Previous issue date: 2014-07-16 / O polietileno de ultra alto peso molecular (PEUAPM) é um polímero com
excelentes propriedades físicas e mecânicas tendo como mais notáveis a sua
inércia química, a lubricidade, resistência impacto e resistência a abrasão.
Exibe um papel relevante no sucesso na Artroplastia Total de Joelho (ATJ) e
Artroplastia Total de Quadril (ATQ). A Artroplastia se caracteriza pela
substituição da articulação natural por uma articulação artificial. As articulações
artificiais são formadas por um conjunto de componentes, sendo fabricados de
matéria prima metálica ou cerâmica e um componente, normalmente feito em
PEUAPM, que é interposto entre componentes metálicos. Alguns fatores
contribuem para um maior desgaste do componente de polietileno. Os
componentes fabricados em polietileno PEUAPM não possuem
fotoestabilizadores. E em inspeções realizadas em fabricantes e distribuidores
destes produtos foi observado que durante seu processamento fabril e
comercialização eles não são adequadamente protegidos da iluminação direta.
As características consideradas importantes para o desempenho in vitro destes
dispositivos estão definidas em normas técnicas, mas muitas destas normas
somente recomendam determinados requisitos considerados importantes do
ponto de vista técnico. Atualmente a área de materiais de uso em saúde
trabalha em regulamentos que visam instituir os requisitos mínimos para
registro de implantes ortopédicos. Desta forma, este trabalho propôs avaliar as
possíveis alterações de componentes fabricados em polietileno de ultra alto
peso molecular utilizados em procedimentos cirúrgicos de artroplastia e joelho
e quadril, exposto a radiação ultravioleta. O trabalho foi dividido em 4 grupos:
grupo 1 (material não exposto a UV); grupo 2 (material exposto a UV por 1h);
grupo 3 (material exposto a UV por 24h) e grupo 4 (material exposto a UV por
48h). Todas as amostras foram caracterizadas por Difração de Raio X (DRX),
Espectroscopia de Absorção de Infravermelho por Transformada de Fourier
(FTIR), Microscopia Ótica (MO), Microscopia Eletrônica de Varredura (MEV)
com mapeamento por EDS e Microscopia de Força Atômica (AFM) com o
intuito de avaliar as possíveis degradações do PEUAPM submetidos a radiação
UV. Os resultados demonstraram que a exposição do polímero a radiação UV
em todos os tempos testados neste trabalho promoveu alterações nas
amostras, pois foi observado, principalmente, nos ensaios de DRX e EDS que
ocorreu respectivamente, um discreto aumento da cristalinidade do material e
diminuição no percentual de oxigênio. Desta forma, pode-se concluir que o
aumento da cristalinidade pode, pela ação da radiação UV, afetar nas
propriedades do material contribuindo para o aumento do desgaste e
diminuição do tempo de vida útil do implante. / The polyethylene, ultra high molecular weight (UHMWPE) is a polymer with
excellent physical and mechanical properties with the most notable its chemical
inertness, lubricity, impact resistance and abrasion resistance. Displays a
relevant role in the successful Total Knee Arthroplasty (TKA) and Total Hip
Arthroplasty (THA). The arthroplasty is characterized by replacement of the
natural joint with an artificial joint. Artificial joints are formed by a set of
components, being manufactured from metal or ceramic raw material and a
component normally done in UHMWPE, which is interposed between the metal
components. Some factors contribute to increased wear of the polyethylene
component. Components manufactured in polyethylene UHMWPE not have
photostabilizers. And inspections on manufacturers and distributors of these
products was observed that during its industrial processing and marketing they
are not adequately protected from direct light. Characteristics considered
important for the in vitro performance of these devices are set in technical
standards, but many of these standards only recommend certain requirements
considered important from a technical standpoint. Currently the area of
materials for use in healthcare working on regulations that seek to establish the
minimum requirements for registration of orthopedic implants. Thus, this study
aimed to evaluate the possible changes of components manufactured in
polyethylene, ultra high molecular weight used in surgical procedures and knee
arthroplasty and hip, exposed to ultraviolet radiation. The work was divided into
4 groups: group 1 (material not exposed to UV); group 2 (material exposed to
UV for 1 h); group 3 (material exposed to UV for 24 h) and group 4 (material
exposed to UV for 48 h). All samples were characterized by X-Ray Diffraction
(XRD), Absorption Spectroscopy Fourier Transform Infrared (FTIR), optical
microscopy (OM), scanning electron microscopy (SEM) with EDS mapping and
Atomic Force Microscopy (AFM) in order to evaluate the possible degradation of
UHMWPE subjected to UV radiation. The results showed that exposure of the
polymer to UV radiation at all times tested in this study modified the samples, it
was observed mainly in the experimental XRD and EDS which occurred
respectively a slight increase in crystallinity and decrease in the percentage
oxygen. Thus, it can be concluded that the increase in crystallinity can by the
action of UV radiation, affect the material properties contributing to increased
wear and reduced useful life of the implant.

