The relationship between corticosterone and personality in red junglefowl

Stress cannot be fully avoided in an animal´s life, and the way that animals handle stress can be crucial for their health. Coping styles in animals are describing consistent sets of behaviour and physiological stress responses. Two forms of coping are typically described; a more proactive type that is more bold, active, and aggressive, and, in contrast, a less active, more shy reactive type. Coping styles can be used to learn how animals react to stress, and thus improve our handling of stress in captive animals. To investigate the existence of the reactive-proactive coping styles in the red junglefowl (Gallus gallus), both male and female red junglefowl were exposed to behavioural assays measuring boldness, activity, exploration, and aggression. These behavioural measures were compared to blood plasma concentrations of the stress hormone corticosterone, hence exploring the link between behaviour and physiological response to stress. I show positive correlations between boldness, activity, and exploration measured in a novel arena test, and boldness and activity measured in a novel object test. This somewhat confirms the inter-related behavioural aspect of reactive-proactive coping styles. Further, some of these behaviours correlated with post-stress concentrations of corticosterone, which somewhat links behaviour to the physiological aspect of coping styles. This study tentatively supports the reactive-proactive coping styles by showing a link between behaviours and the physiological stress response in the red junglefowl, with some similarities to what have been observed in other species across taxa.

Significant increase in liver and heart mass found in post hatching red junglefowls (Gallus gallus).

Significant heart and liver increase in hatching and neonates has been found evident in multiple different species such as the emu, Peking Duck, and pigs. The relative mechanism behind heart growth could be tied to closing of the ductus arteriosus, however there is still a debate whether it’s significant impact in avian species. Liver and heart mass were measured at four different transition stages before and after hatching on Red junglefowls (Gallus gallus). Heart mass was found to vary between 83-170mg, with the lowest values most often found in pre pipped chickens while higher values found more often in hatchlings. The relative heart mass was found to be significant (P<0,05) across all groups except between internally pipped and externally pipped chickens. Therefore, the results can conclude that an increase in relative heart mass was found to be directly tied to age. Liver mass was found to be significant (P<0,05) but changes in mass was found to occur only between EP and hatchling stage. Further analysis on absolute heart and liver mass showed in both cases hatchlings having a higher mean mass compared to the other three stages. Both absolute liver and heart mass was found to be statistically significant (P<0,05) which indicates there are no major differences between analysis on relative or absolute mass. These findings suggest that red junglefowls follow the same growth pattern found in other precocial birds such as emu or Peking duck.

Genomics-Based Analysis of Antibody Response to Sheep Red Blood Cells in Chickens

Immune response provides vertebrates an important mechanism to fight pathogens and to reduce the incidence of diseases. Defining the molecular basis of antibody response may facilitate genetic improvement in the immune response of animals to pathogens. For almost 4 decades, antibody titers in response to challenge by sheep red blood cells (anti-SRBC) have provided an investigative tool in the efforts to define molecular mechanisms that underlie vertebrate immune response. The overall objective of this dissertation research was to identify DNA markers associated with anti-SRBC response in chickens. Specific objectives were: to develop a resource population for QTL analysis for anti-SRBC, to identify DNA markers and genes associated with primary anti-SRBC, and to evaluate the allelic frequencies in non-selected chicken populations of candidate markers associated with either high or low anti-SRBC response. These objectives tested the hypothesis that genetic control of a chicken's response to SRBC is polygenic. The resource population developed consisted of F1, backcross, and F2 derived from reciprocal crosses of birds from parental lines in the 28th generation of divergent selection for low (L) and high (H) anti-SRBC. The mean anti-SRBC titers of the parental lines were significantly different, with 11.5 for H and 2.6 for L (P<0.05). That for the 4 groups of F2 progeny ranged from 6.3 to 7.5, while those of the 8 groups of backcross progeny ranged from 3.9 to 13.3. Four of 555 random primers used to screen the parental H and L anti-SRBC lines were informative by amplifying seven line-specific fragments (P<0.0025). Each of the 7 line-specific fragments was converted to a sequence characterized amplified region (SCAR) within which single nucleotide polymorphisms (SNPs) were identified and tested for association with anti-SRBC. Only two of the seven SCARs in the parental lines were associated (P<0.05) with anti-SRBC level in the backcross resource population. Additionally, from analysis of the parental L and H anti-SRBC lines using microarrays, a total of 57 line-specific SNPs were also identified. Twenty of the line-specific SNPs were in and/or near genes previously reported to have immunity-related function. Microarray-based gene expression profiling of pooled RNA samples from L and H anti-SRBC birds identified three differentially expressed genes. In summary, this dissertation describes resources that include candidate SCARs and SNPs as well as differentially expressed genes that may be useful for the identification of genes that underlie antibody response. / Ph. Etude de la réponse immunitaire innée induite par les virus de la grippe aviaire dans les cellules épithéliales pulmonaires et les cellules endothéliales de poulets / Study of innate immune response induced by avian influenza viruses in chicken lung epithelial cells and chicken endothelial cells

