Metabolomics investigation of microbial cell factories

The stream of new technological advancements and their integration into the field of microbiology have contributed significantly towards our understanding of life in the micro-scale world, making the fields of microbiology and biotechnology shine like never before. Since 1980, the recombinant protein-based therapeutics industry has become one of the fastest growing sectors in the biopharmaceutical market. Nearly 30% of commercially available recombinant proteins are produced in Escherichia coli, making this species one of the most commonly used bacterial expression systems for the production of recombinant biotherapeutics. However, when it comes to the production of enzymes and bioactive secondary metabolites (antibiotic, antifungal, antiviral and immunosuppressant), Streptomyces species remain the major producer within this sector. Meeting the high demand for such products requires a clear and in-depth understanding of the bioprocesses involved to achieve high yield and quality products, whilst keeping the process industrially attractive. It is generally accepted that the metabolome, as a down-stream process to the genome and proteome, may provide a clearer picture of a biological system. Thus, in this thesis a series of metabolomics approaches were adopted to obtain a deeper insight into the metabolic effects of recombinant protein production in E. coli and Streptomyces lividans. Furthermore, a Geobacter-based biomagnetite nanoparticle production system which displayed a prolonged lag phase upon scale-up was investigated by employing metabolic profiling and fingerprinting approaches combined with multivariate analysis strategies, to identify growth-limiting metabolites. The results of this analysis identified nicotinamide as the growth limiting metabolite. Nicotinamide-feeding experiments confirmed the above findings, leading to improved biomass yield whilst restoring the lag phase to bench-scale level. Raman and Fourier transform infrared spectroscopies combined with stable isotopic probing strategies were also employed to demonstrate the application of metabolic fingerprinting in providing detailed biochemical information for quantitative characterisation and differentiation of E. coli cells at community and single-cell levels. The single-cell approach proved promising, offering detailed biochemical information and perhaps accompanying other cultivation-free approaches such as metagenomics for further future investigations. It is hoped that the advances made in these studies have proved the potential applications of metabolomics strategies to aid the optimisation of microbially-driven bioprocesses.

<p>Syftet med denna uppsats är att undersöka analysverktyg med fokus på LUS (LäsUtvecklingsSchema) samt att söka kunskap om pedagogers nytta med användandet. Vi vill även ta reda på vilken hänsyn man som pedagog tar i användandet av analysverktyget, med tanke på elevers modersmål, tidigare erfarenheter och förståelse av skriftspråket. Genom att använda oss av relevant litteratur och enkäter med öppna frågor, vill vi ge ett kvalitativt resultat av problemområdet. De pedagoger som deltagit i studien har olika erfarenheter av LUS och kan därför ge en nyanserad bild av LUS i praktiken. Uppsatsen börjar med en bakgrund bestående av vilken betydelse läsning har i samhället, vad styrdokumenten säger samt beskrivning av analysverktyg. Med de ämnen vi behandlar i teoridelen ger vi en bild av läsinlärning, tidigare forskning och Nya LUS-boken. Resultatet och analysen visar att LUS-användningen är utbredd i skolor och att LUS är ett bra bedömningsinstrument om man tar i beaktande dess begränsningar, såsom att LUS inte tar hänsyn till att alla elever har olika bakgrund.</p> / <p>The purpose of this investigation is to look into analytical tools biased towards reading development. Our aim is also to seek knowledge about how teachers actually use the analytical tools. We also want to find out what considerations are made in the usage of the tools, regarding children’s native language, earlier experience and understanding written language. We will, with relevant literature and an opinion poll which contains questions without given answers, create a quality based study that answers the investigations question at issue. The teachers who has participated in this essay, can with their different experience in using analytical tools, bring light to different perspectives and therefore give a good picture of analytical tools in use. Our investigation begins with a background which contains the significance in reading, curriculums and a escription of analytical tools. In the following part you can read about learning how to read, former research and a description of the book named, “Nya LUSboken”, which is about one sort of analytical tools focusing reading. The results show that almost every school use LUS and that it’s a good tool if you consider it’s limits regarding different pupils backgrounds.</p>

BINARY BRIGHT-LINE DECISION MODELS FOR GOING CONCERN ASSESSMENT: ANALYSIS OF ANALYTICAL TOOLS FOR BANKRUPTCY PREDICTION CONSIDERING SENSITIVITY TO MATERIALITY THRESHOLDS

In August, 2014, the Financial Accounting Standards Board issued an update concerning the disclosure of uncertainties about an entity’s ability to continue as a going concern. The standard requires an entities management to evaluate whether there is substantial doubt about the entity’s ability to continue as a going concern and to provide related footnote disclosures in certain circumstances. One consequence of this regulation is the need for guidance for audit testing of management’s assessments in each phase of the audit. This research evaluates the usefulness of bankruptcy prediction models as analytical tools in the planning stage of an audit for going concern assertions and questions the use of precision as the only measure of a model’s effectiveness. I use simulation to manipulate the fundamental accounting data within five bankruptcy prediction models, explore failure rates in an environment with materiality concerns, and consider the total change in market value due to simulated errors. Given the inherent limitations of the information environment and/or current prediction models, my results indicate auditors’ current failure rates are not an indication of audit failure. The results suggest that bright-line testing using bankruptcy prediction models are sensitive to materiality and that the cost trade-off between Type I and Type II errors is an important indicator of model choice.

