Ausência do receptor Toll-Like 2 ocasionou a formação de lesões periapicais mais extensas e com maior número de osteoclastos em camundongos / Silence of toll-like receptor 2 promoted superior size of periapical lesion and number of osteoclasts in mice

O objetivo deste trabalho foi caracterizar a formação e progressão de lesões periapicais induzidas experimentalmente em dentes de camundongos knockout para receptores toll-like 2 (TLR2 KO) comparados a animais wild-type (WT). As lesões periapicais foram induzidas nos primeiros molares inferiores de 28 camundongos WT e de 27 camundongos TLR2 KO. Decorridos 7, 21 e 42 dias da indução da lesão periapical, os animais foram submetidos à eutanásia em câmara de CO2, as mandíbulas foram removidas e submetidas ao processamento histotécnico. A seguir, cortes representativos foram corados com hematoxilina e eosina (HE), para descrição do tecido pulpar e das regiões apical e periapical, em microscopia óptica convencional, e mensuração da área das lesões periapicais, em microscopia de fluorescência. Espécimes sequenciais foram avaliados por meio de: histoenzimologia para a atividade da TRAP, para identificação de osteoclastos; coloração de Brown & Brenn, para localização de bactérias; e imunoistoquímica, para identificação de marcadores da osteoclastogênese (RANK, RANKL, OPG). Os resultados numéricos obtidos da análise morfométrica da extensão da área das lesões periapicais e do número de osteoclastos foram submetidos à análise estatística por meio dos testes não-paramétricos de Mann-Whitney e Kruskal-Wallis, utilizando o software SAS (Statistical Analysis System) for Windows versão 9.1.3. O nível de significância adotado foi de 5%. Os resultados da coloração de Brown & Brenn e Imunoistoquímica foram expressos de maneira qualitativa. O grupo de animais WT apresentou diferença significante na extensão da área das lesões periapicais e no número de osteoclastos entre os períodos experimentais de 7 e 42 dias (p<0,05) e entre 21 e 42 dias (p<0.05). Por outro lado, no grupo de animais TLR2 KO, as diferenças para a extensão da área das lesões periapicais e número de osteoclastos foram encontradas entre os períodos experimentais de 7 e 21 dias (p<0,05) e entre 7 e 42 dias (p<0,05). Quando os períodos dos grupos foram comparados entre si, foram encontradas diferenças estatísticas entre todos os períodos experimentais, tanto para a análise morfométrica da extensão da área das lesões periapicais, quanto para o número de ostoclastos (p<0,05). A análise descritiva do tecido pulpar e das regiões apical e periapical, por meio da coloração de HE, bem como da localização das bactérias, por meio da coloração de Brown & Brenn, não mostrou diferenças entre os dois grupos de animais. Com relação à Imunoistoquímica, as marcações foram semelhantes entre os dois grupos de animais, exceto para as marcações de RANK, as quais não foram encontradas nas lesões periapicais do grupo de animais TLR2 KO. A partir das metodologias empregadas e dos resultados obtidos pode-se concluir que na ausência do TLR2, os animais desenvolveram lesões periapicais significantemente maiores (com maior presença de osteoclastos) quando comparados aos animais WT, sugerindo o importante papel desse receptor na resposta imune e inflamatória do organismo no sentido de combater a infecção do sistema de canais radiculares e dos tecidos perirradiculares. / The aim of the present study was to characterize the formation and progression of periapical lesions experimentally induced in the teeth of toll-like receptors 2 knockout (TLR2 KO) mice compared to wild-type (WT) mice. Periapical lesions were induced in the lower first molars of 28 WT and 27 TLR2 KO mice. After 7, 21 and 42 of periapical lesion induction, the animals were euthanized in a CO2 chamber, and the mandibles were removed and subjected to histotechnical processing. Representative histological sections were stained with hematoxylin and eosin (HE) for description of the features of the pulp tissue and the apical and periapical regions under conventional optical microscopy, and for determination of the size of the periapical lesions under fluorescence microscopy. Sequential specimens were evaluated by: TRAP histo-enzymology for identification de osteoclasts; Brown & Brenn staining for localization of bacteria; and immunohistochemistry for identification of osteoclastogenesis markers (RANK, RANKL, OPG). Data from the morphometric evaluation of the size of periapical lesions and the number of osteoclasts were subjected to statistical analysis by the nonparametric Mann-Whitney and Kruskal-Wallis tests, using the SAS (Statistical Analysis System) software for Windows version 9.1.3. A significance level of 5% was set for all analyses. Data from the Brown & Brenn staining and immunohistochemical analysis were displayed qualitatively. The group of WT mice presented statistically significant difference in the periapical lesion size and number of osteoclasts between the 7- and 42-day experimental periods (p<0.05) as well as between 21 and 42 days (p<0.05). On the other hand, in the group of TLR2 KO mice, significant differences in the periapical lesion size and number of osteoclasts were found between the 7- and 21-day experimental periods (p<0.05) as well as between 7 and 42 days (p<0.05). Comparison of the periods within each group revealed statistically significant differences among all experimental periods for the morphometric evaluation of the size of the periapical lesions and number of osteoclasts (p<0.05). Descriptive analysis of pulp tissue and apical and periapical regions by HE staining and localization of bacteria by Brown & Brenn staining did not show significant differences between the two groups of animals. The immunohistochemical results showed similar immunostaining in both groups of animals, except for RANK expression, which was not observed in the periapical lesions of the TLR2 KO mice. Based on the employed methodology and the obtained results it may be concluded that in the silence of TLR2, the animals developed superior size of periapical lesions (with higher presence of osteoclasts) compared to WT animals, suggesting the important role of this receptor during the immune and inflammatory response against the infection of root canal system and periapical tissues.