Les virus influenza aviaires faiblement pathogènes (IAFP) ciblent principalement les épithéliums des voies respiratoires et intestinales chez les poulets (Gallus gallus) infectés. Cependant, les virus influenza aviaires hautement pathogènes (IAHP) mènent à une maladie systémique fatale avec une localisation particulière aux endothéliums. L’objectif de cette thèse a été d’explorer les relations entre la réplication des virus influenza aviaires (IA) et la réponse antivirale de l’hôte dans deux modèles cellulaires originaux obtenus chez le poulet : des cellules épithéliales pulmonaires (CLEC213) et des cellules endothéliales d’aortes (chAEC). Les résultats clés sont les suivants : (i) la réplication productive des virus IA dans les chAEC dépend du clivage de l’hémagglutinine et de l’échappement viral à la réponse immunitaire innée ; (ii) les CLEC213 sont très permissives aux virus IA et présentent une faible réponse antivirale médiée par la signalisation TLR3 et MDA5 ; (iii) les fonctions régulatrices de SOCS1 et SOCS3, sur le signal des interférons et des cytokines, sont conservées chez le poulet. Nous proposons que certains virus IA peuvent exploiter les fonctions pro-virales de SOCS1 et SOCS3 à leur avantage de manière spécifique au type cellulaire. / Low pathogenic avian influenza (LPAI) viruses essentially target the epithelia of the respiratory and intestinal tract in the infected chicken host (Gallus gallus). However, highly pathogenic avian influenza (HPAI) viruses induce a peracute fatal systemic disease and exhibit a striking endothelial cell tropism. The objective of the present thesis was to explore the interdependencies of AI virus replication and the antiviral host response in two novel avian cell culture models: chicken lung epithelial cells (CLEC213) and chicken aortic endothelial cells (chAEC). The salient findings from this study are that (i) productive AI virus replication in chAEC is dependent on hemagglutinin cleavability and appears to be related to innate immune escape; (ii) CLEC213 are highly permissive to AI virus infection, due to a cell type-specific diminished TLR3- and/or MDA5-mediated antiviral signaling response; (iii) the interferon and cytokine regulatory functions of SOCS1 and SOCS3 are conserved in the chicken. Based on our data, we propose a model that predicts that certain AI viruses may exploit the proviral functions of SOCS1 and SOCS3 in a cell type-specific manner.

