L’addiction à l’alimentation (AA) est un concept ancien, mais encore sujet de controverses qui toucherait 18 à 24 % des obèses. Dans cette thèse, nous nous sommes intéressés aux intrications entre régulations du comportement alimentaire, addiction et AA, afin de mieux comprendre les mécanismes liés à la désinhibition de la prise alimentaire. Notre travail a conduit à plusieurs résultats : 1) Une revue de la littérature a permis de montrer l’existence d’interaction étroite entre les niveaux de régulation du comportement alimentaire et comment AA pourrait en être un exemple de dérégulation. 2) Une étude de cohorte a permis de démontrer l’existence de caractéristiques cliniques communes entre addictions et AA et celle d’un probable transfert d’addiction de la nicotine vers l’alimentation. 3) Sur le plan expérimental, nous avons montré qu’il existe des perturbations des ondes P300 et N200, dans l’obésité et la désinhibition alimentaire, proches de celles observées dans les addictions. 4) Enfin, nos résultats suggèrent le rôle potentiel de la ghréline comme marqueur de risque de trouble du comportement alimentaire. Ce travail ouvre des perspectives sur le plan expérimental, avec la suggestion de groupes contrôles plus pertinents ; cliniques, avec la création d’un potentiel test de dépistage rapide ; thérapeutiques, avec la mise en place d’un essai thérapeutique par tDCS / Food addiction (FA) is an old concept, but still subject to controversy. In this thesis, we are interested in overlaps between food behavior regulation, addiction and FA, in order to better understand the mechanisms linked to food intake disinhibition. Our work leads to several results: 1) A review of the literature has shown the existence of close interaction between eating behavior regulation levels and how FA could be an example of their disturbance. 2) A cohort study demonstrated the existence of common clinical features between addiction and FA and a probable addiction transfer from nicotine to food. 3) From an experimental point of view, we have shown that there are disturbances of P300 and N200 ERP, in obesity and food disinhibition, close to those observed in addictions. 4) Finally, our results suggest the potential role of ghrelin as a marker for eating disorders increased risk. Orbitofrontal metabolic impairment is characteristic of the frontal variant of frontotemporal dementia (fv-FTD), as are early changes in emotional and social conduct. Two main types of behavioral disturbances have been distinguished in fv-FTD patients: apathetic and disinhibited manifestations. In this study, we searched for relationships between brain metabolism and presence of apathetic or disinhibited behavior. Metabolic activity and behavioral data were collected in 41 fv-FTD patients from European PET centers. A conjunction analysis of the PET data showed an expected impairment of metabolic activity in the anterior cingulate, ventromedial and orbital prefrontal cortex, the dorsolateral prefrontal cortex and the left anterior insula in fv-FTD subjects compared to matched controls. A correlation was observed between disinhibition scores on the Neuropsychiatric Inventory scale and a cluster of voxels located in the posterior orbitofrontal cortex (6, 28, –24). Comparison of brain activity between apathetic and nonapathetic fv-FTD patients from two centers also revealed a specific involvement of the posterior orbitofrontal cortex in apathetic subjects (4, 22, –22). Neural mechanisms of short-term visual plasticity and cortical disinhbitionParks, Nathan Allen 06 April 2009 (has links)
Deafferented cortical visual areas exhibit topographical plasticity such that their constituent neural populations adapt to the loss of sensory input through the expansion and eventual remapping of receptive fields to new regions of space. Such representational plasticity is most compelling in the long-term (months or years) but begins within seconds of retinal deafferentation (short-term plasticity). The neural mechanism proposed to underlie topographical plasticity is one of disinhibition whereby long-range horizontal inputs are "unmasked" by a reduction in local inhibitory drive. In this dissertation, four experiments investigated the neural mechanisms of short-term visual plasticity and disinhibition in humans using a combination of psychophysics and event-related potentials (ERPs). Short-term visual plasticity was induced using a stimulus-induced analog of retinal deafferentation known as an artifical scotoma. Artificial scotomas provide a useful paradigm for the study of short-term plasticity as they induce disinhibition but are temporary and reversible. Experiment 1 measured contrast response functions from within the boundaries of an artificial scotoma and evaluated them relative to a sham control condition. Changes in the contrast response function suggest that disinhibition can be conceived of in terms of two dependent but separable processes: receptive field expansion and unrestricted neural gain. A two-process model of disinhibition is proposed. A complementary ERP study (Experiment 2) recorded visual evoked potentials elicited by probes appearing within the boundaries of an artificial scotoma. Results revealed a neural correlate of disinhibition consistent with origins in striate and extrastriate visual areas. Experiment 3 and 4 were exploratory examinations of the representation of space surrounding an artificial scotoma and revealed a neural correlate of invading activity from normal cortex. Together, the results of these four studies strengthen the understanding of the neural mechanisms that underlie short-term plasticity and provide a conceptual framework for their evaluation.