Cette thèse est consacrée à l'étude expérimentale du bruit quantique électronique d'un système mésoscopique. Dans une première partie, nous nous sommes intéressés au bruit de partition d'un conducteur unidimensionnel balistique : un contact ponctuel quantique (QPC). Nous avons montré que, lorsque l'un des contacts du QPC est modulé par une onde radio-fréquence, il apparaît un bruit de partition en l'absence de courant moyen à travers le conducteur. Nous avons ainsi validé la théorie de la diffusion appliquée au bruit photo-assisté, d'une part en mesurant le facteur de Fano en l'absence de tension appliquée au QPC, et d'autre part en mesurant le bruit en présence d'une tension continue et d'une irradiation micro-onde. Dans une seconde partie, nous avons testé le système de mesure d'une nouvelle expérience qui, à terme, permettra de mesurer le bruit à haute fréquence d'un conducteur mésoscopique, ainsi que la statistique des photons qu'il émet dans le circuit de mesure. Le test a consisté à réaliser des expériences de type Hanbury-Brown et Twiss (interférométrie d'intensité) avec deux types de sources de photons micro-ondes. D'abord, nous avons utilisé une source thermique incohérente (résistance macroscopique de 50 Ohms) qui présente une statistique super-poissonnienne : les fluctuations de puissance sont proportionnelles au carré de la puissance moyenne émise par la source. Puis nous avons mis en évidence la statistique poissonnienne d'une source classique monochromatique, et nous avons montré que le facteur de Fano géant mesuré est parfaitement expliqué par le bruit des chaînes d'amplification.

From Transcendental Subjective Vision to Political Idealism: Panoramas in Antebellum American Literature

This dissertation explores the importance of the panorama for American Renaissance writers' participation in ideological formations in the antebellum period. I analyze how Ralph Waldo Emerson, Henry David Thoreau, Nathaniel Hawthorne, William Wells Brown, Henry Box Brown, and Harriet Beecher Stowe use the panorama as a metaphorical site to contest their different positions on epistemological and sociopolitical agendas such as transcendentalism, masculinist expansionism, and radical abolitionism. Emerson uses the panorama as a key metaphor to underpin his transcendental idealism and situate it in contemporary debates on vision, gender, and race. Connecting the panorama with optical theories on light and color, Emerson appropriates them to theorize his transcendental optics and makes a hierarchical distinction between light/transparency/panorama as metaphors for spirit, masculinity, and race-neutral man versus color/opacity/myopic vision for body, femininity, and racial-colored skin. In his paean to the moving panorama, Thoreau expresses his desire for Emersonian correspondence between nature and the spirit through transcendental panoramic vision. However, Thoreau's esteem for nature's materiality causes his panoramic vision to be corporeal and empirical in its deviation from the decorporealized vision in Emerson?s notion of transparent eyeball. Hawthorne repudiates the Transcendentalists' and social reformers' totalizing and absolutist idealism through his critique of the panorama and the emphasis on opacity and ambiguity of the human mind and vision. Hawthorne reveals how the panorama satisfies the desire for visual and physical control over the rapidly expanding world and the fantasy of access to truth. Countering the dominant convention of the Mississippi panorama that objectifies slaves as a spectacle for romantic tourism, Box Brown and Wells Brown open up a new American subgenre of the moving panorama, the anti-slavery panorama. They reconstruct black masculinity by verbally and visually representing real-life stories of some male fugitive slaves and idealizing them as masculine heroes of the anti-slavery movement. In Uncle Tom's Cabin, Stowe criticizes how the favorable representation of slavery and the objectification of slaves in the Mississippi panorama and the picturesque help to construct her northern readers' uncompassionate and hard-hearted attitudes toward the cruel realities of slavery and presents Tom's sympathetic and humanized "eyes" as an alternative vision.