Uma população F2 foi desenvolvida através do cruzamento entre uma linhagem de frangos de corte (TT) e uma de postura (CC). Sete machos TT foram acasalados com sete fêmeas CC para produzir uma população F1 TC. Cada macho F1 foi acasalado com três fêmeas F1 não-aparentadas para produzir aproximadamente 100 descendentes por família de irmãos completos. Neste estudo foram utilizadas as 5 famílias que apresentaram maior número de marcadores informativos nos cromossomos 1 a 5, em estudos realizados anteriormente nesta população. Foram coletados dados fenotípicos de peso ao nascer (PN), peso aos 35 (PV35) e 41 (PV41) dias de idade, ganho de peso (GP35-41), consumo de ração (CR35-41) e eficiência alimentar (EF35-41) dos 35 aos 41 dias de idade. Os indivíduos parentais e F1 foram genotipados com 35 marcadores microssatélites posicionados nos cromossomos 9, 10, 12, 14, 15, 16, 17 e 18. Os marcadores informativos (22) foram genotipados nos indivíduos F2 das famílias escolhidas. O cromossomo 16 não foi mapeado, pois o único marcador disponível não foi informativo. No cromossomo 17 apenas um marcador foi informativo, logo foi utilizada uma análise de marca simples para associação do marcador com as características selecionadas. O mapa de ligação de cada cromossomo foi construído e comparado ao mapa consenso. Os mapas de ligação apresentaram o mesmo ordenamento dos marcadores em relação aos mapas consenso, no entanto foram encontradas pequenas discrepâncias nas distâncias entre os marcadores A análise empregada no estudo de mapeamento de QTL por intervalo utilizou o método de regressão linear e estimação de quadrados-mínimos, utilizando o programa QTL Express, na opção análise de F2. No cromossomo 17, duas características foram associadas ao genótipo do marcador: GP35-41 (P<0,05) e EF35- 41 (P<0,01). Foram encontrados 4 QTL sugestivos no cromossomo 10: para PV35, PV41, GP35-41 e EF35-41. Houve interação do QTL com família para EF35-41. Os QTLs para PV35, PV41 e GP35-41 apresentaram efeito aditivo, apenas EF35-41 apresentou efeito de dominância. / An F2 chicken population was developed by crossing broiler line (TT) with a layer line (CC). Seven males TT were mated with seven females CC to generate an F1 population. Each male F1 was then mated with three unrelated females F1 generating approximately 100 individual per family of dam. On the present study 5 families were used, which showed the greatest number of informative markers in previews studies of chromosomes 1 to 5 of the resource population. Phenotypic dada were collected for body weight at 1 (BW1), 35 (BW35) and 41 (BW41) days of age, weight gain (WG35- 41), feed intake (FI35-41) and feed efficiency (FE35-41) from 35 to 41 days of age. The parental and F1 individuals were genotyped for 35 microsatellite markers distributed on chromosomes 9, 10, 12, 14, 15, 16, 17 and 18. The informative markers were genotyped on the F2 from the selected families. Chromosome 16 was excluded, because the only available marker was not informative. On chromosome 17 only one marker was informative, therefore a single marker analysis was used to associate the marker with the selected characteristics. The linkage map of each chromosome was constructed and compared to the consensus map. The linkage maps showed the same marker ordering compared to the consensus map, however small discrepancy between markers distances were presented. Interval mapping using regression methods was applied to a line-cross and least square analysis, using the QTL Express program, on the F2 analyses option. On chromosome 17, two characteristics were associated to the marker genotype: WG35-41 (P<0.05) and FE35-41 (P<0.01). Suggestive QTLs were detected for BW35, BW41, WG35-41 and FE35-41 on chromosome 10. A QTL x family interaction effect was statistically significant for FE35-41. QTLs for BW35, BW41 and WG35-41 showed addictive effects, only FE35-41 showed dominance effect.