Incontinência Urinária de Esforço e apresenta elevada taxa de cura e melhora da
qualidade de vida com baixos índices de complicações. Este trabalho teve como
objetivo desenvolver e confeccionar, uma tela de policaprolactona, para uso na
técnica de Sling e realizar análises comparativas deste material com a tela de
polipropileno, utilizada comercialmente. A pesquisa foi desenvolvida no Laboratório
de Avaliação e Desenvolvimento de Biomateriais do Nordeste – CERTBIO. Utilizouse duas amostras de tela, uma confeccionada no CERTBIO e outra de uso
comercial. Foram avaliadas as composições químicas e físicas das telas de
Policaprolactona (PCL) e Polipropile no (PP), através das técnicas de:
Espectroscopia na Região de Infravermelho com Transformada de Fourier (FTIR),
Difração de Raios X (DRX) e Microscopia Eletrônica de Varredura (MEV), Tensão
superficial, Ensaios Mecânicos de Tração e Teste de Citotoxicidade. Pode-se
comprovar pela análise de FTIR, que o material formador das telas era apenas de
polipropileno e policaprolactona. A análise de DRX corroborou com os resultados
obtidos na análise de FTIR, que indicavam que o material das telas tratava de PP e
PCL, apresentando um espectograma característico d estes polímeros. Também,
através da análise de DRX, foi possível observar o grau de cristalinidade indicando o
PP que possui maior grau de cristalinidade que o PCL. Pode-se comprovar também,
que o PP possui maior resistência mecânica que o PCL, verificando assim que o PP
é mais frágil e quebradiço do que o PCL. A análise de tensão superficial mostrou
que as duas telas possuem molhabilidade parcial devido as cavidade presentes na
superfície das telas. A biocompatibilidade dos compostos foi confirmada com o teste
de Citotoxicidade. / The sling technique by transobturator approach is effective for the treatment of
women with Stress Urinary Incontinence and has a high cure rate and improved
quality of life with low complication rates. This work aimed to develop and produce,
polycaprolactone, screen for use in Sling surgery and to perform comparative
analyses of this material with a polypropylene bladder, used commercially. The
survey was developed in the laboratory of evaluation and development of
Biomaterials in the Northeast-CERTBIO. Two samples were used to screen; a made
in the laboratory and another used commercially. Been evaluated physical and
chemical compositions of the Polycaprolactone (PCL) and Polypropylene (PP)
screens, through the following techniques: infrared Spectroscopy in the region with a
Fourier transform (FTIR), x-ray Diffraction (XRD) and scanning electron microscopy
(SEM), surface tension, traction mechanical essays and Cytotoxicity test. You can
check by the FTIR analysis, that the trainer material of fabrics was just poliproplieno
and polycaprolactone, for examining the computer spectrogram. The XRD analysis
corroborated with the results obtained in the FTIR analysis, which indicated that the
material was PP screens and PCL, showing a characteristic you might like to look of
these polymers. Also, through the analysis of DRX, it was possible to o bserve the
degree of crystallinity indicating the PP has higher degree of crystallinity the PCL.
One can prove that the PP has higher mechanical strength than the PCL, checking
so that the PP is more brittle than the PCL. The analysis of surface tension showed
that both screens have partial wettability due to cavity present on the surface of the
canvas. The biocompatibility of the compounds has been confirmed with the
Cytotoxicity test and it showed low cytotoxicity in two screens.

Nanoporous calcium carbonate-based substrates for the controlled delivery of functional materialsLevy, Charlotte Luanne Victoria January 2017 (has links)
The overall aim of this project was to study 'functionalised' calcium carbonates (FCCs) for use as a carrier for the controlled release of 'actives,' by permeation and diffusion, and is being proposed as an environmentally friendly and non-toxic pharmaceutical excipient, nutraceutical, and flavour carrier. The delivery of a drug to its target site in the appropriate amount and time-frame in order for it to have a controlled release effect whilst achieving the maximum therapeutic effect remains a topic of design and development for novel drug delivery systems. FCCs encompass a family of new pharmaceutical excipients in which the conditions of manufacture follow strict process regulations with respect to the grade of reagents that are employed and the microbiological environment under which they are produced, and include freedom from organic polymers. Adjustments to the FCC production process can be used to produce a wide range of different morphologies, and raise the possibility of tailoring the void structures of the particles to provide controlled release delivery vehicles for actives across many fields, including drugs and flavours. However, such tailoring can only be fully optimised by a fundamental characterisation of the way in which a drug, loaded into an FCC, then flows and diffuses out over a period of time to provide the delayed release. It was found that adsorption on the FCC surface is selective, for example, saccharin does not become adsorbed from 4-(2-hydroxyethyl)piperazine-1-ethanesulfonic acid (HEPES) buffer solution, and neither does anethole from ethanol. FCC also does not adsorb the cationic probe benzyltrimethylammonium bromide (BTMAB) or the anionic probe sodium 2-naphthalenesulphonate (Na2NS). However, it was found that vanillin adsorbs onto the FCC in an amount of 2.00 ± 0.59 mg g^-1. Aspirin and vanillin adsorption from ethanolic solutions with various additions of water onto FCC TP was investigated and fitted with the Tóth isotherm. It was estimated that vanillin adsorbed onto around 17 %, and aspirin onto around 39 %, of the overall FCC TP surface area without the addition of any water. An equation was formulated in order to approximate the adsorption as a function of the FCC's surface coverage by the water. This is discussed in Chapter 4 and has also been published in a peer-reviewed academic journal (Levy et al., 2017). Chapter 5 discusses the preliminary steps of the loading of vanillin and saccharin into FCC, and the results were inconclusive for a majority of samples, concluding that the loading and analysis methods need refining. The modelling of the diffusion profiles of vanillin loaded FCC S07 and S10 was successful, and resulted in diffusion coefficients of 231.9 x 10^-16 m^2 s^-1 and 248.44 x 10^-16 m^ s^-1, respectively. This is outlined in Chapter 6. Chapter 7 describes the 'zero length column' (ZLC) technique, which was used as a way to characterise the diffusivity of the intraparticle pores of each FCC grade. However, it was established that there are many experimental artefacts present with such a method. This work outlines the development of the novel 'finite length column' (FLC), which was developed as a means to overcome the limitations of the ZLC (Levy et al., 2015). Effective diffusivity coefficients in the long-term region of the diffusion curves of the FCC samples range from 1.06-106 x 10 ^-16 m ^2 s^-1. The FLC was then used in preliminary trials to dilute FCC with an inert solid in order to further refine the ZLC technique, and is discussed in Chapter 8. Two mathematical methods were also developed to aid in the refinement. The reported effective diffusivity coefficient for FCC 03 in the long-term region of the diffusion curve is 49.5 x 10^-16 m^2 s^-1. In conclusion, this work confirms that FCC has potential for use as a carrier for the controlled release of 'actives' by diffusion. The utilisation of mathematical modelling in conjunction with experimental methods in the study of drug release and delivery is steadily increasing due to its enormous future potential; it will enable the optimisation of novel dosage forms and the elucidation of release mechanisms at a major reduction in cost and time compared with the number of experimental studies required to do so.