versteuring met hiperaktiwiteit(ATHV/H) verband hou met probleme
met voortgesette aandag, terwyl aandagtekort versteuring sander
hiperaktiwiteit(ATHV/SO) verband hou met probleme met gefokusde
aandag. Neurosielkundige toetse waarmee voortgesette- en
gefokusde aandag gemeet word, word gebruik om drie groepe,
naamlik ATHV/H(n=24), ATHV/SO(n=l5) en 'n kontrole groep(n=21),
met mekaar te vergelyk. Resul tate dui daarop dat die groep
ATHV/SO beduidend swakker presteer as die ander twee groepe op
sommige van die toetse van gefokusde aandag, en ook neig om
swakker te presteer op die toets van voortgesette aandag. Die
groep ATHV/SO se probleme blyk gekoppel te wees aan 'n stadiger
spoed van kognitiewe prosessering en 'n toestand van hipoopwekking.
Die groep ATHV/H presteer nie beduidend swakker as
die ander twee groepe op die toets van voortgesette aandag nie,
maar presteer wel swakker as die kontrole groep op die Stroop
Colour-Word Test waar inhibisie van ontoepaslike response vereis

disorder with hyperactivity(ADHD/H) is related to problems with
sustained attention, while attention deficit disorder without
hyperactivity (ADHD/WO) is related to problems with focussed
attention. The performance of three groups: ADHD/H (n=24),
ADHD/WO(n=15) and a control group(n=21) were compared. Results
indicated that the ADHD/WO group performed significantly poorer
than the other two groups on some of the tests of focussed
attention and also tended to perform poorer on the test assessing
sustained attention. This groups' symtoms appear to be related
to a slowed speed of processing and a state of hypo-arousal. The
ADHD/H group did not perform poorer than the other two groups on
the test assessing sustained attention. The ADHD/H groups'

Ces travaux abordent la question de l'impact du pouvoir et du contexte privé ou public sur la sensibilité à l’influence normative. Nous défendons la thèse selon laquelle le contexte régule les effets du pouvoir sur la sensibilité à l’influence normative. Dans une première partie, nous avons testé l'hypothèse selon laquelle la possession de pouvoir, par la désinhibition qu’il suscite, rendrait les individus moins sensibles à la norme. Nous présentons deux études qui montrent que la possession de pouvoir occasionne des comportements moins conformes aux normes ainsi qu’une présentation de soi socialement moins désirable. Dans une seconde partie, nous testons l’hypothèse selon laquelle l’activation d’un pouvoir élevé entrainerait une plus forte désinhibition des conduites en privé et une plus forte inhibition des conduites en public que l’activation d’un pouvoir faible. Deux études ont été réalisées et tendent à soutenir cette idée. Une dernière recherche a été menée en vue d’examiner le rôle de l’activation contextuelle de la responsabilité sociale sur la relation entre le pouvoir et la sensibilité à l’influence normative. Toutefois, il semble que l’activation de la responsabilité sociale ne médiatise pas les effets modérateurs du contexte sur la normativité des jugements effectués. Dans l'ensemble, ces travaux contribuent à éclaircir les effets paradoxaux du pouvoir qui ont été observés dans la littérature, en proposant un modèle permettant de comprendre comment le contexte génère des processus différents en fonction du pouvoir, quand la désinhibition et l’inhibition sont susceptibles de se produire, et quelles en sont les conséquences sur les conduites individuelles. / This work investigates the impact of social power and private or public context on normative influence. We defend the thesis according to which the context (private vs. public) regulates the effects of power on normative influence. In a first series of research, we tested the hypothesis that the possession of power, by generating disinhibition, would make individuals less sensitive to social norms. Two studies are presented and show that the possession of power causes less normative behaviors as well as less socially desirable self-presentations. In a second series of research, we argue that high power primed individuals would exhibit greater disinhibition in private and greater inhibition in public than low power primed individuals. Two studies were conducted and tend to support this idea. A last study examined social responsibility as a potential moderator of the effects of social power and context on normative influence. However, it seems that social responsibility does not mediates the inhibition and disinhibition patterns we observe. Overall, these studies help to clarify the paradoxical effects of power underlined in the literature, by proposing a model which allows to understand how the context generates different processes depending on social power, when disinhibition and inhibition are likely to occur, and what are the consequences for individual behavior.

