Emprego da flotação lamelar de alta taxa e convencional como pós-tratamento do efluente de sistema constituído de reator anaeróbio seguido de reator aeróbio tratando esgoto sanitário / Lamella design (high rate) and conventional flotation units applied to the post-treatment of the effluent from a system made up anaerobic reactor followed by aerobic reactor treating domestic sewage

O presente trabalho teve como objetivo o estudo da potencialidade de aplicação da flotação por ar dissolvido (FAD) lamelar (de alta taxa) e convencional como parte final de sistema de tratamento sequencial de esgoto sanitário, constituído de reatores anaeróbios de manta de lodo (UASB) seguidos de tanque de aeração. O trabalho, desenvolvido na Estação de Tratamento de Esgotos Jardim das Flores da cidade de Rio Claro, foi dividido em três etapas. Na primeira etapa, realizada com equipamento Flotateste, foram investigados os valores mais adequados de dosagens de polímero sintético catiônico, amido de araruta, cloreto férrico e associações entre cloreto férrico e polímero ou amido de araruta, de razão ar/sólidos (A/S), bem como de tempo e gradiente de mistura rápida e de floculação. Nessa etapa, os melhores desempenhos da FAD foram obtidos com o emprego de polímero catiônico, tanto isoladamente quanto associado ao cloreto férrico, sendo esta última condição imprescindível para maior remoção de fósforo. A flotação de liquor misto utilizando apenas o cloreto férrico como coagulante, apresentou desempenho extremamente insatisfatório, com a formação de flocos com características ruins de flotabilidade. Além disso, observou-se que a flotação desse tipo de efluente requisitou condições mais amenas de mistura, sendo que, em muitos casos, os melhores resultados foram obtidos mediante a eliminação da mistura rápida ou da floculação. Nas segunda e terceira etapas, realizadas com unidades piloto de flotação lamelar de alta taxa (FADAT) e convencional, respectivamente, e empregando apenas polímero como auxiliar de floculação/flotação (dosagem em torno de 1 g de polímero/kg de SST), foram investigados os valores mais adequados de Taxa de Aplicação Superficial (TAS), Taxa de Aplicação de Sólidos (TS) e quantidade de ar fornecida à flotação, calculada através da razão A/S ou da dosagem de ar (Dar), em g de ar/'M POT.3' de esgoto afluente, dependendo da concentração de sólidos suspensos totais afluentes à flotação (SSTafl). Através dos resultados dos ensaios pôde-se observar que a razão A/S é mais adequada para o cálculo da quantidade de ar na flotação de suspensões concentradas (SSTafl acima de 1100 mg/L no presente estudo), ao passo que Dar é mais representativo no caso da flotação de suspensões diluídas. Empregando a unidade FADAT, alimentada com liquor misto de tanque de aeração em boas condições de biofloculação, foi possível obter resultados satisfatórios para TAS de até 350 'M POT.3'/'M POT.2'.dia e TS entre 120 e 250 kg de SST/'M POT.2'.dia, desde que fornecida quantidade adequada de ar ao processo (Dar na faixa de 15,0 a 18,0 g de ar/'M POT.3' de afluente), para SSTafl entre 800 e 1100 mg/L. Na flotação lamelar de alta taxa, constatou-se que o bom desempenho esteve diretamente relacionado a baixos valores de SSTafl. As recomendações para a flotação com unidade de FAD convencional tiveram que ser mais conservadoras, devido às condições ruins de biofloculação apresentadas pelo liquor misto do tanque de aeração durante a realização da terceira etapa do trabalho. Nesses ensaios, os resultados mais satisfatórios foram obtidos com o emprego de TAS em torno de 200 'M POT.3'/'M POT.2'.dia, TS menor que 200 kg de SST/'M POT.2'.dia e A/S em torno de 0,020 (Dar de 21,0 g de ar/'M POT.3' de afluente), para SSTafl em torno de 1400 mg/L / The aim of this research was the study of the lamella design (high rate) and conventional dissolved air flotation (DAF) process potentiality of application as the final part of a domestic sewage sequential treatment system made up anaerobic reactors (UASB) followed by aeration tank. The work, developed at the Jardim das Flores WWTP of Rio Claro city, was separated in three phases. In the first phase, using the flotatest apparatus, the most suitable values of chemical (synthetic cationic polymer, arrow-root starch, ferric chloride and ferric chloride combined with polymer or arrow-root starch) dosages, air to solids ratio (A/S), as well as rapid mix and flocculation time and mean velocity gradient values, were investigated. In this phase, the best DAF performances were obtained with the application of cationic polymer, alone and associated to ferric chloride. The use of ferric chloride was essential to obtain better phosphorus removal results. The mixed liquor flotation using ferric chloride alone as primary coagulant presented extremely unsatisfactory performance, with the formation of flocks showing bad characteristics of flotability. Moreover, it was observed that flotation of this kind of effluent had requested gentler mixture conditions. In many cases, the best results were obtained by means of rapid mix or flocculation step elimination. In the second and third phases, using the lamella design and conventional flotation units respectively and cationic polymer as the only flocculation/flotation aid (dosage around 1 g of polymer/kg TSS), the most suitable values of Overflow Rate (OR), Solids Application Rate (SAR) and amount of air supplied to the flotation process, expressed as the A/S ratio or air dosage (AD, in g of air/'M POT.3' of influent) depending on influent total suspended solids concentration (TSSinfl), were investigated. The flotation essays results showed that the A/S ratio is more suitable for the air supply estimation in flotation of concentrated suspensions (TSSinfl above 1100 mg/L in this study). Contrarily, the parameter AD is more representative in cases of flotation of diluted suspensions. Using the lamella design unit, fed with aeration tank mixed liquor presenting good bio-flocculation conditions, it was possible to obtain adequate results applying OR as high as 350 'M POT.3'/('M POT.2'.day) combined with SAR values between 120 and 250 kg of TSS/('M POT.2'.day), since the adequate amount of air has been provided for the process (AD values between 15,0 and 18,0 g of air/'M POT.3' of influent), for TSSinfl between 800 and 1100 mg/L. For the lamella flotation, it was noticed that the best performance has been directly associated to lower values of TSSinfl. The recommendations for flotation using the conventional DAF unit had to be more conservative, taking into account the poor bio-flocculation conditions presented by the mixed liquor during the third phase of work. In theses essays, the more adequate results were obtained by applying OR around 200 'M POT.3'/('M POT.2'.day), SAR under 200 kg of TSS/('M POT.2'.day) and A/S ratio around 0,020 (AD of 21,0 g of air/'M POT.3' of influent), for TSSinfl around 1400 mg/L

