What impact does the conversation between citizen, market and governance have on inherent meaning in cultural heritage? How does it manifest and unfold concerning values it presents in the given context of Slussen´s preservation and reconstruction? As cultural beings, what significance does these values have on democracy? The aim of this studie is to measure how the concept of cultural heritage is negotiated and established by the city of Stockholm, Skanska Sweden and relating practitioners towards the significance of democracy. This with the approach of archival studies from e-archive Stockholm and an analysis of records from Skanska Sweden in combination with an ethnological perspective. This conversation exposes that the representational democracy which city of Stockholm represents endangers its senior citizens with exclusion. The values that late modern society evokes is in discrepancy with which values Stockholm´s senior citizens stands up for, relating back to the aftermath from World war II. The results points out that Stockholms collective memory aims to wipe out values which the ladder stands for, acting within Slussens cultural heritageprocess.

”Ett nationalromantikens ’wonderland’” : Användandet av autenticitetsbegreppet i samband med ombyggnationen av Nationalmuseum

This thesis aims to investigate how the meaning of the concept of authenticity has been used in arguments about the reconstruction of Nationalmuseum in Stockholm, Sweden. The museum, which opened for the first time in 1866, was closed for restoration, renovation and other engagements during 2013-2018. After the reopening, a debate took place in the Swedish newspaper Svenska Dagbladet. In the debate there were people who argued that the reconstruction had erased the museum’s history and replaced it with modern design. Other people argued that the reconstruction had made the building more true to its history as older reconstructions had distorted the original building. The main source material for the present study consists of a final report of the project made by the National Property Board, selected debate articles from the debate on the reconstruction in Svenska Dagbladet, and an interview and two mail conversations that have been made with persons involved in the project. Through argumentation analysis, arguments from the final report, the debate in Svenska Dagbladet, the interview and the mail conversations have been analyzed. The arguments selected from the source material are arguments that either indicate that the authenticity has been preserved or that the authenticity has not been preserved. The thesis does not intend to determine whether the reconstruction of Nationalmuseum has preserved the authenticity or not, it is the argumentation about this issue that is to be investigated. The reason is that the perception differs between different people, different contexts and different views. The analysis shows that the arguments claiming that the authenticity has not been preserved above all relate to the building's younger reconstructions with its additions that were removed during the current reconstruction. Some debaters argue that the building's younger history also has a value that should be protected. In the end it comes down to what kind of authenticity one intends to preserve. In the project, the experience values have been prioritized before the material values, which indicates that the authenticity from a material perspective may have decreased while the functional authenticity of the building has increased. The arguments that favour that the authenticity has been preserved are primarily based on the view that great emphasis has been placed on the preservation of the building's cultural-historical value. During the project one has also been inspired by the building's original drawings made by the building's architect Friedrich August Stüler, which indicates that the authenticity of the original building has been strengthened. The thesis ends with a final discussion where the result of the analysis is presented in relation to the theoretical framework of the thesis. This is done in order to answer the research questions. The main research question for the thesis has been; "How has the meaning of authenticity been presented in the final report, in the debate and by people who have worked with the reconstruction of Nationalmuseum?”.

Dada la internalización y normalización de los estándares de belleza que implantan un tipo de cuerpo ideal para todas las mujeres en la actualidad, y el crecimiento de una población más consciente sobre aquello que es real o no, el movimiento Body Positive se mantiene luchando contra los cánones clásicos de belleza que parecen no dejar de expandirse, optando por representar al cuerpo de la mujer como bello, sin importar el aspecto, color, talla,etc. Por lo que el presente trabajo analiza la influencia de esta representación del cuerpo bajo la tendencia Body Positive en la percepción de autenticidad de marca. De esta forma, y bajo el análisis de literatura, se busca analizar a las marcas que han optado por una estrategia publicitaria Body Positive y sus repercusiones a nivel de usuario. Así mismo, cómo estas repercusiones podrían estar vinculadas directamente con la percepción de una marca como auténtica y/o real. De la misma forma, se estudia los aspectos de una posible marca auténtica y cómo podría afectar esto en el comportamiento del consumidor. / Given the internalization and normalization of beauty issues that implant an ideal body type for all women today, and the growth of a population more aware of what is real or not, the Positive Body movement continues to fight against classic canons of beauty that seem to not stop expanding, choosing to represent the woman's body as beautiful, regardless of appearance, color, size, etc. So the present work analyzes the influence of this representation of the body under the positive body trend in the perception of brand authenticity. In this way, and under the analysis of literature, we seek to analyze the brands that have opted for a positive body public strategy and its repercussions at the user level. Likewise, how these repercussions can be directly linked to the perception of a brand as authentic and / or real. In the same way, study the aspects of a possible authentic brand and how this could affect consumer behavior. / Trabajo de investigación