Psychological and Cognitive Factors that Influence Post-Exercise Energy Intake in Normal Weight and Overweight Sedentary MalesHarris, Cristen Lynn 26 March 2008 (has links)
The primary purpose of this study was to evaluate the effects of a single bout of moderate-intensity exercise on acute (ad libitum lunch) post-exercise energy intake (PE-EI) and 12-hour energy intake in normal-weight and overweight sedentary males. Accuracy in estimating energy intake (EI) and energy expenditure (EE), solid vs. liquid carbohydrate intake, mood, and perceived hunger were also assessed. The study consisted of two conditions, exercise and rest, with each subject participating in each condition, in a counterbalanced-crossover design on two days. The participants were randomly assigned to either the exercise or resting (seated) control condition on the first day of the experiment, and then the condition was reversed on the second day. Exercise consisted of walking on a treadmill at moderate-intensity for 60 minutes. Eighty males, mean age 30+8 years were categorized into five groups according to weight status (overweight/normal-weight), dietary restraint status (high/low), and dieting status (yes/no). The main effects of condition and group, and the interaction were not significant for acute (lunch) or 12-hour PE-EI. Overall, participants estimated EE for exercise at 46% higher than actual exercise EE, and they estimated EE for rest by 45% lower than actual resting EE. Participants significantly underestimated EI at lunch on both the exercise and rest days by 43% and 44%, respectively. Participants with high restraint were significantly better at estimating EE on the exercise day, and better at estimating EI on the rest day. Mood, perceived hunger, and solid vs. liquid carbohydrate intake were not influenced by dietary restraint, weight, or dieting status. In conclusion, a single bout of moderate-intensity exercise did not influence PE-EI in sedentary males in reference to dietary restraint, weight, and dieting status. Results also suggested that among sedentary males, there is a general inability to accurately estimate calories for moderate-intensity physical activity and EI. Inaccurate estimates of EE and EI have the potential to influence how males manage their weight.

Sharp wave-ripple Komplexe (SWRs) sind kurze Ereignisse von kohärenter Netzwerkaktivität im Hippocampus.
SWRs spielen eine wichtige Rolle bei der Konsolidierung von expliziten Gedächtnisinhalten, die Mechanismen sind aber bis heute ungeklärt.
Pyramidenzellen (PYR) und Parvalbumin-positive Korbzellen (PV+BCs) feuern während SWRs besonders häufig, wohingegen sie außerhalb beinahe inaktiv sind.
SWRs treten spontan auf, und können durch Stimulation von PYR und PV+ Zellen hervorgerufen werden.
Um die Rolle von PV+ Zellen in SWR Generierung zu klären, untersuche ich wie das Zusammenspiel von exzitatorischen Neuronen (PYR) und zwei Klassen von Interneuronen (PV+BCs und derzeit unbekannte Anti-SWR-Zellen) die Entstehung und die Häufigkeit von SWRs beeinflusst.
Erstens entwickle ich ein Netzwerk aus feuernden Neuronen, das spontane Übergänge vom Anti-SWR-Zustand zum SWR-Zustand zeigt. Die Aktivität von PV+BCs, die die Aktivität von PYR disinhibieren, dominiert den SWR-Zustand.