A proteína A de superfície de pneumococo (PspA) é indispensável para a virulência da bactéria e foi escolhida para a elaboração de uma nova vacina conjugada contra S. pneumoniae. Para tanto foi desenvolvido um processo industrial de produção e purificação do fragmento recombinante da PspA clado 3 em E. coli. Cultivos descontínuos alimentados foram estabelecidos com glicose ou glicerol em reator de 5L, obtendo-se 62g/L de células secas e 3g/L de PspA3. As células foram lisadas por homogeneizador contínuo de alta pressão com eficiência de 96,7%. A centrifugação foi definida como etapa de clarificação. A sequência cromatográfica troca aniônica seguida de afinidade por Ni+2 rendeu os melhores resultados de pureza (81%) e recuperação (70%). A cromatografia de troca catiônica foi selecionada como terceira etapa do processo, definindo assim um processo de produção e purificação escalonável que possibilitou a obtenção de PspA3 com alto grau de pureza (90%). / The pneumococcal surface protein A (PspA) is indispensable for virulence of S. pneumoniae and it was the first choice as carrier for a new conjugated vaccine against S.pneumoniae. Hence, the purpose of this work was to develop an industrial production and purification process of a recombinant fragment PspA clade 3 (rfPspA3) in E. coli. Fed-batch cultivations in 5 L bioreactors with defined medium were carried out using glucose or glycerol as carbon sources. It was obtained 62 g/L of dry cell weight and 3 g/L of rfPspA3. Cells were disrupted with 96.7% of efficiency by high pressure continuous homogenizer. Centrifugation was defined for the clarification step. The sequence with Q- followed by IMAC-Sepharose yielded the best purity and recovery of rfPspA3 (81 and 70%, respectively). Cation exchange was chosen for the last chromatography. In conclusion, an industrial production and purification process was developed and rfPspA3 was obtained with high purity (90%).

