Mutações em genes da via de reparo de DNA por excisão de nucleotídeo estão associados a doenças genéticas raras, como Xeroderma pigmentosum, Síndrome de Cockayne e Tricotiodistrofia. Este trabalho teve como objetivo identificar mutações responsáveis por essas Síndromes em pacientes brasileiros. Inicialmente o projeto teve como foco o estudo da comunidade de Araras, no interior de Goiás, onde existe uma das maiores incidências de pacientes XP no mundo. Duas mutações foram identificadas no gene POLH, associadas a dois efeitos fundadores independentes na região. Os dezessete pacientes, entre 10 e 80 anos, caracterizados geneticamente apresentam uma grande heterogeneidade das manifestações clínicas, com fenótipos brandos à extremamente agressivos. Esse estudo se estendeu a mais dezessete pacientes de todo o Brasil, incluindo outros grupos de complementação de XP, CS e TTD. Na segunda parte deste trabalho avaliamos a resposta das linhagens XP-V ao tratamento com cisplatina. Foi possível observar que essas linhagens são mais sensíveis aos danos induzidos pela cisplatina, encontram problemas na fase G1/S de replicação e possuem um aumento da marcação high-level para histona gH2AX. / Mutations at DNA repair genes are associated with rare genetic diseases such as Xeroderma pigmentosum, Cockayne syndrome and Trichothiodystrophy. This study aimed to identify mutations responsible for these syndromes in Brazilian patients. Initially the project focused on the study of Araras community in the state of Goiás, where there is one of the densest known areas of XP patients. Two mutations were identified at POLH gene, associated with two independent founders effects in the region. The seventeen patients between 10 and 80, characterized genetically a wide range of heterogeneity of clinical symptoms with mild to extremely aggressive phenotypes. Brazilian patients from other places were studied, including seventeen other patients with XP, CS and TTD phenotypes. In the second part of this work, we evaluated the response of XP-V cell lines treated with cisplatin. It was observed that Pol eta deficient cells are more sensitive to damage induced by cisplatin, as well as encounter problems in the S phase replication after 24 hours of treatment, and have increased high-level fluorescence for gH2AX.

The aim of this study was to evaluate the efficacy of heat-treated nickel-titanium reciprocating instruments and complementary techniques in retreatment of flattened canals. Material and methods: Twenty-six mandibular incisors were prepared up to size 25/.08 rotary file and filled using the single-cone technique. Subsequently, the teeth were divided into 3 groups (n=12) according to retreatment procedures: Reciproc Blue: R25 and R40; ProDesign R: PDR 25/.06 and PDR 35/.05; WaveOne Gold: WOG Primary and WOG Medium. The apically extruded material was collected in microtubes and the volume (mm³) was measured by Micro-CT. After this, the specimens received complementary cleaning with XP-Endo Shaper, PUI and 60° oscillatory instrumentation with #30 Hedströem file. Micro-CT scans were performed before and after removing filling material. After quantification of the volume of remaining filling material, the teeth were sectioned and scanning electron microscopy (SEM) images of the cervical, middle and apical thirds were obtained with a 700X magnification, and evaluated by scores based on the amount of filling material in the surface of dentine walls. Results: No reciprocating instrument promoted complete removal of filling material. All systems showed extrusion, however, WOG25 presented the higher extrusion of material (P<.05). XP-Endo Shaper significantly reduced the amount of filling material in the apical and middle thirds (P<.05). In the SEM analysis, there was no statistical difference among the cleaning methods (P> .05). Conclusions: The reciprocating instruments studied did not promote complete removal of root canal filling materials. XP-Endo Shaper, used as a complementary instrument in retreatment, improved removal of filling material. / O objetivo deste estudo foi avaliar a eficácia de instrumentos reciprocantes em níquel-titânio tratados termicamente e métodos complementares no retratamento de dentes com raízes achatadas. Materiais e métodos: Trinta e seis incisivos inferiores foram preparados com instrumentos rotatórios até