Influência de tratamentos térmicos nas propriedades mecânicas e metalúrgicas em liga odontológica à base de níquel-cromo. / Influence of heat treatment on the mechanical and metallurgical properties in the dental alloy nickel-chromium.André Rocha Pimenta 25 November 2009 (has links)
É crescente a preocupação com o desenvolvimento de materiais adequados a trabalharem interagindo com o corpo humano. Diversas pesquisas têm sido realizadas no desenvolvimento de biomateriais aplicáveis na odontologia, este empenho é justificável pelo grande número de intervenções cirúrgicas para extração de dentes realizadas em todo o mundo. Durante o processo de fabricação de reconstruções dentárias, que utilizam sistemas metalocerâmicos, é utilizado um tratamento térmico que tem a função de promover a adesão da porcelana ao metal. Entretanto, sabe-se que tratamentos térmicos podem alterar a microestrutura do material metálico, modificando suas propriedades. Este trabalho avaliou as modificações causadas em propriedades mecânicas e microestruturais da liga à base de níquel (FIT CAST-SB) utilizada para fins odontológicos, quando a mesma é submetida ao tratamento térmico para adesão da porcelana (denominado de queima). A liga foi inicialmente fundida através da técnica de centrifugação e cera perdida. Posteriormente, um grupo de amostras (grupo TT) foi submetido ao tratamento térmico de queima para adesão da porcelana e o outro grupo (grupo F), permaneceu apenas submetido ao processo de fundição. Os grupos F e TT foram submetidos a ensaio de tração. Nos grupos F e TT, e no material como recebido pelo fabricante (grupo CR), foram realizados ensaios de microdureza e caracterização microestrutural, esta ultima através da técnica de microscopia eletrônica de varredura (MEV). Os grupos F e CR foram submetidos à análise química quantitativa (em um espectrômetro de emissão atômica) e semi-quantitativa por um sistema de Energy Dispersive Spectroscopy (EDS) acoplado ao MEV, sendo que esta ultima técnica também foi aplicada ao grupo TT. A técnica de tratamento digital de imagem foi aplicada às micrografias dos grupos F e TT, para a determinação de possíveis modificações quantitativas nas fases presentes, antes e após o tratamento térmico. Todos os resultados dos ensaios foram submetidos ao teste de hipótese nula (H0), para a distribuição t de Student. Concluiu-se que, para as amostras testadas, o limite de resistência foi superior ao fornecido pelo fabricante, respectivamente 559,39 e 545,55 MPa para os grupos F e TT, contra 306 MPa do fabricante. Enquanto o limite de escoamento foi ligeiramente inferior, 218,71 e 240,58 MPa para os grupos F e TT, respectivamente, contra 258 MPa do fabricante. Os resultados de microdureza ficaram entorno de 70HV, superior aos 21HV fornecido pelo fabricante. Pode-se afirmar, com 95% de confiabilidade, que não houve variação nas propriedades mecânicas e na microestrutura (quantidades de fases presentes e tamanho) antes e após a queima para adesão da porcelana, para os corpos de prova testados. A microestrutura da liga, quando observada em MEV no modo elétrons retroespalhados (modo BSE), é formada por uma matriz de estrutura dendrítica e coloração cinza, uma segunda fase interdendrítica de coloração branca e aspecto rendilhado, e precipitados de coloração preta, apresentando também porosidades. / There is a growing concern with the development of suitable materials for interactive working with the human body. Many searchs have been carried in the development of biomaterials for odontology applications and this dedication is justifiable for the large number of surgical interventions for tooth extractions in the entire world. During the manufacturing process of dental reconstructions, which use metal-ceramic systems, a heat treatment is used to promote adhesion of the porcelain in the metal. However, it is known that heat treatment can change the metallic materials microstructure, changing their properties. This work evaluated the mechanical properties and microstructural changing of the nickel base alloy (FIT CAST-SB) used in odontology, when the alloy is submitted to the heat treatment for the porcelain adhesion (demented of firing). First the alloy was cast using the technical of centrifugation and lost-wax. After that, one samples group (TT group) was submitted for porcelain firing heat treatment and another group (F group), remained only cast. The F and TT groups were submitted to tensile test. The F and TT groups, and the material as received by the manufacturer (CR group), were submitted to microhardness test and microstructural characterization, this latter using scanning electron microscopic. The F and CR groups were submitted to quantitative chemical analysis (atomic emission spectroscopy) and semi-quantitative by Energy Dispersive Spectroscopy (EDS) in the MEV, and this last technique was applied to TT group too. The digital images process technique was applied to micrographics of F and TT groups, to determine the changes in the material phases before and after heat treatment. All results were submitted to the hypothesis test of Students t-distribution. For the samples tested, it was concluded that the tensile strength was higher then the manufacturer information, respectively 559,39 and 545,55 MPa for F and TT groups, against 306 MPa informed by the manufacturer. However, the yield point was slightly lower, 218,81 and 240,58 MPa for F and TT groups, respectively, against 258 informed by the manufacturer. The microhardness results were about 70HV, higher than 21HV informed by the manufacturer. It was concluded, with 95% of confiability, that there was no mechanical properties and microstructure (percent and size of the phases) changes, before and after porcelain firing, in the specimens used. The alloy microstructure, when observed by scanning electron microscopy (SEM) in backscattered electron mode (BSE mode) is formed by a matrix with dendritc structure and gray color, a second phase interdendritc with white color and lacy appearance, and black precipitates, and porosity are also found.

