In the light of this scope, this work was elaborated and articulated in three chapters whose purpose was to point out the origins of the controversy that involves the concept of thing itself, the main objectors and their respective objections to the thing itself. Given these questions to the thing itself, we’ve approached Allison's proposal to defend a non-polemical use of the concept of thing itself, from the understanding that the distinction phenomena and thing itself are two ways of considering the same thing. Next, it deals with the construction of the concept of thing itself within the Critique of Pure Reason, addressing the implications of texts of the Preface, Transcendental Aesthetics, Analytic Concepts and Analytic of the Principles for the consolidation of the meaning of the thing considered in itself it. In this sense, the differences between the thing itself, phenomenon, transcendental object and noumenon, and especially in what sense some of these concepts can be used as equivalents in the light of the Critique of Pure Reason, stand out. Finally, the importance of the concept of thing itself in Transcendental Idealism was analyzed, since the criticisms were incisive, but Kant remained with the concept of thing itself in his Theory of Knowledge. Thus, the task of the concept of thing itself in Transcendental Idealism was observed, which led to the conclusion that the thing considered in itself, in spite of the controversies surrounding it, is essential for the success of Kant's critical program, named as Transcendental Idealism whose thesis is the access to the objects of the senses, since these depend on the pure forms of sensibility and the impossibility of the cognoscent subject to scan things considered in themselves, since their existence does not depend on the human faculty of knowledge / Essa dissertação tem como objetivo estudar o conceito de coisa em si na Crítica da Razão Pura e as origens da polêmica que o envolve. À luz desse escopo, o trabalho foi elaborado e articulado em três capítulos, cujo propósito foi de apontar as origens da polêmica que envolve o conceito de coisa em si, os principais objetores e seus respectivos questionamentos à coisa em si. Dado estes questionamentos à coisa em si, abordou-se a proposta de Allison em defender um uso não polêmico do conceito de coisa em si, a partir da compreensão que a distinção fenômeno e coisa em si são dois modos de considerar a mesma coisa. Em seguida, trata-se da construção do conceito de coisa em si no interior da Crítica da Razão Pura, abordando as implicações de textos do “Prefácio”; “Estética Transcendental”; “Analítica dos Conceitos” e “Analítica dos Princípios” para a consolidação do significado da coisa considerada em si mesma. Nesse sentido, destacam-se as diferenças entre coisa em si, fenômeno, objeto transcendental e númeno e, especialmente, em que sentido alguns desses conceitos podem ser utilizados como equivalentes à luz da Crítica da Razão Pura. Por fim, analisou-se a importância do conceito de coisa em si no Idealismo Transcendental, visto que as críticas foram incisivas, mas Kant permaneceu com esse conceito em sua Teoria do Conhecimento. Sendo assim, observou-se a tarefa do conceito de coisa em si no Idealismo Transcendental o que conduziu à conclusão de que a coisa considerada em si mesma, a despeito das polêmicas que a envolve, é imprescindível para o êxito do programa crítico de Kant, nomeado como Idealismo Transcendental cuja tese é o acesso aos objetos dos sentidos, pois estes dependem das formas puras da sensibilidade e a impossibilidade do sujeito cognoscente esquadrinhar as coisas consideradas em si mesmas, porque o modo de existir dessas não dependem da faculdade humana de conhecimento