o diâmetro 25./08 e obturados pela técnica do cone único. Em seguida, os dentes foram divididos em 3 grupos (n=12) de acordo com os sistemas utilizados para desobturação no retratamento: Reciproc Blue: R25 e R40; ProDesign R: PDR 25./06 e PDR 35/.05; WaveOne Gold: WOG Primary e WOG Medium. O material extruído apicalmente foi coletado em microtubos e o volume (mm³) foi mensurado com Micro-CT. Na sequência, os espécimes receberam limpeza complementar com XP-Endo Shaper ou com irrigação passiva ultrassônica (PUI) ou com instrumentação com lima #30 Hedströem em movimento oscilatório. Escaneamentos em Micro-CT foram realizados antes e após a remoção do material obturador. Após quantificação do volume de remanescente de material obturador, os dentes foram seccionados e imagens com microscópio eletrônico de varredura (MEV), dos terços cervical, médio e apical, foram obtidas com aumento de 700X, e avaliadas através de escores baseados na quantidade de material obturador remanescente nas paredes dentinárias. Resultados: Nenhum instrumento reciprocante promoveu remoção completa do material obturador. Todos os sistemas demonstraram extrusão, no entanto, WOG25 proporcionou maior extrusão (P<.05). XP-Endo Shaper reduziu significantemente a quantidade de material obturador nos terços médio e apical (P<.05). Na análise da limpeza das paredes dentinárias, em MEV, não houve diferença estatística entre os métodos de limpeza (P>.05). Os instrumentos reciprocantes estudados não promoveram a remoção completa do material obturador. XP-Shaper, utilizado como instrumento complementar no retratamento, aumentou a remoção de material obturador.

Although genetic and epigenetic changes can activate proto-oncogenes, inactivate tumor suppressor genes and repair mechanism genes, the cell has several mechanisms that contribute to the maintenance of genomic stability. Mutations in repair genes can lead to tumor progression and loss of genome integrity leading to the onset of cancer. Nucleotide excision repair (NER) is an important mechanism primarily used to repair injuries caused by UV. The objective of this study was to evaluate single nucleotide polymorphisms (SNP) of XPA and XPC genes and the risk of developing BCC. One hundred samples of paraffined tissue from patients from the State of Paraíba with histopathological diagnosis of BCC were analyzed for each polymorphism. The results were obtained by a newly developed genotyping method, the Dideoxy Unique Allele Specific - PCR, a method that presents high sensitivity and low cost. Graphpad Prism 6.01 software was used for the statistical analysis and application of Chi-square and Fisher's exact test. The SNP rs535425175 of the XPC gene showed a significant association with the BCC in the analyzed samples (X2 = 14.51 and P <0.005). Whereas the SNPs rs745769173 of the XPA gene and rs761106780 of the XPC gene are in the Hardy-Weinberg equilibrium, not showing any association with the neoplasia. The results suggest that the SNP rs535425175 of the XPC gene may be considered a risk factor associated with the development of BCC. / O Carcinoma Basocelular (CBC) é uma neoplasia frequente em seres humanos e seu principal fator etiológico é a exposição à radiação solar. Embora alterações genéticas e epigenéticas possam ativar proto-oncogenes, inativar genes supressores de tumor e genes do mecanismo de reparo, a célula apresenta vários mecanismos que contribuem para a manutenção da estabilidade genômica. Mutações em genes de reparo podem levar a progressão tumoral e à perda da integridade do genoma levando ao surgimento do câncer. O reparo por excisão de nucleotídeo (NER) é um importante mecanismo utilizado principalmente para reparar lesões causadas por UV. O objetivo deste trabalho foi avaliar polimorfismos de nucleotídeo único (SNP) dos genes XPA e XPC e o risco de desenvolver CBC. Foram analisadas 100 amostras de tecido parafinado de pacientes do Estado da Paraíba com diagnóstico histopatológico de CBC para cada polimorfismo. Os resultados foram obtidos por um método de genotipagem recentemente desenvolvido, o Didesoxi Único Alelo Específico – PCR, método que apresenta alta sensibilidade e de baixo custo. O software Graphpad Prism 6.01 foi utilizado para as análises estatísticas e aplicação de teste Qui-quadrado e Exato de Fisher. O SNP rs535425175 do gene XPC apresentou associação