Influência do estresse pré-abate na expressão gênica e qualidade da carne de frango (Gallus gallus) / Influence of stress on gene expression and broiler meat quality

A produção de carne de frango no Brasil é um importante setor do agronegócio. O setor passou por décadas de aprimoramentos, culminando desde 2004, no mais alto posto de exportação de carne de frango e ocupando a terceira maior produção mundial. A expansão, conquista e manutenção de novos mercados está inevitavelmente relacionada com a qualidade da matéria-prima. Entre os fatores que influenciam a qualidade do produto final, encontra-se o estresse no manejo pré abate. O presente estudo objetivou estabelecer relações entre a qualidade da carne de frangos e a expressão gênica sob condições de estresse térmico. Para tal, frangos de corte de linhagens comerciais foram submetidos a estresse térmico e abatidos. A carne oriunda dessas aves teve suas propriedades funcionais mensuradas. Os genes expressos de forma diferenciada para a qualidade da carne foram identificados pelo método de arranjos de alta densidade (macroarranjos) e validados com o emprego de PCR quantitativo em tempo real. Para os parâmetros de qualidade de carne avaliados (pH, cor e capacidade de retenção de água), os animais estressados apresentaram peito mais escuro a 1 hora pós abate (valor de L* de 52,46 contra 55,00; p<0,05), menor valor de pH as 24h pós abate (5,82 contra 5,99; p<0,05) e maior capacidade de retenção de água a 1h pós abate (0,29 contra 0,22; p<0,05). O estudo de expressão gênica por macroarranjos de DNA evidenciou um total de 259 genes diferencialmente expressos entre os tratamentos. 149 desses genes apresentaram indução de expressão (p<0,05) nos animais submetidos ao estresse térmico. Alguns desses genes fazem parte da rede de proteínas responsáveis pelo metabolismo energético celular. Lactato desidrogenase A (LDHA), glicose fosfomutase e piruvato quinase são enzimas que atuam na degradação da glicose e demonstraram aumento da expressão sob condição de estresse térmico (p<0,05). Tal fato sugere que durante o estresse térmico, houve uma maior mobilização do glicogênio muscular, fornecendo maior aporte de glicose na célula muscular, acarretando em aumento da taxa glicolítica nesses animais. Outros genes tidos como induzidos pela condição de estresse térmico foram os genes de choque térmico HSP70 e 90 (p<0,05), evidenciando a atividade protetora de suas proteínas na atividade celular frente a condições de elevação da temperatura. Miostatina, Actina 3 e IGF II, proteínas envolvidas no desenvolvimento da musculatura esquelética se apresentaram igualmente elevadas pelas condições de estresse (p<0,05). 9 genes foram selecionados para validação por PCR quantitativo em tempo real. Miostatina, Lactato desidrogenase A, HSP 70 e HSP 90 foram selecionados com base nos resultados obtidos com as hibridizações. Além destes, outros 5 genes foram escolhidos com base na evidência biológica e do papel na resposta fisiológica ao estresse térmico e atividade glicolítica: Glicogênio fosforilase, citrato sintase e lactato desidrogenase B. Miostatina, HSP 70 e 90 e citrato sintase apresentaram maiores níveis de expressão nos animais estressados (p<0,05). Já Glicogênio fosforilase teve sua expressão inibida com o estresse térmico (p<0,05). Esses achados sugerem importante papel do metabolismo energético durante condições de estresse térmico, além da possível influência que a temperatura exerce no desenvolvimento da musculatura esquelética. / Brazilian broiler production yields a considerable amount of income for the nation. Brazil is currently the world&#39;s third broiler producer and the first chicken meat exporter, contributing with 41% of the whole world market. To reach new market frontiers it is inevitable the improvement of meat quality. Among the reasons that influences final products quality is the preslaughter stress. This study scoped establishing the relationships between meat quality and gene expression under thermal stressful conditions. Commercial broiler lines were submitted to high thermal stress prior to slaughter. Breast meat from these animals had their proprieties evaluated. Differentially expressed genes for stressed or non-stressed birds were investigated by macroarrays and further validated by qRT-PCR. Concerning to the meat attributes, breast meat from stressed birds was darker (L* values of 52,46 against 55,00; p<0,05), with lower pH at 24h postmortem (5,82 against 5,99; p<0,05) and had greater water holding capacity 1h after slaughter (0,29 vs. 0,22; p<0,05). Macroarray analysis revealed 259 differentially expressed genes between stressed and non-stressed control birds. From these, 149 were induced by heat stress (p<0,05). Some of these induced genes play a role in carbohydrate metabolism such as lactate dehydrogenase A (LDHA), glucose phosphomutase and piruvate kinase. Such enzymes acts on degradation of glucose in order to provide energy to the muscle cells which has direct effect upon meat quality processing. These findings suggest great mobilization of muscle glycogen stores, offering opportunity of a not efficient muscle-to-meat conversion. Genes that responds to heat stress damage also were induced with heat exposure. That provides a sort of heat tolerance and ensures the birds&#39; survival under heat conditions. Some muscle-related genes were also induced by heat stress. Myostatin, actin 3 and IGFII showed greater amounts of mRNA under this condition (p<0,05). Among all genes differentially expressed, 9 were picked up for further studies with qRT-PCR. Myostatin, lactate dehydrogenase A and B, HSF1, HSF3, HSP70, HSP90, glycogen phosphorylase and citrate synthase were studied and only myostatin, HSP 70 and 90 confirmed the differentially expressed patterns between treatments (p<0,05). Unlikely, glycogen phosphorylase appeared repressed in heat stressed animals (p<0,05). These findings suggest an important role of energetic metabolism of birds at hot environments.

