Infuence of volatiles transport in disks on giant planets composition / L'influence du transport des volatiles dans les disques sur la composition des planètes géantes

Ce manuscrit présente des travaux originaux sur la théorie de la formation des planètes.Le but fondamental est de connecter la composition chimique des planètes géantes etdes petits corps avec les processus physiques et chimiques prenant lieu dans le disqueprotoplanétaire.1. Dans le chapitre 1 j'introduis les propriétés fondamentales des disques protoplané-taires ainsi que les bases de la théorie de formation des planètes.2. Dans le chapitre 2 j'attaque le problème du rapport C/O supersolaire mesurérécemment dans WASP 12b. J'élabore un modèle qui suit la distribution et transportde l'eau et du CO gazeux et solides à travers leurs di_usion, condensation,coagulation, gaz drag et sublimation afin de quantifer la variation du rapport C/Odans le disque en fonction du temps et de la distance. Mon modèle montre que,au fur et à mesure du temps, les vapeurs vont être enlever de l'intérieur de leurlignes de glaces respectives, avec le vapeur CO enlevé beaucoup plus lentement quela vapeur d'eau. Cette effet va augmenter le rapport C/O à l'intérieur de la lignede glace de l'eau d'une valeur initiale solaire (0.55) vers une valeur au voisinagede l'unité, permettant de former des planètes géantes avec des rapports C/O _ 1,comme WASP 12b. Je fnis ce chapitre en discutant les preuves observationnellesde cette enlèvement des vapeurs à l'intérieur des lignes de glaces.3. Dans le chapitre 3 j'utilise le même modèle pour interpréter la composition chimiqued'Uranus et Neptune. Je montre comment la formation de ces deux planètessur la sur-densité de glaces prédite par mon modèle sur la ligne de glace de CO peutexpliquer pourquoi ces planètes sont à la fois riches en carbone, pauvres en azote etavec des valeurs D/H sous-cométaires.4. Dans le chapitre 4 je change de sujet vers les propriétés chimiques des météoriteschondritiques, surtout leurs rapports D/H. J'utilise un modèle de disques à 2 couches(actif et morte) avec une code d'évolution D/H pour vérifier si les profiles thermiquesnon monotone trouvés dans ces disques peuvent expliquer la large gamme des valeursD/H trouvé entre les différents familles chondritiques. Je finis ce chapitre en discutantles implications de ce modèle des disques contenant des zones mortes sur laformation de Jupiter.5. Finalement je résume nos résultats dans Conclusions & perspectives, et finis enposant des questions que j'espère voir résolus prochainement. / In this manuscript I present multiple original works on planets formation theory. Themain goal is to connect the chemical composition of giant planets and small bodies to thephysical and chemical processes taking place in the protoplanetary disk.1. In chapter 1 I introduce the fundamental properties of disks and the basics ofplanets formation theory.2. In chapter 2 I tackle the supersolar C/O and subsolar C/H ratios measured recentlyin WASP 12b. I elaborate a model that tracks water and CO vapors and icesevolution through di_usion, condensation, coagulation, gas drag and sublimation inorder to quantify the variation of the C/O ratio as a function of distance and time.My model shows that, over time, vapors will get permanently depleted inside oftheir respective snowlines with CO getting depleted much slower than water. Thiswill increase the C/O ratio inside of the water snowline from the solar value of 0.55to near unity, allowing the formation of giant planets with C/O _ 1, such as WASP12b. I end this chapter by discussing the observational proofs for the existence ofsuch vapor depletions inside the icelines3. In chapter 3 I use the same model to interpret the chemical composition of Uranusand Neptune. I show how the formation of both planets on the CO snowline's icesoverdensity predicted by this model can explain why both planets are rich in carbon,poor in nitrogen and have subcometary D/H ratios.4. In chapter 4 I shift the discussion to the chemical properties of chondritic meteorites,mainly their D/H ratios. I use a snapshot from a layered (active + dead)zones disk model with a D/H ratio evolution code to check if the non monotonicthermal pro_les in these disks can explain the wide range of D/H ratios measuredin the di_erent chondritic families. I end this chapter by discussing the implicationsof the dead zone disk models for the formation of Jupiter.5. I _nally summarize my results in Conclusions & perspectives, and _nish bypointing out several relevant open questions to be hopefully resolved soon.