Erwin Bodky as Musicologist, Pedagogue, and Performer: A German Émigre in The United States

Before 1933, Erwin Bodky actively participated in musical life in Berlin. When he was a student, the Prussian Government had given him grants in 1920 and 1921 to study with the distinguished composers Richard Strauss and Ferruccio Busoni. His international performing career was launched when, at the last minute, Bodky was asked to replace a pianist for a performance with Wilhelm Furtwängler. When he became a professor at the Staatlich Akademie für Kirchen- und Schulmusik in Charlottenburg in 1926, he immersed himself in early music, performing on harpsichord and clavichord and founding his own collegium musicum. His publications, Der Vortrag alter Klaviermusik (Berlin: Hesse, 1932) and Das Charakterstück (Berlin: Vieweg, 1933), promised a future wealth of scholarship. However, in 1933, Bodky was expelled from his teaching position because he was a Jew. After officials came to seize part of his instrument collection, he and his family fled to Amsterdam, where they were in exile for five years. In 1938, he managed to escape Europe by securing a position at the Longy School of Music in Cambridge, Massachusetts. Bodky would become one of the most important proponents of early music in the United States. Over the last twenty years of his life, he promoted historical instruments, performance practice, and forgotten early music repertoire. His many accomplishments in the United States include the founding of the Music Department at Brandeis University and the Cambridge Society for Early Music, the introduction and promotion of the collegium musicum, and the publication of the treatise The Interpretation of Bach’s Keyboard Works (Cambridge, MA: Harvard University Press, 1960). This thesis provides the first detailed examination of his influence in the United States. His roles as performer, scholar, and teacher are each explored. Relying upon unpublished archival material, academic records, memoirs, and numerous concert programs in addition to Bodky’s published writings, this study reveals that Bodky had a significant impact on how early music was performed, taught, and received in the United States.

The gatekeepers of Moroccan cultural heritage : Amazigh women and the status quo

This study aims to determine the role of Amazigh/Berber women in maintaining and preserving the cultural heritage of their people through carpet weaving, as well as how this tradition contributes to gender norms and tourism in rural Morocco. The research method consisted of conducting semi-structured interviews and a literature review of intangible cultural heritage preservation, traditional gender norms in rural areas, and nomadic tourism theories. Analysis of the interviews concluded that Amazigh women keep their traditional culture alive through carpet weaving, and they are simultaneously valued and marginalized within their society. The necessity and pressure placed upon them leads to issues related to gender equality and girls’ rights/accessibility to a quality education in the modern context. Though in a constant state of change and shifting with time, the nomadic lifestyle and traditional Berber carpet weaving are still participated in today and nevertheless hold deep significance for those who partake in these activities. Traditional mentalities regarding gender norms remain intact, although they are challenged more frequently. Women’s weavings are critical in preserving Amazigh culture and symbolize a heritage identity in an overt manner, making women central to the Berber story and Berber heritage.