The research has the theoretical-methodological support of Systemic-Functional Linguistics (SFL), which involves the notion of evaluation (MARTIN, 2000, 2003), the ethical, aesthetic and emotional evaluation – in the construction of a "textual world" that contributes to the context construction. Scholars, who deal with language and text (although the fact has been clear to rhetoric practitioners for millennia), have accepted it, that the implicit presentation of content may facilitate persuasion of the audience. Language works in social processes, actively constructing context, and, at the same time, it is its expression (LUKIN, 2013). In this process, meanings are influenced by the cultural context – genre – and by the situational context – register – in which they are exchanged (HALLIDAY, 1994, 2004). Language is a "way of action" and not just an "expression of thought" as it relates to the relationships between text, context and their extensions (MALINOWSKI, 1935). Relating to these questions, research also has the support of Critical Linguistics (FOWLER, 1991), for which any aspect of the linguistic structure carries ideological significance. For sociologists, journalistic news is a commodity in the business process of providing forms of consciousness. The research, of a critical nature, answers the following questions: (a) What "context-building" work does the editorial genre examined do? (B) What role does the construction of context play in the persuasive process that runs through the editorial? The results show the construction of a context that is based on the knowledge of the reader, present in its frame, in a process of intersubjectivity, which makes it complicit in the intentions of the newspaper. Thus, the editorial constructs a discourse that is coherent in this "textual world" (DOWNING 2003), although unilateral in the underlay of the text, made by the news provider in line with ideas of controlling groups in an industrial-capitalist society, because the newspaper is an industry with its own commercial interests / O objetivo desta dissertação de mestrado é o exame crítico da função da língua na construção de contexto no processo persuasivo presente no editorial intitulado “O Quinto Centenário”, publicado no jornal Folha de S.Paulo, em 24.04.2000. A pesquisa tem o apoio teórico-metodológico da Linguística Sistêmico-Funcional (LSF), que envolve a noção de avaliatividade (MARTIN, 2000, 2003), a avaliação ética, estética e de emoções – na construção de um “mundo textual” que contribui na construção de contexto. Tem sido aceito por estudiosos que tratam da língua e do texto (embora o fato já estivesse claro para os profissionais da retórica, por milênios) que a apresentação implícita de conteúdo pode facilitar a persuasão da audiência. A língua trabalha nos processos sociais, construindo ativamente o contexto, ao mesmo tempo em que ela é também sua expressão (LUKIN, 2013) e, nesse processo, os significados são influenciados pelo contexto cultural – gênero – e pelo contexto situacional – registro – em que são intercambiados (HALLIDAY, 1994, 2004). A língua é um modo de ação e não apenas uma expressão do pensamento no que se refere às relações entre texto, contexto e suas extensões (MALINOWSKI, 1935). Nessas questões, a pesquisa conta também com o apoio da Linguística Crítica (FOWLER, 1991), para a qual qualquer aspecto da estrutura linguística carrega significação ideológica. Para os sociólogos, o noticiário jornalístico é uma mercadoria no processo empresarial de fornecimento de formas de consciência. A pesquisa, de natureza crítica, responde às seguintes perguntas: (a) Que trabalho de “construção de contexto” faz o gênero editorial examinado? (b) Que papel exerce a construção de contexto no processo persuasivo que percorre o editorial? Os resultados mostram a construção de um contexto que se apoia no conhecimento do leitor, presente em seu frame, num processo de intersubjetividade, que o faz cúmplice das intenções do jornal. However, new analysis and methodologies have been emerging, especially with the advent of standard statements based on the International Accounting Standard (IAS), providing reliability and quality for researching. This study aims to analyze how companies listed on the B3 financed the working capital and identify the impacts generated on company´s profitability and gains between 2010 and 2016. The dynamic model also known as the Fleuriet’s Model, proposes a different view of analysis of these statements, mainly by using a new type of accounting groups for working capital. This model improved the quality of analysis regarding the financing of working capital, the liquidity situation of companies and the possibility to identify financial risks. The research also identified the scissors effect, which is a characteristic situation of companies facing financial risks. Through statistic and econometric models, it was identified the interference of working capital in the profitability and gains of these companies. The sectorial analysis demonstrated how companies reacted differently regarding the investment in working capital within the period / O objetivo principal dos demonstrativos financeiros é fornecer informações sobre a situação patrimonial e financeira da empresa num determinado período, permitindo que gestores, acionistas e conselheiros tomem decisões que garantam a valorização da empresa e sua perpetuidade. Novas metodologias de análise vêm surgindo ao longo do tempo, principalmente com o advento da padronização desses demonstrativos com base no IAS – International Accounting Standard, proporcionando também maior confiabilidade aos trabalhos de pesquisa. Este estudo visa analisar o comportamento de empresas brasileiras listadas na Bolsa de Valores de São Paulo (B3) quanto à forma de financiamento do capital de giro e os impactos gerados na lucratividade e rentabilidade dessas empresas no período compreendido entre 2010 e 2016. Optou-se por utilizar o modelo dinâmico, também conhecido como modelo Fleuriet, o qual propõe uma visão diferenciada de análise desses demonstrativos, principalmente por utilizar uma nova forma de agrupamento das contas circulantes que permitem maior visibilidade quanto à origem do financiamento do capital de giro, à liquidez das empresas e à identificação de eminentes riscos financeiros. Identificou-se o efeito tesoura, característico em empresas em situação de risco financeiro e por meio de cálculos estatísticos e econométricos, constatou-se a interferência do capital de giro na lucratividade e rentabilidade das empresas. A Culturally and Linguistically Responsive Framework for Improving Academic and Postsecondary Outcomes of Students with Moderate or Severe Intellectual Disability

The needs of culturally and linguistically diverse (CLD) students with moderate or severe intellectual disability (ID) are quite unique and complex. CLD students with moderate or severe ID face many of the same issues as their non-disabled CLD peers; however, due to the nature of their disability this may lead to even less access to the general curriculum, appropriate services, materials, and meaningful collaboration between families and educators. The purpose of this article is to provide a culturally responsive framework for facilitating academic instruction for CLD students with moderate or severe ID that also includes appropriate supports in an effort to increase access to postsecondary outcomes for this population. Suggestions for accessing the general curriculum and a discussion about increasing parental involvement and accessing appropriate adult agencies to further enhance these outcomes are provided.