Simulation of physical and chemical processes in reservoirs: Two case studies.

Managing water quality aspects requires the use of integrative tools that allow a holistic approach to this problem. Water quality models coupled to hydrodynamic models are these tools. This study presents the application of the water quality model WASP coupled to the hydrodynamic model DYNHYD for two distinct reservoirs: Lake Texoma and Tocoma Reservoir. Modeling the former included simulations of water velocities, water level, and four chemical and physical compounds: chlorides, dissolved oxygen (DO), biochemical oxygen demand (BOD), and total suspended solids (TSS); and validation of the results by comparing with observed values during March - May, 1997. The latter is still under project status and the simulation was performed in a prospective way. The analysis included simulations of water velocities under current and for expected conditions, DO and BOD. Both models, DYNHYD and WASP, fitted pretty well to observed conditions for Lake Texoma and for where Tocoma Reservoir has been planned. Considering management and decision support purposes, the role of boundary and loading conditions also was tested. For Lake Texoma, controlling boundary conditions for chlorides is a determinant factor for water quality of the system. However, DO and TSS in the reservoir are governed by additional process besides the condition of the boundary. Estimated loadings for this system did not provided significant effects, even though the allocation of a load for chlorides resulted in significant changes in the trend for expected chloride concentrations at the Washita River Arm of Lake Texoma. For Tocoma Reservoir, the expected concentration of DO all over the reservoir is going to driven by boundary conditions, as well as by the management of autochthonous BOD loadings provided by vegetation decomposition. These two factors will be determinant for the resulting water quality of the future reservoir.

Esta pesquisa teve como objetivo verificar a alteração do campo de vento na área de estudo, decorrente da alteração de gabarito quanto à ventilação natural, e avaliar o impacto nas condições de conforto dos pedestres. A verificação das condições de ventilação natural nas cidades, com estudos em modelos, auxilia na elaboração de projetos arquitetônicos, ou de planejamento, possibilitando uma análise da influência do efeito do vento em determinados locais da área em análise. O objeto de estudo da tese é a relação entre a alteração de gabarito (altura dos edifícios) e as mudanças ocasionadas na ventilação natural em espaços urbanos na área entre o Canal 1 e 2 (Bairro Pompéia) na cidade de Santos/SP. Partiu-se da hipótese que a ventilação natural em áreas urbanas depende da altura dos edifícios, da direção e velocidade dos ventos. Foi utilizado método experimental com simulação da configuração urbana em túnel de vento, medidas in loco para a verificação das condições climáticas, simulação com software CFD (Computer Fluids Dynamics) e a aplicação de um índice de neutralidade térmica. Trata-se de uma tese de caráter experimental e exploratório, onde os métodos utilizados demonstraram ser aplicáveis para o entendimento das condições de ventilação natural em meio urbano. / The objective of this research is to verify alterations of the wind pattern in the study area, stemming from changes to the grids natural ventilation, and evaluate the impact to the pedestrians comfort conditions. The verification of natural ventilation conditions in cities, with the study of models, aids in the creation of architectural or planning - designs, enabling an analysis of the influence of the effects of wind in certain points of the area in study. This thesis object of study is the relation between changes to the framework (building height) and changes to the natural ventilation in urban environments in the area between Canal 1 and 2 (Bairro Pompéia), in the city of Santos/SP. The hypothesis is that natural ventilation in urban areas depends on the height of buildings, direction and velocity of winds. The study involved an experimental method, with wind-tunnel simulation of the urban grid, in loco measurements of the climate conditions, simulation with CFD software and application of a thermal neutrality index. This thesis is experimental and exploratory in its character, and the methods used proved applicable to the understanding of the natural ventilation conditions in an urban environment.

