fluid, bone marrow and blood samples collection before and after the treatment, then the RT-PCR of the samples was proceeded. The negative were analysed according to the hn-PCR technique. The canine distemper virus were found in 95% of blood samples, 90% of bone marrow and 53,3% of CSF before the treatment. The adverse effect of ribavirin and its association with prednisone was anemia. Prednisone, at its immunosuppressive dose, led to the increase of protein and decrease of cellularity in CSF, and increase of leukocytes blood count. The antiinflammatory dose led to the CSF protein concentration's decrease. Considering the survival and clinical improvement rates, the most successful treatment was the one applied to the G2 (80%); followed by G1 (70%); G5 (70%) and G3 (70%); G6 (60%); and the lowest rate G4 (30%). Pesquisa de agentes virais de doenças hemorrágicas em cervídeos brasileiros : Imunodetecção de células-tronco tumorais em neoplasias mamárias caninas /

Orientador: Karin Werther / Banca: Eliana Reiko Matushima / Banca: Eveline dos Santos Zanetti / Resumo: Os cervídeos têm sido acometidos por doenças hemorrágicas virais, tais como, doença epizoótica hemorrágica (DEH), língua azul (LA) e doença hemorrágica por adenovírus (DHA). Como as lesões macroscópicas, entre elas, enterite hemorrágica, edema pulmonar, petéquias e sufusões em diversos órgãos, são observadas nas três doenças, há necessidade de técnicas acuradas para realizar o diagnóstico definitivo. A partir do material de arquivo (blocos de parafina) existente no Departamento de Patologia Veterinária da FCAV - Unesp, 42 cervídeos brasileiros, tanto de vida livre como de cativeiro, foram selecionados, por apresentarem sinais clínicos e/ou lesões macroscópicas sugestivas de doenças virais hemorrágicas. Das amostras analisadas, utilizando técnica de imunoistoquímica, todas apresentaram resultado negativo para adenovírus. Utilizando técnica de RT-PCR em tempo real para vírus da doença epizoótica hemorrágica, os resultados foram também negativos. A mesma técnica aplicada para vírus da língua azul revelou sete animais positivos (16,66%) confirmados com eletroforese em gel de agarose 4% e sequenciamento. Todos os casos positivos foram de animais provenientes de cativeiro, sendo três fêmeas (duas jovens, uma adulta) e quatro machos jovens. As principais alterações macroscópicas observadas nesses animais foram conteúdo intestinal hemorrágico, mucosas avermelhadas do trato gastrointestinal, úlceras em língua e petéquias em diversos órgãos. Na histologia observou-se principalmente infiltrado inflamatório, hemorragia e congestão em diversos órgãos. Os vírus da DHA e DEH não estão envolvidos nos óbitos dos cervídeos estudados. A relevância deste trabalho está no fato de ser a primeira descrição de material genético do vírus da LA em cervídeos brasileiros / Abstract: Cervids have been affected by viral hemorrhagic diseases, such as Epizootic hemorrhagic disease (EHD), Bluetongue (BT), and Adenoviral hemorrhagic disease (AHD). Once that gross lesions, among them, hemorrhagic enteritis, pulmonary edema, petechiae and suffusions in several organs, are similar, it is necessary to use accurate techniques to the definitive diagnosis. From the archival material (paraffin blocks) available in the Department of Veterinary Pathology of FCAV - Unesp, 42 Brazilian deer, both free living and captive, were selected because they had lesions suggestive of hemorrhagic viral disease. The samples analyzed, using Immunohistochemistry, were all negative for adenovirus. Using real time RT-PCR for EHD, the results were also negative. The same technique applied to BT virus revealed seven positive animals (16,66%) confirmed after agarose 4% gel electrophoresis and gene sequencing. The main macroscopic changes observed in these animals were hemorrhagic intestinal contents, reddish mucous membrane of the gastrointestinal tract, ulcers on tongue and petechiae in various organs. Mostly histological changes observed were inflammatory infiltrate, hemorrhage, and congestion in various organs. All positive cases were from captive animals, three females (two young and one adult), and four young males. The AHD and EHD virus are not involved in the deaths of the deers studied in this research. Análise histopatológica e imunoistoquímica de encéfalo de cães com cinomose tratados com ribavirina /