O processo de desencantamento do mundo iluminista, que prometia liberdade, certezas científicas e progresso, transformou a razão em instrumento de dominação e mascarou a realidade, instaurando, assim, um sistema administrado. Esta estrutura provocou o estreitamento da racionalidade crítica e eliminou a tensão entre conceito e realidade. Escondeu também a real política, pois o sistema administrado não considera as singularidades, o não idêntico, aquilo que aparece de contraditório na vida humana e na sociedade. Dessa forma, o objetivo da política fica encoberto, torna-se um conceito abstrato que está a serviço do sistema instrumental burguês. A real política é mais do que aparece de imediato na totalidade da sociedade administrada, por isso, nosso objetivo é fazer um exercício, nas obras de Theodor Adorno, da estrutura que apreende e direciona a política, compreendendo como este posicionamento leva o indivíduo e a sociedade à decadência. Mas também, pretendemos, a partir do pensamento de Adorno, apontar perspectivas que podem levar a humanidade e a sociedade para processos emancipatórios através do que, seguindo a terminologia de Adorno, denominamos de política negativa. Para encontrar as perspectivas, acreditamos que é preciso tirar a casca ao redor da política, expor a realidade na qual ela está inserida e dar dinamicidade para que o heterogêneo viva. Nesse sentido fica a pergunta: isso poderia ser feito através da dinâmica da dialética negativa? Na teoria de Adorno, a dialética não se completa, ela se detém no momento da negatividade, articulando e tencionando a razão permanentemente e à medida que ocorre a reflexividade, a razão vai iluminando e qualificando a realidade sem torná-la abstratamente conceitual, ou seja, os elementos se cristalizam por um movimento objetivo projetando novas constelações. Essa dinâmica, de dialética negativa, pode tensionar, articular e iluminar a política, trazendo à tona possibilidades emancipatórias da vida e de uma sociedade melhor sem torná-la administrada. Esta pesquisa analisa se a política negativa pode desobstruir a camada ideológica que oculta as contradições e trazer à tona uma nova configuração, uma outra realidade, ou ainda, a verdadeira realidade. Também tenciona compreender se uma política negativa pode manter a tensão, um campo de forças e pode ir além da superfície imediata das coisas ao não se contentar com as aparências, e se pode resistir e contrapor a estrutura burguesa e a falsidade do todo pela verdade. Com estas questões abertas, nos desafiamos a construir uma reflexão, através da política negativa na obra de Theodor Adorno, por acreditarmos que não existe uma teleologia da sociedade e nem uma racionalidade dos acontecimentos previamente fixados. / The following thesis aims to thematize the possibility of a Negative Politics in Adorno in order to think about the emancipated life. The disenchantment process of the enlightenment world, that promised freedom, scientific certainty and progress, transformed reason into a tool of domination and masked the reality, establishing, a managed system. This structure caused the narrowing of critical rationality and eliminated the tension between concept and reality. It also hid the real politics, because the managed system does not consider the singularities, the non identical, something that appears contradictory in human life and in society. In this way, the objective of politics is covered, it becomes an abstract concept, at the service of the instrumental bourgeois system. The real politics is more than what immediately appears in the totality of the managed society, thats's why, our objective is to make an exercise, in the works of Theodor Adorno, the structure that apprehends and directs the politics understanding how this positioning takes the individual and the society to the decay. But also, we intend, from Adorno thoughts, point out some perspectives that can lead humanity and society to emancipatory processes which, following the terminology of Adorno, we call negative politics. To find the perspectives, we believe that it is necessary to take the bark around politics, to expose the reality in which it is inserted and to give dynamicity so that the heterogeneous could live. In this sense the question remains: could this be done through the dynamics of negative dialectics? In Adorno's theory, the dialectic is not complete, it stops at the moment of negativity, articulating and intending the reason permanently as the reflexivity occurs, the reason illuminates and qualifies reality without making it an abstract concept, the elements crystallize by an objective movement projecting new constellations. This dynamics, of negative dialectics, can envisage, articulate and illuminate politics, bringing the emancipatory possibilities of life and a better society without making it administered. This research analyzes can negative politics erase the ideological layer that hides contradictions and bring out a new configuration, another reality, or a real reality. It also compromises san a negative policy keep a tension, a force field, and can it go beyond the immediate surface of things by not being content with appearances, can it resist and oppose the bourgeois structure and the falsity of the whole by the truth. With these open questions, we challenge ourselves to construct a reflection in the books of Theodor Adorno, through negative politics, believing, that there is no teleology of society and no rationality of events previously fixed.