Previous issue date: 2011-08-29 / Universidade Federal de Minas Gerais / Due to limited availability of autogenous bone and of the risks associated with the use of bone allografts, new synthetic materials have been developed in order to replace the bone tissue lost due to trauma or pathological process. The bioactive materials in the form of scaffolds are synthetic materials promising for bone grafting. Several studies suggest that these biomaterials are able to stimulate the proliferation of osteoblasts and osteogenesis at the site of fracture. However, the feasibility of these biomaterials to a clinical application requires the investigation of their biocompatibility. In this context, this study aimed to evaluate the biocompatibility of a scaffold synthesized from a fully crystallized glass-ceramic bioactive quaternary system P2O5-Na2O-CaO-SiO2 (Biosilicate®), through histopathological analysis of the biomaterial implanted in the subcutaneous tissue of rats and the cytotoxicity and genotoxicity analysis of the biomaterial in cell cultures (OSTEO-1 and L929 cells). Histopathologic analysis of the biomaterial was performed using 65 Wistar rats male (210- 260 g), randomly divided into two groups, Control group (n = 3 animals per period) and Biosilicate group (n = 10 animals per period), evaluated at 7, 15, 30, 45 and 60 days after surgery. The animals of Biosilicate group underwent surgery and received a subcutaneous implant of Biosilicate® scaffolds. The animals of Control group underwent surgery but did not receive any biomaterial implant. The cytotoxicity analysis was performed to assess the effect of products leaching from Biosilicate® scaffolds (extracts) on cellular proliferation (MTT). The extracts were evaluated in various concentrations (100, 50, 25 and 12.5%) in experimental periods of 24, 72 and 120 hours in two cell lines (OSTEO-1 and L929). The genotoxicity analysis (comet assay) was performed to assess DNA damage in cells OSTEO-1 and L929 grown in contact with the Biosilicate® scaffolds in different periods of 24, 72 and 96 horas. The statistical analysis of parametrics data was performed by analysis of variance (ANOVA) followed by Tukey post-hoc and the analysis of nonparametrics data was performed by Mann-Whitney test. Both statistical tests were performed with a significance level of 5%. The results of histopathological analysis showed that the animals of the Control group did not present inflammation process, necrotic tissue and fibrous tissue. The animals of Biosilicato group showed a granulation tissue after 7 days of implantation. In the other periods (15, 30, 45 and 60 days) a chronic inflammation process of foreign body, marked by the presence of fibrous tissue and giant cells was observed. No infection or necrotic tissue was observed in any animal. In the analysis of cytotoxicity, it was observed that extracts of Biosilicato® scaffolds did not have any significant effect in reducing cell proliferation OSTEO-1 and L929, and that lower concentrations of the extracts (12.5 and 25%) stimulated the proliferation of both cells in periods of 72 and 120 hours. The analysis of genotoxicity showed that the Biosilicate® scaffolds did not induce DNA damage in the cell lines tested in all experimental periods. The results of this study showed that the Biosilicate® scaffolds presented biocompatibility in vivo and in vitro. / Devido a limitada disponibilidade de osso autógeno e dos riscos associados ao uso de osso alógeno, novos materiais sintéticos vêm sendo desenvolvidos com o objetivo de substituir o tecido ósseo perdido em decorrência de traumatismos ou processos patológicos. Os materiais bioativos na forma de scaffolds são materiais sintéticos promissores para enxertia óssea. Vários estudos sugerem que estes biomateriais são capazes de estimular a proliferação de osteoblastos e a osteogênese no local da fratura. No entanto, a viabilização destes biomateriais a uma aplicação clínica requer o emprego de testes que avaliem a sua biocompatibilidade. Dentro deste contexto, o presente estudo teve como objetivo avaliar a biocompatibilidade do scaffold sintetizado a partir de uma vitrocerâmica bioativa totalmente cristalizada do sistema quaternário P2O5-Na2O-CaO-SiO2 (Biosilicato®), por meio da análise histopatológica do biomaterial implantado no tecido subcutâneo de ratos, e pelas análises de citotoxicidade e genotoxicidade do biomaterial em cultura de células da linhagem OSTEO-1 e L929. A análise histopatológica do biomaterial foi realizada utilizando 65 ratos machos da linhagem Wistar (210-260 g), distribuídos aleatoriamente em dois grupos, Controle (n = 3 animais por período) e Biosilicato (n = 10 animais por período), avaliados em períodos distintos de 7, 15, 30, 45 e 60 dias. Os animais do grupo Biosilicato foram submetidos a uma cirurgia no tecido subcutâneo e receberam um implante de scaffold de Biosilicato®. Os animais do grupo Controle foram submetidos à mesma cirurgia, mas não receberam o implante do biomaterial. A análise de citotoxicidade foi realizada para avaliar os efeitos dos produtos da lixiviação dos scaffolds de Biosilicato® (extratos) na proliferação celular pelo ensaio MTT. Os extratos foram avaliados em várias concentrações (100, 50, 25 e 12,5%) em períodos experimentais de 24, 72 e 120 horas, utilizando duas linhagens celulares (OSTEO-1 e L929). A análise de genotoxicidade (ensaio cometa) foi realizada para avaliar os danos no DNA de células OSTEO-1 e L929 cultivadas em contato com scaffolds de Biosilicato® em períodos distintos de 24, 72 e 96 horas. A análise estatística dos dados paramétricos foi realizada pelo teste de variância (ANOVA), seguido do post-hoc de Tukey, e a análise dos dados não paramétricos foi realizada pelo teste de Mann-Whitney. Ambos os testes estatísticos foram realizados com nível de significância de 5%. Os resultados da análise histopatológica demonstraram que os animais do grupo Controle não apresentaram processo inflamatório, tecido necrótico ou tecido fibroso. Já os animais do grupo Biosilicato apresentaram um tecido de granulação após 7 dias de implantação e nos demais períodos (15, 30, 45 e 60 dias) apresentaram um processo inflamatório crônico de corpo estranho, marcado pela presença de tecido fibroso e células gigantes multinucleadas. Em todos os animais avaliados não foi evidenciado foco de infecção ou tecido necrótico. Na análise de citotoxicidade foi observado que os extratos dos scaffolds de Biosilicato® não possuem efeito significativo na redução da proliferação de células OSTEO-1 e L929, e que as menores concentrações dos extratos (12,5 e 25%) estimularam a proliferação de ambas às células nos períodos de 72 e 120 horas. Na análise de genotoxicidade foi evidenciado que os scaffolds de Biosilicato® não induzem danos do DNA de células de ambas às linhagens testadas em todos os períodos experimentais. Os resultados obtidos neste estudo demonstraram que os scaffolds de Biosilicato® apresentaram biocompatibilidade em experimentos in vivo e in vitro.