The role of protein arginine methylation in T-lymphocyte activation

T-lymphocytes are an essential cell type of the adaptive immune system. Due to their importance in immune responses and disorders, the molecular mechanisms leading to T-lymphocyte activation have been the subject of extensive research which has translated into important therapeutic developments. Early signalling events involving tyrosine phosphorylation are well characterised. However, later events involving other post-translational modifications are less well understood. Several studies have provided evidence suggesting a role for protein arginine methylation in T-lymphocyte activation. Arginine methylation is an essential post-translational modification in mammals and yet has not been extensively studied. No large scale analysis of arginine methylation sites has been performed. To gain insight into the role of protein arginine methylation in T-lymphocyte activation, the aims of this work were to: 1. Establish whether levels of arginine methylation are altered during Tlymphocyte activation 2. Use mass spectrometry based proteomics to identify arginine methylated proteins in the T-lymphocyte proteome 3. Further characterise an arginine methylated protein important to Tlymphocyte activation Arginine methylation was found to be induced after long term (>20 hours) stimulation of primary T-lymphocytes. Large increases in the main protein arginine methyltransferase, PRMT1, were also observed. Enrichment and labelling methods were developed to detect arginine methylated peptides from T-lymphocytes by mass spectrometry. This resulted in the identification of 265 unique arginine methylation sites in 141 proteins. 204 of the methylation sites were novel and 103 of the proteins had not previously been described as arginine methylated. Individual arginine methylation sites were characterised before and after activation of T-lymphocytes, with some sites showing significant changes in abundance. Among the novel arginine methylated proteins discovered were Dynamin II, WASp and WIPF1. These proteins are involved in re-organisation of the actin cytoskeleton at the immunological synapse formed between a Tlymphocyte and an antigen presenting cell. The functional consequences of the arginine methylation sites inWASp were characterised. WASp is essential for T-lymphocyte activation and some initial evidence showed that one of the arginine methylation sites is important for WASp activation.

Dynamique évolutive des symbioses protectrices chez les insectes / Evolutionary dynamics of protective symbioses in insects