Vaccination as a form of prevention to this disease, has not been effective, due to the great genetic variability found. As in the world, in Brazil there are already several studies characterizing the genetic variability found in this virus. In Rio de Janeiro, poultry farming suffers from the disease, but there is no recent study to analyze the genetic variability of the strains found, as a way of supporting epidemiological studies that may help in the future, in the choice of Protocols. The objective of this study was to characterize the S1 gene of the Chicken Infectious Bronchitis virus (vBIG) in chickens farms in the southern region of Rio de Janeiro, as well as to evaluate the genetic variability of the S1 gene in a BIG outbreak on a farm Of laying hens of the northern region of. Tissue samples were collected from animals presenting BIG-compatible clinical signs, such as weight loss, respiratory problems and low productivity. Samples of collected tissues were stored in solution of RNA Later and formalin 10% buffered for molecular and histopathological diagnoses, respectively. In order to perform the molecular analyzes, RNA extraction techniques, as well as RT-PCR (Reverse Transcriptase, Polymerase Chain Reaction), real-time PCR (RT-qPCR), Nested PCR (RT-nPCR), and Sequencing of some samples collected. For the histopathological analyzes, cleavage and preparation of microscopy slides were performed. The results obtained from the sequencing were compared to the sequence of the Ma5 vaccine strain as reference strain, in addition to other sequences deposited on GenBank, exhibiting a variable identity percentage of 72.41% to 100%. Among the samples from each farm, there was variability, even within the same bird. All sequences analyzed were described as genotype 1, belonging to strains 1 and 11; In the comparison of amino acids, there were changes in the hypervariable regions of the virus in the sequences classified as BR-1, being able to explain the low cross-protection that have been occurring in Brazilian plants. These results demonstrated the importance of virus identification, since it becomes an important tool in epidemiological surveillance and in the elaboration of efficient vaccine protocols. / O v?rus da Bronquite Infecciosa das Galinhas (vBIG) ? um agente viral altamente contagioso entre a esp?cie Gallus gallus, sendo considerado uma das patologias que mais acomete a avicultura mundial. No Rio de Janeiro, a cria??o av?cola sofre com a doen?a, por?m ainda n?o existe um estudo recente com o objetivo de analisar a variabilidade gen?tica das cepas encontradas, como uma forma de apoio a estudos epidemiol?gicos, que possam auxiliar no futuro, na escolha de protocolos vacinais mais adequados. Este estudo teve como objetivo realizar a caracteriza??o molecular do gene S1 do v?rus da Bronquite Infecciosa das Galinhas (vBIG) em granjas de frangos de corte na regi?o Sul fluminense, como tamb?m avaliar a variabilidade gen?tica do gene S1 em um surto de BIG em uma granja de poedeiras da regi?o Norte fluminense. Foram realizadas coletas de tecidos dos animais que apresentavam sinais cl?nicos compat?veis com BIG, como perda de peso, problemas respirat?rios e baixa produtividade. Amostras de tecidos coletados foram armazenadas em solu??o de RNA Later e formalina 10% tamponada para os diagn?sticos molecular e histopatol?gico, respectivamente. Para realiza??o das an?lises moleculares foram utilizadas t?cnicas de extra??o de RNA, bem como RT-PCR (?Reverse Transcriptase, Polymerase Chain Reaction?), PCR em tempo real (RT-qPCR), ?Nested? PCR (RT-nPCR), e sequenciamento de algumas amostras coletadas. Para as an?lises histopatol?gicas foram realizadas clivagem e elabora??o de l?minas de microscopia. Os resultados obtidos do sequenciamento foram comparados a sequ?ncia da cepa vacinal Ma5, como cepa refer?ncia, al?m de outras sequencias depositadas no GenBank, apresentando um percentual de identidade vari?vel de 72,41% a 100%. Dentre as amostras de cada granja, houve variabilidade, at? mesmo dentro de uma mesma ave. Todas as sequencias analisadas foram descritas como gen?tipo 1, pertencentes as linhagens 1 e 11; Na compara??o de amino?cidos, houve mudan?as nas regi?es hipervari?veis do v?rus nas sequ?ncias classificadas como BR-1, podendo explicar a baixa prote??o-cruzada que v?m ocorrendo nos plant?is brasileiro. Diversité génétique structurale et fonctionnelle du CMH chez le Poulet : Implication pour la résistance aux maladies

