Perfil protéico e uso de marcadores moleculares relacionados à qualidade de panificação em trigo / Protein profile and use of molecular markers related to baking quality of wheat

In recent decades researchers have shown concern over acquisition of cultivars with good baking quality, a characteristic which initially refers to the protein composition of the endosperm, composed principally of the proteins making up gluten. Various methodologies have been employed in the evaluation of this characteristic, including SDS-PAGE (polyacrylamide gel electrophoresis), SE-HPLC (size exclusion high performance liquid chromatography), baking tests and employment of genetic markers. In an attempt to compare the different existing methodologies and propose a strategy for wheat genetic improvement programs based on industrial quality, four different techniques were tests with 45 national cultivars. Six allele-specific markers were used for verification of the presence of the HMW subunits: Glu-D1-1d (5+10) and Glu-B1-2a (7+8), one LMW subunit: Glu-A3d, the presence or absence of 1B/1R translocation: by the presence of Glu-B1 (long arm of the wheat chromosome) or ω- secalin (gene of the short arm of rye) and puroindoline (Pinb-D1b). Also employed where SDS-PAGE, SE-HPLC and baking test methods. As a result of applying the markers, the presence in the genotypes observed was: 71% for subunit Glu-D1-1d (5+10), 16% for Glu-B1-2a (7+8), 60% for Glu-B1 (absence of translocation) and 8% for Pinb-D1b and Glu-A3d. The results of SDS-PAGE suggested a prevalence of the subunits 2* and 1 in chromosome 1A; 7+8, 7+9 and 17+18 in 1B; and 5+10 in 1D. The average values of SE-HPLC were 35.99% and 44.99% for the polymeric protein in protein (PPP) and unextractable polymeric protein (UPP), respectively, and 1.29 for the ratio between gliadins and glutenins (GLI/GLU), with significant variation among the genotypes (p≤0.05). The baking test also showed a significant difference (p≤0.05) between the cultivars under the same conditions. The UPP data were independent of the PPP percentage data, signifying that a portion of the unextractable polymeric protein (UPP) does not depend on the greater percentage of total polymeric proteins. However there was a positive correlation between the score of HMW subunits, evaluated by SDSPAGE, and the UPP values; this indicates that scoring has apparently been efficient in classifying the genotypes of greatest quality. The cultivars without translocation with rye also showed better results for UPP, PPP and GLI/GLU in relation to those possessing translocation. The GLI/LGU ratio is correlated with the specific volume of the bread, but apparently does not guarantee quality of the qualitative characteristics, which may indicate the action of gliadins in extensibility, but with a lack of structure in the gluten network. These results corroborate for the selection of HMW subunits 5+10, cultivars without translocation with rye, with high values of UPP and an appropriate GLI/GLU ratio with the objective of obtaining greater wheat baking quality. / O trigo é um dos principais cereais em todo o mundo, sendo processado para uma gama de produtos. Surgiu nas últimas décadas a preocupação dos pesquisadores com a obtenção de cultivares com boa qualidade de panificação, característica que refere-se primordialmente a constituição protéica do endosperma, composta principalmente pelas proteínas formadoras de glúten. Várias metodologias vêm sendo empregadas na avaliação dessa característica, dentre as quais, estão o SDS-PAGE (eletroforese em gel de poliacrilamida), SE-HPLC (cromatografia líquida de alta eficiência por exclusão molecular), teste de panificação e o emprego de marcadores genéticos. Buscando comparar as diferentes metodologias existentes e propor uma estratégia a programas de melhoramento de trigo voltados à qualidade industrial, testaram-se quatro diferentes técnicas em 45 cultivares nacionais. Usaram-se seis marcadores alelo-específicos direcionados à verificação da presença das subunidades de HMW: Glu-D1-1d (5+10) e Glu-B1-2a (7+8), uma subunidade de LMW: Glu-A3d, a presença ou ausência de translocação 1B/1R: pela presença de Glu-B1 (braço longo do cromossomo de trigo) ou de ω-secalin (gene do braço curto do centeio) e puroindolina (Pinb-D1b). Também foram empregadas as metodologias de SDS-PAGE, SE-HPLC e teste de panificação. Como resultado da aplicação de marcadores obteve-se presença nos genótipos de: 71% para subunidade Glu-D1-1d (5+10), 16% para Glu-B1-2a (7+8), 60% para Glu-B1 (ausência de translocação) e 8% para Pinb-D1b e Glu-A3d. Os resultados de SDS-PAGE apontam uma prevalência das subunidades 2* e 1 no cromossomo 1A; 7+8, 7+9 e 17+18 no 1B; e 5+10 no 1D. Enquanto, os valores médios de SE-HPLC foram de 35,99% e 44,99% para as frações de proteínas poliméricas totais (PPP) e não-extraíveis (UPP), respectivamente, e 1,29 para a relação entre gliadinas e gluteninas (GLI/GLU), com variação significativa entre os genótipos (p≤0,05). O teste de panificação também apontou diferença significativa (p≤0,05) entre as