Orientador: Antonio Carlos Paes / Banca: Marcio Garcia Ribeiro / Banca: Osimar de Carvalho Sanches / Resumo: A cinomose é uma doença infecto-contagiosa viral, sistêmica e letal causada pelo Morbilivírus que acomete cães de todas as idades e apresenta sinais clínicos respiratórios, gastrintestinais, dermatológicos, oftálmicos e neurológicos, que podem ocorrer sequencialmente ou isoladamente. Este estudo teve como objetivo comparar lesões encefálicas utilizando a histopatologia e avaliar a quantidade de vírus pela imunoistoquímica em cães tratados com Ribavirina na dose de 30mg/Kg por via oral uma vez ao dia por 15 dias e animais não tratados com esse fármaco, e que devido a evolução desfavorável da doença foram eutanasiados. Os animais foram atendidos e tratados durante a rotina do Setor de Enfermidades Infecciosas dos Animais Domésticos- FMVZ- UNESP/ Botucatu, SP. A análise histopatológica constituiu na avaliação de córtex cerebral, cerebelo, tálamo, tronco encefálico, hipocampo e visualização de manguitos perivasculares em meninges. A partir dessas regiões foi possível identificar e classificar lesões como: desmielinização; presença de manguitos perivasculares, edema, necrose neuronal, alterações vasculares (congestão, hemorragia e trombose) e alterações inflamatórias (meningite, meningoencefalite e meningoencefalomielite). Foi observado que a desmielinização se manteve altamente presente nos dois grupos, e que quando comparados estatisticamente não existe diferença significante entre os grupos, assim como na formação de manguitos perivasculares. Houve diminuição considerável do edema em todas as estruturas avaliadas, e a ribavirina® se mostrou eficaz na redução da inflamação encefálica, assim como a necrose neuronal foi menor em quase todas as estruturas avaliadas no grupo tratado, com exceção do tálamo. A análise estatística comparativa da marcação viral entre os grupos se mostrou significante somente para o cerebelo, tálamo e tronco encefálico / Abstract: Distemper is a viral infectious disease, caused by systemic and lethal morbillivirus that affects dogs of all ages and clinical signs respiratory, gastrointestinal, dermatological, ophthalmic and neurological disorders, which may occur sequentially or separately. This study aimed to compare brain lesions using histopathology and assess the amount of virus by immunohistochemistry in dogs treated with Ribavirin at a dose of 30mg/Kg orally once daily for 15 days and animals not treated with this drug, and that due to the unfavorable development of the disease were euthanized. The animals were treated and handled during routine Division of Infectious Diseases of Domestic Animals-FMVZ-UNESP / Botucatu, SP. Histopathologic analysis was to evaluate the cerebral cortex, cerebellum, thalamus, brainstem, hippocampus and viewing perivascular cuffs in the meninges. From these regions it was possible to identify and classify lesions such as demyelination, the presence of perivascular cuffing, edema, neuronal necrosis, vascular changes (congestion, hemorrhage and thrombosis) and inflammatory changes (meningitis, meningoencephalitis and meningo-encephalomyelitis). It was observed that the demyelination remained strongly present in both groups, and compared statistically no significant difference between groups, as well as the formation of perivascular cuffs. There was a considerable decrease of the edema measured in all structures, and ribavirin ® was effective in reducing brain inflammation, and neuronal necrosis was lower in almost all structures evaluated in the group treated with the exception of the thalamus. Avaliação da patogenicidade e da imunidade cruzada de estirpe variante do vírus da bronquite infecciosa aviária isolada no Brasil /

Orientador: Hélio José Montassier / Banca: Antônio Carlos Paulillo / Banca: Ricardo Luís Moro de Sousa / Resumo: Nesse estudo, um isolado de campo do vírus da bronquite infecciosa (VBI) no Brasil (IBVPR-12), previamente classificado como um genótipo variante, foi caracterizado de forma comparativa com a estirpe M41 do VBI, sendo levantadas as características de patogenicidade em diferentes órgãos como a traqueia, o pulmão, os rins, as gônadas e as tonsilas cecais (patotipo) e a imunidade cruzada com relação à estirpe vacinal H120 do VBI (protectotipo), incluindo as respostas imunes humorais sistêmicas e locais induzidas. Para tanto, foram utilizados grupos experimentais de galinhas "specific pathogen free" (SPF) previamente vacinadas ou não com a estirpe H120 do VBI e depois desafiados com essa variante viral, ou com a estirpe M41. Para essas duas estirpes virais foram avaliadas a capacidade de replicação e as lesões produzidas em diferentes órgãos, a atividade inibidora do movimento ciliar no epitélio traqueal e as respostas imunes humorais desenvolvidas no soro sanguíneo e na secreção lacrimal dessas