Les associations symbiotiques entre microorganismes et eucaryotes sont omniprésentes dans le monde vivant. Ces microorganismes peuvent jouer un rôle crucial dans l’évolution et l’écologie de leurs porteurs en modifiant leur phénotype. Ces symbiotes étant le plus souvent héritables, les phénotypes étendus résultant de ces associations symbiotiques peuvent se transmettre aux générations suivantes. Certains microorganismes vont permettre l’accès à une ressource alimentaire, d’autres conférer une protection contre un ennemi. Une telle protection symbiotique est rencontrée chez le puceron du pois (Acyrthosiphon pisum) en interaction avec la bactérie Hamiltonella defensa. Cette symbiose confère au puceron une résistance face à l’attaque de son principal ennemi : le parasitoïde Aphidius ervi. Les populations de ce ravageur des Légumineuses sont structurées en biotypes (populations spécialisées sur des plantes hôtes). La distribution du symbiote protecteur au sein des populations de pucerons est singulière. De nombreux individus vivant sur la luzerne, la bugrane ou les genêts abritent ce symbiote alors qu’il est peu fréquent dans les populations d’autres biotypes d’A. pisum comme le pois ou le trèfle. Nous avons cherché à comprendre pourquoi H. defensa n’était pas retrouvé chez tous les biotypes du puceron du pois. Afin de prédire la dynamique de la symbiose protectrice et le potentiel de résistance dans les populations aphidiennes naturelles, nous nous sommes intéressés à plusieurs processus écologiques et évolutifs. L’incidence de la pression des parasitoïdes sur la composition des populations symbiotiques a été mesurée chez trois biotypes (luzerne, pois et trèfle) à travers une approche terrain. La distribution du symbiote H. defensa dans les populations est directement dépendante de la variabilité du phénotype associé exprimé dans les différentes populations, j’ai identifié les phénotypes associés au symbiote pour des pucerons issus de différents biotypes ainsi que l’influence du contexte local sur ces phénotypes. L’absence d’H. defensa chez certains individus peut s’expliquer par la redondance d’une fonction protectrice en place chez ces biotypes comme un alternative symbiotique autre que H. defensa ou encore une immunité forte. Enfin, nous avons testé si le cumule des protections symbiotiques conférées par deux bactéries du cortège du puceron du pois pouvaient se cumuler créant ainsi des super-organismes. Mon travail met en évidence l’implication de nombreux facteurs dans la prédiction des fréquences symbiotiques d’une bactérie facultative dans les populations d’hôte. / Symbiotic associations between microorganisms and eukaryotes are ubiquitous in the living world. These microorganisms can play a crucial role in the evolution and ecology of their hosts by altering their phenotypes. Since these symbionts are usually heritable, extended phenotypes resulting from these symbiotic associations may be transmitted to subsequent generations. Some microorganisms will allow access to a food source; others will provide protection against natural enemies. Such symbiotic protection is found in the pea aphid (Acyrthosiphon pisum) in its interaction with the bacteria Hamiltonella defensa. This symbiosis provides the aphid with a resistance against the attack of its main parasitoid enemy: Aphidius ervi. The populations of the pea aphid, a legume pest insect, are structured in different biotypes (specialized populations on host plants). The distribution of this protective symbiont within pea aphid populations is singular: many individuals living on Medicago sativa (alfalfa), Ononis spinosa or Genista sagittalis and G. tinctoria host plant with H. defensa while it is rarely found in other populations of A. pisum biotypes such as Pisum sativum (pea) or Trifolium sp. (clover). We sought to understand why H. defensa was not found in every pea aphid biotype. In order to predict the dynamics of the protective symbiosis and the resistance potential in natural aphid populations, we focused on several ecological and evolutionary processes. We measured the consequence of parasitoid stress in the composition of symbiotic populations in three different biotypes (alfalfa, clover and pea) using a field approach. The distribution of H. defensa symbiont in populations dependent directly on the variability of the associated phenotype expressed in different populations. We identified the phenotypes associated with this symbiont in aphids from different biotypes, and the influence of the local context on these phenotypes. The lack of H. defensa in some individuals can be explained by the redundancy of a protective function already in place in these biotypes, such as an alternative symbiotic species or a strong immunity. Finally, we tested whether the symbiotic protections provided by two different bacteria in the pea aphid could be cumulated, thus creating super-organisms. My work highlights the many factors involved in predicting the frequencies of facultative symbiotic bacteria in host populations.