Establishing Guidelines for Usability Testing when Using Web Analytical Tools

Usability and User Experience has become increasingly important. Users should be able to achieve their set out goals with ease, whether that is to purchase a book on an e-commerce site or to find articles about current events in Europe on a news site. Achieving good usability is a task most developers and designers face today and to address that task they employ the use of usability evaluation methods. Lately, web analytical tools have shown some potential in usability evaluation. This thesis aims to explore if usability can be evaluated with Web Analytical Tools by comparing their benefits and limitations with classical usability evaluation methods. The result of this thesis is a set of guidelines meant to help smaller companies with their design phase. It was found that Web Analytical Tools can be used in some cases to realize usability problems, but to understand and fix the problems it is essential to conduct usability tests with users, so called user testing.

Carbon footprint decomposition in MRIO models: identifying EU supply-chain hot spots and their structural changes over time

Politics' demand for informative consumption-based emission assessments based on multi-regional input output (MRIO) databases is steadily increasing. Based on the MRIO database EXIOBASE 3, we exemplify the utility of a range of analytical tools and discus their potential insights for consumption-based policies. The analysis decomposes the overall EU carbon footprint into product groups as well as into emitting regions. Subsequently, we illustrate the potential of applying production layer decomposition (PLD) and structural path analysis (SPA) for the assessment of global supply-chains related to the EU carbon footprint and their structural changes over time. We close with some policy ecommendations on reducing carbon footprint hot spots. / Series: Ecological Economic Papers

Few studies have a focus on progression in teaching and learning mathematics. An assumption for this study was that progression in teaching between school stages was important. The approach of the study was based on action research. Four teachers from preschool to 9th grade (age 1-16) were invited to a temporary team, a community of practice. The overall aim of the study was to develop knowledge about teaching fractions when teachers used students’ understandings as a point of departure for their action plans. A second aim was to illuminate what influences progression in their teaching. The team of teachers used the four phases of action research: plan, act, observe and reflect, during their learning processes. The teachers’ learning sessions were videotaped and transcribed and this provided the main data that formed the basis of the results. Wenger’s dimensions of social learning were used as an analytical tool: joint enterprise, mutual engagement and shared repertoire. Four themes that described teachers’ negotiation of qualities in mathematics instruction were identified: interpreting students’ understandings, basing instruction on students’ understandings, visualizing fractions and ensuring students’ understanding. When teachers, regardless of what stage was involved, reified similar instructions, it did not benefit students’ learning opportunities. In order to improve progression in teaching fractions, it was important that teachers succeeded in identifying students’ understandings and that the team negotiated different qualities in their community of practice. The shared repertoire (the pre-tests and the video recordings) formed the core of negotiating progression based on students’ understandings. The team showed a mutual engagement, with students’ learning as their joint enterprise. An implication of the study is that teachers from different educational stages can negotiate progression and improve it.

This thesis deals with the issue of social networks in terms of business support and their use for commercial purposes. Based on long-term statistics from the most widely used social networks in the Czech Republic, this thesis describes the main principles of using these social networks as a means of increasing profit or turnover. Particular attention is paid to Facebook, the leading social network in the online world. This study concerns the combination of marketing and the IT world, which nowadays has a key role in the sale of products and services. This study should show the importance of the combination of these two worlds. The practical part of the study is the analysis of the communication of the RunCzech Running League (formerly the Prague International Marathon), including a detailed analysis of one their competitors on the market. For the analysis appropriate metrics were selected, which were measured using special tools. The conclusion includes a comprehensive evaluation and presents suggestions for more effective future communication of the RunCzech running leagues in the field of social networking.

The CRM system is relevant for various reasons: the development of business processes, the role of the Internet and the use of mobile devices in business, the development and reduction of the cost of data processing technologies and IT infrastructure. And, of course, the fact that the technical capabilities of CRM systems have reached perfection played an important role. They allow you to track the path of the buyer from the moment of the first contact with him for resale. It creates sales technology by linking advertising, analytical tools and the sales system itself into a single system. The results of the final qualifying work will be used in the photo studio when implementing the AppEvent CRM system.