aves. Foram observadas diferenças marcantes na patogenicidade e no tropismo tecidual desses vírus, sendo que a estirpe M41 apresentou replicação mais intensa e lesões mais pronunciadas no trato respiratório, especialmente na traqueia, enquanto que a estirpe variante foi encontrada de forma mais distribuída em vários dos órgãos analisados, tendo-se replicado e provocado menos lesões na traqueia, mas alcançando maior replicação e tendo causado lesões mais severas nos rins e nos testículos. Nas regiões teciduais mais afetadas por lesões, a presença do VBI foi detectada por marcação específica com anticorpos policlonais contra a nucleoproteína do VBI pela técnica de imuno-histoquímica. As aves vacinadas com a estirpe H120 do VBI, revelaram proteção parcial contra a estirpe variante em órgãos como traqueia e... (Resumo completo, clicar acesso eletrônico abaixo) / Abstract: In this study a Brazilian field isolate of infectious bronchitis virus (IBV), previously classified as variant genotype, was characterized comparatively with the M41 strain of IBV, by evaluating the pathogenicity in different organs (trachea, lung, kidney, gonads and caecal tonsil) and the cross-immunity with H120 vaccine strain, including the systemic and local humoral immune responses. Experimental groups of specific pathogen free (SPF) chickens were vaccinated or not with H120 strain of IBV and challenged with this variant isolate. The viral replication and histopathology in different tissues and organs, the ability to inhibit ciliar movement of tracheal epithelial cells, and local and systemic humoral immune responses were evaluated in these chickens. The pathogenicity and tissue tropism of these IBV strains showed marked differences, and while the M41 strain damaged more the respiratory tract, specially the trachea, the variant isolated has a wide tissue distribution, showing less replication and lesions in the trachea, but affecting more severely the kidney and the testicles. In the most affected tissue regions, the presence of IBV was detected by immunohistochemistry technique, using an anti-nucleoprotein polyclonal antibodies. The H120 vaccine induced against this variant isolate a partial protection with regard to the infection of trachea and kidney and no cross-protection to the infection of testicles. No Brasil, os casos de encefalite por BoHV-5 são a segunda maior causa de óbitos no rebanho. A infecção viral geralmente resulta em alteração no processo de morte celular programada, pela via mitocondrial, estimulando ou inibindo genes relacionados à apoptose, favorecendo a replicação e disseminação viral. O presente estudo teve como objetivo a análise da expressão de genes relacionados à apoptose (Apaf-1, α, FasL e citocromo c) em células MDBK infectadas ou não com o BoHV-5. Foi possível observar um aumento (p<0,05) na expressão dos genes estudados para o grupo infectado comparado ao controle, em diferentes momentos pós-infecção experimental. No caso da família Herpesviridae, a modulação da apoptose parecer ser uma etapa chave para sua patogênese, apesar da associação das atividades anti e pró-apoptóticas com o BoHV-5 ser completamente desconhecida. Este estudo demonstra a capacidade do BoHV-5 em modular, seja ativando ou inibindo, a via extrínseca da apoptose / Abstract: Belonging to the family Herpesviridae, subfamily Alphaherpesvirinae, bovine Herpesvirus type 5 ( BoHV-5) is the causative agent of bovine non-suppurative meningoencephalitis, which predominantly affects young cattle. In Brazil, cases of encephalitis caused by BoHV-5 are the second leading cause of deaths in the herd. Viral infection usually results in a change in the programmed cell death via mitochondrial, stimulating or inhibiting apoptosis related genes, promoting viral replication and dissemination process. The present study aimed to analyze the expression of apoptosis related genes (Apaf-1, TNFα-R1, FasL and cytochrome c) in infected or uninfected MDBK cells with BoHV-5. We observed an increase ( p < 0.05 ) in expression of the genes studied for the infected group compared with controls, in different times post-infection experimental. In the case of the Herpesviridae family, modulation of apoptosis appears to be a key step in its pathogenesis, despite the association of anti and pro-apoptotic activities with BoHV-5 is completely unknown. Prevalência dos principais vírus respiratórios em bovinos da raça holandesa, no estado do Paraná /