Le complexe majeur d'histocompatibilité (CMH) est une région génomique complexe des Vertébrés, encore imparfaitement connue chez le poulet, qui présente à certains locus une très grande variabilité génétique et qui joue un rôle central dans l'organisation de la réponse immunitaire d'un animal aux pathologies infectieuses. Le CMH est également une région de choix pour étudier le déterminisme génétique de l'adaptation aux agents pathogènes dans un contexte évolutif. De plus une meilleure connaissance du déterminisme génétique de la réponse immunitaire contre les agents pathogènes est un atout important pour développer une stratégie globale de lutte contre les maladies infectieuses. Nous avons donc entrepris d'utiliser les récents outils de la génomique, notamment des marqueurs génétiques de type SNP (Single Nucleotide Polymorphism) afin de caractériser la région B du CMH du poulet. En premier lieu, la diversité génétique a été évaluée dans plus de 80 races ou populations en utilisant le marqueur LEI0258. Ensuite, afin de couvrir l'ensemble de la région à l'aide de marqueurs SNPs, nous avons choisi d'identifier ces polymorphismes par reséquençage de 11 gènes d'intérêt et par comparaison des séquences obtenues entre elles et avec la séquence du génome et les séquences de référence disponibles dans les bases de données. En parallèle, ces travaux ont également permis d'améliorer la connaissance de certains gènes, notamment trois gènes de classe II non classiques de type DM. Effect of daytime and age on the avoidance and approach behaviour of commercial Danish broiler chicken

