Lalancette, Simon 20 April 2018 (has links)

Examination of Native American remains in east central Indiana through mitochondrial DNA analysis

Rapier, Brandon S. January 2006 (has links)
Mound building was a common practice in the Midwestern United States among the Adena and Middle Mississippian tribes from approximately 500 B.C. to 1500 A.D. Though they varied greatly in size, shape, and complexity between cultures, the mounds served a common purpose as means to bury the deceased. Mounds representing both cultures have been found dispersed throughout Indiana, in areas such as Randolph County, Henry County, Madison County, Knox County, and Vanderburgh County.Of particular interest in this investigation is an Adena burial plot, known as Windsor Mound, which is 2000 yrs old and located in neighboring Randolph County. An amateur excavation of the mound from 1986 to 1988 unearthed the fragmented remains of 44 individuals and several artifacts, all of which were loaned to the Ball State University Anthropology Department. Documentation of the excavation was poor and the exact location of artifacts and remains within the mound were not recorded, leaving anthropologists with many unanswered questions that could only be answered through genetic analysis. Pressing questions were the degree to which the 44 individuals were related and which of the five Native American lineages (haplotypes) they belonged to.For this analysis, dentin was recovered from the teeth of three Native American individuals exhumed from Windsor Mound. Two of the individuals (87.17.5 and 87.17.10) were found in the lower mound portion (70 B.C.), while the third individual (87.17.22) was found in the upper mound cap (1180 A.D.). Ancient DNA (aDNA) was extracted from each dentin sample, yielding an average of 0.072 ug/mg of dentin. A highly polymorphic portion of the mitochondria) DNA control region (nt 16,049 - nt 16,221) termed hypervariable region 1 (HVI) was amplified via PCR to generate 172 bp amplicons which were cloned into a plasmid vector. Following a transformation, 10 clones from each individual were sequenced and aligned to identify consistent mutations, as opposed to random post-mortem damage that may have occurred.Sequencing of the HVI region for Individual 87.17.5 revealed a T —~ C base substitution at nt 16,189, a C — T substitution at nt 16,192, and a deletion at nt 16,203. Identical mutations were seen in Individual 87.17.22 from the upper mound cap. The alignment for Individual 87.17.10 revealed a unique T --~ C mutation at nt 16,126 as well as the deletion at nt 16,203. A literature search revealed that the substitution at nt 16,189 is specific to Native American 1-laplogroups B and X. Haplogroup B first appeared in central Asia 60,000 yrs ago while Haplogroup X appeared in western Asia 30,000 yrs ago. The substitution at nt 16,192 is indicative of East Asian origin and specific to Japanese populations. To our knowledge the deletion at nt 16,203 has not been reported before in a Native American, thus its presence was thought to indicate kinship between the individuals. However, an alignment of the ancient consensus sequences to that of the three investigators revealed that the deletion was present in two of the modern samples and was not novel. Nothing is known about the substitution at nt 16,126. Further sequencing downstream of nt 16,221 is needed to identify additional mutations characteristic of Haplogroup B or X and a novel kinship marker. / Department of Biology

Vanishing Acts: Absence, Gender, and Magic in Early Modern English Drama, 1558-1642

Dell, Jessica 19 November 2014 (has links)
This dissertation examines how early modern English playwrights employ absence to enrich their representations of the unknown, including witchcraft and the supernatural. Throughout the sixteenth and seventeenth centuries magical themes were often dramatized through visual and linguistic excess. Whether this excess was manifested through the use of vibrant costumes, farcical caricatures, or exaggerated dialogue, magic was often synonymous with theatricality. Playwrights such as William Rowley, Ben Jonson, and William Shakespeare, however, challenge stereotypical depictions of magic by contrasting excessive magic with the subtler power of restrained or off-stage magic. Embedded in the fantastical events and elaborate plots of Shakespeare and his contemporaries, absence, whether as an unstaged thing or person or an absent ideology, becomes a crucial element in understanding how playwrights represented and understood occult issues during the early modern period. Further, when gendered feminine, magical absences serve to combat oppressive silences within scripts and provide female subjects with an unimpeded and inherently magical space from which to challenge pre-established patriarchal systems of control. Each chapter in this dissertation, therefore, appraises the magical possibilities that theatrical absences provide to women as a platform from which to develop their narrative voice. Partnered with a complementary discussion of Jonson’s The Masque of Queens and two thematically linked witchcraft cases, my first chapter argues that Mistress Ford uses the complete stage absence of both a witch and a queen in The Merry Wives of Windsor to reform her community and critique her society’s unjust categorization of women. In chapter two, I examine a series of “vanishing acts” in The Birth of Merlin and argue that Rowley’s female characters use their final moments on stage to contextualize their impending absences for audiences as moments of magical defiance rather than defeat in the face of male tyranny. In my final chapter, I look at how magical objects, such as the handkerchief in Shakespeare’s Othello or the belt in Jonson’s The Sad Shepherd resist the absence of their female creators and continue to provide physically absent or dead women with magical agency. By structuring my dissertation on these three specific gradations of absence, I provide a nuanced analysis of the purposes these dramatic omissions serve by focusing on how these shades of absence subtly alter the ways in which we interpret and define early modern magical belief. / Thesis / Doctor of Philosophy (PhD)

Estimation of the characteristic in-situ compressive strength class of concrete structures - A case study of the Skuru bridge / Bedöming av den karaktäristiska tryckhållfasthetsklassen för betong konstruktioner - en fallstudie av Skuru bron

Ahmadiyan, Sara, Mehari, Daniel January 2021 (has links)
It is inevitable that Structures become older and their intended use changes or the structuralcodes regulations change. In some regions the damage from seismic activities is a possibility. It becomes, therefore crucial to assess the structural capacity of such structures. The purposeof this study is to assess the different methods used for testing and estimating thecharacteristic in-situ compressive strength which is the most vital parameter required instructural assessment. The focus of the study is for existing structures where there is no prior knowledge about theconcrete strength. This study first investigates and evaluates the merits and demerits of thesemethods for investigation of the condition of in-situ compressive strength of concrete inexisting structures. A case study of the Skuru bridge that was built in 1914 was utilized forthis study. The study is based on information of the construction data and some results fromprior investigation performed by the company COWI. Afterwards, non-destructive tests werecarried out with the UPV and Rebound hammer to assess the quality of the concrete. In addition, the study assesses the use of different interpretation methods with regards toreliability and practical application. The results were interpreted in accordance to theEuropean codes, Swedish codes and other interpretation methods. The difference of theresults from the different interpretation methods are compared and evaluated for reliabilityand efficiency. The test results confirmed that the concrete consisted of the same strength class. However, the results from the different interpretation methods are dissimilar. The reason for obtainingdifferent results is because the methods depend on different methodologies. The studyshowed that some methods can sometimes overestimate the results and become unsafe forstructural assessment. On the contrary, the other methods can yield lower but safer estimates. Moreover, the use of small number of cores is evaluated for various methods. The reasons arebecause in practice, the preference is to avoid large number of cores. As a result, it isrecommended to apply care and proper judgment in selection of the methods andinterpretation of the results. It is also recommended to consider the methods with respect tothe aim of the investigation, their limitations and assumptions. Åandra sidan, resulterade vissa tolkningsmetoder i lägre men säkrare uppskattning avtryckhållfastheten.