cultivares nas mesmas condições. Os dados de UPP foram independentes dos dados da proporção de PPP, significando que a porção de proteínas poliméricas não-extraíveis (UPP) não depende de maior proporção de proteínas poliméricas totais. Enquanto houve correlação positiva entre o escore dado às subunidades de HMW, avaliadas por SDS-PAGE, e os valores de UPP; isto indica que a pontuação aparentemente vem sendo eficaz em classificar os genótipos de melhor qualidade. As cultivares sem translocação com centeio também obtiveram melhores resultados para UPP, PPP e GLI/GLU em relação as que a possuem. A relação GLI/GLU está correlacionada ao volume específico dos pães, mas aparentemente não garantiu boa qualidade nas características qualitativas, o que pode indicar a ação de gliadinas na extensibilidade, mas com falha na estruturação da rede de glúten. Esses resultados corroboram para uma seleção a favor das subunidades 5+10 de HMW, de cultivares sem translocação com centeio, com alto valor de UPP e razão apropriada de GLI/GLU, quando se objetiva melhorar a qualidade de panificação do trigo.

Vid köp av konfigurerbara produkter som till exempel hus eller sängar på internet är det viktigt att kunden kan erbjudas en realistisk visualisering för att kunna få en känsla för hur den färdiga produkten kommer att se ut. En metod för att skapa visualiseringen är att från en miljö uppbyggd av 3d-modeller som anpassas efter kundens val skapa en rendering i realtid. För att den renderade bilden ska se realistisk ut och få hög kvalitet är det nödvändigt att i förväg utföra tunga beräkningar för hur ljuset sprider sig i scenen, så kallad bakning. Hur hanteringen av innehållet och bakningen effektivt ska göras är ett område där det saknas studier men där det finns stor potential för effektivisering. Därför har denna designstudie utförts där syftet är att öka kunskapen om hur hanteringen av innehållet och ljusberäkningar kan effektiviseras. Utvecklingen har utförts med Action Design Research där tre iterationer har fullföljts. Principer från Continuous Delivery har använts för att utveckla en IT-artefakt som automatiserar processen som krävs från det att en ny ändring har lagts in i versionshanteringssystemet till det att det finns ett färdigt paket med innehållet som behövs för att göra en visualisering. Resultatet består av nya komponenter skrivna i C# för Unity och skript skrivet med Python. Resultatet från studien visar att det är möjligt att med hjälp av principer från Continuous Delivery effektivisera processen inom det här området och att antalet manuella steg och den krävda arbetstiden kraftigt kan reduceras. Utifrån designstudien har fem nya designprinciper kunnat formuleras som är lämpliga att använda där program används som inte är har ett bra stöd för att köras automatiskt och att steg i processen kan vara beräkningsmässigt tunga. De går att applicera på fallet med hantering av innehållet i Unity men bör också vara relevanta för att hantera ett mer generellt fall där det finns en process med många manuella steg och beräkningsmässigt tunga steg. / When purchasing configurable products (for example, houses or beds) on the Internet, it is important that the customer can be offered a realistic visualization in order to get a feel for how the finished product will look. One method for creating the visualization is to create a real-time rendering from an environment built of 3D models that is adapted to the customer's choice. In order for the rendered image to look realistic and get high quality, it is necessary to perform heavy calculations in advance for how the light spreads in the scene (baking). How to effectively manage the content and baking is an area where there is little research but where there is great potential for improving the work flow. Therefore, this design study has been carried out where the purpose is to increase the knowledge of how the handling of the content and light calculations can be improved. The development has been carried out with Action Design Research, where three iterations have been completed. Continuous Delivery principles have been used to develop an IT artifact that automates the process required from the time a new change is added to the version management system until there is a complete package of content needed for visualization. The result consists of new components written in C# for Unity and scripts written with Python. The results of the study show that it is possible to streamline the process in this area using principles from Continuous Delivery and that the number of manual steps and the required working time can be greatly reduced. Based on the design study, five new design principles have been formulated that are suitable for use where programs are used that do not have good support for automating this and that contains steps in the process can be computationally heavy. They can be applied to the case of handling the contents of Unity but should also be relevant to dealing with a more general case where there is a process with many manual steps and computationally heavy steps.