The diploma thesis is devoted to the analysis of the connection between mindfulness, work engegament and authentic behavior. The theoretical part of the diploma thesis is divided into three chapters. The first chapter describes the sources of work motivation. The second chapter deals with the relationships between employees and personal resources that lead to engaged behavior. In the third chapter, I describe the appropriate interventions and impacts that lead to increased engagement and work related well-being. The work thus evaluates a specific theory of personal, work and motivational resources that affect further work engagement and satisfaction. The research part of the work consists of quantitative research, in which we have verified the mediation analysis of the relationships between mindfulness and engagement, where authenticity is considered a mediator of these relationships. Based on the results of the mediation analysis, it was proved that the relationship between mindfulness and engagement is fully mediated by authenticity. While the direct relationship between mindfulness and engagement ceased to be significant (β = -.0632, p = .543), the indirect relationship was significant (β = -.2196, p < .001). Moderní postupy hodnocení kvalitativních parametrů potravin / Modern Approaches in Qualitative Aspects of Foods Assessment

Antioxidant properties and some other characteristics of selected organic and conventional products (wine, spelt flours) were evaluated in view of their potential utilization as markers suitable for food unambiguous differentiation according to the affiliation to production system. Beside these, study of fruit juices stability during storage in dependence on different technological procedures of juice processing was performed. In both cases, combination of dominantly EPR and UV/VIS spectroscopy but also other methods (e.g., HPLC, AAS, electrophoresis) was applied to evaluate the stability and antioxidant properties of selected samples. Statistical methods, especially methods of multivariate statistics were used to assess the influence of production and technological-processing operations on antioxidant status of foods and their selected characteristics. Qualitative analysis of Slovak and foreign organic and conventional wines proved that their antioxidant properties are influenced by production system, vintage and variety. On the basis of statistical analysis, descriptors of AAE, kTEMPOL and %RS were selected as the most powerful markers for Slovak organic and conventional wines discrimination. The discrimination of white and red organic and conventional wines possessed 100% and 95.2% correctness, respectively. High correctness of differentiation of Slovak and foreign wines was also achieved. Monitored descriptors of antioxidant activity of spelt flours varied in dependence on season, origin, variety, production system and depend also on way of spelt treatment, although this effect is not so significant. It was also confirmed that the type of extraction agent substantially affects the composition of extracts including polyphenols and flavonoids concentration, affecting thus also their antioxidant activity. Mixture of 50% ethanol/water was selected as the most promising extraction system. Although the results of individual spectroscopic assays revealed high variability and often ambiguous trends, the differences found were sufficient for successful differentiation of both wines and spelt flour samples according to the selected criteria by multivariate statistical processing of the data. Influence of changes in technological procedures of fruit juice processing (nitrogen atmosphere application, utilization of caps with oxygen scavengers) on radical-scavenging properties of 100% pineapples juices with pieces of pineapple and 100% orange juices with pulp was assessed as well. Results obtained proved that antioxidant activity of juices varied in dependence on storage conditions (temperature, storage time, light exposure) and depends also on origin, variety and quality of fruit used. However, the application of nitrogen atmosphere and active packaging materials is not sufficient enough to eliminate the oxidation reactions in juices, but changes in antioxidant activity are better predictable. It can be concluded that EPR spectroscopy could be effectively applied for wines and spelt flours differentiation according to the way of production and to assess the stability of food products, either alone or in combination of UV/VIS spectroscopy and the other methods, utilizing multivariate statistics for processing of experimental data.

Ověření pravosti razítek v dokumentu / Verification of Authenticity of Stamps in Documents

Klasická inkoustová razítka, která se používají k autorizaci dokumentů, se dnes díky rozšíření moderních technologií dají relativně snadno padělat metodou oskenování a vytištění. V rámci diplomové práce je vyvíjen automatický nástroj pro ověření pravosti razítek, který najde využití zejména v prostředích, kde je nutné zpracovávat velké množství dokumentů. Procesu ověření pravosti razítka musí přirozeně předcházet jeho detekce v dokumentu - úloha zpracování obrazu, která zatím nemá přesvědčivé řešení. V této diplomové práci je navržena zcela nová metoda detekce a ověření pravosti razítka v barevných obrazech dokumentů. Tato metoda zahrnuje plnou segmentaci stránky za účelem určení kandidátních řešení, dále extrakci příznaků a následnou klasifikaci kandidátů za pomoci algoritmu podpůrných vektorů (SVM). Evaluace ukázala, že algoritmus umožňuje rozlišovat razítka od jiných barevných objektů v dokumentu jako jsou například loga a barevné nápisy. Kromě toho algoritmus dokáže rozlišit pravá razítka od kopií.