Além disso, a análise de corante de cabelo em fios de cabelo tingidos, poderia favorecer informações úteis para análise forense. Em virtude de alto consumo de corantes de cabelo, que podem ser encontrados na forma de tinturas permanente, semipermantes e temporárias, com diferentes estruturas e comportamento físico-químico, a demanda por métodos analíticos sensíveis e robustos para sua quantificação é altamente pertinente. Diante disso, o presente trabalho descreve a construção de sensores eletroquímicos para quantificação de corantes de cabelo do tipo temporário, pois são classe de corantes facilmente removidos do cabelo e, consequentemente, podem ser liberados para o meio ambiente. O sensor eletroquímico consiste no desenvolvimento de um eletrodo compósito com partículas magnéticas modificadas como grupos carboxílicos (CFMP) para determinação dos corantes basic brown 16 (BB16), basic red 51 (BR51) e basic yellow 57 (BY57) em amostras de extratos de fios de cabelos, amostras comercias de tintura temporária, água de lavagem, amostras de água de rio e de torneira. Os três corantes apresentam grupos azos como cromóforos. No presente trabalho, investigou-se a oxidação de aminas e hidroxilas presentes como grupos auxocromos nos corantes. Os resultados mostraram que a oxidação do grupo funcional nos corantes sobre o eletrodo compósito modificados por grupos carboxílicos pode ocorrer em potenciais de 0,44 V, 0,86 V e 0,65, respectivamente para os corantes BB16, BR51 e BY57 e a corrente de pico aumenta significativamente em relação ao eletrodo sem modificação. A interação entre corante + CFMP foi comprovada por espectroscopia de UV/Vis e FTIR. Foram construídas curvas analíticas para os corantes BB16, BR51 e BY57, usando as condições otimizadas na técnica de voltametria de onda quadrada (VOQ): frequência de aplicação dos pulsos (f) = 30 s-1, amplitude de pulsos (a) = 50 mV e incremento de varredura (ΔEs) = 2 mV para os corantes. Relações lineares foram obtidas nos intervalos de 2,0 x 10-7 mol L-1 a 9,0 x 10-7 mol L-1, 1,5 x 10-6 mol L-1 a 5,5 x 10-5 mol L-1, 2,0 x 10-7 mol L-1 a 1,2 x 10-7 mol L-1 para os corantes BB16, BR51 e BY57, respectivamente. Foram obtidos limites de detecção de 1,01 x 10-8 mol L-1, 1,19 x 10-7 mol L-1 e 2,89 x 10-7 mol L-1 para os corantes BB16, BR51 e BY57, respectivamente. O método foi aplicado em amostras de extratos de fios de cabelo, água de lavagem, água de torneira e formulações comerciais. A determinação simultânea dos corantes BB16, BR51 e BY57 foi também proposto usando o método descrito e otimizado. O método foi aplicado em amostras de água de torneira e água de rio com recuperações de aproximadamente 100% e em amostra da tintura de cabelo comercial (COLOR EXPRESS (Vermelho intenso). O método eletroquímico foi comparado com o método de referência usando cromatografia líquida de alta eficiência com detector de arranjo de diodo (HPLC/DAD). / Hair dyes, mainly coming from commercial hair colorants in beauty salons, when not properly discarded, can generate effluents that are difficult to treat. In addition, hair dye analysis in dyed hair strands could be used as useful information for forensic analysis. Due to the high consumption of hair dyes, which can be found in the form of permanent, semipermanent and temporary dye, with different structures and physicochemical behavior, the demand for sensitive and robust analytical methods for their quantification is highly relevant. Therefore, the present work describes the construction of electrochemical sensors for the quantification of hair dyes of the temporary type, because they are class of dyes easily removed from the hair and, consequently, are released in great quantity for the environment. The electrochemical sensor consists of the development of a composite electrode using modified magnetic particles with carboxylic groups (CFMP) to determine the dyes basic brown 16 (BB16), basic red 51 (BR51), and basic yellow 57 (BY57) in samples of yarn extracts Hair samples, commercial samples of temporary tincture, wash water, samples of river water and tap water. The three dyes analyzed have azide groups as chromophores. In the present work, the oxidation of amines and hydroxyls present as auxochromic groups in the dyes was investigated. The results showed that the oxidation of the functional group in the dyes on the composite electrode modified by carboxylic groups can occur in Ea = 0.44 V, Ea = 0.86 V and Ea = 0.65, respectively for the dyes BB16, BR51, and BY57 and the peak current increases significantly with respect to the electrode without modification. The interaction between dye + CFMP was confirmed by UV/Vis and FTIR spectroscopy. Analytical curves for the BB16, BR51, and BY57 dyes were constructed using the optimized conditions in the square wave voltammetry (SWV) technique: pulse frequency (f) = 30 s-1, pulse amplitude (a) = 50 mV and scanning increment (ΔEs) = 2 mV for the dyes. Linear relationships were obtained at intervals of 2.0 x 10-7 mol L-1 to 9.0 x 10-7 mol L-1, 1.5 x 10-6 mol L-1 at 5.5 x 10-5 mol L-1, 2.0 x 10-7 mol L-1 to 1.2 x 10-7 mol L-1 for the dyes BB16, BR51, and BY57, in this order, using the SWV technique. Detection limits of 1.01 x 10-8 mol L-1, 1.19 x 10-7 mol L-1, 2.89 x 10-7 mol L-1 were obtained for the dyes BB16, BR51, and BY57, respectively. The method was applied to hair strands, wash water, tap water, and commercial formulation samples. The simultaneous determination of the dyes BB16, BR51, and BY57 was also proposed using the method described and optimized. Evaluation psychometrischer Methoden zur Verbesserung der Diagnostik der Zwangsstörung