Novel Regulators of the TGF-β Signaling Pathway

<p>The transforming growth factor-β (TGF-β) superfamily consists of related multifunctional cytokines, which include TGF-βs, activins, and bone morphogenetic proteins (BMPs) and coordinate several biological responses in diverse cell types. The biological activity of TGF-β members is executed by transmembrane serine/threonine kinase receptors and intracellular Smad proteins. The effects of TGF-β on the epithelium are of high interest. Carcinomas (tumors of epithelial origin) are the most common type of human cancer and frequently exhibit aberrant responses to TGF-β. Therefore, TGF-β can be defined as tumor suppressor as it inhibits growth of normal epithelial cells. However, TGF-β also induces an epithelial-mesenchymal transition (EMT), a key component of metastasis, and thus promotes cancer spread.</p><p>The scope of this thesis is the mechanism of TGF-β signaling in epithelial cells. We established that only TGF-β, but not BMP pathways can elicit EMT. Moreover, we found that Smad signaling is critical for regulation of EMT. In a transcriptomic analysis, we identified a large group of novel genes, whose regulation is pivotal for TGF-β-induced EMT and metastasis. We focused on two of such genes, <i>Id2</i> and <i>Id3</i>. Interestingly, we found that TGF-β-induced repression of <i>Ids</i> is necessary for inducing EMT and potent cell cycle arrest. BMP increases expression of <i>Ids</i> and therefore it cannot induce the same biological responses as TGF-β. Hence, knock-down of endogenous Id2 and Id3 proteins sensitized epithelial cell to BMP-7. We proposed a model, in which Id2 and Id3 are important components controlling concerted regulation of cell proliferation and EMT downstream of TGF-β pathways.</p><p>Furthermore, we identified a serine/threonine kinase, <i>SNF1LK</i>, whose mRNA is rapidly induced by TGF-β in epithelial cells. We found that SNF1LK is a negative regulator of the TGF-β pathway and it promotes TGF-β receptor turnover. Subsequently, we demonstrated that SNF1LK together with Smad7 and Smurf2 targets TGF-β receptor for ubiquitin-dependent degradation. Furthermore, SNF1LK interacts with proteasomes, suggesting that SNF1LK serves as bridge between ubiquitinated receptors and proteasomes, helping proteasomes to recognize the ubiquitinated cargo destined for degradation. We therefore established a novel negative feedback regulatory mechanism of TGF-β signaling. </p>

Authentification dans les réseaux maillés sans fils avec la cryptographie basée sur l'identité

De nos jours, l'authentification dans les réseaux maillés sans fils fait appel aux certificats ou aux secrets partagés. Dans les environnements sans fils, la gestion des certificats est désavantageuse. En effet, les certificats nécessitent le déploiement d'une infrastructure à clés publiques (ICP) et la définition d'une autorité de certification (AC). La AC définit toute une politique qui permet de contrôler la génération, la transmission et la révocation des certificats. Cette politique ne prend pas en considération les limites en termes de puissance et de mémoire que peuvent avoir les stations des clients dans un réseau maillé. Afin de ne pas utiliser les certificats et ne pas déployer une ICP, nous avons étudié dans cette thèse les utilisations possibles de la cryptographie basée sur l'identité (CBI) pour la définition de nouveaux schémas d'authentification pour les réseaux maillés sans fils. La CBI propose de dériver, directement, la clé publique d'une station à partir de son identité. Par conséquent, nous n'avons plus besoin de passer par des certificats pour associer l'identité de la station à sa paire de clés (publique et privée). Par contre, la CBI définit un générateur de clé privée (GCP) qui gère le calcul des clés privées des différentes stations sur le réseau. Par conséquent, ce GCP est capable de réaliser une attaque d'usurpation d'identité (escroc de clés) à l'encontre de toutes les stations légitimes. Pour diminuer le risque de cette attaque, les chercheurs ont tendance à supposer que le GCP est digne de confiance. Dans cette thèse, nous présentons tout d'abord un protocole d'authentification basée sur l'utilisation conjointe d'un mot de passe et de la CBI. En effet, nous proposons d'utiliser le serveur d'authentification de notre réseau maillé comme GCP. Ensuite, nous étudions une liste de mécanismes qui permettent de contrer l'attaque de l'escroc qui caractérise le GCP.