DNA-BASED METHODS FOR AUTHENTICITY AND TRACEABILITY OF PLANTAND MICROBIAL SPECIES AND DURUM WHEAT VARIETIES

Qualità e sicurezza degli alimenti, inclusa la loro tracciabilità ed autenticità, è diventato ngli ultimi anni obiettivo primario per la salute e il benessere dei consumatori. Il progetto è diretto allo sviluppo e applicazione di metodiche DNA-based per la tracciabilità di specie vegetali, varietà di frumento duro e microorganismi a difesa della qualita’, salubrita’ ed autenticità della filiera grano e prodotti processati. Le attività progettuali dello studio sono finanziate da industria (Barilla S.P.A.) con il coinvolgimento di enti pubblici di ricerca (Università Cattolica Del Sacro Cuore Di Piacenza, CREA-GB di Fiorenzuola D’arda). Il progetto si articola in tre argomenti principali: WP1.Tracciabilità di specie vegetali nella filiera pasta. Tracciabilità di varietà di frumento duro nella filiera pasta A questo scopo sono state intraprese nel secondo anno di attività due azioni dirette allo sviluppo e validazione di due diverse metodiche di fingerprinting varietale. Partendo dalle direttive UPOV in materia di impiego di marcatori molecolari per la caratterizzazione varietale è stata applicata e validata su di un pool di 26 varietà di interesse per l’industria un’ analisi basata su di una combinazione di marcatori SSR (Simple Sequence Repeats) che ha consentito di identificare in maniera univoca ciascuna delle varietà in esame. Il saggio è stato trasferito ai laboratori dell’industria che lo applica attualmente nella routine per il controllo di partite di granella. A fronte della robustezza dell’analisi SSR si pongono però i lunghi tempi analitici. Per ottimizzare questo aspetto si è completata un’attività di sequenziamento parziale del genoma di 28 varietà presenti in due repliche biologiche (52 campioni) e 12 mix di DNA di due varietà (4 differenti percentuali per ogni coppia di varietà miscelate). I dati ottenuti hanno fornito circa 15.000 marcatori molecolari DArT-seq (Diversity Array Technology ) e SNP (Simple Nucleotide Polimorphisms). Dall’intero set di marcatori è stato quindi individuato, attraverso una procedura bioinformatica, un set ridotto di marcatori ad alta informatività in grado di identificare univocamente le singole varietà e di predire la presenza di altre varietà in miscela e la percentuale di contaminazione. WP2.Tracciabilità Di Microrganismi Fungini Nella Filiera Pasta Questo studio è volto al controllo e identificazione di specie microbiche patogene che possono svilupparsi lungo la filiera grano con conseguente impatto negativo sulla salubrità di granella, di semole e dei prodotti finiti. A questo scopo sono stati prodotti campioni di granella a contaminazione controllata. Si è costituita attraverso l’analisi delle sequenze Barcode una ceppoteca che comprende i maggiori patogeni fungini che possono contaminare la granella durante la crescita della pianta in campo o durante lo stoccaggio della granella. Dopo l’inoculo artificiale dei singoli ceppi in due varietà di frumento duro, sono stati raccolti, a tempi crescenti, campioni di spighe e granella. La metodica è risultata rapida e sensibile nell’identificazione di DNA fungino fin dalle prime fasi dell’infezione, quando i sintomi della malattia risultavano ancora non ancora visibili. Le informazioni di sequenza Barcoding, in prospettiva, potranno essere utilizzate per sviluppare nuovi metodi di identificazione fungina più sensibili e rapidi. WP3. Identificazione molecolare di microrganismi vivi o morti in pesto L’obiettivo di questo lavoro è stato quello di sviluppare metodiche di V-qPCR (Viability-PCR), per permettere l’identificazione, quantificazione e discriminazione cellule microbiche vive o morte in alimentiprocessati, come il pesto. Per lo studio è stato scelto il batterio patogeno B.cereus, microrganismo ubiquitario, patogeno e sporigeno di difficile identificazione soprattutto in matrici