As activity levels of intensively managed broiler chicken vary over time, detailed knowledge on their influence is potentially useful to further adjust welfare assessment schemes. Therefore, this study investigated the influence of daytime and age on the performance of broiler chicken in two applied fear tests.On-farm studies were carried out in 14 flocks of intensively managed broiler chicken. A forced and a voluntary approach test were conducted during morning and evening hours and at three different ages (1: 6-12d; 2: 21-24d; 3: prior to slaughter). At each observation, avoidance distances (AD) and the number of animals voluntarily approaching (VA) an observer were collected.No significant correlation was found between daytime and AD or VA. When tested at different ages, daytime and VA were significantly correlated (1: r= 0.56*; 2: r= 0.40*; 3: r= 0.64*), but the results were not consistent. At three weeks the VA was found to be higher in the morning but at younger age and prior to slaughter, more birds approach an observer in the evening. Both, approach and avoidance peaked at three weeks and decreased prior to slaughter. Further, the effect of differing observer locations in the chicken house on the behavioural response of broiler chicken was studied. A higher proportion of birds voluntarily approached an unknown human in the area close to the main entrance door, but the avoidance behaviour was not affected.From the results of this study I conclude, that the time of assessment has no effect on avoidance or approach behaviour of commercial slow-growing broiler chicken. However, fear related behaviour changed depending on age. This makes the applied tests potentially applicable independent of daytime restrictions when performed with birds of the same age.

Foram utilizados 560 pintos machos (Cobb), com um dia de idade, distribuídos aleatoriamente em tratamentos: controle (dieta vegetal), os demais tratamentos tiveram a inclusão de 0,5; 1,0; 1,5; 2,0; 2,5; 3,0; 3,5; 4,0 e 8,0% de FV na dieta. A cada 7 dias foram separadas ao acaso oito aves (n = 8) por tratamento e coletados sangue e músculo peitoral para análise da razão isotópica do carbono e nitrogênio. Tanto no sangue quanto no músculo peitoral das aves aos 7 dias de idade a FV foi detectada a partir de 3,5%, valor que é mantido para o sangue aos 14 dias de idade. No músculo peitoral aos 14 dias a detecção foi a partir de 4,0%. Com 21 dias de idade no sangue a FV foi detectada a partir 2,0% e no músculo peitoral acima de 0,5% de inclusão. Aos 28, 35 e 42 dias no sangue foi detectado a partir de 4,0; 2,5 e 3,0%; respectivamente. No músculo peitoral aos 28 e 35 dias de idade foi identificada a FV em nível a partir de 2,0% e oas 42 dias a partir de 1,5%. A coleta de amostras ao longo do período de criação é necessária para acompanhar a variação do nível de inclusão de FV na alimentação de frangos de corte, devido as mudanças de dieta de acordo com as fase de criação de aves. O músculo peitoral comparado ao sangue das aves identifica a FV na alimentação de frangos de corte em menores níveis. A análise estatística linear discriminante é boa ferramenta para identificação de menores níveis de inclusão de FV na alimentação de frangos de corte comparada a análise estatística manova / The aim of this paper was to identify the increasing levels of poultry offal meal (POM) in feed for broiler chickens using stable isotopes of carbon and nitrogen in the blood analysis and breast muscle of birds at each week. They were used 560 male chicks (Cobb), one day age. They were randomly assigned to treatments: control (vegetable diet), all other treatments had the inclusion of 0.5, 1.0; 1.5; 2.0; 2.5; 3.0; 3.5; 4.0 and 8.0% of POM in the diet. For analysis of the isotope ratio carbon and nitrogen at every 7 days were separated at random eight birds (n = 8) per treatment and collected blood and breast muscle. Both in blood as in breast muscle at 7 days old POM was detected at 3.5%, which is maintained for the blood at 14 days old. In the breast muscle at 14 days the detection was from 4.0%. Within 21 days of age in the POM blood was detected from 2.0% in the breast muscle and above the 0.5% inclusion. At 28, 35 and 42 days in the blood was detected from 4.0, 2.5 and 3.0%, respectively. In the breast muscle at 28 and 35 days of age was identified in the POM level from 2.0% and 42 days from 1.5%. The collection of samples throughout the growing period is needed to monitor the level of inclusion variação POM in feed for broiler chickens, because of changes in diet according to the phase of poultry. The breast muscle compared to the blood of birds identified the POM in the supply of broilers at lower levels. Statistical analysis of linear discriminant is a good tool for identification of lower levels of inclusion of POM in feed for broiler chickens compared to Manova statistical analysis