Epigenetic Regulation of Replication Timing and Signal Transduction

Upon fertilization the paternal and maternal genomes unite, giving rise to the embryo, with its unique genetic code. All cells in the human body are derived from the fertilized ovum: hence they all contain (with a few exceptions) the same genetic composition. However, by selective processes, genes are turned on and off in an adaptable, and cell type-specific, manner. The aim of this thesis is to investigate how signals coming from outside the cell and epigenetic factors residing in the cell nucleus, cooperate to control gene expression. The transforming growth factor-β (TGF-β) superfamily consists of around 30 cytokines, which are essential for accurate gene regulation during embryonic development and adult life. Among these are the ligands TGF-β1 and bone morphogenetic (BMP) -7, which interact with diverse plasma membrane receptors, but signal via partly the same Smad proteins. Smad4 is essential to achieve TGF-β-dependent responses. We observed that by regulating transcription factors such as Id2 and Id3 in a specific manner, TGF-β1 and BMP-7 achieve distinct physiological responses. Moreover, we demonstrate that CTCF, an insulator protein regulating higher order chromatin conformation, is able to direct transcription by recruiting RNA polymerase II to its target sites. This is the first mechanistic explanation of how an insulator protein can direct transcription, and reveals a link between epigenetic modifications and classical regulators of transcription. We also detected that DNA loci occupied by CTCF replicate late. The timing of replication is a crucial determinant of gene activity. Genes replicating early tend to be active, whereas genes replicating late often are silenced. Thus, CTCF can regulate transcription at several levels. Finally, we detected a substantial cross-talk between CTCF and TGF-β signaling. This is the first time that a direct interplay between a signal transduction pathway and the chromatin insulator CTCF is demonstrated.

From Physicochemical Features to Interdependency Networks : A Monte Carlo Approach to Modeling HIV-1 Resistome and Post-translational Modifications

The availability of new technologies supplied life scientists with large amounts of experimental data. The data sets are large not only in terms of the number of observations, but also in terms of the number of recorded features. One of the aims of modeling is to explain a given phenomenon in possibly the simplest way, hence the need for selection of suitable features. We extended a Monte Carlo-based approach to selecting statistically significant features with discovery of feature interdependencies and used it in modeling sequence-function relationships in proteins. Our approach led to compact and easy-to-interpret predictive models. First, we represented protein sequences in terms of their physicochemical properties. This was followed by our feature selection and discovery of feature interdependencies. Finally, predictive models based on e.g., decision trees or rough sets were constructed. We applied the method to model two important biological problems: 1) HIV-1 resistance to reverse transcriptase-targeted drugs and 2) post-translational modifications of proteins. In the case of HIV resistance, we were not only able to predict whether the mutated protein is resistant to a drug or not, but we also suggested some new, previously neglected, mutations that possibly contribute to drug resistance. For all these mutations we proposed probable molecular mechanisms of action using literature and 3D structure studies. In the case of predicting PTMs, we built high accuracy models of modifications. In comparison to other methods, we were able to resolve whether the closest neighborhood of a residue (the nanomer) is sufficient to determine its modification status. Importantly, the application of our method yields networks of interdependent physicochemical properties of amino acids that show how these properties collaborate in establishing a given modification. We believe that the presented methods will help researchers to analyze a large class of important biological problems and will guide them in their research.

Réalisation et optimisation d'une électronique intégrée basse consommation pour la mesure de gaz polluants.

Afin de réaliser un appareil innovant pour la mesure de gaz polluants, la société SVS@CAP s'est associée avec le laboratoire de physique corpusculaire en 2009 pour la création du projet EREBUS. Ce projet a pour but la réalisation d'un ensemble de dispositifs sans fil permettant d'effectuer une surveillance de la concentration de gaz polluants. L'autonomie et la compacité d'un tel dispositif étant essentielles, la problématique principale porte sur la réduction de la consommation. A partir d'une première étude menée sur les différentes technologies existantes, les capteurs électrochimiques ont été identifiés comme les moins consommateurs d'énergie. Pour chacun des gaz cibles, un modèle électrique du capteur associé a été déterminé. A partir de ces modèles, une architecture dédiée et épurée a pu être déduite. Pour atteindre et même dépasser les objectifs de consommation, les efforts ont aussi été portés sur un dimensionnement avec la méthode gm/id. La réalisation de cette électronique intégrée a permis d'atteindre une consommation de l'ordre du microwatt pour chaque voie de mesure.