Target Practice: Exploring Student TL/L1 Use in Paired Interactions

Teachers of foreign languages typically encourage their students to speak in the target language (TL) often, but due to various factors, this is not always achieved. Some reasons might include insufficient vocabulary, lack of topic knowledge, embarrassment, or simple unwillingness. Much of the existing research observes uses of the TL or native language (L1). The purpose of this study was to examine how often students use the TL in paired interactions and whether that amount has any relationship to the students' oral proficiency at the end of the course. In a SPAN 105 course at Brigham Young University, 27 students participated in this project by recording themselves during six in-class, paired interactions, after which they provided comments via questionnaires. This study was conducted using a mixed-methods approach, with both quantitative and qualitative data. The quantitative data revealed information about the time spent in the TL, as well as the time spent in the L1, and what relationship these variables had with listening comprehension and oral proficiency. The qualitative data exposed emergent findings related to TL/L1 use: helpful tools, effects of task type, pitfalls the students experienced, struggles and benefits of partnering, effects of recording, and student perceptions about the L1. The results of the study indicate a need for teacher strategies to encourage TL use in the classroom.

The aim of the present paper was to study the pragmatic ability in terms of request for clarification in children with Hearing Impairment (HI) in conversation with children with no known hearing impairment, henceforth normally hearing (NH). The present study also describes requests for clarification sequentially in detail, and also highlights points where there may be observable differences between children with and without hearing impairment. Eight conversational pairs consisting of one child with HI and one NH child were studied. Children’s Communication Checklist (CCC) was filled out by the parents and teachers of the children with HI in order to complement the description of the pragmatic ability in children with HI. Conversation Analysis was used in order to make the study of interaction as open-minded as possible. The conversation extracts were grouped according to task or non-task oriented interaction based on the activities that occurred.</p><p>The results of the present study showed that the children with HI and NH both used non-specific and specific requests for clarification, and that these types of requests were used in both task and non-task oriented interaction. Children with HI used the non-specific request "sorry?" to greater extent in the extracts than children with NH did. In general, no differences in occurrence of specific requests were seen between children with HI and children with NH. As regards to what happened before and after requests for clarifications, no distinct differences could be established between children with HI and children with NH. In conclusion, it is not certain if and how a hearing impairment have affected the pragmatic ability, although tendencies in the conversation extracts indicated that hearing impairment sometimes affected the interaction.</p> / <p>Pragmatik är beroende av många förmågor, däribland hörseln. "Har han blå eller röda ögon?" : En studie om referentiell kommunikation och ordförråd hos personer med utvecklingsstörning