Orientador: Roberto Calderon Gonçalves / Coorientador: Ivan Roque de Barros Filho / Banca: Simone Biagio Chiacchio / Banca: José Paes Oliveira Filho / Banca: Sílvia Cristina Osaki / Banca: Andreza Amaral da Silva / Resumo: Realizou-se levantamento sorológico dos seguintes vírus respiratórios Herpesvírus Bovino 1 (BoHV-1), Vírus da Diarreia Viral Bovina (BVDV), Vírus da Parainfluenza Bovina Tipo 3 (bPIV-3) e Vírus Respiratório Sincicial Bovino (BRSV) em bovinos da raça Holandesa variedade Preta e Branca (HPB), criados no estado do Paraná. Colheram-se 714 amostras de sangue da veia jugular de bovinos, não vacinados para os agentes estudados, com mais de seis meses de idade, de 26 propriedades leiteiras, distribuídas em 17 municípios do estado do Paraná. Paralelamente à colheita, aplicou-se um questionário para cada propriedade estudada, com a finalidade de avaliar os fatores de risco associados aos vírus examinados. As amostras sorológicas foram submetidas ao teste de diagnóstico de virusneutralização no Laboratório de Viroses de Bovinos do Instituto Biológico de São Paulo. Foram observadas prevalências de 22,3%; 35,8%; 80,4% e 93,5% de bovinos sororreagentes e 65,3%, 88,5%, 96,1% e 100% das propriedades reagentes para BoHV-1, BVDV, bPIV-3 e BRSV, respectivamente. A coinfecção que ocorreu com maior prevalência foi a bPIV-3 e BRSV (37,1%). Após análise de regressão logística, os fatores de risco associados ao BoHV-1 foram: bovinos com idade maior que 48 meses (OR=15,012; IC95% 7,500 - 30,051), área da propriedade 26 a 50 hectares (OR=11,328; IC95% 2,482 - 51,697), lotação maior de três bovinos por hectare (OR=1,026; IC95% 0,370 - 2,847), percentual de entrada maior que 10% de bovinos (OR=52,520; IC95% 13,269 - 207,876); para o BVDV foram: bovinos com idade maior que 48 meses (OR=4,407; IC95% 2,456 - 7,906), região Sudoeste (OR=52,388; IC95% 4,629 - 5892,873), sistema de produção semi-intensivo (OR=1,333; IC95% 0,261 - 6,815), propriedade com número de animais menor ou igual a 50 bovinos (OR=16,682; IC95% 3,218 - 86,481), percentual de entrada maior que 10% de bovinos (OR=17,56; IC95% 7,613 - 40,506) e ... / Abstract: A serological survey of the following respiratory viruses: Bovine Herpesvirus 1 (BoHV-1), Bovine Viral Diarrhea Virus (BVDV), Bovine Parainfluenza Virus Type 3 (bPIV-3) and Bovine Respiratory Syncytial Virus (BRSV) in Holstein cattle black and White (HPB), raised in Paraná state was performed. 714 blood samples were collected from the jugular vein of cattle not vaccinated for the studied agents, over six months of age, from 26 dairy herds, distributed in 17 municipalities of Paraná state. Alongside the collection, a questionnaire was used for each property studied in order to assess the risk factors associated with the viruses examined. The serum samples were subjected to the diagnosis test of virus neutralization at the Laboratory of Viral Diseases of Cattle of the Biological Institute of São Paulo. Frequencies of 22.3%; 35.8%; 80.4% and 93.5% of seropositive cattle were observed and 65.3%, 88.5%, 96.1% and 100% of the seropositive properties BoHV-1, BVDV, bPIV-3 and BRSV, respectively. The co-infection that occurred most frequently was the bPIV-3 and BRSV (37.1%). Assim, um novo tratamento que evite perdas financeiras irreparáveis em criações de cavalos é bastante relevante. Por outro lado, os produtos naturais são alvos das mais diversas pesquisas para formulação de novos medicamentos, dentre eles os antivirais por possuírem um elevado potencial terapêutico. Neste trabalho, realizamos ensaios biológicos em cultivo celular com células Vero para determinar o efeito citotóxico e os potenciais antivirais de 8 compostos (quatro substâncias puras, um extrato de microrganismo e três extratos vegetais de uma mesma espécie). As citotoxicidades foram determinadas por microscopia óptica e confirmadas usando uma variação do método colorimétrico baseado na redução do sal MTT. Com o objetivo de investigar in vitro o potencial antiviral de cada tratamento sobre o EHV-1, foi feita a avaliação da redução de título viral por visualização do efeito citopático seguida de titulação viral em três ensaios antivirais, o de “Inativação Direta”, o “End Point” e o “Timing of Addition”. O primeiro teve como objetivo determinar se os tratamentos possuem ação sobre as partículas virais desestabilizando-as ou se ligando a elas de forma irreversível. O segundo tem como objetivo determinar se as substâncias puras ou presentes nos extratos se ligam aos receptores celulares ou a outras estruturas moleculares causando modificações celulares duradouras. O terceiro foi realizado com o objetivo de determinar a ação dos compostos sobre quatro etapas do ciclo infectivo viral, avaliando se apresentam ação antiviral em fases específicas