SWRs können hervorgerufen werden durch Stimulation von PYR oder PV+BCs, und durch Inaktivierung von Anti-SWR-Zellen.
Durch Kurzzeitdepression der synaptischen Verbindung von PV+BCs zu Anti-SWR-Zellen wird die Dauer der SWRs reguliert.
Die Koexistenz von Anti-SWR- und SWR-Zuständen bei konstanten Stärken der synaptischen Depression erlaubt die Untersuchung der Bistabilität des Netzwerks. Durch eine Mean-field-Näherung können Voraussetzungen für bistabile Netzwerkaktivität analytisch hergeleitet werden.
Das Modell prognostiziert die Existenz von Anti-SWR-Zellen. Im letzten Teil dieser Arbeit zeige ich erste experimentelle Ergebnisse, die die Existenz von CA3-Interneuronen belegen, die anti-moduliert sind bezüglich SWRs.
Durch die Untersuchung der Rolle von Interneuronen hinsichtlich der Generierung von SWRs trägt diese Arbeit zu einem tieferen Verständnis der neuronalen Schaltkreise im Hippocampus bei, die essentiell für den Erwerb und die Konsolidierung expliziter Gedächtnisinhalte sind. / Sharp wave-ripple complexes (SWRs) are events of coordinated network activity originating in the hippocampus. SWRs are thought to mediate the consolidation of explicit memories, but the mechanisms underlying their occurrence remain obscure.
Pyramidal cells (PYR) and parvalbumin-positive basket cells (PV+BCs) preferentially fire during SWRs and are almost silent outside. SWRs emerge spontaneously or by activating PYR or PV+ cells. To understand how the activation of PV+ interneurons can result in an increase of PYR firing, I explore how the interaction of excitatory neurons (PYR) and two groups of interneurons (PV+BCs and a class of anti-SWR cells) contributes to the initiation, termination, and incidence of SWRs.
First, I show that a biophysically constrained network of spiking neurons can exhibit spontaneous transitions from a non-SWR state to a SWR state, in which active PV+BCs disinhibit PYR by suppressing anti-SWR cells. SWR events can be triggered by activating PYR or PV+BCs, or inactivating anti-SWR cells. Short-term synaptic depression at the PV+BCs-to-anti-SWR cells connections regulates the termination of SWR events.
The coexistence of states for intermediate values of the depression allows to study the network behavior in terms of bistability. To this end, I consider a mean-field approximation of the spiking network, where conditions for the emergence of a bistable configuration are derived analytically. This allows to unveil the mechanisms regulating the existence of bistable disinhibitory networks.
The model predicts the existence of a class of anti-SWR cells. In the last part of this work, I show the first experimental evidence for CA3 interneurons anti-modulated with respect to SWRs, and discuss their involvement in the SWR generation process.
Overall, the results of this thesis elucidate the role of interneurons in SWR generation and broaden our understanding of the microcircuits supporting the dynamics of memory-related networks.

Régulation des sous-types d’hétérodimères du récepteur GABAB dans la moelle épinière en conditions de douleurs neuropathiques : rôle des protéines partenairesPapon, Marie-Amélie 04 December 2009 (has links)
Dans le système nerveux central, le récepteur inhibiteur GABAB est un archétype des RCPGs hétérodimériques. Il est composé en effet de deux sous-unités, la sous-unité GABAB1 (B1a ou B1b) qui lie l’agoniste et la sous-unité GABAB2 couplée aux protéines G. L’activation de ce récepteur a un effet antinociceptif bien établi concernant les douleurs aiguës mais son effet reste cependant très limité en cas de douleurs neuropathiques. Notre hypothèse est que son activation et sa signalisation peuvent être altérées par des protéines partenaires, aboutissant à des processus de désinhibition dans la moelle épinière en conditions de neuropathie. Nos résultats mettent en évidence le rôle de deux protéines partenaires qui sont surexprimées en conditions douloureuses et qui diminuent l’activation du récepteur GABAB via deux mécanismes différents. D’un part, la protéine cytosolique 14-3-3? induit la dissociation de l’hétérodimère B1b/B2. Cette action a lieu principalement dans les compartiments post-synaptiques. D’autre part, la fibuline-2, protéine de la matrice extracellulaire diminue l’activation de l’hétérodimère B1a/B2. Il s’agit cette fois préférentiellement d’une action dans les compartiments pré-synaptiques. Des stratégies anti-sens (siRNA anti-14-3-3? ou anti-fibuline-2) ou des peptides de compétition sélectifs de l’interaction B1b/14-3-3? permettent de potentialiser