Ett flertal studier har undersökt användandet av olika typer av efterfrågan om förtydligande i en referentiell kommunikationsuppgift. Bland annat har skillnader i kommunikationsstrategier mellan barn med cochleaimplantat och normalhörande barn påvisats. Syftet med föreliggande studie var att undersöka huruvida kommunikationsstrategier skiljer sig åt i en referentiell kommunikationsuppgift hos barn med lindrig till måttligutvecklingsstörning och en mentalt åldersmatchad kontrollgrupp. Vidare syfte var att undersöka hur barn med lindrig till måttlig utvecklingsstörning reagerar vid kommunikationssammanbrott, när personen inte har tillgång till en specifik referent. Studien har också undersökt vokabulär hos de båda grupperna och om detta har någon betydelse för vilka kommunikationsstrategier de väljer. I studien ingick två grupper. Den ena gruppen bestod av personer med utvecklingsstörning som har en kronologisk medelålder på 14:9 år, och kontrollgruppen bestod av barn utan utvecklingsstörning med en kronologisk medelålder på 7:8 år. Båda grupperna hade en mental medelålder på 8:6 år. Barnets kommunikativa strategier skattades av logopedstudenter i en referentiell kommunikationsuppgift och ordförrådsdelen i WISC-IV användes för att kunna jämföra ordförråd. Då idén till föreliggande studie föddes ur Ibertsson et al. studie från 2009 Deaf teenagers with cochlear implants in conversation with hearing peers har samma typer av efterfrågan om förtydligande använts som utgångspunkt i analys. Resultaten visade att personer med utvecklingsstörning kan skilja sig i användandet av olika typer av efterfrågningar från en mentalt åldersmatchad grupp. Vissa typer av efterfrågningar korrelerade signifikant med varandra i högre grad inom kontrollgruppen än inom målgruppen. Prestation på ordförrådstestet skiljde sig inte signifikant mellan grupperna. The purpose of this study was to examine whether communication strategies differs between children with developmental disabilities and mental age matched peers in a referential communication task, and how the children react to a communication breakdown. We tested vocabulary to see if vocabulary correlates with different types of requests for clarification. Our study included two groups. One group consisted of people with development disabilities with chronological mean age of 14:9 years. The other group consisted of mental age matched children with no development disabilities. The mental mean age of both groups was 8:6 years. The children’s communicative strategies were assessed in a referential communication task. The vocabulary was examined with the vocabulary subtest of WISC-IV. Our results indicate that children with mental disabilities differ from mental age matched peers in the use of types of requests for clarification in this study. Different types of requests correlates with each other in both groups. This indicates that the persons in the groups use different strategies in solving the task. The vocabulary was not different between the groups.

CALLing all learners: An explanatory integrative research study of EFL learner-learner corrective feedback patterns within on-line synchronous environments

This mixed methods research study centers on learner-learner interactions; thus, contributing to the on-going investigation within negotiation and interaction, computer-mediated-communication and its role in second language learning. The specific aim was to investigate corrective feedback types, incidences, and the relationship between error and feedback type among peers within online synchronous environments in EFL classes in Slovenia, Europe. Interactional characteristics of corrective feedback with learners having a documented special need (SN) also were explored using qualitative analyses. The study encompassed 208 students that were randomly placed into 104 dyads within intact classes of Grades 7, 8, 10, and 11. There were 32 dyads in Grade 7, 16 dyads in Grade 8, 24 dyads in Grade 10, and 32 dyads in Grade 11. Three participants had a documented special need. Quantitative analysis did not reveal statistical significant difference in the incidence of corrective feedb feedback and grade level, the relationship among the type of corrective feedback and grade level, or the relationship between learner error and type of corrective feedback across grade levels. Corrective feedback types were similar to those studied in traditional classroom research (i.e., explicit corrections, recasts, negotiation of form). However, descriptive statistics and qualitative analyses revealed conversational techniques that are specific to text-based online discourses providing insight into interactional characteristics among interactants within a discourse environment that differs both from speech and written texts. Consequently, an additional corrective feedback type emerged from the data, coded as feedback request. The most frequent corrective feedback type provided was explicit corrections. Frequency data revealed that corrective feedback tended to decrease as the grade level increased. Data with SN learners indicated distinctive discourse techniques.Overall, low incidences of corrective feedback and error types might have been affected by the learner's developmental levels, social readiness, and/or psychological readiness (Oliver, 1998), as well as the learner's individual conversational styles and socio-cultural factors. Consequently, further research is warranted in examining these factors. In addition, longitudinal studies are warranted in examining whether online negotiated work lead towards L2 acquisition. Finally, the role of phantom corrective moves when coding qualitative online text data also need to be examined further.