Die Zwangsstörung ist durch sehr verschiedenartige Symptome charakterisiert. Diese Symptomheterogenität stellt Herausforderungen für die psychologische Diagnostik dar. Die Yale-Brown Obsessive Compulsive Scale (Y-BOCS) gilt als der Goldstandard bei der Messung von Zwangssymptomen. Mittels einer großen Stichprobe von Zwangspatienten hatte diese Arbeit drei Ziele zur Weiterentwicklungen psychometrischer Methoden: Erstens wurden die Messmodelle von Zwangssymptomen evaluiert und verbessert. Dabei fand eine umfassende Überarbeitung statt, wobei Bayesianische Messmodelle genutzt wurden. Zweitens wurde getestet, ob die Messeigenschaften der Y-BOCS über die Zeit und für weitere klinische relevante Variablen konstant sind. In einer breit angelegten Messinvarianzanalyse wurden nur wenige Verletzungen der Messinvarianz gefunden. Falls solche vorliegen, werden Gruppenvergleiche verzerrt und eventuell falsche Schlussfolgerungen gezogen. Für diese Situationen wurde drittens eine Erweiterung partieller Messinvarianzmodelle entwickelt und angewendet. Partielle Messinvarianzmodelle ermöglichen valide Gruppenvergleiche auch dann, wenn Messinvarianz nur für einige wenige Items hält. Es wurde eine Methode zur Modellmittelung entwickelt, die die Unsicherheit berücksichtigt, die in der Auswahl von Items für partielle Messinvarianz liegt. Die vorgelegten Studien dienen der Weiterentwicklung psychometrischer Analysen in der klinischen Diagnostik im Allgemeinen und stärken die Validität der Messung durch die Y-BOCS im Besonderen. Klinische Studien können vertrauenswürdige Ergebnisse nur aufbauend auf soliden Messverfahren erzielen. Außerdem werden weitere neuere Entwicklungen in der psychometrischen Theorie im Hinblick auf ihren Nutzen in der klinischen Diagnostik und dem Verständnis der Zwangsstörung diskutiert. / Obsessive-compulsive disorder (OCD) is characterized by heterogeneous symptoms. Like for many clinical phenomena, this heterogeneity in symptoms poses challenges to psychological assessment. The Yale-Brown Obsessive Compulsive Scale (Y-BOCS) has developed into a gold standard within the past three decades. Using a large sample of patients suffering from OCD, we worked towards three goals in order to advance psychometric methods: First, evaluating and improving the measurement models associated with OCD symptoms by means of an exhaustive overhaul and the usage of Bayesian measurement modeling. Second, to test whether the Y-BOCS' measurement properties are stable across time and other features relevant to clinical research. In a broad analysis of measurement invariance (MI), we found only few instances where MI did not hold. Under such circumstances, group comparisons may be biased and conclusions could be misleading. For such situations, we thirdly derived and applied a procedure extending partial MI modeling. In partial MI models, group comparisons are still valid, even if MI holds only for a few items. We developed a model averaging approach that appropriately reflects the uncertainty stemming from choosing items for partial MI models. The developed Bayesian procedure makes decisions made during the analysis fully transparent and thus open to discussion. The presented studies form a research program that advances psychometrical analyses in clinical assessment and increases the validity of the assessment of OCD by means of the Y-BOCS. Clinical trials require such sound measurements in order to provide trustworthy conclusions. Furthermore, we discuss other recent advances in the field of psychometry and their usability for clinical research as a whole and the understanding of OCD specifically.