les effets antinociceptifs d’un agoniste du récepteur sur un modèle animal de neuropathie. L’ensemble de ces résultats suggèrent que l’état d’oligomérisation des RCPGs peut être modulé in vivo par des protéines partenaires endogènes impliquées dans le développement ou le maintien d’états pathologiques de sensibilisation à la douleur. / In the central nervous system, the inhibitory GABAB receptor is an obligate heterodimeric GPCR that requires the association between GABAB1 (B1a or B1b) and GABAB2 subunits. The heterodimeric GABAB receptor activation has a well-known antinociceptive action in acute pain but its effect appears limited in pathological states. Our hypothesis is that the GABAB activation and signaling could be altered by partner proteins, thus resulting in desinhibition processes in the spinal cord. In the present study, we investigated the role of two partner proteins overexpressed in neuropathic states which decrease GABAB activation through two different mechanisms. On the one hand, the cytosolic 14-3-3? protein induces the dissociation of the heterodimer B1b/B2. This effect occurs in post-synaptic compartments. On the other hand, fibulin-2, an extracellular matrix protein, which decreases the activation of the heterodimer B1a/B2 localized preferentially in presynaptic compartments. Anti-sens strategies (anti-14-3-3? or anti-fibulin-2 siRNA) or competing peptides specific of 14-3-3?/B1b interaction, potentiate the antinociceptive effects of GABAB agonist in an animal model of neuropathic pain. Taken together, our data suggest that GPCR oligomeric state can be modulated in vivo by endogenous partners proteins that are involved in the development and the maintenance of pain sensitization.

Individual differences in eating behaviours and their relationship with motivation, cognition and weight controlDavies, Kirsty Mary January 2018 (has links)
A considerable percentage of the UK population are overweight (BMI≥25kg/m2) or obese (BMI≥30kg/m2). However, despite living in the same culture and exposed to a similar “obesogenic” environment, some individuals gain weight while others do not (French et al., 1995). This variability in weight control has been suggested to be associated with individual differences in eating behaviours (French et al., 2012). Certain factors, such as motivation (hedonic hunger and hunger status) as well as cognition (impulsivity and memory) may have an impact on eating behaviours and their relationship with weight control. Thus, the objective of this thesis was to explore individual differences in eating behaviours and investigate their relationship with motivation, cognition and weight control. The first experiment (Chapter 2) investigated the relationship between eating behaviours, motivation (hedonic hunger) and food consumption during an ad-libitum buffet. This study suggests that restrained eating behaviour was associated with higher overall energy intake, greater energy intake from unhealthy foods and greater energy intake from both high and low energy dense foods. However, no interactions between restraint and disinhibition or hedonic hunger was seen. Following this, the second experiment (Chapter 3) examined whether eating behaviours, such as disinhibition, restraint and hunger, change during a weight loss and weight maintenance period and whether they could predict changes in weight during these periods. Indeed, the results suggest that lower baseline restraint could predict greater weight loss during a low-energy liquid diet and interventions which increase restraint and decrease disinhibition may be beneficial for longer term weight maintenance. The third experiment (Chapter 4) was designed to investigate whether motivation and cognition influences eating behaviours. The results suggest that hedonic hunger, restraint and impulsivity may lead to higher levels of disinhibited eating behaviour. This study was also able to replicate the findings of previous literature suggesting that episodic memory is negatively associated with BMI (Cheke et al., 2016). Finally, following on from the previous study results, the fourth experiment (Chapter 5) included a more diverse sample of participants including dieters. The results provide evidence that individuals on a diet have poorer episodic memory ability than those currently not on a diet. This study also extended previous results suggesting that hedonic hunger (but also episodic memory and hunger) are important factors in disinhibited eating. Hedonic hunger was also shown to be important in levels of hunger.