SELF-CARE: A CLARIFICATION OF MEANING AND EXAMINATION OF SUPPORTIVE STRATEGIES

Background Considering ~9 million Canadians have one or more chronic health conditions, and >3.3 million report some level of disability, the burden of care is substantive for individuals and health system. With such conditions, self-care is essential but may pose challenges to both individuals and providers of care. As a concept, self-care is poorly understood. Further, evidence for effective self-care support is diffuse and typically studied relative to specific conditions. Objectives To investigate the concept of self-care three objectives were undertaken: 1) explore and describe the construct of self-care as understood by individuals/families, health care professionals, researchers, policy-makers; and industry; 2) produce new knowledge for health care professionals about interventions for self-care across a range of population groups; 3) develop a provisional framework to inform practice and research. Method A multi-phase enquiry was undertaken. Phase 1 Concept clarification including: 1) synthesis of qualitative evidence on the experience of self-care reported by individuals/families; 2) content analysis and definitional study of the meaning of self-care; 3) concept analysis of self-care; 4) creation of a conceptual schema encompassing these perspectives. Phase 2 Self-care Interventions: Integrative study of systematic reviews, synthesizing evidence for self-care interventions from multiple disease/impairment groupings. Results Three modes of self-care were revealed : ‘Care of self’ self-care performed on one’s own behalf; ‘care by other’ acknowledging individuals with disabilities who guide and direct care provided by another person; and ‘care of other’- care of families and others at a community level. Analysis of 30 self-care interventions across 16 conditions demonstrated that educational sessions and self-care management plans are emerging as effective strategies to support and guide self-care. Conclusion Self-care is a complex care concept that is becoming an expected element in today’s health care environment. A full understanding of how it is viewed, including the individual’s perspective, is vital for enactment and beneficial support. Flexographic deinking with electric field technology by destabilization and flotation

Every year, millions of tons of paper are diverted from landfills and recycled. Newspaper constitutes a large portion of total paper recycled, providing a cheap source of raw material for the paper industry and helping sustainable forestry. The recycling of newsprint paper involves the separation of ink from the newsprint, which is done either by flotation or washing. Conventional flotation processes for separating ink are not adequate for newsprint printed using flexography printing technique and with water-based ink. The removal of these flexographic water-based inks by washing is a better alternative. However, one drawback of washing is that it has lower yield. In addition, the subsequent wash filtrate is difficult and costly to decontaminate. The overall goal is to develop a combination of processes that can remove ink from a feedstock that contains up to 100% flexographic ink newsprint; in the context of process variables with known effects. In the present work the objectives are to (1) demonstrate that incorporating an electric field into a conventional deinking process improves deinking efficiency, (2) propose a mechanism of how incorporating an electric field helps to improve deinking efficiency, (3) demonstrate that an electric field can decontaminate water containing flexographic inks and identify the mechanism behind electric field clarification of water, and (4) demonstrate that by incorporating electric fields into both the flotation deinking stage and water decontamination, the target deinking efficiency can be achieved.