complesse. Durante questo lavoro è stato sviluppato un protocollo analitico che prevede l’estrazione del DNA batterico da pesto, matrice interferente e complessa. Parte del lavoro è stata svolta presso l’Istituto di Microbiologia dell’Università Cattolica e l’Istituto IATA CSIC Institut d’Agroquímica i Tecnologia dels Aliments in Valencia. / Food quality and safety, including food traceability and authenticity, have become crucial in the last decades. Today, molecular and genetic progress can support the agri-food industry, due to the improvement of new analytical tools. Among the available applications, DNA-based methods can detect the presence of a particular species or variety along the food supply chain, verify the genetic identity of food and feed ingredients and detecting the presence of contaminating organisms, thus becoming an essential tool to study patterns, causes, and risk factors of diseases and outbreaks. As a consequence, genetic analysis has become increasingly popular even among non-specialists and highly beneficial for consumers, agricultural farmers, governments, and the private sector (Reid, O’Donnell, and Downey 2006). In this framework, the research developed in this thesis arises by active collaboration between the private company Barilla G. & R. Fratelli S.p.A., the public research institute CREA-GB (Consiglio per la Ricerca in agricoltura e l'analisi dell Economia Agraria) and Università Cattolica del Sacro Cuore, to develop a set of DNA-based methods to improve the traceability and authenticity of plant and microbial species and durum wheat varieties applicable from farm to fork. Following these aims, the research developed in this thesis includes: 1. The optimization and validation of qPCR assay for the discrimination of plant species along the pasta production chain through the organization of a ring test involving nine Italian public and private laboratories. The results obtained in this study were published in the Journal of Cereal Science (Chapter 2); 2. The discrimination of durum wheat varieties by selecting SSRs and DarT molecular markers as reliable methods for variety fingerprinting (Chapter 3). The results confirm the sensitivity of the method and the feasibility to 7 protect the food industry from fraud and ensure the consumer a certified pasta quality; 3. The application of the Barcoding technique and the development of qPCR assay for the identification and quantification of field fungi (Fusarium, Alternaria, Michrodochium, Cochliobolus spp.) and saprophytic fungi (Aspergillus, Penicillium, Rhizopus spp) along the wheat chain (Chapter 3). The sensitivity of the method was investigated by inoculating potted durum wheat plants at full anthesis and wheat kernels (pre and postharvest trials). The DNA-based methods demonstrate a key role in pathogen detection and the application in several points of the wheat chain (e.g., for control of both locally and imported grains, for storage lots, to evaluate the environmental risk associated with grain powder for farmers and workers); 4. The optimization of Viability q-PCR (V-qPCR) for the discrimination of dead and alive Bacillus cereus, a spore-forming bacteria (Chapter 4). The results of PMAxx, combined with qPCR, have demonstrated the selective discrimination of B.cereus viable cells, with no false-positive signals determined by dead cells, a peculiar aspect of thermally treated food; 5. The comparison of two DNA extraction kits (FastDNA® SPIN Kit for Soil – MB and NucleoSpin Tissue - Macherey Nagel) by detecting B.cereus spores in basil pesto sauce, selected as a model food matrix. Despite the limit of detection (LOD) achieved (respectively 1.8x102 spores/gr by using Fast DNA TM SPIN and 2.7 x 105 spores/gr by using NucleoSpin®), the principal challenge remains the spores' DNA extraction from the complex matrix. Lastly, the results obtained during the doctoral research project were globally discussed (Chapter 5).