This study is a preliminary analysis of composites' made of hydroxyapatite (HAP-91®) and polyhydroxybutyrate (PHB) essentials characteristics to be used in orthopedics. The composites were studied in osseous defects concerning their biocompatibility, osseointegration, osseoconduction, and biodegradation. Bone defects were made bilaterally in the olecranon of 30 white rabbits and each filled with a composite, with a total of 12 members per group. Limbs in the 1 group received composite # 1: 10% of HAP-91® and 90% of PHB; limbs in 2 group received composite # 2: 25% HAP- 91® and 75% PHB; and in 3 group the composite # 3: 50% of each biomaterial. Defects in the control group were not filled. Limbs were evaluated in the first eight days after surgery and again at 45 and 90 days afterwards for lameness, circumference, pain, dehiscence, and infection. Radiographs were taken 8, 45 and 90 days after surgeries when four animals in each group were euthanized and samples with defects taken for histology. Significant differences in circumference data were found for all groups before and one day after surgery, what did not happen with four and eight days. No significant differences were found between groups for circumference as well as for the other clinical parameters. Radiographs taken 8 days after surgery showed no bone- composite apposition, but those for the 45th and 90th days after showed that composites were in direct contact with bone. Histological observations indicated a normal repair process in the 8th day, i.e., a vascularized, dense connective tissue and new, early trabecular bone; this evolved to fully organized trabecular bone and the lateral cortical completely formed by the 90th-day. At the three observation dates, tissues in 1, 2, and 3 group samples were found similar to those in control, where more bone than soft tissues were found in the defect. Projections of connective and bone tissues were seen inside composites #2 and 3. No inflammatory cells in any place whatsoever were found at any observation date. Clearly more bone built up in 3 samples, and at its interface, than in the 1 and 2 groups samples. At the 45 and 90th days, in all three groups, osteoclasts were seen in the bone-composite interface, and also composite pieces, detached from the original block, were found within the bone structure and surrounding soft tissues. Conclusion is that the composites are biocompatible, osseoconductive, integrate to bone and can undergo in vivo degradation. / Os biomateriais hoje utilizados no tratamento das fraturas ósseas e da perda do tecido ósseo ainda apresentam deficiências a serem corrigidas. O presente trabalho consta da análise preliminar de características essenciais para utilização de compósitos de hidroxiapatita (HAP-91®) e polihidroxibutirato (PHB) em ortopedia. Estudou-se a biocompatibilidade, osseointegração, osseocondução e biodegradação dos compósitos em defeitos ósseos. Foram implantados três compósitos fabricados com diferentes proporções desses biomateriais em defeitos circulares bilaterais no olécrano de 30 coelhos, onde cada membro recebeu um compósito, com um total de 12 membros por grupo. Os membros do grupo 1 receberam compósitos contendo 10% de HAP-91® e 90% de PHB, do grupo 2 contendo 25% de HAP-91® e 75% de PHB, do grupo 3, 50% de cada biomaterial e os animais do grupo controle não receberam compósito. Os membros foram avaliados quanto à claudicação, circunferência do membro, sensibilidade dolorosa, deiscência e infecção nos oito primeiros dias após a cirurgia e aos 45 e 90 dias. Foram radiografados imediatamente após a cirurgia e aos oito, 45 e 90 dias. Nessas mesmas datas, 4 animais de cada grupo foram eutanasiados, coletando-se material para processamento histológico. Existiu diferença significante entre os valores de circunferência do membro no pré-operatório e do primeiro dia após a cirurgia dentro de todos os grupos, o que não ocorreu em relação aos dias quatro e oito. Não houve diferença entre os grupos para esse parâmetro assim como para os demais. Não foi observado contato direto entre osso e compósito nas radiografias obtidas aos oito dias após as cirurgias, mas aos 45 e 90 dias nos três grupos tratados observou-se este contato. À análise histológica do grupo controle observou-se processo típico de reparação, ou seja, tecido conjuntivo denso vascularizado e trabéculas ósseas novas aos oito dias com evolução para osso trabecular mais organizado aos 90 dias e cortical lateral restabelecida. Os tecidos observados nos grupos 1, 2 e 3 foram semelhantes ao controle, sendo a proporção de tecido ósseo formada no defeito, maior do que a de tecidos moles em todas as datas. Nos grupos dois e três foram observadas projeções de tecido ósseo e conjuntivo no interior dos poros do compósito. Infiltrados inflamatórios não foram observados em nenhum momento. O grupo 3 apresentou interface com maior proporção de tecido ósseo formado na região do defeito e no interior do compósito do que os grupos 1 e 2. Observou- se osteoclastos nas bordas dos compósitos e fragmentos dos mesmos separados do bloco principal aos 45 e 90 dias nos três grupos. Concluiu-se que os compósitos são biocompatíveis, osteocondutores, se integram ao tecido ósseo e são degradados in vivo.