An Online Input Estimation Algorithm For A Coupled Inverse Heat Conduction-Microstructure Problem

<p>This study focuses on developing a new online recursive numerical algorithm for a coupled nonlinear inverse heat conduction-microstructure problem. This algorithm is essential in identifying, designing and controlling many industrial applications such as the quenching process for heat treating of materials, chemical vapor deposition and industrial baking. In order to develop the above algorithm, a systematic four stage research plan has been conducted. </P> <p> The first and second stages were devoted to thoroughly reviewing the existing inverse heat conduction techniques. Unlike most inverse heat conduction solution methods that are batch form techniques, the online input estimation algorithm can be used for controlling the process in real time. Therefore, in the first stage, the effect of different parameters of the online input estimation algorithm on the estimate bias has been investigated. These parameters are the stabilizing parameter, the measurement errors standard deviation, the temporal step size, the spatial step size, the location of the thermocouple as well as the initial assumption of the state error covariance and error covariance of the input estimate. Furthermore, three different discretization schemes; namely: explicit, implicit and Crank-Nicholson have been employed in the input estimation algorithm to evaluate their effect on the algorithm performance. </p> <p> The effect of changing the stabilizing parameter has been investigated using three different forms of boundary conditions covering most practical boundary heat flux conditions. These cases are: square, triangular and mixed function heat fluxes. The most important finding of this investigation is that a robust range of the stabilizing parameter has been found which achieves the desired trade-off between the filter tracking ability and its sensitivity to measurement errors. For the three considered cases, it has been found that there is a common optimal value of the stabilizing parameter at which the estimate bias is minimal. This finding is important for practical applications since this parameter is usually unknown. Therefore, this study provides a needed guidance for assuming this parameter. </p> <p> In stage three of this study, a new, more efficient direct numerical algorithm has been developed to predict the thermal and microstructure fields during quenching of steel rods. The present algorithm solves the full nonlinear heat conduction equation using a central finite-difference scheme coupled with a fourth-order Runge-Kutta nonlinear solver. Numerical results obtained using the present algorithm have been validated using experimental data and numerical results available in the literature. In addition to its accurate predictions, the present algorithm does not require iterations; hence, it is computationally more efficient than previous numerical algorithms. </p> <p> The work performed in stage four of this research focused on developing and applying an inverse algorithm to estimate the surface temperatures and surface heat flux of a steel cylinder during the quenching process. The conventional online input estimation algorithm has been modified and used for the first time to handle this coupled nonlinear problem. The nonlinearity of the problem has been treated explicitly which resulted in a non-iterative algorithm suitable for real-time control of the quenching process. The obtained results have been validated using experimental data and numerical results obtained by solving the direct problem using the direct solver developed in stage three of this work. These results showed that the algorithm is efficiently reconstructing the shape of the convective surface heat flux. </P> / Thesis / Doctor of Philosophy (PhD)

Microalgae as novel ingredients for the formulation of food products

Tesis por compendio / [ES] Las microalgas son organismos unicelulares fotosintéticos microscópicos que se encuentran en gran variedad de ambientes y son muy eficientes en la transformación de energía solar en biomasa. Los estudios realizados hasta el momento hacen referencia a posibles beneficios de la incorporación de microalgas en la dieta, por la mejora del sistema cardiovascular, las propiedades adelgazantes y energizantes, capacidad antioxidante, o la reducción del colesterol y los triglicéridos. La forma más habitual de consumir las microalgas es como suplemento dietético en forma de tabletas, cápsulas o polvo. La incorporación de biomasa de microalgas en productos tradicionales se ha enfrentado al reto de la aparición de colores verdes fuertes, así como su consistencia pulverulenta que puede afectar la textura y percepción del producto. Todos estos aspectos constituyen las principales áreas de mejora para conseguir un mayor grado de aceptación de productos con microalgas, y son la base del reto de este proyecto. El objetivo de la presente tesis doctoral fue el desarrollo de nuevos productos alimentarios incorporando las