Genetic analysis of nitrogen assimilation in the Texas brown tide Aureoumbra lagunensis

The initiation, persistence, and termination of harmful algal blooms (HABs) can all be influenced by nutrient availability. Recent studies have highlighted the role of both organic and inorganic nitrogen sources in HAB dynamics. The pelagophyte Aureoumbra lagunensis causes ecosystem disruptive algal blooms and is responsible for the longest recorded harmful algal bloom (1989-1997). Because of Aureoumbra's small size and its inability to use nitrate, it has been hypothesized that its ability to use ammonium and organic nitrogen, especially at low concentrations, contributed to the unusual persistence of this bloom. This project aimed to assess the response of Aureoumbra to inorganic and organic nitrogen sources by examining the expression of genes responsible for nitrogen assimilation, with an eventual intent of developing expression assays that are indicative of nitrogen source use and/or sufficiency in Aureoumbra. Large volume batch cultures of Aureoumbra were grown with either ammonium or urea as a nitrogen source. Physiological characteristics (C:N, chlorophyll [alpha] cell⁻¹, and Fv/Fm) were monitored throughout the growth period, and the expression of the AMT-1, AMT-2 and UREC genes was assayed at early-, mid- and late-exponential phases. The results show that Aureoumbra can use both ammonium and urea, and that it is well adapted to low-nutrient environments. Only one gene, AMT-1, appeared to be transcriptionally regulated in response to changing nitrogen concentration, and only to ammonium. The results of this study contribute to our understanding of how algae in general cope with low nutrient availability and should ultimately help to define the dynamics of these HAB events. / text

Searching for brown dwarf companions

In this thesis I present the search for ultracool dwarf companions to main sequence stars, subgiants and white dwarfs. The ultracool dwarfs identified here are benchmark objects, with known ages and distances. The online data archives, the two micron all sky survey (2MASS) and SuperCOSMOS were searched for ultracool companions to white dwarfs, where one M9 1 companion to a DA white dwarf is spectroscopically confirmed as the widest separated system of its kind known to date. The age of the M9 1 is constrained to a minium age of 1.94Gyrs, based on the estimated age of the white dwarf from a spectroscopically derived Teff and log g and an initial-final mass relation. This search was extended using the next generation surveys, the sloan digital sky survey (SDSS) and the UK infrared deep sky survey (UKIDSS), where potential white dwarf + ultracool dwarf binary systems from this search are presented. A handful of these candidate systems were followed-up with second epoch near infrared (NIR) imaging. A new white dwarf with a spectroscopic M4 companion and a possible wide tertiary ultracool component is here confirmed. Also undertaken was a pilot imaging survey in the NIR, to search for ultracool companions to subgiants in the southern hemisphere using the Anglo-Australian telescope. The candidates from that search, as well as the subsequent follow-up of systems through second epoch NIR/optical imaging and methane imaging are presented. No systems are confirmed from the current data but a number of good candidates remain to be followed-up and look encouraging. A search for widely separated ultracool objects selected from 2MASS as companions to Hipparcos main-sequence stars was also undertaken. 16 candidate systems were revealed, five of which had been previously identified and two new L0 2 companions are here confirmed, as companions to the F5V spectroscopic system HD120005 and the M dwarf GD 605. The properties of HD120005C were calculated using the DUSTY and COND models from the Lyon group, and the age of the systems were inferred from the primary members. For GD 605B no age constraint could be placed due to the lack of information available about the primary, but HD120005C has an estimated age of 2-4Gyr. In the final part of this thesis I investigate correlations with NIR broadband colours (J - H, H - K and J - K) with respect to properties, Teff , log g and [Fe/H] for the benchmark ultracool dwarfs, both confirmed from the searches undertaken in this work and those available from the literature. This resulted in an observed correlation with NIR colour and Teff, which is presented here. I find no correlation however with NIR colours and log g or [Fe/H], due in part to a lack of suitable benchmarks. I show that despite the current lack of good benchmark objects, this work has the potential to allow UCD properties to be measured from observable characteristics, and suggest that expanding this study should reveal many more benchmarks where true correlation between properties and observables can be better investigated.