Influência de tratamentos térmicos nas propriedades mecânicas e metalúrgicas em liga odontológica à base de níquel-cromo. / Influence of heat treatment on the mechanical and metallurgical properties in the dental alloy nickel-chromium.André Rocha Pimenta 25 November 2009 (has links)
É crescente a preocupação com o desenvolvimento de materiais adequados a trabalharem interagindo com o corpo humano. Diversas pesquisas têm sido realizadas no desenvolvimento de biomateriais aplicáveis na odontologia, este empenho é justificável pelo grande número de intervenções cirúrgicas para extração de dentes realizadas em todo o mundo. Durante o processo de fabricação de reconstruções dentárias, que utilizam sistemas metalocerâmicos, é utilizado um tratamento térmico que tem a função de promover a adesão da porcelana ao metal. Entretanto, sabe-se que tratamentos térmicos podem alterar a microestrutura do material metálico, modificando suas propriedades. Este trabalho avaliou as modificações causadas em propriedades mecânicas e microestruturais da liga à base de níquel (FIT CAST-SB) utilizada para fins odontológicos, quando a mesma é submetida ao tratamento térmico para adesão da porcelana (denominado de queima). A liga foi inicialmente fundida através da técnica de centrifugação e cera perdida. Posteriormente, um grupo de amostras (grupo TT) foi submetido ao tratamento térmico de queima para adesão da porcelana e o outro grupo (grupo F), permaneceu apenas submetido ao processo de fundição. Os grupos F e TT foram submetidos a ensaio de tração. Nos grupos F e TT, e no material como recebido pelo fabricante (grupo CR), foram realizados ensaios de microdureza e caracterização microestrutural, esta ultima através da técnica de microscopia eletrônica de varredura (MEV). Os grupos F e CR foram submetidos à análise química quantitativa (em um espectrômetro de emissão atômica) e semi-quantitativa por um sistema de Energy Dispersive Spectroscopy (EDS) acoplado ao MEV, sendo que esta ultima técnica também foi aplicada ao grupo TT. A técnica de tratamento digital de imagem foi aplicada às micrografias dos grupos F e TT, para a determinação de possíveis modificações quantitativas nas fases presentes, antes e após o tratamento térmico. Todos os resultados dos ensaios foram submetidos ao teste de hipótese nula (H0), para a distribuição t de Student. Concluiu-se que, para as amostras testadas, o limite de resistência foi superior ao fornecido pelo fabricante, respectivamente 559,39 e 545,55 MPa para os grupos F e TT, contra 306 MPa do fabricante. Enquanto o limite de escoamento foi ligeiramente inferior, 218,71 e 240,58 MPa para os grupos F e TT, respectivamente, contra 258 MPa do fabricante. Os resultados de microdureza ficaram entorno de 70HV, superior aos 21HV fornecido pelo fabricante. Pode-se afirmar, com 95% de confiabilidade, que não houve variação nas propriedades mecânicas e na microestrutura (quantidades de fases presentes e tamanho) antes e após a queima para adesão da porcelana, para os corpos de prova testados. A microestrutura da liga, quando observada em MEV no modo elétrons retroespalhados (modo BSE), é formada por uma matriz de estrutura dendrítica e coloração cinza, uma segunda fase interdendrítica de coloração branca e aspecto rendilhado, e precipitados de coloração preta, apresentando também porosidades. / There is a growing concern with the development of suitable materials for interactive working with the human body. Many searchs have been carried in the development of biomaterials for odontology applications and this dedication is justifiable for the large number of surgical interventions for tooth extractions in the entire world. During the manufacturing process of dental reconstructions, which use metal-ceramic systems, a heat treatment is used to promote adhesion of the porcelain in the metal. However, it is known that heat treatment can change the metallic materials microstructure, changing their properties. This work evaluated the mechanical properties and microstructural changing of the nickel base alloy (FIT CAST-SB) used in odontology, when the alloy is submitted to the heat treatment for the porcelain adhesion (demented of firing). First the alloy was cast using the technical of centrifugation and lost-wax. After that, one samples group (TT group) was submitted for porcelain firing heat treatment and another group (F group), remained only cast. The F and TT groups were submitted to tensile test. The F and TT groups, and the material as received by the manufacturer (CR group), were submitted to microhardness test and microstructural characterization, this latter using scanning electron microscopic. The F and CR groups were submitted to quantitative chemical analysis (atomic emission spectroscopy) and semi-quantitative by Energy Dispersive Spectroscopy (EDS) in the MEV, and this last technique was applied to TT group too. The digital images process technique was applied to micrographics of F and TT groups, to determine the changes in the material phases before and after heat treatment. All results were submitted to the hypothesis test of Students t-distribution. For the samples tested, it was concluded that the tensile strength was higher then the manufacturer information, respectively 559,39 and 545,55 MPa for F and TT groups, against 306 MPa informed by the manufacturer. However, the yield point was slightly lower, 218,81 and 240,58 MPa for F and TT groups, respectively, against 258 informed by the manufacturer. The microhardness results were about 70HV, higher than 21HV informed by the manufacturer. It was concluded, with 95% of confiability, that there was no mechanical properties and microstructure (percent and size of the phases) changes, before and after porcelain firing, in the specimens used. The alloy microstructure, when observed by scanning electron microscopy (SEM) in backscattered electron mode (BSE mode) is formed by a matrix with dendritc structure and gray color, a second phase interdendritc with white color and lacy appearance, and black precipitates, and porosity are also found.