propiedades nutricionales de la biomasa de microalgas, incrementando o mejorando con ello, las propiedades nutricionales del alimento original. Para conseguir este objetivo se evaluaron a nivel fisicoquímico, reológico y textural, la incorporación de diferentes especies de microalgas (Arthrospira platensis (Spirulina), Chlorella vulgaris, Dunaliella salina y Nannochloropsis gaditana) en distintas matrices alimentarias (productos horneados, emulsiones y extrusionados). Por otra parte, se evaluó y caracterizó la incorporación de las microalgas utilizando diferentes tecnologías como la impresión 3D o la extrusión. Además de cómo afecta la incorporación de las microalgas a los productos obtenidos, se evaluaron los aspectos nutricionales de su incorporación, en cuanto al aporte de minerales y su biodisponibilidad. Las propiedades reológicas de las masas y emulsiones enriquecidas con microalgas (Spirulina, Chlorella y Dunaliella) indicaron que su comportamiento viscoelástico fue modificado y mejorado, mostrando características aptas para este tipo de productos. La adición de microalgas (Spirulina y Chlorella) a las masas utilizadas para la impresión 3D de galletas y snacks, permitió una mejor extrusión o impresión de éstas, obteniendo muestras impresas en 3D de forma cilíndrica, más precisas en cuanto a sus dimensiones con respecto a la estructura cilíndrica diseñada. Además, las muestras impresas presentaron mayor estabilidad y resistencia, antes y después del proceso de horneado comparadas con la muestra control. Para los productos horneados, tanto para las rosquilletas como los snacks impresos en 3D, la adición de microalgas (Spirulina y Chlorella) permitió mayor estabilidad en términos de textura. Ligeros cambios en los parámetros fisicoquímicos y de expansión se produjeron por la adición de Spirulina y Chlorella en los productos extrusionados. Además, los extrusionados enriquecidos con Nannochloropsis, mostraron parámetros similares a los de la muestra de control. Todos los productos presentaron colores luminosos y apariencias innovadoras y atractivas. En cuanto a los minerales, se observó un aumento de P, K, Ca, Na, Mg, Fe y Se con la adición de Spirulina y Chlorella, junto con el aumento de la concentración adicionada de microalgas. Siguiendo la normativa sobre etiquetado nutricional de los alimentos, el enriquecimiento con microalgas en rosquilletas se puede clasificar como un alimento "rico en hierro (Fe)". De igual forma, las rosquilletas y galletas enriquecidas con microalgas pueden considerarse un alimento "alto en selenio (Se)". Además, la incorporación de Spirulina y Chlorella en las formulaciones de galletas, permitió una mayor bioaccesibilidad del contenido de P, K, Ca, Mg, Fe, Zn y Se para la absorción en el cuerpo comparado con las muestras control. / [CAT] Les microalgues són organismes unicellulars fotosintètics microscòpics. Són molt eficients a l'hora de transformar l'energia solar en biomassa. Els estudis realitzats fins ara fan referència a possibles beneficis de la incorporació de microalgues en la dieta per produir una millora del sistema cardiovascular, per presentar propietats per aprimar i donar energia, per mostrar capacitat antioxidant o per afavorir una reducció del colesterol i els triglicèrids. La forma més habitual de consumir microalgues és com a suplement dietètic en forma de tauleta, càpsula o en pols. La incorporació de biomassa de microalgues en productes tradicionals s'ha afrontat al repte de l'aparició d'un color verd fosc i d'una consistència polsosa que pot afectar a la textura i, per tant, a la percepció del producte. Aquests aspectes constituïxen les principals àrees de millora per aconseguir un major grau d'acceptació de productes amb microalgues i són la base del repte d'aquest projecte. L'objectiu d'aquesta tesi doctoral és el desenvolupament de nous productes alimentaris que incorporen les propietats nutricionals de la biomassa de microalgues, de manera que s'incrementen o es milloren les propietats nutricionals de l'aliment original. Per aconseguir aquest objectiu s'avaluaren a escala fisicoquímica, reològica i de textura la incorporació de diferents espècies de microalgues (Arthrospira platensis (Spirulina), Chlorella vulgaris, Dunaliella salina i Nannochloropsis gaditana) en diferents matrius alimentàries (productes fornejats, emulsions i extrudits). D'altra banda, s'avaluà i caracteritzà la incorporació de les microalgues utilitzant diferents tecnologies com la impressió en 3D o l'extrusió. A banda de valorar com afecta la incorporació de microalgues als productes elaborats, s'avaluaren els aspectes nutricionals, pel que fa a l'aportació i biodisponibilitat de minerals. Les propietats reològiques de les masses i emulsions enriquides amb microalgues (Spirulina, Chlorella i Dunaliella) indicaren que el seu comportament viscoelàstic fou modificat i millorat, de tal manera que mostrà característiques aptes per aquest tipus de productes. L'addició de microalgues (Spirulina i Chlorella) en les masses utilitzades per a la impressió 3D de galetes i