significativa com o CBC nas amostras analisadas (X2=14,51 e P<0,005). Enquanto que os SNP rs745769173 do gene XPA e rs761106780 do gene XPC estão no equilíbrio de Hardy-Weinberg, não apresentando associação com a neoplasia. Os resultados sugerem que o SNP rs535425175 do gene XPC pode ser considerado um fator de risco associado ao desenvolvimento de CBC. Vi har i detta projekt arbetat med att utforma ett robust och säkert system som är byggt med hjälp av klient-server arkitekturen åt Tweakers HB. Tweakers HB är ett nytt företag som utvecklar allt från mobila till stationära applikationer. Produkten som utvecklats är ett ordbaserat frågesportsspel som har multiplayer funktionalitet. Applikationen/klienten var utvecklat för mobiler som använder Android plattformen och server bestod av ett REST API och en MYSQL databas. Klient-sidan byggdes upp med en anpassad tillämpning av MVC mönstret och använde SQLite för att spara data lokalt. Målet med projektet kommer vara att utvecklat ett system som kommer att ha en lång livslängd som även kan byggas ut i framtiden. För att kunna uppnå målet med produkten så användes utvecklingsmetoden XP(Extreme Programming) och test-driven utveckling. Företaget la vissa krav på tekniker som skulle användas men i stort sett så låg ansvaret på utvecklingsgruppens att hitta tekniker och biliotek för att uppnå kraven. En stor del av kraven blev uppfyllda. / We have in this project worked on designing a robust and reliable system that is built using the client-server architecture for Tweakers HB. Tweakers HB is a new company that develops everything from mobile to desktop applications. The product developed is a word based quiz game with multiplayer functionality. The application/client was developed for mobile phones using the Android platform and the server consisted of a REST API and a MySQL database. The client-side was built using a custom implementation of the MVC pattern and used SQLite to store data locally. The goal of the project was to develop a system that will have a long life span that could also be expanded in the future. In order to achieve the goal of the product XP (Extreme Programming) development methodology and test-driven development was used. The company put certain requirements for technologies that could be used but basically the responsibility fell upon development team to find techniques and libraries to achieve the requirements. Much of demands where satisfied.

現今軟體開發專案普遍有時程延宕、預算及人&#63882;&#63847;足、軟體品質低&#63768;、無法應付客戶頻繁的需求變動等問題。為思考相關解決之道，本研究&#63851;考極致編程(Extreme Programming，簡稱XP)軟體開發方法，於校園內選擇特定專案，配合XP的四個核心價值觀與十二項實&#64008;原則實際進&#64008;軟體開發。於專案導入XP後，透過對開發過程的觀察、記&#63807;、資訊蒐集，及針對開發團隊所作的深入訪談，本研究綜合分析與評估XP於校園內軟體開發專案之適用性，並發掘可能的問題。 　　研究中發現XP在校園內&#63847;&#63968;達到完美實踐之目標，適用性&#63847;高，主要受限於時空環境難以配合、團隊專案經驗&#63847;足、缺乏激&#63871;誘因、缺乏團隊約束&#63882;等因素；&#63847;過施&#64008;XP也改善&#63930;團隊內外部的溝通與協調機制，促進團隊成員之間的知&#63996;與經驗分享，亦有&#64023;於系統架構之完整性。 　　本研究排除&#63930;完全無法實施的要項之後，依團隊實際情況提出&#63860;干改善及彈性應變之建議做法，以期將XP進一步應用於業界個案及後續學術研究。

Lean Software Development is a new approach to provide a quality philosophy in software development companies. Lean offers a set of principles that helps companies to identify inefficient processes and waste. Therefore, by applying these principles, we can reduce costs, by shorting development time, incrementing productivity.In this study a software company with seven employees is analyzed. The company develops software and wants to increase its maturity and the quality of its processes. Currently, it has few documented processes. Furthermore, the company want a common way of working for every project. The challenge relies in adapting the set of principles provided by Lean to this particular case of study.We seek to analyze the current situation of the company to find out about the problems and limitations of the current way of working. After that we will state recommendations about the use of Lean combined with Agile practices such as Scrum and XP.As a result we present a proposal for implementation adapted from these philosophies to the needs and characteristics of the company.We have found that there are several ways in which the company can benefit from the implementation of Lean practices in combination with Scrum and XP. The result is a new framework that can be applied for other small software development companies in a similar situation.