Previous issue date: 2006-03-07 / Coordenação de Aperfeiçoamento de Pessoal de Nível Superior - CAPES / The effect of the treatment of the polimeric film preparation with the extracted polyssacharíd of the gum of the Anacardium occidentale L. was evaluated in excisionals cutaneous injuries in mice albinic Swiss, through the accompaniment of the evolution of the healing cutaneous process, under the clinical and histopathological aspects. After the accomplishment of the surgical procedure, 5 groups (n=15/grupo) had been formed in accordance with the employed treatment: 1. NaCl (NaCl 0,15M); 2. AA (acetic acid 4% v/v); 3. POLICAJU/AA (polimeric film I containig acetic POLICAJU 10% m/v in acid 4% v/v) and 4, POLICAJU/NaCl (polimeric film I containing POLICAJU 10% m/v in NaCl 0,15M). Tissue samples had been fixed and staining (Tricrômicro de Masson) and analyzed in 2nd, 7th and 12th pos-surgical (P.S.) days. In group POLICAJU/AA it was observed reduction of the flogistics signals represented by presence of edema, crust and exudate. Histopathological point of view, the groups treatment with POLICAJU had presented partial reepitelization and soft granulation tissue, specially in the POLICAJU/NaCl group. Thus, it is concluded that the polímeric films I containing POLICAJU, applied as oclusive dressings, had been efficient in the process of cutaneous tecidual repair. The use of films obtained of the POLICAJU for tripsina immobilization (DEI), by entrepment, and its application as dressing in cutaneous injuries was evaluated. DEI it demonstrated it to a constant release of the trypsin optimal pH and temperature of 8,6 and 55°C, in the analyzed conditions and revealed stability stored in the conditions retained about 80% (28 days 25°C) of its initial activity. The wound treatment withDEI it had more presented organized morphologic standard when compared with the controls (NaCl 0,15M and acetic acid 4% v/v). These results demonstrate that the POLICAJU can be used as matrix for trypsin immobilization and a biomaterial potential. An experimental design (2) was applied for determination of the best conditions of entrapment of the trypsin in POLICAJU films. For this study the effect of the concentration of trypsin and the temperature on its activity had been analyzed. It was observed that of the analyzed parameters the lesser concentration of trypsin (100ug/mL) was the best condition of immobilization. This system was evaluated how much it influences enviromental (pH and temperature) and topic application. The best values of pH and the temperature had been 9,0 and 55C, demonstrating to stability the range of temperature (25-65C), how much to the storage DEI it retained 30% of its activity (28 days). The results of the hystopathological evaluations had demonstrated an acceleration of the healing process. From the results of the immobilization of trypsin entrappment in POLICAJU/NaCl and POLICAJU/AA, DEI it was applied as oclusive dressing in the treatment of cutaneous wounds being evaluated its effect under the clinical aspects and histopathology. After the surgical procedure 5 groups had been formed in accordance with the type of treatment (n=15/grupo): 1. NaCl; 2. AA; 3. AAT; 4. POLICAJUT/AA and 5, POLICAJUT/NaCl. Tissue samples had been fixed and staining(Tricrômicro of Masson) in the 2nd, 7th and 12th days after the surgery. The wound treatment with POLICAJUT/NaCl how much to the clinical and histopathology parameters they had presented one better when comparative evolution to the too much groups. The results gotten from all the studies of immobilization and biological application demonstrate the effectiveness of the POLICAJUT/NaCl film with oclusive dressing. / Os efeitos do tratamento do filme polimérico preparado com o polissacarídeo extraído da goma do Anacardium occidentale L. foram avaliados em lesões cutâneas excisionais em camundongos albino Swiss, através do acompanhamento da evolução do processo cicatricial cutâneo, sob os aspectos clínicos e histopatológicos. Após a realização do procedimento cirúrgico, 4 grupos (n=15/grupo) foram formados de acordo com o tratamento empregado: 1, NaCl (NaCl 0,15M); 2, AA (ácido acético 4% v/v); 3, POLICAJU/AA (filme polimérico contendo POLICAJU 10% m/v em ácido acético 4% v/v) e 4, POLICAJU/NaCl (filme polimérico contendo POLICAJU 10% m/v em NaCl 0,15M). Amostras de tecido foram fixadas e coradas (Tricrômicro de Masson) e analisada no 2o, 7o e 12o dias P.O.. No grupo POLICAJU/AA foi observado diminuição dos sinais flogísticos representados por presença de edema, crosta e exsudato. Do ponto de vista histopatológico, os grupos tratados com POLICAJU apresentaram reepitelização parcial e tecido de granulação fibroso, especialmente no grupo POLICAJU/NaCl. Assim, conclui-se que os filmes polímericos contendo POLICAJU, aplicados como curativos oclusivos, foram eficazes no processo de reparo tecidual cutâneo. A utilização de filmes obtidos do POLICAJU como matriz para imobilização de tripsina (DEI), por enclausuramento, e sua aplicação como curativo em lesões cutâneas foi avaliada. O DEI demonstrou uma liberação constante da tripsina, pH e temperatura ótimos de 8,6 e 55°C, nas condições analisadas e mostrou-se estável nas condições do meio, retendo cerca de 80% (28 dias a 25°C) da sua atividade inicial. As lesões tratadas com o DEI apresentaram padrão morfológico mais organizado quando comparado aos controles (NaCl 0,15M e ácido acético 4% v/v). Estes resultados demonstram que o POLICAJU pode ser utilizado como matriz para imobilização de tripsina e um potencial biomaterial. Um planejamento experimental (22) foi aplicado para determinação das melhores condições de enclausuramento da tripsina em filmes de POLICAJU. Para esse estudo foram analisados os efeitos da concentração de tripsina e da temperatura sobre sua atividade. Observou-se que dos parâmetros analisados a menor concentração de tripsina (100μg/mL) foi a melhor condição de imobilização. Este sistema foi avaliado quanto a influência do meio (pH e temperatura) e aplicação tópica. Os valores ótimos de pH e a temperatura foram 9,0 e 55oC, demonstrando estabilidade a ampla faixa de temperatura (25-65oC), quanto ao armazenamento o DEI reteve cerca de 30% de sua atividade (28 dias). Os resultados das avaliações histopatológicas demonstraram uma aceleração do processo cicatricial. A partir dos resultados da imobilização de tripsina enclausurada em POLICAJU/NaCl e POLICAJU/AA, o DEI foi aplicado como curativo oclusivo no tratamento de lesões cutâneas sendo avaliado seus efeitos sob os aspectos clínicos e histopatológico. Após o procedimento cirúrgico 5 grupos foram formados de acordo com o tipo de tratamento (n=15/grupo): 1, NaCl; 2, AA; 3, AAT; 4, POLICAJUT/AA e 5, POLICAJUT/NaCl. Amostras de tecido foram fixadas e coradas (Tricrômicro de Masson) nos 2o, 7o e 12o dias após a cirurgia. As lesões tratadas com POLICAJUT/NaCl quanto aos parâmetros clínicos e histopatológicos apresentaram uma melhor evolução quando comparado aos demais grupos. Os resultados obtidos a partir de todos os estudos de imobilização e aplicação biologica demonstram a eficácia do filme POLICAJUT/NaCl com curativo oclusivo.