snacks permeté una millor impressió, ja que s'obtingueren mostres impreses de forma cilíndrica amb unes dimensions més precises respecte a l'estructura cilíndrica dissenyada. A més, les mostres impreses presentaren una major estabilitat i resistència abans i després del procés de fornejat en comparació amb la mostra control. Respecte als productes fornejats, l'addició de microalgues (Spirulina i Chlorella) a les rosquilletes i els snacks impresos en 3D permeté una major estabilitat en termes de textura. Lleugers canvis als paràmetres fisicoquímics i d'expansió es produïren per l'addició d'Spirulina i Chlorella en els productes extrudits. A més, els extrudits que foren enriquits amb Nannochloropsis mostraren paràmetres similars als de la mostra control. Tots els productes presentaren colors lluminosos i aparences innovadores i atractives. Pel que fa als minerals, s'observà un augment de P, K, Ca, Na, Mg, Fe i Se quan s'afegí Spirulina i Chlorella, directament relacionat amb l'augment de la concentració de microalgues. Seguint la normativa sobre etiquetatge nutricional dels aliments, l'enriquiment amb microalgues en rosquilletes ens permet classificar-les com a aliment "ric en ferro (Fe)". These microorganisms are very efficient when transforming solar energy into biomass, due to their cellular structure, which is completely submerged in an aqueous medium, forming an adequate surface for the exchange of nutrients and gases. Microalgae compounds are now known to exhibit cardioprotective, immunomodulatory, anti-proliferative, anti-inflammatory, cognitive, neurobehavioral and antimicrobial properties, amongst others. Researchers have shown possible benefits of the incorporation of microalgae in the diet so far. The most common way to consume microalgae is as a dietary supplement in the form of tablets, capsules or powder. The incorporation of microalgae biomass in traditional products has faced the challenge of the appearance of strong green colours, as well as its powdery consistency that can affect the texture and perception of the product. All these aspects constitute the main areas for improvement the development of microalgae-based products, and they are the challenges faced of this project. The main objective of this PhD Thesis was the development of novel food products incorporating the nutritional properties of microalgae biomass, thereby increasing or improving the nutritional properties of the original food matrix. To achieve this goal, the effect of the addition of different species of microalgae (Arthrospira platensis (Spirulina), Chlorella vulgaris, Dunaliella salina and Nannochloropsis gaditana) on the physicochemical, rheological and textural properties of different food matrices (baked products, emulsions and extrudates) was evaluated. Furthermore, the effect of the incorporation of microalgae using different technologies such as 3D printing or extrusion to obtain food products was studied. In addition, how the incorporation of microalgae affects the nutritional aspects of the food products in terms of the contribution of minerals and their bioavailability was also evaluated. The rheological properties of doughs, batters and emulsions enriched with microalgae (Spirulina, Chlorella and Dunaliella) indicated that their viscoelastic behaviour was modified and improved, showing characteristics suitable for this type of products. The addition of microalgae (Spirulina and Chlorella) to the doughs and batters used for the 3D printing of cookies and snacks, allowed a better extrusion or printing behaviour. This allowed obtaining cylindrical 3D printed samples, more precise in terms of their dimensions with respect to the designed cylindrical structure. In addition, the 3D microalgae-printed sample structures presented greater stability and resistance, before and after the baking process compared to the control sample. For baked products, both for breadsticks and 3D printed snacks, the addition of microalgae (Spirulina and Chlorella) allowed greater stability in terms of texture. Slight changes in the physicochemical and expansion parameters were produced by the addition of Spirulina and Chlorella in the extruded products. In addition, the extrudates enriched with Nannochloropsis, showed similar parameters to those of the control sample. Microalgae-enriched obtained products showed bright colours with appealing appearances. Regarding minerals, an increase in P, K, Ca, Na, Mg, Fe and Se was observed with the addition of Spirulina and Chlorella, along the increase of concentration of microalgae addition. Following the regulations on nutrition labelling for food stuffs, breadstick enrichment with microalgae are a food "high in iron (Fe)" In the same way, breadsticks and cookies enriched with microalgae can be considered a "high in selenium (Se)" food. Going a step further, Spirulina and Chlorella vulgaris incorporation in cookie formulations allowed for greater bioaccessibility of P, K, Ca, Mg, Fe, Zn, and Se content for absorption in the body than control cookies. / Uribe Wandurraga, ZN.