Energy flow and metabolic efficiency attributed to brown adipose tissue

The large capacity of brown adipose tissue (BAT) to expend energy as heat makes it an interesting potential player in weight regulation and other metabolic conditions. This is of particular interest as it has been recognized that adult humans possess BAT. The protein responsible for the heat production is uncoupling protein 1 (UCP1), which, as the name implies, uncouples the respiratory chain from ATP production; instead heat is produced. Cold is the strongest recruiter and activator of BAT. However, also obesogenic food has a low but nonetheless significant effect on the recruitment and activation of UCP1, although the significance of this has been discussed. In the present thesis, I have studied the effect of diet on BAT and the possibilities for it to be obesity-protective. This can be done by comparing responses in wild-type mice and in UCP1-ablated mice. Since the effect of diet on BAT is low, it is of importance to control the temperature and maintain thermoneutrality. Other confounding factors to keep in mind are differences in actual energy and composition of food and also cohort differences. When controlling all the parameters mentioned and giving the mice the same obesogenic diet, the mice possessing UCP1 compared to UCP1-ablated mice had higher energy expenditure, and lower weight gain, despite eating more. This confirms the presence of a UCP1-dependent diet-induced thermogenesis. Thus, the conclusion must be that possessing UCP1 does result in obesity protection at thermoneutrality. β-Adrenergic Signalling Through mTOR

Adrenergic signalling is part of the sympathetic nervous system and is activated upon stimulation by the catecholamines epinephrine and norepinephrine. This regulates heart rate, energy mobilization, digestion and helps to divert blood flow to important organs. Insulin is released to regulate metabolism of carbohydrates, fats and proteins, mainly by taking up glucose from the blood. The insulin and the catecholamine hormone systems are normally working as opposing metabolic regulators and are therefore thought to antagonize each other. One of the major regulators involved in insulin signalling is the mechanistic target of rapamycin (mTOR). There are two different complexes of mTOR; mTORC1 and mTORC2, and they are essential in the control of cell growth, metabolism and energy homeostasis. Since mTOR is one of the major signalling nodes for anabolic actions of insulin it was thought that catecholamines might oppose this action by inhibiting the complexes. However, lately there are studies demonstrating that this may not be the case. mTOR is for instance part of the adrenergic signalling pathway resulting in hypertrophy of cardiac and skeletal muscle cells and inhibition of smooth muscle relaxation and helps to regulate browning in white adipose tissue and thermogenesis in brown adipose tissue (BAT). In this thesis I show that β-adrenergic signalling leading to glucose uptake occurs independently of insulin in skeletal muscle and BAT, and does not activate either Akt or mTORC1, but that the master regulator of this pathway is mTORC2. Further, my co-workers and I demonstrates that β-adrenergic stimulation in skeletal muscle and BAT utilizes different glucose transporters. In skeletal muscle, GLUT4 is translocated to the plasma membrane upon stimulation. However, in BAT, β-adrenergic stimulation results in glucose uptake through translocation of GLUT1. Importantly, in both skeletal muscle and BAT, the role of mTORC2 in β-adrenergic stimulated glucose uptake is to regulate GLUT-translocation. / <p>At the time of the doctoral defense, the following paper was unpublished and had a status as follows: Paper 4: Manuscript.</p>