Previous issue date: 2016-09-30 / Investigar características do processo de integração e biocompatibilidade de
um implante de gel celulósico nas cavidades anoftálmicas de coelhos. Analisar a
proliferação celular em geral, a angiogênese, a proliferação de células
multinucleadas gigantes e a fibrogênese, por técnicas de histomorfométria e
imunohistoquímica. Estudo experimental de intervenção empregando 18 coelhos
divididos em 6 grupos. O olho direito de todos os animais foi eviscerado e os
esquedo não sofreu nenhum tipo de intervenção. Após a evisceração a bolsa
escleral foi suturada e preenchida com gel celulósico gel. Diariamente, todos os
animais foram examinados clinicamente, sob biomicroscopia, até o 7º dia do
implante e uma vez por semana até o dia da eutanásia. Os animais foram
submetidos a eutanásia e enucleações dos olhos no 7º, 30º, 60º, 90º, 120º e 240º
dia após o implante. Os olhos, inclusive os esquerdos, foram avaliados
macroscopicamente e processados para análises histopatológica,
histomorfométricas, imunohistoquímicas e imunofluorescência. Clinicamente, todos
os animais, não exibiram sinais de alergias, intoxicação, extrusão e infecção. Em
todos os grupos ao exame macroscópico, histopatológico e histomordo olho
esquerdo, não revelou alterações anatômicas. No entanto, nesta análise, olho direito
exibiu redução de 8% no volume do bulbo. O corte do saco escleral mostrou um
conteúdo sólido, compacto, elástico, resistente à tração, com superfície lisa e
brancacenta.Não foram observados sinais de necrose, ou liquefação. O tecido
epiescleral estava algo hipertrofiado. As preparações histológicas estudadas, nas
diversas colorações, revelaram uma infiltração linfomonomorfonuclear inicial,
substituída posteriormente por uma proliferação fibrocitária e histiocitária com
formação de células multinucleadas gigantes. Foram também encontrados poucos
polimorfonucleares neutrófilos e eosinófilos. A partir do 30o dia houve proliferação
vascular e deposição de colágeno em todos os espécimes estudados, embora, no
240º dia do experimento, a resposta inflamatória crônica, a neovascularização e a
deposição do colágeno não tinham ainda atingido o centro do implante. Neste
modelo, o gel da celulose produzido pela Zoogleia sp, mostrou-se biocompatível e
integrado às órbitas. Fica, portanto, comprovado que o gel celulósico, utilizadoneste
experimento, além de biocompatível, se integrou às órbitas dos coelhos. / To evaluate histologically the integration process of cellulose gel produced by
Zoogloea sp when implanted into rabbits’ eviscerated eyes. This experimental study
employed 36 eyes of 18 rabbits subjected toEvisceration of their right eyes. The
sclerocorneal bag was sutured and filled withbiopolymer from sugar cane in the gel
state. All animals were clinically examinedby biomicroscopy until the day of their
sacrifice which occurred on the 7º, 30º, 60º, 90º, 120º, or 240º day. The eyeballs
obtained, including the left eyes considered controls were sent for histopathological
study by optical macroscopyand microscopy. Tissue staining techniques used
included hematoxylin-eosin, Masson trichrome (with aniline), Gomori trichrome, Van
Gienson, Picrosirius red and periodic acid-Schiff (PAS). No clinical signs of infection,
allergy, toxicity, or extrusion were observed throughout the experiment. The corneas
were relatively preserved. Macroscopic examination revealed a decrease of ~ 8% in
the volume of the bulbs implanted with the biopolymer. After cutting, the
sclerocorneal bag was solid, compact, elastic, and resistant to traction, with a smooth
and whitish surface, and showed no signs of necrosis or liquefaction. The episcleral
tissues were somewhat hypertrophied. The histological preparations studied in
different colors revealed an initial lymphoplasmacytic infiltration, replaced by a
fibroblastic response and proliferation of histiocytes, along with formation of giant
cells. Few polymorphonuclearneutrophils and eosinophils were also found.
Neovascularization and collagen deposition were present in all animals starting from
day 30; although on the 240º day of the experiment the chronic inflammatory
response, neovascularization and collagen deposition had not yet reached the center
of the implant. In this model, the cellulose gel produced by Zoogloea sp proved to be
biocompatible and integrated into the orbits.

Parmi les bio-polymères issus des ressources renouvelables, les polysaccharides fournissent une alternative intéressante aux polymères de synthèse. Dans ce contexte, l’objectif de ce travail de thèse est basé sur la conception des copolymères amphiphiles pour la préparation de nouveaux biomatériaux. Ainsi, l’hydroxypropylméthylcellulose (HPMC) a été étudiée en raison de ses propriétés remarquables, dont la biocompatibilité, la biodégradabilité, la rétention d'eau et la gélification thermoréversible. Ces propriétés sont utiles pour de nombreuses applications telles que le relargage de médicament, la préparation des membranes et la formation de biomatériaux. L'hydrolyse enzymatique avec des endo cellulases issues de Trichoderma reesei a été étudiée pour produire des fragments d'HPMC ayant une masse molaire (Mw) entre 6000 et 30000 g mol-1. Les paramètres de l’activité enzymatique ont été étudiés en fonction de : la nature de substrat, le temps de réaction et la concentration de l'enzyme. Les polymères obtenus ont été comparés à ceux produits par hydrolyse acide. Il a été constaté que la structure des polymères issus d’un procédé d’hydrolyse, varie en termes de degré de substitution pour un même Mw. Cet effet donne lieu à différentes propriétés de gélification thermoréversible. Des copolymères amphiphiles tels que HPMC-b-poly (propylène glycol) et HPMC-b-PLA ont été préparés par amination réductrice et par couplage click thiol-ene, respectivement. Les propriétés d’agrégation ont été caractérisées par la diffusion de la lumière (DLS), le microscope électronique en transmission (TEM) et par la séparation de phase obtenue par la mesure du point de trouble. / Following the concept of bio-refinery, we propose to produce small fragments of biopolymers that can be used further as building blocks to prepare novel polymeric architectures. In the case of polysaccharides, enzymatic hydrolysis enables to form reducing end groups after each cleavage on the polymer chain. Reaction by reductive amination affords the possibility to introduce polysaccharides fragments in a large variety of materials going from amphiphilic copolymers to more sophisticated devices. Hydroxypropyl methylcellulose (HPMC) was used in this work because of its remarkable properties including biocompatibility, biodegradability, water retention and thermoreversible gelation beneficial for many applications such as drug delivery, film and biomaterial formation. Enzymatic hydrolysis using endo cellulases from Trichoderma reesei was investigated to produce a library of HPMC fragments with molecular weight (Mw) from 6000 to 30000 g mol-1. Mw control was carried out by varying the procedure conditions including the nature of starting HPMC, reaction time and enzyme concentration. The obtained polymers were compared to those produced by acidic hydrolysis.According to the preparation conditions, the structure of short chain polymers regarding substitution degrees varied for the same Mw giving rise to different clouding temperature and thermoreversible gelation properties. Amphiphilic block copolymers HPMC-b-poly(propylene glycol) and HPMC-b-PLA were prepared by reductive amination and by the thiol-ene click reaction, respectively. Self-assembly properties of these novel block copolymer were characterized by dynamic light scattering (DLS), transmission electron microscope (TEM), and clouding point temperature.