Uma das formas de controle dessa praga é por meio do controle biológico, sendo as vespas predadoras uma alternativa viável . Desta forma, objetivou-se avaliar a resposta olfativa da vespa social Polybia fastidiosuscual (Saussure) (Hymenoptera: Vespidae) para voláteis liberados por plantas de milho e/ou da lagarta S. frugiperda. Utilizando um olfatômetro em Y, foram realizados bioensaios comportamentais para verificar a resposta comportamental das vespas sociais aos voláteis liberados por lagartas S. frugiperda, por plantas de milho, por plantas de milho induzidas com lagartas Spodoptera frugiperda e plantas de milho injúrias mecânicas e/ou tratadas com regurgito de lagartas ou água. Observou-se que as vespas sociais não tiveram preferência significativa quando ofertou-se, ar, lagartas, plantas de milho sem indução ou plantas de milho injuriadas mecanicamente tratadas com água. No entanto, as vespas preferiram plantas induzidas por lagartas S. frugiperda ou plantas injuriadas mecanicamente tratadas com regurgito de lagartas durante os intervalos de 5-6 horas e 24-25 horas. O mesmo não foi observado quando as plantas foram induzidas por lagartas ou injuriadas mecanicamente tratadas com regurgito de lagarta no intervalo de 1-2 horas. Estes resultados foram confirmados quando ofertou-se os extratos plantas de milho induzidas por lagartas S. frugiperda ou plantas de milho injuriadas mecanicamente tratadas com regurgito de lagartas após 5-6 horas de indução versus o hexano como controle. Em uma segunda fase os extratos de plantas de milho sem indução, plantas de milho com a presença de S. frugiperda por 1 hora, plantas de milho com a presença continua de S. frugiperda e de plantas de milho injuriadas mecanicamente tratadas com regurgito de lagartas foram quantificados por cromatografia gasosa acoplada a detecção por ionização de chamas (CG-DIC) e analisados por cromatógrafo gasoso acoplado a espectrômetro de massas (CG-EM) utilizando o gás hélio como gás de arraste. Os dados foram coletados e avaliados com o software ChemStation. E a identificação dos compostos dos extratos das plantas de milho foi feita por comparação dos tempos de retenção no CG-DIC com os dos padrões usando coluna apolares e cálculo do índice de Kovats. / The caterpillar Spodoptera frugiperda (Smith, 1797) (Lepdopitera: Noctuidae) is a pest in corn, affecting up to 60%, the grain yield. One way to control this pest is through biological control, and predatory wasps a viable alternative. Thus, we aimed to evaluate the olfactory response of the social wasp Polybia fastidiosuscula (Saussure) (Hymenoptera: Vespidae) to volatiles released by maize plants and / or caterpillar S. frugiperda. Using a olfactometer-Y, behavioral experiments were conducted to verify the behavioral response to volatile social wasps released by S. frugiperda larvae by corn plants, induced by corn plants with caterpillars Spodoptera frugiperda and corn plants mechanical injuries and / or treated with caterpillars regurgitate or water. It was observed that the wasps did not have significant social preferably when offered up, air, caterpillars, corn plants without induction or mechanically injured maize plants treated with water. However, wasps preferred induced by S. frugiperda larvae or mechanically injured plants treated with caterpillars of regurgitation during intervals 5-6 hours and 24-25 hours plants. The same was not observed when plants were induced by caterpillars or injured mechanically treated with caterpillar regurgitate in the range of 1-2 hours. These results were confirmed when offered up corn plant extracts induced by S. frugiperda larvae or corn plants injured overflow, mechanically treated larvae after 5-6 hours of induction versus hexane as control. In a second phase extracts of corn plants without induction, corn plants with the presence of S. frugiperda for 1 hour, corn plants with the continued presence of S. frugiperda and corn plants injured mechanically treated regurgitate Crawler were quantified by gas chromatography-flame ionization detection (GC-FID) and analyzed by gas chromatography coupled to mass spectrometry (GC-MS) using helium as the carrier gas. Data were collected and evaluated with the ChemStation software. And the identification of compounds of extracts of maize plants was made by comparison of retention times in GC-FID with patterns using nonpolar column and Kovats index calculation.

The plots of Billy Budd and The Bonfire of the Vanities are organized entirely around a lawsuit. In The Pickwick Papers the trial is only a part, though an important one, of a series of related adventures in which the main characters of the novel participate. In the three novels there is a trial in which the accused is found guilty, although he is actually innocent. In The Posthumous Papers of the Club Pickwick, the author’s main purpose is to present the operation of the legal system, in which the modus operandi of unscrupulous lawyers, who rely only on cheating and deceiving methods, is atthe beginning of and determines the outcome of the lawsuit. In Billy Budd, an innocent is sentenced to death in order to preserve a supposed higher interest: the common good. In The Bonfire of the Vanities, political factors, personal interests, resentments and other worldly elements determine the outcome of the trial. In the three cases, the watchmaking mechanism that a lawsuit appears to be is completely overcome by factors outside it. / Las tramas de Billy Budd y La hoguera de las vanidades están organizadas íntegramente alrededor de un juicio.