da infecção. Os resultados dos testes demonstraram que as concentrações máximas não tóxicas obtidas foram respectivamente, 70, 100, 120, 110 e 70 μg/mL para a quercetina, morina, rutina, ácido trans-cinâmico e monascus; 40, 40 e 90 μg/mL para os extratos E1, E2, E3, respectivamente e 3,0% (v/v) para o DMSO. Nas concentrações testadas, o DMSO não causou variação significativa no título viral em nenhum dos testes, sendo, portanto, um composto inerte. A rutina não demonstrou ação antiviral significativa em nenhum dos testes. Os extratos apresentaram maior potencial antiviral contra o EHV-1 in vitro, seguidos das substâncias quercetina e morina. Nos resultados do teste de inativação direta, foi observada a ação dos três extratos, da quercetina e da morina. O E3 foi o único a apresentar ação no teste “End Point”. No “Timing of Addition”, todos os compostos apresentaram ação antiviral em pelo menos uma etapa de infecção, com exceção da rutina. A morina e o monascus sobre a adsorção, o ácido trans- cinâmico sobre os receptores celulares e na adsorção, a quercetina e o E2 nas fases de adsorção e penetração viral, e o E1 e o E3 em todas as fases. O processo de adsorção foi o mais afetado pelos compostos, sugerindo ação principalmente sobre as partículas virais e receptores celulares, impedindo momentos iniciais da infecção. Possivelmente, há nos extratos um efeito sinérgico decorrente da ação de mais de um composto. Com os resultados encontrados neste trabalho, concluímos que estes compostos poderão ser de grande importância nas medicinas veterinária e humana para prevenção e tratamento de infecções virais e para formulação de novos medicamentos. / The Equine herpesvírus 1 (EHV-1), a important pathogen of horses, was chosen as target virus because it represents a highly pathogenic family and although we already have vaccines against it, they are inefficient. Thus, a new treatment that avoid irreparable financial loses in creations of horses is quite relevant. On other hand, due to their high therapeutic potential, the natural products are targets of various researches for formulation of new medicines. In this work, we made biological assays in Vero’s cell culture to determine the cytotoxic effect and antiviral potential of 8 compounds (four pure substances, one microorganism extract and three plant extracts from the same specie). The cytotoxicidates were determined by optical microscopy and confirmed by using a variation of colorimetric method based on MTT salt reducing. In order to investigate in vitro the antiviral potential from each treatment on the EHV-1, we did the viral title reduction evaluation by viewing the cytopathic effect followed by viruses titration in three antiviral assays, "Direct Inactivation", "End Point" and "Timing of Addition". The first aimed to determine whether treatments have action on the viral particles, destabilizing them or binding to them on an irreversible way. The second aims to determine whether the pure substances or those who are in extracts bind to cell receptors or other molecular structures causing lasting cellular changes. The third was carried out to determine the action of the compounds on four stages of the viral infective cycle, assessing whether they have antiviral action in specific stages of infection. The assay results showed that the maximum non-toxic concentrations obtained were respectively, 70, 100, 120, 110 and 70 μg/mL to quercetin, morin, rutin, cinnamic acid and monascus; 40, 40 and 90 μg/mL to extracts E1, E2, E3, respectively and 3.0% (v/v) to DMSO. In the tested concentrations, the DMSO didn’t cause significant variation in viral title in any of the assays, being, therefore, an inert compound. The rutin has not shown significant antiviral action in any of the tests. The extracts had greater antiviral potential against EHV-1 in vitro, followed by quercetin and morin. In the direct inactivation assay results, it was observed the action of three extracts, of quercetin and of morin. The E3 was the single to submit action in the assay ‘’End Point ". In the "Timing of Addition," all compounds showed antiviral action in at least one infection stage, except for rutin. Morin and monascus on adsorption, cinnamic acid on cell receptors and adsorption, quercetin and E2 during the adsorption and viral penetration phases, and E1 and E3 at all stages. The adsorption process was the most affected by compounds, suggesting action mainly on the viral particles and cell receptors, preventing infection initial stages. Possibly, there is in extracts a synergic effect resulted from action of more than one compound.
